Regulation of BAP1 tumor suppressor complex by post-translational modifications

Le régulateur transcriptionnel BAP1 est une déubiquitinase nucléaire (DUB) dont le substrat est l’histone H2A modifiée par monoubiquitination au niveau des residus lysines 118 et 119 (K118/K119). Depuis les dernières années, BAP1 emerge comme un gene suppresseur de tumeur majeur. En effet, BAP1 est inactivé dans un plethore de maladies humaines héréditaires et sporadiques. Cependant, malgré l’accumulation significative des connaissances concernant l’occurrence, la pénétrance et l’impact des défauts de BAP1 sur le développement de cancers, ses mécanismes d’action et de régulation restent très peu compris. Cette étude est dédiée à la caractérisation moléculaire et fonctionnelle du complexe multi-protéique de BAP1 et se présente parmi les premiers travaux décrivant sa régulation par des modifications post-traductionnelles. D’abord, nous avons défini la composition du corps du complexe BAP1 ainsi que ses principaux partenaires d’interaction. Ensuite, nous nous sommes spécifiquement intéressés a investiguer d’avantage deux principaux aspects de la régulation de BAP1. Nous avons d’abord décrit l’inter-régulation entre deux composantes majeures du complexe BAP1, soit HCF-1 et OGT. D’une manière très intéressante, nous avons trouvé que le cofacteur HCF-1 est un important régulateur des niveaux protéiques d’OGT. En retour, OGT est requise pour la maturation protéolytique de HCF-1 en promouvant sa protéolyse par O-GlcNAcylation, un processus de régulation très important pour le bon fonctionnement de HCF-1. D’autre part, nous avons découvert un mécanisme unique de régulation de BAP1 médiée par l’ubiquitine ligase atypique UBE2O. en effet, UBE2O se caractérise par le fait qu’il s’agit aussi bien d’une ubiquitine conjuratrice et d’une ubiquitine ligase. UBE2O, multi-monoubiquitine BAP1 au niveau de son domaine NLS et promeut son exclusion du noyau, le séquestrant ainsi dans le cytoplasme. De façon importante, nos travaux ont permis de mettre de l’emphase sur le rôle de l’activité auto-catalytique de chacune de ces enzymes, soit l’activité d’auto-déubiquitination de BAP1 qui est requise pour la maintenance de sa localisation nucléaire ainsi que l’activité d’auto-ubiquitination d’UBE2O impliquée dans son transport nucléo-cytoplasmique. De manière significative, nous avons trouvé que des défauts au niveau de l’auto-déubiquitination de BAP1 due à des mutations associées à certains cancers indiquent l’importance d’une propre regulation de cette déubiquitinase pour les processus associés à la suppression de tumeurs. / BAP1 is a nuclear deubiquitinating enzyme (DUB) that acts as a transcription regulator and a DUB of nucleosomal histone H2AK119. In the recent years, it has become clear that BAP1 is a major tumor suppressor, inactivated in a plethora of hereditary and sporadic human malignancies. Although, we now accumulated a significant body of knowledge in respect to the occurrence, penetrance and impact of BAP1 disruption in cancer, its mechanism of action and regulation remained poorly defined. This work is dedicated to the biochemical and functional characterization of the BAP1 multiprotein complex and presents one of the first cases regarding its regulation by post-translational modifications. First, we defined the initial composition of the BAP1 complex and its main interacting components. Second, we specifically focused on two aspects of BAP1 regulation. We described the cross regulation between the two major components of the complex namely HCF-1 and OGT. We found that HCF-1 is important for the maintenance of the cellular levels of OGT. OGT, in turn, is required for the proper maturation of HCF-1 by promoting O-GlcNAcylation-mediated limited proteolysis of its precursor. Third, we discovered an intricate regulatory mechanism of BAP1 mediated by the atypical ubiquitin ligase UBE2O. UBE2O multi-monoubiquitinates BAP1 on its NLS and promotes its exclusion from the nucleus. Importantly, our work emphasises the role of the autocatalytic activity of both enzymes namely the auto-deubiquitination activity of BAP1, required for the maintenance of nuclear BAP1 and the auto-ubiquitination of UBE2O implicated in its nucleocytoplasmic transport. Significantly, we found that auto-deubiquitination of BAP1 is disrupted by cancer-associated mutations, indicating the involvement of this process in tumor suppression.

1 Abstract Trademarks and their territorial limits The purpose of my thesis is to analyze the ways that are used for overcoming territorial limits of trademarks, to point out the pros and cons of these methods. Likewise, I'd like to point out the fact that the various systems of trademarks - national and international systems and system of Community trade mark are interconnected and they complete each other. Therefore, it is necessary to focus on the quality of national legislation, to match the European Union standards and international standards, as well. I chose this topic, because the issue of trademarks is in my opinion very interesting, I like the fact that they can connect designers and businessmen, I am fascinated by their overlap into the art sphere, too. The topic of marks is very broad and quite specific, for understandable reasons it cannot be given so much time within the subjects taught at the Faculty. Also this was one of the reasons I chose this topic. My work is divided into six major thematic units, together there are 25 chapters, some of them are further divided into subchapters and sections. Jackson Pollock, 1930-1955 : the influence of the Old Masters

The imagery in Jackson Pollock's three extant sketchbooks which date from c.1934-1939 is dependent on that of other artists, especially El Greco, Rubens and Tintoretto. By 1947 however, the painter achieved a mature synthesis, distinctly his, which influenced contemporary painting, and was seminal for the work of a number of artists of the succeeding era. This dissertation is an attempt to document the phases of Pollock's artistic style from the early 1930s through to the middle 1950s, and to investigate the forces which may have catalyzed his temperament and precipitated his late style. The early sketchbooks begun in c.1934 represent Pollock's engagement with the art of the Old Masters and the teaching techniques of Thomas Hart Benton that utilized works from the Renaissance. The third sketchbook from c.1937-1939 induced him to re-examine the work of the Old Masters in a dialectical approach which incorporated new masters with old, but remained preoccupied with the sacred imagery found in the first two books. It is a resolution of these seemingly opposing modes of representation which produced several influential paintings in the early 1940s, including Guardians of the Secret and Pasiphae. At the same time these works display structural emulations related to those of Old Master paintings that would become increasingly prominent in Pollock's art. The canvases of 1947-1950, produced in what is commonly termed the “Classic Poured Period,” appear to represent a quantum leap beyond the concerns of Old Master works and European precedents. By this point Pollock had developed a fluency and assurance in his use of color and line that seems to extend further than the studied paradigmatic repetitions of his early sketchbooks. However, despite the radically new technique his paintings still exhibit pictorial and formal infrastructures derived from Renaissance paintings which were absorbed into Pollock's new idiom with surprising ease. In 1951 Pollock enters what Francis V.O'Connor termed as ‘his fourth phase'. The Black paintings of 1951-1953 betray a further exploration and adaptation of Old Master ideas, both iconographic and aesthetic and were created in Triptychs and Diptychs, typical altarpiece formats. With these paintings Pollock's forms acquired a confident plasticity and invention derived from the sculptural practices of Michelangelo, and progressively fewer individual images are quoted verbatim. An understanding of Pollock's early preoccupation with old Master painting is essential to comprehend the formation of the aesthetics of much of his later art. Significantly the underlying infrastructure remains fixed to old Master precedents and it was precisely these models of Renaissance and Baroque art which became the medium through which his mature synthesis was achieved.

A Multidisciplinary Approach for Determining the Extents of the Beds of Complex Natural Lakes in Louisiana

In Louisiana, the beds of natural lakes are owned by the state. The locations of property boundary lines separating state property from private lands have usually been set by determining the levels of natural monuments known as ordinary high water marks. The term is confusing and subjective, leading to controversy in reference to its determination. Catahoula Lake in central Louisiana was chosen as a study site because of its large size, its 20-foot variation in water levels, and its low-relief perimeter. Geology, geomorphology, hydrology, archaeology, vegetation, dendrochronology and dendrohydrology of the ancient cypress fringe, nineteenth century land survey records, historical records, and time-series statistics were applied to determine the elevation range of ordinary high water. Research suggests that the level of the ordinary high water natural monument used universally in Louisiana to define lake boundaries is not the correct natural monument. This research suggests that, for example, the natural monument that defines the Catahoula Lake boundary is not a vertical monument, but rather an areal monument that was originally recognized by nineteenth century U.S. government surveyors and embedded in the evidence extracted from their original field notes. The solution coincides with the boundaries of regional land patents and offers a consistent solution to determining the boundaries of thousands of acres of disputed lands. The procedure is applicable to other lake and swamp boundaries in Louisiana and other states.

This thesis aims to analyze and interpret a number of central works – Vimpelstaden (1985), Som en gång varit äng (1988), Blomman och människobenet (2003), I en cylinder i vattnet av vattengråt (2005) and Vad hjälper det en människa om hon häller rent vatten över sig i alla sina dagar (2009) – by the Swedish poet Ann Jäderlund (1955-) in the context of phenomenology and affect theory. The analysis consists of three chapters and proceeds chronologically with technical scrutinies of separate phases of Jäderlund’s œuvre – from the aphasic-like treatment of established phraseologies in Vimpelstaden and frozen expressions of the botanical discourse in Som en gång varit äng, to the uncanny focus on perceptual patterns as such in her later works. Throughout these analyses the thesis observes a series of techniques with which the author presents us with a kind of sensory paradox, through a) creating language-based complex appearances, non-appropriable by means of the normal perceptual patterns of embodied perception, while still b) simulating, and thus implicitly emphasizing, these appearances as something already concretely looked at and felt. In short, to experience what cannot be experienced, to live the unlivable. Many of these technical observations made are pinned down analytically using concepts from the field of cognitive poetics, namely George Lakoff and Mark Johnsons findings of experiential image schemata underpinning spoken phraseologies and their influential theories on conceptual metaphors. The interpretative conclusion following these observations is that Jäderlund handles her writing aesthetically as a kind of sensory material in a very literal sense, a “being of sensation” in the terminology of French philosophers Gilles Deleuze and Felix Guattari. Not as means of experiential or intellectual representation, not as some sort of critical enterprise through mere language-gaming of free- floating signifiers – but as a material able to preserve and perform sensory processes immanent to its own material compilation, a tendency that earlier research fails to grasp or simply ignores altogether. Thus the affectivity immanent to the literary material – often being the starting point of studies in affect theory and cognitive poetics – is here proven to be a characteristic, thereby playing the role more of a conclusion than a field of inquiry. The aesthetics of interrogating the limits of sensory experience, introducing a sort of crisis to embodied perception through the experience of poetic language – and the experience of it as having a “metaphysical significance”, as French phenomenologist Maurice Merleau- Ponty puts it – is articulated in the thesis against the background of influential readings of modern art carried out by Merleau-Ponty and Deleuze.

Une question fondamentale en écologie des populations est l’identification des facteurs influençant la dynamique d’une population. L’objectif principal de cette thèse est de déterminer quelles sont les réponses démographiques et populationnelles de prédateurs marins supérieurs face aux fluctuations d’abondance de leurs proies, aux variations climatiques, à la densité-dépendance tout en tenant compte de l’hétérogénéité inter et intra-individuelle (âge, expérience, sexe, qualité ou stratégie). Pour ce faire, nous nous baserons sur l’analyse de suivis à long-terme individuels et populationnels d’oiseaux marins longévifs et prédateurs apicaux phylogénétiquement très proches dans deux biomes contrastés : le labbe de McCormick Catharacta maccormicki sur l’archipel de Pointe Géologie en Antarctique et le labbe subantarctique Catharacta lonnbergi sur l’archipel des Kerguelen en milieu subantarctique. Nous tirerons parti d’estimations d’abondances de leurs proies respectives : le manchot Adélie Pygoscelis adeliae et le manchot empereur Aptenodytes forsteri en Antarctique, et le pétrel bleu Halobaena caerulea et le prion de Belcher Pachyptila belcheri à Kerguelen. Ces jeux de données offrent une opportunité unique de pouvoir déterminer et quantifier simultanément les différentes sources de variabilité dans les changements de taille de populations naturelles occupant l’un des niveaux trophiques les plus élevés des réseaux alimentaires antarctiques et subantarctiques. Nous avons mis en évidence de la variation dans plusieurs traits vitaux des deux populations influencées par les performances des individus et de l’hétérogénéité individuelle latente. Nous discutons des mécanismes par lesquels la variabilité climatique, l’abondance de proie et la densité de population peuvent affecter différentiellement les différentes classes d’âges de chaque trait vital, et les conséquences potentielles de futurs changements environnementaux. / A fundamental endeavor in population ecology is to identify the drivers of population dynamics. The main objective of this thesis is to determine what are the demographic and population responses of superior marine predators to the fluctuations of their prey abundance, to climatic variations, to density-dependence while taking into account inter and intra individual heterogeneity (age, experience, sex, quality or strategy). To do this, we analysed long-term individual and population-based monitoring of long-lived seabirds and phylogenetically close apical predators in two contrasting biomes: the south polar skua Catharacta maccormicki at Pointe Géologie archipelago, Antarctica, and the brown skua Catharacta lonnbergi on the sub-Antarctic Kerguelen Archipelago. We will use direct abundance of their respective prey: Adélie penguin Pygoscelis adeliae and emperor penguin Aptenodytes forsteri in Antarctica, and the blue petrel Halobaena caerulea and the thin-billed prion Pachyptila belcheri prion in Kerguelen islands. These datasets provide a unique opportunity to simultaneously disentangle and quantify the different sources of variability driving variation in natural populations occupying one of the highest trophic levels of the Antarctic and sub-Antarctic food webs. We found variation in several vital traits of both populations influenced by individual performance and latent individual heterogeneity. We discuss the mechanisms by which climatic variability, prey abundance, and population density can differentially affect the different age classes of each age class, and the potential consequences of future environmental changes.

Nonviolent atonement : a theory -praxis appraisal of the views of J Denny Weaver and S Mark Heim

Violence in traditional “satisfaction” atonement theologies is addressed here. An alternative non-violent view follows in discussion with Weaver / Heim. Weaver outlines a nonviolent Jesus narrative focussing on God’s rule made visible in history. Jesus’ saving death stems not from God but Jesus’ opposing evil powers. For viability violent biblical texts are disregarded. Church history interpretation is nonconventional. Early church is nonviolent. The subsequent Constantinian “fall” births the violent satisfaction model. Weaver’s problematical violence definition receives attention. Girard’s scapegoating philosophy and Jesus’ rescuing humankind from this evil undergirds Heim’s approach. Scapegoating establishes communal peace preventing violence. The bible is antisacrificial giving victims a voice. Jesus becomes a scapegoating victim, yet simultaneously exposes and reverses scapegoating, his death stemming from evil powers not God. Nonviolent atonement influences numerous theological concepts with Incarnational theology demonstrating Jesus’ humanness impacting upon atonement. 台灣消費者物價指數的預測評估與比較 / The evaluations and comparisons of consumer price index's forecasts in Taiwan

本篇論文擴充Ang et al. (2007)之基本架構,分別建構台灣各式月資料與季資料的物價指數預測模型,並進行預測以及實證分析。我們用以衡量通貨膨脹率的指標為 CPI 年增率與核心CPI 年增率。我們比較貨幣模型、成本加成模型、6 種不同設定的菲力浦曲線模型、3 種期限結構模型、隨機漫步模型、 AO 模型、ARIMA 模型、VAR 模型、主計處(DGBAS)、中經院(CIER) 及台經院(TIER) 之預測。藉由此研究,我們可以完整評估出文獻上常用之各式月資料及季資料預測模型的優劣。 我們實證結果顯示,在月資料預測模型樣本外預測績效表現方面, ARIMA 模 型對 2 種通貨膨脹率指標的樣本外預測能力表現最好。至於季資料預測模型樣本外預測績效表現, ARIMA 模型對未來核心 CPI 年增率的樣本外預測能力表現最好; 然而,對於 CPI 年增率為預測目標的預測模型則不存在最佳的模型。此外,實證分析中我們也發現本研究所建構的模型預測表現仍遜於主計處的預測,但部份模型的樣本外預測能力表現則比中經院與台經院的預測為佳。 / This paper compares the forecasting performance of inflation in Taiwan. We conduct various inflation forecasting methods (models) for two inflation measures(CPI growth rate and core-CPI growth rate) by using monthly and quarterly data. Besides the models of Ang et al. (2007), we also consider some macroeconomic models for comparison. We compare some Monetary models, Mark-up models, six variants of Phillips curve models, three variants of term structure models, a Random walk model, an AO model, an ARIMA model, and a VAR model. We also compare the forecast ability of these model with three different survey forecasts (the DGBAS, CIER, and TIER surveys). We summarized our findings as follows. The best monthly forecasting model for both inflation measures is ARIMA model. For quarterly core-CPI inflation, ARIMA model is also the best model; however, when comparing the quarterly forecasts for CPI inflation, there does not exist the best one. Besides, we also found that the DGBAS survey outperforms all of our forecasting methods/models, but some of our forecasting models are better than the CIER and TIER surveys in terms of MAE.
