Tumour-selective apoptosis : identification of NMHCIIa as novel death receptor interactor regulating the response to TRAIL

The cytokine TRAIL is a promising cancer therapeutic candidate as it induces apoptosis selectively in transformed cells. TRAIL-induced clustering of its receptors (DR) is essential for the DISC complex formation, which induces cell death. The mechanism for TRAIL's tumour selective effect is largely unknown. We identified the cytoskeleton proteins non-muscle myosin heavy chain IIa, IIb (NMHCIIa, NMHCIIb), myosin regulatory light chain (MLC2) and ß-actin as novel DR-interactors. An initially weak and TRAIL-induced abrogation of NMHCII/DR interaction correlated with efficient DISC formation in tumour cells. In contrast, a robust NMHCII/DR interaction that was sustained upon TRAIL stimulus was accompanied by incomplete DISC arrangement. Weakening the NMHCII/DR interaction in normal cells using chemical inhibitors enhanced TRAIL-induced apoptosis. Intriguingly, siRNA-mediated NMHCIIa- but not NMHCIIb depletion potently released TRAIL resistance in normal cells and influenced DISC composition. Reduced NMHCII/DR interaction in transformed cells was characterised by diminished MLC2 phosphorylation and altered protein expression of upstream regulatory kinases. Our results suggest that normal cell resistance to TRAIL-apoptosis is based on the interaction of cytoskeleton components with DR that is impaired upon transformation. Since NMHCII function in cell adhesion and migration, it will be interesting to study possible roles of the interaction in cell detachment and altered TRAIL sensitivity; moreover this link may provide clues as to the cause of TRAIL resistance in some cancers.

INTRODUÇÃO: A nefrite lúpica é uma complicação frequente e de alta morbimortalidade do lúpus eritematoso sistêmico (LES). A evolução para insuficiência renal crônica terminal varia entre 8 e 15% dos casos, após um período de 5 anos. A fase inicial da nefrite se deve a uma atividade imunológica exacerbada que leva a sequelas renais, como a fibrose intersticial, sinéquias glomerulares, e glomeruloesclerose. Uma vez instalada, vários fatores aceleram a velocidade de progressão da insuficiência renal, como a presença de proteinúria residual, hipertensão arterial sistêmica e a etnia do paciente. Estudos recentes mostraram que a presença de polimorfismos do MYH9 são altamente prevalentes em pacientes com GESF (glomeruloesclerose focal e segmentar), nefropatia do HIV e em pacientes com doença renal crônica não diabética. Os polimorfismos do MYH9 mais relacionados com essas doenças são os do haplótipo E1, causados pelos polimorfismos rs4821480, rs2032487, rs4821481 e rs3752462, presentes principalmente na população negra e de hispano-americanos. No Brasil não há estudos sobre a prevalência desse gene. MÉTODOS: Nosso estudo analisou retrospectivamente 196 pacientes com nefrite lúpica, acompanhadas no ambulatório de glomerulopatias do Hospital das Clínicas da USP. Foram recuperados os dados clínicos e laboratoriais dos pacientes de janeiro de 1999 a dezembro de 2010. Foi feita análise dos polimorfismos do haplótipo E1 do gene do MYH9 (rs4821480, rs2032487, rs4821481 e rs3752462) e correlacionados com suas características clínicas e laboratoriais, apresentando como desfecho a duplicação da creatinina ou a evolução para doença renal crônica terminal. RESULTADOS: O tempo de seguimento médio dos pacientes foi de 6,1 anos, com a creatinina inicial média de 1,6 g/dL e proteinúria média de 3,9 g/dia. Dezenove pacientes não recuperaram função renal, mantendo-se em diálise. Dos 177 pacientes restantes 43 (24%) apresentaram o desfecho de duplicação (DC) da creatinina, ou necessidade de diálise (DRCT). Pacientes progressores eram tinham maior SLEDAI renal (10 vs 8,9 p=0,04), maior índice de cronicidade renal à biópsia (5 vs 2, p<0,001) e maior frequência de reativações da doença renal (flare renal) (82,9% x 53,8%, p=0,002), assim menores índices de remissão completa ou parcial (p<0,0001). Os 4 polimorfismos se segregam em conjunto, ou seja, como um haplótipo, pelo modelo de Hardy-Weinberg. Analisando separadamente cada polimorfismo, apenas o rs3752462, apresenta associação com o desfecho DC/DRCT, na análise por genótipo (CC/CT/TT, p=0,03) e quando feita análise TT/CT vs CC (p=0,02). Não houve relação dos polimorfismos com a etnia negra ou parda. Pacientes com haplótipo E1 eram progressores em 28% dos casos, conferindo um OR de 1,79 (IC 1,02 a 3,0) de DC/DRC. DISCUSSÃO: A presença do haplótipo E1 têm alta prevalência em pacientes portadores de nefrite lúpica no Brasil, sendo fator de risco para progressão da doença renal crônica / BACKGROUND: Lupus nephritis (LN) is a frequent complication with high morbidity and mortality of systemic lupus erythematosus (SLE). Chronic renal failure is observed in 8 to 15% of the patients after 5 years of follow up. LN is an inflammatory disease after a systemic autoimmune activation. Once inflammation is shutdown several renal and nonrenal factors, such as residual proteinuria, hypertension and ethnicity of the patient, may emerge and impose to the kidney a chronic phenotype (interstitial fibrosis, glomerular adhesions and glomerulosclerosis. Recently E1 haplotype (rs4821480, rs2032487, rs4821481 and rs3752462 polymorphisms) of the MYH9 gene was associated to progressive kidney diseases in patients with FSGS (focal segmental glomerulosclerosis), HIV nephropathy and non-diabetic chronic kidney disease, in african american and spanic american patients. In Brazil there is no data on this subject. METHODS: Retrospective analysis of 196 patients with LN followed in our outpatient glomerular disease ward were enrolled glomerulopathies. Patients clinical data from January 1999 to December 2010 were retrieved and MYH9 rs4821480, rs2032487, rs4821481 and rs3752462 polymorphisms were genotyped. Outcome was defined as doubling of serum creatinine, or end stage renal disease (ESRD). RESULTS: The mean follow-up of patients was 6.1 years, with an initial mean creatinine of 1.6 g/dL and mean proteinuria 3.9 g/day. On enrollment nineteen patients were on dialysis and did not recover renal function, they were withdraw from analyses of progressive kidney disease. On follow up, from 177 remaining patients, 43 (24%) showed the composite outcome: dialysis, or doubling creatinine. Progressors had higher renal SLEDAI (10 vs 8.9, p = 0.04), higher chronicity index at biopsy (5 vs 2, p <0.001) and more frequently renal flares (82, 9% vs. 53.8%, p=0.002), as well as lower rates of complete or partial remission (p <0.0001). The four polymorphisms segregate as a haplotype, according the Hardy-Weinberg model. Analysing each polymorphism, only TT/CT genotype from rs3752462 polymorphism was associated with the outcome of DC/ESRD (p = 0.02). E1 haplotype were associated with progression with an OR of 1.79 (CI 1.02 to 3.0). DISCUSSION: The presence of the E1 haplotype is associated with worse prognosis of chronic renal failure in lupus nephritis patients

A isoforma 3 do trocador Na+ /H+ (NHE3), presente em membrana apical, é a proteína de transporte que medeia a maior parte da reabsorção de NaCl e NaHCO3- em túbulo proximal renal. A fosforilação direta do NHE3 por PKA na serina 552 é um dos mecanismos pelos quais a sua atividade pode ser inibida. A ligação da angiotensina II (Ang II) ao receptor AT1 (AT1R) em túbulo proximal estimula a atividade do NHE3 por diferentes vias de sinalização. Entretanto, não foram ainda bem estabelecidos os efeitos da ativação da via AT1R/Gi, com consequente diminuição nos níveis de cAMP, na regulação do NHE3. A Ang II pode ainda estimular a atividade do NHE3 por promover a sua translocação da base para o corpo das microvilosidades, entretanto, o papel da proteína motora miosina IIA nesta translocação em resposta à Ang II ainda não foi estabelecido. Sendo assim esta tese teve como objetivos: (1) testar a hipótese de que a Ang II diminui os níveis de fosforilação do NHE3 mediados pelo cAMP/PKA na serina 552 aumentando a sua atividade por reduzir os níveis de cAMP e (2) testar a hipótese de que a miosina IIA participa da redistribuição do NHE3 da base para o corpo das microvilosidades em túbulo proximal renal em condições de estímulo da reabsorção de sódio, como ocorre em resposta à Ang II. Visando avaliar os efeitos da ativação da via AT1R/Gi na regulação do NHE3, verificamos, por meio da técnica de recuperação do pH dependente de Na+, que, em condições basais, a Ang II estimulou a atividade do NHE3, mas não alterou a atividade da PKA e nem afetou os níveis de fosforilação do NHE3 na serina 552 em uma linhagem de células de túbulo proximal (OKP). Entretanto, na presença da forskolin (FSK), agente que eleva os níveis intracelulares de cAMP, a Ang II foi capaz de contrapor-se ao efeito inibitório da FSK sobre o NHE3 por promover redução na concentração de cAMP, diminuição da atividade da PKA e, consequentemente, diminuição nos níveis de fosforilação da serina 552. Todos os efeitos da Ang II foram bloqueados quando um pré-tratamento com Losartan, antagonista do receptor AT1, foi feito nas células OKP, destacando a contribuição da via AT1R/proteína Gi no aumento da atividade do NHE3 pela Ang II. Observamos que a inibição da proteína Gi com PTX (toxina pertussis) diminuiu a atividade do NHE3 em células OKP e que a PTX diminuiu a atividade do NHE3 assim como preveniu o efeito estimulatório da Ang II sobre a atividade do NHE3 em túbulo proximal de ratos Wistar. Adicionalmente, com a intenção de avaliar os efeitos da miosina IIA na redistribuição do NHE3, constatamos que a blebistatina, inibidor da miosina IIA, preveniu completamente o aumento de atividade do NHE3 mediado pela Ang II em ratos Wistar e que o uso da blebistatina foi capaz de prevenir o aumento do NHE3 na superfície de células OKP tratadas com Ang II. Em conjunto, nossos resultados sugerem que a Ang II contrapõe-se aos efeitos do cAMP/PKA sobre a fosforilação e a atividade do NHE3 pela ativação da via AT1R/Gi e que a miosina IIA desempenha um papel na mediação da regulação da atividade do NHE3 em túbulo proximal renal de ratos em resposta à Ang II. Sugerem ainda que a desfosforilação do NHE3 na serina 552 pode representar um evento chave na regulação do manuseio de sal tubular proximal pela Ang II na presença de hormônios natriuréticos que promovem o aumento dos níveis de cAMP e da fosforilação do transportador e que a miosina IIA está envolvida na regulação do tráfego do NHE3 em túbulo proximal renal / The Na+/H+ exchanger isoform 3 (NHE3), expressed on the apical membrane, is responsible for most NaCl and NaHCO3 - reabsorption in the renal proximal tubule. Direct phosphorylation of NHE3 by PKA at serine 552 is one of the mechanisms by which its activity is inhibited. Binding of angiotensin II (Ang II) to the AT1 receptor (AT1R) in the proximal tubule stimulates NHE3 activity through multiple signaling pathways. However, the effects of AT1R/Gi activation and subsequent decrease in cAMP accumulation on NHE3 regulation are not well established. Ang II can also stimulate NHE3 activity by promoting its translocations from the base to the body of the microvilli, however, the role of the myosin IIA motor protein in this translocation in response to Ang II is not yet established. Therefore, the aims of this thesis are: (1) to test the hypothesis that Ang II decreases the cAMP/PKA-mediated NHE3 phosphorylation levels at serine 552 increasing its activity by reducing cAMP levels and (2) to test the hypothesis that myosin IIA participates in the NHE3 redistribution from the base to the body of the microvilli in the renal proximal tubule under conditions in which sodium reabsorption is stimulated, such as in response to Ang II. In order to evaluate the effects of AT1R/Gi pathway activation on NHE3 regulation, by means the intracellular pH recovery technique, we verified that under basal conditions, Ang II stimulated NHE3 activity but did not affect PKA-mediated NHE3 phosphorylation at serine 552 in opossum kidney (OKP) cells. However, in the presence of the cAMP-elevating agent forskolin (FSK), Ang II counteracted FSK-induced NHE3 inhibition, reduced intracellular cAMP concentrations, lowered PKA activity, and prevented the FSK-mediated increase in NHE3 serine 552 phosphorylation. All effects of Ang II were blocked by pretreating OKP cells with the AT1R antagonist Losartan, highlighting the contribution of the AT1R/Gi pathway in Ang II-mediated NHE3 upregulation under cAMP-elevating conditions. We also verified that Gi protein inhibition by pertussis toxin treatment decreased NHE3 activity both in vitro and in vivo and, more importantly, prevented the stimulatory effect of Ang II on NHE3 activity in Wistar rat proximal tubules. Additionally, we assessed the effects of myosin IIA on NHE3 redistribution, and found that blebbistatin, a myosin IIA inhibitor, completely prevented the increase of Ang II-mediated NHE3 activity in Wistar rats and that blebbistatin was able to prevent the increase of NHE3 on the Ang II-treated OKP cells surface. Collectively, our results suggest that Ang II counteracts the effects of cAMP/PKA on NHE3 phosphorylation and inhibition by activating the AT1R/Gi pathway and that myosin IIA plays a role in mediating the NHE3 activity regulation in the rat renal proximal tubule in response to Ang II. Furthermore, these findings support the notion that NHE3 dephosphorylation at serine 552 may represent a key event in the regulation of renal proximal tubule sodium handling by Ang II in the presence of natriuretic hormones that promote cAMP accumulation and transporter phosphorylation, and that myosin IIA is involved in NHE3 trafficking regulation in the renal proximal tubule

Characterizing mechanical properties of living C2C12 myoblasts with single cell indentation experiments : application to Duchenne muscular dystrophy / Caractérisation expérimentale par indentation des propriétés mécaniques de myoblastes : application à la dystrophie musculaire de Duchenne

Cette thèse interdisciplinaire a été dédiée à la caractérisation des propriétés mécaniques de myoblastes (murins et humains) et de myotubes (murins) à l'aide de la microscopie à force atomique (AFM). En modifiant ou en inhibant la dynamique du cytosquelette (CSK) d’actine de ces cellules, nous avons pu montrer que ces propriétés mécaniques variaient. L’enregistrement de courbes de force indentation nous a permis de montrer que la présence de cellules adhérentes introduisait sur les leviers d’AFM un amortissement visqueux supplémentaire à celui d’une paroi solide, et que cet amortissement visqueux dépendait de sa vitesse d’approche et que celui-ci restait non négligeable pour les plus faibles vitesses (1μm/s). Nous avons observé que les propriétés mécaniques des précurseurs de muscles devenaient non linéaires (comportement plastiques) pour des grandes déformations (>1μm) et qu’elles dépendaient de l’état, du type de cellule et de leur environnement. En combinant des expériences d’AFM, des modèles visco-élastiques et des méthodes d'analyse multi-échelle basées sur la transformation en ondelettes, nous avons illustré la variabilité des réponses mécaniques de ces cellules (de visco-élastiques à visco-plastiques). À l'aide de courbes de force-indentation, de l’imagerie morpho-structurale (DIC, microscopie à fluorescence) et de traitements pharmacologiques, nous avons éclairé le rôle essentiel des processus actifs (dépendants de l’ATP) dans la mécanique de myoblastes, en discutant tout particulièrement ceux des moteurs moléculaires (myosine II) couplés aux filaments d’actine. En particulier, nous avons montré que les fibres de stress du cytosquelette d’actine situées autour du noyau pouvaient présenter des évènements de remodelage soudains (ruptures) et que ces ruptures étaient une mesure indirecte de l’aptitude de ces cellules à tendre leur CSK. Nous avons enfin montré qu’il était possible de généraliser cette approche à des cas cliniques humains, en l’occurrence des myoblastes primaires de porteurs sains et de patients atteints de dystrophie musculaire de Duchenne, ouvrant la voie à des études plus larges sur d’autres types cellulaires et pathologies. / This interdisciplinary thesis was dedicated to the atomic force microscopy (AFM) characterization of the mechanical properties of myoblasts (murine and human) and myotubes (murine). We reported that the mechanical properties of these cells were modified when their actin cytoskeleton (CSK) dynamics was inhibited or altered. Recording single AFM force indentation curves, we showed that adherent layers of myoblasts and myotubes introduced on the AFM cantilever an extra hydrodynamic drag as compared to a solid wall. This phenomenon was dependent on the cantilever scan speed and not negligible even at low scan velocities (1μm/s). We observed that the mechanical properties of the muscle precursor cells became non-linear (plastic behaviour) for large local deformations (>1μm) and that they varied depending on the state, type and environment of the cells. Combining AFM experiments, viscoelastic modeling and multi-scale analyzing methods based on the wavelet transform, we illustrated the variability of the mechanical responses of these cells (from viscoelastic to viscoplastic). Through AFM force indentation curves analysis, morpho-structural imaging (DIC, fluorescence microscopy) and pharmacological treatments, we enlightened the important role of active (ATP-dependent) processes in myoblast mechanics, focusing especially on those related to the molecular motors (myosin II) coupled to the actin filaments. In particular, we showed that the perinuclear actin stress fibers could exhibit some abrupt remodelling events (ruptures), which are characteristic of the ability of these cells to tense their CSK. Finally, we showed that this approach can be generalized to some human clinical cases, namely primary human myoblasts from healthy donors and patients affected by Duchenne muscular dystrophy, paving the way for broader studies on different cell types and diseases.

Active gels in vivo : patterns and dynamics in cytokinetic rings and their functions in cell division / Gels actifs in vivo : structures et dynamiques dans l'anneau de cytokinètique et leurs fonctions dans la division cellulaire

Les structures d'acto-myosine sont impliquées dans de nombreuses fonctions cellulaires. Comprendre leur organisation et leur comportement collectif est toujours difficile. Nous avons étudié l'anneau cytokinétique dans les cellules de mammifères et dans les levures de fission, en orientant les cellules dans les microcavités, ce qui permet de voir l'anneau dans un seul plan focal. Avec cette configuration, nous révélons de nouvelles structures et des dynamiques distinctes pour les deux systèmes cellulaires. Dans les cellules de mammifères, nous trouvons des motifs réguliers de la myosine et la formine. Les caractéristiques de ces motifs sont stables tout au long de sa fermeture et leur apparition coïncide avec la constriction. Nous proposons que ce phénomène est une propriété inhérente du réseau d'acto-myosine et que la formation de ces motifs entraîne une augmentation du stress. Ces hypothèses sont confirmées par notre modèle en champ moyen. Par contraste, l'anneau de levure de fission montre des inhomogénéités tournantes de l'actine, de la myosine, des protéines de la construction de la paroi (Bgs) et d'autres protéines. La dynamique des inhomogénéités de myosine est inchangée, si la croissance de la paroi est inhibée. Cependant, l'inhibition du mouvement des inhomogénéités conduit à l'arrêt de la fermeture. Nous proposons que la fermeture de l'anneau est entraînée par la rotation de l'actine et de la myosine qui tirent des protéines Bgs, lesquelles construisent ainsi le septum. Cette hypothèse est confirmée par nos calculs et par des simulations numériques. Nous suggérons que la transition entre les états de différents ordres et dynamiques pourrait être une façon de réguler in vivo les systèmes d'acto-myosine. / Actomyosin structures are involved in many cell functions. Understanding their organization and collective behavior is still challenging. We study the cytokinetic ring in mammalian cells and in fission yeasts, by orienting cells in microcavities. This allows seeing the ring in a single plane of focus. With this setup, we reveal new structures and distinct dynamics for both cellular systems. In mammalian cells we find a pattern of regular clusters of myosin and formin. The characteristics of this pattern are stable throughout closure and its formation coincides with the onset of constriction. We propose that its characteristic is an inherent property of the actomyosin network and that its formation leads to an increase in stress generation. These hypotheses are supported by our theoretical mean field model. In contrast, fission yeast rings show rotating inhomogeneities (speckles), i.e. rotations of actin, myosin, cell wall building proteins (Bgs) and other proteins. Myosin speckles dynamic is unchanged, if wall growth is inhibited. However, the inhibition of speckle motion leads to stalled closure. We propose that the ring closure is driven by the rotation of actin and myosin, which pull Bgs thereby building the septum. This model is supported by our calculations and by numerical simulations. We suggest that the transition between states of different orders and dynamics might be a way to regulate actomyosin systems in vivo.

Novel prognostic biomarkers for renal cell carcinoma

Abstract Background and aims: Stage and grade are the most widely used prognostic parameters for renal cell carcinoma (RCC). The clinical course of this disease is not, however, always predictable by traditional prognostic factors. In the era of new molecular targeted therapies a more accurate prognostication of RCC patient survival is important for the individualization of treatment and follow-up of patients. Despite exhaustive research there are still no prognostic biomarkers for RCC in clinical practice. In order to find novel prognostic tissue markers for RCC, we examined the expression of 14 biomarkers involved in carcinogenesis and clarified their prognostic significance in RCC. Material and methods: Out of 189 consecutive patients who underwent surgery for kidney cancer at Oulu University Hospital in the 1990s, 152 patients with histologically verified RCC were included in this study. The stage distribution was 70 (46%), 12 (8%), 51 (34%) and 19 (12%) patients with stages I-IV, respectively. The majority of the tumours (83 tumours, 55%) were nuclear grade II and 5 (3%), 40 (27%) and 22 (15%) of the tumours were grades I, III and IV, respectively. Clinical and follow-up data were obtained from patient records, the Finnish Cancer Registry and on demand from the Population Register Centre of Finland. The biomarkers studied included markers of the oxidative and neuroendocrine systems as well as proteins related to cell adhesion and migration, invasion, metastasis, inflammation and immune responses. The expression of various biomarkers was characterized via immunohistochemical tests of archival tumour material. The staining intensity was compared to clinicopathological parameters and patient RCC-specific survival. Results: The 5-year RCC-specific survival was 77%. The expression of Toll-like receptor 9 (TLR9) was an independent marker of favourable RCC-specific survival whereas cytoplasmic myosin VI expression was found to be an independent prognostic factor of poor RCC-specific survival. Cell culture experiments showed how cyclooxygenase-2 (COX-2) expression is regulated by HuR in RCC. HuR and COX-2 immunoexpression were also related to decreased RCC-specific survival. Immunostaining of Keap1 was associated with advanced RCC and a marker of a poorer RCC-specific prognosis. The expression of different neuroendocrine markers was evaluated but we could not establish any prognostic value for them. Conclusions: In particular, TLR9, HuR and myosin VI can be regarded as promising novel prognostic biomarkers in RCC. Stage, however, is the most important single prognostic factor for RCC. / Tiivistelmä Munuaissyöpä on vuosikymmenten ajan jatkuvasti yleistynyt. Vaikka se diagnosoidaan nykyisin useimmiten sattumalöydöksenä vatsan alueen kuvantamistutkimuksissa ja hoitomenetelmät ovat viime vuosikymmenten aikana kehittyneet, munuaissyöpäkuolleisuus ei ole laskenut. Munuaissyövän ennusteen määrittäminen voi olla haasteellista. Perinteiset ennustetekijät, levinneisyys ja erilaistumisaste, eivät riitä selittämään kaikkien potilaiden taudinkulkua, eikä munuaissyövälle vielä ole kliinisessä käytössä ennusteellista merkkiainetta. Munuaissyöpähoitojen kehittyessä taudinkulun ennustaminen on yhä tärkeämpää, jotta potilaiden hoito ja seuranta voidaan yksilöidä. Tämän väitöskirjatyön tarkoituksena oli etsiä uusia ennusteellisia kudosmerkkiaineita munuaissyöpäkasvaimille. Väitöskirjatutkimus perustuu 1990-luvulla Oulun yliopistollisessa sairaalassa leikatun 152 munuaissyöpäpotilaan aineistoon. Lähes puolet aineiston kasvaimista edusti levinneisyysluokkaa I, ja yli puolet munuaissyöpäkasvaimista oli hyvin erilaistuneita (tumagradus I ja II). Tutkimuspotilaista kerättiin kattavat seurantatiedot. Leikkauksessa poistettujen munuaissyöpäkasvainten arkistomateriaalista tutkittiin eri merkkiaineiden ilmenemistä. Tutkitut merkkiaineet käsittivät oksidatiivisen ja neuroendokriinisen järjestelmän merkkiaineita sekä valkuaisaineita, jotka liittyvät keskeisiin syövän ominaisuuksiin, kuten solujen välisiin liitoksiin ja solujen liikkumiseen sekä etäpesäkkeiden syntymiseen. Lisäksi tutkittiin merkkiaineita, jotka liittyvät tulehdusreaktioihin ja immuunipuolustukseen. Väitöskirjatutkimus paljasti useita uusia kudosmerkkiaineita, joiden ilmeneminen munuaissyöpäkasvaimessa on yhteydessä potilaan ennusteeseen. Näistä merkittävimpiä ovat myosiini VI, joka liittyy syöpäkasvainten metastasointiin, sekä immuunipuolustuksessa vaikuttava Tollin kaltainen reseptori 9 (Toll-like receptor 9, TLR9). Molemmat merkkiaineet osoittautuivat itsenäisiksi ennustetekijöiksi munuaissyövässä. Muita ennusteeseen vaikuttavia merkkiaineita ovat tutkimuksen mukaan oksidatiivista stressiä aistiva Keap1 sekä immunologisiin reaktioihin liittyvä syklo-oksigenaasi 2 (COX-2) ja sen ilmenemistä säätelevä HuR.

Identification des marqueurs repérables par des capteurs spectroscopiques et significatifs des étapes clefs de la cuisson de viande et poisson / Identification of markers that can be recognised using spectroscopic sensors and which reflect key stages in the cooking of meat and fish

L’un des objectifs du programme de recherche Opticook est d’équiper les fours avec des capteurs spectroscopiques qui permettront de piloter de façon non intrusive la cuisson de viande et de poisson. Le développement de ces capteurs est basé sur l’hypothèse que les protéines de ces aliments sont de bons marqueurs du degré de cuisson. Dans ce contexte, dans un premier temps, plusieurs outils ont été mis en œuvre pour caractériser l’effet de différents barèmes de cuisson sur les propriétés macroscopiques (texture, couleur) d’échantillons de bœuf, de poulet et de dos de cabillaud. Puis, pour étudier les transformations que subissent ces échantillons à l’échelle moléculaire, nous avons utilisé la calorimétrie et différentes spectroscopies. La spectroscopie dans le domaine du visible nous a permis d’observer la dénaturation des hémoprotéines grâce à l’évolution de l’état (oxy à met ). Ces protéines sont responsables du changement de couleur observé entre 50 et 75 °C. Les spectroscopies dans le moyen infrarouge et de fluorescence ont été utilisées pour suivre les changements de structures associées à la dénaturation des protéines fibrillaires. La myosine, purifiée à partir du blanc de poulet, a été étudiée en fonction de la température par fluorescence. Les résultats nous ont montré que des évolutions de la structure protéique se produisent dès 50 °C. À l’échelle microscopique, nous avons réalisé des expériences sur des échantillons de bœuf par RMN bas champ (séquences d’impulsion et gradient de champ), et par tomographie de neutrons pour rendre compte de la contraction des fibres et de la libération du jus selon le degré de cuisson. L’analyse des images 3D enregistrées par tomographie de neutrons et le traitement des données de RMN montrent des changements importants de la morphologie et de la densité de fibres des échantillons après la cuisson à haute température. Pour associer les deux approches (moléculaire et microscopique) des mesures couplées de spectroscopie et d’imagerie de neutrons ont été menées dans le four Opticook pendant la cuisson de tranches de bœuf. Ce couplage nous a permis de suivre l’évolution de la morphologie due à la contraction des fibres, la migration du jus, ainsi que le changement de couleur associé à la dénaturation des hémoprotéines. Nous en concluons qu’il est en effet possible de suivre le degré de cuisson de viande et de poisson à l’aide de différentes spectroscopies (la fluorescence et le visible) grâce aux signatures spectroscopiques de plusieurs marqueurs internes à ces matrices : la myosine, le collagène et les hémoprotéines. / One of the purposes of the Opticook project is to install spectroscopic sensors in ovens, so equipping them with non intrusive tools allowing following cooking process of meat and fish. The hypothesis on which sensors were developed was: are muscular proteins proper probes to discriminate among several cooking degrees? Thus, at the beginning of the project several tools were used to characterise effect of several cooking degrees on macroscopic properties (texture and colour) of beef, chicken and cod fillet samples. Following, calorimetry and spectroscopic techniques were used to study transformations at molecular scale. In particular, visible spectroscopy followed denaturation of haemproteins observing their evolution from oxy to met state. Colour change was observed between 50 and 75 °C. Middle infrared and fluorescence spectroscopies were used to observe evolution of fibrillar proteins which are associated with meat texture changes. One of myofibrils, myosin, was purified from chicken breast and was studied depending on temperature by fluorescence spectroscopy. Observations showed evolution of protein structure occurred around 50 °C. At microscopic scale, low field NMR (spin echo and gradient field configurations) and neutron tomography were used to observe morphology changes and juice release on beef samples according to cooking degree. 3D image analysis and NMR data showed morphology and fibre density changes at high temperature.We associated the two approaches (molecular and microscopic) in the last part of the research. So coupled measures were realised by combining neutron imaging and spectroscopy techniques during cooking process on beef slices by Opticook oven. Coupled measures allowed following morphology evolution because of fibre contractions, juice migration and colour modification as well associated to haemprotein denaturation. In conclusion, it is possible to follow cooking process of meat and fish by following several spectroscopic signatures (fluorescence and visible ones) of matrix own probes: myosin, collage and haemproteins.

Efeitos do treinamento físico por natação sobre o sistema cardiovascular e marcadores moleculares de hipertrofia cardíaca em ratas wistar / Swimming training effects on cardiovascular system and hypertrofic cardiac molecular markesrs in wistar females

O treinamento por natação leva a uma sobrecarga de volume no coração, que induz a hipertrofia cardíaca (HC) excêntrica, com aumento da massa e do diâmetro cardíaco. Neste trabalho foram investigadas as adaptações no sistema cardiovascular e na expressão de genes relacionados à HC patológica, na gênese da HC por treinamento de natação. 42 ratas wistar foram divididas em grupos: sedentário controle (SC) treinado protocolo 1 (P1) e treinado protocolo 2 (P2). O treinamento de P1 foi de 1x60min/dia, 5x/semana, por 10 semanas. O de P2 foi igual ao P1 até a 8ª semana. Na 9ª semana 2x/dia e na 10ª semana 3x/dia. Os grupos treinados, em relação ao SC, apresentaram bradicardia de repouso, melhora no desempenho físico do teste máximo e do consumo máximo de oxigênio e HC, sem alterar a pressão arterial média e a expressão dos genes do fator natriurético atrial e da alfa actina esquelética. O grupo P2 apresentou aumento no diâmetro cardíaco e redução da expressão do gene da beta miosina de cadeia pesada. Este último resultado é contrário à literatura para a HC patológica, que mostra o aumento não só da expressão deste gene como a dos outros genes estudados. Os resultados de HC de P2 assemelham-se aos encontrados em estudos recentes com atletas de modalidades de maior componente aeróbio, sendo este um bom modelo para investigação dos mecanismos envolvidos na HC destes atleta / Swimming training leads to a cardiac volume overload that induces excentric cardiac hypertrophy (CH) with an increase in cardiac mass and diameter. Cardiovascular system adaptations and expression of genes relatated with pathological CH were investigated in swimming training CH. We studied 42 wistar females, divided in sedentary control (SC) group, protocol 1 trained group (P1) and protocol 2 trained group (P2). The P1 training program was once a day for 5 times/week for 10 weeks. P2 was the same as P1 until 8th week. In 9th week it was twice a day and in 10th week 3 times a day. Trained groups, in contrast with SC, showed rest bradycardia, improvement in physical performance, maximum oxygen uptake and CH, with no alteration in the medium arterial pressure and in the expression of atrial natriuretic factor and skeletal alpha actin genes. Moreover, P2 showed an increase in cardiac diameter and decrease in the expression of beta myosin heavy chain gene. This expression result is different of patological CH literature wich shows an increase of this gene expression and also in the others genes we had investigated. P2 CH results were similar to those recently found in endurance-type athletes, sugesting this is a good model to investigate mechanisms involved in endurance-type athletes CH
