The aim of the work is to determine the noise pollution of the weekend house area around Dehtář. Receiver locations were chosen to find out noise levels in places with the highest intensity of human activity and at each observed source of noise. Noise levels were recorded in four seasons of the year. The equivalent continuous sound pressure levels were quantified. and compared with local noise law limits.

A expressão do receptor andrógeno em uma série de tumores de mama triplo-negativos

INTRODUÇÃO: Carcinomas de mama triplo-negativos (CMTN) constituem um grupo de tumores heterogêneos caracterizados por sobrevida pobre dos pacientes e falta de terapia-alvo. Receptor andrógeno (AR) tem sido descrito no CMTN, mas o impacto prognóstico da expressão nesse subgrupo não está bem claro. OBJETIVO: Investigar a associação do status de expressão AR analisado por imunohistoquímica em casos de CMTN com parâmetros clínicos (idade, sobrevida) e variáveis patológicas (tamanho do tumor, grau do tumor). MÉTODOS: Foram analisados 62 CMTN por imuno-histoquímica automatizada para receptor andrógeno. A imuno-histoquímica foi avaliada por dois diferentes patologistas e a expressão do biomarcador foi avaliada por H-Score (intensidade mais percentagem de coloração). Kaplan-Meyer foi o método usado para avaliar a sobrevida global, tendo as diferenças nas distribuições sido avaliadas com base na expressão do marcador. RESULTADOS: 26% dos CMTNs foram AR positivos (n = 16) e 74% AR negativos (n = 46). Todos os casos de AR positivos ocorreram em mulheres ≥ 40 anos enquanto 13% dos casos de AR negativos foram vistos em mulheres ≤ 40 anos. 93% (52/56) de todos os CMTNs foram carcinoma ductal invasivo do tipo não especial (ICNST) e 93% (14/15) de AR positivos dos casos de ICNST. O tamanho do tumor variou de 2 a 4,9 cm na maioria dos cânceres AR positivos (n = 8/13; 62%). 60% (n = 9/15) dos casos de AR positivos mostraram grau histológico 3 seguidos de 27% de tumores de grau 2. Não se observou diferença entre pacientes AR positivos e AR negativos quando comparados por idade, tamanho do tumor, grau do tumor e tipo do tumor (p > 0,17). A positividade imuno-histoquímica para ARs também não foi associada com melhor sobrevida (p = 0,737; n = 51) ou sobrevida livre da doença (p = 0,552; n = 45) em CMTNs. CONCLUSÃO: Sob o ponto de vista do prognóstico, a imunorreatividade do AR tem sido associada com melhor sobrevida das pacientes. Esse resultado não foi confirmado na presente série. Isso poderia estar relacionado com o pequeno tamanho amostral ou com uma baixa prevalência de casos de AR positivos especificamente nesse subtipo de câncer de mama. Estudos realizados com maiores amostras são necessários para investigar esse biomarcador em CMTNs. / INTRODUCTION: Triple negative breast carcinomas (TNBC) are a heterogeneous group of tumors characterized by poor patient survival and lack of targeted therapies. Androgen receptor (AR) has been described in TNBC but the prognostic impact of the expression in this subgroup of tumors is not clear. OBJECTIVE: To investigate the association of AR expression status by immunohistochemistry in TNBC cases with clinical (age, survival) and pathological variables (tumor size, tumor grade). METHODS: 62 TNBC were analyzed by automated immunohistochemistry for androgen receptor. Immunohistochemistry was scored by two investigators and biomarker expression was assessed by H-Score (intensity plus the percentage of staining). Kaplan-Meier was used to evaluate overall survival, where differences in distributions were evaluated based on marker expression. RESULTS: 26% of TNBC were AR-positive (n = 16) and 74% AR negative (n = 46). All AR-positive cases occurred in women ≥ 40 years, while 13% of AR-negative cases were seen in women ≤ 40 years. 93% (52/56) of all TNBC were infiltrating ductal carcinomas of no special type (ICNST) and 14/15 (93%) of AR-positive cases were ICNST. Tumor size varied from 2-4.9 cm in the majority of AR-positive cancers (n = 8/13; 62%). 60% (n = 9/15) of AR-positive cases showed histological grade 3 tumors, followed by 27% of grade 2 tumors. No differences were observed between AR-positive and AR-negative patients when compared for age, tumor size, tumor grade and tumor type (all p > 0.17). AR immunohistochemical positivity was also not associated with better overall survival (p = 0.737, n = 51) or disease-free survival (p = 0.552; n = 45) in TNBCs. CONCLUSION: From the prognostic point of view, AR immunoreactivity has been associated with better overall patient survival. Unfortunately, this result could not be confirmed in our series. This could be related to the relatively small series of analyzed samples and to the low prevalence of AR-positive cases in this specific breast cancer subtype. Further studies with bigger samples are needed to investigate this biomarker in TNBC.

Word-of-mouth negativo e o mercado financeiro : repercussões no desempenho das ações no curto e no longo prazo

Seguindo as linhas de pesquisa que visam relacionar o impacto do relacionamento com os clientes aos resultados financeiros da empresa, o presente estudo procura avaliar o impacto do word-of-mouth (WOM) negativo no retorno da ação da empresa. O comportamento de falar mal da empresa por parte dos consumidores insatisfeitos pode destruir o valor dos market-based assets (customer equity e brand equity), comprometendo o desempenho das ações da companhia. Considerando a literatura existente, esta pesquisa busca identificar o impacto do WOM negativo no desempenho das ações das empresas no curto e no longo prazo. O primeiro estudo replica o modelo de Luo (2007) para identificar o impacto do WOM negativo nos retornos de curto prazo da ação das empresas. O segundo estudo busca verificar o impacto de longo prazo a partir da comparação do desempenho de portfólios compostos por ações de empresas em relação ao seu nível de reclamações. Os resultados do primeiro estudo convergiram em certa medida com os resultados apresentados por Luo (2007) para o setor bancário, apontando para um impacto negativo das reclamações no retorno da ação, mas foram mistos quando as empresas foram avaliadas individualmente. O segundo estudo demonstrou que o portfólio composto por ações de empresas menos reclamadas teve um retorno acumulado superior ao portfólio composto pelas ações mais reclamadas, além de apresentar um risco inferior ao risco de mercado. As implicações e limitações do estudo são discutidas e são sugeridas pesquisas futuras a partir deste estudo. / Following previous research that seek to integrate customer relationship impact to firm value, the present study focus to assess the impact of negative word-of-mouth (WOM) on stock returns. Dissatisfied customers that spread negative WOM about the company can ruin market-based assets (customer equity and brand equity) and hurt its performance. Based on present literature, this research seeks to identify the impact of negative WOM on stocks’ short and long term performances. The first study replicates Luo’s (2007) model to identify the short term impact of negative WOM on stock performance. The second aims to identify the long term impact of negative WOM by comparing the performance of portfolios created based on companies level of complaints. The first study accomplished to partially replicate the results found by Luo (2007) for banking industry, finding a negative impact of negative WOM on firm stock returns, although it found mixed results when the impact was assessed for each company individually. The second study demonstrates that a portfolio with less complained companies has a superior performance over a portfolio with more complained companies, while presenting a risk below market’s level. The results implications and limitations are discussed and further studies suggestions are made.

INTRODUÇÃO: O câncer de mama é uma importante causa de morbimortalidade, é conhecido por ser uma doença heterogênea. A caracterização clínica e molecular de seus subtipos é fundamental para nortear o prognóstico e o tratamento dessas pacientes. Novos estudos são necessários na melhor caracterização dos tumores triplo-negativos. O estudo da expressão das claudinas pode auxiliar na caracterização desses tumores. OBJETIVO: Investigar a associação da expressão das claudinas 1, 3, 4 e 7 e E-caderina com variáveis clínico-patológicas e fatores prognósticos, em uma série de tumores de mama triplo-negativos (RE-, RP- e HER2-). MÉTODOS: 80 tumores triplo-negativos foram analisados por imunoistoquímica automatizada para as claudinas 1, 3, 4, 7 e E-caderina. A expressão imunoistoquímica foi avaliada pelo escore H (intensidade multiplicada pela porcentagem de marcação). Foram avaliadas as associações entre características clínico-patólogicas e o escore H. Para a avaliação prognóstica das pacientes, curvas de Kaplan-Meier foram construídas a partir dos dados de seguimento das pacientes e do escore H. RESULTADOS: Foi encontrada associação significativa entre o alto escore H da CLDN-1 (HCLDN-1) e pacientes mais idosas e com a presença de necrose, alto escore H da E-caderina (HE-CAD) em pacientes mais jovens e baixo escore H da CLDN-7 (HCLDN-7) e Ki67 positivo. Além disso, pacientes com elevado HCLDN-1 tiveram menor sobrevida geral. Por outro lado, o elevado HCLDN-3 apresentou uma tendência à associação com maior sobrevida geral e sobrevida livre de doença. CONCLUSÕES: A expressão diferencial das claudinas e E-caderina podem auxiliar na caracterização clinico-patológica dos tumores triplo-negativos. Além disso, as claudinas 1 e 3 parecem ser fatores prognósticos para esses tumores. / INTRODUCTION: Breast cancer is a major cause of morbidity and mortality, is known to be a heterogeneous disease. The clinical and molecular characterization of its subtypes is critical to guide the prognosis and treatment of these patients. Further studies are needed for the best characterization of triple-negative tumors. The study of the expression of claudinas can aid in the characterization of these tumors. OBJECTIVE: To investigate the association of expression of claudinas 1, 3, 4 and 7 and E-cadherin with clinicopathological variables and prognosis in a series of triple-negative breast cancers (ER-, PR- and HER2-). METHODS: 80 triple negative tumors were analyzed by automated immunohistochemistry for the claudins 1, 3, 4, 7 and E-cadherin. The immunohistochemical expression was assessed by H-Score (intensity multiplied by the percentage of staining). We evaluated the associations between clinicopathological characteristics and H-Score. For the prognostic assessment of patients, Kaplan-Meier curves were constructed from the follow-up data of patients and H-Score. RESULTS: We found a significant association between high H-Score of CLDN-1 (HCLDN-1) and older patients and the presence of necrosis, high H-Score of E-cadherin (H-CAD) in younger patients and low H-Score CLDN-7-(7-HCLDN) and Ki67 positive. Furthermore, patients with high-HCLDN1 had a lower overall survival. On the other hand, the high HCLDN-3 showed a trend toward association with greater overall survival and disease-free survival. CONCLUSIONS: Differencial expression of claudins and E-cadherin can help in clinic-pathological characterization of triple-negative tumors. Futhermore, claudin 1 and 3 appear to be prognostic factor for these tumors.

Ce travail de recherche propose d’explorer différentes scènes thérapeutiques (psychodrame, psychothérapie en institution, médiation thérapeutique basée sur la confection et la mise en scène de masques et expression corporelle) pour décliner le travail du négatif – du pôle oppositionnel jusqu’au vide de représentation- et ses conséquences sur le processus adolescent. Il sera supposé que l’accueil des mouvements négatifs, destructeurs, informes ou marqués par la béance, des adolescents et leur élaboration à travers la matière, le corps et le processus de création permet de soutenir le processus de subjectivation. Seront abordés le remaniement identificatoire, l’appropriation subjective qui passe par le corps et l’analyse du contre-transfert en vue de mieux saisir la problématique adolescente lorsque le thérapeute se constitue comme médium malléable. En référence au négatif photographique retrouvé chez Freud et chez Anzieu, une attention sera portée à la modélisation des enveloppes psychiques et au destin des pulsions, notamment dans l’attachement au négatif. Le travail du négatif sera analysé à partir de la matrice proposée par Winnicott et reprise par Roussillon : destructivité, réflexivité, créativité et subjectivation. Trois figures potentielles du négatif s’incarneront dans le masque, la destructivité et l’obésité. Il sera également question du double dans les identifications et la considération de l’axe narcissico-objectal théorisé par Jeammet pour mieux saisir les enjeux intrapersonnels et intersubjectifs. La dimension groupale sera aussi analysée dons son rapport à la négativité. / This research proposes to explore different therapeutic scenes (psychodrama, institutional psychotherapy, therapeutic mediations based on the making / staging of masks and body expression) in order to analyse the work of the Negative – from the oppositional pole to the absence of representation (blankness) and its consequences on the Adolescent Process. It will be assumed that being receptive to adolescents’ negative, destructive movements as well as to formlessness or blankness expressions enables the therapist, through their elaboration via the process of creation, to support and accompany the process of subjectivation. We will address the identificatory reorganisation, the subjective appropriation which pass through the body and the analysis of the countertransference in order to better understand the adolescent problem when the therapist is in the position of a malleable medium. In reference to the photographic negative found in Freud and Anzieu’s work, attention will be paid to the modelling of psychic envelopes together with the destiny of the drives, more precisely in the attachment to the Negative. The work of the Negative will be analysed according to the frame proposed by Winnicott and taken up by Roussillon : Destructivity, Reflexivity, Creativity and Subjectivation. Three potential figures of the Negative will be embodied in the mask , destructivity and obesity. In order to better grasp the intrapersonal and intersubjective issues at stake, we will also consider the Figure of the Double in the identifications as well as the narcissistic-objectal axis as it was theorized by Jeammet. The groupal dimension will also be analysed in relation to Negativity.

Novel Aminoglycoside Polymers and Combination Treatments in Triple Negative Breast Cancer Studies

abstract: In the United States, 12% of women are typically diagnosed with breast cancer, where 20-30% of these cases are identified as Triple Negative Breast Cancer (TNBC). In the state of Arizona, 810 deaths occur due to breast cancer and more than 4,600 cases are diagnosed every year (American Cancer Society). The lack of estrogen, progesterone, and HER2 receptors in TNBC makes discovery of targeted therapies further challenging. To tackle this issue, a novel multi-component drug vehicle is presented. Previously, we have shown that mitoxantrone, a DNA damaging drug, can sensitize TNBC cells to TRAIL, which is a protein that can selectively kill cancer cells. In this current study, we have formulated aminoglycoside-derived nanoparticles (liposomes) loaded with mitoxantrone, PARP inhibitors, for delivery to cancer cells. PARP inhibitors are helpful in preventing cancer cells from repairing their DNA following damage with other drugs (e.g. mitoxantrone). Various treatment liposome groups, consisting of lipid-containing polymers (lipopolymers) synthesized in our laboratory, were formulated and characterized for their size, surface charge, and stability. PARP inhibitors and treatment of cells for in-vitro and in-vivo experiments with these liposomes resulted in synergistic death of cancer cells. Finally, studies to evaluate the pre-clinical efficacy of these approaches using immuno-deficient mouse models of TNBC disease have been initiated. / Dissertation/Thesis / Masters Thesis Chemical Engineering 2018

Connecting past and present: the historical anthropology of the small-scaled minning production in porco, bolivia Who studies and creates the narratives that surround the city of Lima’s archaeological sites, known locally as huacas? Traditionally, this has been the responsibility of professional archaeologists, who in their research and conservation efforts, as well as in their efforts to convert sites into open-air museums, tend to focus almost exclusively on the prehispanic period, when these sites were initially designed, built, used, and transformed. This approach marginalizes and even renders invisible the role these huacas had during the Colonial and Republican periods. This particular narrative is problematic, since it subjectively “freezes” sites into limited time frames, and implies that the value and significance of sitez lies solely in a very specific past. Following this narrative, huacas become static entities, instead of dynamic spaces that change over time, in which all historical periods contributed significantly to their current state. Inspired by the research, conservation, and conversion of Huaca Huantinamarca (in Lima’s San Miguel district) into a public space and open-air museum, this paper proposes to go beyond the traditional narrative and include all historical periods, including those periods perceived as “despised history”, in order to construct a narrative that is more comprehensive, authentic, and inclusive. / ¿Quién investiga y construye la narrativa de los sitios arqueológicos en la ciudad de Lima, conocidos localmente como huacas? Tradicionalmente, esta tarea ha sido responsabilidad de los arqueólogos profesionales, que, en la investigación, conservación y, sobre todo, en los trabajos de «puesta en valor», suelen privilegiar el período de construcción, uso original y transformaciones de estos sitios durante la época prehispánica, marginando e incluso haciendo invisible el papel que las huacas tuvieron durante la Colonia y la República. Esta narrativa es problemática, puesto que, de manera subjetiva, «congela» a las huacas en un periodo delimitado, y sugiere que su valor y significado se encuentra solamente en un pasado específico. Siguiendo esta narrativa, las huacas son presentadas como espacios estáticos, en vez de espacios dinámicos que cambian con el tiempo, donde todos los períodos históricos —incluido el período de ruina— contribuyen de manera significativa a su estado actual. Inspirados en la estrategia de investigación, conservación y puesta en valor aplicada en Huantinamarca en el distrito de San Miguel, proponemos ampliar la narrativa de las huacas e incluir todos los períodos históricos, incluso aquella historia percibida como «negativa» para así construir una narrativa más completa, auténtica e inclusiva.

Abnormal daily situations and negative affects in Peru / Situaciones cotidianas anómalas y afectos negativos en el Perú

Four hundred seventy nine Peruvian students (188 mal e and 291 female) rated the frequency of negative social emotions in themselves, in their neighborhood, and in the Peruvian society; and the frequency of anomalous daily situations. Verbal aggressions and gossiping obtained the highest averages in the evaluations of affects in the neighborhood, but solidarity was rated as high than the negative affects. In the Peruvian society, envy and physical aggression were rated asthe highest, and the positive affects obtained the smallest averages. To be a victim of an assault,to be cheated, and to be insulted obtained the highest averages in anomalous situations. Resentment and anger were the most salient negative affects. / Se investigó a 479 universitarios peruanos (188 hombres y 291 mujeres) a través de una autoevaluación sobre la frecuencia de afectos y conductas negativas en ellos mismos, en su vecindad y en la sociedad peruana, y la de situaciones anómalas en su vida cotidiana. Agresiones verbales al otro y hablar mal de los demás obtuvieron los promedios más elevados en la valoración de los afectos y las conductas negativos en el barrio, pero la solidaridad fue asimismo vista como muy elevada. En la sociedad peruana, la envidia y la agresión verbal fueron evaluadas como las más altas y los promedios de los afectos positivos (solidaridad y confianza en los demás)f ueron los más bajos. Ser víctima de un robo, ser víctima de un engaño y ser insultado obtuvieron los promedios más altos en situaciones anómalas. El resentimiento y la cólera fueron los afectos negativos de mayor magnitud.

A Prospective Study of Childhood Negative Events, Temperament, Adolescent Coping, and Stress Reactivity in Young Adulthood

abstract: Accumulating evidence implicates exposure to adverse childhood experiences in the development of hypocortisolism in the long-term, and researchers are increasingly examining individual-level mechanisms that may underlie, exacerbate or attenuate this relation among at-risk populations. The current study takes a developmentally and theoretically informed approach to examining episodic childhood stressors, inherent and voluntary self-regulation, and physiological reactivity among a longitudinal sample of youth who experienced parental divorce. Participants were drawn from a larger randomized controlled trial of a preventive intervention for children of divorce between the ages of 9 and 12. The current sample included 159 young adults (mean age = 25.5 years; 53% male; 94% Caucasian) who participated in six waves of data collection, including a 15-year follow-up study. Participants reported on exposure to negative life events (four times over a 9-month period) during childhood, and mothers rated child temperament. Six years later, youth reported on the use of active and avoidant coping strategies, and 15 years later, they participated in a standardized psychosocial stress task and provided salivary cortisol samples prior to and following the task. Path analyses within a structural equation framework revealed that a multiple mediation model best fit the data. It was found that children with better mother-rated self-regulation (i.e. low impulsivity, low negative emotionality, and high attentional focus) exhibited lower total cortisol output 15 years later. In addition, greater self-regulation in childhood predicted greater use of active coping in adolescence, whereas a greater number of negative life events predicted increased use of avoidant coping in adolescence. Finally, a greater number of negative events in childhood predicted marginally lower total cortisol output, and higher levels of active coping in adolescence were associated with greater total cortisol output in young adulthood. Findings suggest that children of divorce who exhibit better self-regulation evidence lower cortisol output during a standardized psychosocial stress task relative to those who have higher impulsivity, lower attentional focus, and/or higher negative emotionality. The conceptual significance of the current findings, including the lack of evidence for hypothesized relations, methodological issues that arose, and issues in need of future research are discussed. / Dissertation/Thesis / Ph.D. Psychology 2013
