The diploma thesis deals with Integrated public transport system in South Moravia region. The author describes the term of integrated public transport system and its advantages in the theoretical part of the thesis. The practical part deals with constitution and development of the system in the South Moravia region and describes its subsystems: organizing and economical, tariff and transport subystem. The final chapter describes transport systems in the regions of the Czech republic. The last chapter shows the shortages of the current transport system. The main aim of the thesis is to make analysis of the current system, to compare theoretical findings with the current situation, to situate the transport system in South Moravia into the frame of the public transportation in the Czech republic and to compare it with other systems.

Syfte: Syftet är att redovisa den nuvarande verkligheten av mellanstora konsertarrangörer i Stockholm Stad. Genom en analys av intressenter, nätverk och omvärldsfaktorer som kan påverka organisationen så vill vi tillföra en bredare förståelse av hur de samverkar med en utvecklande miljö.  Metod: Vår metod är aktör-nätverksteorin, där empirin har framtagits via semistrukturerade intervjuer och insamling av källkritiskt utvald sekundär data.  Sammanfattning: Mellanstora konsertarrangörer i Stockholm ingår i ett flertal olika nätverk, varav samtliga aktörer inom dessa nätverk har individuella intressen gentemot konsertarrangörens verksamhet. Nätverkens betydelse är väldigt stor för konsertarrangörerna då de organisationer som de samarbetar med har resurser som korrelerar med konsertarrangörens syfte; till exempel artister. Nätverken och dess aktörers intressen är dock dynamiska och föränderliga vilket problematiserar definieringen av dessa nätverk och intressenter. Detta beror på att konsertarrangörerna ständigt jobbar med att etablera och underhålla nätverk. Den omvärld som de mellanstora konsertarrangörerna ingår i har dessutom andra aktörer vars intresse ligger för Stockholms stads utveckling. Dessa intressen korrelerar inte alltid med konsertarrangörernas och på grund av den mångfald av intressen som stadens politiker och myndigheter behöver beakta skapas konflikter som i dagsläget skapat en väldigt aktuell debatt om Stockholms kulturliv. / Purpose: The purpose is to present a current view of the reality in being a middle sized concert organizer in Stockholm. By analyzing their stakeholders, network connections and external factors that can affect their organizations, we want to contribute with a broader understanding of how they cooperate in a environment that is constantly evolving.  Method: Our method for this essay is the actor-network-theory (ANT) in which we have chosen to collect our empiricism through semi structured interviews and secondary data with thoroughly criticised sources.  Summary: Middle sized concert organizers in Stockholm interacts in many different networks in which all of the included actors have individual interests in collaborations with the organizer. Working with networks is very important for the concert organizer because of the resources that is essential for the organizers purpose; artists is an good example. The networks and the different interests are difficult to define and map out because of the complexity that comes with their dynamics. They are dynamic because concert organizers are continuingly working with establishing new networks as well as maintaining the current ones. The environment that the midsized concert organizers are acting in also includes other actors in which the interests lies within the development of Stockholm. These interests doesn’t always correlate with the concert organizers. And because of the massive amount of interests that Stockholms politicians and other authorities have to take in consideration, conflicts are created. This is something that has led to a big debate about the culture development in Stockholm.

Mechanism of cell adhesion at the midbrain-hindbrain neural plate in the teleost Danio rerio

The correct development of multicellular organisms is tightly regulated by intrinsic and extrinsic factors at specific time points. Disturbance on any level of these multiple processes may result in drastic phenotypes or eventually death of the organism. The midbrain-hindbrain boundary (also termed isthmic organizer) is a region of high interest as well in early as also in later development. The isthmic region carries organizer identity by the expression and subsequent release of FGF8. False patterning events of this region in early developmental stages would therefore display dramatic results over time. As it has been shown that the midbrain-hindbrain boundary (mhb) in the zebrafish is a compartment (or lineage restriction) boundary I tried to understand the underlying molecular mechanism for its correct establishment. In this work I focused both on embryological, molecular and genetic means to characterize involved molecules and mechanisms. In the first part of the thesis I followed in vivo cell transplantation assays, having started with an unbiased one. Cells of either side the mhb were challenged with this boundary by bringing them into direct cell contact with their ectopic counterpart. In a biased approach, cells overexpressing mRNA of specific candidate genes were transplanted and their clonal distribution in host embryos was analyzed. In the second part of the thesis I started interfering with specific candidate genes by transiently knocking down their protein translation. The adhesion molecules of the Eph/ephrin class had been shown to restrict cell mixing and thereby creating compartment boundaries in other tissues, such as the hindbrain, in the zebrafish and other organisms. Additionally, we generated several stable genetic mutant lines in cooperation with the Tilling facility at the Max-Planck-Institute. The only acquired potential null mutant ephrinB2bhu2971 was analyzed and characterized further. I observed that a knock down or knock out of only one of the ephrinB2 ligands does not seem to be sufficient for a loss of compartment boundary formation. The combinatory approach of blocking translation of EphrinB2a in ephrinB2bhu2971 mutants gave very complex and interesting phenotypes, which need to be investigated further.

Notre recherche vise à expliquer et comprendre la manière dont 3 formateurs chevronnés parviennent, à travers le développement de leur activité, à "absorber" des prescriptions issues d’un nouveau paradigme de professionnalisation, à l’origine d’une discontinuité culturelle. Comment dans ce contexte, parviennent-ils à mettre en place un nouveau dispositif, le "tutorat partagé" entre l’ESPE et les établissements scolaires, considéré comme un des piliers de cette nouvelle réforme et de la formation en alternance ?En reposant sur une méthodologie interventionniste, les résultats de cette recherche montrent la nécessité, pour ces trois formateurs expérimentés, de faire appel à leur expérience afin de trouver les compromis nécessaires leur permettant d’être efficaces « malgré tout ». Nous montrons comment ils dégagent un certain nombre d’implications au niveau de la formation, notamment en passant par une phase de transition professionnelle, correspondant à un usage débutant du genre formateur leur permettant une plus grande maîtrise de la situation et l’obtention d’un confort relatif. Prendre au sérieux la permanence du changement à l’œuvre dans les prescriptions adressées aux formateurs, nous conduira à dégager des conditions d’un accompagnement plus efficient propres à les aider à traverser des périodes de transition complexes. C’est à travers la création d’un milieu de travail "extra-ordinaire" sur le travail ordinaire que nous visons à faire des différentes pratiques de "tutorat partagé", fondé sur l’inter-métier de formateurs aux statuts différents, un objet de controverses professionnelles afin d’accroître les ressources mobilisables et le pouvoir d’agir des intéressés. / Our research aims at explaining and at including how three experienced trainers, through the development of their activity, succeed in "absorbing" prescriptions arising from a new paradigm of professionalisation, at the origin of a cultural discontinuity. How do they manage to set a new device, the "shared tutoring" between the ESPE and the schools, in this context, considered as one of the pillars of this new reform and of work-linked training courses ?Based on an interventionist methodology, the results of this research show the need, for these three experienced trainers, to use their experience to find compromises necessary to be effective "despite everything". We show how they have a number of training implications, in particular through a professional transition phase, corresponding to a beginner use of the trainer type, allowing them a greater control of the situation and the procurement of a relative comfort. Taking into account the permanence of the change in work in the prescriptions addressed to the trainers will lead us to identify the conditions of a more efficient support capable of helping them through complex transition periods. It is through the creation of an “extraordinary” work environment on the ordinary work that we aim to make various practices of "shared tutoring", based on inter-trade of the different trainers, an object of professional controversies in order to increase the mobilizable resources and the power to act of the interested parties.

Porém, mesmo tendo sido vítima do controle ideológico, de perseguições, de prisões e processos, manteve-se convicto de suas posições teórico-práxicas, atuando na organização da cultura e na luta em defesa do projeto societário e de formação humana. / This thesis aimed at analyzing the trajectory of José Rodrigues Vieira Netto, privileging the explanation of his action in the organization of the social and educational project of communist character as well as his work mobilizing the society for the fight defending this ethical-political proposal. The analytical process aims at understanding Vieira Netto’s qualification from his family inheritance, emphasizing a view on his school education and his university graduation; discussing his projection and the context of his professional activity as a professor, lawyer and public prosecutor; characterizing his main activism fields, highlighting the Liberal Alliance, the Brazilian Communist Party and the Legislative Assembly of the State of Parana, as well as the consequences of this activism with the restriction of his rights and persecution; and, finally, discussing Vieira Netto’s social and educational project in close relation to the Brazilian Communist Party’s educational project, privileging an analysis of the social and educational view present in his actions and productions. The analyses comprise the period from 1945 and 1973, including the moment Vieira Netto’s joined the Brazilian Communist Party (1945) and the year he died (1973). This thesis integrates the Intellectual History, in a movement to understand the subjects called intellectuals due to their posture and action when facing social issues, mainly, when they adopted positions of defence of the so-called universal rights such as justice, truth and democracy. Regarding our analysis, we chose Antonio Gramsci’s theory, mainly the concept of organic intellectual – culture organizer – who is a determining political agent, since his work is not exhausted in the production of scientific, artistic or philosophical knowledge, but is extended, mainly to the duty of the organizing function, which is characteristic of the social and political fights. This thesis is also supported by sources found in the minutes of the Federal University of Parana, the archives of the Political and Social Order Agency in Parana, the archives of the digital platform ‘Brasil: Nunca Mais’ (Brazil: Never More), interviews, and memoir books to explain how Vieira Netto worked to systematize a social project of human education, as well as to mobilize the working class to engage the fight for this project. Our view defends that Vieira Netto acted as an organic intellectual in the construction of the communist social and educational program in Parana, since he was a professor, lawyer and public prosecutor, member of the Liberal Alliance, an activist in PCB (Brazilian Communist Party) becoming a State deputy and acting in social movements. This activism had consequences, since from 1945 to 1973 he was booked and constantly monitored, despite having been acquitted in the processes that he had faced during the military dictatorship. His rights as a politician were also restricted in 1948, but his mandate as a State deputy was symbolically restored in 2013. However, even having been a victim of the ideological control, persecutions, imprisonment and law suits, he kept his theoretical approach and his praxis, acting as a culture organizer and in the fight defending the social and human educational project.

Este trabalho tem como objetivo analisar o projeto de criação de uma aristocracia do trabalho a partir do ensino industrial, através do estudo de caso da Escola Técnica de Curitiba, posteriormente Escola Técnica Federal do Paraná, entre os anos de 1942 a 1963. Num primeiro momento, faz-se um breve apanhado da citada instituição quando era Escola de Aprendizes Artífices do Paraná, considerando que a concepção de ensino então desenvolvida era destinada aos desfavorecidos da fortuna, portanto direcionada principalmente para a disciplinarização das chamadas classes perigosas. Mas ao longo da década de 1920 e de 1930, tal concepção foi sendo modificada, até que em 1942, com a Lei Orgânica do Ensino Industrial, consolidou-se a idéia de formação de uma aristocracia do trabalho pela via do ensino industrial. Essa elite ocuparia postos no setor fabril que exigissem uma maior qualificação, acompanhando o processo de desenvolvimento industrial do Brasil, não bastando, porém, um melhor adestramento técnico, mas que fosse bem disciplinada, através de um processo educacional marcado pela presença intensiva de conteúdos de moral e civismo, notadamente durante o Estado Novo (1937-1945), servindo assim de exemplo para a classe trabalhadora como um todo. O pós-guerra foi marcado pela Guerra Fria e pela influência cada vez maior do american way of life na vida brasileira. A presença da Comissão Brasileiro-Americana de Educação Industrial (CBAI), programa de cooperação firmado entre os governos do Brasil e dos Estados Unidos em 1946, veio caracterizar esse momento no ensino industrial do país, através da adoção de métodos sistemáticos na formação de docentes para esse ramo do ensino, baseados na Racionalização Científica. A fim de dinamizar o treinamento dos professores, a CBAI criou, em 1957, o Centro de Pesquisas e Treinamento de Professores (CPTP), na Escola Técnica de Curitiba, cuja função era preparar docentes para atuarem nas escolas industriais e técnicas de todo o Brasil, de forma a capacitá-los para a tarefa de formação da aristocracia do trabalho. Em 1963 o acordo entre a CBAI e o governo brasileiro não foi renovado, e o Centro encerrou suas atividades no ano seguinte. A Lei nº 3.552/59 estabeleceu uma reforma no ensino industrial, fixando um curso único de 1º Ciclo com finalidade propedêutica para os cursos técnicos de 2º Ciclo. A Lei de Diretrizes e Bases de 1961 promoveu a equivalência entre o ensino técnico e o ensino secundário. Tais modificações, ligadas à nova etapa vivida pela industrialização, inclusive em relação às demandas do mercado de trabalho, aliadas à possibilidade de chegar ao ensino superior ao mesmo tempo em que se obtém uma formação de nível médio em escolas de elevada qualidade de ensino, promoveram alterações no perfil dos alunos que procuravam o ensino industrial, que gradativamente foi sendo ocupado por estudantes originários da classe média, deixando de ter validade a concepção de formação de uma aristocracia do trabalho exclusivamente a partir da classe trabalhadora. / This thesis have the objective of analyze the project of creation of the labour aristocracy based in the industrial teaching, by case study of the Escola Técnica de Curitiba, posteriorly Escola Técnica Federal do Paraná, between 1942 and 1963. On a first moment, making a succinct summary of the named institution when denominated Escola de Aprendizes Artífices do Paraná, seeing that the educational intention developed was destined to luckless, therefore reserved to the discipline the dangerous classes. But during the 1920 and 1930 decades, this intention was going modify, and in 1942 the idea of conformation of the labour aristocracy based in the industrial education was consolidated, with the Organizer Law of the Industrial Teaching. This elite must occupy functions in the manufacturing sector what demanding a superior qualification, accompanying the industrial development of Brazil, however dont being enough a better technical training, but was very much disciplined, in through of the educational process marked for intensive carriage of moral and civic contents, notedly during the Estado Novo (1937-1945), what a example for all the working class. The period after the Second World War was marked for the Cold War and to the growing influence of American way of life in Brazil. The presence of the Comissão Brasileiro-Americana de Educação Industrial (CBAI), a cooperation program signed between Brazilian and American governments in 1946, marked this moment in the industrial teaching of Brazil, through the adoption of systematical methods to the formation of instructors for this kind of teaching, based on Scientific Management. With the aim of to pep the instructors training, in 1957 CBAI created Centro de Pesquisas e Treinamento de Professores (CPTP), on the Escola Técnica de Curitiba, and your function was to prepare instructors to work on the Industrial and Technical Schools in Brazil, capacitating them to the assignment of to educate the labour aristocracy. In 1963 the accord between CBAI and the Brazilian government is not renovated, and the CPTP was closed your activities in 1964. The 3.552/59 Act determined a reform at the Industrial Teaching, fixing a unique course of First Level with propaedeutics finality to the technical courses of Second Level. The Guidelines and Basis Act of 1961 furthered the equivalency between Technical and Secondary Teaching. These modifications, linked with the new stage of industrialization and connected with the work markets demands, allied to the possible incoming of students go to College while graduate in high quality schools of Second Level, make changes in the profiles of students that searched the Industrial Teaching, which was occupied for students of midclass, invalidating the conception of conformation of the labour aristocracy originating exclusive of working class.
