Immunophenotypic Variation in Neonatal Pigs and Immunomodulating or Anti-allergic Effects of Microbial Treatments

Due to the intrauterine environment required to maintain pregnancy it may be that neonatal animals are born type-2 immune response (IR) biased, which consequently may increase susceptibility to certain infectious and immune mediated diseases, such as allergy. Recently, the prevalence of both allergic and autoimmune diseases has increased, leading to the development of the hygiene hypothesis. The hypothesis states that lack of early environmental stimulus leading to inappropriate development and bias in IR, may contribute to this increase. The objectives of this thesis, therefore, were to: (a) Determine the IR bias of neonatal pigs. (b) Investigate the effect of heat-killed Escherichia coli, lipopolysaccharide (LPS) and muramyl dipeptide (MDP) on the IR phenotype and the frequency of allergy in pigs sensitized to the egg white allergen ovomucoid (Ovm). (c) Establish IR phenotypes of pigs allergic or clinically tolerant to Ovm. Immune response bias was determined using an established phenotyping protocol and compared between two groups of pigs, (A) and (B). A difference in IR bias was observed. Bias in IR was not consistently towards type-2. Increase in indicators of type-1 IR, were greater in A and the frequency of type-2 IR correlates were greater in B. It’s likely that unidentified environmental variables may have induced this change, although etiology was not pursued. Treatment with heat-killed Escherichia coli, LPS and MDP had an effect on IR bias and frequency of allergy. Muramyl dipeptide-treatments promoted type-2 bias and were associated with increased frequency of allergy. Pre-treatment with E. coli did not affect allergic frequency, but did elicit the production of a relatively balanced allergen-specific IR phenotype. Lipopolysaccharide-pre-treatment was associated with decreased frequency of allergy. Correlates of an allergic IR phenotype in pigs were also established. The measurement of allergen-specific IgG, IgG1 and/or IgE activity and evaluation of late-phase intradermal skin tests were proposed to be useful in identifying allergic IR phenotypes. This thesis emphasizes the importance of considering the potential for variation in IR in terms of pig health and experimental reproducibility. Contamination des solutions d’hyper-alimentation intraveineuses (HAIV) néonatales, effet de l’ascorbylperoxyde au foie

Introduction : Chez les nouveau-nés prématurés, l’hyper-alimentation intraveineuse (HAIV) contribue à leur survie, mais elle est aussi une source importante de molécules oxydantes. L’absence d’une protection adéquate contre la lumière ambiante génère in vitro, via la photo-excitation de la riboflavine, du H2O2, des peroxydes organiques et un dérivé peroxydé de la vitamine C, l’ascorbylperoxyde (AscOOH). Plusieurs données du laboratoire associent l’infusion d’HAIV à des désordres lipidiques dans notre modèle animal. L’hypothèse est donc que l’AscOOH a un pouvoir oxydant et est responsable de certains des effets biologiques observés. Mes objectifs sont les suivants : 1) développer une méthode de dosage de l’AscOOH; 2) démontrer, à l’aide du modèle animal bien établi au laboratoire, des relations entre la concentration tissulaire de cette molécule et des paramètres métaboliques et l’état redox au foie et dans la circulation; et 3) confirmer l’effet physiologique de l’AscOOH dans un modèle cellulaire. Méthode : Différents étalons internes potentiels ont été testés pour le dosage de l’AscOOH par spectrométrie de masse après séparation sur HPLC (LC-MS). Les phases mobiles et conditions chromatographiques ont été optimisées. Pour l’objectif 2, des cobayes de 3 jours de vie (n=11) ont reçu par voie intraveineuse une dose d’AscOOH (entre 0 et 3,3mM). Les animaux ont été sacrifiés au 4e jour de traitement pour le prélèvement de tissus. Les concentrations tissulaires d’AscOOH ont été déterminées au LC-MS. La triglycéridémie et la cholestérolémie ont été mesurées à l’aide d’un kit commercial par spectrophotométrie. Le glutathion oxydé et réduit ont été mesurés par électrophorèse capillaire. Les relations linéaires obtenues sont exprimées par le ratio des carrés (r2), et traitées par ANOVA. Résultats : La validation du dosage de l’AscOOH par LC-MS a été réalisée. Chez les animaux, la concentration urinaire d’AscOOH par créatinine corrèle positivement avec la dose reçue, négativement avec la lipidémie, et négativement avec le redox sanguin et érythrocytaire, indiquant un milieu moins oxydé. Conclusion : La concentration urinaire d’AscOOH peut donc être un reflet de l’oxydation de l’HAIV en clinique. Nos données chez l’animal suggèrent une interaction de l’AscOOH avec le métabolisme hépatique produisant une chute de la concentration plasmatique de cholestérol et de triglycérides. Le modèle cellulaire n’a pas permis d’élucider le mécanisme moléculaire de l’action de l’AscOOH sur le métabolisme. / Introduction: Intravenous hyperalimentation (IVHA) often contributes to the survival of preterm newborns, but it is also an important source of oxidizing molecules. The lack of adequate protection from ambient light generates, in vitro, through the photo-excitation of riboflavin, H2O2, organic peroxides and a peroxidated derivative of vitamin C: ascorbylperoxide (AscOOH). Certain data from our laboratory linked the infusion of IVHA to lipid disorders in our animal model. The hypothesis is that AscOOH is an oxidant that is responsible for some of the biological effects observed. My objectives are: 1) to develop a method for quantitation of AscOOH, 2) to demonstrate, using the guinea pig model used by our laboratory, relations between the tissue concentration of this molecule and metabolic and redox parameters in the liver and plasma, and 3) to confirm the physiological effect of AscOOH in a cell culture model. Method: Different promising internal standards were tested for AscOOH quantitation by mass spectrometry after HPLC separation (LC-MS). Mobile phases and chromatography conditions have been optimized. For objective #2, 3 days old guinea pig pups (n = 11) received an intravenous dose of AscOOH (between 0 and 3.3mM). Animals were sacrificed on the 4th day of treatment for tissue gathering. Tissues AscOOH concentrations were determined by LC-MS. The triglyceride and cholesterol levels were measured by spectrophotometry using a commercial kit. The oxidized and reduced glutathione were measured by capillary electrophoresis. The linear relations obtained are expressed by the square of the correlation coefficient (r2), and processed by ANOVA. Results: The validation of the LC-MS method for AscOOH quantification has been achieved. In animals, the concentration of urinary AscOOH by creatinine correlates positively with the dose received, negatively with blood lipids, and negatively with blood and erythrocyte redox, indicating a less oxidized environment. Conclusion: The urinary AscOOH concentration may be a good indicator of the oxidation state of clinical IVHA. Our data in animals suggest an interaction between AscOOH and liver metabolism producing a drop in plasma concentration of cholesterol and triglycerides. The cell model was not able to clarify the molecular mechanism of AscOOH action on metabolism.

Détermination des effets de l'administration des probiotiques sur l'attachement d'Escherichia Coli Entérotoxinogène F4 et l'expression de cytokines chez le porcelet sevré

Les diarrhées post-sevrages causées par des infections à Escherichia coli entérotoxinogène positif pour le fimbriae F4 (ETEC F4), entraînent des pertes économiques importantes chez les producteurs de porc. Depuis quelques années, l’utilisation de probiotiques, comme additif alimentaire pour prévenir ce type d’infection entérique et réduire les traitements aux antimicrobiens, suscite un intérêt grandissant en production porcine. Le but du présent travail est de déterminer l’influence de l’administration des probiotiques Pediococcus acidilactici (PA) et Saccharomyces cerevisiae boulardii (SCB) sur la colonisation et l’attachement des ETEC F4, l’accumulation de fluide intestinal et l’expression de cytokines dans l’iléon de porcelets sevrés. Dès la naissance, différentes portées de porcelets ont été affectées aux traitements suivants : PA, SCB, PA + SCB, témoin et témoin avec antibiotiques (ATB). Une dose quotidienne de probiotiques (1 × 109 UFC) a été administrée aux porcelets des groupes probiotiques durant la lactation et après le sevrage. Sept jours après le sevrage, à 28 jours d’âge, des porcelets positifs pour le récepteur intestinal spécifique pour F4 ont été infectés oralement avec une souche ETEC F4. Les porcelets ont été euthanasiés 24 heures après l’infection (jour 29) et différents échantillons intestinaux ont été prélevés. Chez les porcelets recevant des probiotiques, l’attachement des ETEC F4 à la muqueuse iléale était significativement diminué chez les groupes PA ou SCB en comparaison avec le groupe ATB. Finalement, l’expression de cytokines intestinales était plus élevée chez les porcs du groupe PA + SCB en comparaison avec les porcelets témoins. En conclusion, les résultats de cette étude suggèrent que l’administration de probiotiques pourrait être une alternative pour limiter les infections à ETEC F4 chez le porc. / Postweaning diarrhea (PWD) associated with F4-positive enterotoxigenic Escherichia coli (ETEC F4) causes important economic losses in swine production. Since a couple of years, the use of probiotics as feed additives to prevent such enteric infections and reduce the use of antimicrobial treatments, has gained in interest. The aim of the present study is to evaluate the effects of Pediococcus acidilactici (PA) and Saccharomyces cerevisiae boulardii (SCB), on ETEC F4 colonization and attachment, accumulation of intestinal fluid and cytokines expression in weaned pig’s ileum. At birth, different litters of pigs were allocated to the following treatments: PA, SCB, PA + SCB, control (CTRL) and control with antibiotics (ATB). Probiotics (1 × 109 CFU) were administered daily to probiotics group during the lactation period and after weaning. One week after weaning, at 28 days of age, all F4-receptor-positive pigs were orally challenged with an ETEC F4 strain. Pigs were slaughter 24 hours later (day 29) and different intestinal samples were collected. In pigs treated with PA or SCB, the attachment of ETEC F4 to the ileal mucosa was significantly reduced in comparison with the ATB group. Finally, intestinal cytokines were upregulated in PA + SCB group in comparison with the CTRL group. In conclusion, these results suggest that administration of probiotics could be an alternative to attenuate ETEC F4 infection in pigs.

Étude sur le Staphylococcus aureus résistant à la méthicilline chez le porc à l'abattoir au Québec, Canada

Le Staphylococcus aureus résistant à la méthicilline (SARM) est un pathogène important qui a été identifié comme agent d‟infection chez les animaux d‟élevage et les travailleurs exposés à ces animaux. Au Canada, très peu d‟informations sont disponibles concernant les SARMs d‟origine porcine. L‟objectif de cette étude était de déterminer la prévalence des SARMs provenant de porcs à l‟abattoir, de caractériser leur résistance aux antibiotiques ainsi que d‟évaluer le niveau de séroconversion des porcs envers le S. aureus chez les animaux porteurs ou non du SARM. Un total de 107 isolats ont été identifiés positifs aux SARMs sur 660 échantillons. La prévalence de SARMs à l‟abattoir A était de 30,8% et de 23,8% à l‟abattoir B. La susceptibilité aux antibiotiques a été déterminée en utilisant la méthode de micro-dilution de Sensititre. Tous les isolats ont démontré une sensibilité envers la ciprofloxacine, la gatifloxacine, la gentamicine, la lévofloxacine, le linézolide, la quinupristine/dalfopristine, la rifampicine, la streptomycine, le triméthoprime/sulfaméthoxazole et la vancomycine. De la résistance a été observée envers la daptomycine (0,93%), l‟érythromycine (29%), la clindamycine (29%), la tétracycline (98,1%). De plus, 30% des SARMs isolés étaient résistants à plus de deux antibiotiques autres que les β-lactamines. Par typage, deux clones prédominants ont été obtenus ainsi que deux types de SCCmec (type V et possiblement un nouveau type comprenant les cassettes III et IVb). 15 clones ont été identifiés par typage MLVA, comprenant les clones prédominants VI (40.1%; 43/107) et XI (17.7%; 19/107). Deux souches de SARMs ont été caractérisées par biopuce à ADN et des gènes d‟antibiorésistance, de typage (SCCmec et MLST) et de virulence ont été identifiés. Sans considération pour le site de colonisation, les porcs SA-/MRSA- (n=34) et les porcs SA+ (n=194) montrent, respectivement, des taux de séroconversion de 20.6% et 32.5%. Les porcs colonisés par un SARM à un site de iv prélèvement et non colonisés par un SA à l‟autre site (n=18) montrent une séroconversion (5.6%) significativement (P < 0.05) plus faible comparativement aux porcs colonisés par SA à un ou deux sites de prélèvement et n‟ayant pas de SARM. Nos résultats démontrent que les porcs provenant d‟abattoir peuvent être colonisés par des SARMs multi-résistants aux antibiotiques. De plus, ces SARMs sont possiblement capable de coloniser leurs hôtes sans stimuler la production d‟anticorps et ce par l‟atténuation de la réponse immunitaire ou par la colonisation de porcs qui sont moins immunocompétents. / Methicillin-resistant Staphylococcus aureus (MRSA) found in food producing animals is a major public health concern. Transmission to humans has been reported and MRSA represents a reservoir of antimicrobial resistance genes. Little is known on how MRSA successfully establishes colonization and how it is able to persist in the host. This study was conducted to determine the occurrence and the antimicrobial resistance profile of MRSA from abattoir pigs and their level of seroconversion toward S. aureus (SA). A total of 107 isolates were identified as MRSA from 660 samples. Antimicrobial susceptibilities were determined by broth microdilutions. Fifteen clones were identified by MLVA with clones VI (40.1%; 43/107) and XI (17.7%; 19/107) being the most predominant. All MRSA isolates were pvl-, tst-, eta- and etb-negative. Most isolates were SCCmec type V (70.1%; 75/107). All MRSA isolates were susceptible to ciprofloxacin, gatifloxacin, gentamicin, levofloxacin, linezolid, quinupristin/dalfopristin, rifampin, streptomycin, trimethroprim/sulfamethoxazole and vancomycin. However, resistance was observed toward clindamycin (29%), daptomycin (0.9%), erythromycin (29%) and tetracycline (98.1%). Multi-resistance was confirmed in MRSA since 28% of all isolates were resistant toward three antimicrobials other than β-lactams. The effect of MRSA carriage on seroconversion was examined to see whether the host responded differently to MRSA or SA colonization. The presence of SA-specific antibodies in pig serums was measured for each animal using indirect ELISA and a mixture of two widespread SA antigens (IsdH [HarA] and IsdB). Regardless of the colonization site, SA-/MRSA- pigs (n=34) and SA+ pigs (n=194) showed 20.6% and 32.5% seroconversion, respectively. Notably, pigs colonized by MRSA at one body site and no SA at the other sampling site (n=18) showed a significantly lower (5.6%) seroconversion (P < 0.05) compared to pigs colonized by SA at one or both vi sites without MRSA. The findings of the study show that the nares and axillae of abattoir pigs can harbor MRSA strains with multiple antimicrobial resistances. In addition, these MRSA were possibly able to colonize the host either without stimulating antibody production, by attenuating the immune response or by colonizing pigs that are less immunocompetent. This may explain the success of MRSA colonization and persistence in pigs. Further studies are required to better elucidate MRSA colonization in abattoir pigs and their public health risk.

Impact of neonatal total parenteral nutrition and early glucose-enriched diet on glucose metabolism and physical phenotypes in Guinea Pig

Les oxydants infusés avec la nutrition parentéral (NP) néonatale induisent une modification du métabolisme des lipides et du glucose, donnant lieu à l’âge adulte à un phénotype de carence énergétique (faible poids, baisse de l’activité physique). L’hypothèse qu’une diète précoce riche en glucose prévient ces symptômes plus tard dans la vie, fut évalué chez le cobaye par un ANOVA en plan factoriel complet à deux facteurs (p < 0:05) : NP du jour 3 à 7, suivit d’une nourriture régulière (chow) (NP+) vs. chow à partir du 3ième jour (NP-), combiné avec une eau de consommation enrichie en glucose (G+) ou non (G-) à partir de la 3ième semaine. Les paramètres suivant ont été mesurés à l’âge de 9 semaine: taux de croissance, activité physique, activité de phosphofructokinase-1 et glucokinase (GK), niveau hépatique de glucose-6-phosphate (G6P), glycogène, pyruvate et potentiel redox du glutathion, poids du foie, glycémie, tolérance au glucose, concentrations hépatiques et plasmatiques en triacylglycérides (TG) et cholestérol. Le groupe G+ (vs. G-) avait un taux de croissance plus bas, une activité de GK et une concentration en G6P plus élevée, et un potentiel redox plus bas (moins oxydé). Le niveau plasmatique de TG était moins élevé dans le groupe NP+ (vs. NP-). Les traitements n’eurent aucun effet sur les autres paramètres. Ces résultats suggèrent qu’indépendamment de la NP, une alimentation riche en glucose stimule la glycolyse et déplace l’état redox vers un statut plus réduit, mais ne surmonte pas les effets de la NP sur le phénotype physique de carence énergétique. / Neonatal exposure to oxidant molecules from total parenteral nutrition (TPN) alters future lipid and glucose metabolism, resulting in an energy deficient phenotype characterized by lower body weight and physical activity. Using a guinea-pig model, the hypothesis that early diet supplementation with glucose could overcome such symptoms at week 9 of age was tested in a two-factor full-factorial ANOVA design (p<0:05): TPN day 3-7, chow thereafter (TPN+) vs: chow from day 3 (TPN-), combined with glucose-enriched diet from week 3 (G+) vs: plain chow throughout (G-). The growth rate, physical activity, phosphofructose kinase-1 and glucose kinase (GK) activities, glucose-6-phosphate (G6P), glycogen and pyruvate concentrations, relative liver weight, fasting blood glucose, glucose tolerance, hepatic and plasma triacylglyceride and cholesterol levels, individual glutathione levels and GSH/GSSG-based redox potential were determined at 9 weeks. Glucose supplementation (vs: the lack thereof) resulted in a lower growth rate, higher GK activity, and higher G6P concentration at week 9. Plasma triacylglycerides at week 9 were lower in TPN+ (vs: TPN-) subjects. Hepatic GSH=GSSG-derived redox potential shifted to a more reduced state in G+ (vs: G-) subjects. No other parameters showed significant differences. Independently of TPN, an early glucose-rich diet stimulated the glycolysis pathway, shifted the redox potential towards a more reduced status ; however, it did not overcome the effects of TPN on future physical and metabolic phenotype.

Carmen Klein: Sonographie der Lunge und Analyse der Atmungsmechanik mittels Impuls-Oszilloresistometrie beim lungengesunden und pneumoniekranken Ferkel und Läuferschwein Institut für Bakteriologie und Mykologie der Veterinärmedizinischen Fakultät der Universität Leipzig Bundesinstitut für gesundheitlichen Verbraucherschutz und Veterinärmedizin (BgVV), Fachbereich 4 in Jena eingereicht im Mai 1999, verteidigt am 16. November 1999 (157 Seiten, 52 Abbildungen, 34 Tabellen, 285 Literaturstellen; Anhang mit: 62 Seiten, 15 Abbildungen und 50 Tabellen) Mit der Sonographie der Lunge und der Impuls-Oszilloresistometrie (IOS - engl.: Impulse Oscilloresistometry System) wurden erstmals zwei nichtinvasive diagnostische Verfahren auf ihre Anwendbarkeit und Aussagefähigkeit bei lungengesunden und pneumoniekranken Absatzferkeln und Läuferschweinen bis zu einer Körpermasse von ca. 50 kg geprüft. Die sonographische Untersuchung der Lunge wurde an insgesamt 45 lungengesunden bzw. pneumoniekranken Schweinen durchgeführt. Dazu wurden die Tiere mit 2,0 mg/kg KM Diazepam sediert. Eine Studie an sechs klinisch gesunden Schweinen gab Aufschluß über die Ausdehnung des bei dieser Tierart sonographisch erreichbaren Lungenfeldes. Die bei Pneumonien häufig betroffenen kranioventral gelegenen Lungenabschnitte waren sonographisch gut zu erreichen. Die bei der sonographischen Untersuchung der Lunge des Schweines nachgewiesenen Befunde zeigten eine gute Übereinstimmung mit den in der Literatur an anderen Spezies beschriebenen Resultaten. Beim Schwein wurden folgende Befundbilder abgegrenzt: ? keine Veränderungen : sonographisch o.b.B. ? geringgradige Veränderungen: Kometenschweifartefakte in geringer Ausprägung ? mittelgradige Veränderungen: Kometenschweifartefakte und Konsolidierungen oder echogenes Grundmuster mit Kometenschweifartefakten ? hochgradige Veränderungen: echogenes Grundmuster oder echogenes Grundmuster mit Abszeß oder Nekrose Die Befunde der sonographischen Untersuchung der Lunge wurden bei 34 Schweinen den entsprechenden pathologisch-anatomischen Befunden zugeordnet. Pathologisch-anatomisch unverändertes Gewebe stellte sich mit einer Wahrscheinlichkeit (Sensitivität) von 95,65 % ohne bzw. mit nur geringradigen sonographischen Veränderungen dar. Bei pathologisch-anatomisch verändertem Gewebe wurden mit einer Wahrscheinlichkeit von 74,62 % (Sensitivität) mittel- und hochgradig veränderte sonographische Befundbilder festgestellt. In die histologische Untersuchung wurden insgesamt 49 Gewebeproben einbezogen. Rückschlüsse vom sonographischen Befund auf die Art der histologisch diagnostizierten Pneumonie waren beim Schwein nicht möglich. Es wurde jedoch eine statistisch gesicherte Beziehung zwischen dem Ausprägungsgrad der histologischen Veränderung und den sonographischen Befunden festgestellt. Der Ausprägungsgrad der histologischen Veränderung stand u.a. in direkter Beziehung zum Luftgehalt im Lungengewebe. Die Impuls-Oszilloresistometrie ist ein Verfahren zur Analyse der Atmungsmechanik und wurde am Schwein ebenfalls unter Sedation mit Diazepam (1,5 bis 2,0 mg/kg KM) oder aber nach entsprechendem Training der Tiere ohne jede medikamentelle Ruhigstellung vorgenommen. Die Schweine wurden in einer Hängematte streßarm fixiert und das Meßsystem über eine starre, luftdicht am Kopf abschließende Atemmaske an das Tier adaptiert. Insgesamt 26 Schweine wurden in die Studien zur Untersuchung verschiedener Einflüsse auf das Verhalten der IOS-Parameter einbezogen. Folgende Parameter wurden berücksichtigt: Atmungsfrequenz (Af), Atemzugvolumen (Vt), die Spektralparameter Resistance (R), Reactance (X) und Kohärenz (K) jeweils bei 5, 10, 15 und 20 Hz (R5, ...R20, X5, ...X20, K5, ...K20) sowie die Modellparameter zentrale (Rz) und periphere (Rp) Resistance, Lungencompliance (Cl) und zentrale Inertance (Lz). In einer Variabilitätsanalyse an fünf nicht sedierten, klinisch gesunden Schweinen wurde die Reproduzierbarkeit der Ergebnisse bei drei unmittelbar innerhalb weniger Minuten aufeinander folgend sowie bei an drei Tagen nacheinander zur selben Tageszeit durchgeführten IOS-Messungen überprüft. Innerhalb des kurzen Zeitraumes von wenigen Minuten wurde die Variabilität der Meßergebnisse der meisten IOS-Parameter überwiegend durch inter-individuelle Einflüsse verursacht, während bei der über mehrere Tage reichenden Betrachtungsweise der intra-individuell bedingte Anteil der Variabilität zunahm. Die Entwicklung von Alter und Körpermasse konnten im Rahmen der an elf klinisch gesunden, sedierten Schweinen über einen Zeitraum von 62 Tagen vorgenommenen IOS-Messungen nur teilweise die mit dem Wachstum einhergehenden Veränderungen der IOS-Parameter erklären. Der an acht klinisch gesunden, sedierten Schweinen festgestellte Einfluß der Tageszeit auf die IOS-Parameter war nur gering, stimmte aber von der Tendenz her mit den in der Literatur beschriebenen Angaben überein. An 16 klinisch gesunden Schweinen wurde der Einfluß der Sedation mit Diazepam auf die IOS-Parameter überprüft. Dieses Medikament führte zu einer signifikanten Verringerung von Af und Vt und zu einer Verbesserung der Qualität der IOS-Messungen (K5, K10, K20). An sieben klinisch gesunden, nicht sedierten Schweinen wurden durch Inhalation von Aerosolen mit bestimmten pharmakologisch wirksamen Substanzen (Carbachol, Fenoterolhydrobromid) definierte Zustände am respiratorischen System wie Bronchospasmus und Bronchodilatation ausgelöst und mittels IOS-Messungen nachgewiesen. Die während der bronchospastischen Reaktion vorgefundenen Veränderungen der IOS-Parameter, insbesondere der starke Anstieg der Resistance im unteren Frequenzbereich (R5), das deutliche Absinken der Reactance, insbesondere bei 5 Hz, und die beträchtliche Erhöhung des Modellparameters Rp sind für diesen Zustand des Bronchialsystems charakteristisch. Neun klinisch gesunde, sedierte Schweine wurden zunächst bei physiologischer und anschließend bei stark distal abgebeugter Kopfhaltung untersucht. Durch die starke Flexion im Kopf-Hals-Bereich wurde eine extrathorakal gelegene Obstruktion erzeugt. Hierbei waren die spektrale Resistance (R5 bis R20) und der Modellparameter Rz bei abgebeugter Kopfhaltung signifikant erhöht. Am Beispiel einer experimentell ausgelösten Infektion mit Actinobacillus pleuropneumoniae wurde der Verlauf einer respiratorischen Erkrankung an sieben sedierten Schweinen unter regelmäßiger Durchführung von IOS-Messungen, über einen Zeitraum von 23 Tagen verteilt, beobachtet. Die Resultate der IOS-Messungen 3 Tage p.i. (Resistance im gesamten Frequenzbereich erhöht, starker Anstieg von Rz und Lz ) wiederspiegelten vor allem eine Beteiligung der extrathorakal und zentral gelegenen Bereiche des Respirationstraktes. Durch die zumindest tendenziell vorhandenen Veränderungen bestimmter IOS-Parameter (Verstärkung der negativen Frequenzabhängigkeit, Erhöhung von Rp, Abnahme von Cl) wurde zusätzlich eine Beteiligung der peripheren Bereiche in der Lunge angezeigt. Für einen definierten Zeitraum wurden die IOS-Meßergebnisse den Befunden der sonographischen Untersuchung der Lunge gegenübergestellt. Dabei wurden hohe Korrelationen zwischen auf periphere Veränderungen verweisenden IOS-Parametern und sonographisch ermittelten Veränderungen an der Lunge nachgewiesen. Abschließend ist festzustellen: 1.) Beide getesteten diagnostischen Verfahren sind beim Schwein anwendbar und liefern eine wertvolle Ergänzung der konventionellen Diagnostik am Respirationstrakt dieser Tierart. 2.) Nach sonographischer Untersuchung der Lunge sind mit hoher Wahrscheinlichkeit Aussagen über das Vorhandensein pathologisch-anatomisch sichtbarer pneumonischer Verän-derungen und deren Ausdehnung im sonographisch erreichbaren Lungenfeld möglich. 3.) Die Impuls-Oszilloresistometrie liefert in einem Parameter-Komplex Informationen über den funktionellen Zustand des gesamten respiratorischen Systems. Dieser ist von zahlreichen physiologisch wirkenden Faktoren (u.a. Kopfhaltung, Wachstum, Tageszeit), applizierten pharma-kologisch wirksamen Substanzen (z.B. Diazepam, Carbachol, Fenoterolhydrobromid) und dem Vorhandensein einer Erkrankung des respiratorischen Systems (z.B. Actinobacillus pleuropneumoniae) abhängig. 4.) Da beide Verfahren verschiedene Sachverhalte, den morphologischen (Sonographie) bzw. den funktionellen Zustand (Impuls-Oszilloresistometrie) des respiratorischen Systems widerspiegeln, liefern sie einander ergänzende Aussagen. 5.) Beide diagnostischen Verfahren bieten sich beim Schwein vor allen Dingen für den Einsatz zu wissenschaftlichen Zwecken an. Aufgrund ihres nichtinvasiven Charakters ist ein schonender Umgang mit den Probanden möglich. 6.) Die Anwendung der Sonographie der Lunge ist auch in ausgewählten Fällen der tierärztlichen Praxis, z.B. beim Ankauf von Zuchttieren, denkbar. / Carmen Klein: Lung Sonography and Analysis of the Respiratory Mechanics by means of Impulse Oscilloresistometry in Healthy and Pneumonic Piglets and Porkers Institute of Bacteriology and Mycology Faculty of Veterinary Medicine University of Leipzig Germany Federal Institute for Health Protection of Consumers and Veterinary Medicine Jena Branch Germany submitted in May 1999 (157 pages, 52 figures, 34 tables, 285 references, appendix with 62 pages, 15 figures, 50 tables) Two non-invasive diagnostic methods, i.e. lung sonography and impulse oscillometry, were examined for the first time with regard to their applicability and evidence in clinically healthy as well as pneumonic weaned piglets and porkers of up to ca. 50 kg b.w. A total of 45 healthy or pneumonic swine was subjected to lung sonography. For this purpose, the animals were sedated with 2.0 mg/k b.w. diazepam. Six clinically healthy swine were studied to establish the lung area accessible to sonography in this animal species. Sonography was found to cover well the cranioventral sections of the lung often affected by pneumonias. The findings established by lung sonography in the swine showed a good coincidence with results described in literature for other species. The findings in swine revealed the following picture. ? No changes: sonography NAD ? Low-grade changes: little pronounced comet-tail artefacts ? Moderate changes: Comet-tail artefacts and consolidations or echogenic basic pattern including comet-tail artifacts ? High-degree changes: basic echogenic pattern or basic echogenic pattern plus abscess or necrosis Lung sonography findings in 34 swine were assigned to the corresponding pathological-anatomical findings. Tissue considered as unchanged from the pathological-anatomical angle was found to be without or with only minor sonographic changes, with a probability (sensitivity) of 95.65 %. In tissue exhibiting pathological-anatomical changes, sonographic findings showed pictures of moderate and high-degree changes, with a probability (sensitivity) of 74.62 % A total of 49 tissue samples was subjected to histological examination. It was not possible to infer, from the sonographic findings, the type of pneumonia diagnosed histologically in the swine. Nevertheless, a statistically confirmed correlation between the degree of pronouncedness of the histological changes and the sonographic findings was established. The pronouncedness of the histological changes was, like other factors, directly related to the air content of the lung tissue. Impulse oscillometry being a method used to analyze respiratory mechanics was applied to swine either sedated by means of diazepam (1.5 - 2.0 mg/kg b.w.) or non-sedated by medication but trained correspondingly. The animals were fixed in a canvas sling with openings for the limbs, avoiding stress, and the measuring system was adapted to the animal using a rigid face mask hermetically fitting to the animal's head. A total of 26 animals was included in the studies to examine a variety of factors influencing the IOS parameters. The following parameters were considered: Respiratory rate (Af), tidal volume (Vt), the spectral parameters resistance (R), reactance (X) and coherence (K) at 5, 10, 15 and 20 Hz each (R5, ...R20, X5, ...X20, K5, ...K20) as well as the model parameters, central (Rz) and peripheral (Rp) resistance, lung compliance (Cl) and central inertance (Lz). A variability analysis involving five non-sedated, clinically healthy swine served to examine the reproducibility of the results of three IOS measurements either performed consecutively within a few minutes, or on three consecutive days at the same time of the day. During the brief period of a few minutes, the variability of the results of measurements, for most parameters, was due to inter-individual factors while in the case of an analysis over several days, the intra-individual share of variability increased. In the context of the IOS measurements performed in eleven clinically healthy, sedated swine over a period of 62 days, development in terms of age and body weight could only partially explain the variations of the IOS parameters taking place with growth. The influence of the time of the day on IOS parameters studied in eight clinically healthy, sedated swine was found to be small; coincidence with data given in literature, however, was seen as a tendency. The influence of diazepam sedation on the IOS parameters was examined in 16 clinically healthy swine. This medication resulted in a significant reduction of Af and Vt and improved quality of IOS measurements (K5, K10, K20). In seven clinically healthy, non-sedated swine, defined respiratory situations, i.e. bronchospasm and bronchodilatation were generated by aerosol inhalation of pharmacologically active substances such as carbachol and fenoterolhydrobromide and demonstrated by IOS measurements. This state of the bronchial system is characterized by the changes in IOS parameters found at the time of the bronchospasmic reaction, in particular the vehement rise of resistance in the lower frequency range (R5), the obvious drop in reactance, especially at 5 Hz and the noteworthy rise in the model parameter, Rp. Nine clinically healthy, sedated swine were examined first with their heads in a physiological and then, in a vertical position of their heads. The severe bending in the head-neck area produced an extrathoracic obstruction. Spectral resistance (R5 - R20) and the model parameter, Rz were significantly higher at a vertical position of the head. An experimental infection of seven sedated swine with Actinobacillus pleuropneumoniae served as a model to study the course of respiratory disease. The animals were subjected to regular IOS measurements over a period of 23 days. The results of IOS measurements performed 3 days p.i. (elevated resistance over the entire frequency range, vehement rise of Rz and Lz) reflected, above all, an involvement of the extrathoracic and central areas of the respiratory tract. As a result, the modifications of defined IOS parameters (intensification of negative frequency dependency, rise in Rp, drop in Cl) which were present at least as a tendency, indicated an additional involvement of the peripheral lung areas. The results of IOS measurements over a defined period were compared with the findings from lung sonography. High correlations between IOS parameters indicating peripheral changes and lung changes detected by sonography were demonstrated. Conclusions 1.) Both diagnostic methods studied may be used in swine and will provide a valuable completion of conventional diagnosis in the respiratory tract of this species. 2.) With a high probability, lung sonography will provide evidence of the presence of visible pathological-anatomical changes in the lung and their extent in the lung area that is accessible to sonography. 3.) Impulse oscillometry provides a complex of parameters that contain information about the functional state of the entire respiratory system. This state is dependent upon numerous physiological factors (among them position of the head, growth, time of the day), pharmacologically active substances administered (e.g. diazepam, carbachol, fenoterolhydrobromide) and the presence of respiratory disease (e.g. caused by Actinobacillus pleuropneumoniae). 4.) Since the two methods reflect different facts, i.e. the morphological (sonography) and the functional (impulse oscillometry) state of the respiratory system, they supply mutually supplementing information. 5.) Both diagnostic methods offer themselves above all for use in scientific research on swine. Owing to their non-invasive character, it is possible to handle test animals in a non-offensive way. 6.) In specific cases, lung sonography may also be used in the veterinary practitioner's surgery, e.g. for the examination of animals kept for breeding.

The Effect of CO2-Pneumoperitoneum on Ventilation Perfusion Distribution of the Lung

Laparoscopic operations are a common and popular way for abdominal procedures. They are usually performed by insufflation of carbon dioxide (CO2) into the abdominal cavity. However, insufflation of CO2 may interfere with cardiac and circulatory as well as respiratory functions. The CO2-pneumoperitoneum (PP) may cause hypercarbia and acidosis. The direct effects of CO2 and acidosis lead to decreased cardiac contractility, sensitization of the myocardium to arrhythmogenic effects of catecholamines and systemic vasodilatation. There may even be long-lasting post-operative effects on breathing control. The pneumoperitoneum may also cause several respiratory changes, e.g. decreased functional residual capacity (FRC) and vital capacity (VC), formation of atelectasis, reduced respiratory compliance and increased airway pressure. Still, arterial oxygenation is mostly maintained or even improved during PP. In view of the apparently contradictory results in respiratory mechanics and gas exchange, the present studies were performed to evaluate respiratory changes on gas exchange and ventilation-perfusion distributions during PP in a porcine model. It was demonstrated that atelectasis during anaesthesia and PP may be estimated by an increased arterial to endtidal PCO2-gradient (study I). Perfusion was redistributed away from dorsal, collapsed lung regions when PP was established. This resulted in a better ventilation-perfusion match (study II). Increasing abdominal pressure shifted blood flow more and more away from collapsed lung tissue, decreased pulmonary shunt and improved oxygenation from 8 to 16 mmHg PP, despite an increase of atelectasis formation (study III). CO2-PP enhanced the shift of blood flow towards better ventilated parts of the lung compared to Air-PP. Moreover, sodium natriumprusside worsened the ventilation-perfusion match even more and blunted the effects previously seen with carbon dioxide. CO2 should therefore be the mediator of enhancing HPV during PP. In conclusion, pneumoperitoneum with CO2 causes atelectasis with elimination of ventilation in the dependent lung regions. However, an efficient shift of blood flow away from collapsed, non-ventilated regions results in a better ventilation-perfusion matching and better oxygenation of blood than without PP. A prerequisite for the beneficial effect is the use of carbon dioxide for the abdominal inflation, since it enhances HPV.

El present treball analitza al microscopi òptic i al microscopi electrònic de transmissió les glàndules sexuals accessòries de Sus domesticus (raça Landrace - varietat anglesa) a partir de mascles reproductors porcins adults i sans. Un millor coneixement dels patrons estructural i ultraestructural normals de las glàndules sexuals accessòries permetrà diagnosticar amb facilitat quina ha estat l'estructura o la funció glandular afectada en mascles en els que s'observa una disminució de la qualitat del semen. Per altra banda, els estudis anatomopatològics s'han de complementar amb tècniques histoquímiques que generalment permeten confirmar o excloure un diagnòstic histopatològic previ.Les glàndules sexuals accessòries del mascle reproductor porcí estan molt desenvolupades i inclouen les glàndules vesiculars, la pròstata i les glàndules bulbouretrals. L'epiteli secretor de les glàndules vesiculars està format per cèl·lules columnars, cèl·lules basals i mastòcits. Les cèl·lules columnars es caracteritzen per presentar tres morfologies diferents que es consideren diferents estadis d'un mateix tipus cel·lular: les cèl·lules principals, les cèl·lules clares i les cèl·lules denses. Les cèl·lules principals secreten activament glicoproteïnes N- i O- glicosilades amb residus d'&#945;-L-fucosa, &#945;(1&#61543;6)fucosa, &#945;-D-mannosa, &#945;-D-glucosa, &#945;- i &#61538;-D-N-acetilgalactosamina, &#61538;-D-galactosa-&#61538;(1&#61543;3)-D-N-acetilgalactosamina, &#945;-D-galactosa, galactosa-&#61538;(1&#61543;4)-N-acetilglucosamina, D-N-acetilglucosamina i àcid neuramínic. Aquestes glicoproteïnes afavoreixen les interaccions entre l'espermatozoide i l'occit i regulen la permeabilitat de la membrana espermàtica. La pròstata està formada per dues porcions glandulars, el cos de la pròstata (BP) y la pròstata disseminada (DP), entre las quals s'observen diferencies estructurals, ultraestructurals, histoquímiques i funcionals. En ambdues porcions, l'epiteli secretor està constituït per cèl·lules columnars principals, denses i cèl·lules basals, i també per cèl·lules cúbiques en el BP i per cèl·lules mucoses en la DP. En ambdues porcions glandulars, se sintetitzen i secreten N- i O- glicoproteïnes neutres i àcides. Aquestes glicoproteïnes s'alliberen mitjançant un mecanisme regulat en el BP y mitjançant un mecanisme regulat i un constitutiu en la DP. Aquests residus proporcionen resistència a la proteolisi a les O-glicoproteïnes secretades, les quals participen en la lubrificació y protecció de l'epiteli, i intervenen en el control de la permeabilitat del plasmalemma dels espermatozoides i en el transport d'ions al seu través. / The present study evaluates by means of light and transmission electron microscopy, the accessory sex glands coming from adult and healthy boars (Sus domesticus Landrace breed - British variety). A deeper knowledge of the normal structural and ultrastructural patterns of the accessory sex glands will allow diagnosing easily the structure or the glandular function affected when a low spermatic quality is observed. On the other hand, anatomopathological studies must to be complemented with histochemical techniques which generally allow confirming or rejecting a previous histopathological diagnostic. The boar accessory sex glands were very developed and formed by the vesicular glands, the prostate and the bulbourethral glands. Columnar cells, basal cells and mast cells composed the secretory epithelium of the vesicular glands. Columnar cells showed three morphologies, which were considered different stages of the same cell typology and named: principal cells, clear cells and dense cells. Principal cells secreted actively N- and O- glycoproteins that exhibited residues of: &#945;-L-fucose, &#945;(1&#61543;6)fucose, &#945;-D-mannose, &#945;-D-glucose, &#945;- and &#61538;-D-N-acetilgalactosamine, &#61538;-D-galactose-&#61538;(1&#61543;3)-D-N-acetilgalactosamine, &#945;-D-galactose, galactose-&#61538;(1&#61543;4)-N-acetilglucosamine, D-N-acetilglucosamine and neuraminic acid. These glycoproteins improve sperm-oocyte interactions and regulate plasma membrane permeability.The prostate was constituted by two glandular portions, the prostate body (PB) and the disseminate prostate (DP). Both of them showed structural, ultrastructural, histochemical and functional differences. Principal columnar cells, dense columnar cells and basal cells composed the secretory epithelium of these two glandular parts. The PB contained also cubic cells and the DP contained mucous cells. Both glandular portions synthesised and secreted N- and O- neutral and acid glycoproteins. These glycoproteins were thought to be released by a regulated pathway in the BP while in the DP the glycoproteins seemed to follow a regulated and a constitutive pathway. The BP luminal glycoproteins exhibited residues of: fucose, mannose, &#945;- and &#61538;-D-N-acetilgalactosamine, galactose-&#61538;(1&#61543;4)-N-acetilglucosamine, D-N-acetilglucosamine and neuraminic acid. The DP glycoproteins also contained &#61538;-D-galactose-&#61538;(1&#61543;3)-D-N-acetilgalactosamine and &#945;-D-galactose residues.The BP and the DP glycoproteins secreted by the regulated pathway participate in the control of sperm membrane stability, avoid uterine immune response and sperm agglutination and, improve their progressive motility. The DP glycoproteins secreted by the constitutive pathway protect and lubricate the pelvic urethra.Principal and dense piramidal cells composed the secretory epithelium of the bulbourethral glands. The principal cells mainly synthesised and secreted carboxylated and sulfated acid O-glycoproteins with residues of: N-acetilgalactosamine, &#61538;-D-galactose-&#61538;(1&#61543;3)-D-N-acetilgalactosamine, &#945;-D-galactose, D-N-acetilglucosamine and neuraminic acid. These O-linked oligosaccharides make these proteins extremely resistant to proteolysis, provide lubrication and protection to the epithelium, and participate in ion transport throughout the plasma membrane.

In folk medicine, the decoction of its roots is used as abortive (AGRA, 1991). In a preliminary study, Santos et al. (2003) demonstrated that crude ethanolic extract of aerial parts from S. agrarium (SA-EtOH) showed non-selective spasmolytic activity on guinea-pig ileum and rat uterus. The aim of the study was evaluate on the cytotoxic potential of SA-EtOH in rats erythrocytes and to investigate the spasmolytic action mechanism on guinea-pig ileum and rat uterus. SA-EtOH extract showed moderate hemolytic activity only at concentration of 243 and 500 μg/mL (Emax = 21.2 ± 3.2%). On guinea-pig ileum, SA-EtOH extract inhibited the cumulative concentration-response curves to carbachol (CCh) and histamine, and these were shifted to the right, in a non-parallel manner, with depression of the maximal effect (Emax), suggesting a noncompetitive antagonism. SA-EtOH extract relaxed in a significant and concentration dependent manner the ileum pre-contracted with 40 mM KCl (EC50 = 17.4 ± 3.5 μg/mL), 10-6 M CCh (EC50 = 119.4 ± 24.3 μg/mL) or 10-6 M histamine (EC50 = 18.7 ± 4.6 μg/mL), suggesting that SA-EtOH extract could be acting on voltage-gated Ca2+ channels (Cav). This assumption was confirmed by observation that SA-EtOH extract antagonized the CaCl2 induced contractions in the depolarizing medium without Ca2+. The finding that SA-EtOH extract relaxed (EC50 = 187.2 ± 27.3 μg/mL) the guinea-pig ileum pre-contracted with S-(-)-BAY K8644 suggests that the Ca2+ channel subtype involved is the CaV1. As the SA-EtOH extract was more potent in relaxing the body pre-contracted with KCl than by S-(-)-Bay K8644, it is suggestive of indirect blockade of the Cav1. We decided to investigate the participation of K+ channels in the spasmolytic action of SA-EtOH extract. The relaxation curve induced by SA-EtOH extract was rightward displacement only in the presence of tetraethylammonium (TEA+) 1 mM, this concentration is a selective blocker of the large conductance clacium-activated K+ channels (BKca) (EC50 = 37.7 ± 4.9 μg/mL) and 4-aminopyridine, selective blocker of the voltage activated K+ channels (Kv) (EC50 = 48.9 ± 6.2 μg/mL). On rat uterus, SA-EtOH extract inhibited the cumulative concentration-response curves to oxytocin, and these were shifted to the right, in a non-parallel manner, with depression of the maximal effect (Emax) suggesting a noncompetitive antagonism. SA-EtOH extract did not effective at relax the uterus pre-contracted with 60 mM KCl (Emax = 10.5 ± 5.0%), however relaxed in a significant and concentration dependent manner the rat uterus pre-contracted with 10-2 UI/mL (EC50 = 56.3 ± 8.2 μg/mL). Therefore, suggesting that SA-EtOH extract could be promoting its spasmolytic effect of indirect blockade influx Ca2+ by Cav, as observed in guinea-pig ileum. However, the relaxation curve induced by SA-EtOH extract was rightward displacement only in the presence of glibenclamide (EC50 = 124.8 ± 21.0 μg/mL), selective blocker of the ATP-sensitive K+ channels (KATP), different from that observed in guinea-pig ileum. These results suggest that SA-EtOH extract is not promoting the membrane injury on rat erythrocytes and that the spasmolytic action mechanism of SA-EtOH extract involves modulation/activation of BKCa and KV on guinea-pig ileum, and of KATP on rat uterus. / Solanum agrarium Sendtner é conhecida popularmente por babá , gogóia e melancia da praia . O decocto da raiz é empregado popularmente como abortivo (AGRA, 1991). Em estudos anteriores, Santos et al., (2003) demonstraram que o extrato etanólico bruto obtido das partes aéreas de S. agrarium (SA-EtOH) apresentou atividade espasmolítica não seletiva em íleo de cobaia e útero de rata. O objetivo desse estudo foi avaliar o potencial citotóxico do extrato SA-EtOH em eritrócitos de rato e investigar o mecanismo de ação espasmolítica em íleo de cobaia e útero de rata. O extrato SA-EtOH apresentou uma atividade hemolítica moderada apenas nas concentrações de 243 e 500 μg/mL (Emax = 21,2 ± 3,2%). Em íleo de cobaia, o extrato SA-EtOH inibiu as curvas cumulativas ao carbacol (CCh) e à histamina e estas foram desviadas para a direita de maneira não paralela com redução do Emax, sugerindo um antagonismo não competitivo. O extrato SA-EtOH relaxou de maneira dependente de concentração e significante o íleo de cobaia pré-contraído com 40 mM de KCl (CE50 = 17,4 ± 3,5 μg/mL), ou com 10 6 M de CCh (CE50 = 119,4 ± 24,3 μg/mL) ou de histamina (CE50 = 18,7 ± 4,6 μg/mL), sugerindo que o extrato SA-EtOH deve estar agindo bloqueando os canais de Ca2+ dependentes de voltagem (CaV). A confirmação desta hipótese veio com a observação de que o extrato SA-EtOH antagonizou as contrações induzidas por CaCl2 em meio despolarizante nominalmente sem Ca2+. O fato do extrato SA-EtOH ter relaxado (CE50 = 187,2 ± 27,3 μg/mL) o íleo pré-contraído com S-(-)-BAY K 8644 sugere que o canal de Ca2+ envolvido é o tipo CaV1. Como a potência relaxante do extrato SA-EtOH foi maior quando o órgão foi pré-contraído com KCl do que pelo S-(-)-Bay K8644, isto é sugestivo de que há um bloqueio indireto dos Cav1. Decidiu-se investigar a participação dos canais de K+ na ação espasmolítica do SA-EtOH. A curva de relaxamento induzida pelo extrato SA-EtOH foi desviada para direita apenas na presença de tetraetilamônio (TEA+) 1 mM, nessa concentração um bloqueador dos canais de K+ ativados por Ca2+ de grande condutância (BKCa) (CE50 = 37,7 ± 4,9 μg/mL) e na de 4-aminopiridina, bloqueador dos canais de K+ operados por voltagem (KV) (CE50 = 48,9 ± 6,2 μg/mL). Em útero de rata, o extrato SA-EtOH inibiu as curvas cumulativas à ocitocina e esta foi desviada para a direita de maneira não paralela com redução do Emax, sugerindo um antagonismo não competitivo. O extrato SA-EtOH não foi eficaz em relaxar o útero pré-contraído com 60 mM de KCl (Emax = 10,5 ± 5,0%), entretanto relaxou de maneira dependente de concentração o útero pré-contraído com 10-2 UI/mL ocitocina (CE50 = 56,3 ± 8,2 μg/mL). Diante deste fato, pode-se sugerir que o extrato SA-EtOH pode estar promovendo seu efeito espasmolítico pelo bloqueio indireto do influxo de Ca2+ através dos CaV, assim como o observado em íleo de cobaia. Entretanto, a curva de relaxamento induzida pelo extrato SA-EtOH foi desviada para direita apenas na presença de glibenclamida (CE50 = 124,8 ± 21,0 μg/mL), bloqueador seletivo dos canais de K+ sensíveis ao ATP (KATP), diferente do observado em íleo de cobaia. Esses resultados são sugestivos de que o extrato SA-EtOH não esteja promovendo injúrias a membrana dos eritrócitos de ratos e que o mecanismo de ação espasmolítica do extrato SA-EtOH se dá pela modulação/ativação dos BKCa e KV em íleo de cobaia e pelos KATP em útero de rata.

Avaliação e modelização da dinâmica de carbono e nitrogênio em solo com o uso de dejetos de suínos / Evaluation and simulation of soil carbon and nitrogen dynamics with the use of pig manure

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / The managedment of the pig manure, in the slurry or solid form (deep bedding), and how it is applied to the soil, in the surface or incorporated, must affect the dynamics of the C and the N in the soil. The objectives of this work were: 1) to evaluate the dynamics of the C and the N in soil and to evaluate the N supply to maize which were applied pig manure in the slurry and solid form, in no-tillage and minimum tillage system; and 2) to use the STICS model (BRISSON et al., 1998) to simulate the dynamics of N and water in the soil-plant system with the use of pig slurry in maize under no-tillage system. To reach the first objective, three experiments were realized. Two of them in the field conditions, during the period of 2002/03 in the Experimental Area of the UFSM/RS Soil Department in a typic Hapludult and one of them in laboratory conditions. The treatments consisted in the application or no application of pig manure in the slurry and solid form (deep bedding) on the crop oats residues with and without incorporation to the soil in the maize. Aiming to reach the second objective, a data set generated in the Experimental Area of the UFSM Soil Department during the period of 1998 to 2002, involving the N dynamics in the soil-plant system, with the use of pig slurry in notillage system was used. The gotten results had indicated that the N losses by NH3 volatilization were higher with the application of pig slurry than with the application of solid manure and that these losses decrease with the incorporation of the manure to the soil. The amounts of mineral N in the soil layer of 0-90 cm did not differ between the treatments with and without incorporation of the manure in spite of the lost N amount by volatilization has been higher in the treatment without incorporation. The incorporation of the manure to the soil did not increase the straw C decomposition and nor the N immobilization. The presence of the oats straw stimulated the immobilization of the ammoniacal N applied with the pig slurry. The solid manure presented low nitrogenous fertilizing otential to the maize when compared to the pig slurry in terms of the low mineralization rate of the organic N, present in the deep bedding. The N-N2O emission increased with the application of the pig slurry on the oats straw in relation to the system with straw and without pig slurry. The pig slurry ammoniacal 15N recovery by the maize in physiological maturation stage was only 14.9%. In the same period, only 49.2% of the ammoniacal 15N applied with the slurry pig slurry were recuperated in the soil and the plant (aerial part + roots), indicating that more than a half of the ammoniacal N applied with the pig slurry was lost through the volatilization, lixiviation and denitrification processes. The STICS model underestimates the amount of N lost by ammonia volatilization after the pig slurry application. The use of a factor in STICS to express the N losses by ammonia volatilization provided an improvement in the simulations of the N dynamics in the soil. The water dynamics in the soil layer of 0 - 60 cm was simulated by STICS with an error lower than 13.5%. These work results showed that the STICS reparametred model presents potential to be used in subtropical weather conditions in the simulation of the N and water dynamics in no-tillage system with the use of pig manure. / A forma como os dejetos de suínos são manejados, se na forma líquida ou sólida (cama sobreposta), e o modo de aplicação dos mesmos no solo, se na superfície ou incorporados, devem afetar a dinâmica do C e do N no solo. Os objetivos deste trabalho foram: 1) avaliar a dinâmica do C e do N no solo e o fornecimento de N ao milho com uso de dejetos de suínos na forma líquida e sólida, em sistema plantio direto e preparo reduzido do solo; e 2) utilizar o modelo STICS (BRISSON et al., 1998) para simular a dinâmica do N e da água no sistema solo-planta com o uso de dejetos líquidos de suínos em sistema plantio direto de milho. Para atingir o primeiro objetivo, foram realizados três experimentos, sendo dois em condições de campo, no período de 2002/03 na área Experimental do Departamento de Solos da UFSM/RS, em solo Argissolo Vermelho distrófico arênico, e um em condições de laboratório. Os tratamentos consistiram da aplicação ou não de dejetos de suínos na forma líquida e sólida (cama sobreposta), sobre os resíduos culturais de aveia com e sem incorporação ao solo, na cultura do milho. Com vistas ao alcance do segundo objetivo, foi utilizado um conjunto de dados gerados na área Experimental do Departamento de Solos da UFSM no período de 1998 a 2002 envolvendo a dinâmica do N no sistema solo-planta com o uso de dejetos líquidos de suínos em sistema plantio direto. Os resultados obtidos indicaram que as perdas de N por volatilização de NH3 foram maiores com a aplicação de dejetos líquidos de suínos do que com a aplicação de dejetos sólidos e que essas perdas diminuíram com a incorporação dos dejetos ao solo. As quantidades de N mineral na camada de 0-90 cm do solo não diferiram entre os tratamentos com e sem incorporação dos dejetos apesar da quantidade de N perdida por volatilização ter sido maior no tratamento sem incorporação. A incorporação dos dejetos ao solo não aumentou a decomposição do C da palha e nem a imobilização de N. A presença da palha de aveia estimulou a imobilização do N amoniacal aplicado com os dejetos de suínos. Os dejetos sólidos apresentaram baixo potencial fertilizante nitrogenado ao milho, comparado aos dejetos líquidos, em função da baixa taxa de mineralização do N orgânico presente na cama sobreposta. A emissão de N-N2O aumentou com a aplicação dos dejetos líquidos sobre a palha de aveia em relação ao sistema com palha e sem dejetos. A recuperação do 15N amonical dos dejetos líquidos pelo milho no estádio de maturação fisiológica foi de apenas 14,9%. Nesse mesmo período, apenas 49,2% do 15N amoniacal aplicado com os dejetos líquidos de suínos foram recuperados no solo e na planta (parte aérea + raízes), indicando que mais da metade do N amoniacal aplicado com os dejetos foi perdido através dos processos de volatilização, lixiviação e desnitrificação. O modelo STICS subestimou a quantidade de N perdida por volatilização de amônia, após a aplicação dos dejetos de suínos. O uso de um fator em STICS para expressar a perdas de N por volatilização de amônia proporcionou uma melhora nas simulações da dinâmica do N no solo. A dinâmica da água na camada 0 60 cm do solo foi simulada por STICS com um erro inferior a 13,5%. Os resultados desse estudo evidenciaram que o modelo STICS reparametrado apresenta potencial para ser utilizado em condições de clima subtropical na simulação da dinâmica do N e da água em sistema plantio direto com o uso de dejetos de suínos.
