Near Field Communication (NFC) är en trådlös kommunikationsteknik för korta avstånd, vanligast med cirka 10 cm vilket medför en säkrare förbindelse. NFC är en relativt ung teknologi som är billig att implementera samt användarvänlig. NFC är lämpad främst för mobiltelefoner där en bred implementering av applikationer är möjlig. Tekniken medför snabbare och enklare utbyte av information genom att ställa enheter sida vid sida. I dagsläget används tekniken mest i Asien. I Europa har ingen stor satsning påbörjats än, men den är på väg. I framtiden förväntas det att NFC kommer att finnas i de flesta mobiltelefoner.Denna uppsats behandlar praktisk implementering av en ”verktygslåda” med NFC teknologi. Målet är att skapa en ”verktygslåda” med olika komponenter. ”verktygslådan” är tänkt att användas som grundskelett för implementering av NFC applikationer och för experiment med de olika komponenterna. Hårdvarukomponenterna består av mobiltelefon, tagg, RFID läsarmodul och mikroprocessor. Programvarukomponenterna består av ett antal kodexempel.Vårt arbete har resulterat i tre prototyper som tillsammans utgör ”verktygslådan”. I prototyperna sker kommunikation mellan en kombination av komponenter i ”verktygslådan”: mobiltelefon och tagg, läsare och tagg samt läsare och mobiltelefon. / Near Field Communication (NFC) is a contactless short distance (approximately 10 centimeters) communication technology, which conveys a more secure connection. NFC is a relatively young technology which is cheap to implement, and user-friendly. NFC is directed to mainly cellphones, where opportunities for implementation of applications are wide. The technology conveys a faster and easier exchange of information, by putting units side by side. At present, the technology is used mostly in Asia. In Europe, no major effort has begun yet, but is expected to. NFC is expected to exist in a majority of cellphones, in the future.This essay treats practical implementation of a “toolbox” with the NFC technology. The goal was to make a “toolbox” with different components. The “toolbox” is intended as a base for implementing NFC applications and for experimenting with the different components. The hardware components are: cellphone, a tag, a RFID module and a microprocessor. The program components consist of a number of code examples.Our work has resulted in three prototypes which together constitute a “toolbox”. The prototypes communicate between any combinations of the components in the “toolbox”: cellphone with tag, reader with tag and reader with cellphone.

När sociala nätverk blir en del av vår vardag öppnas uppenbara möjligheter till interaktion och nätverkande på Internet med vänner, kollegor och bekanta. Samtligt som sociala nätverk erbjuder användare intressanta redskap, med många fördelar, för att kunna interagera utsätts samtidigt användares personliga integritet för risker och hot. Denna uppsats undersöker vad som oroar användare mest på Internet, värdet av en skyddad privat sfär utan insyn, varför vi delar med oss av privat information på sociala nätverk, vad kan man ta reda på utifrån den information som finns tillgänglig samt hur kan vi bära oss åt för att inte förlora vår integritet på Internet. Utöver ett stort fokus på sociala nätverk behandlas även positionsbaserade tjänster och hur dessa tjänster kan användas utan att integriteten går förlorad. Med detta i åtanke genomfördes en enkätundersökning av svenska studenters syn på integritet och medvetenhet i sociala nätverk samt deras inställning och vanor i positionsbaserade tjänster. Slutsatserna av undersökningen resulterade i reflektioner och diskussioner om vår syn på integritet och antaganden inför framtiden. / When social networking becomes a part of our everyday life opportunities for interaction and networking with friends, colleagues and other acquaintances opens up. While social networking offers and interesting approach to interacting it exposes users' personal privacy to external threats. This thesis examines elements that worries users on the Internet, the value of having a protected personal sphere, why we disclose personal information on social networks, what information can be obtained through data mining of personal profiles and how we can carry on with our online presence without losing our online privacy. Beyond the immediate focus on social networks, privacy in location-based services gets a review and a closer look into how it can be used without forfeiting privacy. With this in mind, a survey including Swedish students' views on privacy and general awareness on social networks and attitudes towards location-based services was conducted. The conclusion of the survey resulted in reflections and discussions about outlooks on privacy and assumptions about the future.

Membrane Domain of Plant 3-Hydroxy-3-Methylglutaryl Coenzyme A Reductase: Targeting, Topology, and Function

The rate limiting step in isoprenoid biosynthesis is catalyzed by 3-hydroxy-3-methylglutaryl CoA reductase (HMGR, EC 1.1.1.34). In plants, HMGR is encoded by small gene families whose members are differentially expressed. In tomato, hmg2 was previously isolated and sequenced. We report the isolation and sequence analysis of a clone (pCD4) encompassing exon I of tomato hmg1 which encodes the putative membrane domain. Sequence comparisons of plant HMGR proteins reveal two hydrophobic stretches within the amino terminus which are highly conserved among species. Using in vitro transcription and translation systems, the membrane domain structure of two tomato HMGR isoforms, HMG1 and HMG2, were analyzed. Results from these experiments reveal that tomato HMGRs are targeted to microsomal membranes in a cotranslational fashion that does not involve cleavage of an N-terminal targeting peptide. Membrane topography of HMGR was revealed by protease protection studies, indicating that both tomato HMGRs span the membrane two times such that both the C- and N-termini are located within the cytosol. HMG2 but not HMG1 was glycosylated in the in vitro system. Deletion of the hmg1 5' untranslated regions and sequences encoding the first six highly charged amino acids resulted in inefficient translation in vitro. However, targeting to microsomes was unchanged. HMG1 membrane domain was tagged with a FLAG epitope to facilitate in vivo studies. Agrobacterium-mediated transformation was used to introduce the tagged hmg1 gene into two Nicotiana tabacum cell lines, BY-2 and KY-14. The slow growth kinetics of KY-14 prevented effective recovery of transformed lines, however, Northern analyses of BY-2 showed that the hmg1 transgene was expressed. Comparisons of BY-2 and KY-14 revealed differences in defense responses to elicitor treatment. BY-2 cells showed minimal defense capabilities, whereas KY-14 cells were rapidly induced as indicated by increased HMGR enzyme activity and browning of the cells. HMGR enzyme activity was decreased in both KY-14 and BY-2 cells following sterol treatment, but the reduction was more pronounced in KY-14 cells. Thus transgenic BY-2 cells may be useful in future in vivo immunolocalization studies, but analyses of HMGR transcriptional regulation and regulated degradation will require use of the more responsive KY-14 cells.. / Ph. Systemization of RFID Tag Antenna Design Based on Optimization Techniques and Impedance Matching Charts

The performance of commercial Radio Frequency Identification (RFID) tags is primarily limited by present techniques used for tag antenna design. Currently, industry techniques rely on identifying the RFID tag application (books, clothing, etc.) and then building antenna prototypes of different configurations in order to satisfy minimum read range requirements. However, these techniques inherently lack an electromagnetic basis and are unable to provide a low cost solution to the tag antenna design process. RFID tag performance characteristics (read-range, chip-antenna impedance matching, surrounding environment) can be very complex, and a thorough understanding of the RFID tag antenna design may be gained through an electromagnetic approach in order to reduce the tag antenna size and the overall cost of the RFID system. The research presented in this thesis addresses RFID tag antenna design process for passive RFID tags. With the growing number of applications (inventory, supply-chain, pharmaceuticals, etc), the proposed RFID antenna design process demonstrates procedures to design tag antennas for such applications. Electrical/geometrical properties of the antennas designed were investigated with the help of computer electromagnetic simulations in order to achieve optimal tag performance criteria such as read range, chip-impedance matching, antenna efficiency, etc. Experimental results were performed on the proposed antenna designs to compliment computer simulations and analytical modelling.

Influenza A Viren besitzen ein segmentiertes, einzelsträngiges RNA-Genom, welches in Form viraler Ribonukleoprotein (vRNP)-Komplexe verpackt ist. Während das virale Genom im Zellkern repliziert wird, finden Assemblierung und Knospung reifer Viruspartikel an der apikalen Plasmamembran statt. Für die Virusbildung müssen die einzelnen viralen Komponenten hierher gebracht werden. Während intrinsische apikale Signale der viralen Transmembranproteine bekannt sind, sind der zielgerichtete Transport und der Einbau des viralen Genoms in neuentstehende Virionen noch wenig verstanden. In dieser Arbeit wurden potentielle Mechanismen des vRNP-Transportes untersucht, wie die Fähigkeit der vRNPs mit Lipidmembranen zu assoziieren und die intrinsische subzellulären Lokalisation des viralen Nukleoproteins (NP), eines Hauptbestandteils der vRNPs. Es konnte gezeigt werden, dass vRNPs nicht mit Lipidmembranen assoziieren, was mittels Flotation aufgereinigter vRNPs mit Liposomen unterschiedlicher Zusammensetzung untersucht wurde. Die Ergebnisse deuten jedoch darauf hin, dass das virale M1 in der Lage ist, Bindung von vRNPs an negativ-geladene Lipidmembranen zu vermitteln. Subzelluläre Lokalisation von NP wurde des Weiteren durch Expression fluoreszierender NP-Fusionsproteine und Fluoreszenzphotoaktivierung untersucht. Es konnte gezeigt werden, dass NP allein nicht mit zytoplasmatischen Strukturen assoziiert, stattdessen aber umfangreiche Interaktionen im Zellkern eingeht und mit hoher Affinität mit bestimmten Kerndomänen assoziiert, und zwar den Nukleoli sowie kleinen Kerndomänen, welche häufig in der Nähe von Cajal-Körperchen und PML-Körperchen zu finden waren. Schließlich wurde ein experimenteller Ansatz etabliert, welcher erlaubt, den Transport vRNP-ähnlicher Komplexe mittels Fluoreszenzdetektion aufzuzeichnen und Einzelpartikelverfolgungsanalysen durchzuführen. Unterschiedliche Phasen des vRNP-Transportes konnten beobachtet werden und ein 3-Phasen-Transportmodell wird skizziert. / Influenza A viruses have a segmented single-stranded RNA genome, which is packed in form of viral ribonucleoprotein (vRNP) complexes. While the viral genome is replicated and transcribed in the host cell nucleus, assembly and budding of mature virus particles take place at the apical plasma membrane. Efficient virus formation requires delivery of all viral components to this site. While intrinsic apical targeting signals of the viral transmembrane proteins have been identified, it still remains poorly understood how the viral genome is transported and targeted into progeny virus particles. In this study, potential targeting mechanisms were investigated like the ability of vRNPs to associate with lipid membranes and the intrinsic ability of the viral nucleoprotein (NP) – which is the major protein component of vRNPs – for subcellular targeting. It could be shown that vRNPs are not able to associate with model membranes in vitro, which was demonstrated by flotation of purified vRNPs with liposomes of different lipid compositions. Results indicated, however, that the matrix protein M1 can mediate binding of vRNPs to negatively charged lipid bilayers. Intrinsic subcellular targeting of NP was further investigated by expression of fluorescent NP fusion protein and fluorescence photoactivation, revealing that NP by itself does not target cytoplasmic structures. It was found to interact extensively with the nuclear compartment instead and to target specific nuclear domains with high affinity, in particular nucleoli and small interchromatin domains that frequently localized in close proximity to Cajal bodies and PML bodies. An experimental approach was finally established that allowed monitoring the transport of vRNP-like complexes in living infected cells by fluorescence detection. It was possible to perform single particle tracking and to describe different stages of vRNP transport between the nucleus and the plasma membrane. A model of three-stage transport is suggested.

<p>The Company Siemens Industrial Turbo Machinery AB manufactures and launches in operation among other things gas turbines, steam turbines, compressors, turn-key power plants and carries out service for components for heat and power production. Siemens also performs research and development, marketing, sales and installations of turbines and completes power plants, service and refurbish.</p><p>Our thesis for the engineering degree is to develop an automatic status log which will be used as a tool to control how the status of the machine is before and after technical service at gas turbines. Operational disturbances will be registered in a structured way in order to get a good possibility to follow up the reliability of the application.</p><p>An automatic log function has been developed and will be activated at start, stop and shutdown of the turbine system. Log files are created automatically and get a name with the event type, the date and the time. The files contain data as timestamp, name, measured values and units of the signals which are going to be analyzed by the support engineers. They can evaluate the cause of the problem using the log files.</p>
