1	Effects of Site Directed Mutagenesis of the Second Exon of the Adenovirus 5 E1A Gene on Transcriptional Activation / Mutagenesis of the Second Exon of the AD5 E1A Gene Skiadopoulos, Mario 06 1900 (has links) The early region 1a oncogene of adenovirus 5 codes for proteins that can activate transcription of viral and cellular genes. This study describes the construction of three deletions and one point mutation that together span the entire coding region of the second exon of E1A. The exon-2 mutants were tested for their ability to activate transcription from the adenovirus early region 3 promoter (E3) in transient expression assays. Dl1116 (dl aa 205-221) did not affect transactivation of E3 in pKCAT-23. Sub1117 (dl exon-2 aa) and dl1115 (dl aa 188-204) were unable to activate transcription. Pm1131 (SER-219 to stop) had a reduced transactivating efficiency but was still able to stimulate transcription. These results define the 3' boundary of a transactivation domain on the E1A proteins as being between positions 188 and 204. Results obtained in our lab define the 5' boundary as being between 138-147 (Jelsma et al., 1988). The mutants that could not transactivate were tested for their ability to block wildtype E1A transactivation of the E3 promoter in assays similar to those described by Glenn and Ricciardi (1987). Dl1115 and sub1117 appeared to block transactivation by WT E1A. In transient expression assays, the fatty acid sodium butyrate was found to stimulate transcription of the CAT gene, when added to the medium of HeLa cells transfected with pKCAT-23. 2	Sequences in Adenovirus 5 E1A Gene that are Required for Transcriptional Activation, Enhancer Repression, and Oncogenic Transformation / Functional Domains in Adenovirus 5 E1A Jelsma, Anthony 09 1900 (has links) The E1A gene of adenovirus 5 carries out a number of functions in infection and oncogenic transformation, including the transcriptional activation of viral and cellular genes, the repression of transcriptional enhancers, and cooperation with the adenovirus E1B gene or with the ras oncegene to transform primary cells. The purpose of this work was to investigate the mechanism of action of E1A, by determining the regions of the proteins that are required for these functions. Deletion and point mutations were made in the region unique to the larger E1A mRNA, by exonuclease digestion and deletion loop mutagenesis respectively. These mutants and a series of mutants which delete sequences spanning the entire coding region, were examined for their effect on transcriptional activation, enhancer repression, and transformation. The region which, when deleted, rendered E1A defective for transcriptional activation was found to be confined to the region unique to the 13s mRNA and the beginning of exon 2. Mutations in three regions, all within the 12s exon 1, affected repression activity. The first two, the N terminal region of the protein, and a region, CR1, conserved between adenovirus serotypes, were essential for repression activity. The third region, at the end of exon 1 of the 12s mRNA, was probably only indirectly involved in repression. Deletions in three regions of exon 1 resulted in a loss of the transforming function of E1A. The first two corresponded to the regions required for repression, suggesting that enhancer repression is a component of transformation. The third region, containing CR2, also conserved between adenovirus serotypes, is functionally distinct from the other two and appeared to affect the morphology of the transformants. These two functions did not operate efficiently when present ion separate plasmids. 6	Friction Intended : Exploring the overlooked potential of designing for effort. Zuilhof, Daphne January 2014 (has links) Ease, comfort and efficiency are assumed desirables; they form the established norm of unquestioned values in commercial product design. The norm shapes our everyday. Those daily things considered mundane and commonplace, form how we go about our daily doings. How we actually live our lives. Design needs to be there to defend human interest; to approach the user differently than a passive consumer, to create space for human qualities in contemporary everyday life. I have been exploring the potential of designing for effort, and argue for its value by giving examples. I have developed a series of three products under the shared name Friction Intended. The proposals evoke effort of different kinds, each creating space for other alternative values. Object A is a light concept working with reflections. Reflecting from one surface to another the light can be followed and its behavior studied. The reflective elements are tools for exploration and active learning; to actively perceive the daily phenomenon of light. Object B is a backpack to be assembled from a large sheet of textile and a set of straps. Over time, the usage of the bag can become a personal ritual. Wearing the bag can also be a statement; how will people react when the large cloth is dramatically folded open in a public environment? Object C is a cup with rounded base. The cup moves; never fully finding its balance it sways back and forth ever differently depending on the amount of liquid inside and the qualities of the gestures it has been handled with. The attention is drawn to the moment by giving careful attention to a simple daily ritual. Designing for effort in everyday products creates space to design for an engaging and stimulating environment. Once deciding simple things are worth more time, strain and patience there is the opportunity to enrich those activities. Effort has the potential to create space for the development of contemporary rituals, active engagement and everyday curiosity. The design space of the potential of effort is a rich and varied. The examples given by the Friction Intended series, are representatives for a field where much more potential still lays. 7	Inflammasome : Investigating the effect of NEK7 in the activation of the NLRP3 Inflammasome Adindu Uzowuru, Cosmas January 2020 (has links) Inflammation is a biological defence mechanism applied by living organisms against foreign invaders. In the response to DAMPs and PAMPs, organisms use inflammatory multi-protein complexes to fight the attackers. The most studied inflammasome proteins are NLRP3, ASC and Caspase-1. This study is aimed at understanding the role of NEK7 protein in the NLRP3 inflammasome’s activation, using CRISPR/Cas9 system. To determine the effect of CRISPR/Cas9 and transfection, mRNA expression was analyzed. The results obtained suggest that neither the transfection nor the NEK7 protein knockout have sufficiently worked. This study could not experimentally establish that NEK7 triggers NLRP3 inflammasome activation because ELISA was not conducted to verify the levels of cytokines emitted, due to there being no statistical differences between the samples. Above all, the research question in this thesis project was not answered because the instability of the ACTB reference gene negatively influenced the results. However, previous related studies conclude that NEK7 plays a crucial role in the activation of the NLRP3 inflammasome. Pohořálková, Pavla January 2013 (has links) The diploma thesis focuses on teaching grammar in the context of in-company language courses. What needs to be taken into consideration are both the psychological aspects of adult learners and the sociological characteristics of a typical course participant, their position of a client which is connected to high expectations of the service provided - language tuition. The role of the teacher in an in-company course is much less authoritative than in a traditional course and teachers become rather partners or coaches for the learners. The pedagogical research conducted among 172 Czech in-company learners with the help of an electronic questionnaire shows that the main aim of in-company students is to be able to communicate efficiently in real-life situations from their private and working lives. It is the role of the teacher, or the coursebook author, to choose which grammar structures are necessary to succeed in the respective situations. Therefore, the most suitable method for teaching grammar in the context of in-company courses is the Engage, Study, Activate method combined with the eclectic method.

10	Hur blir du en framgångsrik tiggare i Sverige? : en undersökning av tiggandets och givandets bilder 2011 till 2016 / How do you become a successful beggar in Sweden? : an inquiry into the images of begging and giving 2011 to 2016 Parsberg, Cecilia January 2016 (has links) Mitt första möte med en tiggande föranledde mig att under fem år undersöka den nya situationen för tiggeriet och giveriet i Sverige. Förutsättningen är att vardagliga handlingar och reaktioner gentemot en annan människa kan synliggöras estetiskt med en etisk klangbotten. Min undersökning utspelar sig i första hand i gaturummet och i medierna. Det är hela tiden bilderna som är utgångspunkten för resonemangen och de gestaltande verken. Bilder som både separerar och länkar samman kroppar. Vilka bilder är i spel i tiggeriets och giveriets sociala koreografi? Hur kan bilder i detta sammanhang aktiveras på nya sätt? Hur kan nya genereras? Tiggandet är en uppmaning till social interaktion, och vare sig givaren socialt interagerar eller inte med den tiggande människan på gatan så involveras givaren i det europeiska samfundets asymmetriska värdesystem. I min första gestaltning anlitas en professionell marknadsundersökare för att ta reda på hur en tiggare i Sverige skulle kunna göra för att bli framgångsrik. Det blir en film som jag sedan visar mittemot en film där tiggande pratar om hur givare ger. Ur detta verk följer så en rad gestaltningar och en interdisciplinär teoretisk diskussion med bland andra Judith Butler, Sara Ahmed och Hannah Arendt, samt med en rad konstnärers arbeten, kring hur bilder – och kroppsliga handlingar – är kopplade till samhällsbilden och samhällskroppen? The premise is that every-day actions and reactions to another person can be made visible through aesthetics with ethical underpinnings. My investigation takes place mainly in the urban landscape and in the media. The images always constitute the point of departure for the reasoning and for the staged works. Images that separate as well as connect bodies. Which images are at play in the social choreography of begging and giving? In this context, how can images be activated in new ways? How can new images be generated? Begging is a call to social interaction, and regardless of whether the giver interacts socially with the begging person on the street, the giver is implicated in the asymmetrical value systems of the European Union. In my first staged work I hire a professional market researcher to find out how a beggar in Sweden should behave to be successful. This becomes a film that I then show opposite another film in which begging people talk about how givers give. This is followed by a number of staged works and an interdisciplinary theoretical discussion involving, among others, Judith Butler, Sara Ahmed, and Hannah Arendt, as well as a number of artistic works concerning how images – and bodily actions – are linked to the social image and the body politics. The arrangement of the choirs in the staged work The Chorus of Begging and The Chorus of Giving, indicates a space for social interaction and thus demonstrates a different order that demands different actions in terms of language, movement, and attitude toward each other. It’s a social choreography: when the choirs rehearsed and sung together a political form emerged. My hope is to make visible a space for action between the begging and the giving that can be used for continued ethical negotiations and new staged works. / <p>Föreliggande doktorsarbete har genomförts och handletts i forskarutbildningen i Fri konst vid Konsthögskolan, Umeå universitet.
