Decolorization of an azo and anthraquinone textile dye by a mixture of living and non-living Trametes versicolor fungus

Wastewater from the textile industry is difficult to treat effectively due to the prevalent use and wide variety of synthetic dyes that are resistant to conventional treatment methods. White-rot fungi, such as Trametes versicolor, have been found to be effective in decolorizing many of these synthetic dyes and current research is focusing on their application to wastewater treatment. Although numerous studies have been conducted on the ability of both living and nonliving Trametes versicolor to separately decolorize textile dyes, no studies were found to have investigated the use of a mixture of live and dead fungus for decolorization. This study explored potential interactions between live and dead, autoclaved Trametes versicolor biomass in a mixed system by utilizing a series of batch tests with two structurally different synthetic textile dyes. Samples were analyzed by spectrophotometer and compared with controls to determine the effect of any interactions on decolorization. The results of this study indicate that an interaction between living and nonliving biomass occurred that affected the specific dye removal for both Reactive Blue 19, an anthraquinone textile dye, and Reactive Orange 16, an azo textile dye. This interaction was seen to improve the specific dye removal during the first 10-46 hours of experimentation but then diminish the specific dye removal after this period. This effect could be due to hydrophobins, which are surface-active proteins excreted by live fungi that may alter hydrophobicity. Additionally, the presence of adsorptive dead biomass could affect dye contact with degrading enzymes released from the live fungus. By expanding current knowledge of the interactions that take place in a fungal bioreactor and their effect on textile dye decolorization, this research aims to inspire more effective and less costly bioreactor designs for the treatment of textile wastewater.

The thesis entitled “New Supramolecular Ion Sensing Probes and their Application in the Detection of Environmentally Relevant Ions” deals with the design and synthesis of several small molecular probes which can specifically sense environmentally relevant ions of (anion or cation) particularly in aqueous or biological medium. The probes have been designed using four different molecular entities which include anthraquinone, oxidized bis-indolyl system, pyrene and rhodamine. The probes afford naked eye detection of a particular ion in the aqueous medium. This work has been divided into six chapters. Chapter 1. Introduction The first chapter gives a brief idea of ion sensor. It provides the description of various approaches used for designing molecular sensors. The chapter further presents an overview of the four different dyes (anthraquinone, oxidized-bis-indole, pyrene and rhodamine) used for designing probes in this work. The properties of these probes, their advantages and disadvantages to use as a signaling subunit have been discussed. This chapter also describes the use of micellar medium for solubilizing different organic dyes in water. Chapter 2. Colorimetric Probes based on Anthraimidazolediones for Selective Sensing of Fluoride and Cyanide ion via Intramolecular Charge Transfer. The second chapter describes the design and synthesis of four different probes based on anthra [1, 2-d] imidazole-6, 11-dione. The anthraquinone part of each molecule has an acceptor moiety whereas substituted nitrogen linked aromatic unit forms the donor site. Each probe acted as strong colorimetric sensor for fluoride and cyanide ion detection and exhibited intramolecular charge transfer (ICT) band which showed significant red-shifts after addition of either the F¯ or CN¯ ion. One of the probes 2 showed selective colorimetric sensing for both cyanide and fluoride ions. In organic medium 2 showed selective color change with fluoride and cyanide, whereas in aqueous organic medium it showed a selective ratiometric response towards cyanide ion. The effect of anionic charge (on the donor moiety) on ICT has been discussed. Among the various donor moieties, the donor site having negative charges on them was found to disperse greater electron density on them. Figure 1. Molecular structures of the sensors Chapter 3 deals with chemodosimetric detection of cyanide ion in water using various oxidized bis-indole based compounds. Chapter 3A. A Chemodosimetric Probe based on a Conjugated and oxidized Bis¬ indolyl System for Selective Naked Eye Sensing of Cyanide ion in Water. The chapter 3A describes the design and synthesis of a new water-soluble bis-indolyl based probe, 5 which possesses two –COOH groups. This probe specifically reacted with the CN¯ ion in pure water at ambient temperature and produced a remarkable change in color from red to colorless. The mechanism of this process was investigated by NMR (1H, 13C and DEPT-135) spectroscopy, mass spectrometry and kinetic studies. The mechanism investigation showed that the cyanide ion reacts with the probe and removes the conjugation of the bis-indolyl moiety of the probe with that of the 4-substituted aromatic ring which renders the probe colorless. Taken together a plausible mechanism of the reaction was presented which showed to operate via a Michael type adduct formation under ambient conditions of pH and temperature in water. The probe gave a detection limit of 0.38 ppm for detection of cyanide ion in water. Figure 2. Molecular structure of the probe 5. Chapter 3B. Micelle Assisted ppb level Detection of Cyanide ion in Water by Chemodosimetry and Visual detection of the Endogenous Cyanide. The chapter 3B deals with the synthesis of a bis-indole based colorimetric probe 6. The probe showed selective detection of the cyanide ion in water at ppb level and a visible detection of endogenous cyanide from cassava (a major staple food in the developing world) by chemodosimetry. The cyanide ion binds with the probe 6 in a chemodosimetric fashion and follows pseudo first-order kinetics in water under appropriate conditions. It showed a highly sensitive detection of the cyanide ion in water with a detection limit of 0.33 ppm. The use of the micellar medium improved the detection limit drastically and a ppb level detection limit was achieved. The probe also showed the detection of the endogenously bound cyanide in cassava both visually and by spectrophotometer. Figure 3. Molecular structure of the probe 6. Chapter 3C. Ratiometric Cyanide ion probe in Water and for the detection of the Endogenously bound cyanide. Chapter 3C presents the synthesis of two new bis-indolyl (7 and 8) based probes for colorimetric detection of cyanide ion in pure water. Compound 8 showed a ratiometric response with cyanide in water and a visual detection of the endogenously bound cyanide ion in cassava. Using compound 8 the selective detection of the cyanide ion in water was achieved with a detection limit of ~ 17 ppb which is almost 13 times lower than the permitted limit as specified by EPA, United States. 7; R = H 8; R = -(OCH2CH2)3CH3 Figure 4. Molecular structures of the probes 1 and 2. Chapter 4 deals with the colorimetric and ratiometric detection of the Cu2+and Hg2+ions using different small synthetic molecular probes. Chapter 4A. Colorimetric Sensors for Ratiometric Detection of Copper and Mercury ions in Biological media and below ppm level in Water. The chapter 4A deals with the synthesis of two novel colorimetric probes (9, 10) using bispicolyl unit as the binding moiety and anthraimidazolediones and bis-indolyl system as a signaling sub-unit. Using the two sensors, Cu2+ion can be detected below the permitted limit (1.3 ppm) in both drinking water and at physiological pH 7.4. Sensor 9 can detect both Cu2+and Hg2+ in water with very low detection limit. It showed specific binding with Cu2+ at physiological pH 7.4 and in presence of serum albumins. Chemosensor 10 can be used for the specific detection of both Cu2+and Hg2in water as well as for the contamination in microorganisms. Figure 5. Molecular structure of the sensors 9 and 10. Chapter 4B. A New Molecular Probe for the Selective Sensing of Cu2+ and Hg2+ ions in Micellar Media and in Live ells.This chapter describes a synthesis of a novel bispicolyl based sensor 11 which can detect Cu2+ ion specifically in water medium and both Cu2+ and Hg2+ ions selectivelyin Brij-58 micellar medium. In micellar medium both the ions can be detected in the ppb level. Using fluorescence spectroscopy these two metal ions can be discriminated.The probe is also be useful for checking metal ion contamination in cellular samples. Figure 6. Molecular structure of the sensor 11. Chapter 4C. Rhodamine based Sensors for Cu2+ and Hg2+ ions in Water and in Biological media. The chapter 4C presents the synthesis and the sensing properties of the three positional isomers of the pyridine end of the rhodamine-pyridine compounds (12-14). The three isomers only differ in the position of nitrogen of the pyridine moiety. Sensor 12, which contains the pyridine nitrogen at the ortho-position showed selective sensing toward Cu2+ ion in both pure water and in buffered physiological media of pH 7.4. It gave a detection limit of ~13 ppb which is 100 times lesser than the EPA permitted limit. The other two sensors 13 and 14, which possessed the pyridine ends with the nitrogen atom at the meta- and the para- positions respectively showed the selective sensing of Hg2+ ion in water and did not show any interaction with the Cu2+ ion. Probes 2 and 3 showed ‘turn-on’ detection of Hg2+ ion both in the UV-vis and the fluorescence emission spectroscopy. Compound 2 and 3 showed a detection limit of ~ 9 and 4 ppb respectively. The NMR titration showed the change in color was due to the opening of the spirolactam ring of the rhodamine. The sensors can also be used for the detection of Cu2+ and Hg2+ ion in real life water samples and in the live cells. Figure 7. Molecular structure of the sensors 12, 13 and 14. Chapter 5. Ratiometric and ppb level Detection of Toxic Transition Metal ions using a Single Probe in Micellar media. This chapter describes the selective sensing of multiple ions using a single probe 15. The probe incorporates pyrene and pyridine as signaling and interacting moiety respectively. The sensor showed different responses towards different metal ions just by varying the medium of detection. In organic solvent (acetonitrile), the probe showed selective detection of Hg2+ ion. In water the fluorescence quenching was observed with three metal ions, Cu2+, Hg2+ and Ni2+. Further just by varying the surface charge of different micellar media, the probe showed selective interaction with Hg2+ ion in neutral micelles (Brij-58). However, in anionic micellar medium (SDS), the probe showed selective changes with both Cu2+ and Ni2+ in the UV-vis spectroscopy. The discrimination between these two ions was achieved by emission spectroscopy, where it showed selective quenching only with Cu2+. Thus using a single probe all the three metal ions Cu2+, Hg2+ and Ni2+ can be detected and discriminated just by varying the surface charge of the micellar medium. Figure 8. Molecular structure of the sensors 15. Chapter 6. Highly sensitive Rhodamine Based Dual Probes for the Visual detection of F¯ and Hg2+ ions in Water. This chapter deals with the design and synthesis of two new rhodamine based probes (16-17) which act as dual probes for the ppb level selective detection of Hg2+ and F¯ ions in water and at physiological pH 7.4. The two probes were synthesized by coupling tert-butyldiphenylsilyl (TBDPS) protected forms of 4-hydroxybenzaldehyde and 2, 4- dihydroxy benzaldehyde with rhodamine hydrazone. The F¯ ion detection is based on the desilylation of the probe, whereas the spirolactam ring opening leads to the detection of Hg2+ ion. The two probes gave turn-on detection of both Hg2+ and F¯ ion selectively in aqueous medium with the detection limit well below the EPA permitted limits. The probes showed detection of both the ions by dual mode with visibly different color and fluorescence under UV-lamp. The F¯ ion interacts with the silyl bond of probe and the cleavage results into yellow color whereas; the addition of Hg2+ ion to the probe solution opened the spirolactam ring and resulted into appearance of pink color. Studies on the regulation of conidiation in species of Trichoderma

A characteristic feature of species of Trichoderma is the production of concentric rings of conidia in response to alternating light-dark conditions. In response to a single burst of light, a single ring of conidia forms at what was the colony perimeter. On the basis of these observations, competency to photoconidiate has been proposed to be due to the age and metabolic rate of the hyphal cell. In this study, conidiation was investigated in five biocontrol isolates (T. hamatum, T. atroviride, T. asperellum, T. virens and T. harzianum) using both a morphological and molecular approach. All five isolates produced concentric conidial rings under alternating light-dark conditions on potato-dextrose agar (PDA), however, in response to a 15 min burst of blue light, only T. asperellum and T. virens produced a clearly, defined conidial ring which correlated with the colony margin at the time of light exposure. Both T. harzianum and T. hamatum photoconidiated in a disk-like fashion and T. atroviride produced a broken ring with a partially filled in appearance. On the basis of these results, it was postulated that competency to photoconidiate is a factor of the metabolic state of the hyphal cell rather than chronological age or metabolic rate. The influence of the source of nitrogen on photoconidiation was assessed on pH-buffered (pH 5.4) minimal medium (MM) amended with glutamine, urea or KNO₃. In the presence of glutamine or urea, T. asperellum and T. harzianum conidiated in a disk, whereas, when KNO₃ was the sole nitrogen source, a ring of conidia was produced. Further, in the presence of increasing amounts of glutamine, the clearly defined photoconidial ring produced on PDA by T. asperellum became disk-like. These results clearly demonstrated that primary nitrogen promotes photoconidiation in these isolates and strongly suggests that competency of a hyphal cell to conidiate in response to light is dependent on the nitrogen catabolite repression state of the cell. The experiments were repeated for all five isolates on unbuffered MM. Differences were apparent between the buffered and unbuffered experiments for T. atroviride. No photoconidiation was observed in T. atroviride on buffered medium whereas on unbuffered medium, rings of conidia were produced on both primary and secondary nitrogen. These results show that photoconidiation in T. atroviride is influenced by the buffering capacity of the medium. Conidiation in response to light by T. hamatum and T. virens was absent in all nitrogen experiments, regardless of the nitrogen source and buffering capacity, whereas both isolates conidiated in response to light on PDA. These results imply that either both sources of nitrogen are required for photoconidiation, or a factor essential for conidiation in these two isolates was absent in the minimal medium. Mycelial injury was also investigated in five biocontrol isolates of Trichoderma. On PDA, all isolates except T. hamatum conidiated in response to injury. On nitrogen amended MM, conidiation in response to injury was again observed in all isolates except for T. hamatum. In T. atroviride, injury-induced conidiation was observed on all medium combinations except the pH-buffered MM amended with glutamine or urea and T. virens conidiated in response to injury on primary nitrogen only, regardless of the buffering capacity. These results have revealed conidiation in response to injury to be differentially regulated between isolates/species of Trichoderma. On unbuffered MM amended with glutamine or urea, conidiation in response to injury occurred at the colony perimeter only in T. atroviride. It was hypothesised that the restriction of conidiation to the perimeter may be due to changes in the pH of the agar. The experiment was repeated and the pH values of the agar under the growing colony measured at the time of light induction (48 h) or injury (72 h). The areas under the hyphal fronts were acidified to below the starting value of the medium (pH 5.4) and the centres of the plates were alkalinised. The region of acidification at the time of stimuli correlated with the production of conidia, which implicates a role for crossregulation of conidiation by the ambient pH. The influence of the ambient pH on injury-induced conidiation was investigated in T. hamatum and T. atroviride on MM amended with glutamine and PDA, pH-buffered from pH 2.8 to 5.6. Thickening of the hyphae around the injury site was observed at the lowest pH values on MM in both T. atroviride and T. hamatum, however no conidia were produced, whereas both Trichoderma species conidiated on pH-buffered PDA in a strictly low pH-dependent fashion. This is the first observation of injury-induced conidiation in T. hamatum. The influence of the ambient pH on photoconidiation was assessed in T. hamatum, T. atroviride and T. harzianum using both buffered and unbuffered PDA from pH 2.8 to 5.2. On buffered PDA, no conidiation in response to light was observed above pH 3.2 in T. hamatum, above 4.0 in T. atroviride and above 4.4 in T. harzianum, whereas on unbuffered PDA it occurred at all pH values tested. It was postulated that conidiation at pH values above 4.4 on unbuffered PDA was due to acidification of the agar. The pH values of the agar under the growing colony were measured at the time of light exposure and in contrast to the MM with glutamine experiments, alkalisation of the agar had occurred in both T. atroviride and T. hamatum. No change in medium pH was recorded under the growing T. harzianum colony. These results indicate that low pH-dependence of photoconidiation is directly related to the buffering capacity of the medium. Recent studies have linked regulation of conidiation in T. harzianum to Pac1, the PacC orthologue. In fungi, PacC regulates gene expression in response to the ambient pH. In these studies pH-dependent photoconidiation occurred only on buffered PDA and on unbuffered PDA conidiation occurred at significantly higher ambient pH levels. It is proposed that the influence of ambient pH on conidiation in the isolates used in this study is not due to direct Pac1 regulation. The T. harzianum isolate used in this study produced profuse amounts of the yellow anthraquinone pachybasin. Production of this secondary metabolite was strictly pH-dependent, irrespective of the buffering capacity of the medium. Studies in T. harzianum have linked Pac1 regulation to production of an antifungal α-pyrone. pH-dependence on both buffered and unbuffered media strongly suggests that pachybasin production may also be under the control of Pac1. Photoconidiation studies on broth-soaked filter paper, revealed rhythmic conidiation in the pachybasin producing T. harzianum isolate. Diffuse rings of conidia were produced in dark-grown cultures and, in cultures exposed to light for 15 min at 48 h, the rings were clearly defined. These results show that conidiation is under the control of an endogenous rhythm in T. harzianum and represent the first report of circadian conidiation in a wild-type Trichoderma. A Free-Running Rhythm (FRR) assay was used to investigate rhythmic gene expression in T. atroviride IMI206040 and a mutant derivative, in which the wc-2 orthologue, blr-2, was disrupted. Over a 3 d period, expression of gpd, which encodes the glycolytic enzyme glyceraldehyde-3-phosphate dehydrogenase, oscillated with a period of about 48 h. In the Δblr-2 mutant, the gpd rhythm was absent. These results revealed that in T. atroviride, gpd expression is under the control of an endogenous clock and that clock-regulated expression of gpd is associated with a functional BLR complex. Using degenerate primers, a portion of frq, which encodes the N. crassa clock oscillator FREQUENCY, was isolated from T. atroviride and used to probe the FRR assay northern blots. No frq expression was detected at any time point, which suggests that the circadian clock in Trichoderma does not involve FREQUENCY. In a concurrent study, orthologues of rco-1 (rcoT) were isolated and sequenced from T. atroviride and T. hamatum using a combination of degenerate, inverse and specific PCR. RcoT is an orthologue of the yeast global co-repressor Tup1 and in the filamentous fungi, RcoT orthologues have been demonstrated to negatively regulate conidiation. Genomic analysis of all available rcoT orthologues revealed the conservation of erg3, a major ergosterol biosynthesis gene, upstream from rcoT in ascomycetous filamentous fungi, but not in the ascomycetous yeast or in the basidiomycetes. These studies have significantly contributed to our understanding of the regulatory factors controlling conidiation in Trichoderma and have multiple implications for Trichoderma biocontrol; most notable the promotion of conidiation by primary nitrogen and low pH. Incubation conditions can be altered to suit the nitrogen and pH preferences of a biocontrol strain in order to promote cost effective conidial production, however this is not easily achieved in the soil, where the biocontrol strain must perform in a highly buffered environment optimised for plant growth. Successful use of Trichoderma biocontrol strains may involve the screening and targeting of strains to the appropriate pH conditions or the selection of new strains on the basis of capacity to perform under a given range of conditions.

Etude sur la biosynthèse de Naphtoquinones végétales et bactériennes

Les quinones sont des composés largement répandus dans la nature, aussi bien dans le règne végétal, qu'animal ou microbien. Ces molécules sont biosynthétisées à partir d'un petit nombre d'intermédiaires simples. Le noyau des naphtoquinones, en particulier, peut être synthétisé à partir de quatre précurseurs principaux l'acétate, la toluhydroquinone, le p-hydroxybenzoate et le shikimate. L'objet du présent travail est l'étude de la biosynthèse des naphtoquinones végétales ou bactériennes dérivant de l'acide shikimique (acide trihydroxy-3,4,5 cyclohexène-1 carboxylique), et la recherche des intermédiaires de cette voie de biosynthèse. Etant donné, à l'origine, l'absence quasi-totale de résultats expérimentaux dans ce domaine et le développement important qu'il a acquis en même temps que nous commencions notre travail, celui-ci a été constamment lié dans son développement aux données les plus récentes parues dans la littérature. Quelques études antérieures avaient montré que l'acide shikimique devait être le précurseur commun des acides aminés aromatiques et de plusieurs facteurs de croissance aromatiques (acide dihydroxy-2,3 benzoique, dihydroxy-3,4 benzoique, p-aminobenzoique et p-hydroxybenzoique) ainsi que de la ménaquinone de Escherichia coli (16,17). Lorsque nous commençons ce travail, on sait seulement que l'acide shikimique est incorporé in toto dans les ménaquinones bactériennes, de telle sorte que son carboxyle devient un des carbonyles de la quinone. Trois questions se posent alors: - Sur lequel des carbones 2 ou 6 adjacents au carboxyle, le cycle naphtoquinonique se ferme-t-il ? - Quelle est la molécule qui fournit les trois carbones nécessaires à la fermeture du cycle quinonique de la naphtoquinone, et quel est le mécanisme de cette fermeture ? - Le cycle naphtoquinonique ainsi formé est-il incorporé de façon symétrique ou non dans les ménaquinones, c'est-à-dire le carboxyle de l'acide shikimique est-il incorporé indifféremment dans les deux carbonyles quinoniques, ou sélectivement dans l'un des deux ? Pour résoudre la première question, LEISTNER et ZENK d'une part et M. LEDUC dans notre laboratoire d'autre part, ont utilisé l'acide shikimique [14 C-1,6]. Cette méthode nécessite une dégradation laborieuse des quinones formées. Nous avons, quant à nous, synthétisé un acide shikîmique [3H-3] marqué au tritium en une position spécifique, ce qui permet une dégradation simple et univoque. Une réponse partielle à la deuxième question a été donnée par CAMPBELL. Il montrait, en 1969, que le glutamate est capable d'apporter ses carbones 2, 3 et 4 pour la fermeture du cycle naphtoquinonique, mais il ne disposait pas alors des résultats de notre laboratoire lui permettant d'induire correctement le mécanisme de cette biosynthèse. Nous avons pu postuler la formation d'un intermédiaire, I'acide ortho-succinoylbenzoique OBS, dont nous avons pu prouver l'existence par synthèse et incorporation dans les quinones étudiées . L'existence de cet intermédiaire et la possibilité que nous avons eue de réaliser sur l'OSB différents marquages spécifiques nous ont permis de répondre à la troisième question, à savoir, l'incorporation orientée ou non de l'OSB dans les quinones. En même temps, différentes équipes montraient que d'autres quinones existant dans les végétaux telles que la lawsone, la juglone et différentes anthraquinones dérivaient aussi de l'acide shikimique; il nous a semblé intéressant d'appliquer nos techniques à l'étude très voisine de la biosynthèse de ces quinones. Pour la clarté de l'exposé, nous présentons dans le premier chapître les méthodes de synthèse des précurseurs radioactifs, dans le deuxième, les méthodes d'isolement et de dégradation des quinones. Les trois chapîtres suivants rapportent l'incorporation dans les ménaquinones, les naphtoquinones végétales et les anthraquinones végétales. Dans le sixième chapître, nous discutons nos résultats et proposons un schéma général de biosynthèse des quinones dérivant de l'acide shikimique. La partie expérimentale de ce travail est rassemblée après l'exposé théorique.

Etude chimique et caractérisation de principes colorants historiquement employés dans l'impression des indiennes en Provence

Ces travaux portent principalement sur l'étude de trois plantes tinctoriales : la garance, le nerprun et la gaude. Ces végétaux ont fait l'objet de nombreuses cultures en Provence et constituaient la principale matière première en colorants rouges et jaunes pour les teinturiers et les artistes. Une optimisation des conditions d'extraction des colorants de la garance assistée par ultrasons a été effectuée en utilisant un modèle statistique. Ce procédé d'extraction simple, rapide et efficace, a été comparé à deux autres techniques utilisées conventionnellement. Une étude cytohistologique des racines de garance a permis d'examiner les effets apportés par les différents procédés d'extraction. Les cellules traduisent après extraction par ultrasons, de profondes déstructurations fournissant une explication au plus important rendement en colorant extraits en comparaison aux extractions classiques. Une étude fondamentale sur l'identification des colorants extraits à partir des fruits immatures d'espèces appartenant au genre Rhamnus a été effectuée. Une approche chromatographique utilisant la CLHP/UVVisible/ SM a permis d'identifier la partie flavonol. Elle présente principalement des composés glycosylés dont la partie rhamninoside est liée sur le flavonol en position 3 ou 4'. Des flavonols 3-O-acétyl-rhamninoside ont également été caractérisés et sont spécifiques de Rh. saxatilis. Les fruits matures renferment aussi des anthraquinones qui ont été séparées des flavonols et concentrées en utilisant l'Extraction sur Phase Solide (SPE). Après analyse par RMN, des dérivés rhamnoside et arabinoside acétylés de l'émodine, jamais décrits dans la littérature, ont été identifiés dont le 6-O-(3',4' diacétyl)-arabinopyranoside d'émodine et 6-O-(2',3',4'-triacétyl)- arabinopyranoside d'émodine présents seulement dans Rh. alaternus. Les colorants jaunes de la gaude (Reseda luteola) ont été analysés par électrophorèse capillaire. En comparaison avec la CLHP, un gain important de la durée d'analyse a été observé tout en conservant une séparation convenable. L'ensemble de ces résultats expérimentaux a pu être appliqué avec succès à l'étude de colorants extraits à partir d'objets et d'échantillons historiques provenant de collections muséales et comprenant notamment des indiennes du XIXème siècle. Enfin, des essais de teintures ont été réalisés, en collaboration avec la société Les Olivades dans le but de développer une gamme de tissus à base de colorants naturels

No / Novel 1- and 1,4-substituted chloroethylaminoanthraquinones with DNA binding and alkylating properties along with their respective hydroxyethylaminoanthraquinone intermediates were synthesized. Selected chloroethylaminoanthraquinones were shown to cross-link DNA and alkylate guanines (at low nM concentration) with a preference for reaction sites containing 5'-PyG. A compound (Alchemix) with the bis-chloroethyl functionality confined to one side chain alkylated but did not cross-link DNA. All the 1,4-disubstituted chloroethylaminoanthraquinones were potently cytotoxic (nM IC50s) against cisplatin-resistant ovarian cancer cell lines.

Zusammenfassung In der vorliegenden Promotionsschrift werden Strategien zur Erzeugung von Linkermolekülen beschrieben, die geeignet sind, um als Bauelemente für sogenannte \"Metal-Organic rameworks\" (MOF) eingesetzt zu werden. Diese Stoffe bestehen aus einem organischen \"Linker\"-Teil, welcher die Metalleinheiten (\"Sekundäre Baueinheit\", SBU) miteinander verbindet, und dadurch die Entstehung von dreidimensionalen porösen Netzwerken ermöglicht. Dieses Projekt wurde an der Universität Leipzig in der Arbeitsgruppe von Prof. Dr. rer. nat. Dr. h.c. Evamarie Hey-Hawkins am Institut für Anorganische Chemie durchgeführt. Seit den frühen neunziger Jahren genießen MOFs großes Interesse aufgrund ihrer besonderen Eigenschaften als Festkörper mit wohldefinierter und permanenter Porosität. In der Einleitung dieser Promotionsschrift werden das Anwendungspotential oder bereits existierende Anwendungen für MOFs und verwandte poröse Feststoffe näher beschrieben. Mögliche Anwendungsgebiete sind unter anderem Gasspeicherung, Trennverfahren, zur Katalyse, in der Medizin und für Detektoren in der analytischen Chemie. Die Auswahl der organischen Baueinheiten für die Synthese von MOFs richtet sich nach Kriterien wie z.B. thermischer Stabilität, Geometrie der koordinierenden Einheiten, Rigidität des Linkermoleküls. Weiterhin können potentielle sekundäre Effekte berücksichtigt werden, z.B. Eignung für katalytische Prozesse, Ladungspufferung oder Lichtsammelvermögen. Alle Linker enthalten aromatische Einheiten; es werden Anthracen, Binaphthyl (verkörpert durch chirale BINAPO-Einheiten), Biphenyl, Pyren und Perylen als Mittelstück des Linkers eingesetzt. Die Promotionsschrift beleuchtet auch in einem Streifblick die Geschichte, das Vorkommen, mögliche Anwendungen und Giftigkeit von Molekülen mit polycyclischen aromatischen Kohlenwasserstoffgruppen. Die Synthese der Linker wurde mittels der folgenden Route realisiert: Bromierung der aromatischen Bestandteile und Anfügen von Endstücken (\"capper\") - welche die koordinierenden Einheiten enthalten - durch die Suzuki-Miyaura-Kupplung. Die Syntheserouten für den bromierten aromatischen Mittelteil des Linkers, die Boronsäuren und die vollständigen Linker, werden im experimentellen Teil beschrieben, und die erhaltenen Linker werden charakterisiert. Für den anorganischen Teil des MOF wurden außerdem ausgewählte komplexe Einheiten erzeugt, die als Startmaterialien für den sogenannten \"controlled SBU approach\" (CSA) dienen. Ein Ausblick auf weiterführende Untersuchungen an diesen Verbindungen wird ebenfalls gegeben. / Abstract This thesis describes approaches to obtain linker molecules that are suitable to build a so-called \"metal-organic framework\", MOF (or porous coordination polymer, PCP). These materials consist of an organic linker part that connects the metal units, \"secondary building units\" (SBU), with each other to form threedimensional, hollow networks. This project was performed at the University of Leipzig in the multinational workgroup of Prof. Dr. rer. nat. Dr. h.c. Evamarie Hey-Hawkins, at the Faculty for Inorganic Chemistry. These MOF materials have gained a lot of interest since the early 1990s for their special properties as solid materials with a well-defined, permanent porosity. The introductory part of this thesis elaborates potential or already current usage of MOF materials and related porous solids, such as gas storage, separation processes, catalysis, medical use and use in analytic detection. The choice of organic building blocks for the synthesis of MOFs followed certain criteria, such as thermal stability, geometry of coordinating units, rigidity of the linker molecule, and potential secondary effects such as suitability for catalytic purposes, charge buffering or light harvesting. All linkers feature aromatic units, and among the cores are moieties like anthracene, binaphthyl (represented by the chiral BINAP), biphenyl, pyrene and perylene. A glance is thrown on the history, occurrence, possible use and potential toxicity of molecules with polycyclic aromatic hydrocarbon moieties. Synthesis of the linkers is implemented by use of brominations of the aromatic core units and attaching capper units - carrying the coordinating units - by means of Suzuki-Miyaura-coupling. The synthesis routes to these molecules, brominated aromatic core, boronic acids and finished linkers, are described in the experimental part. The obtained linkers are characterised and described. Furthermore, selected complex units for the inorganic part of a potential MOF were created as starting materials for a so-called \"controlled SBU approach\" (CSA); wherein a building block, very similar to the desired unit in the final framework, is prepared and then used as a precursor in the synthesis. Various attempts at the generation of MOFs were conducted: Slow diffusion in liquids or gels, and, especially, reactions by tempering the organic and inorganic building blocks or starting materials in a solvent - partially also under solvothermal conditions. Several polymers were obtained in crystalline form, suitable for structure determination by X-ray crystallography. These structures are presented and discussed in this work, as well as the further research on these solids and the results being achieved.
