Substratbindung und -freigabe während des Katalysezyklus eines biotinspezifischen ECF-Transporters

ECF (Energy-Coupling Factor)-Transporter sind prokaryotische Aufnahmesysteme für Mikronährstoffe, die eine spezielle Gruppe von Transportern mit ATP-Bindekassette (ABC) darstellen. Sie beinhalten zwei asymmetrische Membranproteine, von denen eins (S) für die spezifische Bindung und Translokation des Substrates und das andere (T) für die Kopplung mit den ATPasen (A1,A2) zuständig ist. Bei ECF-Transportern der Subklasse I bilden diese Komponenten eine Einheit, während bei Vertretern der Subklasse II ein AAT-Modul mit wechselnden S-Einheiten interagiert. In der vorliegenden Arbeit wurde der Transportmechanismus, der eine Drehung der kompletten S-Einheit in der Membran beinhaltet, anhand des Biotintransporters BioMNY erstmals experimentell validiert. Durch Rekonstitution in Lipid-Nanodiscs, chemische Quervernetzung, fluoreszenz- und ESR-spektroskopische Techniken sowie einen Bindungstest mit radioaktivem Biotin wurde gezeigt, dass (i) die ATP-Bindung an die ATPasen zu einer Aufrichtung der S-Einheit (BioY) führt, (ii) diese Bewegung die Substratbeladung ermöglicht und (iii) BioY dabei ununterbrochen mit der T-Einheit (BioN) interagiert. Dies stellt einen Gegensatz zu Systemen der Subklasse II dar, für die ein ATP-abhängiger Austausch von S-Einheiten im Transportzyklus gezeigt worden war. Darüber hinaus wurde ein Escherichia coli-Stamm konstruiert, der durch Blockierung seines hochaffinen Biotintransporters und des -synthesewegs auf Spuren von Biotin nicht wachsen kann. Dieser Stamm ermöglichte einen eindeutigen Nachweis der Transportaktivität einiger solitärer BioY-Proteine. Aufgrund der einheitlichen Topologie von S-Einheiten ist ein Kippen auch für solitäre BioY-Varianten wahrscheinlich. Auch die metallspezifischen S-Einheiten CbiM und NikM besitzen ohne AAT-Modul eine basale Co2+- bzw. Ni2+-Transportaktivität. Ein ESR-spektroskopischer Kobaltnachweises zeigte, dass die aus nur zwei Membranhelices bestehende CbiN-Einheit für die Metallbeladung von CbiM essentiell ist. / ECF (Energy-Coupling Factor) transporters are a subgroup of ABC transporters that mediate uptake of micronutrients into prokaryotic cells. In contrast to canonical ABC importers, ECF transporters comprise two unrelated membrane proteins, one of which is responsible for specific and high affinity substrate binding (S) and the other one constitutes the coupling component (T) between S and the cytosolic ABC-ATPases (A1,A2). Subclass I transporters consist of four dedicated components whereas in subclass II transporters, a central AAT-module may interact with various S units. The biotin specific subclass I ECF transporter BioMNY was used to experimentally verify the hitherto hypothetic transport mechanism, which involves a rotation of the S unit within the membrane. With a series of experiments including reconstitution of BioMNY into lipid nanodiscs, site-specific cross-linking, a substrate binding assay with radioactive biotin and both fluorescence and EPR spectroscopic techniques, the ATP-dependent rotation of BioY (S) as a prerequisite for substrate binding and release was shown for the first time for an ECF transporter. Unlike subclass II transporters, for which an ATP-dependent release of the S unit was proposed, BioY interacts continuously with BioN (T) during the transport cycle. In a second focus of the work, an Escherichia coli reporter strain for biotin transporters was constructed. Due to inactivation of both biotin synthesis and the intrinsic high affinity biotin transporter, this strain was not capable of growing on trace amounts of biotin. With the use of this strain, transport activity of recombinantly produced solitary BioY proteins that naturally lack other ECF components was evidenced. Transport activity in the absence of AAT modules is also a feature of the Co2+ and Ni2+ specific S components CbiM and NikM. An EPR spectroscopic Co2+ detection assay helped underscoring the essential role of the small membrane protein CbiN for Co2+ loading of CbiM.

A citricultura é de grande importância para as atividades agrícolas brasileiras, uma vez que o Brasil é o principal produtor e exportador de suco de laranja. O cancro cítrico, causado pela bactéria Xanthomonas axonopodis pv. citri (Xac) é um grave problema nesse setor, causando um elevado prejuízo na produção de frutos e seus derivados. O fator sigma é a subunidade da RNA polimerase que tem a função de direcionar o núcleo da RNA polimerase a uma classe específica de sequências promotoras. Como a maioria das bactérias sintetiza diversos fatores sigma, essa característica proporciona à bactéria a oportunidade de manutenção basal da sua expressão gênica, assim como, a regulação em resposta a alterações ambientais e a sinais durante o desenvolvimento bacteriano. O genoma de Xac codifica para 14 fatores sigma. Nesse presente trabalho, detectamos interações dos fatores σECF (Xac2814. Xac3989, Xac0922, Xac1319, Xac1380, Xac1682, Xac4129 e Xac2191) e seus fatores anti-σ cognatos (Xac2815. Xac3988, Xac0921, Xac1320, Xac1379, Xac1681, Xac4130 e Xac2192). Além disso, observamos interações entre o fator σFliA (Xac1933) e o anti-σFlgM (Xac1989), seu fator anti-σ cognato. A caracterização das cepas nocautes para alguns fatores σ apontaram o envolvimento do fator σ54Xac1969 no mecanismo de formação de flagelo, a contribuição do fator σECFXac1682 na resposta ao choque térmico e a participação do fator σECFXac2191 no crescimento bacteriano em condições de carência de ferro. / Citriculture is an important sector of the economy of the State of São Paulo. The sigma factor is the subunit of RNA polymerase that serves to direct the RNA polymerase core to a specific class of promoter sequences. Most bacteria code for more than one sigma factor, which provides the cell with the means by which to maintain basal gene expression while at the same time modulate the expression of specific genes in response in environmental changes and signals during bacterial growth. The Xac genome codes for 14 sigma factors which are the objects of study in this thesis. We demonstrate that many of the sigma factors of the σECF family (Xac2814, Xac3989, Xac0922, Xac1319, Xac1380, Xac1682, Xac4129 e Xac2191) interact with cognate anti- factors (Xac2815, Xac3988, Xac0921, Xac1320, Xac1379, Xac1681, Xac4130 e Xac2192). These sigma-anti-sigma pairs are all coded by neighboring genes. Interactions between the sigma factor σFliA (Xac1933) and anti-σFlgM (Xac1989) were also observed. Xac strains with gene knockouts for several sigma factors were produced. The characterization some these knockout strains point to the involvement of σ54Xac1969 in the biosynthesis of flagella, participation of σECFXac1682 in the ability to survive heat shock and involvement of σECFXac2191 in the response to iron deficiency.

CnrX est un senseur périplasmique, ancré à la membrane, appartenant au complexe CnrYXH qui contribue à réguler l'expression des gènes impliqués dans la résistance au nickel et au cobalt chez Cupriavidus metallidurans CH34. La résistance est induite par la libération de CnrH, un facteur sigma de type ECF (Extracytoplasmic Function), par le complexe CnrYX en réponse à Ni et Co. Nous avons cherché à comprendre la manière dont CnrXs, le domaine senseur de CnrX, détecte les ions métalliques, les stratégies utilisées pour sélectionner spécifiquement Ni ou Co ainsi que la nature du signal engendré par cette interaction. Les techniques spectroscopiques et biophysiques telles que l'UV-visible, la RPE, le XAS et l'ITC ont permis d'étudier les sites métalliques en solution. Le dimère de CnrXs possède quatre sites de liaison au cobalt. Deux des sites (sites F) sont retrouvés dans la protéine entière dont nous avons maintenant un excellent modèle avec le mutant CnrXs-H32A. Les deux autres sites (sites E) ont un signal spectroscopique atypique probablement dû à la formation d'un complexe binucléaire de cobalt. Nous présentons également des structures à haute résolution de CnrXs dans ses formes apo et métallées par le nickel, cobalt ou zinc. Nous avons établi que la forme zinc est la forme inactive de la protéine et que le mécanisme de détection est engendrée par la substitution du zinc par le nickel et le cobalt dans le site F, conduisant à une modification majeure du site de liaison au métal. Tandis que le zinc est pentacoordiné dans une sphère 3N2O, Ni et Co recrutent le soufre de la seule méthionine (Met123) comme sixième ligand pour former un site octaédrique. Nous suggérons que Met123 soit l'interrupteur moléculaire dont la liaison avec le métal fait évoluer la structure de la protéine vers une conformation active. A notre connaissance, ces résultats constituent la première étude structurale et spectroscopique d'un senseur de métal périplasmique impliqué dans un système de transduction du signal dépendant d'un facteur sigma de type ECF. / CnrX is the membrane-anchored periplasmic sensor of the CnrYXH complex that contributes to regulate the expression of the genes involved in cobalt and nickel resistance in Cupriavidus metallidurans CH34. This resistance is induced by the release of the ExtraCytoplasmic Function (ECF) sigma factor CnrH from the CnrYX complex upon sensing of Ni or Co. We addressed the metal sensing mechanisms of CnrXs, the strategies used to select Ni or Co and the nature of the signal onset. Biophysical and spectroscopic techniques allowed us to study the metal binding sites in solution. The CnrXs dimer contains four cobalt binding sites. Two (F sites) are present in the full-length protein which H32A-CnrXs mutant is an excellent model of. The two other sites have an unusual spectroscopic signal that might be due to the formation of a binuclear cobalt complex. We present also high-resolution structures of CnrXs in the apo, Ni-, Co-, and Zn-bound forms. We propose that Zn-bound CnrX typifies the resting state of the complex and that the sensing mechanism is triggered by the substitution of Zn for Ni or Co in the F site. This substitution leads to dramatic changes in the metal-binding site. While the Zn ion is pentacoordinated in a 3N2O sphere, Ni or Co ions recruit the thioether sulfur of the only methionine (Met123) residue as a sixth ligand to form an octahedral site. We propose that the Met123 side chain recruitment is the qualitative change that switches on the sensing mechanism by remodeling the four-helix bundle that accommodates the metal-binding site. To our knowledge these results represent the first structural and spectroscopic study of a periplasmic metal sensor involved in transmembrane signal transduction for the activation of an ECF-type sigma factor.

Perceptions of Swedish Fund Managers of Equity Crowdfunding

Equity crowdfunding (ECF) is the smallest of four main types of crowdfunding (CF) and has emerged as a new form of investment, where investors receive equity of a company in return for capital. Over the past years, the development of CF has given entrepreneurs the option to seek funding from a new source, instead of relying solely on venture capital (VC) and other sources of funding (Mollick, 2013). However, the implementation of regulations for ECF has not yet developed enough to make such investing equally consistent for everyone.   Equity crowdfunded ventures can potentially attract a large number of investors that can possibly create corporate governance issues between the entrepreneur and investors. Additionally, individual investors might both lack the competence or incentive to extensively research and assess a venture and make an investment. Due to the growth in ECF and the lack of knowledge of it, the purpose of this thesis was to discover the perceptions of Swedish fund managers (FMs) regarding the nature of ECF and their interest in it.   In order to fulfill the purpose of this study, previous studies were carefully examined to gain a thorough understanding of the area of study. Furthermore, the empirical findings were gathered through a course of seven semi-structured interviews of Swedish FMs.   Empirical findings suggested that ECF has remained as a means of providing only seed-stage funding for ventures and that it is a marginal phenomenon in Sweden. ECF was believed to potentially disturb the VC industry if it grew and moved to a later stage. Additionally, ECF was found to be a good option for companies that were unable to receive funding from traditional funding methods. Risks of fraud and other scandals should be prevented for ECF to maintain its popularity. The findings implied that the future of ECF remains uncertain. Based on the perceptions of Swedish FMs, ECF neither would replace nor be a threat to traditional methods.

Salsberry / This dissertation examines an understudied group according the American Council on Education: the tenure-track early career faculty (ECF). The focus is on the culturalization, socialization, academic culture, and emergent themes discerned from ten semi-structured interviews with tenure-track ECF. This qualitative bounded system case study was conducted in the context of a Midwestern Carnegie I Research Land-Grant Institution, an institution with emphasis on teaching, research, and service. Ten semi-structured interviews were conducted and transcribed. From these, codes, categories, patterns, and themes were found. Additional documentation was also considered such as participants’ resumes and the faculty handbook, COE: Orientation to COE A Guide for Faculty 2008-2009. The themes included a reliance on socialization from the parent degree granting institution, and an overall request for balance between professional and personal life. In addition the ECFs need mentoring at their new institution in research, writing and publication, in professional identity, and socialization to tacit knowledge via cognitive apprenticeship. A recommendation is that there be a bi-directional conversation on socialization, rather than a top down approach.
