An Evaluation Approach for Regional Logistics Abilities

The continued growth of regions in a state of economic expansion is often stunted by their inability to assess, maintain and expand logistics infrastructure. The need for a comprehensive, detailed and intuitively understandable logistics capability model to aid in the assessment phase of logistics maintenance and expansion provides motivation for this thesis. Logistics benchmarking can also be presented to attract prospective clients and provide basis for further development. Through the process of extensive literature review, we perform objective evaluations and provide a single recommendation for regional logistics capability modeling within a limited research scope. We recommend a 12 category logistic evaluator model with an interview process and expert scoring. The scoring is performed on the basis of logistics data and statistics. An implementation of the model is performed on Gävle, Sweden and is partially validated with a case study on GEVALIA Coffee. Implementations on SWOT and a discussion of the similarities of the 12 category logistics evaluator model and AHP will be given.

Developing Tools for Introducing Modifications into the Chicken Genome

The chicken is a classic model organism that has provided key insights into embryonic development. Chicken embryos can be directly manipulated and observed during development while retaining the potential to reach adulthood. Despite this benefit, the utility of the chicken in studying development has been limited by the difficulty of introducing genetic changes to the genome. The recent development of cell culture conditions for chicken primordial germ cells (cPGC) has made it feasible to produce transgenic chickens, but there is still a lack of tools for introducing genetic modifications into cPGCs. Recombinase Mediated Cassette Exchange (RMCE) is a technique that has been utilized in traditional genetic systems to generate multiple alleles at a given locus but has not yet been adapted to the chicken. In order to use RMCE in the chicken, we inserted Lox sites into cPGC using CRISPR/Cas9. We targeted the ovalbumin locus and potential genomic safe harbor sites (GSH) identified using genomic data. We performed RMCE to exchange green fluorescent protein (GFP) into these loci. We observed RMCE efficiency as less than 1% at each loci. We then designed a system using a drug inducible Caspase 9 (iCasp9) to select for cells that underwent cassette exchange. This method enabled us to obtain a population of 100% edited cells. We anticipate that this tool will increase the utility of the chicken as a model organism, livestock, and bioreactor.

A dissertação de mestrado apresenta as metodologias utilizadas com a integração de sistemas dos equipamentos de campo ao Centro de Controle, Comunicação, Operações e Segurança - CCCOS, visto ser o concentrador que recebe, gerencia, armazena e envia todas as informações dos equipamentos, seja do sistema de controle de acesso, através dos dados enviados e recebidos pelas placas de comunicação das leitoras das catracas, cancelas, torniquetes, sensores, sistema OCR, leitura automática de placas, contêineres, biometria facial, etc, ou do sistema de CFTV, através das imagens enviadas pelas câmeras fixas e móveis, via encoders e switches ou do sistema de comunicação VOIP, rádio ou telefonia. O sistema de segurança, preferencialmente, deverá estar com todos os subsistemas integrados e, na medida do possível, com um sistema de acesso especial e restrito para entrada de pessoas não autorizadas na área do centro de controle, com níveis de permissões em cada terminal de computador, acesso ou servidor, e neste trabalho, se descreve as tecnologias aplicadas dos equipamentos e dos subsistemas utilizados no sistema de segurança integrada e, como resultado, demonstra melhoria substancial da gestão, quando utilizados gerenciadores integrados, visto que o processo de entrada e saída para atender o sistema de segurança ou ISPS Code é otimizado e auxilia na gestão operacional e de segurança dos terminais e portos. Como proposta de customização, se terá vários Centros de Controles integrados, em portos públicos e privados de cada unidade da federação, com política de segurança criptografada, com confidencialidade, integridade e disponibilidade, tornando possível integrar todas as autoridades portuárias, alfandegárias e demais autoridades federais, estaduais e municipais, respeitando suas atribuições legais, em um único centro de controle global virtual, que permitirá inclusive se comunicar on line com demais sistemas portuários do mundo, que tenha o mesmo sistema de segurança aplicado. O Porto de Santos está localizado no litoral Paulista, na costa Sudeste do Brasil e é o maior e mais importante porto do país. Desde 1859 vem sofrendo adaptações e modernizações tanto em suas instalações físicas quanto em sua estrutura administrativa. As variáveis com as quais a Engenharia Civil deve lidar podem ser resumidas basicamente aos fenômenos naturais, pois estes determinam como serão projetadas e construídas as estruturas e instalações. Em projetos portuários, diversas características naturais devem ser mensuradas como, por exemplo, ventos, correntes marítimas, incidência de ondas, tipo de solo em que se encontra o porto, as temperaturas médias do local e, sobretudo, o nível do mar. Os mares e oceanos não possuem superfícies estáveis e apresentam cotas que estão em movimentos constantes. Estas variações se dão basicamente por ondas de curto ou longo período. A maré é uma onda de longo período que causa a variação diária do nível do mar. Atualmente é cada vez maior o interesse da comunidade científica pela temática relacionada às variações do nível do mar. Buscando contribuir com o tema, o presente estudo tem como objetivo principal a análise das variações do nível do mar no Porto de Santos ao longo dos últimos 60 anos, utilizando-se de dados coletados nos três marégrafos da região. Foi elaborada a média móvel desta série com a finalidade de detectar alguma variação significativa. A análise dos dados implicou que fossem considerados períodos de 18,61 anos, correspondentes ao intervalo do ciclo de precessão lunar, de modo a eliminar o efeito da variação astronômica no nível do mar. A elevação do nível máximo, médio e mínimo do mar no período de 1953 a 2008, apresentou uma elevação média de 2,5 mm/ano, resultado semelhante a outros estudos. Foram avaliados também o impacto da variação do nível do mar sobre as estruturas portuárias, indicando os efeitos da elevação nas estruturas, nos cais, nos aparelhos de defensa e ancoragem, nos equipamentos de descarga e outros. / Santos Harbor is at São Paulo\'s coast, in Brazil\'s southeast coast and it is the biggest and the most important port of the country. Since 1859 had been suffering adaptations and modernizations in its installations such as in its administrative structure. The variables that Civil Engineering has to deal with can be resumed basically to the natural phenomena because its determines how projected and built the structures will be. In docks projects, several natural characteristics must be measured, as example, winds, ocean currents, waves incidence, kind of soil that supports the harbor\'s structures, the local medium temperatures and, mainly the sea level. The seas and oceans doesn\'t have stables surfaces and shows levels that are in constant movements. These variations are products of short and long period waves. The tide is a long period wave that causes the daily sea level variation. Actually, the interest of the scientific community in sea level variation is increasing. Aiming to contribute with this theme, the present study has as main objective analyzing the sea level variations at Santos Harbor above the last 60 years, using data collected from three tidal gauges in the region. It was applied the moving average of the series to detect some significant variation. The data analysis demanded that the periods of 18,61 years, which corresponds to the interval of the moon precession cycles, was considered to purge the astronomic variation over the sea level. The maximum sea level, average and minimum sea level, in the period of 1953 and 2008 showed an average increase of 2.5 mm/year, which is similar to other studies. It was evaluated the impact of this sea level rise over the port\'s structures, showing the structural effects, over the docks; in the defense and anchorage equipment, unload equipment and others.

Similarities among harbor developments : An analysis of case-studies in Hamburg, Dublin, Copenhagen and Stockholm

Harbor developments have been realized in several European cities over the last years, becoming an important topic in the field of urban and regional planning. Despite their presence in innumerous sites with unlike contexts and backgrounds, those developments look pretty similar in terms of design and form. Are we witnessing the born of a generic and similar city? The aim of this paper is to investigate how similar are these areas and their positive or negatives effects upon the existing cities. Through a review on the main urban theories of the twentieth century it is possible to identify what are the contemporaneous trends in urban planning and how they have been employed in harbor developments. Four case studies were selected to illustrate these similarities, two almost completed: Dublin Docklands and Hamburg HafenCity, and two newly ones: Stockholm Norra Djurgårdsstaden and Copenhagen Nordhavnen. The findings indicated the case-studies share several similarities in terms of discourse, implementation strategy, sustainability, publicity and as well social and income inequalities. Contemporaneous urban discourse preaches for diversity which it is still far to be reached by the case-studies
