Thesis (MPhil)--Stellenbosch University, 2005. / ENGLISH ABSTRACT: Two literary conversion narratives with much historical detail, are compared in this thesis: the
Confessiones written by the renowned fourth century church-father, St. Augustine, and
Surprised by Joy written by the twentieth century writer and scholar, C.S. Lewis. In order to
understand St. Augustine's conversion to the Christian faith, Christian religion as a social
phenomenon in the Ancient World is discussed. As background for the discussion and
comparison of the two conversion narratives, a brief biography is given of St. Augustine and
of Lewis, as well as a description of each one's course of conversion. The research is
structured In terms of beacons that St. Augustine identified during the course of his
conversion: people who played a significant role, events that influenced his life, and inner
conflict that spurred him on his way. By means of an analysis regarding theme and content, it
is shown that there are clear similarities between the beacons identified by Augustine and
Lewis in their conversion narratives. / AFRIKAANSE OPSOMMING: In hierdie tesis word twee literêre bekeringsverhale waarin die historiese werklikheid deur die
skrywers weergegee word, met mekaar vergelyk: die Confessiones van die vierde eeuse

Lewis. Om Augustinus se bekeringsverhaal histories te kan plaas teen die agtergrond waarin
hy geleef en tot bekering gekom het, word 'n uiteensetting gegee van die Christelike
godsdiens as 'n sosiale fenomeen in die Antieke Wêreld. 'n Kort lewensbeskrywing van
Augustinus en Lewis en 'n oorsig van die weg wat elkeen se bekering gevolg het, dien as
agtergrond vir die bespreking en vergelyking van die twee bekeringsverhale. Die navorsing
word gestruktureer aan die hand van bakens wat Augustinus op sy bekeringsweg uitgelig het:
persone wat 'n beduidende rol gespeel het, gebeure wat hom beïnvloed het, innerlike konflik
wat hom voortgedryf het. Die tesis toon deur 'n analise op grond van inhoud en tematiek aan
dat daar duidelike ooreenkomste is tussen die bakens op Augustinus se bekeringsweg en dié

Synthèse d'une librairie d'analogues monomériques et dimériques du sLe XCalosso, Mickael 09 1900 (has links)
Dans cet ouvrage sera décrite la synthèse de nouveaux analogues du sialyl
Lewis X (sLex). A cet effet, nous avons préparé une librairie d’analogues synthétisée
à partir d’une approche mettant en jeu un «espaceur» acyclique permettant d’avoir un
biais conformationnel que nous avons défini comme la stratégie ATC-B.
Nous avions déjà démontré que certains analogues portant un groupe benzoate
en C-2 et en C-4 du galactose présentent une activité 50 fois supérieure à celle du
sLex. Nous avions par ailleurs démontré qu’en l’absence du benzoate en C-2,
l’activité devient alors trois fois plus faible. A présent, il paraissait interessant de
synthétiser des analogues ayant seulement un groupe benzoate en C-4 pour evaluer
l’impact de ce groupement sur la puissance de nos analogues.
Par le passé, nous avions également mis en évidence le rôle des esters sur
l’activité des analogues portant un «espaceur» acyclique dans le cadre de la stratégie
ATC-B. Nous effectuerons donc des variations à ce niveau pour en évaluer l’impact.
Enfin, nous avons préparé une nouvelle famille d’analogues de type dimère.
Ceux-ci seront constitués de 2 unités des composés monomériques synthétisés
précédemment. La synthèse de ces dimères fera l’emploi de la «Click Chemistry».
Cette étude nous mènera a vous présenter la synthèse de ces composés et la
méthodologie employée. / This work describes the synthesis of novel sialyl Lewis X (sLex) analogues.
To this end, we have prepared a library of analogues by implementing a strategy that
makes use of an Acyclic Tether which allows for defined Conformational Bias (ATCB
strategy).
We have previously shown that analogues bearing a benzoate group at both
positions C-2 and C-4 of the galactose exert an activity 50-fold greater than that of
sLex. Indeed, removing the benzoate at position C-4 while keeping the one at C-2 led
to a three fold decrease in potency. We are currently preparing the corresponding
analogues that have the benzoate only at position C-4 in order to fully evaluate its
impact on the potency of the analogue.
We have also previously elucidated the role of esters on the activity of
analogues that have acyclic tether as part of the ATC-B strategy. Variations have
been made at the level of the ester and their impact has been evaluated.
Lastly, we have prepared a novel family of analogues consisting of two
identical monomeric subunits linked together. Synthesis of these dimers was
performed via «Click Chemistry». This study describes the synthesis of the
compounds and the methodology employed.

Addition stéréosélective de nucléophiles sur un centre acétal : synthèse de nucléosides 1’,2’-cisSt-Jean, Olivier 04 1900 (has links)
Plusieurs analogues de nucléosides thérapeutiques (Ara-C, Clofarabine), utilisés
pour le traitement de leucémies, présentent un arrangement 1’,2’-cis entre la nucléobase
reliée au centre anomère et le substituant (électroattracteur) en C-2’. Récemment, notre
laboratoire a développé une approche synthétique pour former sélectivement des
analogues de nucléosides et de thionucléosides 1’,2’-trans et 1’,2’-cis à partir de
précurseurs acycliques.
Ce mémoire présente une nouvelle méthodologie pour accéder efficacement aux
analogues de nucléosides 1’,2’-cis à partir de furanosides. Différents groupements en
position anomérique ont été examinés, sous conditions cinétiques en utilisant le bromure
de diméthylbore pour générer sélectivement des produits acycliques ou cycliques. Les
intermédiaires cinétiques de différents furanosides de méthyle formés en présence de
Me2BBr ont été piégés in situ par un thiol pour générer des thioacétals acycliques avec
de bonnes voire d’excellentes diastéréosélectivités. Les produits générés sont en accord
avec une rétention globale de l’information stéréochimique du centre acétal et deux
déplacements SN2 consécutifs ont été suggérés pour rationaliser ces résultats. Toutefois,
l’objectif de synthétiser des analogues de nucléosides à partir de furanosides de méthyle
a échoué.
Tel que démontré par le Dr Michel Prévost, l’activation par Me2BBr des lactols
des quatres différents furanosides suivie d’une addition in situ d’une base silylée a
permis de former diastéréosélectivement les analogues de nucléosides 1’,2’-cis
correspondants avec d’excellents rendements. Nous avons démontré que d’autres
substrats peuvent être employés et que l’induction stéréochimique est sous contrôle du
substituant électroattracteur en C-2. D’autres acides de Lewis, tel que TMSBr, peuvent
également être utilisés.
Cette méthodologie a également été étendue à d’autres nucléophiles tels que des
Grignards ou des éthers d’énols silylés, conduisant à de bonnes sélectivités. / Many therapeutically relevant nucleoside analogs (Ara-C, Clofarabine) for the
treatment of leukemia have a 1’,2’-cis arrangement between the nucleobase attached at
the anomeric center and the non-hydrogen substituent at C-2’. Recently, our laboratory
has developed a versatile approach to the synthesis of 1’,2’-trans and 1’,2’-cis
nucleoside and thionucleoside analogues from acyclic scaffolds.
This work will present a new methodology to access efficiently 1’,2’-cis
nucleoside analogues from cyclic furanoside. Activation of various anomeric groups by
Me2BBr was investigated, and under kinetic control acyclic substrates or cyclic ones
could be generated selectively. Trapping the kinetic product of methyl furanoside
formed in presence of Me2BBr by thiol in the presence of base led to the formation of
acyclic thioacetal in good to excellent diastereoselectivity. The results obtained are in
accordance with total retention of the stereochemical information of the acetal moiety
and thus suggested that the mechanism of these two reactions is two successive SN2
displacements. The objective of synthesizing nucleoside analogs from methyl
furanoside was unsuccessful.
As shown recently by Dr Michel Prévost, activation of all four furanoside lactol
scaffolds by Me2BBr with an in situ addition of silylated nucleobase afforded 1’,2’-cis
pyrimidine nucleoside analogues in very good yields and with diastereoselectivities
greater or equal to 20:1. Expending this methodology to other scaffolds provided
evidence of stereoelectronic control of the C-2 electron-withdrawing substituent. Other
Lewis acids such as TMSBr can be used.
This methodology was also applied to other nucleophiles such as allyl Grignard
and silylated enols ethers, which were successfully alkylated in good yield and 1,2-cis
diastereoselectivity.

Étude de l’issue diastéréomérique impliquant la réduction radicalaire d’α-bromoesters adjacents à un tétrahydropyrane substituéViens, Frédérick 09 1900 (has links)
Cet ouvrage traite de la formation diastéréosélective de tétrahydropyranes 3,7-cis et 3,7-trans polysubstitués. Des méthodologies de cycloétherification et de C-glycosidation en tandem avec une réduction radicalaire y sont décrites (pyranes 3,7-
trans) tandis qu’une haloéthérification en tandem avec le même processus radicalaire
conduit aux pyranes 3,7-cis.
Suite à des travaux antérieurs du laboratoire, des études ont été réalisées afin de comprendre l’influence de la stéréochimie des substituants en position C3, C6, C7 et C8 sur l’issue diastéréochimique lors d’un transfert d’hydrure sous contrôle exocyclique sur un centre radicalaire adjacent à un tétrahydropyrane (C2). Ces études ont permis de solutionner les problèmes de diastéréosélectivité rencontrés lors des réductions radicalaires des centres en C2 de divers fragments élaborés d’ionophores (e.g. zincophorine, salinomycine, narasine…) réalisées par notre groupe. Des études conformationnelles réalisées sur des pyranes di- ou tri-substitués ont permis de comprendre les diastéréosélectivités notées lors du processus radicalaire en fonction des stéréochimies relatives des centres précédemment cités. En particulier, l’utilisation d’un bicycle rigide (trans-octahydrochromène) a permis de montrer l’importance du positionnement spatial (axial ou équatorial) de la chaîne portant le
centre radicalaire.
Par la suite, nous avons pu mettre en évidence une amplification des ratios en faveur du produit de réduction radicalaire 2,3-anti lorsque la réaction est réalisée en
présence d’un acide de Lewis monodentate encombré (MAD).
L’optimisation du contrôle endocyclique lors du transfert d’hydrure a permis de générer la stéréochimie complémentaire 2,3-syn. L’utilisation du TTMSS comme source d’hydrure combinée à l’utilisation d’un acide de Lewis bidentate tel que MgBr2·OEt2 a permis l’obtention d’excellentes sélectivités en faveur du produit endocyclique. Des études RMN 13C ainsi que le titrage des ions Mg2+ en solution ont
été effectués afin de comprendre la nature des complexes impliqués.
Finalement, ces études ont permis la formation stéréocontrôlée de centres
stéréogéniques adjacents à un THP, motifs fréquement rencontrés dans certains
policétides. Elles permettent ainsi d’envisager la synthèse de polyéthers de type
ionophore et d’autres molécules d’intérêt biologique. / This work describes the diastereoselective formation of polysubstituted 3,7-cis
and 3,7-trans tetrahydropyrans. Tandem radical reduction/cycletherification and
tandem radical reduction/C-glycosidation methodologies are depicted for 3,7-trans
pyrans formation while tandem radical reduction/haloetherification lead to 3,7-cis
pyrans.
Following previous work in the laboratory, studies have been conducted to better understand the stereochemical influence that substituents in the C3, C6, C7 and C8 positions have on the diastereoselectivity of a hydride transfer radical reaction on a
radical center with an adjacent tetrahydropyran (exocyclic control). These studies have helped to solve diastereoselectivity problems encountered with C2 radical reductions in different ionophore fragments (e.g. zincophorin, salinomycin, narasin ...) that were
investigated by our group.
Conformational studies performed on di-or tri-substituted pyrans have helped to
understand the diastereoselectivities observed in the radical process of the
stereochemical centers mentioned above. In particular, the use of a bicycle frame
(trans-octahydrochromene) showed the importance of the spatial positioning (axial or equatorial) of the radical center chain.
Subsequently, we were able to demonstrate an increase ratios for the 2,3-anti radical reduction product when the reaction was performed in the presence of a bulky
monodentate Lewis acid (MAD).
Optimization of hydride transfer under endocyclic control has generated the
complementary 2,3-syn stereochemistry. Use of TTMSS as a hydride source combined
with the use of a bidentate Lewis acid such as MgBr2·OEt2 allowed for the formation of
endocyclic products with excellent selectivities. 13C NMR studies and titration of Mg2+ ions in solution were performed to understand the nature of the complexes involved.
Finally, these studies led to the stereocontrolled formation of stereogenic
centers adjacent to a THP, motifs frequently encountered in polyketides. This work
thus involves the synthesis of polyether ionophore-type fragments and other molecules
of biological interest.

Synthèse totale du méthyl ester de la zincophorine et étude DFT du transfert d’hydrogène diastéréosélectif impliquant des intermédiaires radicalaires vicinaux à un esterGodin, François 04 1900 (has links)
Cet ouvrage traite d’une méthodologie pour l’induction de centres stéréogènes au sein des motifs propionates par la chimie des radicaux et de son application à la synthèse totale du méthyl ester de la zincophorine. Cet ionophore, aux propriétés biologiques intéressantes, présente plusieurs défis synthétiques dont une séquence de type polypropionate anti, anti, anti, anti difficilement accessible ainsi qu’un tétrahydropyrane trans trisubstitué. Récemment, l’intérêt renouvelé pour ces composés polyéthers, en tant qu’agents anticancéreux, accentue l’importance de stratégies versatiles permettant l’accès à ces structures ainsi qu’à leurs analogues.
Depuis quelques années, notre groupe s’intéresse à la synthèse d’unités propionates acycliques par une séquence réactionnelle contrôlée uniquement par le substrat. La première étape découle d’une aldolisation de Mukaiyama entre un aldéhyde alpha-chiral et un énoxysilane tétrasubstitué portant une liaison carbone-halogène, et où l’issue stéréochimique dépend de la nature monodentate ou bidentate de l’acide de Lewis employé. La seconde réaction-clé implique la formation d’un radical tertiaire, vicinal à un ester, pouvant être réduit diastéréosélectivement en présence d’hydrure d’étain.
La première section décrit l’élaboration de motifs tétrahydropyranes trisubstitués et l’induction des centres stéréogènes vicinaux par une réduction radicalaire. Nous avons révélé que l’issue diastéréosélective de la réaction de cyclisation par une iodoéthérification était dictée par le groupement gamma-méthyle des esters alpha,beta-insaturés de départ. Nous avons ensuite démontré que les produits de la réaction radicalaire anti et syn pouvaient être obtenus sélectivement à partir d’un intermédiaire commun, respectivement en prenant appui sur l’effet exocyclique ou endocyclique lors de la réduction. Par une stratégie complémentaire, nous avons révélé que le précurseur radicalaire pouvait également être obtenu par une réaction de cycloéthérification en présence d’un énoxysilane tétrasubstitué. Une étude systématique des substituants du cycle a révélé que certaines relations stéréochimiques conduisaient à une perte de sélectivité au détriment du produit anti.
La seconde section concerne l’étude DFT au niveau BHandHLYP/TZVP des intermédiaires radicalaires impliqués lors du transfert d’hydrogène. Par une étude de décomposition de l’énergie d’activation, nous avons été en mesure de rationaliser l’issue diastéréosélective de la réaction sur la base des énergies de distorsion (∆Ed‡) et d’interaction (∆Eint‡) requises pour accéder à la paire d’états de transition pro-anti et pro-syn. De plus, nous avons démontré qu’une analyse NBO permettait de relativiser l’impact des interactions stéréoélectroniques. Par la suite, l’évaluation des intermédiaires radicalaires borinates et aluminates nous a permis de révéler que l’encombrement stérique de la chaîne propionate était la cause principale de la formation sélective des produits anti ou syn lors d’une réduction en présence d’un acide de Lewis.
La dernière section décrit l’élaboration de la séquence polypropionate de la zincophorine, et de ses isomères, à partir du fragment tétrahydropyrane substitué. Au cours de notre étude, nous avons identifié que le nombre de sites de chélation potentiels devait être limité à trois lors de l’aldolisation en condition Cram-chélate. De plus, nous avons démontré que les différents motifs acétates sont accessibles sélectivement par l’utilisation d’un énoxysilane encombré. Par ailleurs, nous avons révélé qu’une même séquence réactionnelle pouvait être employée pour la synthèse du fragment C17–C25 de son analogue naturel CP-78,545, et avons complété la plus récente synthèse totale du méthyl ester de la zincophorine. / The focus of this thesis is the use of free radical chemistry for the introduction of stereogenic centres in propionate motifs and application of this methodology to the total synthesis of zincophorin methyl ester. This ionophore, with interesting biological properties, displays several synthetic challenges including an anti, anti, anti, anti polypropionate sequence and a trisubstituted trans tetrahydropyran. Recently, the renewed interest for polyether compounds as anticancer agents emphasizes the significance of versatile strategies to access these targets, as well as analogues thereof.
Over the last few years, our research group has been interested in the synthesis of acyclic polypropionates via a substrate-based strategy relying on two key reactions. The first one consists of a Mukaiyama aldol reaction between an alpha-chiral aldehyde and a tetrasubstituted enoxysilane bearing a carbon-halogen bond, where the stereochemical outcome depends on the monodentate or bidentate nature of the Lewis acid. The second key step involves the generation of a tertiary free radical, vicinal to an ester, which can be reduced diastereoselectively in presence of trialkyl tin hydride.
The first section illustrates the elaboration of trisubstituted tetrahydropyran motifs and the induction of vicinal stereogenic centres via a radical reduction. We discovered that the diastereoselective outcome of the iodoetherification cyclization reaction was dictated by the gamma-methyl group of alpha,beta-unsaturated esters. Moreover, we showed that both anti and syn reduction products could be accessed from a common radical intermediate, respectively by relying on the exocyclic or endocyclic effect during the reduction reaction. Using a complementary strategy, we also revealed that the radical precursor could be accessed via a cycloetherification reaction in presence of a tetrasubstituted enoxysilane. A systematic investigation of the substituents on the ring indicated that certain stereochemical relationships led to a decrease in anti-selectivity.
The second section presents a DFT study, at the BHandHLYP/TZVP level of theory, of radical intermediates involved in hydrogen transfer. Using an energy decomposition analysis, we were able to rationalize the diastereoselective outcome of the reaction on the basis of the distortion energy (∆Ed‡) and interaction energy (∆Eint‡) required to access pro anti and pro syn transition states. Moreover, we have demonstrated that a Natural Bond Orbital (NBO) analysis can be used to interpret stereoelectronic interactions. Subsequently, an evaluation of radical intermediates bearing a borinate or an aluminate revealed that steric hindrance of the propionate chain was mainly responsible for the selective formation of anti and syn reduction products in the presence of a Lewis acid.
The last section describes the elaboration of the polypropionate sequence of zincophorin, and analogues thereof, from the substituted tetrahydropyran fragment. In our analysis, we have highlighted the necessity to limit the number of potential chelation sites to three during the aldol step under Cram-chelate conditions. Moreover, we demonstrated that the different aldol acetate motifs are accessible using a bulky enoxysilane. Finally, we showed that a similar reaction sequence can be used to access the C17–C25 fragment of its natural analogue CP-78,545 and have completed the most recent total synthesis of zincophorin methyl ester.

Glossolalie, traduction et dissonance : l'écriture hors-la-loi d'Antonin ArtaudBradette, Marie-Eve 08 1900 (has links)
Dans nombre de ses œuvres, Antonin Artaud n’a eu de cesse de formuler les termes de ce que serait une poésie anarchique, une forme langagière qui permettrait d’abolir la distance entre les mots et les choses, et à laquelle sa poésie aspire sans toutefois parvenir à une telle réalisation. Devant le constat que l’anarchie décrite par Artaud ne peut rendre compte de ce qui se réalise véritablement dans son écriture, ce mémoire pose les questions suivantes : que fait donc la poésie d’Artaud? Existe-t-il une figure qui est à même de rendre compte des modalités d’écriture qui sont à l’œuvre dans les différents textes du poète français? À ces interrogations, la figure du hors-la-loi m’est apparue des plus pertinentes dans la mesure où elle offrait tout un espace à la réflexion et à la conceptualisation. À partir de cette figure, ce mémoire formule donc l’hypothèse que ce qui se joue dans la textualité d’Antonin Artaud est une poétique hors-la-Loi, c’est-à-dire une manière de subvertir, par l’usage de différents procédés littéraires, la Loi. En d’autres termes, la poétique hors-la-Loi produit des effets de dissonance au sein du Symbolique. Pour consolider cette hypothèse, je propose deux axes de réflexion étayés à partir de l’analyse des procédés d’écriture traductologique et glossolalique d’Artaud. Dans un premier temps, la subversion est entrevue à partir des notions d’espace et de territoire. Par la suite, j’étudie la subversion dans son rapport à la temporalité. / In a number of his writings, Antonin Artaud formulated, repeatedly, the terms of what might be considered an anarchic poetry, a form of language that would allow the abolition of distance between words and things, which is what he ultimately hopes to achieve through his writings. However, Artaud was never able to achieve this sense of literary anarchy. Therefore, since the concept of anarchy put forth by Artaud cannot fully explain what is really at work in his writings, this thesis raises the following questions: What does Artaud’s poetry ultimately achieve? Is there a figure which can illustrate the writing modalities at work in the French poet’s various texts?
The figure of the outlaw is relevant in this case because it opens up the realm of reflection and conceptualisation. With the figure of the outlaw as the main starting point, this thesis formulates the hypothesis that what is played out in Antonin Artaud’s textuality is an outlaw poetic – a subversion of the Law through the use of different literary devices. In other terms, the outlaw poetic produces dissonant effects within the Symbolic. In order to reinforce this hypothesis, I shall work on two different axes of reflection which were brought forth through the analysis of Artaud’s traductologic and glossolalic literary devices. Subversion will first be seen through the notions of space and territory, and it will then be studied in its relation to temporality.

Phosphines, carbènes N-hétérocycliques (NHCs) et nouveaux précurseurs de NHCs pour la catalyse organique de réactions (macro)moléculairesFèvre, Maréva 29 September 2012 (has links)
Dans ce travail de thèse, plusieurs approches ont été développées pour permettre une utilisation plus variée des carbènes N-hétérocycliques (NHCs) et des phosphines en tant que catalyseurs/activateurs organiques.Les précurseurs de NHCs étudiés dans un premier temps, c'est-à-dire les hydrogénocarbonates d’azolium, peuvent être synthétisés en une seule étape, à l’inverse des NHCs dont la synthèse et l’isolation sont souvent compliquées. Nous avons démontré que ces espèces sont stables à l’air et sont à l’équilibre en solution avec leurs homologues carboxylates d’azolium (adduits NHC-CO2). Leur utilisation permet donc de faciliter la manipulation des NHCs tout en conservant une activité catalytique satisfaisante tant en synthèse moléculaire qu’en chimie des polymères.Des paires de Lewis silane ou borane/NHC ont ensuite été employées afin d’augmenter le potentiel des NHCs pour des réactions “modèles” de chimie (macro)moléculaire par un effet de double assistance (acide/base de Lewis).Enfin, une phosphine commerciale a été utilisée pour catalyser la polymérisation par transfert de groupe des (méth)acrylates d’alkyle de façon “vivante/contrôlée”. / In this thesis work, some points are adressed in order to broaden the scope of the application of N-heterocyclic carbenes (NHCs) as organic catalysts/activators.The novel NHC precursors studied first, i.e. azolium hydrogen carbonates, are synthesized in a one-step undemanding process, in contrast to NHCs whose synthesis and isolation is often a tedious procedure. We then showed that these species are air-stable and are at the equilibrium, in solution, with their azolium-2-carboxylates homologues (NHC-CO2 adducts). The use of such precatalysts thus allows facilitating the manipulation of NHCs, while maintaining an efficient catalytic activity in molecular chemistry as well as in polymer synthesis.We then proposed to use NHCs in conjunction with organic Lewis acids (silanes or boranes) as a possible means to induce a cooperative dual activation mechanism (Lewis acid/base) in order to increase the potential of NHCs for “model” (macro)molecular reactions.Finally, a commercial phosphine was used to trigger the group transfer polymerization of alkyl (meth)acrylates in a “controlled/living” fashion.
