Previous issue date: 2012-09-14 / This study aims to evaluate the immunohistochemical expression of antibodie MAGE-(Y18) in potentially malignant oral lesions and malignant lesions in patients with or without factor risk, and also to check the interexaminers variability on histological classification of oral epithelial dysplasia. Paraffin-embedded samples of 20 cases of severe dysplasia/carcinoma in situ and 20 cases of intraoral squamous cell carcinoma (SCC) were used in the study, and in both groups the samples had been distributed between patients with and without other risk factors associated with the occurrence of injury (tobacco and alcohol). The positive immunohistochemical expression of MAGE(Y-18) was defined by a cytoplasmic staining pattern, and it was used a scoring system considering the homogeneity of the reaction. In order to verify the interexaminer variability when performing a histological classification of oral epithelial dysplasia, 75 cases of potentially malignant oral lesions were selected from a referral service in oral disease. Slides were read independently and blindly by three pathologists in two stages. In the first stage it was made available to examiners the slides stained with H & E along with the clinical information of each case, and pathologists had to classify the lesions: no dysplasia, mild dysplasia, moderate dysplasia, severe dysplasia or carcinoma in situ.In the second stage, it was not given access to clinical information to the examiners, and they had to classify slides into two categories: low risk (non-dysplastic lesions / mild dysplasia) and high risk (moderate or severe dysplasia / carcinoma in situ). . The results were positive in 4/20 PML and 17/20 SCC. Through the Kruskal-Wallis test, it was observed a statistically significant difference (p<0.05) between groups of CPB and the group of potentially malignant lesions (PML) without the risk factor. The Mann-Whitney test found a statistically significant difference between carcinomas and potentially malignant lesions (p<0.001). The Fisher Exact test showed no statistically significant difference between the lesions that had the same histological diagnosis when compared in subgroups of associated risk factor (p=1). Detection of MAGE antigen may be useful in identifying LPM that has a higher risk of progression to invasive SCC, but it seems to be unrelated to exposure to popular risk factors The agreement between the examiners and the gold standard ranged from low to moderate, regardless of the classification system that was used. In the first stage, the intraclass correlation coefficient showed a moderate agreement between examiners 1 and 3 (r = 0.479 and r = 0.544) and, a low agreement of the examiner 2 (r = 0.384). In the second phase, it was observed a moderate agreement between examiners 2 and 3 and the gold standard (k = 0.433 k = 0.422) and, a low concordance between the examiner 1 and the gold standard (k = 0.186).Taking into account that grading dysplasia is a subjective process and still there is no test that outperforms the histological observation on diagnosis of oral epithelial dysplasia, it is understood that the process of grading made by two or more examiners should be encouraged, in order to assist in defining more precisely the diagnostics / O objetivo deste trabalho foi avaliar a expressão imuno-histoquímica do MAGE (Y-18) em LPM e malignas de pacientes com e sem fator de risco associado, bem como verificar a variabilidade interexaminadores na classificação histológica das displasias epiteliais orais. Foram utilizadas amostras incluídas em parafina de 20 casos de displasia severa/carcinoma in situ e 20 casos de CEC intraoral, sendo que em cada grupo as amostras estavam distribuídas entre pacientes com e sem fator de risco associado à ocorrência da lesão (tabaco e álcool). A expressão imuno-histoquímica positiva de MAGE (Y-18) foi definida por um padrão de coloração citoplasmático, sendo utilizado um sistema de pontuação considerando a homogeneidade da reação. Para classificar as displasias epiteliais orais, 75 casos de lesões orais potencialmente malignas foram selecionados de um serviço de referência em patologia bucal. Três patologistas analisaram as lâminas de forma independente e cega em dois momentos.Na primeira etapa foram disponibilizadas aos examinadores as lâminas coradas em H&E juntamente com as informações clínicas de cada caso e os patologistas classificaram as lesões em sem displasia, displasia leve, moderada, severa ou carcinoma in situ. Na segunda etapa, os avaliadores não tiveram acesso às informações clínicas e classificaram as lâminas em duas categorias: baixo risco (lesão não displásica/displasia leve) e alto risco (displasia moderada/severa/carcinoma in situ). A análise das reações imuno-histoquímicas demonstrou positividade em 4/20 LPM e 17/20 CEC. Pelo teste de Kruskall-Wallis observou-se diferença estatisticamente significativa (p<0,05) entre os grupos de CEC e o grupo das lesões potencialmente malignas sem fator de risco. O teste de Mann-Whitney verificou diferença estatisticamente significativa entre os carcinomas e as lesões potencialmente malignas (p<0.001). O teste Exato de Fischer não demonstrou diferença estatisticamente significativa entre as lesões de mesmo diagnóstico histológico quando comparadas nos subgrupos de fator de risco associado (p=1). Nossos resultados sugerem que a detecção do antígeno MAGE pode ser útil na identificação de LPM com maior risco de progressão para o CEC invasivo, mas parece não ter relação com a exposição aos fatores de risco mais conhecidos. Quando na classificação das displasias, independente do sistema de classificação utilizado, a concordância entre os avaliadores e o padrão ouro variou
de fraca a moderada.Na primeira etapa, o Coeficiente de Correlação Intraclasse demonstrou uma concordância moderada entre os avaliadores 1 e 3 (r=0,479 e r=0,544) e uma concordância baixa do avaliador 2 (r=0,384). No segundo momento observou-se uma concordância moderada entre os examinadores 2 e 3 e o padrão ouro (k=0,433 e k=0,422) e uma baixa concordância entre o avaliador 1 e o padrão ouro (k=0,186). Tendo em vista que a graduação da displasia é um processo subjetivo, e que ainda não dispomos de nenhum exame que supere a observação histológica no diagnóstico das displasias epiteliais orais, entendemos que o processo de graduação entre dois ou mais observadores deve ser incentivado, a fim de auxiliar na definição mais precisa do diagnóstico

Instability Studies In Amorphous Silicon Based AlloysOzdemir, Orhan 01 January 2004 (has links) (PDF)
The pixel element which is an integrated combination of a p-i-n diode with a thin film transistor (TFT) is used to produce image sensor arrays in scanning and displays technologies, necessitating the deposition of hydrogenated silicon based semiconducting and insulating thin films such as a-Si:H, a-SiNx:H over large area. The widely used techniques to achieve this goal is the plasma enhanced chemical vapor deposition (PECVD) due to its large area and low temperature (& / #61603 / 300 & / #61616 / C) abilities. In particular, PECVD has proved to be able to deposit both high quality insulator (a-SiNx:H) and active layer of p-i-n diode (intrinsic hydrogenated amorphous silicon carbide, a-SiCx:H) and by sequential deposition, it is possible to minimize the interface related problems, which play an important role in metal insulator semiconductor (MIS) and TFT structures.
PECVD deposited a-SiCx:H films over p-type crystal Si and metal substrates (MIS and MIM) were investigated by both admittance spectroscopy (Capacitance or conductance vs. voltage, temperature or frequency measurements) and Deep Level Transient spectroscopy (DLTS) to investigate the interface related problems. In this respect, instability phenomena (due to the creation of metastable states and charge injection into the gate electrode) were studied via the c-Si/a-SiCx:H (and/or a-SiNx:H) heterojunction. Specially, capacitance voltage kinetics were worked out and then the enrolled trap energies were identified with temperature mode DLTS.
The expertise gathered as a result of these studies were used in the fabrication and characterization of TFT& / #65533 / s. In this respect, inverted gate staggered type Thin Film Transistor produced and characterized for the first time after Combo-251 Pattern Generator was implemented.

On the Impact of Bariatric Surgery on Glucose HomeostasisAbrahamsson, Niclas January 2016 (has links)
Obesity has grown to epidemic proportions, and in lack of efficient life-style and medical treatments, the bariatric surgeries are performed in rising numbers. The most common surgery is the Gastric Bypass (GBP) surgery, with the Biliopancreatic diversion with duodenal switch (DS) as an option for the most extreme cases with a BMI>50 kg/m2. In paper I 20 GBP-patients were examined during the first post-operative year regarding the natriuretic peptide, NT-ProBNP, which is secreted from the cardiac ventricles. Levels of NT-ProBNP quickly increased during the first post-surgery week, and later established itself on a higher level than pre-surgery. In paper II we report of 5 patient-cases after GBP-surgery with severe problems with postprandial hypoglycaemia that were successfully treated with GLP-1-analogs. The effect of treatment could be observed both symptomatically and in some cases using continuous glucose measuring systems (CGMS). In paper III three groups of subjects; 15 post-GBP patients, 15 post-DS, and 15 obese controls were examined for three days using CGMS during everyday life. The post-GBP group had high glucose variability as measured by MAGE and CONGA, whereas the post-DS group had low variability. Both post-operative groups exhibited significant time in hypoglycaemia, about 40 and 80 minutes per day <3.3mmol/l and 20 and 40 minutes < 2.8mmol/l, respectively, longer time for DS-group. Remarkably, only about 20% of these hypoglycaemic episodes were accompanied with symptoms. In Paper IV the hypoglycaemia counter regulatory system was investigated; 12 patients were examined before and after GBP-surgery with a stepped hypoglycaemic hyperinsulinemic clamp. The results show a downregulation of symptoms, counter regulatory hormones (glucagon, cortisol, epinephrine, norepinephrine, growth hormone), incretin hormones (GLP-1 and GIP), and sympathetic nervous response. In conclusion patients post bariatric surgery exhibit a downregulated counter regulatory response to hypoglycaemia, accompanied by frequent asymptomatic hypoglycaemic episodes in everyday life. Patients suffering from severe hypoglycaemic episodes can often be treated successfully with GLP-1-analogues.

Régulation du contrôle de qualité de NKCC2 par les interactions protéine-protéine / Regulation of NKCC2 quality control by protein-protein interactionsSeaayfan, Elie 27 September 2017 (has links)
Le co-transporteur Na+-K+-2Cl- spécifique du rein et sensible au bumétanide, NKCC2, joue un rôle essentiel dans l’homéostasie hydro-électrolytique et acido-basique de l’organisme. Les mutations inactivatrices de NKCC2 induisent le syndrome de Bartter anténatal de type 1, une grave maladie rénale caractérisée par une hypotension artérielle associée à des anomalies électrolytiques. À l’opposé, une activité accrue de NKCC2 est associée à une hypertension artérielle sensible au sel. Pourtant, peu est connu sur la régulation moléculaire de NKCC2. Le but de ces travaux de thèse a donc été l’identification des déterminants moléculaires impliqués dans la régulation de l’expression et du trafic intracellulaire de NKCC2, plus spécifiquement dans le contrôle de qualité de ce co-transporteur. Suite au criblage par la technique de double hybride chez la levure d’une banque d’ADNc de rein humain, nous avons identifié OS-9 en tant que partenaire de NKCC2. La léctine OS-9 est un facteur clé de régulation du contrôle de qualité des protéines au niveau du RE. Les analyses de co-immunoprécipitation dans les cellules rénales ont montré qu’OS-9 interagit principalement avec la forme immature de NKCC2. De plus, les expériences d’immunofluorescence ont révélé que cette interaction aurait lieu au niveau du RE. La surexpression d’OS-9 diminue l’abondance totale de NKCC2. Cet effet est aboli suite à l’inhibition de la voie de dégradation protéique par le protéasome par le MG132. De plus, les expériences pulse-chase et cycloheximide-chase ont montré que cette diminution est secondaire à l’augmentation de la dégradation de la forme immature de NKCC2. A l’inverse, le knock-down d’OS-9 endogène augmente l’expression du co-transporteur en augmentant la stabilité de sa forme immature. Enfin, la mutation du domaine MRH (Mannose 6-phosphate Receptor Homology) d’OS-9 n’altère pas son effet sur NKCC2, alors que la mutation des deux sites de N-glycosylation de NKCC2 abolie l’effet d’OS-9. L’ensemble de nos résultats démontre l’implication de la lectine OS-9 dans le système ERAD de NKCC2. Le deuxième volet de ce travail a porté sur l’identification de nouveaux mécanismes Moléculaires impliqués dans le Syndrome de Bartter. Nous avons découvert des mutations dans le gène MAGE-D2, situé sur la chromosome X, responsables d’une nouvelle et très sévère forme du syndrome de Bartter anténatal, caractérisé par un polyhydramnios très précoce avec un risque élevé d’accouchement prématuré et de mortalité. Nous avons montré que les anomalies de MAGE-D2 entraînent un défaut de maturation et d’expression membranaire de NKCC2 ainsi que celle du co-transporteur Na-Cl, NCC, du tubule distal. La comparaison in vitro de l’interactome de MAGED2 sauvage et mutée a révélé que la protéine MAGE-D2 sauvage interagit spécifiquement avec DNAJB1 (HSP40) et/ou GNAS, suggérant l’implication de ces deux partenaires protéiques dans la régulation de NKCC2 et NCC par MAGE-D2 pendant la grossesse. Le troisième volet de ce travail a porté sur l’étude de l’effet de DNAJB1/HSP40, partenaire de MAGE-D2, sur l’expression de NKCC2. HSP40 a été identifiée aussi comme partenaire de NKCC2 par la technique de double hybride réalisée par notre équipe. Nous avons montré que HSP40 et son co-chaperon HSPA1A (HSP70) interagissent avec la forme immature de NKCC2 au niveau du RE. La co-expression de HSP40 et HSP70 augmente l’expression de NKCC2 en augmentant sa stabilité et sa maturation. De plus, ces deux co-chaperons régulent l’expression de NCC de la même manière. Ces observations suggèrent que MAGE-D2 coopère avec DNAJB1/HSP40 et HSPA1A/HSP70 pour protéger NKCC2 et NCC contre la rétention et la dégradation de NKCC2 au niveau du RE durant la grossesse, révélant ainsi une nouvelle voie de régulation du trafic intracellulaire de NKCC2 et NCC. Mutations of NKCC2 cause antenatal type 1 Bartter syndrome, a life-threatening kidney disease characterized by arterial hypotension associated with electrolyte abnormalities. In contrast, an increase in NKCC2 activity is associated with salt-sensitive hypertension. Yet the mechanisms underlying the regulation of NKCC2 trafficking in renal cells are scarcely known. The aim of this work was to identify the protein partners involved in the regulation of the expression and the intracellular trafficking of NKCC2, specifically in the quality control of this co-transporter. Using the yeast tow-hybrid system, we identified OS-9 as a specific binding partner of NKCC2. Lectin OS-9 is a key factor in the regulation of protein quality control at ER. Co-immunoprecipitation assay in renal cells showed that OS-9 interacts mainly with NKCC2 immature forms. Accordingly, immunocytochemistry analysis showed co-localization of the proteins mainly in the ER. Overexpression of OS-9 decreased the total abundance of NKCC2. This effect is abolished following the inhibition of the proteasome protein degradation pathway by MG132. In addition, the pulse-chase and cycloheximide-chase assays demonstrated that the marked reduction in the co-transporter protein levels was essentially due to increased protein degradation of NKCC2 immature forms. Conversely, knock-down endogenous of OS-9 increased the expression of the co-transporter by increasing the stability of its immature form. Finally, inactivation of the Mannose 6-phosphate Receptor Homology domain had no effect on its action on NKCC2, while mutation of the two NKCC2 N-glycosylation sites abolished the effect of OS- 9. In summary, our results demonstrate the involvement of lectin OS-9 in the ERAD of NKCC2. The second part of this work focused on the identification of new molecular mechanisms involved in Bartter Syndrome. We found that MAGE-D2 mutations caused X-linked new and severe form of antenatal Bartter's syndrome, characterized by a very early polyhydramnios with a high risk of premature delivery and mortality. We have shown that MAGE-D2 abnormalities lead to a lack of maturation and membrane expression of NKCC2 as well as that of the Na-Cl co-transporter, NCC, of the distal tubule. In vitro comparison of the wild-type and mutated MAGED2 interactome revealed that wild-type MAGE-D2 interacts specifically with DNAJB1 (HSP40) and / or GNAS, suggesting involvement of these two protein partners in NKCC2 and NCC regulation by MAGE-D2 during pregnancy. The third part of this work focused on the study of the effect of DNAJB1 / HSP40, partner of MAGE-D2, on the expression of NKCC2. HSP40 was also identified as a specific binding partner of NKCC2 by the yeast two-hybrid system realized by our team. We have shown that HSP40 and its co-chaperone HSPA1A (HSP70) interact with the immature form of NKCC2 at the ER. The co-expression of HSP40 and HSP70 increased the expression of NKCC2 by increasing its stability and maturation. In addition, these two co-chaperones regulate the expression of NCC in the same way. These findings suggest that MAGE-D2 cooperates with DNAJB1 / HSP40 and HSPA1A / HSP70 to protect NKCC2 and NCC against retention and degradation of NKCC2 at ER during pregnancy, revealing a new pathway for regulating NKCC2 and NCC intracellular trafficking. A better understanding of NKCC2 and NCC regulatory pathways would help to better understand the pathophysiology of sodium retention and ultimately would provide a new target for a pharmaceutical approach to preventing and / or treating kidney disease related to sodium balance.

This study analyses two non-western novels used in the subject of Swedish in upper secondary school: Fatou Diomes The Belly of the Atlantic and Chimamanda Ngozi Adichies Half a Yellow Sun. Looking at how the books female main character relate to Stuart Halls theory of cultural identity, I come to the conclusion that they somewhat differently relate to an essential ”authentic” self. Salie talks explicit about a generic African soul that she possesses. Olanna never talks about anything ”authentic”, but her narrative and contrary subject positions can be read as a way of demasking her European ”white” self in favour of a truer Igbo self. I also come to the conclusion that both novels use themes of alienation related to gender structures and positioned westernness and that this kind of reading could contribute to interesting classroom discussions about a dynamic interpretation on culture and identity.

Diplomová práce je zaměřena na problematiku detekce a sledování polohy hlavy v obraze jako jednu s možností jak zlepšit možnosti interakce mezi počítačem a člověkem. Hlavním přínosem diplomové práce je využití inovativních hardwarových a softwarových technologií jakými jsou Microsoft Kinect, Point Cloud Library a CImg Library. Na úvod je představeno shrnutí předchozích prací na podobné téma. Následuje charakteristika a popis databáze, která byla vytvořena pro účely diplomové práce. Vyvinutý systém pro detekci a sledování polohy hlavy je založený na akvizici 3D obrazových dat a registračním algoritmu Iterative Closest Point. V závěru diplomové práce je nabídnuto hodnocení vzniklého systému a jsou navrženy možnosti jeho budoucího zlepšení.
