運動產業之品牌銷售公司,產品須配合四季推出,生命週期短且汰換率高,此外擴張產品策略到休閒服飾,也因此面臨休閒服飾國際品牌紛紛進入之衝擊及流行趨勢挑戰。在高度競爭的運動產業中,消費者的購買行為強烈受到品牌認知影響,品牌建構成為產品差異化的重要關鍵。業者為了提昇業績成長和提高市佔率,如何有效地將有限的行銷資金投資於品牌之建構,成為重要的管理議題。藉由在人潮聚集的主要商圈投資經銷商或自營的銷售店點,開設品牌形象專賣店,是業者達成業績成長的主要策略之一。在快速投資的過程中如何提升採購效率,降低開店成本,以達到盈餘成長,攸關業者的生存發展。因此,開設銷售店點之營運流程,也成為成本控制的重要關鍵流程,藉以在精簡的人事組織中持續簡化流程,增加營運效率,確保服務品質,降低成本,提高企業競爭力。 本研究首先運用企業流程資訊有機體( PWIO)模式分析個案公司開設銷售店點交易循環的營運流程,透過分析作業流程對內、對外之資訊聯絡管道和接觸點,以實作表詳實描述各項作業的原貌。根據PWIO分析結果,辨識出個案公司管理議題缺失,針對各項作業提出具體的流程改進建議,另一方面在實作表中建立各該流程的目標,並提出具體衡量各作業流程的績效衡量指標。依據目標與績效衡量指標的因果關係進一步探討個案公司的平衡計分卡並描繪策略地圖,提供個案公司持續改善流程的績效管理架構基礎,以實現公司願景。 / The products of the sports brand companies need to be launched seasonally. The life cycle of sports products is short. The market strategy has expanded beyond active users to encompass the leisurewear market which makes the sports industry encounter not only the threat of international leisurewear’s brand but also the challenge of fashion trend. Facing such sharp competition, branding has become a key success factor for differentiation. As consumers are strongly affected by brand images, in order to increase both sales and market share, it is essential to allocate marketing resources effectively to brand building activities. By contributing brand-specific fixtures and signage to franchised stores or direct retail stores in shopper-populated districts, running branded stores has become the main sales growth strategies. In the course of fast investment, how to enhance the efficiency of purchase and how to lower the cost of setting up shops to achieve earning growth become the key to growth. Therefore, the business process of setting up retail shops becomes critical to cost control. In short, the key factors to success lies in how to reengineer the business process in simplified organization, in order to improve performance efficiency, to ensure service quality, to lower cost for the purpose of strengthen industry competition power. This study attempts to analyze the information flow underlying the business process of setting up retail shop for a subsidiary of an international sports brand in Taiwan. Based on the PWIO approach, the operation process was restored and descried in the business table by identifying the practice of contact point (CP) and information channel(IC). The related suggestions to improve management gaps of this process are proposed according to the PWIO analysis. The practices table exploring also help to set up the objectives and concreted performance measures to enhance the value adding degree of this strategic business process. A proposed balanced scorecard and strategy map are developed to serve as the management framework of evaluation and management of the strategic performance to assuring sustained process improvement. Company vision will be achieved through the business process reengineering.

Functional Role Of Heat Shock Protein 90 From Plasmodium Falciparum

Molecular chaperones have emerged in recent years as major players in many aspects of cell biology. Molecular chaperones are also known as heat shock proteins (HSPs) since many were originally discovered due to their increased synthesis in response to heat shock. They were initially identified when Drosophila salivary gland cells were exposed to a heat shock at 37°C for 30 min and then returned to their normal temperature of 25°C for recovery. A “puffing” of genes was found to have occurred in the chromosome of recovering cells, which was later shown to be accompanied by an increase in the synthesis of proteins with molecular masses of 70 and 26 kDa. These proteins were hence named “heat shock proteins”. The first identification of a function for HSPs was the discovery in Escherichia coli that five proteins synthesized in response to heat shock were involved in λ phage growth. The products of the groEL and groES genes were found to be essential for phage head assembly while the dnaK, dnaJ and grpE gene products were essential for λ phage replication. It was later shown that GroEL and GroES are part of a chaperonin system for protein folding in the prokaryotic cytosol while DnaK is a member of the Hsp70 family that works in conjunction with the DnaJ (Hsp40) co-chaperone and the nucleotide exchange factor GrpE to promote phage replication by dissociating the DnaB helicase from the phage-encoded P protein. Since then, a large number of other proteins collectively referred to as HSPs have been discovered. However, heat shock is not the only signal that induces synthesis of heat shock proteins. Stress of any kind, such as nutrient deprivation, chemical treatment and oxidative stress among others causes increased production of HSPs and therefore, they are also known as stress proteins. The term “molecular chaperone” was originally used to describe the function of nucleoplasmin, a Xenopus oocyte protein that promotes nucleosome assembly by binding tightly to histones and donating the bound histone to chromatin. However, since then, chaperones have been defined as “a family of unrelated classes of proteins that mediate the correct assembly of other proteins, but are not themselves components of the final functional structure”. This view of molecular chaperones, though undoubtedly correct, doesn’t capture the multifaceted roles they have since been discovered to play in cellular processes. In recent years, molecular chaperones have been shown to perform other functions in addition to the maintenance of protein homeostasis: translocation of proteins across organelle membranes, quality control in the endoplasmic reticulum, turnover of misfolded proteins as well as signal transduction. As a result, many chaperones are also essential under non-stress conditions and play crucial roles in cell growth and development, cell-cell communication and regulation of gene expression. Heat shock protein 90 (Hsp90) is one of the most abundant and highly conserved molecular chaperones in organisms ranging from bacteria to all branches of eukarya. It has been shown to be essential for cell viability in Saccharomyces cerevisiae, Schizosaccharomyces pombe and Drosophila melanogaster. Although the bacterial homolog HtpG is dispensable under normal conditions, it is important for cell survival during heat shock. In addition to its role as general chaperone in protein folding following stress, Hsp90 has a more specialized role as a chaperone for several protein kinases and transcription factors. Many Hsp90 client proteins are signaling proteins involved in regulation of cell growth and survival. These proteins are critically dependent on Hsp90 for their maturation and conformational maintenance resulting in a key role for Hsp90 in these processes. Recent reports have also highlighted a role for Hsp90 in linking the expression of genetic and epigenetic variation in response to environmental stress with morphological development in Drosophila melanogaster and Arabidopsis thaliana. In Candida albicans, Hsp90 augments the development of drug resistance, implicating a role for Hsp90 in the evolution of infectious diseases. The malarial parasite, Plasmodium falciparum, is the causative agent of the most lethal form of human malaria. The parasite life cycle involves two hosts: an invertebrate mosquito vector and a vertebrate human host. As the parasite moves from the mosquito to the human body, it experiences an increase in temperature resulting in a severe heat shock. The mechanisms by which the parasite adapts to changes in temperature have not been deciphered. Our laboratory has been interested in investigating the role of heat shock proteins during acclimatization of the parasite to such temperature fluctuations. Heat shock proteins of the Hsp40, Hsp60, Hsp70 and Hsp90 families have been characterized in the parasite and are being examined in our laboratory. This thesis pertains to understanding the functional role of Plasmodium falciparum Hsp90 (PfHsp90) during adaptation of the parasite to fluctuations in environmental temperature. The parasite expresses a single gene for cytosolic Hsp90 on chromosome 7 (PlasmoDB accession no.: PF07_0029) coding for a protein of 745 amino acids with a pI of 4.94 and Mw of 86 kDa. Eukaryotic Hsp90 regulates several protein kinases and transcription factors involved in cell growth and differentiation pathways resulting in a crucial role for Hsp90 in developmental processes. A role for PfHsp90 in parasite development, therefore, seems likely. Indeed, PfHsp90 has previously been implicated in parasite development from the ring stage to the trophozoite stage during the intra-erythrocytic cycle. Pharmacological inhibition of PfHsp90 function using geldanamycin (GA), a specific inhibitor of Hsp90 activity, abrogates stage progression. These experiments suggest that PfHsp90 may play a critical role in parasite development. This is further substantiated by the fact that several pathogenic protozoan parasites such as Leishmania donovani, Trypanosoma cruzi, Toxoplasma gondii and Eimeria tenella depend on Hsp90 function during different stages of their life cycles. It appears, therefore, that a principal role of Hsp90 in protozoan parasites may be the regulation of their developmental cycles. However, the precise functions of PfHsp90 during the intra-erythrocytic cycle of the malarial parasite are not clear. In this study we have carried out a functional analysis of PfHsp90 in the malarial parasite. We have examined the role of PfHsp90 in parasite development during repeated exposure to febrile temperatures. We have investigated its involvement in parasite development during a commonly used synchronization protocol involving cyclical changes in temperature. We have examined the interaction of GA with the Hsp90 multi-chaperone complex from P. falciparum as well as the human host. Finally, we have carried out a systems level analysis of chaperone networks in the malarial parasite as well as its human host using an in silico approach. We have analyzed the protein-protein interactions of PfHsp90 in the chaperone network and predicted putative cellular processes likely to be regulated by parasite chaperones, particularly PfHsp90.

Biopolitics and Reflexivity : A Study of GMO Policymaking in the European Union / Biopolitik och reflexivitet : En studie av tillkomsten av GMO policy i den Europeiska Unionen

The political discourse that has emerged as a consequence of establishing a European regulatory framework for GMOs has not been without problems. This dissertation addresses the political and regulatory challenges created by the development and use of genetically modified organisms (GMOs) in the EU. The underlying hypothesis in the dissertation is that the emergence of a European policy in the field of GMOs has appeared through new reflexive forms of governance. The aim of this research is to understand how these reflexive forms of regulation have emerged and operate within the EU, with a particular focus on the two GMO directives 90/220/EEC and 2001/18/EC. However, the study scrutinises the regulatory regimes from the 1970s onwards by investigating how the regulatory framework regarding GMOs has been developed and implemented in the EU. This is done through an analysis of the notion of ‘risk’ and the ‘precautionary principle’ since these concepts have been at the forefront of the GMO regulation debate. The empirical approach focuses on how the European Commission, the European Parliament and the European Council as well as other actors and institutions articulate ambivalence, interpretations and uncertainties in the decision-making processes regarding regulative measures for GMOs, with an accent on risk and the precautionary principle. The main empirical material has been documents concerning the inter-institutional process involved in the regulatory process of making the two directives. The analysis indicates that during the process of implementing GMO regulations, new steering strategies have appeared within the EU’s decision-making institutions when the objective of the regulation has taken centre stage in political and scientific controversies. / Den politiska diskursen som har uppstått som en följd av upprättandet av ett europeiskt regelverk för GMO har inte varit utan problem. Denna avhandling behandlar de politiska utmaningar som alstrats av skapandet och tillämpningen av genetiskt modifierade organismer (GMO) inom EU. Den underliggande hypotesen i avhandlingen är att framväxten av en europeisk politik på området för GMO har uppstått genom nya reflexiva regleringsformer. Syftet med denna forskning är att förstå hur reflexiva regleringsformer har uppkommit och opererar inom EU, med särskilt fokus på de två GMO direktiven 90/220/EEG och 2001/18/EG. Studien analyserar lagstiftning från 1970-talet och framåt genom att undersöka hur lagstiftningen om genetiskt modifierade organismer har utvecklats och implementerats i EU. Detta görs genom en analys av begreppen ‘risk’ och ‘försiktighetsprincipen’ eftersom dessa begrepp har varit centrala för debatten om GMO lagstiftningen. Den empiriska metoden fokuserar på hur Europeiska kommissionen, Europaparlamentet och Europeiska rådet samt andra aktörer och institutioner har uttryckt ambivalens, tolkningar och osäkerhet i beslutsfattandet gällande reglerings åtgärder för genetiskt modifierade organismer, med tonvikt på risk och försiktighetsprincipen. Analysen visar att implementeringen av GMO lagstiftningen har skapat nya styrningsstrategier i synnerhet i de fall där ändamålet med lagstiftningen har varit föremål för politiska och vetenskapliga kontroverser.

"Mais um vizinho", a Floresta Nacional : mobilização e controvérsias na gestão ambiental em Mato Castelhano-RS

Esta dissertação é resultado da pesquisa etnográfica desenvolvida junto a os atores envolvidos na gestão ambiental da Unidade de Conservação da Natureza (UC) chamada Floresta Nacional de Passo Fundo (Mato Castelhano/RS). Este universo compõe-se de cientistas, servidores públicos, agricultores e índios kaingang, em diálogo sobre o uso e a preservação da floresta. Nos encontros do Conselho Gestor da UC são expressas visões de mundo que se refletem em formas diferenciadas de apropriação dos elementos do ambiente próprias de cada grupo. Em um contexto de introdução de lavouras transgênicas nas propriedades próximas à área protegida, o estudo visa a apreender pontos de vista sobre a "natureza" e a "sociedade" e sobre o ambiente de Mato Castelhano entre os participantes do Conselho Gestor da UC, pesquisadores, especialistas da área ambiental e agricultores, abordando ainda a perspectiva de índios kaingang acampados na BR-285. Foram realizadas entrevistas semi-estruturadas com índios e conselheiros e observação participante de suas atividades e reuniões, além da análise de documentos. Evidencia-se que o diálogo em relação aos elementos objetos de manejo e conservação é dificultado pela desconsideração da complexidade da maneira de conceber o ambiente entre os diferentes atores locais. / This dissertation results from an ethnographic field work among actors who are involved in a process of public management of an Environmental Protected Area, which is called National Forest of Passo Fundo (Mato Castelhano/RS). This universe is composed of scientists, public servants, farmers and a kaingang indigenous group, dialoging about the use of "nature" and its conservancy. In the gatherings of the Management Council of the National Forest are expressed different perspectives in what concerns the appropriation of the area and its elements, which are related to each one's ways to engage themselves in that environment. In a context of transgenic agriculture in the farms around the protected area, the study focus the apprehension of points of view related to "nature" and "society", as well as the environment of Mato Castelhano, among members of the Council, professional researchers, experts on environment issues, farmers, and includes the perspective of the kaingang indians. In a qualitative approach, were made analysis of documents, semi-directive interviews and participant observation during ordinary meetings and tasks of the councilors, and within the indigenous encampment on the highway BR-285. The research evidences that the dialog about use and conservancy of "natural" elements do not reach to consider the different ways of conceive environment among the local actors.

In this work we focused on the application of physical activity, which aimed to reduce the negative impact of physical deprivation and associated functional and structural changes in the musculoskeletal system. The principal criterion was the diagnosis of the organ systems of the locomotor system, focusing on the muscular system. The subsequent application of appropriate forms of exercise intervention was designed to mitigate the effects of the aging process and functional changes in the body of the senior individual. Finally, we wanted to assess the impact of changes in body composition in its own self- sufficiency and independence of older people and the quality of life of older people in terms of physical and psychosocial aspects. The most significant is the loss of muscle mass, sarcopenia development, increase fat mass with frequent reductions in body weight. The focus of our work and its specific purpose is to clarify the influence of the applied physical interventions. To assess changes in body composition are needed for our work used the method of multi Bioanalysis. Results in body composition in older adults have been evaluated and intra-individual measured data were compared with the general senior population. This fieri is hereby taken as representative of what to be made or to become in a science not arrested by its own previous conceptions. As to advance this research: it has been identified the phases among the theoretical fieri of Carl Rogers; the understanding of Rogers concerning the Organism as presented in each of his intellectual phases; considered the main theorists and academic influences Rogers recognized upon his own work; made explicit the broader epistemological frameworks which informed Rogers´ understanding of Organism. This thesis has been inspired by the historical-method developed by Jean Piaget as an epistemological approach for scientific research which focus on recognizing the ideas that have supported the development of a concept or a theory. This methodology was based in a deductive formulation as for what may have grounded the notion of Organism as formulated in the original texts as well as from those who were theoretically influential on Rogers´ system. Coming from a selection of those pieces of work explicitly referring to the concept of Organism, we may infer Rogers was profoundly part of this cultural moment. Not only restricted to assimilating and elaborating these diffused influences at his experience, his Organismic conceptualization turned possible the integration of different dimensions as such Personality (I/Me), Society (interrelations) and Nature (Cosmos). At each stage of his thought, Rogers indicated the following influences on the construction of that conception: (1) in non-directive couseiling – the functionalism-pragmatism north american and new psychoanalysis of Rank, Horney and Sullivan; (2) in client-centered therapy – repeat the previous influences and increases scientism and its north american applied Psychology, Gestalt Psychology and Kurt Lewin, educacional, social and politic philosophies, and theorists of personality; (3) in transition from client-centered therapy and person-centered approach, the impact of the experiencing concept of Gendlin, experience with groups, the performance in education, reflections of alternative perspectives and behavioral sciences and studies on organism and self-actualization Goldstein, Maslow, Angyal and allusions to Whyte; (4) in person-centered approach, it adds to studies of organism and self-actualization, the research of Szent-Gyoergy and the emergency of Capra, Prigogine and Maruyama systemic paradigm and holistic. This present thesis led us to reveal a new intelligibility traced with Rogers´ notion of Organism where emergency a cosmological approach for the person which does not restrain itself to the Personality. We understood Rogers was tracking his person-centered academic ground largely dependent on the new findings of the emergent contemporary science. / Esta dissertação tem como objetivo investigar, mediante uma perspectiva epistemológica, a noção de organismo no fieri teórico de Carl Rogers. Por fieri entende-se o devir ou o fato que faz uma ciência não ficar estagnada em suas concepções. Para realizar esta investigação foram: identificadas as fases do fieri teórico de Rogers; pesquisado o fato de como Rogers concebeu a noção de organismo; examinados os principiais teóricos e influências que Rogers reconheceu a este respeito; verificadas as linhas epistemológicas gerais da noção de organismo em Rogers. Neste trabalho adotou-se uma abordagem epistemológica de pesquisa, inspirada no uso que Jean Piaget fez do método histórico que, segundo ele, consiste em determinar como procedeu à invenção de um conceito ou teoria, reconhecendo as ideias que a tornaram possível. O método utiliza-se de um sistema dedutivo do que foi levado a imaginar a criação da ideia de organismo em Rogers. Foram utilizados como fontes de pesquisa os textos escritos de Rogers e de suas influências. A seleção destes seguiu o critério de conter aspectos relativos à concepção organísmica de Rogers. Como resultado, ficou evidente que Rogers foi um pensador ligado às questões de sua época. Ele assimilou e elaborou, com suporte em sua experiência com essas influências, uma concepção organísmica que integrou as dimensões da personalidade (eu), sociedade (inter-elações) e natureza (cosmos). Em cada fase do seu pensamento, Rogers indicou as seguintes influências para essa concepção: (1) no aconselhamento não-diretivo o funcionalismo-pragmatismo dos Estados Unidos e a psicanálise neofreudidana de Rank, Horney e Sullivan; (2) na terapia centrada no cliente repetem-se as influências anteriores acrescidas do cientificismo estadunidense e sua Psicologia aplicada, a Psicologia da Gestalt, Kurt Lewin, a filosofia educacional, social e política estadunidense e os teóricos da personalidade; (3) na transição entre terapia centrada no cliente e abordagem centrada na pessoa - o impacto do conceito de experienciação de Gendlin, as experiências com grupos, a atuação no campo da educação, as reflexões de perspectivas alternativas às ciências do comportamento e os estudos sobre organismo e auto-realização de Goldstein, Maslow, Angyal e alusões a Whyte; (4) na abordagem centrada na pessoa – se acresce aos estudos de organismo e auto-realização, as pesquisas de Szent-Gyoergy, e a emergência do paradigma sistêmico e holístico de Capra, Prigogine e Maruyama. Considera-se que é possível traçar uma nova inelegibilidade de Rogers com base em noção de organismo. Por esta é possível pensar no avanço de uma abordagem cosmológica da pessoa que não se restringe somente à personalidade. Percebe-se que Rogers deu pistas de como desenvolver um solo científico para a abordagem centrada na pessoa, com arrisco paradigma emergente de ciência contemporânea.

Em um contexto de introdução de lavouras transgênicas nas propriedades próximas à área protegida, o estudo visa a apreender pontos de vista sobre a "natureza" e a "sociedade" e sobre o ambiente de Mato Castelhano entre os participantes do Conselho Gestor da UC, pesquisadores, especialistas da área ambiental e agricultores, abordando ainda a perspectiva de índios kaingang acampados na BR-285. Foram realizadas entrevistas semi-estruturadas com índios e conselheiros e observação participante de suas atividades e reuniões, além da análise de documentos. Evidencia-se que o diálogo em relação aos elementos objetos de manejo e conservação é dificultado pela desconsideração da complexidade da maneira de conceber o ambiente entre os diferentes atores locais. / This dissertation results from an ethnographic field work among actors who are involved in a process of public management of an Environmental Protected Area, which is called National Forest of Passo Fundo (Mato Castelhano/RS). This universe is composed of scientists, public servants, farmers and a kaingang indigenous group, dialoging about the use of "nature" and its conservancy. In the gatherings of the Management Council of the National Forest are expressed different perspectives in what concerns the appropriation of the area and its elements, which are related to each one's ways to engage themselves in that environment. In a context of transgenic agriculture in the farms around the protected area, the study focus the apprehension of points of view related to "nature" and "society", as well as the environment of Mato Castelhano, among members of the Council, professional researchers, experts on environment issues, farmers, and includes the perspective of the kaingang indians. In a qualitative approach, were made analysis of documents, semi-directive interviews and participant observation during ordinary meetings and tasks of the councilors, and within the indigenous encampment on the highway BR-285. The research evidences that the dialog about use and conservancy of "natural" elements do not reach to consider the different ways of conceive environment among the local actors.

A presente pesquisa tem como objetivo desenvolver um estudo acerca da noção aristotélica de constituição orgânica, no qual procurarei propor um exame da filosofia da natureza de Aristóteles referente ao aspecto, atribuído ao organismo, de ser substância natural no seu mais elevado grau, estabelecendo uma análise comparativa entre os compostos elementares, os compostos homogêneos inanimados e os compostos orgânicos. Para tanto, pretendo, primeiramente, promover uma análise a respeito do caráter substancial dos organismos vivos. Em seguida, delimitar uma investigação em torno dos tipos de composições naturais, levando em conta a maneira pela qual a necessidade natural estaria envolvida no processo constitutivo de tais composições. E, por fim, oferecer uma interpretação relativa ao capítulo 12 do livro IV dos Meteorológicos, a qual tem por base discernir a natureza formal associada, por um lado, às propriedades características dos corpos homogêneos em geral, considerados em si e por si mesmos, e, por outro, às propriedades características da constituição orgânica, de modo a precisar as diferenças entre elas. Penso que, com este estudo, também será possível compreender de um modo claro o motivo pelo qual, nesta concepção de natureza, toma-se o organismo vivo como o paradigma de substâncias (ousiai) naturais. / This research aims to develop a study on the aristotelian notion of organic constitution, which seek to propose an examination of the natural philosophy of Aristotle concerning the aspect, assigned to the body, to be natural substance in its highest degree, establishing an analysis comparative between elementary compounds, inanimate homoeomerous compounds and organic compounds. Therefore, I intend, first, to promote an analysis regarding the substantial nature of living organisms. Then delimit an investigation around the kinds of natural compositions, taking into account the way in which natural necessity would be involved in the constitutive process of such compositions. And finally, offer an interpretation relating to Chapter 12 of De Caelo IV, which relies on discerning the formal nature associated, on the one hand, the characteristic properties of homoeomerous bodies generally considered in themselves and for themselves, and, on the other hand, the characteristic properties of the organic constitution in order to clarify the differences between then. I think that, with this study, also will be possible understand in a clearly way the reason, in this conception of nature, it takes the living organism as the paradigm of natural substances (ousiai).

Esta dissertação é resultado da pesquisa etnográfica desenvolvida junto a os atores envolvidos na gestão ambiental da Unidade de Conservação da Natureza (UC) chamada Floresta Nacional de Passo Fundo (Mato Castelhano/RS). Este universo compõe-se de cientistas, servidores públicos, agricultores e índios kaingang, em diálogo sobre o uso e a preservação da floresta. Nos encontros do Conselho Gestor da UC são expressas visões de mundo que se refletem em formas diferenciadas de apropriação dos elementos do ambiente próprias de cada grupo.
