Caracterização morfológica e molecular de Strongyloides ophidiae (nematoda, strongyloididae) parasitas de serpentes

A primeira foi descrita em serpente Mastigodryas bifossatus e a segunda em lagartixa Hemidactylus mabouya. Na descrição de S. ophidiae apenas a fêmea partenogenética foi caracterizada em 1929 e, desde então, nenhum outro relato sobre esta espécie foi publicado. Estudos moleculares foram realizados com o objetivo de contribuir para a caracterização de Strongyloides spp. foram realizados, porém estes não incluíram S. ophidae. No presente estudo foi realizada a caracterização morfológica e molecular de S. ophidiae coletados em serpentes da região de Botucatu, Estado de São Paulo, Brasil. De um total de 125 animais, apenas quatro encontraram-se parasitados por S. ophidiae, porém fêmeas partenogenéticas foram recuperadas de apenas um animal. Coproculturas foram realizadas para obtenção das larvas L1, L2 e L3, bem como machos e fêmeas de vida livre, os quais foram morfologicamente analisados. Os produtos da PCR apresentaram 350 pb e as seqüências de nucleotídeos (número de acesso Genbank EU287935) isoladas deste nematódeo de Oxyrhopus guibei apresentaram 98% de identidade com Strongyloides procyonis (número de acesso AB272234.1 e AB205054.1) e 97% de identidade com Strongyloides cebus, Strongyloides stercoralis, Strongyloides sp. “ex snake” e Strongyloides fuelleborni (Genbank AB277236.1, AF279916.2, AJ417031.1 e AJ407030.1, respectivamente). A análise filogenética das espécies da família Strongyloididae e Rhabdiasidae demonstrou que, embora apresente 97% de similaridade com outras espécies, S. ophidiae encontra-se isolado em um ramo da árvore filogenética, caracterizando uma identidade diferente em relação a outras espécies analisadas. O presente estudo caracterizou morfológica e molecularmente S. ophidiae, parasita de serpentes da região de Botucatu, São Paulo, Brasil. / In Brazil the genus Strongyloides includes two reptile parasite: S. ophidiae and S. cruzi. The former was described in Mastigodryas bifossatus snake and the other in Hemidactylus mabouya gecko. In the description of S. ophidiae just the partenogenetic females was characterized in 1929 and, ever since, no other report about this species was published. Molecular studies for the characterization of Strongyloides spp. were reported, however these did not include S. ophidae. In the present study the morphological and molecular characterization of S. ophidiae obtained from snakes in Botucatu region, São Paulo State, Brazil was accomplished. Among the 125 studied animals only four snakes were found infected by S. ophidiae, however partenogenetic females were recovered in only one animal. Fecal exams had been performed for obtaining L1, L2 and L3 larvae, as well as free-living males and females, which were morphologically analyzed. PCR products amplified from S. ophidiae partenogenetic female samples presented about 350 bp. Nucleotide sequences (Genbank accession numbers EU287935) isolates from these nematodes from Oxyrhopus guibei presented 98% identity with Strongyloides procyonis (Genbank acession number AB272234.1 and AB205054.1) and 97% identity with Strongyloides cebus, Strongyloides stercoralis, Strongyloides sp. ex snake and Strongyloides fuelleborni (Genbank AB277236.1, AF279916.2, AJ417031.1 and AJ407030.1, respectively). The analysis of the phylogenetic relationship among the species of Strongyloididae and Rhabdiasidae demonstrated that, although there are similarity above 97% with the other species, S. ophidiae was allocated in an isolated branch of the phylogenetic tree, characterizing a different identity in relation to other species analyzed. The present study contributed for the morphological and molecular characterization of S. ophidae from the Botucatu region, São Paulo State, Brazil.

Eficácia dos fungos nematófagos Duddingtonia flagrans e Arthrootrys robusta na profilaxa das infecções naturais por nematódeos gastrointestinais em ovinos

O uso indiscriminado de anti-helmínticos teve como conseqüência a seleção de populações de helmintos resistentes. A utilização de fungos nematófagos predadores como controle biológico é uma das alternativas pesquisadas para auxiliar na profilaxia das helmintoses. O objetivo do trabalho foi avaliar a eficácia de fungos nematófagos e a administração de diferentes dosagens na profilaxia das nematodioses gastrintestinais em ovinos naturalmente infectados em Botucatu - SP. O experimento foi dividido em duas etapas, nas quais primeiramente avaliou-se o fungo mais eficaz (Duddingtonia flagrans e Arthrobotrys robusta) e posteriormente duas dosagens do fungo A. robusta. As variáveis avaliadas foram: peso, volume globular (VG), ovos de estrongilídeos por grama de fezes (OPG), coproculturas e larvas infectantes (L3) na pastagem. Não houve diferença significativa no peso médio e VG entre os grupos estudados em ambos experimentos. O número médio de OPG não diferiu significativamente entre os grupos na maioria das colheitas, entretanto o grupo que recebeu o fungo A. robusta (Experimento I) foi o que recebeu menor quantidade de tratamentos com anti-helmínticos, logo foi o fungo utilizado para o Experimento II. Neste houve redução significativa no número de L3 de Trichostrongylus spp. por quilo de matéria seca de forragem nos piquetes pastejados pelo grupo tratado diariamente com fungo A. robusta. Haemonchus contortus foi a espécie predominante em cordeiros traçadores utilizados em ambos os experimentos. Nas condições experimentais do estudo o controle biológico não foi eficaz na profilaxia das helmintoses. / Parasitic gastroenteritis is one of the major problems confronted of the sheep industry. The indiscriminate use of anthelmintics has had as consequence the resistant selection of helminth population. The use of nematophagous fungi as biologic control is one of the alternatives strategies to gastrointestinal nematode infection prophylaxis. The aim of this work was to evaluate the efficacy of the nematophagous fungi and its use in different doses to the prophylaxis of natural infection by gastrointestinal nematodes in sheep kept on pasture in Botucatu – SP, Brazil. The experiment was carried out in two phases: in the first trial, the comparative efficacy of Duddingtonia flagrans and Arthrobotrys robusta fungi was evaluated and in the second, two different strategies of administration of A. robusta fungi. The measurements were: body weight, packed cell volume (PCV), faecal egg counts (FEC), and numbers of third stage larvae (L3) on pasture. There were no significant differences regarding body weight and PCV means between the studied groups of both experiments. The mean FEC did not differ significantly between the groups at the most of the sample collections, however, the daily feed A. robusta group (Experiment I) received the smaller quantity of anthelmintics treatments, so it was the fungi used in the Experiment II. Trichostrongylus spp. L3 numbers per kilogram of dry matter was significantly lower on pasture grazed by the daily treated group with A. robusta fungi. Haemonchus contortus was the predominant species recovered from the tracer lambs in both experiments. At the experimental conditions of the study, the biologic control was not efficient to the helminth prophylaxis in sheep.

Expressão imunoistoquímica de ácido retinóico, CRABP2, NFkB p100 e p105, TRAP1 e TWEAK em nefroblastomas

Apesar do prognóstico favorável uma parcela dos pacientes evolui para recidiva e óbito. Biomarcadores atuantes na progressão tumoral, proliferação, diferenciação, apoptose e recentemente descrita ação na resposta imune e inflamação, tais como ácido retinóico (AR), CRABP2, NF?B p100/52, NF?B p105/50, TRAP1 e TWEAK tem sido descritos. Este estudo tem por objetivo a avaliação da expressão imunoistoquímica de ácido retinóico (AR), CRABP2, NF?B p100/52, NF?B p105/50, TRAP1 e TWEAK em amostras parafinadas de nefroblastomas correlacionando-a com fatores prognósticos. Foram selecionados 77 espécimes de nefroblastoma provenientes de serviços de patologia. A imunoexpressão foi avaliada através de análise semiquantitativa e quantitativa. Para a análise semiquantitativa empregou-se o sistema escore de Allred e para a avaliação quantitativa, a análise morfométrica da área corada. Para a comparação de dois grupos foi utilizado o teste não-paramétrico de Mann-Whitney, enquanto para a comparação de três ou mais grupos utilizou-se o teste não-paramétrico de Kruskal-Wallis. A imunopositividade para AR, CRABP2, NF?B p100/52 e NF?B p105/50 foi encontrada tanto no núcleo quanto citoplasma. A imunoexpressão de TRAP1 e TWEAK esteve presente apenas no citoplasma das células cancerosas. Todos os tipos histológicos do nefroblastoma (blastema, epitélio e estroma) foram positivos para as seis proteínas estudadas. AR pela análise morfométrica e CRABP2 pela análise semiquantitativa apresentaram maior expressão imunoistoquímica nos pacientes com metástase (p= 0.0247 e p= 0.0128, respectivamente). Semelhante ao encontrado em relação à análise quantitativa de AR que apresentou maior imunopositividade em amostras tumorais submetidas a quimioterapia pré-cirúrgica (p= 0.0330). Observou-se a maior expressão de NF?B p105/50 em nefroblastomas em estádio I/II (p= 0.0415). NF?B p105/50 e TRAP1 através da análise semiquantitativa e TWEAK de acordo com análise quantitativa, apresentaram menor positividade em nefroblastomas tipo blastematoso (p= 0.046, p= 0.003 e p= 0.066, respectivamente). A anaplasia se associou a maior escore total de Allred de TRAP1 (p= 0.005). Não houve correlação estatística da imunoexpressão dos marcadores estudados, comprometimento nodal e evolução clínica. A análise semiquantitativa e quantitativa dos marcadores AR, CRABP2, NF?B p105/p50, TRAP1 e TWEAK apontam para potenciais biomarcadores para progressão tumoral e para a participação destas proteínas na tumorigênese do nefroblastoma. Palavras-chave: nefroblastoma; ácido retinóico; CRABP2; NF?B; TRAP1; TWEAK. / Abstract: Nephroblastoma, or Wilms' tumor, is the commonest renal cancer in children and accounts for about 98% of renal tumors in this age group. Despite of the favorable prognosis, a parcel of patients evolved to tumor relapse and death. Biomarkers acts in tumor progression, proliferation, differentiation, apoptosis and the most recent described immune and inflammation response action, as retinoic acid (RA), CRABP2, NF?B p100/52, NF?B p105/50, TRAP1 and TWEAK. The aim of this study was to evaluate the immunohistochemical expression of RA, CRABP2, NF?B p100/52, NF?B p105/50, TRAP1 and TWEAK in paraffin-embedded samples of nephroblastomas to determine whether expression correlates with prognostic factors. A total of 77 specimens from pathology services were selected. The immunoexpression was evaluated through semiquantitative and quantitative analysis. For the semiquantitative analysis, the Allred score system was used and for the quantitative evaluation, the morphometric analysis of the stained area was applied. For the comparison of two groups, the non-parametric test of Mann-Whitney was used, while for the comparison of three or more groups the Kruskal-Wallis nonparametric test was used. Immunopositivity to RA, CRABP2, NF?B p100/52 and NF?B p105/50 was found in the nucleus and cytoplasm. Immunoexpression of TRAP1 and TWEAK was present only in the cytoplasm of cancer cells. All histological types of nephroblastoma (blastema, epithelium and stroma) were positive for the six proteins studied. RA morphometric analysis and CRABP2 semiquantitative analysis presented higher immunohistochemical expression in patients with metastasis (p= 0.0247 and p= 0.0128, respectively). This is similar to the results of the quantitative analysis of RA, which showed greater immunopositivity in tumor samples of patients subjected to pre-surgical chemotherapy (p= 0.0330). The highest expression of NF?B p105/50 was observed in stage I/II nephroblastomas (p= 0.0415). NF?B p105/50 and TRAP1 semiquantitative analisys and TWEAK quantitative analisys presented lower positivity in nephroblastomas blastematal type (p= 0.046, p= 0.003 and p= 0.066, respectively). Anaplasia were associated with high TRAP1 total Allred score (P= 0.005). No significant correlation was found between the markers and variables studied, such as nodal involvement, and clinical evolution. The semiquantitative and quantitative analysis of RA, CRABP2, NF?B p100/p52, NF?B p105/p50, TRAP1 and TWEAK point to potential biomarkers for tumor progression and for the participation of these proteins in nephroblastoma tumorigenesis. Helmintos parasitos de mocós (Kerodon rupestris Wied, 1820), de vida livre e de cativeiro, criados no semi-árido nordestino

Poucas pesquisas foram realizadas com essa espécie, principalmente as relacionadas aos seus parasitos. Dessa forma, este trabalho objetivou conhecer a fauna helmintológica de mocós criados na região semi-árida do Rio Grande do Norte. Foram utilizados 16 animais (sete machos e nove fêmeas), sendo sete jovens e nove adultos. Nove animais foram capturados no seu habitat natural e sete procedentes do cativeiro localizado no Centro de Multiplicação de Animais Silvestres da Escola Superior de Agricultura de Mossoró. Após o sacrifício, os animais foram necropsiados e o conteúdo resultante de cada segmento do trato digestório foi fixado e envasado em frascos individuais, identificados e enviados ao Laboratório de Helmintologia da Faculdade de Ciências Agrárias e Veterinárias da Universidade Estadual Paulista - Câmpus de Jaboticabal. Ainda foram observados quanto à presença de parasitos a traquéia, coração, pulmão, fígado e rins. Para classificação dos helmintos seguiu-se a metodologia descrita na literatura, além de cortes histológicos para o estudo da sínlofe. Dos 16 animais necropsiados, foram coletados 113 helmintos, sendo cinco espécies de nematódeos (Paraspidodera uncinata, Trichostrongylus colubriformis, Trichuris gracilis, Trichuris muris e Vianella lenti) e uma de cestódeo (Thysanotaenia congolensis), sendo o T. colubriformis o nematódeo mais prevalente. Não houve relação significativa entre os parasitos e a idade do hospedeiro (p>0,05). Em relação à carga parasitária e espécies de helmintos os animais de vida livre estavam mais parasitados. / The brazilian rodent rock cavy (Kerodon rupestris) belongs to the Family Caviidae. Only a few researches were accomplished with this species, and just a little is known about its parasites. This paper aims to study the helminthfauna of free ranging and captivity Kerodon rupestris growth in the semi-arid area of Rio Grande do Norte state, Brazil. Sixteen animals, (seven male and nine females), seven immature and nine adults, were studied. The nine free-ranging animals were caught in their natural habitat, and the seven captivity animals were procedent from the Centro de Multiplicação de Animais Silvestres da Escola Superior de Agricultura de Mossoró, Rio Grande do Norte State, Brazil. These animals were necropsied and the gastrointestinal content of each segment was fixed and bottled in individual flasks and sent to the Laboratório de Helmintologia da Faculdade de Ciências Agrárias e Veterinárias da Universidade Estadual Paulista - Câmpus de Jaboticabal. Also, trachea, heart, lung and liver were examined in search of worms. The classification of the helminths was based in methodology found in literature and in histological sections for synlophe study. From the sixteen animals necropsied, were collected 113 helminths, five nematode species (Paraspidodera uncinata, Trichostrongylus colubriformis, Trichuris gracilis, Trichuris muris and Vianella lenti) and one cestode (Thysanotaenia congolensis). T. colubriformis was the most prevalent species. There was not statistical relationship between the parasites and the age of the host (p>0,05). Free-ranging animals were more intensely parasitized than the captive ones, as much for number of worms as for diversity of species.

Caracterização da interação entre a proteína prion e a vimentina

Defesa: Curitiba, 21/03/2017 / Inclui referências : f. 93-97 / Resumo: A proteína prion celular, ou PrPC, é uma proteína ancorada externamente à membrana plasmática por uma molécula de glicofosfatidilinositol. Sua conversão de uma forma fisiológica para uma isoforma mal dobrada e infecciosa, denominada scrapie, é responsável pelo aparecimento das doenças priônicas, também conhecidas como Encefalopatias Espongiformes Transmissíveis. Para elucidar as funções fisiológicas de PrPC, são realizados estudos para se descobrir seus possíveis parceiros moleculares. Recentemente, resultados obtidos por nosso grupo identificaram dez possíveis ligantes para PrPC, dentre os quais destaca-se a vimentina, uma proteína de filamento intermediário. Em células com quantidade excessiva de proteína mal dobrada ou com inibição de proteassoma, há a formação de agressomos, estruturas pericentriolares com agregados de proteínas em seu centro e envoltos por uma gaiola de filamentos intermediários compostos principalmente por vimentina. Para esmiuçar a relação entre as proteínas PrPC e vimentina, verificar a formação de agressomos e suas possíveis consequências para as células, fizemos experimentos de Pull-Down, imunofluorescência, Western Blot, solubilidade em metanol e MTT em células HeLa e N2a. Foram testados tratamentos com DMSO, MG132 e Ciclosporina A (CsA), e transfecções com plasmídeos expressando GFP, GFP-PrPC, GFP-PrPcy, e mutantes de PrPC P104L e V179I. A interação entre as proteínas prion celular e vimentina pôde ser confirmada in vitro pela técnica de Pull-Down nas células HeLa. Inesperadamente, a viabilidade celular de HeLa e N2a não pareceu ser afetada por nenhum dos tratamentos testados. Nas imunofluorescências com ambas as células foi possível observar a formação de agressomos, principalmente nos tratamentos com MG132 e CsA, sendo o primeiro um inibidor de proteassoma e o segundo um inibidor da via das ciclofilinas. A formação desses agressomos parece estar ligada a uma redução na expressão de vimentina de ambas às células, e a um aumento da insolubilidade da proteína prion nas células N2a, mas não nas células HeLa. Scrapie. / Abstract: The cellular prion protein, or PrPC, is a protein anchored to the external leaflet of the plasma membrana by a glycophosphatidylinositol molecule. Its conversion from a physiological isoform to a misfolded and infeccious one, named scrapie, is responsible for the prionic diseases, also known as Transmissible Spongiform Encephalopathies. To elucidate the physiological functions of PrPC, studies are conducted to discover its possible molecular partners. Recently, studies done by our group found ten possible ligants for PrPC, among of them is the vimentin, a intermediate filament protein. In cells showing high levels of misfolded proteins or with proteasome inhibition, there is the formation of aggresomes, pericentriolar structures with protein aggregates on its center wrapped by a cage of intermediate filaments composed mostly by vimentin. To deeply investigate the relation between PrPC and vimentin proteins, to verify the formation of aggresomes and its possible consequences to the cells, we did Pull-Down, immunofluorescence, Western Blot, methanol solubility and MTT experiments in HeLa and N2a cells. We tested treatments with DMSO, MG132 and Cyclosporine A (CsA), and transfections with plasmids expressing GFP, GFP-PrPC, GFP-PrPcy, and PrPC mutants P104L and V179I. The interaction between cellular prion protein and vimentin could be confirmed in vitro by the Pull-Down technique in HeLa cells. Unexpectedly, the HeLa and N2a cell viability was not affected by none of the tested treatments. Immunofluorescence with both cells showed the formation of aggresomes, mainly in MG132 and CsA treatments, the first being a proteasome inhibitor and the second a cyclophilins route inhibitor. The formation of these aggresomes seems to be linked to a reduction in vimentin expression in both cells, and to an increase in insolubility of prion protein in N2a cells, but not in HeLa cells. Caracterização dos mecanismos de resistência ao sistema complemento humano e microvesiculação em diferentes cepas de Trypanosoma cruzi

Considerada como uma doença tropical negligenciada, esta doença acomete milhões de pessoas do mundo e atinge também mamíferos silvestres e domésticos. Para produzir uma infecção o parasito T. cruzi precisa evadir o sistema imune inato do hospedeiro. O sistema imune inato é a primeira linha de defesa contra patógenos; e o sistema complemento representa o principal mecanismo da defesa inata. O sistema complemento é formado por um conjunto de proteínas que são ativadas em cascata e, que culmina na formação de um poro na membrana do patógeno, provocando a sua lise. Todavia, muitos organismos patogênicos como o T. cruzi têm desenvolvido vários mecanismos para escapar o ataque do sistema complemento afin de progredir no seu ciclo de vida, e portanto estabelecer a infecção. Recentemente, foi descrito um novo mecanismo de evasão empregado por patógenos, que é a liberação de microvesículas. Microvesículas são estruturas bicamadas lipídicas complexas (0.1-1 ?m), oriundo da brotamento e fissão da membrana plasmática e que transportam várias moléculas derivadas das células de origem tais como proteínas e ácidos nucleicos. Nosso grupo de pesquisa recentemente descreveu alguns aspectos das características e papel das microvesículas nos mecanismos de evasão do sistema imune inata e durante os eventos de infecção, utilizando como modelo as microvesículas liberadas durante a interação de apenas uma cepa de T. cruzi com células hospedeiras. Desse modo, o presente trabalho teve como objetivo analisar microvesiculação em diferentes cepas de T. cruzi durante a interação com células hospedeiras e availar seu efeito. Apresentamos evidências de que a liberação de microvesículas é induzida durante a interação entre tripomastigotas metacíclicos de diferentes cepas de T. cruzi, G (TcI) e Y (TcII), e células sanguíneas (monócitos) e que existem diferenças na produção de microvesículas entre as diferentes cepas. Os nossos resultados também demostram o envolvimento das microvesículas isoladas nos mecanismos de resistência de T. cruzi à lise mediada pelo complemento e os mecanismos de invasão. A adição de microvesículas a 6,25% de soro humano normal aumentou a resistência dos parasitos da cepa Y à lise mediada por complemento de 50% para 80%; este fenômeno foi mais pronunciado em parasitos da cepa G, pois este aumento quase dobrou. Em termos do papel das microvesículas na infectividade de parasito, verificamos que as microvesículas induziram um aumento de invasão celular pelo parasito. Na presença de microvesículas, o número de parasitos da cepa Y invadindo células mamíferas aumentou por aproximadamente 50%, entretanto mais uma vez este fenômeno foi mais evidente para parasitos da cepa G, o número de parasitos que penetraram as células mamíferas duplicou. Curiosamente, foi observado que o papel das microvesículas na resistência à lise do complemento e na invasão é de classe específica, uma vez que as microvesículas de uma classe não tinha efeito sobre os parasitos de outra classe. Isto indica que as cepas têm mecanismos de inibição independentes e que os componentes de microvesículas poderiam ser diferentes. Finalmente foi comprovado que as microvesículas carreiam proteínas parasitárias envolvidas na interação parasitohospedeiro e que soro de pacientes chagásicos possuem anticorpos que reagem com componentes das microvesículas. Isto fornece evidências de que as microvesículas poderiam ser utilizadas por T. cruzi para transferir fatores de virulência a células hospedeiras e do potencial das mesmas como biomarcador para o diagnóstico da doença. Com esse trabalho foi possível demostrar novos aspectos da relevância de microvesículas durante a interação de diferentes cepas de T. cruzi com células hospedeiras. Palavras-chave: microvesículas. sistema complemento. invasão. / Abstract: The protozoan parasite Trypanosoma cruzi is the etiologic agent of Chagas disease. Considered a neglected tropical disease, this disease affects millions of people worldwide as well as wild and domestic mammals. In order to produce an infection, the T. cruzi parasite has to evade the host's innate immune system. The innate immune system is the first line of defense against pathogens; and the complement system represents the main mechanism of innate defense. The complement system is composed of a set of proteins that are activated in cascade, culminating in the formation of a pore in the membrane of the pathogen, resulting in its lysis. However, many pathogenic organisms such as T. cruzi have developed several mechanisms to escape the attack of the complement system, in order to progress in their life cycle, and thus establish an infection. Recently, a new evasion mechanism used by pathogens has been described, this is, the release of microvesicles. Microvesicles are complex lipid bilayer structures (0.1-1 ?m), originating from the outward blebbing and fission of plasma membrane, and which carries molecules derived from the parent cell such as proteins and nucleic acids. Our research group recently described some aspects of the characteristics and role of microvesicles as a mechanism of evasion of the innate immune system and during the infection process, using as model, the microvesicles released during the interaction of only one strain of T. cruzi with host cells. Accordingly, the objective of the present work was to analyse microvesiculation during interaction of different T. cruzi strains with host cells and to evaluate their effect. We present evidence that microvesicle release is induced during the interaction between metacyclic trypomastigotes from different T. cruzi strains, G (TcI) and Y (TcII), and blood cells (monocytes). Our results also demonstrate the involvement of microvesicles, isolated from T. cruzi and host cells interaction, in parasite resistance to complementmediated lysis and invasion mechanisms. Addition of microvesicles to 6,25% normal human serum increased parasite resistence to complement-mediated lysis from 50% to 80% in parasites of Y strain; this inhibition was more pronounced in G parasites since this increase was almost doubled. In terms of the role of microvesicles in parasite infectivity, we verified that the microvesicles induced an increase in cellular invasion by the parasite. In the presence of microvesicles, the number of parasites of the Y strain invading mammalian cells increased by approximately 50%, however once again this phenomenon was more evident for parasites of G strain, the number of parasites that penetrated the mammalian cells duplicated. Interestingly, it was observed that the role of microvesicles in resistance to complement lysis and invasion is class-specific, since microvesicles of one class had no effect on the parasites of the other class. This indicates that the strains have independent inhibition mechanisms and that the components of microvesicles could be different. Lastly, it was demonstrated that microvesicles carry parasite proteins involved in the parasite-host cell interaction and that sera from chagasic patients contain antibodies that react with microvesicular components. This provides evidence that microvesicles could be used by T. cruzi to transfer virulence factors to host cells and their potential application as a biomarker for the diagnosis of this disease. With this work, it was possible to demonstrate new aspects of the relevance of microvesicles released during the interaction of different T. cruzi strains with host cells. Isolamento e bioprospecção de actinomicetos endofíticos de Vochysia divergens Pohl

Entre estas, a planta Vochysia divergens, conhecida como Cambará, é utilizada em tratamentos de resfriados, tosse, febre e pneumonia, e se mostra promissora para a prospecção de microrganismos. Dos quais os endófitos isolados de plantas medicinais têm sido associados a uma ampla gama de metabólitos secundários com atividade biológica. Neste contexto, o presente trabalho objetivou o isolamento e a bioprospecção de actinomicetos endofíticos da planta medicinal V. divergens. Um total de 10 isolados obtidos foram identificados por meio do sequenciamento parcial do gene 16S rRNA e análise morfológica, como Aeromicrobium ponti, Williamsia serinedens, Streptomyces thermocarboxydus, Microbacterium sp., Sphaerisporangium sp., Micrococcus sp., Microbispora sp. e Actinomadura sp.. Afim de selecionar a melhor condição de cultivo para produção de metabólitos secundários, foram avaliadas duas condições diferentes de meio de cultura e temperatura. Todas as condições utilizadas resultaram na produção de metabólitos com atividade biológica, porém os melhores resultados foram no cultivo em caldo SG (Soja e glucose) a 36°C. Nestas condições, o extrato a partir da fermentação da linhagem Aeromicrobium ponti (LGMB491) apresentou atividade contra Staphylococcus aureus sensível à meticilina (MSSA) e S. aureus resistente à meticilina (MRSA), com CIM de 0,02mg/mL e 0,04mg/mL, respectivamente. O extrato oriundo da linhagem Microbacterium sp. (LGMB471) apresentou inibição total do crescimento micelial do fitopatógeno Phyllosticta citricarpa com CIM de 0,16mg/mL. Neste estudo, o conhecimento da comunidade endofítica da planta medicinal V. divergens foi ampliado por meio do isolamento de novos gêneros associados a esta planta, alguns deles isolados pela primeira vez como endófitos. MRSA. / Abstract: The Pantanal is a periodic flood plain with a unique biodiversity, and has several plants with traditional knowledge associated with its medicinal properties. The plant Vochysia divergens, known as Cambará, is used for treatment of colds, cough, fever and pneumonia, and is very promising for microorganisms' prospection. Endophytic microorganisms isolated from medicinal plants have been studied due to the production of a wide range of secondary metabolites with biological activity. In this context, the present work aimed at the isolation and bioprospection of endophytic actinomycetes from medicinal plant V. divergens. Ten strains of endophytic actinomycetes were identified by partial 16S rRNA gene sequencing and morphological analysis, as belonging to Aeromicrobium ponti, Williamsia serinedens, Streptomyces thermocarboxydus, Microbacterium sp., Sphaerisporangium sp., Micrococcus sp., Microbispora sp. and Actinomadura sp. In order to select the best culture conditions for production of secondary metabolites, two different culture media and two temperatures were evaluated. All the conditions used resulted in the production of metabolites with biological activity, but the best conditions are cultivation in SG broth at 36°C. In these conditions, the extract from the fermentation of Aeromicrobium ponti (LGMB491) showed high activity against methicillin-sensitive Staphylococcus aureus (MSSA) and methicillin-resistant S. aureus (MRSA), with MIC of 0.02mg/mL and 0.04mg/mL, respectively. The extract from the Microbacterium sp. (LGMB471) showed total inhibition of the mycelial growth of phytopathogen Phyllosticta citricarpa with MIC of 0.16mg/mL. In this study, we broadened the knowledge about the endophytic community of the medicinal plant V. divergens through the isolation of rare actinomycetes, some of them isolated for the first time as endophytes. Eficácia dos fungos nematófagos Duddingtonia flagrans e Arthrootrys robusta na profilaxa das infecções naturais por nematódeos gastrointestinais em ovinos /

Resumo: Um dos maiores problemas enfrentados pelos criadores de ovinos são as helmintoses gastrintestinais. O uso indiscriminado de anti-helmínticos teve como conseqüência a seleção de populações de helmintos resistentes. A utilização de fungos nematófagos predadores como controle biológico é uma das alternativas pesquisadas para auxiliar na profilaxia das helmintoses. O objetivo do trabalho foi avaliar a eficácia de fungos nematófagos e a administração de diferentes dosagens na profilaxia das nematodioses gastrintestinais em ovinos naturalmente infectados em Botucatu - SP. O experimento foi dividido em duas etapas, nas quais primeiramente avaliou-se o fungo mais eficaz (Duddingtonia flagrans e Arthrobotrys robusta) e posteriormente duas dosagens do fungo A. robusta. As variáveis avaliadas foram: peso, volume globular (VG), ovos de estrongilídeos por grama de fezes (OPG), coproculturas e larvas infectantes (L3) na pastagem. Não houve diferença significativa no peso médio e VG entre os grupos estudados em ambos experimentos. O número médio de OPG não diferiu significativamente entre os grupos na maioria das colheitas, entretanto o grupo que recebeu o fungo A. robusta (Experimento I) foi o que recebeu menor quantidade de tratamentos com anti-helmínticos, logo foi o fungo utilizado para o Experimento II. Neste houve redução significativa no número de L3 de Trichostrongylus spp. por quilo de matéria seca de forragem nos piquetes pastejados pelo grupo tratado diariamente com fungo A. robusta. Haemonchus contortus foi a espécie predominante em cordeiros traçadores utilizados em ambos os experimentos. Nas condições experimentais do estudo o controle biológico não foi eficaz na profilaxia das helmintoses. / Abstract: Parasitic gastroenteritis is one of the major problems confronted of the sheep industry. The indiscriminate use of anthelmintics has had as consequence the resistant selection of helminth population. The use of nematophagous fungi as biologic control is one of the alternatives strategies to gastrointestinal nematode infection prophylaxis. The aim of this work was to evaluate the efficacy of the nematophagous fungi and its use in different doses to the prophylaxis of natural infection by gastrointestinal nematodes in sheep kept on pasture in Botucatu - SP, Brazil. The experiment was carried out in two phases: in the first trial, the comparative efficacy of Duddingtonia flagrans and Arthrobotrys robusta fungi was evaluated and in the second, two different strategies of administration of A. robusta fungi. The measurements were: body weight, packed cell volume (PCV), faecal egg counts (FEC), and numbers of third stage larvae (L3) on pasture. There were no significant differences regarding body weight and PCV means between the studied groups of both experiments. The mean FEC did not differ significantly between the groups at the most of the sample collections, however, the daily feed A. robusta group (Experiment I) received the smaller quantity of anthelmintics treatments, so it was the fungi used in the Experiment II. Trichostrongylus spp. L3 numbers per kilogram of dry matter was significantly lower on pasture grazed by the daily treated group with A. robusta fungi. Haemonchus contortus was the predominant species recovered from the tracer lambs in both experiments. Both species are rodent’s parasites and the human infection is considered incidental. On humans, A. cantonensis cause eosinophilic meningitis and A. costaricensis cause abdominal angiostrongiliase. Since there is no elimination of parasitic forms in human infection, diagnosis becomes difficult and molecular methods are necessary. After years of discovery of angiostrongilíases, there are still many efforts in the study and development of a specific and sensitive diagnostic test capable of discriminating Angiostrongyliases of the other parasites. In this context, cross-reactivity becomes a problem in specificity of serologic tests. The main objective of this study was to analyze the sharing of antigens between Angiostrongylus spp. and parasites Strongyloides spp. , Fasciola hepatica, Ascaris lumbricoides, Hymenolepis diminuta and Toxocara canis, and identify these molecules shared.To obtain antigens, some rats were captures and their feces analyzed by the method of Baerman addition to being seeded Agar Plates. Were obtained larvae of Strongyloides spp. and Angiostrongylus spp. In pulmonary arteries were found 11 female worms and 2 male worms of A. cantonensis, which led to the first report of the occurrence of this parasite in Rio Grande do Sul. In the small intestine was obtained a Strongyloides spp worm and analysis of small intestine were obtained Hymenolepis diminuta worms. From the adult worms of parasites were obtained antigens used in this study. Furthermore, we used antigens of allergens such as peanut, tomato, strawberry and pollen. To identify shared antigens, proteins were separated by one and two dimensional electrophoresis technique assayed by Western blot using sera of patients with angiostrongyliases. All antigens were recognized by sera from patients infected with angiostrongyliases, being a total of fourteen bands identified as being immunogenic.The fourteen bands were cut out of polyacrylamide gels and analyzed by mass spectrometry. The proteins identified were: Phosphoenolpyruvate carboxykinase, heat shock protein 70 (HSP70), DAPPUDRAFT hypothetical protein, 60 kDa Chaperonin 5, Glyceraldehyde-3-phosphate dehydrogenase, sigma class GST Chain A, Glucose-6-phosphate isomerase, pyruvate dehydrogenase subunit E1, Glutamate dehydrogenase 2, arachin Ahy-1, Full = Allergen Ara h 1, clone P41B, Gly1. The data generated in this study show that there is sharing of antigens between organisms of different taxonomic groups, but also with allergens tested. Moreover, the description and analysis of shared molecular components can help in understanding the evolutionary history and phylogeny of these organisms. / Duas espécies de parasitos nematódeos da família Angiostrongylidae, do gênero Angiostrongylus de localização intra-arterial são capazes de produzir doença em humanos: Angiostrongylus costaricensis e Angiostrongylus cantonensis. Ambas as espécies são parasitos próprios de roedores e a infecção humana é considerada acidental. No homem, A. cantonensis é o causador da meningite eosinofílica e A. costaricensis causador da angiostrongilíase abdominal. Como não há eliminação de formas parasitárias na infecção humana, o diagnóstico se torna difícil e métodos moleculares se tornam necessários. Depois de anos da descoberta das angiostrongilíases, ainda há muitos esforços no estudo e desenvolvimento de um teste de diagnóstico específico e sensível capaz de discriminar as Angiostrongilíases de outras parasitoses. Neste contexto, a reatividade cruzada se torna um problema na especificidade de testes sorológicos. O objetivo principal deste trabalho foi analisar o compartilhamento de antígenos entre Angiostrongylus spp. e os parasitos Strongyloides spp. , Fasciola hepatica, Ascaris lumbricoides, Hymenolepis diminuta e Toxocara canis, assim como identificar essas moléculas compartilhadas.Para obtenção de antígenos, algumas ratazanas foram capturas e suas fezes analisadas através do método de Baerman além de terem sido semeadas em Placas de Ágar. Foram obtidas larvas de Strongyloides spp. e Angiostrongylus spp. Nas artérias pulmonares de uma ratazana foram encontrados 11 vermes fêmeas e 2 vermes machos de A. cantonensis, que deu origem ao primeiro relato da ocorrência deste parasito no Rio Grande do Sul. No intestino delgado foi obtido um verme de Strongyloides spp e da análise do intestino delgado foram obtidos vermes de Hymenolepis diminuta. A partir dos vermes adultos dos parasitos, foram obtidos os antígenos utilizados neste trabalho. Além disso, foram utilizados antígenos de alergenos como o amendoim, tomate, pólen e morango. Para identificação dos antígenos compartilhados, as proteínas foram separadas por eletroforese uni e bidimensional ensaiadas pela técnica de Western-Blot utilizando-se soro de pacientes com angiostrongilíases.Todos os antígenos foram reconhecidos pelo soro de pacientes infectados com angiostrongilíases, sendo um total de quatorze bandas identificadas como imunogênicas. As quartorze bandas foram recortadas dos géis de poliacrilamida e analisadas por espectrometria de massas. As proteínas identificadas foram: Fosfoenolpiruvato carboxiquinase, Proteína de choque térmico 70 (HSP70), Proteína hipotética DAPPUDRAFT, 60 kDa Chaperonina 5, Gliceraldeído-3-fosfato-desidrogenase, Cadeia A sigma classe GST, Glucose-6-fosfato isomerase, Piruvato desidrogenase subunidade E1, Glutamato desidrogenase 2 , arachin Ahy-1, Full=Allergen Ara h 1, clone P41B , Gly1. Os dados gerados no presente trabalho demonstram que há compartilhamento de antígenos entre organismos de diferentes grupos taxonômicos, como também com os alergenos testados. Além disso, a descrição e a análise de componentes moleculares compartilhados podem ajudar na compreensão da história evolutiva e da filogenia destes organismos.

Estudo das proteínas HSP70, SOD e catalase de Angiostrongylus cantonensis em hospedeiros habituais e acidentais

In humans, they can cause eosinophilic meningitis. Although the parasite have been described in 1935, little is known about the mechanisms of adaptation to its hosts. A. cantonensis owns molecules that help the parasite in the evasion of the host´s immune system, among these are SOD, catalase and HSP70. The main goal of this work was to study the profiles of HSP70, SOD and CAT of L5 produced in different hosts and analyze the diagnostic potential of the extracts. For the obtainment of fifth stage larvae (L5), rats and mice were infeceted with 104 and 50 third stage larvae, respectively, and euthanized after 21 days. The L5 were recovered from the brain of the animals and homogenized in different buffers to obtain the protein extracts. The quantification of the total protein suggests no significant difference between extracts. For the identification of HSP70, it was performed dot blot and Western blot, showing that in both samples HSP70 were present. ELISA assays were used for the quantification of HSP70 and analyze antigen-antibody binding with from individuals with confirmed diagnosis for angiostrongyliasis. L5 from usual hosts were recognized by the sera showing superior titles to the L5 from the accidental hosts. Levels of stress proteins HSP70, SOD (339U versus 0. 08U of SOD/mg of protein) and catalase (184. 4 versus 75. 6 pmol of CAT mg-1 proteins) were higher p<0. 05, in A. cantonensis larvae from usual hosts in comparison to the larvae obtained from accidental hosts. These data suggest that the levels of stress proteins may be associated to the adaptation of A. cantonensis to the usual hosts. / Angiostrongylus cantonensis é um nematódeo da família Angiostrongylidae. Em humanos, podem causar meningite eosinofílica. Apesar de o parasito ter sido descrito em 1935, pouco se sabe dos mecanismos de adaptação aos seus hospedeiros. A. cantonensis possui moléculas que o auxilia na evasão do sistema imune do hospedeiro, dentre estas estão SOD, catalase e HSP70. O objetivo principal deste trabalho foi estudar o perfil de HSPs 70, SOD e CAT de L5 produzidas em diferentes hospedeiros e analisar o potencial diagnóstico dos extratos. Para a obtenção das larvas de quinto estágio (L5), ratos e camundongos foram infectados com 104 e 50 larvas de terceiro estágio, respectivamente, e eutanasiados após 21 dias. As L5 foram recuperadas do cérebro dos animais e homogeneizadas em diferentes tampões para obtenção dos extratos protéicos.A quantificação de proteínas totais sugere não haver diferenças significativas entre os extratos. Para a identificação de HSP70 foram realizados dot blot e Western blot, mostrando que em ambas as amostras HSP70 estavam presentes. Ensaios de ELISA foram utilizados para a quantificação de HPS70 e para analisar a ligação antígeno anticorpo com soros de indíviduos com diagnóstico confirmado para a angiostrongiliase, sendo que L5 provenientes de hospedeiros habituais apresentam reconhecimento pelos soros com títulos superiores às L5 provenientes de hospedeiros habituais. Os níveis das proteínas de estresse HSP70, SOD (339U versus 0,08U de SOD/mg de proteína) e catalase (184,4 versus 75,6 pmole de CAT mg-1 proteínas) foram superiores p<0,05, em larvas de A. cantonensis provenientes de hospedeiros habituais em comparação com as larvas obtidas de hospedeiros acidentais. Estes dados sugerem que os níveis de proteínas de estresse podem estar associados a adaptação do A. cantonensis aos hospedeiros habituais.
