Couplage entre introduction et réparation des cassures double brin pendant les réarrangements programmés du génome de Paramecium tetraurelia / Ku-mediated coupling of DSB introduction and repair during programmed genome rearrangements in Paramecium tetraurelia

L’élimination programmée d’ADN spécifique de la lignée germinale pour former un nouveau noyau somatique a été décrite chez les eucaryotes. Ces réarrangements sont initiés par l’introduction de cassures double brin (CDB) de l’ADN et la préservation de l’intégrité du génome requiert une réparation efficace. Chez Paramecium tetraurelia, le génome est largement réarrangé pendant le développement du nouveau noyau somatique, après l’introduction de milliers de cassures double brin programmées par la transposase domestiquée PiggyMac (Pgm)Ces réarrangements consistent en l’excision précise de dizaines de milliers de séquences uniques et non codantes (IES) qui interrompent 47% des gènes dans la lignée germinale ; et l’élimination hétérogène de séquences répétées qui mène à des délétions internes de taille variable ou à la fragmentation des chromosomes avec addition de télomères aux extrémités.L’implication de la voie du Non Homologous End Joining (NHEJ) dans l’excision précise des IES a été prouvée. Dans des cellules déplétées de Ligase IV ou XRCC4, les cassures aux bornes des IES sont introduites normalement mais il n’y a pas de jonctions d’excision formées et les extrémités cassées s’accumulent sans être dégradées. Mais la voie de réparation impliquée dans les réarrangements imprécis est encore inconnue. L’hypothèse d’une réparation par la voie NHEJ alternative (alt-NHEJ), indépendante de Ku et impliquant la résection des extrémités et l’utilisation de microhomologie, a été émise. C’est pourquoi pendant ma thèse je me suis intéressé à ma thèse au rôle des protéines Ku.Deux gènes KU70 et trois gènes KU80 ont été identifiés dans le génome de la paramécie. KU70a et KU80c sont spécifiquement induits pendant les réarrangements programmés du génome et les protéines localisent dans les noyaux somatiques en développement. Des expériences d’extinction de ces gènes par ARN interférence ont prouvé que ces gènes étaient indispensables. Au niveau moléculaire, l’ADN non réarrangé est amplifié dans les cellules déplétées de Ku. De plus, les cassures double brin programmées ne sont pas introduites aux bornes des IES.Mes résultats suggèrent que Ku fait partie d’un complexe de pré-excision, avec la transposase domestiquée Pgm, et est nécessaire pour l’introduction des cassures double brin programmées pendant les réarrangements programmés du génome. / Programmed elimination of germline specific DNA has been described in several eukaryotic organisms. These rearrangements are initiated through introduction of DNA double strand breaks (DSB). To ensure genome integrity, efficient repair is needed. In Paramecium tetraurelia, the genome is widely rearranged during development of a new somatic nucleus after introduction of tens of thousands of DSBs by the domesticated transposase PiggyMac (Pgm)These rearrangements consist in: the precise excision of thousands of unique and non coding sequences called IESs that interrupt 47% of genes in the germline; and the heterogeneous elimination of repeated sequences. It leads to internal deletions of variable sizes or to chromosome fragmentation with telomere addition at DNA ends.Implication of the Non Homologous End Joining Pathway (NHEJ) in precise IES excision has been proved. In cells depleted for Ligase IV or XRCC4, DSBs at IES boundaries are introduced normally but broken DNA ends accumulate without being repaired nor degraded. The repair pathway implicated in heterogeneous rearrangements is still unknown. An hypothesis would be that heterogeneous rearrangements involve a Ku independent alternative NHEJ (alt-NHEJ) pathway characterized by end resection and use of microhomologies. During my thesis I studied the role of Ku proteins in programmed genome rearrangements.Two KU70 genes and three KU80 genes has been identified in the Paramecium genome. KU70a and KU80c are specifically induced during programmed genome rearrangements. Encoded proteins localize in developing somatic nuclei. Gene extinction by RNA interference experiments proved that these genes are necessary for programmed genome rearrangements. At molecular level, non rearranged DNA is amplified in cells depleted for Ku. And more surprisingly, no programmed DSBs are introduced at IES boundaries in these cells.My results indicate that Ku is a part of a pre excision complex with the domesticated transposase Pgm and necessary for the introduction of programmed DSB during programmed genome rearrangements.

Debating the efficacy transitional justice mechanisms : the case of national healing in Zimbabwe, 1980-2011

This study is an exploration of transitional justice mechanisms available to post conflict communities. It is a context sensitive and sustained interrogation of the effectiveness of endogenous transitional justice mechanisms in post-colonial Zimbabwe. The study utilised Ruti Teitel’s (1997: 2009-2080) realist/idealist theory as its theoretical framework. Using the case of Africa in general and Zimbabwe in particular, it analyses the application of imported idealist transitional justice mechanisms, mainly International Criminal Court (ICC) trials. It also debates the efficacy of realist transitional justice mechanisms, mainly the South African model of a Truth and Reconciliation Commission (TRC).The study explores the application of what it terms broad realist transitional justice mechanisms used mostly in rural areas of Zimbabwe to achieve peace building and reconciliation. These modes of everyday healing and reconciliation include the traditional institutions of ngozi (avenging spirit), botso (self-shaming), chenura (cleansing ceremonies), nhimbe (community working groups) and nyaradzo (memorials). The key finding of this exploration is that local realist transitional justice mechanisms are more efficacious in fostering peace building and reconciliation than imported idealist mechanisms such as the ICC trials and imported realist mechanisms such as the TRC. More value can be realised when imported realist mechanisms and local realist transitional justice mechanisms complement each other. The study contributes to the literature on transitional justice in general and bottom-up, victim-centred reconciliation in particular. It offers a different approach to the study of transitional justice in post conflict Zimbabwe by recasting the debate away from the liberal peace paradigm which critiques state centric top-down approaches such as trials, clemencies, amnesties and institutional reform. The study considers the agency of ‘ordinary’ people in resolving the after effects of politically motivated harm. It also lays the foundation for further research into other traditional transitional justice mechanisms used for peace building and reconciliation elsewhere in Africa / Political Sciences / D. Abstract The topic of this graduation theses is the protection of property as embodied and provided by European Convention of Human Rights and its authorities. The author also pays attention to how the European Court of Human Rights dealt with number of Czech and Slovak complaints concerning the restitution process taking place after the fall of the communism with purpose to rectify at least some of the last regime's wrongs as to the acts of deprivation occurred before the restoration of democracy. Emphasis is primarily put on Strasbourg authorities case law.

The problem is the search around for reflections on new strategies of access to higher education, through the policy of quotas, specifically for the black students . Intend question and analyse the main meanings of a legal process and academic inconclusive, under one of the institutions members of the State System of Higher Education in Goiás: the UEG, by the fact be in the single IES to deploy until now, the achievement of a legal right. Bi-to reconstruct the trajectory of the construction of Law No. 14.832, July 12, 2004 / Goias, their journeys, their problems, their deployment. With the deployment of Law, democratizing up access for blacks in higher education in Goias, leaving them, however, without the Program of Support and Monitoring. Consequently without the financial resources so these social agents could succeed in its path university, it is concluded that, in practice, have not been consolidated effective ways for the exercise and enjoyment of rights established by Law This fact can be observed by the absence of public interventions with the IES belonging the system. The standard is inert, the implementation of the Quota Law in higher education in Goias gave up in a limited way. / O presente estudo é de natureza exploratória com abordagem qualitativa aportada em uma pesquisa empírica, constituída por entrevistas e significativos registros documentais. À luz do método dialético, procurou-se orientar a investigação a partir das categorias filosóficas, essência e aparência, mediatizadas pela lei da contradição. A temática de estudo abarca a configuração do processo histórico de exclusão social e cultural do negro brasileiro no ensino superior. O problema de pesquisa volta-se para as reflexões sobre novas estratégias de acesso à educação superior, por intermédio da política de cotas, especificamente às destinadas aos estudantes negros. Pretendeu-se problematizar e analisar as principais significações de um processo legal e acadêmico inconcluso, no âmbito de uma das instituições integrantes do Sistema Estadual de Educação Superior em Goiás: a UEG, pelo fato de constituir-se na única IES a implantar, até o momento, a conquista legal de um direito. Pretendeu-se reconstruir a trajetória da construção da Lei nº 14.832, de 12 de julho de 2004 / Goiás, seus percursos, seus problemas, sua implantação. Com a da implantação da Lei, democratizou- se o acesso para negros na educação superior em Goiás, deixando-os, contudo, sem o Programa de Apoio e Acompanhamento. Conseqüentemente sem os recursos financeiros, para que esses agentes sociais pudessem ter êxito em sua caminhada universitária, conclui-se que, na prática, não foram consolidadas formas efetivas para o exercício e gozo de direitos estabelecidos por Lei. Tal fato pode ser observado pela ausência de intervenções públicas junto as IES jurisdicionadas ao Sistema. Com o engessamento da norma, a implantação da Lei das Cotas na educação superior em Goiás deu-se de forma limitada.

Réflexions sur les statuts de victime en droit international pénal / Victim’s status in international criminal law

À ce moment charnière où les T.P.I. accomplissent leurs Stratégies d’achèvement et où la C.P.I. va rendre son premier jugement, il est essentiel de faire un point sur le droit international pénal et particulièrement sur les statuts de victime selon ce droit. En effet, ceux-ci symbolisent les tenants et aboutissants de l’ensemble du droit international pénal. Tantôt oubliée, tantôt sacralisée, la victime ne laisse pas indifférent, et encore moins lorsqu’elle a subi un crime international. Sa reconnaissance a d’ailleurs bouleversé l’économie générale du droit international pénal qui lui accorde une place de plus en plus grande. Une étude approfondie des droits des différentes juridictions permet de constater qu’il n’y a pas un mais plusieurs statuts de victime. La pluralité de ces statuts réside dans l’existence de plusieurs qualités juridiques de victime et de plusieurs régimes juridiques applicables à celle-ci. L’analyse des différents facteurs de cette pluralité donne lieu de voir qu’elle est une force et non une faiblesse du droit international pénal. Elle est la voie médiane entre le rejet de la considération des victimes comme une masse informe et leur impossible prise en compte individuelle. Mais, comme pour le statut du particulier en droit international public général, l’équilibre est parfois difficile à trouver et les statuts sont perfectibles. C’est pourquoi, des propositions d’enrichissements des statuts de victime sont faites et, pour chacune d’entre elles, il est observé si elle est réalisable et souhaitable. Un système de justice pénale internationale serait en construction. La victime pourrait finir par trouver une place en son sein qui satisferait l’ensemble des protagonistes de la répression des crimes internationaux, y compris et surtout, les États. / While the ICT carry out their Completion Strategies, the ICC will soon pronounce its first decision. At this point, itis important to assess international criminal law and especially victim’s status according to this law. Actually, theysymbolise the ins and outs of international criminal law. Once forgotten, once sacralised, the victim never leavesus indifferent, above all a victim of international crimes. His or her recognition has deeply altered internationalcriminal law, which now gives him or her an increasing place. Studying the laws of the international criminal courtsshows that there is not one status but several status of victim. This plurality comes from the fact that there areseveral legal characterisations of victim and several legal conditions. The analysis of the different factors leadingto this multiplicity of legal status enables us to envision this legal diversity as a power of international criminal law,rather than one of its weaknesses. It is the middle way between considering victims as a shapeless, undefinedand undifferentiated crowd and considering each victim individually. But, as with the status of private persons ininternational public law, reaching and keeping a well-balanced status is difficult ; and these are perfectible andcan be improved. This is the reason why some suggestions for the enrichment of the different status are madeand, for each of them, there is a thorough questioning and analysis of whether it is feasible and desirable. As aninternational criminal justice system is under construction, the victim could find a place in it which would satisfy allthe protagonists of the legal proceedings against international crimes, including and particularly States.

Les Inhibiteurs de PARP dans le Traitement des Cancers Chimio-Résistants. Etude pré-clinique sur la Dépendance à PARP

Introduction Le cancer bronchique est un problème de santé publique en étant la première cause de décès par cancer dans le monde. Il reste de mauvais pronostic avec une résistance au Cisplatine qui est inéluctable dans l'histoire naturelle de la maladie. Nous nous sommes intéressés à l'association du CDDP aux inhibiteurs de la Poly(ADP-ribose) polymérase. Les inhibiteurs pharmacologiques de PARP sont source d'optimisme en oncologie clinique en monothérapie pour des tumeurs déficientes pour une voie de réparation de l'ADN et en association aux cytotoxiques classiques.Matériel et méthodes Nous avons généré 9 clones résistants au CDDP après culture de la lignée A549 dans des faibles doses de CDDP. Deux inhibiteurs pharmacologiques de PARP, CEP8983 (CEP) et PJ34 (PJ), ainsi que des siRNA spécifiques de PARP1 sont utilisés pour l'inhibition de PARP. L'apoptose est mesurée en cytométrie de flux par l'intermédiaire du potentiel membranaire de la mitochondrie DiOC6(3) et la perméabilisation de la membrane plasmique est évaluée par l'iodide de propidium. Le test de clonogénicité permet d'évaluer la capacité des cellules à échapper à la mort et à former une colonie. L'activité métabolique des cellules est mesurée par la mesure de clivage du sel de tetrazolium WST-1. L'immunofluorescence sur cellules fixées a permis d'étudier les dommages de l'ADN (γH2AX), la voie intrinsèque de l'apoptose (l'activation de la caspase 3 et la libération du cytochrome c) et la recombinaison homologue (BRCA1, RAD51). En Western Blot nous avons mesuré l'expression et l'activité de PARP (PAR) ainsi que l'expression d'acteurs de la réparation par excision de base (BER) (XRCC1 and polymérase β). Nous avons développé une méthode de détection de PAR en immunohistochimie sur des tissus inclus en paraffine. Résultats Nous avons trouvé un effet synergique pour l'association du CDDP aux inhibiteurs de PARP in vitro. De façon inattendue nous avons observé que les clones résistants au CDDP développent une addiction à PARP et sont spécifiquement tués par l'inhibition de PARP contrairement à la lignée parentale. Ces clones exhibent une hyperexpression et une hyperactivité de PARP. La réponse aux inhibiteurs de PARP corrèle plus précisément avec l'activation plutôt qu'avec l'expression de PARP, pointant que PAR est un biomarqueur plus précis que PARP. Nous avons observé que l'hyperactivation de PARP accompagne une résistance induite au CDDP et prédispose à une sensibilité aux inhibiteurs de PARP dans d'autres lignées de cancer bronchique (H460 et H1650), de mésothéliome (P31), de cancer de l'ovaire (TOV112D) et de col (HeLa). Dans des expériences in vivo nous avons noté que dans les xénogreffes obtenues à partir de clones résistants au CDDP, l'expression de PAR est stablement retrouvée en immunohistochimie. Ces tumeurs répondaient à l'inhibition de PARP par le PJ en diminuant l'expression de PAR. Les clones résistants au CDDP sensibilisés aux I PARP ont une recombinaison homologue conservée, cependant ont un déficit dans les étapes terminales du BER.Conclusion Nous avons identifié un effet synergique pour l'association des inhibiteurs de PARP au CDDP de des lignées de cancer bronchique. Nous avons observé une dépendance à PARP dans des lignées de cancer bronchique résistantes au CDDP et déficientes pour l'élongation du BER. Nous postulant que PAR est un biomarqueur spécifique de la réponse aux inhibiteurs de PARP.

On 11 June 2008 the Government of Canada delivered an official apology to former students of Indian residential schools for its participation in the schools’ creation and administration. The morally infused discourses of political apologies may at first seem to symbolize a progressive step towards a better and more egalitarian future. This thesis, however, will challenge and problematize such perspectives by presenting not only a critical analysis of the 2008 apology itself but also by contextualizing the apology’s narratives with the colonial framing strategies which have historically served to marginalize and dominate the Indigenous nations and peoples of Turtle Island. Through the critical exploration of the 2008 apology’s operability and political significance in Canada’s colonial context, this thesis intends to reveal both the message(s) that the apology got across to the Canadian general public and the forms of domination and political distraction that the apology’s seemingly moral and progressive narratives effectively belie.

The moderation function of in-group status position on the relationship between group-based guilt and reparation intention was tested in a 2 (group-based guilt: low versus high) x 2 (status loss: weak versus strong) factorial between-subjects design, using an online survey software program named Qualtrics. The target population was white South African undergraduate students born after 1988 and registered at the University of South Africa. The results of the first Experiment confirmed the hypothesis, that the relationship between group-based guilt and reparation intention becomes less significant the more participants perceive a loss of status for their in-group. However, the hypothesis could not be confirmed in Experiment 2. The results are presented and discussed in detail / Psychology / M.A. (Psychology)
