Molecular Characterization of Early Dedifferentiation in Newt Forelimb Regeneration

Newts have the incredible ability to regenerate many different organs and tissues as adults, including the limbs. Limb regeneration occurs via the dedifferentiation of stump tissue and the formation of a blastema, which provides the majority of cells for the regenerate. Despite all that we have learned about dedifferentiation and blastema formation, the cellular and molecular mechanisms underlying these processes are still poorly understood. We used representational difference analysis (RDA) to identify genes involved in the early dedifferentiation process in newt forelimb regeneration. Our analysis identified approximately 410 unique genes that were differentially regulated during this process. Microarray analysis was used to determine the expression profile of these genes throughout limb and tail regeneration. We used quantitative PCR (qPCR) to validate the expression of a subset of these genes [β-catenin, wntless, dapper, thymosin-β 4 (Tβ4), and thymosin-β 10/15 (Tβ10/15)] in regenerating limb and tail tissue, as well as in differentiating newt myoblasts. We also verified the expression of these genes in the regenerating newt limb using immunohistochemistry (IHC) and in situ hybridization (ISH). Finally, we performed a functional analysis on β-catenin, wntless, dapper, and Tβ4 by overexpressing these genes in mouse myoblasts to examine their effects on differentiation and potential roles in dedifferentiation. Quantitative PCR verified the expression of β-catenin, wntless, dapper, and Tβ4 during limb regeneration and IHC/ISH localized the β-catenin and Tβ4 proteins to the blastema during regeneration. Tβ10/15 was shown by qPCR to be expressed in the tail during regeneration. Overexpression of newt β-catenin, wntless, dapper, and Tβ4 in mouse myoblasts showed that each of these genes has an inhibitory effect on the differentiation of myoblasts into myotubes and, therefore, may play a role in promoting or maintaining the dedifferentiated state. Our work has identified a large number of genes with potential roles in regulating the dedifferentiation process during newt forelimb regeneration. We have also laid a framework from which much more work can be done by drawing on the genes we have identified and the microarray data, which indicate ideal follow-up candidates. Our analysis of specific genes has also increased our understanding of the molecular events occurring during the dedifferentiation process in the regenerating newt limb.

O agente etiológico da Febre Maculosa das Montanhas Rochosas (RMSF), conhecida no Brasil como Febre Maculosa Brasileira, é a bactéria Rickettsia rickettsii. Essa bactéria é transmitida ao homem pela picada de diferentes espécies de carrapatos ixodídeos. No Brasil, os vetores são Amblyomma cajennense e A. aureolatum. As taxas de prevalência de R. rickettsii nas populações de carrapatos de áreas endêmicas para RMSF são baixas, em geral abaixo de 1%. Essa baixa prevalência parece estar associada a menores taxas reprodutivas e de sobrevivência de linhagens infectadas, sugerindo que R. rickettsii seja patogênica também para os seus vetores. Infecções experimentais demonstraram que 80-100% dos indivíduos de uma colônia de A. aureolatum mantida em laboratório são infectados por R. rickettsii, enquanto apenas 10-60% de A. cajennense adquirem a bactéria. Esses dados indicam que as respostas dessas duas espécies de carrapatos à infecção sejam diferentes, resultando em diferentes taxas de prevalência da bactéria. Dessa maneira, a caracterização molecular das interações entre carrapatos do gênero Amblyomma e a bactéria R. rickettsii é importante, podendo gerar informações não somente para o esclarecimento acerca dos mecanismos de patogenicidade de R. rickettsii para os carrapatos, mas também para um melhor entendimento dos mecanismos responsáveis pela aparente restringência de A. cajennense à infecção. Assim, os objetivos do presente estudo foram: (i) analisar os efeitos da infecção por R. rickettsii sobre o perfil de expressão gênica de carrapatos A. cajennense por hibridação subtrativa por supressão (SSH), (ii) validar os dados de SSH por reação em cadeia de polimerase quantitativa precedida por transcrição reversa (RT-qPCR) e (iii) caracterizar funcionalmente dois genes com expressão induzida pela infecção por RNA de interferência (RNAi). Após a análise bioinformática dos dados de SSH, 44 sequências únicas foram obtidas, das quais 36 representam genes com expressão induzida e 8 genes com expressão reprimida pela infecção. A indução dos genes codificadores da subunidade I da citocromo c oxidase (COX1), da subunidade IV da NADH desidrogenase, de uma proteína com domínio de inibidor de serina-proteases Kunitz-type (papilina-like), identificados por SSH, e de um peptídeo antimicrobiano (hebraeína), foi confirmada por RT-qPCR. O silenciamento gênico da hebraeína e da papilina-like não teve nenhum efeito na aquisição de R. rickettsii pelo vetor, indicando que, isoladamente, não são responsáveis pela proteção de A. cajennense contra a infecção. Os dados gerados pelo presente estudo abrem perspectivas para que outros genes sejam avaliados quanto ao seu papel na aquisição de R. rickettsii, os quais, no futuro, podem ser considerados como alvos para o desenvolvimento de vacinas. / The etiologic agent of the Rocky Mountain Spotted Fever (RMSF), also known as Brazilian Spotted Fever in Brazil, is the bacterium Rickettsia rickettsii. This rickettsia is transmitted to humans by the bite of various tick species. In Brazil, Amblyomma cajennense and A. aureolatum are known as vectors. The prevalence rates of R. rickettsii infected ticks in RMSF endemic areas are low, oscillating around 1%. These low prevalence rates seems to be associated with lower reproductive and survival rates of infected ticks, suggesting that R. rickettsii is also pathogenic to its vectors. Experimental infections with R. rickettsii have demonstrated that 80 to 100% of A. aureolatum ticks from a laboratory colony acquire this bacterium, whereas only 10 to 60% of A. cajennense ticks become infected. These results indicate that the responses of these two tick species against infection are different, resulting in different prevalence rates of the bacterium. Therefore, the elucidation of the interactions between ticks of the genera Amblyomma and the bacterium R. rickettsii at a molecular level is important to provide information to better understand the mechanisms of pathogenicity of R. rickettsii against ticks as well as for the elucidation of the mechanisms responsible for the apparent refractoriness of A. cajennense against infection. Therefore, the objectives of the current study were: (i) analyze the effets of the infection with R. rickettsii on the gene expression of ticks A. cajennense by suppression subtractive hybridization (SSH), (ii) validate SSH data by reverse transcription quantitative polymerase chain reaction (RT-qPCR), and (iii) functionally characterize two genes induced by infection using RNA interference (RNAi). After bioinformatics analysis of SSH data, 44 unique sequences were obtained, among which 36 represent genes with expression induced and 8 repressed genes by infection. The induction of genes encoding subunit I of cytochrome c oxidase (COX1), the NADH dehydrogenase subunit IV, a protein containing Kunitz-type inhibitor domain (papilin-like), identified by SSH, and an antimicrobial peptide (hebraein), was confirmed by RT-qPCR. The effects of knockdown of hebraein and papilin-like encoding genes had no effect on the acquisition of R. rickettsii by the vector. Data of the current study may be used to evaluate the role of other genes in acquisition of R. rickettsii, which, in the future, may be considered as target for vaccine development.

Desde a década de 80, o nucleopoliedrovírus Anticarsia gemmatalis (AgMNPV) tem sido utilizado no Brasil como agente de controle biológico no combate à lagarta-da-soja, resultando para o país significativos benefícios econômicos e ecológicos. Este vírus envelopado, pertencente à família Baculoviridae, possui DNA circular de fita dupla (132.239 pb) contido em um capsídeo protéico, que pode estar ocluído em uma matriz para-cristalina. Neste trabalho, analisamos a expressão temporal de seus genes em duas linhagens celulares (UFL-AG-286 e IPLB-SF-9), por PCR em tempo real. Outro objetivo foi o estudo do efeito da multiplicação viral na malha gênica celular (GRN), visando analisar a expressão gênica celular diferenciada durante a infecção, através da técnica de hibridização subtrativa. Verificamos que todas as ORFs (exceto ORFs 64 e 83, que provavelmente não codificam a genes) foram expressas, com diferenças significativas entre as linhagens, principalmente em relação ao nível de expressão. Apesar disso, o grupo de genes ligados a replicação apresentou perfil de expressão similar nas duas linhagens, possivelmente por este ser um processo essencial à replicação viral. De uma forma geral, todos os genes apresentaram um perfil de expressão mais precoce do que o relatado na literatura, o que poderia ser tanto devido à replicação precoce do DNA do AgMNPV quanto até mesmo consequência da sensitividade do método utilizado. O agrupamento dos genes por k-means seguiu, em sua maioria, a hora pós-infecção (p.i.) onde a expressão de cada gene foi detectada, o que é coerente com a expressão gênica em cascata de baculovírus. Entretanto, por esta classificação não foi possível predizer função gênica para os genes pouco caracterizados. Em relação ao efeito da infecção do AgMNPV na GRN da UFL-AG-286, observamos que em 20h p.i., uma grande diversidade de genes e funções celulares foram hipo-expressas. / Since the 80s, the Anticarsia gemmatalis nucleopolyhedroviruses (AgMNPV) has been used in Brazil as a biological control agent against the Anticarsia gemmatalis caterpillar in soybean fields, resulting in considerable economic and ecological benefits. This enveloped virus belongs to the Baculoviridae family. It has circular double-stranded DNA (132239 bp) enclosed in a capsid, which can be occluded in a crystalline matrix. In this work we elucidated the temporal gene expression profile of the AgMNPV-2D in two cell lines (UFL-AG-286 and IPLB-SF-9), using a real time PCR. Another objective was to study the effect of viral replication on the cellular gene regulatory network (GRN), in order to analyze the differential cellular gene expression during infection, using subtractive hybridization method. We found that most ORFs (except 64 and 83 ORFs that probably do not encode genes) were expressed, with significant differences between cell lines, mainly in expression intensity. However, the group of genes associated with viral DNA replication had similar expression profile in both lineages, possibly because replication is an essential process for viral multiplication. In general, most genes had earlier expression than reported in the literature, probably due to the early DNA replication in AgMNPV. Moreover, this could be a consequence of the method sensitivity used herein. We clustered genes with the k-means algorithm according to the time pos infection (p.i.) in which each gene expression was first detected and found it to be consistent with the typical cascade of gene expression known for baculovirus. Nonetheless, following this classification, it was not possible to predict gene function for poorly characterized genes. When looking at the impact of viral replication on the host GRN using subtractive hybridization, we found considerable inhibition of cellular transcription at 20h p.i. Furthermore at this time, a large and diverse set of cellular genes and functions were found to be hypo-regulated, indicative of an extensive effect of AgMNPV infection on the UFL-AG-286 GRN.

Le groupe ‘M. mycoides’ constitue une branche phylogénétique homogène des mycoplasmes regroupant 6 taxons pathogènes des ruminants, responsables pour la plupart de maladies inscrites sur la liste de l’OIE. L’identification taxinomique sur laquelle repose le diagnostic reste délicate à cause de réactions antigéniques et génétiques croisées et d’un manque d’universalité intra-taxon des PCR, notamment pour les taxons Mcc, MmmLC et Mbg7. Une approche par hybridation soustractive sélective a été développée pour 1) appréhender les différences moléculaires entre ces 3 taxons ; 2) analyser globalement la diversité au sein du groupe ‘M. mycoides’ et 3) rechercher de nouveaux marqueurs d’intérêt diagnostique. Nos résultats montrent un important partage de séquences entre ces taxons, MmmLC et Mcc étant très polymorphes par rapport à Mbg7, plus homogène et qui représente une sorte de chimère entre les taxons Mcc et MmmSC. Nos données nous ont permis de développer un test PCR spécifique pour Mcc mais la diversité génétique du groupe ‘M. mycoides’ dépasse les frontières entre taxons rendant difficile et peu pertinente l’identification taxinomique. Un typage des souches en fonction de la virulence indépendamment de l’espèce serait l’approche diagnostique alternative. La faisabilité d’une telle approche a été explorée dans le cas du taxon MmmLC mais aucun critère susceptible de différencier les souches issues de foyers de celles issues de portage dans des troupeaux sans antécédent clinique n’a pu être mis en évidence. Ce continuum génétique entre souches, probablement lié à des transferts génétiques horizontaux, imposera à l’avenir une surveillance globalisée des mycoplasmoses / The ‘M. mycoides’ cluster, a homogenous phylogenetic branch of the Mollicutes, includes 6 taxa which are responsible for diseases in ruminants, most of which are listed by the OIE. Their taxonomic identification, on which current diagnosis is based, is impaired by antigenic and genetic cross-reactivity and by the lack of a universal, intra-taxon PCR assay, especially for the Mcc, MmmLC and Mbg7 taxa. A suppression subtractive hybridization approach was developed to: 1) define molecular differences between these 3 taxa; 2) analyze the overall genetic diversity within the ‘M. mycoides’ cluster and 3) search for new markers useful for diagnosis. Results obtained here showed that several sequences are shared across taxa, with Mcc and MmmLC being very polymorphic compared to Mbg7 which is more homogeneous, representing a sort of chimera between Mcc and MmmLC. From these analyses, a specific PCR assay was designed for Mcc identification but, because of the genetic diversity existing within the ‘M. mycoides’, the taxonomic identification of new strain appears less and less relevant. Instead, regardless of their species, strain typing on the basis of their virulence would offer an alternative approach for diagnosis. We assessed this type of approach for the MmmLC taxon but so far, our attempts to uncover markers that would distinguish pathogenic strains from carrier strains, isolated from herds with no clinical history, have failed. The genetic continuum observed between strains is remnant of horizontal gene transfers and imposes the development of a more global approach for mycoplasmosis surveillance

Identificação de genes de maracujá azedo diferencialmente expressos durante a interação com Xanthomonas axonopodis / Identification of differentially expressed genes during the yellow passion fruit- Xanthomonas axonopodis interaction

O Brasil é o maior produtor mundial de maracujá azedo (Passiflora edulis f. flavicarpa) sendo esta a espécie de maior expressão comercial dentre as passifloras cultivadas. A bacteriose do maracujazeiro, causada por Xanthomonas axonopodis pv. passiflorae (Xap), é uma das doenças mais severas da cultura, acarretando grandes prejuízos aos produtores. Atualmente, é incipiente o conhecimento sobre a interação maracujá azedo-Xap. Diante disso, a identificação e a caracterização dos genes envolvidos no processo de defesa são passos importantes para dar suporte ao desenvolvimento de variedades resistentes. Assim, o objetivo deste trabalho foi identificar e caracterizar genes de maracujá azedo diferencialmente expressos durante a resposta de defesa à Xap, bem como mensurar a sua expressão. Para isso, foram construídas duas bibliotecas subtrativas de cDNA (forward e reverse) usando o método SSH a partir de transcritos de folhas, que foram inoculadas com o patógeno ou solução salina (controle). Após o sequenciamento dos clones, o processamento e a montagem das sequências, as unisequências foram anotadas através da Plataforma PLAZA e do programa computacional Blast2GO. Genes envolvidos em diversos processos biológicos foram selecionados para a validação das bibliotecas por PCR quantitativo. Usando a Plataforma PLAZA, 78 % (764) das unisequências mostraram similaridade com proteínas de Arabidopsis thaliana, enquanto 87 % (866) delas apresentaram similaridade com proteínas putativas de diversas espécies vegetais, quando se utilizou Blast2GO. Na biblioteca forward, foram identificadas 73 proteínas relacionadas à resposta de defesa, dentre as quais estão proteínas envolvidas na sinalização intracelular, na ativação da transcrição e regulação da expressão de genes de defesa, bem como proteínas de defesa, de resistência e relacionadas à patogênese (PRs). Dentre os 22 transcritos validados, 95 % foram diferencialmente expressos em pelo menos um dos três períodos avaliados; os genes mais expressos em resposta à infecção pelo patógeno são os que codificam as enzimas lipoxigenase, (+)-neomentol desidrogenase e quitinase, as quais participam diretamente nas respostas de defesa vegetal. Dos genes cuja expressão foi mais reprimida, dois codificam proteínas relacionadas à fotossíntese e dois codificam proteínas envolvidas na detoxificação da amônia e do H2O2. Nossos resultados sugerem que a planta utiliza um arsenal de transcritos para responder à infecção; entretanto, este arsenal não é eficiente para impedir a ação do patógeno e, consequentemente, o desenvolvimento da bacteriose nas condições estudadas. Nosso estudo é inédito e gerou informações sobre a reprogramação transcricional durante a interação maracujá azedo-Xap, o que constitui um importante passo para o melhor entendimento sobre este patossistema. / Brazil is the main producer of yellow passion fruit (Passiflora edulis f. flavicarpa) worldwide, which is the most widely commercialized crop among the cultivated passifloras. The bacterial leaf spot induced by Xanthomonas axonopodis pv. passiflorae (Xap) is one of the most severe diseases of the crop, causing great losses to producers. Currently, we understand very little about the yellow passion fruit-Xap interaction. Therefore, the identification and characterization of genes involved in the defense process are important steps to support the development of resistant varieties. Thus, the objective of this study was identify and characterize differentially expressed genes during the defense response to Xap, as well as to measure their expression. For that, we constructed two subtractive cDNA libraries (the forward and the reverse) by performing the SSH method from leaf transcripts, which were inoculated with the pathogen or saline solution (control). After sequencing the clones and sequence data processing, sequences were assembled into unique sequences, which were annotated using the PLAZA Platform and the computational program Blast2GO. Genes involved in several biological processes were selected to validate the libraries by quantitative PCR. When PLAZA was used for sequence similarity searches, 78 % (764) of the yellow passion fruit unique sequences showed similarity to proteins of Arabidopsis thaliana; when Blast2GO was used, 87 % (866) of the unique sequences showed similarities to putative proteins of several plant species. For the forward library, 73 proteins related to defense response were identified, such as those involved in intracellular signaling, transcription activation and regulation of defense gene expression, as well as defense and resistance proteins, and pathogenesis-related proteins (PRs). Of the 22 validated transcripts, 95 % were differentially expressed during at least one of the three periods evaluated; the genes up-regulated in response to the pathogen infection were those that code for the enzymes lipoxygenase, (+)-neomenthol dehydrogenase and chitinase, which participate directly in plant-defense responses. Out of down-regulated genes, two code for photosynthesis-related proteins, and two for ammonia and H2O2 detoxification. Our results suggest the plant uses an arsenal of transcripts to respond to infection; however, this arsenal is not effective to prevent pathogen action and consequently the occurrence of bacterial leaf spot under the evaluated conditions. The present study is the first to produce information on the transcriptional reprogramming during the passion fruit-Xap interaction, which represents an important step for a better understanding of this pathosystem.

"Clonagem e caracterização de genes regulados por glicose em ilhotas pancreáticas humanas" / Cloning and characterization of glucose-regulated genes in human pancreatic islets

O Diabetes mellitus (DM) do tipo 1 é uma doença causada pela destruição, por mecanismo auto-imune, das células beta das ilhotas pancreáticas, produtoras de insulina. O tratamento convencional da doença é realizado por meio de injeções diárias de insulina exógena. O transplante de ilhotas pancreáticas inclui-se, atualmente, como uma das alternativas terapêuticas à insulinoterapia. Entretanto, para atingir a insulino-independência, é necessário transplantar um grande número de ilhotas por paciente. O conhecimento do mecanismo de proliferação das células beta pode possibilitar a realização do transplante a partir da expansão celular ex vivo. A glicose é um dos principais indutores da proliferação de células beta. Neste trabalho, foi estabelecida e executada a tecnologia de isolamento e purificação de ilhotas pancreáticas humanas, visando sua estimulação com glicose. Para identificar genes regulados por glicose nestas ilhotas, foi utilizada a técnica de hibridização subtrativa SSH, associada ao rastreamento da biblioteca através de macroarranjos de DNA. Num primeiro rastreamento, foram identificados dois fragmentos gênicos induzidos pela glicose. Um destes apresentou homologia com uma proteína hipotética humana de função desconhecida e o segundo com o receptor de polipetídeo pancreático. Este trabalho permitiu a identificação de novos genes regulados pela glicose em ilhotas pancreáticas humanas, os quais podem estar relacionados à proliferação celular deste tecido. / Type 1 Diabetes mellitus (T1DM) is caused by autoimmune destruction of the insulin-producing pancreatic islet b-cells. Treatment is generally approached by daily subcutaneous injections of exogenous insulin. Nowadays, pancreatic islet transplantation is considered as an effective alternative treatment to insulin therapy. However, in order to reach insulin-independence, a large number of islets is required for each patient. Knowledge of the mechanisms regulating islet b-cell proliferation may allow ex-vivo b-cell expansion prior to transplant. Glucose is considered one of the main inducers of islet b-cells proliferation. We established and executed the technology of human islet isolation and purification. The islets were then stimulated in culture with glucose. In order to identify glucose-regulated genes in cultured human islets, we utilized the suppression subtractive hybridization (SSH) method, followed by cDNA library screening by DNA macroarrays. Preliminary screening allowed us to isolate two cDNAs displaying glucose regulation, one of which is similar to a human hypothetical protein of unknown function and the other shows similarity to the pancreatic polypeptide receptor. This work allowed identification of glucose-regulated genes in human pancreatic islets, which may be related to cell proliferation in this tissue.

Expressão diferencial de genes induzidos por antracnose em feijoeiro em resposta à indução da resistência por silício / Differential expression of genes activated by anthracnose in response to silicon induced resistance

O feijão é importante fonte carboidratos, vitaminas, minerais e fibras. No Brasil, a produtividade desta leguminosa é baixa e um dos fatores é a ocorrência de doenças como a antracnose causada pelo Colletothrichum lindemuthianum, que gera perdas de até 100% da produção. Plantas possuem diversos mecanismos de defesa contra patógenos e relatos apontam que o silício é capaz não só de promover mudanças morfológicas nas folhas, mas também de ativar os genes de resistência. O presente trabalho foi dividido em três estudos que tinham como objetivo: (1) entender a resposta de três cultivares de feijoeiro ao silício disponível na solução nutritiva; (2) identificar a contribuição do Si na expressão de genes relacionados à infecção pelo fungo através da construção de duas bibliotecas subtrativas por supressão (SSH), visando selecionar genes diferencialmente representados durante a infecção da planta com a raça 65 de C. lindemuthianum (a) e durante a infeção da planta na presença de uma maior dose silicato de potássio (75 ppm) no substrato (b); (3) identificar a resposta de dez transcritos selecionados no Estudo 2 para tentar entender a resposta dos mesmos em diferentes períodos (0; 6; 42; 72 h) após a inoculação, com ou sem suplemento de Si. Como resultados, foi observado que para as três cultivares avaliadas o Si começa a ser absorvido 14 dias após o transplante. Também foi identificado por de microscopia de varredura (MEV) que não há diferença significativa entre o número de tricomas e cada cultivar, mas que para o número de estômatos a cultivar IAC-Harmonia destacou-se das demais. Além disso, quando as três cultivares foram suplementadas com Si, houve a formação de uma cera epicuticular descrita como mecanismo de defesa da planta contra fungos; e que através de EDX (Energy-dispersive X-ray spectroscopy) foi possível constatar que plantas tratadas com Si apresentam maior teor deste elemento nas folhas. Através de inoculações com a raça 65 do patógeno verificou-se o efeito do mineral na redução da severidade da doença nas cultivares IAC-Harmonia e Pérola. No segundo estudo, duas bibliotecas de hibridização subtrativa por supressão (SSH), foram construídas visando selecionar os genes diferencialmente expressos entre plantas inoculadas e não-inoculadas (A) e entre plantas inoculadas e tratadas ou não com 75 ppm de Si (B). Foram geradas 991 sequências únicas, anotadas através do GeneOntolgy. Quinze genes de cada biblioteca foram selecionados para os experimentos de validação por RT-qPCR. Para a Biblioteca A, 11/15 genes foram positivamente regulados, e em B, 14/15. No terceiro estudo ficou evidenciado que a inoculação com o patógeno alterou positivamente a expressão de sete genes, enquanto que o tratamento com 75 ppm de Si alterou a expressão de oito genes, em pelo menos um dos tempos avaliados / Beans are an important source of carbohydrates, vitamins, minerals and fibers. In Brazil, this legume still has low productivity and one of the factors involved is the occurrence of diseases such as anthracnose, caused by the fungus Colletothrichum lindemuthianum, which causes losses in production of up to 100%. Plants present several defense mechanisms against pathogens and the reports indicate that silicon does not only promote morphological changes in leaves, but also activates resistance genes. This work was divided into three studies aiming: (1) to understand the response of three bean cultivars to a silicon source in a nutrient solution, (2) to identify the contribution of Si in the expression of genes related to the infection by the fungus by constructing two subtractive suppression libraries (SSH), to select genes differentially represented during infection of the plant with race 65 of C. lindemuthianum (a) and during infection of the plant in the presence of higher dose of potassium silicate (75 ppm) in the substrate (b), (3) to identify the response of ten selected transcripts in Study 2 in various periods (0, 6, 42, 72 h) after inoculation, with or without supplemental Si. As a result, it was observed that for all three cultivars Si begins to be absorbed 14 days after transplantation. Etude des interactions hôte/parasite chez l’huître plate Ostrea edulis et son parasite Bonamia ostreae / Study of host/parasite interactions in the flat oyster Ostrea edulis and the parasite Bonamia ostreae

L’histoire de l’ostréiculture française met en évidence la fragilité de cette production face à la surexploitation des stocks et l’apparition de maladies. En particulier, la production d’huître plate, Ostrea edulis, a fortement diminué suite à l’apparition de deux maladies parasitaires dont la bonamiose. Les moyens de lutte contre la bonamiose sont relativement restreints. Ils sont essentiellement basés sur la surveillance de la santé des huîtres afin de limiter la dissémination et la propagation de la maladie. Cependant l’utilisation de modèles prédictifs de l’évolution de la maladie en zone infectée permettrait d’optimiser la gestion des stocks et minimiser l’impact des agents pathogènes. De plus, le développement d’animaux résistants à l’infection pourrait permettre de relancer cette production. Ces différentes approches nécessitent des outils diagnostiques adaptés, une bonne connaissance du cycle de vie de l’agent pathogène, et, plus particulièrement des interactions du parasite avec son hôte. Dans ce contexte, l’objectif principal du travail de thèse proposé est de comprendre les interactions entre l’huître plate Ostrea edulis et son parasite Bonamia ostreae, et, plus particulièrement les bases moléculaires de la résistance au parasite. Dans un premier temps, la réalisation d’une banque soustractive d’ADNc a permis d’identifier des ESTs différentiellement exprimées chez des hémocytes en réponse au parasite. L’expression de certains gènes dont une galectine a été mesurée en PCR en temps réel dans le contexte d’infections in vitro. En complément, la réponse cellulaire a été étudiée par cytométrie en flux et l’infection contrôlée en microscopie. Ces expériences ont montré une multiplication parasitaire dans les hémocytes au cours du temps associée à une diminution de la production d’EOR et d’estérases. Dans un second temps, il a été entrepris une étude comparative entre une population d’huîtres plates résistantes à la bonamiose et une population naturelle. Les résultats obtenus tendent à montrer qu’une modulation de l’apoptose et une diminution de la phagocytose seraient impliquées dans les mécanismes liés à la résistance à la bonamiose. Ce travail est le premier à étudier la réponse des hémocytes d’huîtres plates à une infection par le parasite Bonamia ostreae au niveau cellulaire et moléculaire. / The history of the French oyster production highlights the fragility of this production against overexploitation and disease outbreaks. In particular, the production of flat oyster, Ostrea edulis, has decreased following the emergence of two parasitic diseases including bonamiosis. The means to fight against bonamiosis are relatively limited. They are mainly based on oyster health surveillance to limit the spread of the disease. However, the use of predictive models of disease progression in infected area would help to improve stock management and minimize the impact pathogens. Moreover the development of resistant animals could help to revive this production. These different approaches require appropriate diagnostic tools, a good knowledge of the life cycle of the pathogen, and the interactions between the parasite and its host. In this context, the main objective of the phD work is to understand the interactions between the flat oyster Ostrea edulis and the parasite Bonamia ostreae, and particularly the molecular basis of the resistance to the parasite. In a first step, a subtractive cDNA bank allowed the identification of ESTs differentially expressed in haemocytes in response to the parasite. Expression of some genes, among which a galectin, was measured by Real Time PCR in the context of in vitro infections. In addition, the cellular response was investigated by flow cytometry and the infection was checked by microscopy. These experiments showed a multiplication of the parasite inside haemocytes associated with a decreased of esterases and of the production of ROS. In a second step, a comparative approach was carried out between a population of oysters resistant to bonamiosis and a natural population. Results suggest that modulation of apopotosis and decrease of phagocytosis could be involved in mechanisms related to resistance to bonamiosis. This work is the first study on the response of haemocytes of flat oysters to an infection with the parasite Bonamia ostreae at the cellular and molecular levels.
