Training intensity distribution, physiological adaptation and immune function in endurance athletes

Exercise intensity and its distribution is probably the most important and most heavily debated variable of endurance training. Training induces adaptation but also induces stress responses. Controlling the training-intensity distribution may provide a mechanism for balancing these two effects. It has been reported that elite endurance athletes train with a high volume and load, relative to the sport. These athletes spend the vast majority (>80%) of training time at relatively low intensities (lower than the lactate threshold, zone one), and therefore <20% of training time above the lactate threshold (zones two and three). Experimental studies support the beneficial effects of a high training volume in zone one, and show detrimental effects of replacing zone one training with training in zone two. This is likely due to enhanced recovery from training in zone one compared with training in zone two. The acute recovery following training sessions in zones two and three has been reported to not be different, but the recovery following training in zone one has been reported to be faster. Improvements in physiological adaptation and endurance performance have been reported to be greater following training programmes with higher exercise intensities. Therefore, it has been suggested that a polarised training model, which includes ~80% of training in zone one with ~20% of training in zone three is more beneficial than a threshold training model, with the majority of training in zone two. However, research into an optimal training-intensity distribution is limited. Therefore, the aims of this thesis were to assess the effectiveness of training-intensity distribution on the improvements in physiological adaptation, endurance performance and assess if manipulating training-intensity distribution had an effect on immune function. Study one revealed that the lactate threshold, the lactate turnpoint and maximal performance measures in swimming, cycling and running, assessed using the methods outlined in the study, are reproducible in trained endurance athletes. These tests can therefore be used by trained endurance athletes as part of a physiological testing programme to assess not only endurance performance, but also to demarcate training intensity zones for exercise intensity prescription and monitor moderate to large adaptations to training. Practitioners should take care when deciding on the duration between tests to test for adaptations from training, as adaptations need to be greater than these detected test-retest variations to be considered physiologically meaningful. To the best of the author’s knowledge, study two was the first study to have assessed training-intensity distribution in a group of multisport athletes. Training was monitored over a 6-month period, and testing took place every two months to assess the effect of the training on physiological adaptation. Although speculative due to the number of variables involved, the results suggest that a greater proportion of training time spent in zone one and a lower proportion of training time spent in zone two is beneficial to physiological adaptation. However, given the number of variables associated with assessing the training-intensity distribution in multisport athletes, it is not easy to draw conclusions as to the effectiveness of the training in the different disciplines on the key measures of adaptation in the different disciplines. Study two highlighted the need for future research to focus on experimental manipulation of training-intensity distribution and thus improve our understanding of its impact on the training-induced adaptations in endurance athletes. Study three manipulated the training-intensity distribution in trained endurance athletes in just one discipline, to reduce the number of variables involved. A polarised training model was compared to a threshold training model on the effectiveness to improve physiological adaptation and endurance performance. Results revealed that a polarised training model is recommended for trained cyclists wishing to maximally improve performance and physiological adaptation over a short-term (six week) training period. The first part of study four assessed the effect of a polarised and a threshold training model on immune function markers in trained cyclists. Both endurance training programmes had similar volume, and were sufficient to induce improvements in performance and physiological adaptation. However, despite likely differences in recovery, both training programmes had no effect on the proportion of low or high differentiated or senescent CD8+ or CD4+ T-cells in blood. Therefore, training adaptation was achieved at no cost to this particular aspect of immune function. From these results and evidence from previous studies, it seems likely that athletes need to be overreached to induce any change in immune function following a period of intensified training. The second part of study four assessed the impact of an ironman triathlon race on Epstein-Barr virus (EBV) and Varicella-Zoster virus (VZV) antibody titres and the frequency of low and high differentiated and senescent blood T-cells in trained endurance athletes. Previous work has revealed that an ironman triathlon race increases the proportion of senescent CD4+ T cells and decreases the proportion of naive CD4+ T cells, and thus induces changes the immune space which could leave an individual at a greater risk of infection. This study however, did not find any changes in the proportions of these T cell subsets following an ironman triathlon race. The mean results of this study suggest that there is no relationship between EBV and VZV-specific antibody concentrations and the proportion of senescent, low and highly differientiated T cells. However, on analysis of individual subject data, it seems possible that subjects with a high antibody titre for EBV or VZV 3 wks before a competition might be more at risk of infection post race. A greater subject number would be needed in order to make a more conclusive statement about this relationship. The results of this thesis suggest that future research is required in the area of training-intensity distribution. Firstly, our understanding of the physiological mechanisms responsible for the effectiveness of a polarised training model in trained endurance athletes is limited, and thus studies should attempt to address this issue. Our current knowledge on the mechanisms underlying a blunted T cell response following strenous exercise is also limited. A change in the immune space to a greater proportion of senescent T cells and a lower proportion of naive T cells might contribute to this blunted response. In the current thesis however, the proportions of these T cell markers were unchanged following the training/racing interventions. It is possible that with a higher training load, there could be changes in these markers, and thus this is an exciting area that could have potential implications on athlete health. Finally, testing for antibody titres in endurance athletes is possibly an avenue to detect individuals at the greatest risk of infection if subjected to a large physical and/or mental stress. This could have implications on maintaining athlete health and therefore, allowing athletes to train consistently.

Ecologies of knowledge : narrative ecology in contemporary American fiction / Strecker

In the 1980s and 1990s, many scientifically cognizant young novelists turned away from the physics-based tropes of entropy and chaos and chose biological concepts of order, complexity, and self-organization as their dominant metaphors. This dissertation focuses on three novels published between 1991 and 1996 that replace the notion of the encyclopedia as a closed system and model new narrative ecologies grounded in the tenets of the emergent science of complex systems. Thus, Richard Powers's The Gold-Bug Variations (1991) explores the marriage of bottom-up self-organizing systems and top-down natural selection through a narrative lens and cautions us against any worldview which does not grasp life as a complex system; Bob Shacochis's Swimming in the Volcano (1993) illustrates how richly complex global behavior emerges from the local interaction of a large number of independent agents; and, David Foster Wallace's Infinite Jest (1996) enacts a collaborative narrative of distributed causality to investigate reciprocal relationships between the individual and the multiple systems in which he is embedded. Unlike many other contemporary authors, the new encyclopedists do not shun the abundance of information in postmodern culture. Instead, as I demonstrate here, the intricate webs of their complex ecologies emerge as narrative circulates through diverse informational networks. Ecologies of Knowledge argues that these texts inaugurate a new naturalism, demanding a reconciliation between humans and the natural world and advocating an increased understanding of life's interdependent patterns and particularities. Grounded in such an awareness of ecological complexity, these large and demanding books are our survival guides for the twenty-first century. / Department of English

Numerical analysis of fluid motion at low Reynolds numbers

At low Reynolds number flows, the effect of inertia becomes negligible and the fluid motion is dominated by the effect of viscous forces. Understanding of the behaviour of low Reynolds number flows underpins the prediction of the motion of microorganisms and particle sedimentation as well as the development of micro-robots that could potentially swim inside the human body to perform targeted drug/cell delivery and non-invasive microsurgery. The work in this thesis focuses on developing an understanding in the mathematical analysis of objects moving at low Reynolds numbers. A boundary element implementation of the Method of regularized Stokeslets (MRS) is applied to analyse the low Reynolds number flow field around an object of simple shape (sphere and cube). It also showed that the results obtained by a boundary element implementation for an unbounded cube, where singularities are presented in the corners of the cube, agrees with more complex solutions methods such as a GBEM and FEM.A methodology for analysing the effect of walls by locating collocation points on the surface of the walls and the object is presented. First at all, this methodology is validated with a boundary element implementation of the method of images for a sphere at different locations. Then, the method is extended when more than one wall is presented. This methodology is applied to predict the velocity filed of a cube moving in a tow tank at low Reynolds numbers for two different cases with a supporting rod similar to an experimental set-up, and without the supporting rod as in the CFD simulations based on the FVM. The results indicate a good match between CFD and the MRS, and an excellent approximation between the MRS and experimental data from PIV measurements. The drag, thrust and torque generated by helices moving at low Reynolds numbers in an unbounded medium is analysed by the resistive force theory, a slender body theory, and a boundary element method of the MRS. The results show that the resistive force theory predict accurately the drag, thrust and torque of moving helices when the resistive force coefficients are calculated from a slender body theory approximation by calculating independently the resistive force coefficients for translation and rotation, because it is observed that the resistive force coefficients depend also of the nature of motion. Moreover, the thrust generated by helices of different pitch angles is analysed calculated by a CFD numerical simulation based on the FVM and a boundary element implementation, an compared with experimental data. The results also show an excellent prediction between the boundary element implementation, the CFD results and the experimental data. Finally, a boundary element implementation of the MRS is applied to predict swimming of a biomimetic swimmer that mimics the motion of E.coli bacteria in an unbounded medium. The results are compared with the propulsive velocity and induced angular velocity measurement by recording the motion of the biomimetic swimmer in a square tank. It is observed that special care needs to be taken when the biomimetic swimmer is modelled inside the tank, as there is an apparent increment in the calculate thrust propulsion which does not represent a real situation of the biometic swimmer which propels by a power supply. However, this increment does not represent the condition of the biomimetic swimmer and a suggested methodology based on the solution from an unbounded case and when the swimmer is moving inside the tank is presented. In addition, the prediction of the free-swimming velocity for the biomimetic swimmer agrees with the results obtained by the MRS when the resistive force coefficients are calculated from a SBT implementation. The results obtained in this work have showed that a boundary element implementation of the MRS produces results comparable with more complex numerical implementations such as GBEM, FEM, FVM, and also an excellent agreement with results obtained from experimentation. Therefore, it is a suitable and easy to apply methodology to analyse the motion of swimmers at low Reynolds numbers, such as the biomimetic swimmer modelled in this work.

Le collagène XXII dans la formation et la fonctionnalité de la jonction myotendineuse chez le poisson zèbre, de l’embryon à l’âge adulte / Collagen XXII in zebrafish myotendinous junction formation and function, from embryogenesis to adulthood

Le collagène XXII (COLXXII) a été décrit, en 2004, comme un marqueur des jonctions tissulaires chez la souris. En particulier, la jonction myotendineuse (JMT) est une matrice extracellulaire spécialisée qui assure une liaison structurale entre le muscle squelettique et le tendon et permet la transmission des forces de contraction au squelette. Mon projet de thèse a visé à caractériser le rôle de COLXXII in vivo; le modèle du poisson zèbre a été choisi. Chez le poisson zèbre, col22a1 code pour COLXXII dont l'expression débute dans tout le somite, puis, se restreint progressivement aux extrémités des fibres musculaires au niveau de la JMT, où est déposée la protéine. Utilisant la technique de morpholino-knockdown, nous avons montré que les morphants développent un phénotype dystrophique et que le COLXXII fait très probablement partie du complexe d'ancrage intégrine α7β1. Cependant, compte tenu de la durée d'efficacité réduite des morpholinos, notre étude était limitée. Utilisant la technologie CRISPR-Cas9, nous avons donc généré deux lignées invalidées pour col22a1. L'analyse ultra-structurale des poissons col22a1-/-, de la jeune larve à l'adulte, montre que les interdigitations du sarcolemme, caractéristiques de la JMT, sont pratiquement absentes chez les mutants (comme chez les morphants), pouvant impacter la transmission des forces et/ou l'attachement du muscle aux myoseptes (tendons). De façon intéressante, pour les deux lignées, la même proportion de larves présente un phénotype très sévère entrainant la mort vers 2 semaines post-fécondation (spf). Les autres larves, elles, survivent et ne montrent pas de phénotype global particulier. En revanche, une forte réduction des interdigitations de la JMT est constatée et dans de rares cas, après challenge des poissons, une rupture musculaire est observée. Une première approche de q- PCR par gène candidat a été réalisée et il semble possible que les différences phénotypiques soient liées à des évènements conjoints d'expressivité variable et de compensation génique. Enfin, utilisant un système original de test de nage à contre-courant par palier, j'ai montré que les poissons col22a1-/- âgés de 6 mois ont une capacité de nage très diminuée et consomment d'avantage d'O2 pendant l'effort comparés aux animaux sauvages. L'efficacité du système musculo-squelettique semble donc moindre en l'absence de collagène XXII. Nos résultats devraient permettre de considérer COL22A1 comme un gène candidat pour les cas de dystrophies musculaires dont la cause génétique est non élucidée / The myotendinous junction (MTJ) is a specialized extracellular matrix which allows the transmission of skeletal muscle forces to bones. My thesis project aimed at characterizing the in vivo role of COLXXII using the zebrafish as a model. The zebrafish col22a1 gene encodes COLXXII whose expression begins in the entire somite, and is then restricted gradually at the MTJ, where the protein is deposited. Morphants (Knockdown) develop a dystrophic phenotype and we have demonstrated that COLXXII is likely a member of the integrin a7ß1 anchoring complex. However, because morpholinos are efficient only few days after injection, our study was limited to early stages of zebrafish development. We thus generated two mutant lines (CRISPR-Cas9) invalidated for col22a1. The typical sarcolemmal interdigitations of the MTJ are almost absent in mutants, from larvae to adult (as in morphants), which could impact the force transmission and/or the muscle attachment to myosepta (tendons). In the two mutant lines, the same proportion of larvae displays a severe phenotype leading to fish death 14 days post-fertilization. On the contrary, other larvae survive without any obvious general phenotype. A first candidate genes approach was realized by qPCR and tends to show that the phenotypic differences may be due to both, variable expressivity and genetic compensation. Finally, I have shown that 6 months col22a1-/- fish show a highly decreased swimming capacity and consume more O2 during effort compared to wild type animals. Thus, we conclude that the musculoskeletal system efficiency seems altered in absence of COLXXII. Our results should allow considering COL22A1 as a candidate gene for muscular dystrophies with unclarified genetic cause.

Efeito antidepressivo e ansiolítico do extrato metanólico de Hibiscus tiliaceus em modelo animal de depressão pós-parto

Hibiscus tiliaceus L. (Malvaceae) é usado popularmente em desordens do pós-parto. O extrato metanólico de flores de H. tiliaceus apresentou atividade antidepressiva-like nos testes de nado forçado e de suspensão da cauda, ambos são amplamente usados como modelos animais preditivos da atividade antidepressiva. Além disso, o extrato demonstrou uma tendência de aumento do tempo gasto nos braços abertos no labirinto em cruz elevado. Considerando que o extrato de H. tiliaceus, que contém fitoesteróis como o estigmasterol, o stigmastadienol e o stigmastadienone, pode ser útil no tratamento ou prevenção da depressão pós-parto relacionadas à retirada crônica de altos níveis de hormônios associados à gestação. Nosso objetivo foi avaliar o efeito de extratos de H. tiliaceus em modelo animal de depressão pós-parto e ansiedade. Utilizamos ratas Wistar fêmeas submetidas ao modelo de depressão pós-parto induzido por hormônios (estradiol e progesterona), administrados por via subcutânea. Após período gestacional induzido, os animais receberam água, veiculo ou extrato de H. tiliaceus nas doses de 100 e 400mg/kg, via oral (gavagem). No período pós-parto, as ratas foram submetidas a testes comportamentais de nado forçado, o labirinto em cruz elevado e de preferência claro-escuro como modelos preditivo de depressão e ansiedade. Foi obsevado um aumento no tempo de imobilidade associado a uma diminuição na tentativa de fuga, no teste do nado forçado, em animais submetidos ao modelo de depressão pós-parto comparado ao grupo controle, sugerindo que este modelo é capaz reproduzir sintomas de depressão pós-parto. Não houve diferença significativa nos testes de ansiedade entre os animais submetidos ao modelo de depressão pós-parto comparado aos controles. A administração de extrato metanólico de H. tiliaceus não foi capaz de alterar o comportamento relacionado à depressão e ansiedade em ratas Wistar. / Postpartum affective disorders are rarely modeled. The depressive-like behavior of hormone withdrawal following hormone-simulated "pregnancy" was described in Long-Evans and Sprague Dawley rats. Our aim was to evaluate the validity of hormone withdrawal following hormonesimulated "pregnancy" method in Wistar rats as a model of depression and/or anxiety. Recently, it was demonstrated an antidepressant-like profile of methanol extract of Hibiscus tiliaceus L., a plant used in postpartum disorders, in adult male Swiss albino mice, then, we also investigated the antidepressant and anxiolytic-like activities of the methanol extract of H. tiliaceus flowers using this animal model of postpartum disorder. Ovariectomized rats received daily injections of the vehicle or hormones (estradiol and progesterone) to simulate the 23-day gestational period in the rat. Days 24-27 were considered the ''post-partum'' period, where the methanolic extract of H. tiliaceus or vehicle were administered by gavage. Rats were submitted to forced swimming, elevated plus-maze test and lightdark box tests. Rats submitted to ''post-partum depression model'' increased the immobility time in forced swimming. The methanolic extract of H. tiliaceus administration did not alter the immobility time in the forced swim test. In the light-dark box test, rats submitted to post-partum depression model showed decreases in number of rearing in dark compartment. In conclusion, our data indicate that Wistar rats may be an adequate model postpartum affective disorders, showing ''depressive-like'' symptoms in the forced swim test without any anxiogenic effect. Besides, we could suggest that decrease on number of rearing in the dark compartment may indicate the motivational state.

Mecanismos subjacentes ao efeito da manipulação neonatal sobre o vínculo mãe/filhote

Ao nascerem, os mamíferos não estão com o sistema nervoso plenamente desenvolvido e os primeiros dias de vida representam uma fase crítica para o desenvolvimento desse sistema. De fato, nesta fase o encéfalo está passando por diversos processos fundamentais como organização funcional das redes neurais, proliferação neuronal, migração, diferenciação, além de gliogênese e mielinização. Em ratos, um procedimento simples, como “manipular” os filhotes por alguns minutos durante a primeira semana de vida, pode marcar decisivamente o desenvolvimento do indivíduo. Assim, a manipulação neonatal tem sido muito utilizada para se examinar os mecanismos pelos quais variações ambientais podem afetar o desenvolvimento do filhote. A manipulação neonatal promove uma série de alterações comportamentais e neuroendócrinas que se caracterizam basicamente por uma diminuição do medo e da resposta ao estresse no adulto. Embora muitos autores até caracterizem a manipulação como uma intervenção positiva ela também pode provocar graves déficits em comportamentos sociais e reprodutivos para a prole aparecendo desde o inicio do desenvolvimento e persistindo até a vida adulta. Além de seu efeito sobre os filhotes, estudos tem demostrado que intervenções na prole no período neonatal também afetam de forma duradoura a resposta ao estresse das genitoras, mas este tema ainda é muito pouco explorado pela literatura Portanto, na primeira parte desta tese, iremos estudar os efeitos da manipulação neonatal sobre a formação do vínculo mãe-filhote, tentando associar mudanças no comportamento da mãe ao longo dos 10 primeiros dias pós-parto com a preferencia pelo odor do ninho em filhotes testados no labirinto em Y. Na segunda parte desta tese iremos abordar os efeitos da manipulação sobre a resposta ao estresse agudo e crônico em ratas que tiveram seus filhotes manipulados no período neonatal, para isso submetemos genitoras dos grupos controle e manipulado após o desmame a um dos dois protocolos descritos a seguir: com estresse (estresse por contenção de movimentos 1h/dia por 7 dias) ou sem estresse (nenhuma intervenção após o desmame) e testamos os animais através do teste de nado forçado para observar mudanças na resposta emocional. Também medimos os níveis de BDNF e corticosterona no plasma após o teste e medimos o peso das adrenais para verificar o efeito da manipulação na resposta ao estresse das genitoras Os resultados dessa tese mostram que a manipulação neonatal afeta a estrutura do comportamento maternal, mudando a sequencia e a sincronia do comportamento da mãe com o filhote, o que poderia ser em parte a causa da alteração no comportamento de preferencia pelo odor do ninho observado em animais manipulados, principalmente nas fêmeas. Além disso, observamos que a manipulação afeta de forma duradoura a resposta ao estresse (agudo e crônico) das genitoras, podendo alterar a resposta emocional desses animais e predispor a sintomas do tipo depressivo em resposta ao estresse agudo Esses resultados reforçam a ideia de que o estudo dos efeitos duradouros da manipulação não só nos filhotes, mas também no organismo materno, podem servir como uma importante ferramenta para elaboração de projetos clínicos, visando a exploração da existência de comportamentos similares em humanos. Isso ajudará na elaboração de politicas de saúde publica que visem minimizar os efeitos de eventos adversos acontecidos no inicio da vida sobre a saúde física e mental tanto da mãe quanto da criança. / Mammals are not born with fully developed nervous system, and the first days of life represent a critical stage in the development of this system. In fact, at this stage, the brain is undergoing many fundamental processes such as functional organization of neural networks, neuronal proliferation, migration, differentiation, gliogenesis and myelination. In rats, a simple procedure such as "handling" the pups for a few minutes during the first week of life can decisively mark the development of the individual. Thus, neonatal handling has been widely used to examine the mechanisms by which environmental adversity can affect the development of the pups. Neonatal handling promotes a series of behavioral and neuroendocrine changes that are characterized primarily by a decrease of fear and stress responses in the adult. Although many authors characterize the handling procedure as a positive intervention, it is also associated with severe deficits in social and reproductive behaviors of the offspring that appear early during development and persist into adulthood. Apart from its effect on the pups, studies have shown that interventions in the offspring during the neonatal period can also induce long lasting effects in the maternal stress response, but this subject is still little explored in the literature Therefore, in the first part of this thesis, we will study the effects of neonatal handling on the mother-pups’ bond formation, trying to associate changes in maternal behavior over the first 10 days postpartum with the preference for the odor of the nest in pups tested the Y maze. The second part of this thesis will address the effects of neonatal handling on the acute and chronic stress response in dams that had their pups handled. For this purpose, we submitted mothers of control and manipulated groups after weaning to: stress (restraint 1h/day for 7 days) or no stress (no intervention after weaning) and tested the animals using the forced swim test to observe changes in emotional response. We also measured plasma BDNF and corticosterone levels after the test and the adrenals’ weight to verify the effect of handling on the dam’s stress response. The results of this thesis show that neonatal handling affects the structure of maternal behavior, changing the behavioral sequence and synchrony of the mother with her pups, which could be in part the cause of the altered social behaviors observed in handled pups, especially in females. Moreover, we observed that handling affects the dam´s response to stress (acute and chronic), and may alter the emotional response of the dams increasing the susceptibility to developing psychiatric disorders such as depression at least in response to acute stress These results reinforce the idea that investigating the long lasting effects of handling not only in the young, but also in the dam’s physiology becomes an important tool for the development of clinical studies, aiming at exploring the existence of similar effects in humans. The final goal will be the elaboration of public health policy to minimize the effects of early life adverse events on physical and mental health of both mothers and their children.

O estudo teve como objetivo investigar o efeito do exercício físico (natação) na redução da esteatose hepática e comorbidades associadas, incluindo a expressão hepática de síntese de ácidos graxos e receptor proliferador de peroxissoma atividade alfa. Camundongos machos C57BL/6 foram divididos em dois grandes grupos de acordo com a dieta durante 22 semanas: dieta padrão (10% de gordura, SC) ou dieta rica em gordura (60% de gordura, HF), caracterizando os grupos sedentários SC-Sed e HF-Sed. Nas últimas 10 semanas do experimento, metade dos grupos sedentários foram submetidos ao protocolo de natação com um aumento progressivo no tempo (6/dia até 60/dia, 5x/semana), caracterizando os grupos exercitados: SC-Ex e HF-Ex. No final do experimento, comparado ao grupo SC-Sed, o grupo HF-Sed teve a massa corporal significativamente superior, hiperglicemia, hiperinsulinemia com resistência à insulina, hipertrofia dos adipócitos (com infiltrado inflamatório), hipertrofia das ilhotas pancreáticas, dislipidemia, alteração das enzimas hepáticas e inflamatórias e NAFLD com mudanças na expressão de proteínas hepáticas lipogênicas e oxidativas. O programa de natação, mesmo concomitante com a dieta rica em gordura, reduziu o excesso de peso e todos os outros resultados, especialmente a NAFLD. Os resultados permitem concluir que a natação pode atenuar os efeitos deletérios de uma dieta rica em gorduras combinado com estilo de vida sedentário em camundongos. Estes dados reforçam a idéia que o exercício físico pode ser considerado uma estratégia terapêutica não farmacológica eficaz no tratamento da NAFLD, obesidade e resistência à insulina. / Chronic accumulation of fat in the liver (nonalcoholic fatty liver disease or NAFLD) is a morbid condition strongly associated with obesity and insulin resistance. The study aimed to investigate the effect of swimming training in reducing the NAFLD and associated comorbidities, including the hepatic expression of fatty acid synthesis and peroxisome proliferator receptor activity-alpha . Male C57BL/ 6 mice were separated into two major groups according to their nutrition and studied during 22 weeks: standard chow (10% fat, SC) or high fat chow (60% fat, HF), characterizing the sedentary groups SC-Sed and HF- Sed. In the last 10 weeks of the experiment, half of the sedentary groups were submitted to a swimming training with a progressive increase in duration, characterizing the exercised groups: SC-Ex and HF-Ex. At the end of the experiment, considering the findings in the SC-Sed group, HF-Sed group had significantly higher body mass, hyperglycemia, hyperinsulinemia with insulin resistance, hypertrophy of the adipocytes (with inflammatory infiltrate), hypertrophy of the pancreatic islets, dyslipidemia, altered liver enzymes and inflammatory cytokines, and NAFLD with changes in gene expression of hepatic lipogenic and oxidative proteins. The swimming program, even concomitant with the high-fat diet, reduced overweight and all the other worst findings, especially NAFLD. The results allow the conclusion that swimming training can attenuate the morbid effects of a high-fat diet combined with sedentary lifestyle in mice. These data reinforce the notion that swimming exercise can be considered an efficient nonpharmacologic therapy in the treatment of NAFLD, obesity and insulin resistance.

EFEITO DO EXERCÍCIO FÍSICO NO EQUILÍBRIO E NO METABOLISMO OXIDATIVO-INFLAMATÓRIO DE RATOS TRATADOS COM VINCRISTINA / EFECT OF THE TREINNING EXERCISE ON BALANCE AND OXIDATIVE-INFLAMATORY METABOLISM IN RATS EXPOSED TO VINCRISTINA

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / The Vincristine is a chemotherapy used for various types of cancers, and is one of the major side effects the impairment of motor function and balance, as well as the oxidative and inflammatory unbalance. Therefore, the present study aimed to evaluate the effect of physical exercise on the balance and in the oxidative metabolism of rats treated with anti-inflammatory Vincristine. For this, rats weighing between 270-300 g were divided into two groups: Trained Group (n = 20, 6 weeks of swimming, 1:00/day, 5 days/week, with 5% overload body weight) and Sedentary Group (n = 20, placebo swimming in a tank with water to 5 cm deep, with no extra charge). After 6 weeks the two groups (n = 20 of each) were treated with a single dose of weekly 0, 5 mg/kg of vincristine sulfate, for two weeks, totaling a cumulative dose of 1 mg/kg intraperitoneal. The rats used in control groups (n = 10 n = 10 trained and sedentary) received the same dose of saline (NaCl). For analysis of motor function and balance were performed after chemotherapy applications, on 1, 7, 8,15 days: Rotarod tests (balance), Grip Test (muscle strength), the Open field (exploration) and the hot plate (peripheral sensitivity). At the end of the assessment (day 15) cerebellum samples were taken for analysis of oxidative stress (DCFH, SOD and CAT) and blood samples for analysis of Picogreen and CBC. In behavioral assessment, there was a decrease in body balance in rats exposed to the VCR and, also, a significant protection of physical exercise. There was no significant decrease in muscle strength and exploitation in the open field of rats exposed to VCR, but there was no effect per se of physical exercise, which increased muscle strength and the distance and the operating time standing in the open field in mice. There was significant change in peripheral sensitivity in mice treated and prior exercise was able to normalize this damage. In biochemical analysis we observed a significant protective effect of exercise in trained/VCR rats, once ROS levels were reduced (DCFH analysis) and antioxidant system enzymes of the cerebellum SOD and CAT modulated positively. In the analysis of observed Picogreen exercise protection against DNA damage in the trained/VCR group. The CBC trained /VCR rats they kept the parameters analyzed within the reference values when compared to sedentary/VCR. Therefore, the exercise proved to be a great ally in protecting the body and balance the immune system front of chemotherapy toxicity Vincristine. / A vincristina é um quimioterápico usado para diversos tipos de cânceres, e tem como um dos principais efeitos colaterais o comprometimento da função motora e equilíbrio, bem como o desbalanço oxidativo e inflamatório. Portanto, o presente estudo teve como objetivo avaliar o efeito do exercício físico no equilibrio e no metabolismo oxidativo-inflamatório de ratos tratadas com Vincristina. Para isso, ratos com peso entre 270-300g foram divididos em dois grupos: Grupo treinado (n=20, 6 semanas de natação, 1h/dia, 5 dias/semana, com sobrecarga de 5% do peso corporal) e Grupo sedentário (n=20, natação placebo em um tanque com água a 5cm de profundidade, sem carga extra). Após as 6 semanas os dois grupos (n=20 de cada) foram tratados com dose única semanal de 0,5mg/kg de sulfato de vincristina, durante duas semanas, totalizando uma dose acumulada de 1 mg/kg, por via intraperitoneal. Os ratos utilizados nos grupos controle (n=10 treinados e n=10 sedentários) receberam a mesma dose de solução salina (NaCl). Para análise da função motora e equilíbrio foram realizados, após as aplicações de quimioterapia, nos dias 1, 7, 8 e 15 os testes Rotarod (equilíbrio), Grip-Test (força muscular), Campo aberto (exploração) e o Placa quente (sensibilidade periférica). Ao final das avaliações (dia 15) foram retiradas as amostras de cerebelo para análise de estresse oxidativo (DCFH, SOD e CAT) e amostras de sangue para análise de Picogreen e hemograma. Na avaliação comportamental, observou-se uma diminuição do equilíbrio corporal nos ratos expostos à VCR e, também, uma proteção significativa do exercício físico. Não houve diminuição significativa da força muscular e da exploração no campo aberto dos ratos expostos à VCR, mas houve efeito per se do exercício físico, que aumentou a força muscular e a distância percorrida e o tempo de exploração em pé no Campo aberto nos ratos treinados. Houve alteração significativa na sensibilidade periférica nos ratos tratados e o exercício físico prévio foi capaz de normalizar este dano. Nas análises bioquímicas observamos um efeito protetor significativo do exercício físico nos ratos treinados/VCR, uma vez que os níveis de ROS foram diminuídos (análise de DCFH) e as enzimas do sistema antioxidante do cerebelo SOD e CAT modularam positivamente. Na análise de Picogreen, observamos uma proteção do exercício físico contra o dano de DNA no grupo treinado/VCR. No hemograma os ratos treinados/VCR, mantiveram os parâmetros analisados dentro dos valores de referência quando comparados aos sedentários/VCR. Sendo assim, o exercício físico mostrou-se um grande aliado na proteção do equilíbrio corporal e do sistema imunológico frente à toxicidade do quimioterápico Vincristina.
