Nutrition of container grown plants with emphasis on the Proteaceae

A range of Proteaceous shrubs and other nursery plants were grown in containers with soilless media and various N levels. Plants demonstrated a range of responsiveness. Supplying soilless media with Osmocote (26% N) and other short term fertilisers proved to be a satisfactory method of studying the comparative nutrition of a wide range of container grown nursery plants using factorial experiments incorporating N, P, K and lime. Nutrient response surfaces were obtained using a central composite incomplete block design. Most Proteaceous shrubs were intolerant of high P levels due to excessive luxury consumption resulting in toxic foliar nutrient levels, especially in the presence of high N. Phosphorus sensitivity in plants appeared to correspond with the soil nutrient levels in their native habitat. This applied to species studied in the 2 main sub-families in the Proteaceae while similar findings were indicated for other Australian genera. A range of optimum N requirements in the Proteaceae was found – lowest needs for Protea which also had the greatest tolerance of very low fertiliser additions, compared to Grevillea robusta with much higher N requirements and strong foliage growth inhibition if nutrient levels are very low. Proteoid root growth on Grevillea rosmarinifolia only occurred at low nutrient levels and was not required for satisfactory foliage growth of container grown plants. Pot plants and seedlings, especially tomato, responded strongly to N and often there were positive NK interactions influencing foliage growth. Lime requirements were studied and in erica increasing lime rates depressed foliage growth. Comparative nutrition studies indicate that general or broad spectrum container media may be unsuitable for some groups of nursery plants and that they could be replaced by potting mixes designed to meet the widely differing needs of the species often grown. The number of specialist mixes would depend on the range of plants and be governed by management considerations.

A structure of the type long-line was used with main rope measuring 25m and 10 sec ondary strings of 2,5m installation to 200m of the coast with depth between 5m and 6m (tide evaporates). 12 seedlings were fastened by secondary rope with spacing of 20cm composing 12 depth lines. With 30 days it accomplished the measure of the weights of the seedlings of the structure identified by secondary rope and depth line. With 60 and 90 days the measure of the weights was just in the depth lines that presented seedlings in better physical conditions. In the tested depths from 0,2 to 2,4m, it was observed a better acting of the growth as well as better appearance and physical condition of the fastened seedlings from 0,2m to 1,2m, while fastened them from 1,4m to 1,8m or they had inferior growth or practically they didn't grow , and fastened them in the inferior limit from 2,0 to 2,4m reduced growth of the seedlings it was verified. The obtained medium weights presented significant statistical difference (F = 187,104; p = 0,05). TCE demonstrated negative correlation with the dept h (r = - 0,95; p = 0,05; n = 120). In the closest depths of the surface from 0,2 to 0,6m favored the growth of the algae reaching weights from 167,5 to 147,0g and TCE 2,92 and 2,69% day-1, among 0,8m to 1,2m weights from 139,0 to 124,0g and TCE fr om 2,29 to 1,90% day-1, and starting from 1,4m until 2,4m weights from 94,5 to 59,5g and TCE 0,9 to 0,53% day-1. The reduction in the weights below 1,2 m was quite accentuated evidencing that the algae had detriment in your growth with the reduction of the intensity brightness presenting reduced growth of some you change fastened in the extremities of the strings. The parameters of the water in the area of the cultivation temperature, salinity and degree of transparency of the water presented averages of 29,6 ± 2,41ºC, 35,6 ± 1,49‰ and 1,20 ± 0,76m. In the end of the experimental period (90 days) the seedlings reached final weight of 937,0g, ideal for crop. For Gracilaria birdiae, the brightness was the environmental factor that it affected your TCE and biomass production indeed, presenting in the conditions of the cultivation, satisfactory growth until the depth of 1,2m where the seedlings reached weight of 124,0 ± 9,94 g and TCE of 1,90 %dia-1. / A presente dissertação analisa o efeito da luminosidade no crescimento da alga marinha Gracilaria birdiae através da taxa de crescimento específico (TCE) e aumento de biomassa em diferentes profundidades. Foi utilizada estrutura do tipo long-line com corda principal medindo 25m e 10 cordas secundárias de 2,5m instalação a 200m da costa com profundidade entre 5 e 6 m (maré seca). Foram fixadas 12 mudas por corda secundária com espaçamento de 20cm compondo 12 linhas de profundidade. Com 30 dias realizou-se a pesagem de todas as mudas da estrutura identificadas por corda secundária e linha de profundidade. Com 60 e 90 dias a pesagem foi apenas nas linhas de profundidade que apresentavam mudas em melhores condições físicas. Nas profundidades testadas de 0,2 a 2,4m, observou-se um melhor desempenho do crescimento como também melhor aparência e condição física das mudas fixadas de 0,2 a 1,2m, enquanto as fixadas entre 1,4 a 1,8m ou tiveram crescimento inferior ou praticamente não cresceram, e as fixadas no limite inferior entre 2,0 e 2,4m verificou-se atrofiamento das mudas. Os pesos médios obtidos apresentaram diferença estatística significativa (F = 187,104; p = 0,05). A TCE demonstrou correlação negativa com a profundidade (r = - 0,95; p = 0,05; n = 120). Nas profundidades mais próximas da superfície de 0,2 a 0,6m favoreceram o crescimento das algas alcançando pesos de 167,5 a 147,0g e TCE de 2,92 e 2,69% dia-1, entre 0,8 a 1,2m pesos de 139,0 a 124,0g e TCE de 2,29 a 1,90% dia-1, e a partir de 1,4m até 2,4m pesos de 94,5 a 59,5g e TCE de 0,9 a 0,53% dia-1. A redução nos pesos abaixo de 1,2m foi bastante acentuada evidenciando que as algas tiveram detrimento em seu crescimento com a redução da intensidade luminosidade apresentando atrofiamento de algumas mudas fixadas nas extremidades das cordas. Os parâmetros da água na área do cultivo temperatura, salinidade e grau de transparência da água apresentaram médias de 29,6 ± 2,41 ºC, 35,6 ± 1,49 ‰ e 1,20 ± 0,76 m. No final do período experimental (90 dias) as mudas atingiram peso final de 937,0g, ideal para colheita. Para Gracilaria birdiae, a luminosidade foi o fator ambiental que afetou efetivamente sua TCE e produção de biomassa, apresentando nas condições do cultivo, crescimento satisfatório até a profundidade de 1,2m onde as mudas atingiram peso médio de 124,0 ± 9,94 g e TCE de 1,90 %dia-1.

Carcinoma adenóide cístico é uma neoplasia maligna de glândula salivar com alto grau de recorrência e metástase. Células tumorais que invadem tecidos subjacentes formam invadopódios, protrusões de membrana ricas em actina, cortactina e MT1-MMP capazes de degradar a matriz extracelular (MEC). Proteólise de moléculas da MEC, como a laminina, promove liberação de fragmentos e peptídeos bioativos. Nesse trabalho, estudamos o papel dos peptídeos AG73 e C16, da laminina-111, na atividade de invadopódios de células derivadas de carcinoma adenóide cístico (CAC2). Nossos resultados mostram que AG73 e C16 regulam atividade de invadopódio e aumentam a expressão de MT1-MMP em células CAC2. Silenciamento da integrina <font face=\"Symbol\">b1 e inibição da via ERK diminuem a atividade de invadopódio induzida por esses peptídeos. Já a inibição da via Rac diminui a atividade de invadopódios induzida apenas por AG73. Propomos que os peptídeos AG73 e C16 regulariam a atividade de invadopódios através da integrina <font face=\"Symbol\">b1 e da via ERK 1/2. Já a via Rac1 transduziria sinais gerados apenas pelo peptídeo AG73. / Adenoid cystic carcinoma is a frequently occurring malignant salivary gland neoplasm with high level of recurrence and metastasis. Tumor cells that invade surrounding tissues rely on invadopodia to degrade extracellular matrix (ECM). Invadopodia are actin and cortactin-rich membrane protrusions that localize MT1-MMP required for ECM degradation. Breakdown of ECM molecules, such as laminins, releases fragments and bioactive peptides. Here we addressed the role of laminin-111 peptides AG73 and C16 in invadopodia activity of cells derived from human adenoid cystic carcinoma (CAC2). Our results show that AG73 and C16 increase invadopodia activity and MT1-MMP expression in CAC2 cells. Silencing of <font face=\"Symbol\">b1 integrin and ERK pathway inhibition decrease AG73 and C16-induced invadopodia. Rac1 pathway inhibition decreases only AG73-induced invadopodia formation. We propose that AG73 and C16 increase invadopodia activity in CAC2 cells through <font face=\"Symbol\">b1 integrin. ERK1/2 pathway would transduce signals generated by both peptides, while Rac1 pathway is related to AG73 signaling.

Gliomas são tumores do sistema nervoso central e o glioblastoma multiforme (GBM) é sua forma mais comum e de pior prognóstico. A proliferação desordenada, migração estão entre os processos biológicos que conferem ao GBM um comportamento altamente agressivo. O objetivo deste estudo foi analisar in vitro o papel de PGE2 na proliferação, migração e apoptose da linhagem de glioma humano T98G através de tratamento com ácido gama-linolênico (GLA), Ibuprofeno (IBU) e adição de prostaglandina E1 (PGE1) e prostaglandina E2 (PGE2) exógeno. Nossos resultados demonstraram através uma diminuição nas taxas de proliferação e de migração e uma indução de apoptose nas células T98G tratadas com GLA ou IBU. Por outro lado, a adição exógeno de PGE1 ou PGE2 resultou num aumentou da proliferação e da migração e numa inibição da apoptose. Esses resultados demonstram a influência de PGE2 na linhagem T98G. / Gliomas are tumors of the central nervous system and glioblastoma multiforme (GBM) is the most common form with the worst prognosis. Uncontrolled cell proliferation, migration are among the biological processes that give GBM a highly aggressive behavior. The aim of this study was to analyze in vitro the role of PGE2 the proliferation, migration and apoptosis of the T98G human glioma cell line through treatment with gamma-linolenic acid (GLA), ibuprofen (IBU) and the addition of exogenous prostaglandin E1 (PGE1) and prostaglandin E2 (PGE2). Our results demonstrated through various tests a decrease in the rates of proliferation and migration and an induction of apoptosis in T98G cells treated with GLA or IBU. On the other hand, the addition of exogenous PGE1 or PGE2 resulted in increased proliferation and migration and inhibition of apoptosis. These results demonstrate the influence of PGE2 in the T98G cell line.

Problemas reprodutivos ocorrem no hipo e no hipertireoidismo, mas pouco se conhece a respeito das bases moleculares destas alterações. Avaliou-se o efeito da administração de triiodotironina na atividade do gonadotrofo em ratos Wistar tireoidectomizados. O hipotireoidismo ocasionou elevação no TSH e do LH sérico, aumento do conteúdo do mRNA e redução do conteúdo protéico do LH e do FSH, redução da cauda poli (A), redução da testosterona e do FSH séricos, redução da expressão do receptor de LH, aumento da expressão do receptor de andrógenos testiculares e epididimais. O tratamento com T3 acarretou a diminuição do mRNA e aumento do conteúdo protéico de LH, aumento do FSH sérico, diminuição do conteúdo do mRNA e aumento do conteúdo protéico do FSH, aumento da expressão do receptor de LH, redução da expressão de receptores de andrógenos testiculares e epididimais. Esses resultados sugerem que o T3 age na modulação da função do gonadotrofo e na modulação da expressão dos receptores em tecidos-alvo. / Reproductive disorders occur in hypo and hyperthyroidism, but little is known about the molecular basis of these alterations. The effect of administration of triiodothyronine (T3) on the activity of gonadotropes in thyroidectomyzed male Wistar rats was evaluated. Hypothyroidism caused increased on TSH and LH serum concentrations, increased of mRNA content and decreased in protein content of LH and FSH, reduction in the length of poli(A) tail of LH, reduction in testosterone and FSH serum concentrations, reduction in the expression of LH receptor, increased in the expression of androgen receptors in the testis and epididymis. T3 treatment caused reduction on mRNA and increased on mRNA of LH and FSH, increased serum FSH, increased the expression of LH receptor, and reduction of the expression of androgen receptors in the testis and epididymis. These results suggest that T3 modulates the function of gonadotrope and the expression of the receptors in target-tissues.

Analysis of dynamical interactions of axon shafts and their biophysical modelling / Analyse des interactions dynamiques des corps d'axones et leur modélisation biophysique

La fasciculation des axones joue un rôle essentiel dans le développement des réseaux neuronaux. Cependant, la dynamique de la fasciculation axonale, ainsi que les mécanismes biophysiques à l’œuvre dans ce processus, demeurent encore très mal compris. En vue d'étudier les mécanismes de fasciculation d'axones ex vivo, nous avons développé un système modèle simple, constitué par des explants d'épithélium olfactif de souris embryonnaires en culture, à partir desquels poussent les axones des neurones sensoriels olfactifs. Grâce à une étude en vidéomicroscopie, nous avons observé que ces axones interagissent de façon dynamique par leur fibre, à la manière de fermetures éclair pouvant se fermer ("zippering") ou s'ouvrir ("unzippering"), ce qui conduit respectivement à la fasciculation ou à la défasciculation des axones. Mettant à profit cette nouvelle préparation expérimentale pour l'étude des interactions dynamiques entre axones, nous avons développé une analyse biophysique détaillée des processus de zippering/unzippering.Nous mettons en évidence dans notre travail l'existence d'un mécanisme biophysique cohérent de contrôle des interactions locales entre fibres axonales. Ce mécanisme local est à mettre en relation avec les changements de la structure globale du réseau axonal (degré de fasciculation) qui s'opèrent sur une échelle temporelle plus longue. Enfin, nous discutons la signification fonctionnelle de nos observations et analyses, et proposons un nouveau rôles de la tension mécanique dans le développement du système nerveux : la régulation de la fasciculation des axones et, en conséquence, de la formation des cartes topologiques au sein des réseaux neuronaux. / While axon fasciculation plays a key role in the development of neural networks, very little is known about its dynamics and the underlying biophysical mechanisms. In a model system composed of neurons grown ex vivo from explants of embryonic mouse olfactory epithelia, we observed that axons dynamically interact with each other through their shafts, leading to zippering and unzippering behaviour that regulates their fasciculation. Taking advantage of this new preparation suitable for studying such interactions, we carried out a detailed biophysical analysis of zippering, occurring either spontaneously or induced by micromanipulations and pharmacological treatments.We show that there is a consistent mechanism which governs local interactions between axon shafts, supported by broad experimental evidence. This mechanism can be reconciled with changes in global structure of axonal network developing on slower time scale, analogically to well-studied relation between local relaxations, and topological changes and coarsening in two-dimensional liquid foams. We assess our observations and analysis in light of possible in vivo functional significance and propose a new role of mechanical tension in neural development: the regulation of axon fasciculation and consequently formation of neuronal topographic maps.

A maioria dos tumores sólidos apresentam características aneuplóides. Porém a relação entre aneuploidia e transformação maligna, ainda não está definida. Nos últimos anos diversas proteínas têm sido descritas como reguladoras de eventos durante a divisão celular, principalmente as relacionadas com a formação do fuso bipolar e segregação equacional dos cromossomos. Neste estudo propomo-nos a analisar os efeitos da interferência em dois pontos críticos da mitose, a segregação cromossômica e a citocinese, em relação à aneuploidia e à instabilidade genética tumoral. Nossos dados mostraram que o tratamento sequencial de Monastrol e Blebistatina determinou o surgimento de fusos mitóticos anormais, amplificação centrossômica, localização ectópica de Aurora A e aumento de micronúcleos. Esta interferência pode levar a um quadro de instabilidade genética e, consequentemente a progressão tumoral, abrindo novas possibilidades para o estudo dos mecanismos moleculares envolvidos na regulação do ponto de checagem mitótico e resistência a quimioterápicos. / Most solid tumors have aneuploid feature. Therefore the relationship between aneuploidy and malignant transformation is not yet understood. In the last years it has been described many proteins involved in regulation of mitosis, mainly those related to bipolar spindle and chromosome segregation. In this work we propose to study the effects of the interference on two mitotic critical points, the chromosome segregation and cytokinesis, in relation to aneuploidy and genetic tumor instability. Our data showed that sequential treatment with Monastrol and Blebbistatin led to abnormal mitotic spindle, centrosome amplification, Aurora A ectopic and micronucleus increased. This interference can lead to genetic instability and may be involved in a tumor progression, opening news possibilities to study the molecular mechanisms involved in regulation the checkpoint mitotic and resistance to chemotherapy found in genetically unstable cells.

O câncer cervical é o terceiro tipo de câncer mais comum entre as mulheres, sendo a infecção por HPV16 o principal fator de risco para sua etiopatogênese. Neste trabalho avaliamos o potencial profilático e terapêutico da vacina proteica E6E7, constituída de epítopos imunogênicos de E6 e E7 do HPV16, com ou sem Ubiquitina (Ub) fusionada (E6E7Ub e UbE6E7), contra tumor de células TC-1. A vacina E6E7 conferiu total proteção contra a formação de tumores em animais C57BL/6 selvagens, CD4 nocautes e TLR4 deficientes previamente imunizados e posteriormente desafiados com células TC-1. Entretanto, em animais imunizados após desafio, a fusão da Ub no antígeno E6E7 foi crucial para a vacina promover a regressão do tumor em 60% dos animais. Além da fusão da Ub na extremidade N-terminal do antígeno (UbE6E7) aumentar a poliubiquitinação in vitro, essa estratégia de fusão promoveu uma melhor resposta imunoterapêutica contra o tumor que E6E7 e E6E7Ub, sendo promissora para o desenvolvimento de vacinas terapêuticas contra câncer cervical e também contra outros tipos de câncer. / Cervical cancer is the third most common cancer women worldwide and HPV16 infection is a major risk factor for its ethiopathogenesis. In this study we evaluated in mice the prophylactic and therapeutic potential of the E6E7 proteic vaccine, consisting of immunogenic HPV16 E6 and E7 epitopes, with or without Ubiquitin-fused (E6E7Ub and UbE6E7), against TC-1 tumor cells. The E6E7 vaccine conferred complete protection against tumor growth in previously immunized wild-type, CD4 knockout and TLR4 deficient mice challenged with TC-1 cells. However, immunization of previously challenged animals with TC-1 cells, Ubiquitin-fused antigens promoted tumor regression in 60% of mice against 0% in non-Ubiquitinfused antigens. Besides, the fusion of Ubiquitin at the N-terminus of the antigen (UbE6E7) seems to enhance polyubiquitination in vitro, this Ubiquitin fusion strategy promoted an effective immunotherapeutic response against the tumor, being promising for the development of therapeutic cervical cancer vaccines and also other types of tumor.

Vertikalt växthus i Kiruna : Med spillvärme från LKABs gruvindustri / Vertical greenhouse in Kiruna : Utilizing waste heat from LKAB's mining industry

Städerna växer och samtidigt ökar efterfrågan på närodlade och ekologiska grödor. För att klara av att producera närodlat och mer hållbart skulle ett alternativ vara att odla i så kallade vertikala växthus. I Kiruna finns Sveriges största malmgruva som drivs av företaget LKAB vars olika processer leder till spillvärme. I Kiruna är klimatet kallt jämfört med större delen av Sverige och därför erfordras uppvärmning om odling ska kunna ske året runt i ett växthus. Examensarbetets uppgift är därför att undersöka hur man kan utnyttja spillvärme till ett vertikalt växthus i samband med en ny kontorsbyggnad på LKABs gruvområde. LKABs nya kontorsbyggnad har ett kvadratiskt avtryck på marken med ett hörn i sydlig riktning. Fördelningen mellan växthus och kontor kan förenklas genom att kvadraten delas på diagonalen där den södra halvan är växthus och den norra halvan kontor. Eftersom solförhållandena för odling är speciella i Kiruna har växthusets väggar en lutning som är anpassad efter solens låga infallsvinkel vilket gör att växthusets area minskar med varje våningsplan. Som yttermaterial till växthuset används glas och som stommaterial används stål. Ett hydroponiskt odlingssystem används där plantorna sätts direkt i en cirkulerande näringslösning och på så sätt kan systemet utformas med horisontella odlingsrör i flera vertikala odlingsplan. För uppvärmning av växthuset kommer spillvärme från LKABs verksamhet att utnyttjas, i examensarbetet undersöks två olika alternativ av spillvärmekällor och resultatet är att båda de alternativ som undersökts kan utnyttjas för uppvärmning av hela växthusdelens volym. / While the cities are expanding the demand for locally grown and organic crops is increasing. To be able to produce locally and more sustainable crops, one option could be to grow in a so-called vertical greenhouse. In Kiruna the largest ore mine in Sweden is operated by the company LKAB. Various processes in the mining industry lead to waste heat. In Kiruna, the climate is cold compared to most parts of Sweden, and therefore requires heating for the cultivation to be able to take place all year round in a greenhouse. The project’s task is to explore how to utilize waste heat from the mine to a vertical greenhouse in the context of a new office building at LKAB's mining area. LKAB's new office building has a square footprint on the ground with one of the corners in the south direction. The division between the greenhouse and the office can be simplified by the square divided diagonally where the southern half is the greenhouse and the northern half the office. Since the sunlight is limited in Kiruna the greenhouse walls has been design to adjust to the sun’s low position. The sun’s low position requires a sloped facade in the south direction. The greenhouse’s floor area decreases with each floor. As the external material for the greenhouse glass is used and as framing material steel is used. A hydroponically system is used where the seedlings are put directly in a circulating nutrient solution and in this way the system can be designed with horizontal pipes in several vertical cultivated floors. The greenhouse will be heated with waste heat from LKAB's industry, the project examines two alternatives of waste heat sources, and the result is that both of the alternatives studied can be used to heat the entire volume of the greenhouse.
