Previous issue date: 2017-08-11 / O objeto do estudo foi a situação vacinai dos Agentes Comunitários de Saúde (ACS) que atuam nas Unidades de Atenção Primária a Saúde (UAPS) de um município da zona da mata mineira. As UAPS estão voltadas para atividades de promoção da saúde e controle de riscos. As equipes de trabalho das UAPS são multidisciplinares sendo compostas por profissionais de áreas diversas e, entre esses, há a presença do ACS, profissional responsável pelo cadastramento das famílias, mapeamento das áreas, identificação de riscos à saúde da comunidade, pela orientação da população para a promoção da saúde e por promover educação sanitária e ambiental, dentre outras atribuições. Desse modo, ele entra em contato com pessoas sadias e também com aquelas acometidas por alguma doença, se expondo a agentes patogênicos. Esta variedade de atribuições, as peculiaridades e complexidade de suas atividades justificam a realização de pesquisas que abordem esses profissionais. Este estudo teve como objetivo analisar a situação vacinai dos ACS das UAPS de um município da Zona da Mata Mineira. Trata-se de um estudo transversal e quantitativo contando com 400 participantes. A coleta de dados ocorreu em dois períodos, sendo o primeiro de julho a outubro de 2015 e o segundo, de outubro de 2016 a fevereiro de 2017. Os ACS dessa investigação foram predominantemente do sexo feminino (91,3%), média de idade de 46 anos, casados ou vivendo em união estável (57,5%), brancos (46,3%), referindo religião católica (58%) e, em sua maioria, possuindo Ensino Médio completo (75,4%). A análise da cobertura vacinai permitiu constatar que 205 (51,3%) ACS possuíam o esquema completo das vacinas exigidas para os trabalhadores da saúde, sendo que 351 (87,8%) estavam vacinados com a 3a dose de Hepatite B, 267 (66,8%) com um reforço da Tríplice Virai, 337 (84,3%) com um reforço da Dupla adulto, 342 (85,5%) contra Influenza, e 308 (77%) com 1 dose e 1 reforço de vacina contra Febre amarela. Na análise bivariada observou-se p-value significativo (<0,05) ao relacionar as variáveis Faixa Etária e Vacina Triplice Virai e Tempo de Trabalho com Hepatite B. A análise multivariada demonstrou que há maior probabilidade do ACS estar com a vacinação em dia se negro, católico ou evangélico, se possui o cartão de vacinação, se realizou o teste AntiHbs, se recebeu orientação em relação às vacinas que deve manter em dia enquanto trabalhador da área da saúde e se trabalha nas regiões Nordeste, Leste, Centro, Oeste ou Rural do município. O estudo permitiu constatar que os participantes, em sua maioria, se preocupavam com ações referentes à vacinação, porém encontravam dificuldades para manterem seus cartões atualizados havendo a necessidade de abordar a temática sobre as doenças que podem ser adquiridas no trabalho do ACS e que são passíveis de prevenção por meio da imunização. / With this study, we aimed at the vaccination situation of the Community Health Agents
(ACS, in Portuguese) that work in the Primary Health Care Units (UAPS, in Portuguese)
of a city in the region of Zona da Mata of Minas Gerais. The UAPS are focused on health
promotion and risk control activities. The work teams of the UAPS are multidisciplinary
and are composed of professionals from different disciplines, among them there is the
ACS, a professional responsible for the registration of families, mapping of areas,
identification of community health risks, orientation of the population for promotion and to
promote health and environmental education, among other attributions. In this way, they
come in contact with healthy people and also with those affected by some disease,
exposing themselves to pathogens. This variety of attributions, the peculiarities and the
complexity of their activities are possible reasons to make people conduct research that
addresses these professionals. The main purpose of this study to analyze the
vaccination situation of the ACS of the UAPS of a city in the Zona da Mata of Minas
Gerais. It is a cross-sectional, quantitative study with 400 participants. Data collection
took place in two periods, the first from July to October 2015 and the second from
October 2016 to February 2017. The ACS of this investigation were predominantly
female (91.3%), mean age 46 year-old, white (46.3%), catholic religion (58%) and most
of them having completed high school (75.4%). The analysis of immunization coverage
showed that 205 (51.3%) ACS have the complete vaccination schedule required for
health workers, and 351 (87.8%) are vaccinated with the third dose of Hepatitis B, 267
(66.8%) with a Triple Viral booster vaccine, 337 (84.3%) with an Adult double booster
vaccine, 342 (85.5%) against Influenza during the last year's campaign, and 308 (77%)
with 1 Dose and 1 booster vaccine against Yellow fever. In the bivariate analysis,
significant p-value (<0.05) was observed when the variables Age Range, Viral Triplice
Vaccine and Working Time with Hepatitis B were analyzed. The multivariate analysis
showed that there is a higher probability of ACS to have the vaccination up to date if they
are black, catholic or evangelical, if they have the vaccination card, if they underwent the
AntiHbs test, if they received orientation regarding the vaccines they should keep up as
a health worker and if they work in the regions Northeast, East, Central, West or Rural.
The study showed that the majority of the participants are concerned with actions related
to vaccination, but they find it difficult to keep their cards up to date and there is a need
to address the issue of diseases that can be acquired in the work of ACS and that are
susceptible of prevention through immunization.

Les anticorps monoclonaux anti-cytokine ont constitué une révolution dans le traitement des maladies inflammatoires chroniques, mais leur utilisation présente des inconvénients (non réponse, résistance, effets secondaires, coûts élevés).Notre équipe développe une stratégie alternative originale, l’immunisation active à base de peptides de cytokines. Elle a pour but de faire synthétiser, par l’organisme même du patient, des anticorps neutralisant les effets pathogènes dus à l’excès de cytokines.Durant ma thèse, j’ai montré que l’immunisation active contre un peptide dérivé de l’IL-6 murine est protectrice dans un modèle murin de sclérodermie systémique. L’immunisation de singes avec l’équivalent humain entraîne une réduction significative des réactions inflammatoires locales suite à l’induction d’une réaction d’HSR. De plus, l’immunisation active contre deux peptides dérivés de l’IL-1β et de l’IL-23 conduit à la réduction de la sévérité de l’EAE.Ces résultats confortent l’intérêt de cibler les cytokines par l’approche d’immunisation active à base de peptides, qui pourra permettre de diversifier l’offre thérapeutique actuellement disponible. / Monoclonal antibodies have been a revolution for the treatment of chronic inflammatory diseases, but their use shows major drawbacks (non-response, resistance, side effects and prohibitive costs).Our team develops an original alternative strategy: anti-cytokine peptide-based active immunization.The aim of the approach is to make the patient’s own organism produce antibodies capable of neutralizing the pathogenic effects of cytokine overproduction.During my PhD, I have demonstrated that active immunization against an IL-6 murine peptide confers clinical protection in a murine model of systemic sclerosis. Monkeys immunized against the human peptide also showed a significant decrease of local inflammatory reactions following a delayed-type hypersensitivity reaction. Moreover, active immunization against an IL-1β and an IL-23 murinepeptide led to a reduction of the severity of the EAE in mice.These results comfort the interest of anti-cytokine peptide-based active immunization, which should eventually widen the choice of therapeutics available for the patients.

Acceptance of an Emergently Released Vaccine by the General Public: 2009 H1N1 Influenza Pandemic VaccineNguyen, Trang January 2012 (has links)
The recent experience with the 2009 H1N1 pandemic has drawn attention to the need to better understand the public’s response to emergently released vaccines (ERV). This study applied a mixed methods approach to examine the causal pathways underlying the vaccination behaviour during a public health emergency. The integrated evidence from empirical and theoretical-based findings highlights a number of factors to consider in interventions to improve vaccination rates with an ERV. These factors include: 1) providing clear risk messages around the disease and the ERV, 2) improving accessibility to the vaccine, 3) encouraging primary healthcare providers to provide recommendations for vaccination, 4) implementing strategies to increase seasonal influenza vaccination prior to the next public health emergency, 5) developing strategies to target sub-populations more reluctant to accept an ERV. Developing theory-based interventions that are behaviour-specific may be more likely to result in behaviour change within the public in future emergency vaccination campaigns.

Using Mathematical Modelling to Evaluate Human Papillomavirus Vaccination Programs in CanadaRogers, Carley January 2013 (has links)
Mathematical models provide unique insights to real-world problems. Within the context of infectious diseases, models are used to explore the dynamics of infections and control mechanisms. Human papillomavirus (HPV) globally infects about 630 million people, many of these infections develop into cancers and genital warts. Vaccines are available to protect against the most prevalent and devastating strains of HPV. The introduction of this vaccine as part of a national immunization program in Canada is a complex decision for policy-makers in which mathematical models can play a key role. We use the current recommendations provided by the World Health Organization to explore the integral role mathematical models have in the decision to incorporate the HPV vaccine within a national immunization program. We then provide a review of the literature discussing the role of mathematical models in the decision to include a vaccine in a national immunization program within the context of the HPV vaccine. Next, we evaluate the current standing of mathematical models used within the context of HPV immunization, to highlight the types of models used, underlying assumptions and general recommendations made about these immunization programs. Then, we create and analyze a model to explore the possibility of bettering the current HPV vaccine strategy in Canada. We focus on the effects of the grade of vaccination and the number of doses required to eradicate the targeted strains of HPV.

O habitat natural da Neisseria meningitis é a nasofaringe humana e a transmissão da bactéria é por contato direto ou por inalação de partículas durante a fase de transmissão N. meningitis é uma bactéria Gram-negativa responsável por uma significante mortalidade em todo o mundo. Embora existam vacinas polissacárides contras os sorogrupos A, C, W135 e Y , não há uma vacina adequada para crianças menores de 4 anos para o sorogrupo B. Estudos estão sendo direcionadas para pesquisa de antígenos vacinais que são derivados da proteínas de membrana externa(NOMV). Entretanto vacinas baseadas em NOMV são consideradas pouco imunogênicas , fazendo com que o uso de adjuvantes seja necessário. Este estudo investiga a imunogenicidade da NOMV de N. meningitidis administrada pela via intranasal/intramuscular em camundongos neonatos BALC/c, usando proteína de membrana externa (NOMC) obtido de uma cepa epidêmica de N. meningitidis B:4:P1:15. As cepas usadas para imunização dos camundongos foram selecionadas por colony-blot, usando anticorpo monoclonal anti L3,7,9 para maior expressão do LPS contra o imunotipo L3,7,9 presente na cepa (B:4:P1:15 3,7,9). Como adjuvantes de mucosa foram utilizados Bordetella pertussis (células íntegras) ou sobrenadante de cultura com 48 horas ou hidróxido de alumínio [Al(OH)3]. O soro dos camundongos imunizados foram analisados pelo método de ELISA à fim de se comparar os diferentes adjuvantes utilisados. O índice de avidez também foi determinado. IgG e IgM foram detectados nos soros dos camundongos após imunização, com índices de intermediária e alta avidez. Todos os adjuvantes foram capazes de aumentar a resposta imune contra NOMV de N.meningitidis. A via intranasal foi adequada para sensibilizar as células do sistema imune que foram rapidamente estimuladas pela via intramuscular usando os adjuvantes utilizados na presente investigação. Dados sugerem que o estudo da NOMV é importante na indução da imunidade de mucosa para N. meningitidis B, e que a qualidade e magnitude da resposta imune gerada pelas vacinas de mucosa são influenciadas tanto pelo adjuvante como pelo antígeno. Concluímos que NOMV juntamente com adjuvantes de mucosa tem considerável potencial no desenvolvimento de vacinas contra o meningococo do sorogrupo B. / The natural habitat of Neisseria meningitidis is the human nasopharynx, and the bacterium is transmitted by direct mouth-to-mouth contact or by the inhalation of released mucous particles during close contact. N meningitidis is a Gram-negative bacterium responsible for significant mortality worldwide. While effective polysaccharide-based vaccines exist against serogroups A, C, W135, and Y, no similar vaccine is suitable for children under 4 years against disease caused by serogroup B strains. Current studies are searching for vaccinal antigens that are derived from the native outer membrane (NOMV). However, vaccines based on NOMV are considered weak, making the use of adjuvants necessary. This study investigated the immunogenicity of NOMV of N. meningitidis administered intranasal/intramuscular in neonate BALB/c mice, using the native outer membrane complex (NOMC) obtained from an epidemic strain of N. meningitidis B:4:P1.15. The strains used for immunization of mice were selected by colony-blot, using anti L3,7,9 monoclonal antibodies, for the highest expression of LPS among the immunotypes (B:4:P1:15 L9á). As mucosal adjuvants, we used Bordetella pertussis (whole cells) or the supernatant of 48 h culture of this bacterium, followed by an intramuscular dose of the same protein adsorbed onto , B. pertussis (whole cells) or 48-h B. pertussis culture supernatant or aluminum hydroxide [Al(OH)3]. Sera of immunized mice were evaluated by ELISA in order to compare the different adjuvants used. We also determined their avidity index. IgG and IgM were detected in the serum of mice after immunization, with avidity indices that ranged from intermediate to high. All adjuvants were capable of increasing the immune response against NOMV of N. meningitidis in the homologous prime/boost schedule used. The intranasal route was suitable for sensitizing the cells of the immune system which were quickly stimulated by the intramuscular route using the adjuvants analysed in the present invertigation. Data suggest that the NOMV studied is important in the induction of mucosal immunity to N. meningitidis B, and that the quality and magnitude of the immune responses generated by mucosal vaccines are influenced by the adjuvant as well as the antigen. In conclusion, nasal delivery of NoMV with mucosal adjuvants has considerable potential in the development of a mucosal vaccine against serogroup B meningococci.

As proteínas de classe 3 são candidatas na preparação de uma vacina contra a doença meningocócica. O objetivo deste estudo é determinar a imunogenicidade da proteína de classe 3 purificada da cepa de Neisseria meningitidis do sorogrupo B juntamente com a capacidade adjuvante de whole cells de Bordetella pertussis. Foram imunizados camundongos BALB/c neonatos em um intervalo de 3 a 12 dias entre 1 e 4 doses da proteína de classe 3 mais adjuvante, pela via intranasal e no 21º dia pela via intramuscular com a proteína de classe 3 emulsificada com hidróxido de alumínio. Os resultados demonstraram que após 2 doses pela via intranasal e 1 dose pela via intramuscular houve rápido estímulo das células imunes nos camundongos adultos BALB/c e neonatos BALB/c e outbred. Todos os soros foram analisados por ELISA e immunoblot. O adjuvante B. pertussis administrado pelas vias intranasal ou intramuscular, aumentou a resposta imune comparada com os controles. Anticorpos bactericidas e de alta afinidade foram produzidos. / Proteins of class 3 sound candidates in the preparation of vaccine against meningococcal illness. The aim of this study was to determine the immunogenicity of class 3 proteins purified of Neisseria meningitidis of the serogroup B along with whole cells of Bordetella pertussis as adjuvant. BALB/c and outbred neonate mice between 3 and 12 days old were immunized with 1 to 4 doses of the purified class 3 proteins with or without adjuvant given by the intranasal route, and on the 21st day the animals received an intramuscular dose of the class 3 proteins with or without aluminum hydroxide. The results demonstrated that after 2 doses by the intranasal route and 1 dose intramuscular there was a rapid stimulation of the immune cells in BALB/c adult mice as well as BALB/c and outbred neonates mice. All sera were analyzed by ELISA and immunoblot. The adjuvant B. pertussis used in the present investigation and given via the intranasal or intramuscular route increased the immune response compared with the controls. High affinity and bactericidal antibodies were produced.

Flutuação de proteínas séricas e diarréia durante o processo de aquisição de proteção passiva em cabritas aleitadas com colostro caprino, bovino in natura e bovino liofilizado / Fluctuation of serum proteins and diarrhea during the process of acquisition of passive protection in newborn goat kids fed goat and bovine colostrum in natura and lyophilized bovineAnali Linhares Lima 22 June 2011 (has links)
Avaliou-se o efeito de diferentes fontes de colostro, caprino e bovino in natura, e bovino liofilizado em 25 cabritas distribuídas aleatoriamente em cinco tratamentos: CCA - colostro caprino adequado (45 a 55 mg/mL de IgG); CCB - colostro caprino baixo (15 a 25 mg/mL de IgG); CBA - colostro bovino adequado (45 a 55 mg/mL de IgG); CBB - colostro bovino baixo (15 a 25 mg/mL de IgG) e CBL - colostro bovino liofilizado (45 a 55 mg/mL de IgG). Os animais receberam 5% do peso vivo de colostro às 0,12 e 24 horas após o nascimento e, em seguida, leite de vaca duas vezes ao dia e concentrado ad libitum até 60 dias de idade. Amostras de sangue foram coletadas nos dias 0, ½, 1, 2 , 5, 10,15, 20, 25, 30, 35, 40, 50 e 60 de idade. O sangue foi analisado quanto ao valor de hematócrito e a concentração de proteína total (PT). As frações protéicas separadas por eletroforese e a condição de diarréia foi avaliada através de cor, consistência, presença de sangue/muco. As variáveis sanguíneas foram analisadas como medidas repetidas no tempo. Com relação ao valor de hematócrito efeito de período foi observado, o valor no primeiro dia de idade, 28,76±1,76, foi maior que nos 10, 30, 35 e 60 dias de idade, com valores médios de 23,88±0,98, 24,18±0,70, 23,73±0,74 e 24,53±0,79%, respectivamente. Interação entre tratamento e período foi verificado para PT sérica, os tratamentos CCB, CBA, CBB e CBL apresentaram aumento gradativo nas concentrações séricas até os 60 dias de idade, enquanto CCA apresentou valores nos dias 10,15, 40, 50 e 60 dias de idade maiores que o valor ao nascimento. Os valores de albumina sérica foram afetados pelo tratamento e período, sendo o valor em CBL, 2,39±0,12 g/dL, significamente inferior ao dos tratamentos CCA, CCB, CBA e CBB, com 3,20±0,12, 2,94±0,12, 3,25±0,12, 3,08±0,12 g/dL, respectivamente. Um aumento gradativo nas concentrações desta variável foi observado até os 60 dias de idade. Ao 0 e ½ dia de idade os valores de globulina sérica foram menores que os observados na última data experimental, 60 dias de idade. A relação albumina/globulinas sérica foi afetada pelo efeito de tratamento e período, com os valores em CBL significativamente superiores do que em CCA, CCB, CBA e CBB. A relação foi maior ao nascimento diminuindo nas datas subseqüentes, ½ e 1 dia de idade. A variável gamaglobulina apresentou efeito de tratamento e período, com valores em CBL significativamente superiores do que em CCB e CBB, não diferindo dos encontrados em CCA e CBA. Os valores ao 0 e ½ dia de idade foram menores que aos 60 dias de idade. Observando a condição de diarréia nos tratamentos verificou-se que os animais CCB e CBB apresentaram maior frequência de diarréia que os animais dos demais tratamentos, indicando uma relação inversa entre anticorpos no colostro e condição de diarréia. Os resultados indicam que o colostro bovino liofilizado pode ser utilizado para caprinos recém-nascidos como fonte alternativa para a aquisição de proteção inicial. / It was to evaluated the effect of different sources of colostrum, goat and bovine in natura, and lyophilized bovine in 25 newborn female goat kids were randomly allocated to five treatments: GCA - goat colostrum adequate (45 to 55 mg/mL of IgG), GCL - goat colostrum low (15 to 25 mg/mL of IgG), BCA - bovine colostrum adequate (45 to 55 mg/mL of IgG), LBC - bovine colostrum low (15 to 25 mg / mL of IgG) and of treatment CBL, lyophilized bovine colostrum (45 to 55 mg / mL of IgG). The animals received 5% of body weight of colostrum at 0, 12, 24 hr after birth and then, bovine milk twice a day and concentrate ad libitum until 60 days of life. Blood samples were collected in 0, ½, 1, 2 , 5, 10,15, 20, 25, 30, 35, 40, 50 e 60 days of age. Blood was analyzed for hematocrit value and total protein (TP). Protein fractions were separated by electrophoresis and the condition of diarrhea was evaluated by color, consistency, blood/mucus presence. Blood variables analyzed as repeated measures on time.Related to hematocrit values effect of period was observed, the value at one day of age, 28.76±1.76, was higher than at 10, 30, 35 and 60 days of age, with average at 23.88±0.98, 24.18±0.70, 23.73±0.74 and 24.53 ± 0.79%, respectively. Interaction between treatment and period was observed for serum TP, treatments GCL, BCA, BCL, and LBC showed a gradual increase in serum concentrations up to 60 days of age, while GCA showed values on days 10, 15, 40, 50 and 60 days of age, greater than the value at birth. The values of serum albumin were affected by treatment and period, being the value in LBC, 2.39±0.12 g/dL, significantly lower than the GCA treatment, GCL, BCA and BCL, with 3.20±0.12 , 2.94±0.12, 3.25±0.12 and 3.08± 0.12 g/dL, respectively. A gradual increase in the concentrations of this variable was observed until 60 days of age. To 0 and ½ day of age values of serum globulin were significantly lower than the observed at last experimental date, 60 days of age. Serum albumin/globulin relation was affected by the treatment and period with the values in CBL significantly higher than observed in GCA, GCL, BCA and BCL. The relation was higher at birth decreasing in the following dates, ½ and 1 day of age. The variable gamma globulin was affected by treatment and period, with values in LBC significantly higher than in GCL and BCL, not differing from the found in GCA and BCA. The values at 0 and ½ day of age were lower than 60 days of age. Observing the diarrhea condition, in the treatments, it was verified that the GCL and BCL animal had showed higher diarrhea incidence than the animals from the other treatments, indicating an inverse relationship between antibodies in the colostrum and condition of diarrhea. The results indicate that lyophilized bovine colostrum can be used to newborns goat kids as an alternative source for the acquisition of initial protection.

Introduction: Complete childhood Immunization remains the most effective way for prevention of Vaccine Preventable Diseases. The 2016 Uganda Demography Health Survey (UDHS) reported that only 55% of children aged 12-23 months had been fully vaccinated. The relationship between maternal education and childhood immunization among Ugandan children remains unclear with most of the studies done being limited in scope. Objective: To investigate the association between maternal education and childhood immunization in Uganda. Methods: This study was based on analysis of data from the UDHS. The study included 2815 children aged 12 – 23 months. Permission and data to conduct the analyses was sought from the DHS Program website. Bivariate and multivariate logistic regression models were used to assess association between the maternal education and full immunization. Adjusted odds ratios (OR) with their 95% confidence intervals (CI) were reported. A p value <0.05 was considered statistically significant. Results: Association between maternal education and childhood immunization was statistically significant (Primary Education - OR = 0.50 95%CI: 0.32 - 0.77, P value = 0.002 and Secondary Education – OR = 0.62 95%CI: 0.39 – 0.97, P value = 0.038). ANC visits, possession of a Child Health Card also had a strong effect on Childhood Immunization. Conclusion: Lower maternal education is associated with reduced completion of childhood immunization. To promote childhood immunization Parents with education lower than Secondary School should be targeted and the usefulness of ANC visits should be emphasized.

Avaliação do programa de uso da imunoglobulina palivizumabe no Estado de São PauloGonçalves, Ivana Regina January 2016 (has links)
Orientador: Cristina Maria Garcia de Lima Parada Parada / Resumo: O programa de uso da imunoglobulina palivizumabe teve início em 2007 no Estado de São Paulo e em 2013 estendeu-se nacionalmente, sendo destinado a crianças com maior risco para a infecção pelo Virus Sincicial Respiratório (VSR). Desde seu início, o programa permanece sem avaliação sistemática. O objetivo dessa pesquisa foi avaliar o programa de uso da imunoglobulina palivizumabe no Estado de São Paulo. Foram avaliados os 16 locais de aplicação: cinco Centros de Referência para Imunobiológicos Especiais (CRIE) e 11 postos de aplicação existentes no Estado de São Paulo, seguindo o referencial de avaliação proposto por Donabedian. Dados de estrutura e processo foram obtidos por entrevista com os responsáveis pela aplicação da imunoglobulina; para avaliação de resultado foram entrevistadas as mães das crianças do programa, visando identificar a associação entre falha na tomada do palivizumabe e a necessidade de hospitalização em Unidade de Terapia Intensiva por doença ou sintomatologia respiratória no período de sazonalidade do VSR. Participaram desta pesquisa 693 crianças/responsáveis (85,1% da população elegível), que ingressaram no programa entre março e agosto de 2014, sendo a coleta de dados realizada entre março e setembro do mesmo ano. Para análise de resultado foi utilizado o odds ratio pontual e intervalar com intervalo de confiança de 95%, considerando-se associações estatisticamente significativas se p < 0,05. Ma thèse a permis de développer une nouvelle approche pour étudier, par imagerie in vivo, le comportement des cellules présentatrices d’antigènes (CPA) de la peau suite à la vaccination par voie intradermique chez le primate non-humain. Le ciblage des CPA a été réalisé par injection in vivo d’un anticorps monoclonal anti-HLA-DR fluorescent. L’effet sur les CPA d’un adjuvant (R-848, ligand du TLR7/8) et l’immuno-ciblage des cellules de Langerhans par une protéine de fusion vaccinale anti-VIH (anti-langérine-VIHGag) ont ainsi été évalués par imagerie in vivo, vidéomicroscopie confocale ex vivo et cytométrie en flux. Ce travail a contribué à améliorer nos connaissances immunologiques sur les effets locaux et précoces des vaccins et /ou adjuvants. / My pHD project allowed the development of in vivo imaging approaches to study the skin antigen presenting cell (APC) behavior post-intradermal vaccination in non-human primates. APC targeting was performed by in vivo injection of fluorescent anti-HLA-DR monoclonal antibody. The effect of an adjuvant (R-848, ligand of TLR7/8) on skin APC and the immunotargeting of Langerhans cells by anti-HIV vaccinal fusion protein (anti-langerin-HIVGag) were assessed by in vivo fluorescent imaging, ex vivo confocal videomicroscopy and flow cytometry. This work contributed to improve immunological knowledge on local and early events post-vaccination with or without adjuvant.
