Caractérisation fonctionnelle et structurale d’une protéine alternative mitochondriale : AltMiD51 / Functional and structural characterisation of a mitochondrial alternative protein : AltMiD51

Contrairement à la vision classique des ARNms eucaryotes qui ne contiendrait qu’une seule séquence codante, de nombreuses évidences expérimentales montrent que ces ARNms contiennent plusieurs séquences codantes qui permettraient l’expression de plusieurs protéines différentes. Les ARNms sont donc multicodants et contiennent des cadres de lectures alternatifs (AltORFs pour alternative open reading frames). Ces ORFs alternatifs sont présents dans les régions non-traduites (UTRs) ou chevauchant le RefORF (cadre de lecture ouvert de référence) dans les cadres de lectures non-canoniques +2 et +3. Le protéome est donc plus complexe que ce que l’on pense. Toutefois, le rôle et la fonction de ces nouvelles protéines restent à être investigués. Au cours de mon projet de recherche à la maîtrise, j’ai commencé la caractérisation de la protéine alternative AltMiD51, codée dans le 5’UTR du gène bicistronique MIEF1/SMCR7L/MID51 et co-exprimée avec sa protéine de référence MiD51. Par des approches variées de biologie moléculaire, cellulaire et biochimique, j’ai d’abord démontré et confirmé la localisation cellulaire de la protéine AltMiD51 à la mitochondrie. Par la suite, j’ai pu démontrer que la présence d’AltMiD51 affecte significativement la morphologie mitochondriale en fragmentant celle-ci. De plus, j’ai pu davantage cibler la région qui contient l’information de sa localisation ainsi que son effet de fragmentation, soit seulement les 23 premiers acides aminés de sa séquence. J’ai également observé que cette région Nterminale (a.a.23) est encore plus efficace pour induire la fragmentation des mitochondries. J’ai pu démontrer que le motif protéique L-Y-R est essentiel pour l’activité de fragmentation. J’ai également validé l’interaction in vivo de AltMID51 avec sa protéine partenaire ACPM (Acyl carrier protein) dans des foci mitochondriaux. En conclusion, mes travaux à la maîtrise ont permis de mettre en évidence que la protéine AltMiD51 est un nouveau facteur impliqué dans la fission mitochondriale. Ces résultats ouvrent de nouvelles perspectives en ce qui concerne la caractérisation de nouvelles protéines alternatives et par leur contribution dans la biologie moléculaire de la cellule. / Abstract : Challenging the dogma that eukaryotic mRNAs contain a single coding sequence, an ever-growing number of studies highlight the possibility for these mRNAs to have several coding sequences, and thereby code for multiple proteins. Thus, eukaryotic mRNAs are multicoding and present alternative open reading frames (AltORFs). These alternative ORFs are present in the non-translated region (UTRs), and within or overlapping the RefORF (reference open reading frame) in non-canonical frames (+2 and +3). The proteome is indeed more complex than we initially thought. However, the role and biological function of these proteins remain to be elucidated. Over the course of my MPhil, I started characterizing the alternative protein AltMiD51, encoded in the 5’UTR of the bicistronic MIEF1/SMCR7L/MID51 gene, and coexpressed with its reference protein (MiD51). Using a wide range of molecular biology, cellular and biochemistry assays, I first demonstrated AltMiD mitochondrial localisation. I then proved AltMiD51 expression alters mitochondrial dynamics, enhancing a fragmented morphology. Moreover, I further characterized the sequence region responsible for AltMiD51 localisation and mitochondrial fragmentation, namely the first 23 amino acids. I also observed that this N-terminal region alone presents a stronger phenotype of mitochondrial fragmentation. In addition, I proved the L-Y-R domain is essential for AltMiD51 fragmentation activity, and I validated AltMiD51 in vivo interaction with ACPM (Acyl Carrier Mitochondrial Protein) within mitochondrial foci. Eventually, the work presented here highlighted AltMiD51 as a novel factor involved in mitochondrial fragmentation. These results shed light on new perspectives regarding the characterisation of alternative proteins and their contribution to the cellular metabolism.

Contraintes sur les violations à la symétrie de Lorentz par analyse des données de télémétrie laser Lune / Constaints on Lorentz symmetry violations using lunar laser ranging observations

La relativité générale (RG) et le modèle standard des particules permettent de comprendre les quatre interactions fondamentales de la nature. La formulation d'une théorie quantique de la gravitation permettrait d'unifier ces deux tenants de la physique moderne. D'après les grandes théories d'unification, une telle union est possible moyennant la brisure de certaines symétries fondamentales apparaissant à la fois en RG et dans le modèle standard telle la symétrie de Lorentz. Les violations de la symétrie de Lorentz peuvent être paramétrées dans tous les domaines de la Physique par une théorie effective du champ appelée extension du modèle standard (SME). Une violation au principe d'invariance locale de Lorentz dans le secteur gravitationnel serait supposée engendrer des perturbations dans la dynamique orbitale des corps présents dans le système solaire, notamment la Lune. Ainsi, à partir des données extrêmement précises de télémétrie laser, l'orbite lunaire peut être minutieusement analysée afin de débusquer d'éventuelles anomalies dans son mouvement. Dans cette optique, ELPN (Ephéméride Lunaire Parisienne Numérique), une nouvelle éphéméride lunaire intégrée dans le cadre du formalisme SME a été développée durant la thèse. ELPN fournit les solutions au problème lunaire sous la forme de séries temporelles datées en temps dynamique barycentrique (TDB). Parmi les solutions numériquement intégrées, mentionnons la position et la vitesse du vecteur barycentrique Terre-Lune, les angles de librations lunaires, la différence entre le temps terrestre et le TDB, ainsi que l'ensemble des dérivées partielles intégrées depuis l'équation aux variations. Les prédictions de l'éphéméride ont été utilisées afin de réduire les observations lunar laser ranging (LLR). Dans le cadre de la RG, la dispersion des résidus s'est avérée en accord avec les dispersions calculées à partir des éphémérides INPOP13b et DE430. Dans le cadre du SME minimal, l'analyse des données LLR a permis de contraindre toutes violations à l'invariance locale de Lorentz. Une grande attention a été portée à l'analyse des incertitudes afin de fournir des contraintes réalistes. Ainsi, dans un premier temps, les combinaisons linéaires de coefficients SME ont été isolées puis ajustées aux observations. Puis, dans un second temps, les incertitudes réalistes ont été déterminées par une méthode de ré-échantillonnage. L'analyse des données de télémétrie laser Lune n'a pas permis de révéler de violations au principe d'invariance locale de Lorentz agissant au niveau de l'orbite lunaire. Les prédictions de la RG ont donc été validées avec des précisions absolues allant de 10-9 à 10-12. / General Relativity (GR) and the standard model of particle physics provide a comprehensive description of the four interactions of nature. A quantum gravity theory is expected to merge these two pillars of modern physics. From unification theories, such a combination would lead to a breaking of fundamental symmetry appearing in both GR and the standard model of particle physics as the Lorentz symmetry. Lorentz symmetry violations in all fields of physics can be parametrized by an effective field theory framework called the standard-model extension (SME). Local Lorentz Invariance violations in the gravitational sector should impact the orbital motion of bodies inside the solar system, such as the Moon. Thus, the accurate lunar laser ranging (LLR) data can be analyzed in order to study precisely the lunar motion to look for irregularities. For this purpose, ELPN (Ephéméride Lunaire Parisienne Numérique), a new lunar ephemeris has been integrated in the SME framework. This new numerical solution of the lunar motion provides time series dated in temps dynamique barycentrique (TDB). Among that series, we mention the barycentric position and velocity of the Earth-Moon vector, the lunar libration angles, the time scale difference between the terrestrial time and TDB and partial derivatives integrated from variational equations. ELPN predictions have been used to analyzed LLR observations. In the GR framework, the residuals standard deviations has turned out to be the same order of magnitude compare to those of INPOP13b and DE430 ephemerides. In the framework of the minimal SME, LLR data analysis provided constraints on local Lorentz invariance violations. Spetial attention was paid to analyze uncertainties to provide the most realistic constraints. Therefore, in a first place, linear combinations of SME coefficients have been derived and fitted to LLR observations. In a second time, realistic uncertainties have been determined with a resampling method. LLR data analysis did not reveal local Lorentz invariance violations arising on the lunar orbit. Therefore, GR predictions are recovered with absolute precisions of the order of 10-9 to 10-12.

SOBREPESO E OBESIDADE EM ESCOLARES: IMPLICAÇÕES E ALTERNATIVAS NO CONTEXTO ESCOLAR / OVERWEIGHT AND OBESITY: IMPLICATIONS AND ALTERNATIVES IN THE SCHOOL CONTEXT

In recent years, the combination of changes in dietary habits with the sedentary lifestyle has contributed significantly to the increased prevalence of overweight and obesity. Thus, there is a need to expand the process of nutritional assessment, aiming at educating young people about the lifestyle and adopt healthy eating and living right, necessary for proper growth and development in this age group. Thus, considering the above, this study aimed to gather information on the percentage of school children with overweight and obesity in primary school students who attend public discipline of Physical Education. A survey of data on body mass index (BMI), waist-hip ratio (WHR), weight, height, waist circumference (PC), hip circumference, sex, age, socioeconomic status, self-image, body perception, eating habits and physical activity in children and adolescents in 6th, 7th and 8th grades, at the State Institute Romaguera Correa municipality of Uruguaiana / RS, with 296 students. From the results obtained, it was observed that the students have high levels of BMI and WHR. In particular, analyzing the prevalence in the 3 series studied, there was a higher percentage of body mass index above 25, where 21% of the students have these ratios and 12% have a BMI above 30, stressing that there was no statistical difference between sexes in relation to BMI and hip. However, in all other variables observed this difference (p <0.05). It was also observed that poor dietary habits and lifestyle of students are directly linked to these values. In the correlation between BMI and WHR with body awareness and self-image of students, students who responded being overweight, the average BMI and WHR confirm this hypothesis, and according to their views on losing weight, it was shown that adolescents are aware of your body. Therefore, this may be a starting point for the proposed change in standards of living habits and physical activity. Then, from the implementation of this research, we verified the importance of including studies on overweight and obesity early in schools, to detect possible health problems as early as possible and suggest actions that can help promote the well being of children and youth. / Nos últimos anos, a combinação de mudanças dos hábitos alimentares com o estilo de vida sedentário tem contribuído significativamente para o aumento da prevalência de sobrepeso e obesidade. Desta forma, há a necessidade de se ampliar o processo de avaliação nutricional, visando orientar aos adolescentes quanto ao estilo de vida mais saudável e a adoção de hábitos alimentares e de vida corretos, necessários ao crescimento e desenvolvimento adequado nesta faixa etária. Assim, considerando os aspectos expostos, este estudo teve como objetivo principal levantar informações sobre o percentual de crianças escolares com sobrepeso e obesidade em alunos do Ensino Fundamental Público que frequentam a disciplina de Educação Física. Foi realizado um levantamento de dados sobre o índice de massa corporal (IMC), relação cintura-quadril (RCQ), peso, estatura, perímetro de cintura (PC), perímetro do quadril, sexo, idade, nível sócio-econômico, autoimagem, percepção corporal, práticas alimentares e de atividade física em crianças e adolescentes de 6ª, 7ª e 8ª séries, no Instituto Estadual Romaguera Corrêa do município de Uruguaiana/RS, contando com 296 alunos. A partir dos resultados obtidos, foi possível observar que os escolares possuem índices elevados de IMC e RCQ. Nomeadamente, analisando-se as prevalências nas 3 séries pesquisadas, observou-se uma maior porcentagem para o IMC acima de 25, onde 21% dos escolares possuem estes índices e 12% possuem IMC acima de 30; salientando também que não houve diferença estatística entre os sexos em relação aos valores de IMC e quadril. No entanto, em todas as demais variáveis observou-se esta diferença (p < 0,05). Observou-se ainda que os hábitos alimentares inadequados e o sedentarismo dos estudantes estão diretamente ligados a estes valores. Na correlação entre IMC e RCQ com a percepção corporal e autoimagem dos alunos, os estudantes que responderam estar acima do peso, as médias de IMC e RCQ confirmam esta hipótese; e de acordo com suas opiniões em perder peso, mostrou-se que os adolescentes têm consciência de seu corpo. Logo, este pode ser um ponto de partida para a proposta de mudança nos padrões de hábitos de vida e atividade física. Então, a partir da execução desta pesquisa, verificou-se a importância de se incluir estudos acerca de sobrepeso e obesidade desde cedo nas escolas, para detectar os possíveis problemas de saúde o mais precocemente possível e sugerir ações que possam auxiliar na promoção do bem-estar da criança e do jovem.

Juridictionnalisation de la répression pénale et institution du ministère public : étude comparative France-Angleterre / Juridicial nature of repression and Public Prosecutor : comparative analysis France-England

En France comme en Angleterre, le ministère public a historiquement vocation à garantir la forme juridictionnelle de la répression-pénale. A l'inverse, la juridictionnalisation de la répression pénale constitue une limite traditionnelle aux prérogatives du ministère public. Ainsi, alors que le premier rend toujours possible la juridictionnalisation et légitime la répression, la seconde limite légitimement le champ des possibles offerts au ministère public. Pourtant, la liaison qui paraît si évidente entre la juridictionnalisation de la répression pénale et l'institution du ministère public s'efface progressivement en nuances. Les législateurs français et anglais, encouragés par la jurisprudence, ont considérablement renforcé les pouvoirs d’enquête et de traitement infrajuridictionnels du ministère public évinçant par là la juridictionnalisation de la répression. Et même si la juridictionnalisation peut paraître préservée lorsqu'il s'agit pour le ministère public d'obtenir une coercition et une rétribution ultimes, celle-ci s'avère en réalité sans substance. Le contrôle à priori du juge de l'autorisation des pouvoirs d'enquête juridictionnalisés est souvent purement formel et ne peut permettre de contredire la nécessité d'ordonner tel ou tel acte qui serait requis par le ministère public, alors que le contrôle a posteriori du juge de l'annulation des actes d'enquête est lui déficient et aléatoire. La garantie d'un usage régulier des pouvoirs d'enquête et de traitement des infractions ne saurait pourtant venir d'une juridictionnalisation de l'institution du ministère public. La comparaison de l'organisation des ministères publics français et anglais laisse apparaître que les solutions qui sont envisagées l'indépendance et impartialité du ministère public français ne seraient pas suffisantes. C'est une restauration de la juridictionalisation des fonctions de l'institution, conforme à sa ratio profonde qu'il convient de mettre en œuvre. / In France, like in England, the Public Prosecutor is historically intended to ensure the judicial nature of criminal justice. Reciprocally, the strengthening of the judicial nature of the criminal justice traditionally restricts the prerogatives of the Public Prosecutor's Office. However, the link between the reinforcement of the judicial nature of the criminal justice and the existence of the Public Prosecutor, albeit obvious, evaporates slowly in multiple shades. The French and the English lawmakers, supported by case law, have come to considerably strengthen the Public Prosecutor's investigation and pre-trial powers, thus ousting the judicial nature of the criminal justice. Although it may seem that the judicial nature of repression is preserved when the Public Prosecutor is required to obtain coercive measures or ultimate retribution, it turns out to be deprived of substance. An a priori control by a judge who grants judicial investigative powers to the Public Prosecutor is always formalistic, whereas an a posteriori control by a judge who may annul decisions taken during the investigation is flawed and random. However, a guarantee of a regular use of investigative powers and of treatment of offenses cannot come out of the strengthening of judicial nature of the Public Prosecutor. A comparative analysis of the structure of the Public Prosecutor’s Office in France and in England reveals that the dues that are to be considered -independence and impartiality of the French Public Prosecutor -do not suffice. It is a reestablishment of the judicial functions of the body, coherent with its true ratio, that should be implemented.

Dissertation thesis focuses on principles of multi-criteria decision-making in public contracts. The theoretical section of the thesis introduces principles and methods of multi-criteria decision-making including the determination of their suitability for public contracts. It also cover characteristics and specifics of purchasing via public contracts and findings of previous research conducted in the Czech Republic and foreign countries. Own empirical research comprises two parts. The first part of research is based on data about public contracts from the Czech Republic between 2011 and 2013 announced via the Information System on Public Contracts. The data allow investigating criteria and their weights used by contracting entities in relationship to characteristics of the public contracts; especially the subject-matter and the value of the contract. The second part of the research is based on sample data file of contract announcements for contracts evaluated according to the economic advantageousness of the tender. The data allow investigating methods of multi-criteria decision making and approaches to qualitative criteria. Findings from the theory and the conducted research were source for a normative model for selection of criteria and methods of multi-criteria decision-making. The goal of the model is to enhance the quality of decision-making in the field of public contracts. The most important contribution of the model is the connection of the three key parts of multi-criteria decision-making; criteria, its weights and methods of multi-criteria decision-making.

Využití teorie fuzzy množin a jejich rozšíření v metodě TOPSIS / The use of the fuzzy set theory and their extensions in the TOPSIS method

This master's thesis deals with extensions of TOPSIS method, which is one of methods for multi-criteria evaluation of alternatives. These extensions use theory of fuzzy sets (FS) and their futher extensions to interval-valued (IVFS), intuitionistic (IFS) and hesitant (HFS) fuzzy sets and their combinations (IVIFS, IVIHFS). Significant part of this thesis explains the principle of fuzzy sets and their generalizations. Descriptions of operators for aggregations of grades of membership has very important role here. Next, very short description of multi-criteria evaluation problems and detailed description of TOPSIS method are contained. The second half of this thesis is dedicated to four existing extensions of TOPSIS metod that uses theories of FS, IVFS, IVIFS and IVIHFS. Every method is illustrated with an example that shows principle of calculations. It illustrates new possibilities of the methods that use new sets theories and potential complications and deviations from the original TOPSIS method. At the end of this thesis, evaluation of usefulness of used approaches is mentioned.

This thesis aims to develop a methodology to translate an ontology to a matrixbased methods that is case independent. In the larger picture this is one of manycomplexities in systems engineering approach. The focus in this work is in the domainof conceptual design, hence, matrix based methods that are most suitable for it areinvestigated. The flexible nature of the ontology always allows for expansion withnew information about the concepts, and from that perspective a parsing scriptwas developed to extract information from the ontology, it was also designed to beflexible. The script can be used to extract any desired type of information by alteringa few keywords as inputs. The advantage is that the same structure of the script canbe used to extract information from many different ontologies.Using this method the matrices relevant for conceptual design were formulated.These matrices were further compared to each other. Also, a few recommendationswere given for the improvement of the method to adapt towards high fidelity. Thecurrent method is capable of solving problems with low fidelity, however this formsa basis to expand the method to high fidelity problems.

Death taxes - an international search for alternatives

The opponents of the death taxes stand in the left hand corner of the ring and the proponents in the right hand corner. When the bell rings, these two opposing sides exchange blows over the death taxes. The death tax debate lingers in the background of all social conversations and flares up at critical times, such as in election years. To date, no amount of arguing has seen an end to the death tax debate. Although both sides agree that death taxes have shortcomings they cannot agree on the appropriate course of action – no side is willing to concede defeat. However, the death tax debate needs to end; precious time and energy should be focused on issues that are relevant to the current times. The only way in which this debate will end, is if an alternative to the death taxes is found – an alternative that is suitable in the eyes of both the opponents and proponents of the death taxes. There is, nevertheless, a lack of contemporary research aimed at finding death tax alternatives. The existing literature has focused on developing approaches to finding alternative taxes, as opposed to studying the alternative taxes themselves. The primary purpose of this study was to conduct a search for alternative taxes that could replace the death taxes. This search transcended national borders in order to identify international solutions to the age-old problem of death taxes. The suitability of the alternative (in the eyes of both the opponents and proponents of the death taxes) was measured by comparing the advantages and disadvantages associated with the tax alternative identified, to the advantages and disadvantages associated with the death taxes. The secondary purpose of this study was to evaluate whether any of the identified alternatives would be suitable for South Africa. Je propose d’étudier un processus central pour les organisations féministes : l’empowerment. L’objectif de ma thèse est tout à la fois de voir en quoi cela permet de comprendre l’organizing féministe et alternatif, et par là-même de penser l’organizing à partir de ses marges.Pour cela, j’ai conduit une ethnographie féministe au sein d’une association féministe, pendant deux ans. Le récit ethnographique que j’élabore décrit trois dimensions de l’empowerment tel qu’il se déploie dans l’association : la conscientisation, la mobilisation et la transformation sociale. Cela me permet de définir l’empowerment comme une praxis collective, dissonante et préfigurative, qui construit performativement l’organisation comme féministe. Je suggère de penser l’empowerment comme un processus d’organizing alternatif, ce qui permet d’incarner et de politiser une ontologie du devenir pour la théorie des organisations. / This thesis is part of a conversation on alternative organizations, and underlines the potential of a feminist approach to understand these organizations. I offer to study a core process of feminist organizations :empowerment. The objective is both to explore how this sheds a new light on feminist and alternative organizing, and to rethink organizing from its margins.To do so, I have conducted a two-years feminist ethnography within a feminist collective. The ethnographic tale describes three dimensions of empowerment as it is practiced within the collective: conscientization, mobilization and social transformation. It entails a definition of empowerment as a collective, dissonant andprefigurative praxis, which performativelyconstructs the organization as feminist. I suggest to think of empowerment as an alternative organizing process, which incarnatesand politicizes an ontology of becoming fororganizations.
