From the contact with activist groups in two cities in the state of São Paulo it was possible to observe situations that alternate between containment and propitiation measures and their consequent interferences in a conviviality supported by the principle of animal protection. Government and activism – as well as the perceptions and conceptions of the domestic animal resulting from their respective actions – allow a theoretical and methodological reflection on the binomial established between culture and nature, when they reveal conflicts and possible ambivalences of animal protection, situated between control and hosting measures that, consequently, are linked to affectivities and legal issues. Ethical arguments, contained in laws and public policies, as well as the appeal to the sensitive dimension, eventually triggered by activism, also provide subsidies to understanding other concepts - such as welfare and animal rights – and these interspecific relationships mediated by emotions and reason, both tangents to the moral condition of the animal as part of a predominantly human socius. / Como uma etnografia acerca de relações interespecíficas – entre humanos e animais – em contextos urbanos, este trabalho visa refletir sobre as ações envolvidas na gestão e tutela da vida animal no âmbito institucional e doméstico. A partir do contato com grupos ativistas em dois municípios paulistas, foram observadas situações que se alternam entre medidas de contenção e de propiciação e suas consequentes interferências num convívio amparado pelo princípio da proteção aos animais. Tanto o poder público como o ativismo – bem como as percepções e concepções sobre o animal doméstico resultantes de suas respectivas ações – possibilitam uma reflexão de ordem teórico-metodológica do binômio estabelecido entre cultura e natureza, ao apresentarem os conflitos e as possíveis ambivalências da proteção animal, situados entre medidas de controle e de acolhimento e, por conseguinte, imbricados a afetividades e juridicidades. Argumentos éticos, presentes em legislações e em políticas públicas, e o apelo à dimensão do sensível, eventualmente acionado pelas atividades militantes, também fornecem subsídios à compreensão de outros conceitos – como bem-estar animal e direitos animais – e dessas relações interespecíficas mediadas por emoções e razões, ambas tangentes à condição moral do animal enquanto parte de um socius predominantemente humano.

Identifying Bovine Respiratory Disease (BRD) through the Nasal Microbiome

<p>Bovine respiratory disease (BRD) is an ongoing health and economic issue in the dairy and beef cattle industry. Also, there are multiple risk factors that make an animal susceptible to BRD and it's diagnosis and treatment is a challenge for producers. Four bacterial species, <em>Mannheimia haemolytica, Pasteurella multocida, Histophilus somni, </em>and<em> Mycoplasma bovis</em> have been associated with BRD mortalities. Hence, this study aims to characterize the cattle nasal microbiome as a potential additional diagnostic method to identify animals suspected to have a lung infection. Quantitative PCR and 16S rRNA gene sequencing were used to determine the bacterial load of these four bacterial pathogens in the nasal microbiome of apparently healthy (N=75) and (N=58) affected by BRD Holstein steers. We then sought to identify a value or equation that could be used to discriminate between BRD and healthy animals using a Linear Discriminant Model (LDA). Additionally, co-occurrence between commensal bacterial and BRD-pathogens were also identified. Cattle diagnosed with BRD presented lower richness, evenness and phylogenetic diversity than healthy pen-mates. Bacterial species and genera <em>Truperella pyrogenes </em>and <em>Bibersteina</em> were increased in the BRD group, and the species <em>Mycoplasma bovirhinis</em> and <em>Clostridium sensu stricto</em> increased in the healthy group. Prevalence of <em>H. somni </em>(98%)<em> </em>and <em>P. multocida </em>(97%) were the highest regardless of disease diagnosis in all the samples. Prevalence of <em>M. haemolytica </em>(81 vs. 61%) and<em> M. bovis </em>(74 vs. 50.7%) were higher in the BRD group. The bacterial density of <em>M. haemolytica</em> and<em> M. bovis </em>was also higher in the BRD group, whereas <em>Histophilus somni</em> was lower in the BRD group. Five different models were tested using LDA, and one model produced a sensitivity and specificity of 60% and 81% agreement with diagnosis based on animal symptoms. Co-occurrence analysis demonstrated that the nasal microbiome members are more likely to interact with each other than associations between BRD-pathogens and nasal microbiome members. This study offers insight into the BRD-pathogens prevalence and difference in nasal microbiome between healthy and BRD animals and provides a potential platform for future studies and potential pen-side diagnostic testing.</p>

Teaching Through the Lens of Humane Education in U.S. Schools

Humane education (HE) is a specialized niche in higher education and adult learning. HE provides a curricular framework positioning environmental ethics, animal protection, human rights, media literacy, culture, and change processes as the nexus for understanding and inspiring social change. Research-derived experiences illuminating how educators conceptualize and implement HE in U.S. schools are absent from the scholarly literature. Facing this gap, practitioners and administrators of HE programs cannot access nor apply research-derived practices to inform instruction. To address this gap, a conceptual framework was advanced weaving together HE teaching experience, Freirean philosophy, hyphenated selves, reflection-in-action, transformative learning, and transformative education to explore and understand what it means to be a practitioner teaching through the lens of HE in U.S. primary, secondary, and post secondary classrooms. A qualitative, multi case study was designed wherein purposeful and maximum variation sampling resulted in the recruitment of 9 practitioners working in Kindergarten to post secondary contexts. Eight practitioners were alumni of HE programming, and 1 practitioner engaged self-study of HE pedagogy. Each bounded system included the HE practitioner, his or her classroom context, and local school community. Interviews, document review, within-case analysis, and cross-case analysis resulted in key themes illuminating the need to design a comprehensive system of field-based learning and ongoing professional support to benefit HE practitioners. A policy recommendation is provided to shape programming, policy development, and resource allocation to improve and sustain HE as a field of study and professional practice.

Thyroid Hormone Metabolism in the Non-Euthyroid Porcine Fetus

<p>Thyroid hormone is essential for regulating adult metabolism and proper fetal development. Under normal conditions, maternal and fetal thyroid hormones are subject to metabolism at the placenta and within fetal tissues through deiodination and sulfation to regulate fetal exposure to the bioactive hormone. Disruptions of the thyroid hormone system can result in non-thyroidal illness syndrome (NTIS), which is classified as the dysregulation of thyroid hormone homeostasis. The exact cause of the alterations in circulating thyroid hormone levels during NTIS is not well- known. In comparison, hypothyroidism results from the absence of thyroid hormone production and presents as low thyroid hormone levels.</p> <p>Porcine reproductive and respiratory syndrome virus (PRRSV) crosses the late gestation placenta and causes suppression of circulating maternal and fetal thyroid hormone. Chapter 2 investigates the potential role of thyroid hormone metabolism in this disruption. Pregnant gilts were challenged with PRRSV2 (n=22) or sham inoculated (n=5) at gestation day 85. Samples were collected on day 106, and viral load was assessed in fetal serum and thymus. From the entire fetal population, three distinct subsets of fetuses representing biological extremes were identified, including uninfected with no detectable viral load (UNIF), high viral load viable (HV-VIA), or high viral load with severe meconium staining (HV-MEC). In addition, control fetuses from sham inoculated gilts (CON) were used as a reference group. Samples of fetal liver, kidney, and the corresponding fetal placenta and maternal endometrium for n=10 fetuses per group were then used to evaluate gene expression. A total of 11 genes associated with thyroid hormone metabolism including deiodinases (DIO1,2,3), sulfotransferases (SULT1A3,1B1,1C2,1E1,2A1), sulfatase (STS), and solute carriers (SLC16A2,16A10) were quantified using absolute quantification qPCR. Evidence of fetal decompensation was observed within the high viral fetuses in the form of decreased DIO1 expression within the fetal liver and increased DIO3 expression in both components of the placenta. Circulating levels of T4 and inactive thyroid hormone metabolites, reverse-triiodothyronine (rT3) and two diiodothyronines (3,5-T2 and 3,3’-T2), were measured in fetal serum. While T4 was depressed, no change was observed in circulating rT3 levels, and neither T2 metabolite reached the lower detection limit. This may suggest that alterations in thyroid hormone metabolism generate a localized effect on hormone metabolites in the respective tissues.</p> <p>Alternatively, the low levels of available T3 and T4 limit the production of downstream metabolites to be found in serum.</p> <p>The cause-and-effect relationship between PRRSV infection, fetal thyroid disruption, and the effects on fetal thyroid hormone metabolism are unclear. Therefore, Chapter 3 developed a non-pathogenic model using methimazole (MMI) to induce hypothyroidism in the late gestation fetus and evaluate the impact on fetal development and thyroid hormone metabolism. Pregnant gilts were either treated with oral methimazole or equivalent sham from gestation day 85-106 (n=4/group), followed by classification of all fetuses as live, live but meconium stained, or dead. Fetuses exposed to MMI in-utero were notably hypothyroid with significantly suppressed serum T3 and T4 and histological evidence of goiter. Surprisingly, fetuses from MMI-treated dams were substantially larger but appeared to exhibit non-allometric growth with an increase in girth but not length. The liver, kidney, and the corresponding fetal placenta and maternal endometrium were collected from a subset of 16 fetuses per group to evaluate the relative expression of five genes associated with thyroid hormone metabolism, including three deiodinases and two solute carriers known to transport thyroid hormone. Compensatory transcription of DIO3 was observed in all tissues evaluated, suggesting increased vertical transfer of maternal thyroid hormone at the placenta and decreased breakdown of thyroid hormone within fetal organs.</p> <p>The evaluation of thyroid hormone metabolism within the fetus and within the placenta has allowed us to differentiate suppressed thyroid hormone levels of the pig fetus under pathogenic and non-pathogenic conditions. In the context of PRRSV infection, the observed decompensation of thyroid hormone metabolism would further exacerbate the hypothyroid state and is therefore consistent with NTIS. In contrast, fetuses with induced thyroid hormone suppression following maternal exposure to MMI showed compensatory thyroid hormone metabolism in the same tissues. This indicates true hypothyroidism and clearly demonstrates a fetal capacity to respond to such endocrine disruption.</p>

Um componente considerado crítico para o manejo bem sucedido de populações silvestres é a manutenção da variação genética. No intuito de avaliar a magnitude e a distribuição da variabilidade genética existente em populações de Caiman latirostris, nove populações distribuídas ao longo do eixo latitudinal da distribuição da espécie no Brasil e uma população em cativeiro foram estudadas com auxílio de onze locos microssatélites. A diversidade gênica média (He=h) e a heterozigosidade média observada (Ho) apresentaram valores elevados, 0,628 e 0,567, respectivamente, denotando existência de elevada variabilidade genética para esta espécie nas regiões de estudo. A população paulista mesmo estando na zona intermediária de distribuição geográfica no Brasil não possui a maior variabilidade genética. O valor FST estimado foi 0,270 e o RST foi 0,342. Ambas as medidas de diferenciação entre as populações foram significativas (P<0,05). As altas estimativas de FST e RST sugerem a ausência ou fluxo gênico restrito entre essas populações, exceção feita entre as populações de Natal (RN) e João Pessoa (PB), onde não houve diferenciação significativa entre as populações, sugerindo assim que há fluxo gênico entre elas, fato confirmado pelo coeficiente de parentesco. Pôde-se concluir neste trabalho que a distância genética entre a população do litoral é afetada pela existência da Serra do Mar como barreira geográfica pelo fato desta população apresentar as maiores diferenciações genéticas e não se agrupar a nenhuma população pelo método de agrupamento UPGMA, mesmo estando próxima das populações paulista. A distância genética entre as populações parece não acompanhar a distância geográfica, em termos de gradiente latitudinal (r=0,206). Entretanto quando retiramos a população da Ilha do Cardoso esta correlação aumenta significativamente (r=0,540), indicando haver um certo padrão espacial da variabilidade genética entre as populações. O coeficiente médio de parentesco foi baixo entre e dentro das populações estudadas. Both differentiation measures among the populations were significant (P < 0,05). The higher estimates of FST and RST suggested a absence or low gene flow among those populations, exception done between the populations of Natal (RN) and João Pessoa (PB), where there was not significant differentiation among the populations, suggesting a gene flow pattern among them, fact confirmed by the related coefficient. It could be concluded in this work that the genetic distance among the population of the coast is affected by the existence of the Mountain of the Sea as geographical barrier for the fact of this population to present the largest genetic differentiations and not to group the any population for the grouping method UPGMA, same being close of the populations from São Paulo. The genetic distance among the populations seems not to accompany the geographical distance, in terms of latitudinal gradient (r=0,206). However when we removed the population of Cardoso's Island this correlation it increases significantly (r= 0,540), indicating there to be a certain space pattern of the genetic variability among the populations. The medium coefficient of related was low among and inside of the studied populations. With these results we can begin to understand the dynamics and it structures social of populations of Caiman latirostris.

殘酷時尚議題已成為當代的芒刺,對動物保護團體及人權組織形成挑戰。善待動物組織(People for the Ethical Treatment of Animals:PETA)是目前規模最大、發展最完善的動物權利組織,其廣告往往能在人們心中喚醒善待動物的意識。PETA首次在時尚雜誌上發起的反對毛皮廣告活動,即獲得「非營利組織最佳廣告獎(Best Ads Produced by a Nonprofit)」。 本研究的宗旨為,展示公益內容廣告運動可在任何市場導向的經濟體中運作。本研究列舉PETA的PSA廣告範例如下:「我寧願裸體」(“I’d rather go naked”, 1992年) 、「毛皮代表死亡」(“Fur is dead”, 2001年)、「對皮膚感到自在」(“Be comfortable in your own skin”, 2003年)、「拯救海豹」(“Save the seals”, 2009年)。「毛皮」 (“Skins”)主題上述廣告皆由知名女性代言。 本研究為質量研究,採用射影技術(projective technique)及深入訪談,調查結合訊息與圖像的PSA廣告在傳達動物權力相關議題時的效果。本研究將指出這些廣告可如何透過1.有創意、逼真的圖像、2.說服力的寓意訊息及3.知名代言人,加強傳遞人與自然關係的概念。 本研究發現,具說服力的訊息必須擁有逼真、富含寓意、簡潔、清楚等特性,且僅有在搭配適當的圖像時,才能發揮效力。此類圖像必須擁有不含混、非情色誘因、有創意、外來風或暗中使用性訴求等特性。廣告必須同時訴諸感性訴求及現實主義,並由知名、受人敬重的代言人出演。當公益廣告意圖以幽默感營造有利的風趣情境時,就不應使用嚴肅形象,或要求觀眾訴諸行動。至於訴諸恐懼類的公益廣告,則結合令人反感的圖像特徵及寓意訊息,仍可營造有利觀眾回應的情境,然而太過強烈的反感特徵有時會造成觀眾無法掌握寓意,導致觀眾無法回應。訴諸恐懼類平面廣告所引起的觀眾回應差異,以及本研究的局限性,可歸因於應答者的文化差異。 本研究做出結論為,擁有優良公眾形象及正面背景的知名代言人,可成功向大眾傳遞寓意訊息。再者,有創意、逼真,但不過度暴力、過度情色的圖像可吸引觀眾目光,促使大眾意識並做出反應。 / In the Modern Age the problem of killing fashion has become a thorny issue for the fashion industry, animal rights protection organizations, animal world and humanity. In this respect, People for the Ethical Treatment of Animals (PETA) performs as the largest and most highly developed animal rights institution in modern countries. PETA’s advertising has made a revolution in people’s minds concerning the ethical treatment of animals and already the first advertising campaign by PETA received the “Best Ads Produced by a Nonprofit” Award for its anti-fur advertising campaign in fashion magazines. The purpose of the study is to show how such social content advertising campaigns can work in all countries that are learning to live under market-driven economies. This research concerns properly selected print PSA samples of “I’d rather go naked” (launched in 1992), “Fur is dead” (launched in 2001), “Be comfortable in your own skin” (launched in 2003), and “Save the seals” (launched in 2009) campaigns presented by a number of celebrity spokeswomen posing for PETA’s “Skins” print advertisements. Based on qualitative research with the use of projective techniques and in-depth interviews, the study investigates the impact of a combination of PSA specific messages and image characteristics concerning animal rights using Public Service Print Advertising. The research will show how these ads hopefully can improve their effectiveness in order to achieve more respect for the human-nature relationship by using 1) Creative, realistic images, 2) A powerful educational message, and 3) A highly respected and appreciated celebrity spokesperson. The study found that a strong persuasive message is defined as realistic, educational, laconic, clear and the advertisement is ultimately effective only when used with proper image characteristics. Such image characteristics are non-vulgar, non-sexually distracting, but creative, exotic, or latent in sexual appeal. An advertisement should use emotional appeal and realism, presented by a celebrity spokesperson with a respected personal background. While PSA uses a high degree of humor to produce favorable and amused attitude, they do not present a serious image or promote a call to action. Regarding using fear appeal, strongly unpleasant image characteristics combined with educational messages still may produce a favorable and responsible attitude. However, it was found that unfavorable perceptions caused by strongly unpleasant images sometimes distracted viewers from the educational message or failed to produce a responsible reaction. The striking differences among public responses regarding fear appeal in print ads and study limitations can be attributed to cultural differences within respondents. The study has concluded that strong educational messages spoken by highly trustworthy celebrities with positive background activity makes the public listen. Furthermore, creative, realistic, but not overly-violent or overly-sexual images catch the viewer’s attention, makes the public more aware and encourages responsible thinking.

The higher estimates of FST and RST suggested a absence or low gene flow among those populations, exception done between the populations of Natal (RN) and João Pessoa (PB), where there was not significant differentiation among the populations, suggesting a gene flow pattern among them, fact confirmed by the related coefficient. It could be concluded in this work that the genetic distance among the population of the coast is affected by the existence of the Mountain of the Sea as geographical barrier for the fact of this population to present the largest genetic differentiations and not to group the any population for the grouping method UPGMA, same being close of the populations from São Paulo. The genetic distance among the populations seems not to accompany the geographical distance, in terms of latitudinal gradient (r=0,206). However when we removed the population of Cardoso's Island this correlation it increases significantly (r= 0,540), indicating there to be a certain space pattern of the genetic variability among the populations. The medium coefficient of related was low among and inside of the studied populations. With these results we can begin to understand the dynamics and it structures social of populations of Caiman latirostris.

Občanská iniciativa v problematice množení zvířat / Civil initiative in the field of animal reproduction

This Diploma Thesis focuses on the issue of reproduction of animals and the non- profit organisations and subjects working in this field. The cat and dog reproduction is a primary cause of their overpopulation in the Czech Republic which becomes an ever- increasing issue as the abandoned animal shelter network fails to provide enough capacity for local demand. The thesis is focused on the activities of such organisations and tools they use for mobilising the necessary resources as well as the issue of animal reproduction itself. The theory basis is in the general theory of resource mobilisation supported with the explanation of the civil initiatives dedicated to this phenomenon as well as the overview of the phenomenon of the animal reproduction as well. The thesis is composed of five qualitative interviews with organisations or movements that are dedicated to effective reduction of animal reproduction. The empiric part of the thesis presents selected organisations and then analyses the collected data. The financial and human resources are mostly utilised by organisations which physically treat animals and the majority of the funds is consumed by providing care for them.
