Essa interação pode levar a produção de diversas citocinas, quimiocinas, moléculas inflamatórias e também estimular a geração de espécies reativas de oxigênio (ERO). Neste trabalho avaliamos se a interação com as células HEp-2 poderia ser genotóxica para os mutantes derivados de Escherichia coli K-12 deficientes em algumas enzimas que fazem parte do sistema de reparo por excisão de base (BER). Além disto, avaliamos a expressão do sistema SOS, que é induzido pela presença de danos no genoma bacteriano. Os resultados obtidos mostraram a presença de filamentos, na interação com células HEp-2, principalmente, no mutante xthA (BW9091) e no triplo mutante xthA nfo nth (BW535). Quando a interação foi quantificada na ausência da D-manose, observamos um aumento das bactérias aderidas. Além disto, a quantidade e o tamanho dos filamentos também aumentaram, mostrando que as adesinas manose-sensíveis estavam envolvidas na filamentação bacteriana. Para comprovar se o aumento da filamentação observada neste ensaio foram uma consequência da indução do sistema SOS, desencadeada pela interação com as células HEp-2, quantificamos a expressão do SOS, na presença e na ausência da D-manose. De fato, observamos que a indução do SOS na ausência da D-manose foi maior, quando comparada, com o ensaio realizado na presença de D-manose. Além disto, observamos que a ausência de xthA foi importante para o aumento da filamentação observada na ausência de D-manose. Diante destes resultados, verificamos se a resposta de filamentação ocorreria quando as bactérias interagiam com uma superfície abiótica como o vidro. Observamos também inúmeros filamentos nos mutantes BER, BW9091 e BW535, quando comparados a cepa selvagem AB1157. Essa filamentação foi associada à indução do SOS, em resposta a interação das bactérias com o vidro. Em parte a filamentação e a indução do SOS observadas na interação ao vidro, foram associadas à produção de ERO. Quantificamos também o número de bactérias aderidas e observamos que as nossas cepas formavam biofilmes moderados. Contudo, a formação de biofilme dependia da capacidade da bactéria induzir o sistema SOS, tanto em aerobiose como em anaerobiose. A tensão do oxigênio foi importante para interação dos mutantes BER, uma vez que os mutantes BW9091 e BW535 apresentaram uma quantidade de bactérias aderidas menor em anaerobiose. Contudo, a diminuição observada não estava vinculada a morte dos mutantes BER. Também realizamos microscopia de varredura na cepa selvagem e nos mutantes, BW9091 e BW535 e confirmamos que as três cepas formavam biofilmes tanto em aerobiose como em anaerobiose. Observamos uma estrutura sugestiva de matriz extracelular envolvendo os biofilmes da cepa selvagem AB1157 e do mutante BW9091. No entanto, a formação desta estrutura por ambas as cepas dependia da tensão de oxigênio, pois nos biofilmes formados em anaerobiose essa estrutura estava ausente. Em conclusão, mostramos que na interação das bactérias com a superfície biótica e abiótica, ocorreu lesão no genoma, com indução do SOS e a resposta de filamentação associada. / The epithelial cell is the first contact between microorganisms and host. This interaction results in production of several cytokines, chemokines, and inflammatory molecules by epithelial cells and also stimulate the generation of reactive oxygen species (ROS). In the present study, we have evaluated whether the interaction to HEp-2 cells causes genotoxicity to mutants derived from Escherichia coli K-12 deficient in some enzymes that are part of the system of base excision repair (BER). Moreover, we measured the expression of SOS system, which is induced by the presence of damage to the bacterial genome. Our results showed mainly presence of filamentous bacterial growth in xthA mutant (BW9091) and triple xthA nfo nth mutant (BW535) when submitted to HEp-2 cells interaction assays. When experiments were performed in the absence of mannose, data showed enhanced interaction of viable bacteria to HEp-2 cells for all strains tested. Furthermore, the removal of D-mannose resulted in an increase in both number and size of bacterial filamentous forms, indicating the involvement of mannose-sensitive adhesins in the filamentation of these strains. In order to verify whether the increased filamentation growth in this assay was a consequence of SOS induction, triggered by interaction to HEp-2 cells, we measured expression of SOS in the presence and absence of D-mannose. Indeed, we observed higher expression of SOS response in the absence of mannose than in experiments performed in the presence of D-mannose. Moreover, we observed that the absence of xthA was important to filamentation increasing in absence of D-mannose. Based on these results, we verified if interaction to abiotic surfaces, like glass, could lead to filamentation of these strains. We also observed numerous filaments in BER mutants, BW9091 and BW535, when compared to wild-type strain AB1157. The filamentation observed was a consequence of SOS induction, triggered by attachment to the glass surface. In part, the filamentation and SOS induction observed in these experiments were related to ROS production. We also quantified interacted bacterial cells and it was observed moderated biofilm formation in all strains tested. However, biofilm formation depended on the ability of the bacteria to induce the SOS response, under both aerobic and anaerobic conditions. The oxygen tension was important factor for interaction of the BER mutants, since these mutants exhibited decreased quantitative adherence under anaerobic conditions. However, this decrease was not related to BER mutants death. Scanning electron microscopy was also performed in the wild-type strain and BER mutants (BW9091 e BW535) and biofilm formation was confirmed under both aerobic and anaerobic conditions. We observed a structure similar to a extracellular matrix which involved biofilms of wild type strain (AB1157) and xthA mutant (BW9091). However, the formation of this structure by both strains depended on the oxygen tension, since biofilm formation, under anaerobiosis condition, did not presented this structure. In conclusion, was provided that bacterial interaction to biotic and abiotic surfaces can lead to damage of bacterial genome, resulting in SOS induction and associated filamentation.

Estudo do reparo das lesões induzidas no DNA de Escherichia coli pela radiação ultravioleta C (UVC) / Study of the repair of lesions induced in Escherichia Coli DNA by ultravioletc radiation (UVC)

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / Didaticamente, podemos dividir o espectro da radiação ultravioleta (UV) em três faixas: UVA (400 a 320 nm), UVB (320 a 290 nm) e UVC (290 a 100 nm). Apesar do UVC ou UV-curto ser eficientemente filtrado pela camada de ozônio da Terra e sua atmosfera, este é uma das faixas do espectro de UV mais usadas para explorar as consequências de danos causados ao DNA, já que a letalidade induzida por este agente está relacionada aos danos diretos no genoma celular, como as lesões dímero de pirimidina, que são letais se não reparadas. Contudo, demonstrou-se que a radiação UVC pode gerar espécies reativas de oxigênio (ERO), como o oxigênio singleto (1O2). Embora, o radical hidroxil (OH) cause modificações oxidativas nas bases de DNA, alguns trabalhos indicam que o 1O2 também está envolvido nos danos oxidativos no DNA. Esta ERO é produzida por vários sistemas biológicos e reações fotossensibilização, quando cromóforos são expostos à luz visível ou são excitados pela luz UV, permitindo que essa energia possa ser transferida para o oxigênio sendo convertido em 1O2, que é conhecido por modificar resíduos de guanina, gerando 8-oxoG, que caso não seja reparada pode gerar uma transversão GC-TA. O objetivo deste trabalho foi o de elucidar a participação de ERO nos efeitos genotóxicos e mutagênicos gerados pela radiação UVC, assim como as enzimas envolvidas no processo de reparação destas lesões em células de Escherichia coli. Nos ensaios as culturas foram irradiadas com o UVC (254 nm; 15W General Electric G15T8 germicidal lamp, USA). Nossos resultados mostram que o uso de quelantes de ferro não alterou a letalidade induzida pelo UVC. A azida sódica, um captador de 1O2, protegeu as cepas contra os danos genotóxicos gerados pelo UVC e também diminuiu a frequência de mutações induzidas no teste com rifampicina. A reversão específica GC-TA foi induzida mais de 2,5 vezes no ensaio de mutagênese. A cepa deficiente na proteína de reparo Fpg, enzima que corrige a lesão 8-oxoG, apresentou menos quebras no DNA do que a cepa selvagem no ensaio de eletroforese alcalina. A letalidade induzida pelo UVC foi aumentada nos mutantes transformados com o plasmídeo pFPG, ao mesmo tempo que representou uma redução na indução mutagênica. Houve dimuição na eficiência de transformação com plasmídeo pUC 9.1 na cepa fpg quando comparado a cepa selvagem. Assim como, um aumento da sensibilidade ao UVC na associação entre mutantes fpg e uvrA. Estes resultados mostram que o 1O2 participa dos danos induzidos pelo UVC, através da geração da lesão 8-oxoG, uma lesão mutagênica, que é reparada pela proteína Fpg / Didactically, we can divide the ultraviolet radiation (UV) spectrum into three bands: UVA (400 to 320 nm), UVB (320-290 nm) and UVC (290-100 nm). Despite the UVC or far-UV be efficiently filtered by Earths ozone layer and its atmosphere, this is one of bands of UV spectrum used to explore the consequences of DNA damages, since the UVC-induced lethality is related to direct damage in genome cells, such as pyrimidine dimers, which are lethal if not repaired. However, it was shown that UVC radiation can generate reactive oxygen species (ROS) such as singlet oxygen (1O2). Although hydroxyl radical (OH) cause oxidative modifications in DNA bases, some works suggests that 1O2 is also involved in oxidative DNA damage. This ROS is produced by several biological systems and photosensitivity reactions when chromophores are exposed to visible light or excited by UV light, allowing that energy can be transferred to the oxygen being converted to 1O2, which is known to modify guanine residues, generating 8-oxoG, if not repaired can lead to a GC-TA transversion. The objective of this work was to elucidate the ROS involvement in the genotoxic and mutagenic effects generated by UVC radiation, as well as the enzymes involved in the repair process of these lesions in Escherichia coli cells. In the assays, cultures were irradiated with UVC (254 nm, 15 W General Electric germicidal lamp G15T8, USA). Our results show that the use of iron chelators did not affect the UVC-induced lethality. The sodium azide, a 1O2 quencher, protected strains against the genotoxic damage produced by UVC and also decreased the frequency of mutations induced in rifampicin assay. Reversal specific GC-TA was induced more than 2.5 fold in the mutagenesis assay. The deficient strain in the repair protein Fpg, an enzyme that corrects 8-oxoG lesions, had less DNA breakage than the wild strain in electrophoresis alkaline assay. The UVC-induced lethality was increased in mutants transformed with the pFPG plasmid, while representing a decrease in mutagenic induction. There was a reduction in the transformation efficiency with plasmid pUC 9.1 on Fpg-strain compared to wild-type strain. Also there was a lethality increase in the association between fpg and uvrA mutants. These results shows that 1O2 participates in UVC-induced damages through the generation of 8-oxoG lesion, a mutagenic lesion that is repaired by the Fpg protein.

Parental separation at birth and maternal depressed mood in pregnancy: associations with schizophrenia and criminality in the offspring

Abstract Early risk factors of the antenatal period and infancy have been increasingly linked to psychiatric disorders. The aim of this thesis was to study the associations between very early parental separation and maternal depressed mood in pregnancy on the other hand, and schizophrenia and criminality in the offspring in adolescence and adulthood, on the other, in two data sets. In the Christmas Seal Home Children Study the index cohort consisted of 3 020 subjects born in Finland in 1945–65 who were temporarily isolated from their family immediately after birth to nursing homes, the Christmas Seal Homes, due to tuberculosis in the family. The average separation time was seven months. For every index subject, two reference subjects were matched for sex, year of birth and place of birth. Data were obtained on schizophrenia from the Finnish Hospital Discharge Register (FHDR) in 1971–98 and on criminal offences from Statistics Finland in 1977–98. The 28-year cumulative incidence of schizophrenia was 1.6% both in the index cohort and in the reference cohort (RR 1.0; 95% CI 0.8–1.4). Both male and female index subjects had committed crimes more commonly than the reference subjects (in men RR 1.3; 95% CI 1.2–1.4; in women RR 1.5; 1.2–2.0). Of the male index subjects 12.1% as compared with only 7.1% of the reference cohort had committed violent offences (RR 1.7; 1.4–2.1). In the Northern Finland 1966 Birth Cohort mothers of 12 058 babies were asked at mid-gestation at the antenatal clinic if they felt depressed. This general population birth cohort of the children was followed up for 31 years being record-linked with the FHDR covering the years 1982–97 and with the criminal register of the Ministry of Justice up to 1998. We divided the schizophrenia patients into those having a psychotic first-degree relative (schizophrenia patients with familial risk for psychosis FR) and those without one. The cumulative incidence of hospital-treated schizophrenia was 1.3% among the offspring of depressed mothers and 0.9% among the descendants of non-depressed mothers (RR 1.5; 95% CI 0.9–2.4). The prevalence of antenatal depression was 35% in mothers of schizophrenia patients with FR. The respective prevalence was 14% both in the mothers of schizophrenia patients without FR and in the mothers of other cohort members. Both male and female offspring of antenatally depressed mothers were more commonly criminal offenders than offspring of non-depressed mothers (in men adjusted OR 1.5; 95% CI 1.2–1.9; in women OR 1.5; 0.8–3.0). In males, 6.5% with depressed mothers and 3.2% with non-depressed mothers had committed violent offences (adjusted OR 1.6; 1.1–2.4). Very early separation and mothers' depressed mood in pregnancy are per se unlikely to increase the risk for schizophrenia in the offspring, but seem to be connected to criminal behaviour, especially violent criminality in men. / Tiivistelmä Raskaus- ja imeväisajan varhaiset tekijät on lisääntyvästi yhdistetty lapsen tuleviin mielenterveyshäiriöihin. Tarkoituksena oli tutkia hyvin varhaisen eron (separaation) ja äidin raskaudenaikaisen masentuneen mielialan yhteyttä lasten skitsofreniaan ja rikollisuuteen nuoruudessa ja aikuisuudessa kahdessa eri aineistossa. Joulumerkkikoti-lasten tutkimuksessa indeksikohortti koostui 1945–65 syntyneistä 3 020 tutkittavasta, jotka erotettiin väliaikaisesti perheistään heti syntymän jälkeen hoitokoteihin, Joulumerkkikoteihin, perheen tuberkuloosin takia. Ero vanhemmista kesti keskimäärin seitsemän kuukautta. Jokaiselle indeksitutkittavalle valittiin kaksi sukupuolen, syntymävuoden ja -paikan mukaan kaltaistettua verrokkitutkittavaa. Tieto skitsofreniaan sairastumisesta hankittiin sairaaloiden poistoilmoitusrekisteristä vv. 1971–98 ja rikoksista Tilastokeskuksesta 1977–98. 28 vuoden kumulatiivinen sairastuvuus skitsofreniaan oli 1,6 % sekä indeksi- että verrokkikohortilla (riskisuhde RR 1.0; 95 %:n luottamusväli CI 0,8–1,4). Sekä miehistä että naisista indeksitutkittavat olivat tehneet useammin rikoksia kuin vertailuryhmä (miehillä RR 1,3; 1,2–1,4; naisilla RR 1,5; 1,2–2,0). Miehistä 12,1 % indeksitutkittavista ja vain 7,1 % vertailuryhmästä oli tehnyt väkivaltarikoksen (RR 1,7; 1,4–2,1). Pohjois-Suomen 1966 syntymäkohortin 12 058 lapsen äideiltä kysyttiin keskiraskauden aikana äitiysneuvolassa, kokivatko he mielialansa masentuneeksi. Tämän väestötason syntymäkohortin (siis lasten) tietoja hankittiin 31-vuotisseurannassa sairaaloiden poistoilmoitusrekisteristä vuosilta 1982–97 ja oikeusministeriön rikosrekisteristä vuoteen 1998. Skitsofreniaan sairastuneet jaettiin niihin, joiden 1. asteen sukulainen oli ollut / ei ollut ollut psykoottinen. Sairaalahoitoa vaatineen skitsofrenian kumulatiivinen sairastuvuus oli 1,3 % masentuneiden ja 0,9 % masentumattomien äitien lapsilla (RR 1,5; 0,9–2,4). Raskaudenaikaisen masentuneen mielialan esiintyvyys oli 35 % niiden skitsofreniapotilaiden äideillä, joilla oli ollut lähisuvussa psykoosia. Vastaavasti masentunutta mielialaa esiintyi 14 %:lla sekä niiden skitsofrenia-potilaiden äideistä, joilla ei ollut sukurasitusta, että muiden kohorttitutkittavien äideistä. Sekä masentuneiden äitien pojista että tyttäristä useampi oli tehnyt rikoksen kuin masentumattomien äitien lapset (miehillä vakioitu vedonlyöntisuhde OR 1,5; 1,2–1,9; naisilla OR 1,5; 0,8–3,0). Masentuneiden äitien pojista 6,5 % ja masentumattomien äitien pojista 3,2 % oli tehnyt väkivaltarikoksen (vakioitu OR 1,6; 1,1–2,4). Hyvin varhainen ero ja äidin masentunut mieliala raskauden aikana eivät todennäköisesti sinänsä lisää skitsofrenian vaaraa lapsilla, mutta näyttävät olevan yhteydessä lasten rikolliseen käyttäytymiseen, erityisesti väkivaltarikoksiin miehillä.

Este estudo se propôs a avaliar o potencial inflamatório das partículas eliminadas na exaustão do diesel (PED) e 1,2-naftoquinona (1,2-NQ) sobre outros compartimentos, como o músculo liso vascular (aorta torácica; RTA) e do corpo cavernoso isolados de ratos (RCC) e os mecanismos envolvidos via ensaios funcionais e bioquímicos. A injeção i.tr. das PED e 1,2-NQ em ratos Wistar causou inflamação e hiporreatividade das vias aéreas associados ao aumento significativo do relaxamento evocado pela ACh na RTA. No RCC desses mesmos animais, tanto o GTN quanto o estímulo elétrico (EFS) causou maior relaxamento. O conteúdo basal de TBARs na RTA e pulmão foi reduzido, embora outros testes indicadores de estresse oxidativo e / ou atividade antioxidante não mostraram diferenças entre os grupos. As taxas de expressão gênica / protéica da nNOS no RCC de ratos não diferiram do grupo controle, mas a expressão da eNOS e iNOS foi reduzida na RTA e RCC. Não foram quantificadas concentrações séricas do TNF<font face=\"Symbol\">&#945 ou IL-1<font face=\"Symbol\">b, sugerindo que os efeitos sistêmicos ocorrem independentemente destas citocinas. Conclui-se que o tratamento agudo de ratos com a mistura de poluentes induziu inflamação das vias aéreas (e hiporreatividade), capaz de afetar outros compartimentos, como a musculatura lisa vascular (RTA) e do RCC. / We tested the hypothesis that local inflammation in the airways evoked by intra-tracheal instillation of the environmental chemical 1,2-naphthoquinone (1,2-NQ) and diesel exhaust particles (DEP) is capable of targeting other systemic compartments, such as vessels (rat thoracic aorta; RTA) and corpus cavernosum (RCC), and possible involved mechanisms. After 3h, this treatment induced airways hyporresponsiveness to ACh and local inflammation. This effect was associated with decreased numbers of leukocyte in the blood and spleen and increased number of leukocytes in the bone marrow. Pollutant treatment also markedly increased ACh-induced relaxation in RTA and by both GTN- and electrical stimulation-induced relaxation in RCC. Exposure to pollutants did not affect FE-induced contraction in RTA. Neither serum levels of cytokines (TNF<font face=\"Symbol\">&#945 e IL-1<font face=\"Symbol\">b) nor basal concentration of total nitrate were different amongst groups. No evidence of increased catalase activity in RTA, RCC and lung was found. The treatment reduced the eNOS e iNOS gene expression in RTA e RCC, without significantly affecting the nNOS gene expression in RCC. Our results are consistent with the hypothesis that DEP-induced airways hiporresponsiveness and inflammation can account to produce systemic changes, such as structural and functional changes in the RTA and RCC by means of substances derived from endothelium or due to the ability of these pollutants to act to stimulate the production of scavenger of free radical.

The impacts of the Canterbury earthquakes on educational inequalities and achievement in Christchurch secondary schools

During 2010 and 2011, major earthquakes caused widespread damage and the deaths of 185 people in the city of Christchurch. Damaged school buildings resulted in state intervention which required amendment of the Education Act of 1989, and the development of ‘site sharing agreements’ in undamaged schools to cater for the needs of students whose schools had closed. An effective plan was also developed for student assessment through establishing an earthquake impaired derived grade process. Previous research into traditional explanations of educational inequalities in the United Kingdom, the United States of America, and New Zealand were reviewed through various processes within three educational inputs: the student, the school and the state. Research into the impacts of urban natural disasters on education and education inequalities found literature on post disaster education systems but nothing could be found that included performance data. The impacts of the Canterbury earthquakes on educational inequalities and achievement were analysed over 2009-2012. The baseline year was 2009, the year before the first earthquake, while 2012 is seen as the recovery year as no schools closed due to seismic events and there was no state intervention into the education of the region. National Certificate of Educational Achievement (NCEA) results levels 1-3 from thirty-four secondary schools in the greater Christchurch region were graphed and analysed. Regression analysis indicates; in 2009, educational inequalities existed with a strong positive relationship between a school’s decile rating and NCEA achievement. When schools were grouped into decile rankings (1-10) and their 2010 NCEA levels 1-3 results were compared with the previous year, the percentage of change indicates an overall lower NCEA achievement in 2010 across all deciles, but particularly in lower decile schools. By contrast, when 2011 NCEA results were compared with those of 2009, as a percentage of change, lower decile schools fared better. Non site sharing schools also achieved higher results than site sharing schools. State interventions, had however contributed towards student’s achieving national examinations and entry to university in 2011. When NCEA results for 2012 were compared to 2009 educational inequalities still exist, however in 2012 the positive relationship between decile rating and achievement is marginally weaker than in 2009. Human ethics approval was required to survey one Christchurch secondary school community of students (aged between 12 and 18), teachers and staff, parents and caregivers during October 2011. Participation was voluntary and without incentives, 154 completed questionnaires were received. The Canterbury earthquakes and aftershocks changed the lives of the research participants. This school community was displaced to another school due to the Christchurch earthquake on 22 February 2011. Research results are grouped under four geographical perspectives; spatial impacts, socio-economic impacts, displacement, and health and wellbeing. Further research possibilities include researching the lag effects from the Canterbury earthquakes on school age children.

Microssistemas eletroforéticos em materiais poliméricos de duplo canal com detecção amperométrica / Electrophoretic microsystems in polymeric materials dual channel with amperometric detection

Electrophoretic microsystems (EM) are powerful tools for the separation of species of microsystems analyzes which can easily be combined with electrochemical detection (ECD) and therefore making it ideal for a method of detection. However, the influence of high voltage at the working electrode used for the separation is a problem to be overcome due to the increased signal/noise ratio and possible damage of the electrode and/or the potentiostat. Thus, it was proposed in this thesis one EM hybrid PDMS / glass configuration with dual-channel potentiostat coupled to an electrically isolated in order to minimize the influence of high potential in the separation channel and improve the separation efficiency of the species and subsequently, improve detection limits. The EM contains two separate parallel channels 200 microns and a channel separation and another reference, and each containing a platinum electrode 15 or 50 μm placed about 1 to 4 μm in the channel. An electrode served as the working electrode, positioned in the separation channel, and another electrode as reference electrode, placed in the reference channel. This configuration associated with the electrically isolated potentiostat allowed the amperometric signals were measured without any change or potential interference arising from the high voltage applied separation. Aiming to evaluate the effectiveness of the methodology proposed in this thesis, samples nitrite, tyrosine and peroxynitrite (reactive nitrogen species – RNS), hydrogen peroxide (reactive oxygen species – ROS), ascorbic acid, glutathione and cysteine were injected into the channel containing the working electrode, while simultaneously boric acid buffer pH 11 containing TTAB was injected into the reference channel containing the reference electrode. From this configuration, we obtained a significant reduction in noise level (about 0.94 pA) and a relative improvement in the resolution ratified by electropherograms, compared with using single channel configuration. The limits of detection (LOD) for the chemical species mentioned above were 0.58 μM, 0.14 μM, 0.75 μM, 0.21 μM, 0.82 μM, was not obtained for cysteine and 1.63 μM, respectively. The efficiency can also be seen by analyzing nitrite performed on samples of perfusate blood of sheeps and rats, where have been detected a concentration of 68.05 μM and 22.04 μM, respectively, by the proposed method. It was also proposed in this thesis, microfabrication and evaluation of a PMMA electrophoretic microsystem with single channel configuration coupled to a base made of the same material to fix the microchip with electrochemical detection using a carbon paste electrode. The purpose of the construction of the base was to obtain, by fixing, reproducibility of events. And the microfabrication of PMMA EM aimed the viability of its use in analysis perspective as having the lowest cost per unit made due to the use of CO2 laser for microfabrication, which has a value considerably lower, compared with photolithographic processes. The evaluation of this system was performed through the analysis standards of serotonin and acetaminophen, which proved that the microfabrication of this system showed good reproducibility and repeatability of events, making it viable processing. / Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / Os microssistemas eletroforéticos (MSE) são ferramentas poderosas para a separação de espécies em microssistemas de análises, onde pode ser facilmente combinada com detecção eletroquímica (DEQ) e tornando-se, portanto, um método de detecção ideal. No entanto, a influência da alta tensão no eletrodo de trabalho utilizada para a separação é um problema a ser contornado devido o aumento da relação sinal/ruído e possíveis danificações do eletrodo e/ou do potenciostato. Assim, foi proposto nesta tese um MSE híbrido de PDMS/vidro com configuração de duplo-canal acoplado a um potenciostato eletricamente isolado com objetivo de minimizar a influência do elevado potencial no canal de separação e melhorar a eficiência de separação das espécies e, subsequentemente, melhorar os limites de detecção. O MSE contém dois canais paralelos separados 200 μm, sendo um canal de separação e outro de referência, e cada um deles contendo um eletrodo de platina de 15 ou 50 μm colocados cerca de 1 a 4 μm dentro do canal. Um eletrodo serviu como eletrodo de trabalho, posicionado no canal de separação, e o outro eletrodo como eletrodo de referência, posicionado no canal de referência. Essa configuração associado ao potenciostato eletricamente isolado permitiu que os sinais amperométricos fossem medidos sem qualquer mudança de potencial ou de interferência oriunda da alta tensão de separação aplicada. Objetivando avaliar a eficiência da metodologia proposta nessa tese, amostras de nitrito e peroxinitrito (espécies reativas de nitrogênio – ERN), tirosina, peróxido de hidrogênio (espécie reativa de oxigênio – ERO), ácido ascórbico, glutationa e cisteína foram injetadas no canal contendo o eletrodo de trabalho, enquanto que simultaneamente o tampão de ácido bórico contendo TTAB pH 11 foi injetado no canal de referência contendo o eletrodo de referência. A partir desta configuração, obteve-se uma significativa diminuição no nível de ruído (cerca de 0,94 pA) e uma relativa melhora na resolução ratificadas pelos eletroferogramas, se comparado com a configuração que utiliza canal único. Os limites de detecção (LOD) para as espécies químicas supracitados foram de 0,58 μM, 0,14 μM, 0,75 μM, 0,21 μM, 0,82 μM, não foi obtida para a cisteína, e 1,63 μM, respectivamente. A eficiência também pode ser vista através das análises de nitrito realizadas em amostras de perfusato de sangue de ovelhas e ratos, onde foram detectados uma concentração de 68,05 μM e 22,04 μM, respectivamente, através da metodologia proposta. Foi proposto também nessa tese, a microfabricação e avaliação de um microssistema eletroforético de PMMA com configuração de canal único acoplado a uma base feita do mesmo material para fixar o microchip, com detecção eletroquímica usando eletrodo de pasta de carbono. O objetivo da construção da base foi obter, através da fixação, reprodutibilidade de eventos. E a microfabricação do MSE de PMMA objetivou a viabilidade do seu uso em análises tendo como perspectiva o baixo custo por unidade confeccionada devido ao uso de laser de CO2 para a microfabricação, o qual possui um valor agregado consideravelmente menor, se comparado com os processos fotolitográficos. A avaliação desse sistema foi feita através das análises de padrões de serotonina e acetaminofeno, onde comprovou-se que a microfabricação desse sistema apresentou boa reprodutibilidade e repetitividade de eventos, tornando-se viável o seu processamento.

Cette thèse porte sur l'étude de micro-plasmas froids à la pression atmosphérique générés à partir de différents réacteurs des configurations basées sur le principe des Décharges à Barrière Diélectrique (DBD) et alimentés par des générateurs de tension impulsionnelle et sinusoïdale. Les plasmas sont formés dans des gaz nobles tels que l'hélium et l'argon (gaz vecteurs), et également dans des mélanges réalisés avec des gaz moléculaires tels que l'azote et l'oxygène afin de produire des Espèces Réactives de l’Azote et de l’Oxygène (ERA, ERO). La (ré)activité chimique du plasma est ainsi supposée être accrue, permettant le traitement de matériaux inertes ou vivants pour diverses applications (fonctionnalisation de surfaces, inactivation de cellules, régénération de tissus vivants, etc.). La caractérisation des plasmas étudiés est réalisée en enregistrant les aspects électriques et optiques en fonction des paramètres élémentaires, comme l’amplitude et la fréquence de la tension, le débit du gaz, la configuration des électrodes, et le rapport cyclique dans le cas du régime pulsé. Ainsi, la (ré)activité chimique des plasmas est évaluée tandis que au même temps les mécanismes de la génération des plasmas et les façons de l’optimisation de la chimie sont dévoilées. Finalement, nous examinons l'efficacité du plasma dans le domaine biomédical en traitant divers systèmes biologiques (bactéries, liposomes, cellules) sans effets thermiques. / The present PhD thesis is devoted to the study of atmospheric pressure cold micro-plasmas produced in different Dielectric Barrier Discharge (DBD) reactors which are driven by pulsed or sinusoidal high voltages. Noble gases such as helium and argon are used as carrier gases, whereas admixtures with nitrogen and oxygen are studied as well. The formation of Reactive Nitrogen and Oxygen Species (RNS, ROS) is thus achieved, and the possibility of improving the chemical (re)activity of the plasmas is demonstrated. This is of interest in the treatment of inert or living materials (e.g. surface functionalization, cell inactivation, living tissue regeneration, etc.). Plasmas are characterized by recording electrical and optical features as a function of principal operational parameters, including voltage amplitude and frequency, gas flow rate, electrode configuration, and voltage duty cycle in the case of pulsed waveform. The physico-chemical (re)activity of the plasmas is thus evaluated, while at the same time mechanisms on the plasma generation and paths for chemistry optimization are unveiled. Finally, the efficiency of the plasma in relation to biomedical applications is tested by treating different biological systems (bacteria, liposomes, cells) while preventing any thermal effect.
