Screening of ligand binding behavior using a high-throughput method and development of guidelines for a learning material / Kartläggning av liganders bindningsbeteende genom en screeningmetod och utvecklandet av riktlinjer för ett utbildningsmaterial

This study is comprised of two distinct parts, a chromatographic study and a didactic study.Chromatographic studyThe need for higher selectivity in chromatography purification has increased in recent years. Multimodal resins, offering novel selectivity, are a possible solution to this demand. The purpose of this study is to develop new multimodal resins with novel selectivity, using an iterative workflow. Therefore, eleven novel multimodal ligands were screened according to binding behavior using a high-throughput (HT) method. The mapping of binding behaviors was comprised of six proteins and 32 different binding buffers, with various salt concentrations and pH-levels, to allow for a wide, but efficient mapping. The data generated from the screenings were presented using binding capacity and partition coefficient and were evaluated against each other using principal component analysis (PCA). The PCA created a ligand diversity map, which separated ligands in respect to binding behavior. The information supplied by the diversity map can be used for selecting ligands for further research. The results from presenting the data in the form of partition coefficient and binding capacity were comparable, which indicated that the effects of initial protein concentrations were low. However, this study found that there are advantages and disadvantages to using both quantities. It is possible that some of the ligands screened in this study will be used in further research and it is likely that the diversity map will facilitate the development of ligands with novel selectivityDidactic studyWith a rising demand for HT screening as a method to evaluate the binding behaviors of novel ligands, in combination with the UN’s goal for quality education for all, the incentives for developing quality educational materials for HT screenings have increased. A first step towards educational materials is to outline common difficulties, thereby creating guidelines for developing educational materials. In this study, guidelines for the development of learning materials for HT screenings have been produced. The guidelines were based on identification of critical steps for the experimental procedure and utilizing employer experience of learning materials in general. Two methods for data collection was used in the didactic study: observations and interviews. The observations were performed during the training of the screening method. For the interviews, three individual and one focus group interview were held. Thereafter, a thematic analysis was performed on the data to generate themes. The results from the observations and interviews indicated that the critical steps of the learning process were the practical parts. The result of the thematic analysis showed the identification of three themes: Cohesion, Facilitation and Interaction. These themes generated four aspirational guidelines: 1. Identify crucial steps of the practical procedure. 2. Adapt to people with different backgrounds. 3. Maintain coherence throughout the learning process. 4. Utilize strategies for interaction, both human interaction and material interaction.

Denna studie har syftat till att nå ökad kunskap om hur myndigheter kan utveckla sin kriskommunikation med barn i åldern 12-13 år, utifrån ett barnrättsperspektiv. Webbplatsen Lilla Krisinfo har utgjort ett exempel för studien. Studiens teoretiska perspektiv har bestått av inkodning- och avkodningsmodellen, användning- och gratifikationsteorin, förväntning-bekräftelseteorin samt retorik, vilka har sammanförts i en egen teoretisk modell. Metoderna som användes var retorisk analys av webbplatsen och fokusgruppsintervjuer med barn. Genom att undersöka barns informationsbehov och informationssökningsstrategier gällande kriser samt deras uppfattning av Lilla Krisinfos webbplats har studien kunnat nå ökad kunskap för att besvara syftet. De huvudsakliga resultaten innefattar att myndigheter bör betona krisers relevans för barn, skapa trygghet men samtidigt inte undanhålla information, anpassa sitt språk efter olika nivåer och sträva efter att vara och upplevas trovärdiga. / This study has aimed to reach further knowledge about how authorities can develop their crisis communication with children aged 12-13, based on a children's rights perspective. The website Lilla Krisinfo has been an example for the study. The study's theoretical perspective has consisted of the encoding and decoding model, the uses and gratification theory, the expectation-confirmation theory and rhetoric, which were brought together in a theoretical model. The methods used were rhetorical analysis of the website and focus group interviews with children. By investigating children's needs of information and strategies for searching information regarding crises as well as their perception of Lilla Krisinfo's website, the study has been able to achieve further knowledge to answer to the study’s purpose. The main results include that authorities should accentuate the relevance of a crisis for children, create a feeling of security but at the same time not withhold information, adapt the language to different levels and strive to be perceived as credible.

As digitalization and technology has quickly advanced over the recent years, marketers are becoming increasingly reliant on collecting user data, and targeted advertising has therefore come to be viewed as a form of data surveillance. Due to marketers' ability to analyze online behavior and target consumers with personalized advertising, users are becoming more concerned about their online privacy. Therefore, the main purpose of this research study is to examine different perceptions, attitudes, and experiences towards online privacy, specifically towards the aspect of targeted advertisement. Furthermore, the fast-developed technological world is constantly changing people's perspectives. Hence, this paper seeks to investigate whether different generations of media users have different perceptions. The empirical material for this study was gathered through a qualitative approach and was collected through two focus group interviews conducted from a comparative angle. The findings revealed that both generations were very negative and critical of data collection and showed mistrust of digital media platforms. Furthermore, despite both generations expressing strong privacy concerns and claiming to value privacy highly, their actions demonstrated the opposite, which is consistent with the theoretical concept of privacy paradox. Moreover, the findings revealed that certain attitudes and perceptions towards online privacy differ between digital natives and digital immigrants. The most significant distinction between the generations is the substantial lack of knowledge and awareness of digital immigrants. The findings suggest that digital immigrants need to be more enlightened regarding the function of new technological aspects, such as targeted advertisements. This is required for them to be more informed, hence reducing the risk of them feeling misled, concerned, and monetized. For users to feel secure using the internet, more laws and regulations are needed. / I takt med att digitaliseringen och tekniken har utvecklats snabbt under de senaste åren, blir marknadsförare alltmer beroende av att samla in användardata, och riktad reklam har på grund av detta kommit att ses som en form av dataövervakning. På grund av marknadsförares förmåga att analysera onlinebeteende och rikta in sig på konsumenter med personlig reklam, blir användarna mer oroade över sin integritet online. Således är det huvudsakliga syftet med denna forskningsstudie att undersöka olika uppfattningar, attityder och erfarenheter av integritet på nätet, särskilt när det gäller aspekten av riktad reklam. Dessutom förändrar den snabbt utvecklade teknologiska världen ständigt människors perspektiv. Därför försöker denna artikel undersöka om olika generationer av medieanvändare har olika uppfattningar. Det empiriska materialet för denna studie samlades in genom ett kvalitativt tillvägagångssätt, och samlades in genom två fokusgruppsintervjuer genomförda in en jämförande vinkel. Resultaten avslöjade att båda generationerna var mycket negativa och kritiska till datainsamling och visade misstro mot digitala medieplattformar. Dessutom, trots att båda generationerna uttryckte stark oro över sin integritet online och hävdade att de värderade integritet högt, visade deras handlingar motsatsen. Detta går i linje med det teoretiska konceptet om integritet paradoxen. Dessutom avslöjade resultaten att vissa attityder och uppfattningar om integritet på nätet skiljer sig åt mellan digitala natives och digitala immigrants. Den viktigaste skillnaden mellan generationerna är den betydande bristen på kunskap och medvetenhet hos digitala immigrants. Resultaten tyder på att digitala immigrants behöver bli mer upplysta om funktionen av nya tekniska aspekter, såsom riktade annonser. Detta krävs för att de ska vara mer informerade, vilket minskar risken för att de känner sig vilseledda, oroliga och övervakade. För att användarna ska känna sig säkra på internet behövs fler lagar och förordningar.

The aim of this African culturally based research is to establish whether a group of unmarried pregnant teenage girls experience rejection during their period of pregnancy as well as to determine whether they experienced rejection for whatever reason before falling pregnant. Seen in the context of the communal and person-centred life view as upheld by the African culture where respect for and well-being of the individual within the tribal and extended family structure is paramount, one would expect that unmarried girls finding themselves in a precarious situation such as premature pregnancy would experience understanding, acceptance and support. The research is quantitative as well as qualitative. The quantitative research focuses on 341 grade 11-leamers at seven secondary schools in the Free State Province of Education as well as in the Gauteng Department of Education. Questionnaires were presented with the aim of establishing the reason for teenage pregnancy; to research the thoughts, feelings and actions of the peer group toward pregnant teenage girls; to determine the presence of feelings of rejection of a group of pregnant teenage girls at the same schools; to determine the thoughts the girls have about themselves as well as about the future. On account of the fact that that the responses of the pregnant girls seemingly contradicted the responses given by the grade 11-peer group sample it was decided to supplement the quantitative research result with a qualitative research investigation by means of focus group interviews conducted with twelve willing to participate pregnant girls at two ofthe secondary schools who voluntarily agreed to participate. The research result indicates that although the pregnant girls did not experience rejection before falling pregnant they did however experience rejection after falling pregnant, especially from the peer group at school, educators, and certain sections of society. Dissertation / This study explores the role of the principal in the management of parent involvement in secondary schools in Northern Province. A literature study investigated existing theories and models of parent involvement; parent involvement in the South African context and the provision of education in South Africa in historical perspective. It also investigated the managerial role of the principal in parent involvement. A qualitative investigation in three secondary schools in Northern Province was done. Data were gathered by means of participant observation in-depth interviews with principals and focus-group interviews with teachers and three parents on the school governing bodies. Analysed data revealed the following findings: principals lack knowledge on parent involvement and schools lack policies and organisational structures dealing with parent involvement as well as home-school relations. Alternative measurement instruments for the evaluation of learner achievement in business management in the Technikon S A

Thesis / This research is mainly concerned with determining whether the existing evaluation methods/measurement instruments used in Business Management at Technikon SA, are still suitable, adequate and acceptable enough for continued use, or whether there is a need for the introduction of alternative methods. The first objective in this research was to determine what the exact role and function of evaluation in the education process is. On the basis of literature, it was determined that evaluation, and by implication measurement, is a crucial component of the education process and that no real teaching or learning can occur without it. Second, an attempt was made to determine what the nature and extent of the shortcomings of the existing measurement instruments were. Also on the basis of literature it was found that there are numerous and serious disadvantages in the existing measurement instruments. Third, the researcher also attempted to determine whether any other (alternative) measurement instruments, in addition to the existing ones, were available. It was discovered that there are a variety of alternative evaluation/measurement instruments available. Fourth, the measurement instruments used in Business Management were analysed and compared to the Business Management curriculum objectives to determine whether they were indeed suitable for the subject. It was found that, although a positive correlation seems to exist between the measurement instruments and the curriculum objectives, there were certain definite shortcomings in the existing measurement instruments in terms of the realization of some of the curriculum objectives. Fifth, focus group interviews were used as a research method to get an understanding of the views of TSA learners on the aspect of evaluation and the methods/instruments currently used. Most of the participants expressed a need for more practical types of measurement/evaluation. However, many participants felt the written examination should not be abolished entirely, but be used in conjunction with other more practical methods. The management of parent involvement in multicultural schools in the Umlazi district

The aim of this study was to examine the management of parent involvement in multicultural schools in the Umlazi District, Kwa-Zulu Natal. A literature study investigated existing programmes and models of parent involvement, multicultural education in South Africa, legislation pertaining to parent involvement in South Africa and the advantages of and barriers to parent involvement. In addition, the role of school management in managing parent involvement was reviewed. A qualitative investigation of parent involvement in three secondary multicultural schools in Umlazi was conducted. The opinions and experiences of thirty three participants were obtained via focus group interviews. Major findings included: all parents, irrespective of race or culture have an interest in their child's education All parents require guidance and support in parenting skills and also school managers lack understanding with regard to their role in managing parent involvement.
