Imunomodulação por levamisol na imunidade inata e adquirida de pacu (Piaractus mesopotamicus) /

Algumas substâncias podem influenciar as respostas do sistema imune de peixes, como o levamisol, através da modulação de parâmetros imunes, aumentando a resistência contra diversos agentes. Foram avaliados os efeitos do levamisol na dieta em dois experimentos. O primeiro experimento avaliou a suplementação com 0, 125, 250 e 500 mg kg-1 do imunoestimulante administrado por sete e 15 dias, no qual foram avaliados os parâmetros da imunidade inata, hematológicos e bioquímicos. A administração do levamisol por sete e 15 dias promoveu alterações em parâmetros imunológicos e hematológicos. A administração do levamisol pode promover imunomodulação, entretanto a determinação do efeito não ficou claro devido às respostas contraditórias em cada parâmetro avaliado. O segundo experimento consistiu em suplementação com 0, 125, 250 e 500 mg kg-1 do imunoestimulante administrada por sete dias conjuntamente com a imunização com bactérias Aeromonas hydrophila. A imunização e a administração de levamisol promoveram aumento do título de anticorpos, atividade bactericida do soro, hematócrito, número de eritrócitos, leucócitos totais e trombócitos nos pacus. A administração de levamisol por sete dias e a imunização de pacus promoveu melhora de alguns parâmetros da imunidade adquirida e inata de defesa. Entretanto outros protocolos devem ser estudados para avaliar o efeito do levamisol sobre o sistema imune de pacu / Abstract: The emergence of diseases in fish farming is a problem faced in Brazil and worldwide. Some substances can influence the immune system responses of fish, like levamisole, increasing resistance against various etiological agents. Were evaluated the effects of levamisole in diet in two experiments. The first experiment evaluated supplementation with 0, 125, 250 and 500 mg kg-1 imunoestimulante administered by seven and 15 days in which have been assessed the parameters of innate immunity, haematological and biochemists. The administration of levamisole by seven and 15 days promoted changes in haematological and immunological parameters. The administration of levamisole can promote immunodulation, however the determination of the effect not clear due to contradictory answers evaluated in each parameter. The second experiment consisted of supplementation with 0, 125, 250 and 500 mg kg-1 imunoestimulante administered by seven days together with immunization with inactivated Aeromonas hydrophila bacteria with formaldehyde. Immunization and administration of levamisole promoted increase antibody titre, serum bactericidal activity, hematocrit, number of erythrocytes, leucocytes and thrombocytes in pacus totals. Administration of levamisole for seven days and immunization pacus promoted improves some parameters of innate and acquired immunity defense. Pectina na ração de frangos de corte: digestibilidade, parâmetros zootécnicos e metabolismo lipídico /

No primeiro experimento, foram avaliados a digestibilidade da ração, a viscosidade e o tempo de trânsito intestinal. Para isso, 240 frangos de corte macho Cobb® foram distribuídos em delineamento inteiramente casualizado (DIC), em 4 tratamentos (0, 1, 3 e 5% de pectina) compostos por 6 repetições de 10 aves por unidade experimental. As análises foram realizadas em duas fases: inicial (14 a 18 dias) e crescimento (27 a 31 dias). Os resultados mostraram que com a ingestão de pectina houve melhora da digestibilidade dos nutrientes na fase inicial dos frangos e piora na fase de crescimento, aumento na viscosidade e no tempo de trânsito intestinal e redução na umidade da excreta. No segundo experimento, foram analisados o desempenho e rendimento de cortes aos 35 e 42 dias de idade, o desenvolvimento semanal das aves e o consumo de água, parâmetros bioquímicos plasmáticos, morfometria do tecido adiposo abdominal, morfometria do fígado e teor de lipídio total hepático e dos músculos peitoral maior e gastrocnêmio. Foram utilizados 720 pintos machos Cobb® distribuidos em DIC, composto por 4 tratamentos (0, 1, 3 e 5% de pectina) e 6 repetições de 30 aves por unidade experimental. Na última semana, o consumo de água aumentou com o aumento do nível de pectina na ração. O consumo de até 1% de pectina na ração mantém o desempenho das aves. Aos 42 dias de idade ocorreu redução nos valores de proteína total plasmática com a ingestão de pectina. Os níveis de 3 e 5% reduziram as concentrações de colesterol total e, o nível de 5% reduziram os níveis de HDL. A ingestão de pectina proporcionou diminuição na concentração de lipídio hepático e menor relação entre peso corporal e gordura abdominal com a ingestão de 5% de pectina aos 42 dias. In the first experiment evaluated the digestibility of feed, the viscosity and intestinal transit time. For this, 240 male Cobb® broilers were distributed in a randomized design in four treatments (0, 1, 3 and 5% pectin) formed by 6 repetitions of 10 birds each. Analyses were performed in two stages: early (14 to18 days) and growth (27 to 31 days). The results showed that the ingestion of pectin had improved the digestibility of nutrients in the initial phase of chickens and worsens during growth, increase in viscosity and intestinal transit time and reduction in moisture of excreta. We used 720 male Cobb® chicks distributed in a completely randomized design with 4 treatments (0, 1, 3 and 5% pectin) consisting of six replicates of 30 birds each. In the last week, water consumption increased with increase level of pectin in the diet. The consumption up to 1% pectin keeps the performance of broilers.. At 42 days of age there was a reduction in total plasma protein values with the intake of pectin. The levels of 3 and 5% reduced the levels of total cholesterol and the level of 5% reduced levels of HDL. The intake of pectin resulted in decreased hepatic lipid content and lower ratio of body weight and abdominal fat with intake of 5% pectin and 42 days. Facteurs influençant la prise de décision sur la vaccination contre le HPV / Study of factors affecting decision-making in HPV vaccination

Le taux de couverture vaccinale contre le papillomavirus humain (HPV) reste faible en France autour de 20%. Ce travail porte sur l’attitude adoptée par la population française afin de comprendre les motivations et les freins à la vaccination HPV.Une série d’études, qualitatives et quantitatives, a été réalisée en 2014-2016 sur deux populations, la première de jeunes filles de 15 à 25 ans et la deuxième de mères ayant des filles âgées de 11 à 16 ans. Notre approche méthodologique est essentiellement basée sur le modèle combiné issu de la théorie du comportement planifié (Theory of Planned Behavior, TPB, Ajzen, 1991) et du modèle des croyances relatives à la santé (Health Beliefs Model, HBM, Rosenstock, 1974). Parmi les modérateurs de la décision les deux systèmes de motivation décrits par Carver et White (1994) ont été retenus. En complément, une étude expérimentale a été conduite basée sur la Théorie des perspectives (Prospect Theory) de Kahneman et Tversky (1979). Les analyses statistiques ont été effectuées avec les logiciels R et SPSS. Plusieurs traitements statistiques ont été opérés sur les données : analyse des comparaisons, analyse en composantes principales (ACP), régressions linéaires multiples, ANCOVA etc.Les résultats apportent de nouveaux éclairages quant au processus de décision d’acceptation ou de refus de vaccination. Le modèle mis en évidence par ces études fait apparaître des facteurs caractéristiques tels que l'attitude favorable à la vaccination, la sécurité du vaccin, le contrôle perçu ainsi que le regret anticipé, qui jouent un rôle déterminant dans le processus de décision. L’environnement médical et familial proche jouent également un rôle essentiel. / The immunization coverage against HPV remains around 20%. The attitude of French girls was studied to understand the motivations and breaks to HPV.Several studies, qualitative and quantitative, were conducted in 2014-2016 on two populations, first sample of young women aged 15 to 25 years and second of mothers of girls aged 11 to 16. Our methodological approach is essentially based on the handset model from The Theory of Planned Behavior (TPB, Ajzen, 1991) and the Health Beliefs Model (HBM, Rosenstock, 1974). To find the moderators of making decision we opted for the two motivational systems described by Carver and White (1994). Finally, the experimental study we were inspired by the Prospect Theory of Kahneman and Tversky (1979). Statistical analyzes were performed using the softwares R and SPSS. Four types of treatment were carried out on the data: Analyze of comparisons (1); Principal Component Analysis (PCA) (2); Linear Regression to explain an intention to be vaccinated by some variables that allows us to make a prediction (3), and ANCOVA (4).The results provide new insights about the process of decision on vaccine acceptance. It appears that the favorable attitude to vaccination, the vaccine safety, the perceived control, and the anticipated regret play an important role in this decision process as well the doctor’s advice and family.

La toxoplasmose est une anthropozoonose cosmopolite due à un protozoaire parasite intracellulaire obligatoire : Toxoplasma gondii. Cette maladie infectieuse est la plus souvent bénigne chez les personnes immunocompétentes mais revêt un caractère de gravité si l’atteinte concerne les femmes enceintes séronégatives ou les personnes immunodéprimées. En plus de cette incidence forte en médecine humaine, la toxoplasmose représente un important problème de santé vétérinaire. A l’heure actuelle, les seuls moyens de lutte contre ce parasite demeurent la chimiothérapie car il n’existe aucune stratégie prophylactique efficace. Le développement d’un vaccin efficace est d’une réelle nécessité et repose sur l’observation qu’une primo-infection par ce parasite confère à l’hôte immunocompétent une réponse immunitaire protectrice efficace à long terme et qui protège lors d’une réinfection et en particulier contre le risque d’une infection congénitale. / Toxoplasmosis is a cosmopolitan anthropozoonosis due to the obligate intracellular protozoan parasite Toxoplasma gondii. This infectious disease is most often benign in immunocompetent individuals but is particularly severe for pregnant women or immunocompromised patients. In addition to its significant impact on human medicine, toxoplasmosis is a major veterinary health problem. Currently, the only means to fight this parasite remain chemotherapy because there is no effective prophylactic strategy. The development of an effective vaccine is a real challenge and is based on the observation that a primary infection of immunocompetent hosts induces a effective and long-term protective immune response and protects during reinfection and in particular against the risk of congenital infection.

Spalničky - možnosti ovlivnění nepříznivé epidemiologické situace v Evropě / Measles - possibilities of managing unsatisfactory epidemiological situation in Europe

This thesis focuses on the epidemiological situation in the incidence of measles in the Czech Republic and possibilities of influencing their unfavourable situation in Europe. Measles is a highly contagious disease of viral origin, which has a significant impact on the health of the child population in particular. The most effective protection against the measles is vaccination. The introduction of measles vaccination positively affected the epidemiological situation in the world. Priority of the WHO (World Health Organization, WHO, hereinafter) is elimination of the measles. The long term goal of the WHO and the European Centre for Disease Prevention and Control (ECDC hereinafter) was the elimination of measles in the European region by 2010. This goal was not achieved. The cause was low immunization coverage of certain population groups in several European countries. The prerequisite of achieving elimination of measles is that the vaccination coverage in the population is high. Many countries, thanks to effective vaccination strategy, succeeded in reducing the incidence of measles. In the years 2007- 2009 the historically lowest incidence, of less than 10 measles cases per million inhabitants, in the European region was achieved. In subsequent years, there was again a rise in measles morbidity. A repeated rise in cases of the disease is attributed to the expansion of social groups where vaccination coverage was inadequate in terms of achieving herd immunity. The thesis is divided into theoretical and practical part. The theoretical part is divided into four main chapters. The first chapter describes the history of measles, the origin of the name of the disease and the first mention of it. The second theoretical chapter is devoted to the clinical characteristics of measles. In subchapters it describes the clinical picture of the disease, possible complications of the disease, how the infection is diagnosed and what treatment options there are. The third chapter defines the epidemiological characteristics of the disease. The subheads are aimed at the cause of the disease, possible ways of transmission, incubation period, period of communicability, susceptibility and incidence of this disease. At last, the fourth chapter of the theoretical part desribes possible epidemiological measures, which include measles surveillance, preventive and repressive measures and vaccination. The practical part of the thesis was elaborated through quantitative research - secondary data analysis. Analysed data were obtained from the information system for reporting and recording infectious diseases EPIDAT on Regional Hygiene Station of South Bohemian Region, based in Budweis and the European Centre for Disease Prevention and Control. The data on population, in terms of age groups and individual regions, were drawn from the demographic yearbooks, which are available on the website of the Czech Statistical Office. The resulting data were subsequently processed in tabular and graphical forms in Microsoft Excel. The research objectives were: 1) to set charting trends in the incidence of measles in the Czech Republic and Central Europe over the period 2004-2013, 2) to compare the incidence of measles in the individual regions of the Czech Republic, and 3) to identify the reasons for gaps in vaccination coverage against measles in the South Bohemian Region. Based on the above stated objectives, the five research questions were defined. The results are intended to highlight the issues of possible adverse influence on the epidemiological situation in the incidence of measles in Europe. This thesis can be used as a source of information on trends in incidence, vaccine development and immunization status for measles.

Hodnocení úrovně pasivní imunizace u masných telat ve vybraném chovu / The evaluation of passive immunization level in meat calves in selected breeding

The aim of the thesis was to evaluate the influence of selected factors on the level of passive immunization of calves (expressed in total protein, g/l in blood serum) in selected beef cattle breeding. In the monitored period 2014-2016, a total of 205 samples of calf serum were evaluated. The selected factors were: the length of dry period and parity, colostrum quality, the time of first intake of colostrum after birth, the weight and sex of the calf. An adequate level of passive immunity was found in 90.2% of calves. This result was also reflected in low calf mortality rates in monitored years: 1.16% (2014), 1.15% (2015) and 1.23% (2016). At the level of passive immunization, the time of the first colostrum intake (P < 0,001), parity (P = 0,0002), colostrum quality (P = 0,0061) and the length of dry period (P = 0,0008) were the most important factors. The other evaluated factors (sex and birth weight of calves) were not statistically significant. Colostrum period management is essential for the rearing of healthy calves and profitability of breeding.

Utilização de lipídio catiônico para neutralização da carga de DNA e aplicação do complexo na imunização de camundongos.

Embora a imunização gênica apresente vantagens sobre as vacinas convencionais, nem sempre a produção de anticorpos e a conseqüente proteção conferida pelo sistema imune contra o antígeno de interesse são satisfatórias. As causas da baixa eficácia da imunização gênica, nestes casos, ainda são pouco conhecidas, porém um dos pontos mais relevantes em todo o processo é a eficiência da transfecção do DNA. O sucesso desta metodologia depende do estabelecimento de um sistema de entrega de gene eficiente e seguro. De acordo com a teoria proposta de Kuhn et al. (1999), baixas concentrações de moléculas anfifílicas podem ser usadas para neutralizar a carga do DNA, sem a formação de lipossomos. Este trabalho teve como objetivo avaliar o uso do lipídio catiônico dimetildioctadecil brometo de amônio (DDAB) como neutralizador da carga do DNA para a imunização de camundongos. Para a realização dos experimentos, um plasmídeo expressando a proteína do capsídeo (CA) do vírus da artrite encefalite caprina (CAEV), sob o comando do promotor citomegalovírus humano (CMV), foi utilizado nas imunizações. Os camundongos foram divididos em cinco grupos e receberam, via subcutânea, três inoculações de 10 μg de DNA linear (grupo 1), DNA linear mínimo (grupo 2), DNA circular (grupo 3) associados a 2,7 μg de DDAB ou DNA circular nu (grupo 4). O grupo controle foi inoculado com 2,7 μg de DDAB em solução fisiológica. Os camundongos imunizados com o plasmídeo na forma linear associado ao DDAB apresentaram maiores títulos de anticorpos CA que os inoculados com a forma circular ou linear mínima. Os dados sugerem que o lipídio catiônico DDAB tem potencial de uso como veículo de transfecção gênica em animais. / Genetic immunization has several advantages over conventional vaccines. However, antibody production and protection against the antigen of interest are often not satisfactory. In these cases, the causes of the low effectiveness of the genetic immunization are still poorly understood. One of the most relevant points in the immunization process is the DNA delivery. A successful DNA vaccine depends on the establishment of an efficient and safe gene delivery system. In accordance with Kuhn et al. (1999), low concentrations of amphiphilic molecules can be used to neutralize the DNA charge without the formation of liposomes. The aim of this study was to examine the effect of cationic lipid dimethildioctadecyl-ammonium bromide (DDAB) as a charge neutralizer for DNA for the immunization in mice. A plasmid expressing the capsid protein (CA) of caprine arthritis encephalitis virus (CAEV) under the control of the human cytomegalovirus immediate early promoter (hCMVie1) was used in the immunization. Mice were grouped and immunized with three injections of either 10 μg of linear DNA (group 1), minimal linear DNA (group 2) or circular DNA (group 3) associated with 2,7 μg of DDAB or naked circular DNA (group 4). The control group received 2,7 μg of DDAB diluted in physiologic solution. Mice immunized with DNA in linear form associated with DDAB developed higher titers of antibodies against CA than the animals immunized with the circular or minimal linear form. The data suggest that lipidic cationic DDAB could be a useful delivery system for genetic immunization.

Um questionário foi aplicado a 489 estudantes do primeiro ao último período do curso. Utilizou-se a regressão logística não condicional para cálculo do odds ratio com um intervalo de confiança a 95% . A prevalência presumível de hepatite B foi baixa (0,44%), e quanto à cobertura vacinal, apesar dos estudantes demonstrarem conhecimento sobre a doença, o índice de vacinação não foi satisfatório - 48,25% dos graduandos não possuíam 3 doses de vacina. Além disso, a análise multivariada demonstrou que as variáveis: semestre (OR=3,52), estágio clínico (OR=3,87), acidentes por exposição ocupacional (OR=2,26) e respeito ao intervalo entre as doses vacinais (OR=8,58) foram consideradas fatores protetores para a amostra avaliada. A partir deste estudo, compreende-se que os esclarecimentos e campanhas passageiras de estímulo à vacinação não são suficientes para uma modificação eficiente do comportamento. Sugere-se aqui uma participação mais efetiva do corpo docente na formação dos acadêmicos de Odontologia, através do ensino teórico/prático das normas de controle e prevenção de infecção, e, da exigência prática dessas atitudes. / Salvador

Controle de anaplasmose bovina através de imunização com Anaplasma Centrale

A anaplasmose bovina é uma das principais causas de perdas produtivas e mortes no Rio Grande do Sul em rebanhos bovinos. O Anaplasma marginale é o principal agente causador da enfermidade e provoca hipertermia, anemia, prostração, abortos e perdas produtivas nos animais acometidos. Tendo em vista o controle deste hemoparasita em uma propriedade leiteira localizada no município de Eldorado do Sul no Rio Grande do Sul, na qual a incidência da doença era alta, 473 animais foram imunizados com Anaplasma centrale na busca de desenvolvimento cruzado para Anaplasma marginale. No experimento foram verificadas que a incidência que normalmente era acima de 30% na propriedade passou para níveis inferires a 5%. No entanto, foram verificados abortos decorrentes da imunização, principalmente nos animais que possuíam menos de 90 dias de prenhes. Já o número de mortes globais na fazenda caiu consideravelmente tendo em vista que a principal causa de morte era a anaplasmose bovina. Dos animais inoculados com A. centrale em torno de 15% apresentaram sintomatologia clínica da enfermidade e precisaram ser tratados com oxitetraciclina no período entre 15 e 30 dias após a imunização. O custo com tratamento empregado na propriedade posterior à imunização caiu em torno de 85% o que provocou impacto significativo economicamente na propriedade. / Bovine anaplasmosis is a major cause of production losses and deaths in Rio Grande do Sul cattle herds. The Anaplasma is the main causative agent of cattle disease. It causes hyperthermia, anemia, prostration, abortions and reduces milk production in affected animals. In order to control this hemoparasite on a dairy farm located in the municipality of Eldorado do Sul in Rio Grande do Sul, where the disease incidence was high, 473 animals were immunized with Anaplasma centrale in the search for cross-protectiv immunity for Anaplasma marginale. The property anaplasmosis incidence, which usually was above 30%, became 5% after the immunization. However, abortions were observed resulting from innoculaition, especially in animals that had less than 90 days of pregnancy. The global number of deaths on the farm dropped considerably given that the main cause of death was the bovine anaplasmosis. 15% of animals inoculated with A. centrale showed clinical symptoms of the disease between 15 and 30 days after immunization and had to be treated with oxytetracycline . The amount of money spent with anaplasmosis treatment decay 85% after the immunization, which caused significant economic impact on the property.

Influenza is a serious illness worldwide, causing high morbidity and mortality. 10-20% of world population fall ill with influenza each year and 250 000 - 500 000 people die annually. The most efficacious protection to date is vaccination. Current vaccines are efficient only one season because of fast mutation rate of influenza virus. The effort to create an effective vaccine faces lack of potent adjuvant, which can adequately stimulate and modulate immune system to protect organism from virus infection.
