Perceptions of school management teams (SMTs) with regard to health and well-being of farm schools/public schools on private property (PSPP) / by Oremeng Lawrens Mashoko

This study was undertaken in five (5) farm schools in the Maquassi Hills Area Project Office (APO) with the intention to assess whether the School Management Teams (SMT's) are given necessary guidance to deal with health and well-being of schools. The Constitution of South Africa stipulates that human values (dignity, freedom and equality) form the most important challenges that condone and maintain that a safe and discipline environment should be created for effective teaching and learning. Focus group interviews were conducted to gather information which was relevant to the study. The findings show that SMT's of farm schools have problems with the managerial tasks as well as the functional tasks of schools' daily activities. The researcher infers that both the landowners and the DoE must ensure that electricity and water is provided where contractual agreements exist, as power affects the usage of visual aids and administering school businesses. The Department of Education (DoE) has no legal structure within which farm schools should operate to guarantee equal educational opportunities, and create a positive disciplined schools where learners and educators not only know what is expected, but feel secure. The officials who are responsible for the dispatch of learning support materials (LSM) hold the SMT's back in assigning duties to the educators because of lack of LSM. The study shows that the official seem to be ignorant of basic education to farm school learners, and how the DoE manages farm schools, reflects how it still clanged to the red-tape of funding farm schools. Other essentials like water and phones are not given attention to communicate problems that need immediate attention like illness, danger or accident. Learners still have to travel long distances which is a safety thread. The researcher eludes that it is in the interest of the landowners and the DoE that the conditions that prevail on farm schools should not impede with the work of the SMT's. The landowners and the DoE should come to a consensus in making the school environment for SMT's inviting. Increasingly adolescents are making unhealthy choices concerning their lifestyle. Behaviour patterns develop which are often carried into adulthood and become an established lifestyle pattern. Examples of these are the abuse of drugs, smoking, alcohol, irresponsible and hazardous sexual behaviour and so forth. It is also reported that young people increasingly contemplate and attempt suicide. It is important to find the reasons for such behaviours. During the last century, spirituality was not regarded as being important in the everyday lives of people. There are however many voices today calling for a reintegration of spirituality into education, health and various other areas of life. The movement of Health Promoting Schools could serve as a basis for reintegrating spirituality into the curriculum. Spirituality can be described as providing primarily meaning, relationship with God and people, a feeling of transcendence and the acquisition of values. These facets of life, neglected and considered unscientific in the positivistic modem era, are now seen as necessary within our post-modem society. The aim of this study was to investigate a possible correlation between the lifestyle choices adolescents make and their spiritual well-being. Both quantitative and qualitative measures were used to investigate the above mentioned correlation. Quantitative results indicated a significant positive correlation between measures of spiritual well-being and risk behaviours. The qualitative analysis indicated that adolescents believe that spiritual values are important when considering meaning in life as well as lifestyle choices. It was concluded that the construct spirituality remains salient in the lives of most adolescents. Health in the Winds: Wind Band Participation as a Health Promoting Activity for Older Men

Using a basic interpretive qualitative research design, this study explores adult musicians’ reflections of participation in wind band as a health promoting activity. Five male participants between the ages of 58 and 76 participated in semi-structured interviews and provided basic demographic information. Data were analyzed thematically and findings represented evocatively. Elements of a wind ensemble were used as a metaphor to provide a rich way of presenting the data as fundamentally tied to the specific act of making music in a wind ensemble. The three main components of the metaphor and the main themes they present are: Instrumentation: Defining Roles (Purpose), Sound: Making Meaning (Physical and Emotional Well-being), and Performance: Extending Self (Challenge, Accomplishment and Connection). The current findings contribute to furthering knowledge and research in the area of music making and health, especially wind ensemble playing participation and healthy aging. Implications for both practice and future research are identified.

What underpins success in a health promoting school in Northeastern Thailand? : a thesis presented in fulfilment of the requirements for the degree of Doctor of Philosophy in Nursing at Massey University

A Health Promoting School (HPS) approach is now widely accepted internationally, with a focus on children’s health, the school curriculum, and whole school environment. In Thailand, the health and well-being of children is a fundamental value. HPS programmes have been implemented in schools as a strategy to focus on young people’s health. A number of barriers to successful HPS have been identified. While there is international evidence to show the steps and the key factors in creating successful HPS, little is known about successful HPS in the Thai context, in particular, in Northeastern Thailand which has been classified the poorest region. Ethnographic methods were used to examine what understanding of the meaning of HPS is necessary for a successful school, and how all those involved acted from the adoption of the HPS programmes by the local school until it achieved HPS status. A rural school which was successful in a HPS programme was selected, in Mahasarakham province, Northeastern Thailand. The data were obtained through participant observation, ethnographic interviews, and ethnographic records, and data analysis took place simultaneously with data collection. In this study, Lofland’s strategy for the analysis of the structure of human interaction was used. A variety of techniques for improving and documenting the credibility of the study such as prolonged engagement, persistent observation, and triangulation were used. This research revealed that the informants’ views reflected diverse understandings of the meaning of HPS. Those views were based on their experiences of HPS which differed according to the degree of participation, different levels of knowledge about HPS, and in the roles they played in the implementation of HPS in the school. Thai culture and school ethos influenced the success of HPS. Community participation was also crucial in supporting the school’s achievement. Key factors that underpinned success are identified. Implications of the findings for the HPS programme, health professionals, the school and community are discussed.

Analysis of the effects of the plant growth promoting substances GR24 and smoke water on abiotically stressed Nicotiana benthamiana seedlings

Thesis (MSc)--Stellenbosch University, 2012. / ENGLISH ABSTRACT: Almost all processes during the life of a plant are affected by the environment. Changes in phytohormone, metabolite and protein levels follow in response to changes in the environment. Plant growth promoting substances can stimulate changes at these levels to facilitate increased plant growth and yields above what the plant would normally establish. In this study, the effects of two growth promoting substances, smoke water (SW) derived from bubbling smoke from the burning of plant material through water, and a synthetic strigolactone analogue, GR24, on plant growth and architecture, as well as the proteome and metabalome of salt stressed Nicotiana benthamiana seedlings were investigated. Physiological studies were conducted to identify the effects of the growth substances on salt stressed seedlings in a tissue culture system. Under non-stress conditions, SW treatment increased seedling fresh mass, root length and leaf area. Under salt stress conditions (100 mM and 150 mM NaCl), SW increased fresh mass, root length, leaf number and lateral root number significantly. Under non-stress conditions, GR24-treated seedlings showed increased fresh mass, leaf number and area and root length. When GR24-treated seedlings were placed under salt stress, the seedlings showed significant increases in fresh mass, leaf number and lateral root number, but only marginal increases in root length and leaf area. Despite these similarities, slight differences were observed in the metabolomes and proteomes of smoke water and GR24-treated seedlings, both with and without the addition of salt stress. Relatively few of the differentially expressed proteins could be identified with the instruments available. Changes in the metabolome indicated that photoassimilation and photosynthesis could be affected in response to smoke water and GR24 treatment. Our results suggest that smoke water and GR24 both promote growth under salt stress conditions in seedlings and we furthermore conclude that, although there are distinct overlaps between treatments, this is accomplished via slightly different mechanisms. / AFRIKAANSE OPSOMMING: Gedurende ‘n plant se lewe word omtrent alle prosesse deur die omgewing geaffekteer. In hierdie studie word die effek van twee groei stimulante, rook water verkry deur rook van plant materiaal deur water te borrel en ‘n sintetiese strigolaktoon, GR24, ondersoek op ‘n morfologiese, metaboliese en ‘n proteomiese vlak in Nicotiana benthamiana saailinge. ’n Studie is onderneem om die veranderinge as gevolg van die onderskeie groei stimulante te ondersoek in ‘n weefsel kultuur sisteem. Rook water het onder normale groei omstandighede vars en droeë massa, blaar aantal asook wortel en blaar lengte verhoog. Rook water het na sout behandeling (100 en 150 mM NaCl) steeds vars massa, wortel lengte, blaai aantal en laterale wortel aantal beduidend verhoog in vergelyking met die sout stres kontrole. Behandeling met GR24 het ook vars massa, wortel lengte, blaar aantal en grootte verhoog en onder sout stres met GR24 is ‘n beduidende vergroting opgemerk in vars massa, blaar grootte en laterale wortel aantal. Ongeag van die veranderinge in groei is klein verskille opgemerk in die metaboliet en protein studies. Net ‘n paar proteine kon positief geidentifiseer word met die apparaat beskikbaar. Verandering in die metaboloom wys na veranderinge in fotoassimilasie en fotosintese in reaksie tot rook water en GR24. Hierdie resultate lei tot die gevolgtrekking dat rook water en GR24 beide groei verbeter in saailing behandel met sout en ook dat alhoewel daar sekere ooreenkomste is tussen die reaksies as gevolg van die plant groei stimulante, dit wel geskiet deur geringe verskillende meganismes.

Mechanism of APC/C activation and substrate specificity in mitosis

In eukaryotes, cell proliferation and cell cycle transitions are strictly controlled by the anaphase-promoting complex/cyclosome (APC/C). The APC/C is an E3 ubiquitin ligase that regulates chromatid segregation at the metaphase to anaphase transition, exit from mitosis and the establishment and maintenance of G1. The APC/C’s catalytic activity and substrate specificity are controlled by its interactions with two coactivators, Cdc20 and Cdh1. In contrast to Cdh1, APC/C activation by Cdc20 during mitosis requires hyper-phosphorylation of APC/C subunits by cyclin-dependent kinase (Cdk) and polo kinase. The aim of the first part of this thesis was to understand how mitotic phosphorylation regulates APC/C activity. Using cryo-electron microscopy and biochemical analysis, we found that an auto-inhibitory segment of the Apc1 subunit acts as a molecular switch that in apo unphosphorylated APC/C interacts with a coactivator-binding site (C-box binding site), thereby obstructing engagement of Cdc20. Phosphorylation of the auto-inhibitory segment displaces it from the C-box binding site to relieve APC/C auto-inhibition. Efficient phosphorylation of the auto-inhibitory segment requires the recruitment of the kinase Cdk-cyclin-Cks to a hyper-phosphorylated loop of Apc3. In addition to regulation of APC/C activity by phosphorylation, ordered cell progression is ensured by the ability of the APC/C to target substrate degradation in a defined order. At mitosis onset, degradation of securin and cyclin B1 is inhibited by the spindle assembly checkpoint, exerted by the mitotic checkpoint complex (MCC), whereas both cyclin A2 and Nek2A are not subject to MCC inhibition. The aim of the second part of the thesis was to elucidate the mechanism of how the APC/C achieves its substrate specificity. Our biochemical analysis showed that the resistance of cyclin A2 to MCC inhibition is due to its ABBA motif and the Cdk-associated Cks2 subunit. Furthermore, we found that it is the Cdc20 molecule of the MCC that binds to the ABBA motif to allow for cyclin A2 ubiquitination. Strikingly, mutating all three known degrons (KEN box, D box and ABBA motif) of cyclin A did not affect its ubiquitination by APC/CCdc20. Deletion of a potential novel degron found within residues 60-80 of cyclin A2 impaired cyclin A2 ubiquitination.

Cette thèse porte sur la valorisation du patrimoine marbrier des Pyrénées grâce aux technologies mobiles et se propose d’interroger le rôle de l’historien de l’art dans le développement de ces activités sur les territoires. Les marbres des Pyrénées sont abordés comme ressource géologique dont l’exploitation a façonné les paysages de la montagne et composé les architectures de palais royaux ; ces marbres intègrent le patrimoine sous toutes ses formes, du matériel à l’immatériel. Leur valorisation est multiforme, convoque les notions de paysage et de développement durable et interroge celle du terroir. D’un point de vue patrimonial, les technologies mobiles offrent de nouveaux moyens de médiation. Elles ouvrent ainsi de nouvelles perspectives, dont peuvent se saisir les acteurs impliqués dans la valorisation. Grâce au soutien de la société Camineo, spécialiste de médiation multimédia mobile, deux projets ont été déployés en regard l’un de l’autre : dans les Pyrénées et au domaine de Trianon à Versailles. Ils permettent de questionner l’apport de l’histoire de l’art à la production de récits de médiation patrimoniale et, plus généralement, au regard porté sur l’environnement. / This thesis focuses on the enhanced promotion of the the French Pyrenees’ marble heritage through the use of mobile technologies. It aims to question the role of the art historian in the development of these activities on such territories. The quarrying of these Pyrenean marbles, a valued geological resource, dramatically transformed the makeup of mountainous landscapes and shaped the architecture of royal palaces; these marbles are an integral part of our heritage, in both tangible and intangible forms. Their promotion is multifaceted, invoking our perception of the landscape, of sustainable development, and of the concept of terroir itself. From a heritage point of view, mobile technologies offer new channels for cultural mediation. They therefore open up new avenues which can be seized upon by those involved in the promotion of this patrimony. Thanks to the support of Camineo, a company specialising in the interpretation of the marble heritage through mobile technologies, two projects were led in conjunction: one in the Pyrenees and the other on the Domaine du Trianon in Versailles. This has enabled us to question the contribution of history of art in the production of outreach material about this heritage, and more generally, on our perception of the environment.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / The present dissertation has the purpose of investigating how, as an affiliate, a broadcast station builds its own identity, distinguish its production from that one made by the network head and, finally, the manner how configures its image, while reinforcing the mother network s identity and, along with it, realizes its own productions in a coherent way. For this, the present work analyses, based on Greimasian discursive semiotics, 13 pieces of the RBS TV s 50th anniversary self-promotional campaign, seeking to verify solutions found by them to respond the challenge of articulate different interests that, commonly, work in opposite directions. Therefore, the main objective of this investigation is to comprehend how the identity construction and the image and brand projection are made during the period of an assumed self-promotional action, verifying which are the discursive procedures adopted by the aforesaid campaign, through the self-talking production, and which forms of expression actualize this strategies of identity construction in the specific case of an affiliate broadcast station. The research, structured in nine sections, after a theoretical foundation about the promotional field and a brief exhibition of the appropriate discursive semiotics concepts that support the methodology used for the selected self-promotional pieces analysis, turns out to the corpus study, considering, as a point of view, its communicative/enunciative context and inter and extra-textual relations. / A presente dissertação propõe-se a investigar como, no contexto de afiliada, uma emissora constrói a sua própria identidade; como distingue sua produção daquela da cabeça de rede; como, finalmente, configura sua imagem, sem deixar de reforçar a identidade da rede mãe e de com ela compatibilizar de forma coerente suas próprias produções. Para tanto, analisa, com base na semiótica discursiva de inspiração greimasiana, 13 peças da campanha autopromocional comemorativa de 50 anos da RBS TV, com vistas a verificar as soluções por elas encontradas para responder ao desafio de articular diferentes interesses que muitas vezes se contrapõem. Assim, o objetivo central da investigação é verificar como se dá a construção identitária e projeção de imagem e marca no desenrolar de uma ação assumidamente autopromocional, verificando quais são os procedimentos discursivos adotados pela referida campanha na produção desse falar de si mesma, que formas de expressão atualizam essas estratégias de construção identitária, no caso específico de uma emissora afiliada. O trabalho, estruturado em nove secções, após uma fundamentação teórica no campo da promocionalidade, e de uma breve exposição dos conceitos apropriados da semiótica discursiva que sustentam a metodologia adotada na análise das peças autopromocionais selecionadas, dedica-se ao exame do corpus, do ponto de vista de seu contexto comunicativo/enunciativo e de suas relações inter e intratextuais.

In Brazil, this crop has received special attention as an alternative for the supply of vegetable oil as a raw material for biodiesel manufacturing, due to the biotechnological potential of its seeds. Several studies aimed at climatic adaptations and productivity have been developed on genetic variability of J. curcas; but few studies have been developed focusing on the analysis of the microbial community associated with this species. The interior of the plant is inhabited by endophyte microorganisms, which can be isolated from disinfected surface and do not put on risk the plant tissues of the host plant. This group of microorganisms has the ability to stimulate the growth of plants by direct mechanisms, such as/biological fixation of nitrogen, solubilization of inorganic phosphate indole acetic acid production (AIA) and indirect mechanisms, such as the antagonism towards phytopathogens. Moreover, endophytic microorganisms are considered promising for bioprospecting of enzymes. The aim of this study was to perform the characterization of seventy-two endophytic bacteria associated with the J. curcas plants, with potential for promoting plant growth and enzyme production. Of this total of tested isolates 40% showed positive results for fixation of atmospheric nitrogen, 43% solubilized inorganic phosphate, 69% have produced AIA, 36% showed some antagonistic activity to phytopathogenic fungi Alternaria alternata, 12% for Ceratocystis paradoxa, 22% for Fusarium proliferatum and 23% for Fusarium verticillioides. Regarding enzymatic activity, 16% of the isolates evaluated showed amyloid activity, 29% endoglycolytic, 29% lipolytic, 20% esterase, 46% pectinolytic (18% poligalacturonase and 43% pectato lyase) 37% proteolytic. From a collection of seventy two bacterial isolates, twenty-five were identified by means of the partial 16S rDNA gene embracing seven different genres: Bacillus, Citrobacter, Curtobacterium, Enterococcus, Microbacterium, Promicromonosporaceae and Sanguibacter were isolated as endophytic bacteria of Jatropha. / O pinhão-manso (Jatropha curcas L.), pertencente à família das Euforbiáceas é uma planta geneticamente próxima a mamona (Ricinus communis, L.), originária da América Central e atualmente está distribuída em todas as regiões tropicais do globo. No Brasil esta cultura tem recebido especial atenção como uma alternativa para o fornecimento de óleo vegetal como matéria-prima para fabricação do biodiesel, devido ao potencial biotecnológico de suas sementes. Vários estudos foram desenvolvidos sobre variabilidade genética do J. curcas visando adaptações climáticas e produtividade; porém poucos estudos foram dedicados a análise da comunidade microbiana associada a essa espécie vegetal. O interior da planta é habitado por micro-organismos endófitos que podem ser isolados dos tecidos vegetais desinfectados superficialmente e que não causam danos aparentes a planta hospedeira. Este grupo de micro-organismos apresenta a capacidade de estimular o crescimento das plantas por mecanismos diretos, tais como fixação biológica de nitrogênio, solubilização de fosfato inorgânico e produção do ácido indol acético (AIA) e por mecanismos indiretos, como antagonismo a fitopatógenos. Além disso, os micro-organismos endofíticos são considerados promissores para a bioprospecção de enzimas. O presente trabalho teve como objetivo, a caracterização de setenta e duas bactérias endofíticas, associadoas à cultura do pinhão-manso, com potencial para promoção de crescimento vegetal e produção de enzimas. Desse total de isolados testados 40% apresentaram resultados positivos para fixação biológica de nitrogênio atmosférico, 43% solubilizaram fosfato inorgânico, 69% produziram AIA, para o teste de antagonismo 36% dos isolados apresentaram alguma atividade antagônica para os fungos fitopatogênicos Alternaria alternata, 12% Ceratocystis paradoxa, 22% Fusarium proliferatum e (24%) e F. Verticillioides. Com relação atividade enzimática 16% dos isolados avaliados apresentaram atividade amilolítica, 29% celulolítica, 29% lipolítica, 20% esterolítica, 46% pectinolítica (18% poligalacturonase e 43% pectato liase) e 37% proteolítica. De uma coleção de setenta e dois isolados bacterianos, vinte e cinco foram identificados por meio do sequenciamento parcial do gene 16S rDNA, abrangendo sete diferentes gêneros: Bacillus, Citrobacter, Curtobacterium, Enterococcus, Microbacterium, Promicromonosporaceae e Sanguibacter foram isolados como bactérias endofíticas de pinhão-manso.
