Analyse des réponses neuronales du cortex visuel primaire du chat à la fréquence spatiale suite à des adaptations répétées

Les neurones du cortex visuel primaire (aire 17) du chat adulte répondent de manière sélective à différentes propriétés d’une image comme l’orientation, le contraste ou la fréquence spatiale. Cette sélectivité se manifeste par une réponse sous forme de potentiels d’action dans les neurones visuels lors de la présentation d’une barre lumineuse de forme allongée dans les champs récepteurs de ces neurones. La fréquence spatiale (FS) se mesure en cycles par degré (cyc./deg.) et se définit par la quantité de barres lumineuses claires et sombres présentées à une distance précise des yeux. Par ailleurs, jusqu’à récemment, l’organisation corticale chez l’adulte était considérée immuable suite à la période critique post-natale. Or, lors de l'imposition d'un stimulus non préféré, nous avons observé un phénomène d'entrainement sous forme d'un déplacement de la courbe de sélectivité à la suite de l'imposition d'une FS non-préférée différente de la fréquence spatiale optimale du neurone. Une deuxième adaptation à la même FS non-préférée induit une réponse neuronale différente par rapport à la première imposition. Ce phénomène de "gain cortical" avait déjà été observé dans le cortex visuel primaire pour ce qui est de la sélectivité à l'orientation des barres lumineuses, mais non pour la fréquence spatiale. Une telle plasticité à court terme pourrait être le corrélat neuronal d'une modulation de la pondération relative du poids des afférences synaptiques. / Primary visual cortex neurons in adult cat are selective to different image properties as orientation, contrast and spatial frequency. This selectivity is characterized by action potentials as electrical activity from the visual neurons. This response occurs during the presentation of a luminous bar in the receptive fields of the neurons. Spatial frequency is the amount of luminous bars in a grating presented from a precise distance from the eyes and is measured in cycles per degree. Furthermore, it was establish until recently that cortical organisation in the adult remains inflexible following the critical period after birth. However, our results have revealed that spatial frequency selectivity is able to change after an imposition of a non-preferred spatial frequency, also called adapter. Following cortical activity recordings, there is a shift of the spatial frequency tuning curves in the direction of the adapter. A second adaptation at the same non-preferred spatial frequency produced a different neural response from the first adaptation. This “short-term plasticity” was already observed in the primary visual cortex for orientation selective neurons but not yet for spatial frequency. The results presented in this study suggest that such plasticity is possible and that visual neurons regulate their electrical responses through modulation of the weights of their synaptic afferences.

L'approche spatio-polyphonique dans les interprétations des pianistes de la deuxième moitié du XIXe siècle et la première moitié du XXe siècle / Spatio-polyphonic approach in the interprétations of pianists of the second half of the 19th Century and the first half of the 20th Century

Le but de ce travail est d'étudier l'art d'interpréter de plusieurs générations de pianistes dont la formation remonte au XIXe siècle, mais dont l'activité s'est étendue jusqu'à la 1re moitié du XXe siècle. L'essor de l'interprétation qui marqua cette époque a prodigué des artistes qui - à la suite de leurs grands maîtres (Chopin, Liszt, A. Rubinstein) - ont déjà laissé un nombre considérable d'enregistrements permettant de saisir à travers leur jeu une image authentique de l’œuvre romantique. En nous référant à l'idée que l'essor du pianisme à la charnière des XIXe - XXe siècles provient de l'expansion remarquable de l'art d'interpréter au XIXe siècle, nous avançons l'hypothèse selon laquelle c'est précisément dans les positions esthétiques et dans la vision du monde propre à cette époque qu'il faut chercher les sources d'un tel essor. Parmi les caractéristiques importantes de la manière d'interpréter des pianistes de la 2e moitié du XIXe siècle, entrent d'abord en jeu la faculté de penser imagée, la liberté et une manière d'improviser en modifiant les textes des œuvres exécutées. Ces données se combinent d'ailleurs avec d'autres, comme l'intellectualisme. Les analyses de certaines particularités de style, des manières de jouer, typiques du XIXe siècle (comme le rubato, le « pointillisme etc.), nous révèlent ainsi ce que fut la réelle pensée polyphonique du Romantisme. Il ne s'agit pas de la seule écriture polyphonique, mais d'un principe polyphonique au sens plus large, s'exprimant à travers le style d'interpréter qui, à son tour, définit la perception spatio-temporelle spécifique de ces quelques décennies. / The aim of this work is to study the performing art of several generations of pianists who were trained in the 19th Century but who extended their artistic activity throught the 1st half of the 20th Century. The rise of the art of interpretation which marked this period gave the artistes who - following their great masters - have left a considerable number of records where their plaiyng conjures up an image of the romantic composition that may be more authentic, though it is quite different from that of today.Based on the idea that the rise of the pianism at the turn of the 20th Century comes largely from the remarkable expansion of the 19th Century performing arts, we hypothesize that it is precisely in the aesthetic positions and the vision of the world inherent in the Romantic era that we should look for the sources of such a rise. Chief among the important features of the style of interpretation of the pianists of the 2nd half of the 19th Century, was their creative thinking, but also the surprising freedom they enjoyed to improvise and modify compositions. Other features include the intellectualism of their approach to the performed composition. Analyses of certain peculiarities of style, of the ways of playing typical of the 19th Century (such as rubato, the « pointillism » etc.) reveal the real polyphonic thougth of the Romantic era. This is not only the polyphonic writing itself, but a polyphonic principle in a broader sense, manifesting itself through the style of interpretation which, in turn, defines the specific spatio-temporal perception of these several decades.

Conselho Nacional do Desenvolvimento Científico e Tecnológico (CNPq) / É implementado em hardware, por meio da linguagem VHDL, um método de avaliação objetiva de qualidade de vídeo digital. Sendo um processo computacionalmente custoso em software, investiga-se sua implementação em hardware. O método implementado, chamado de NRVQA-LM, utiliza seis características espaço-temporais extraídas de diferentes vídeos para chegar a um escore de qualidade. São estudadas essas características e planejada a sua implementação de forma otimizada, a fim de aproveitar as vantagens de plataformas de lógica reconfigurável, como as FPGAS. Durante o desenvolvimento foi necessário o estudo de ferramentas não usuais da linguagem VHDL, tais como as aritméticas de ponto fixo e flutuante e a escrita de funções matemáticas. Os resultados mostram alta correlação com os valores das características e dos escores de qualidade em relação ao método em software. A implementação se mostrou custosa em termos de recursos lógicos, especialmente devido à necessidade de se armazenar um quadro de vídeo inteiro, mas eficiente graças à característica de paralelismo das FPGAs, executando cálculos entre 20 e 40 vezes mais rapidamente que em uma linguagem de alto nível como o MATLAB. A aritmética ponto fixo mostrou-se vantajosa em relação ao ponto flutuante, principalmente no que tange à frequência de operação. / It is implemented in hardware an objective digital video evaluation method, using the VHDL language. As a computationally expensive process in software, it is investigated its implementation in a hardware platform. The implemented method, named NRVQA-LM, employs six spatio-temporal features extracted from different videos in order to obtain a quality score. These features are studied and the implementation is designed to be developed in an optimized way, in order to explore the benefits of reprogammable logic platforms, such as FPGAs. During the development it was necessary to study non-recurrent tools of the VHDL language, such as fixed- and floating-point arithmetics and the writing of math functions. Results shows high correlation between the calculated scores of the hardware and the original software implementations. The hardware implementation revealed to be highly resource expensive, mainly due the need of storing a whole video frame, but efficient in time, thanks to the parallelism feature of FPGA devices, executing quality score calculations between 20 and 40 times faster than a high-level language such as MATLAB. The fixed-point arithmetics revealed to be more efficient than the floating-point, specially regarding operation frequency.

Diploma thesis Background identity is a series of events through which I am trying to find a harmony between the magic of art and life. I decided to initiate the situation, "performance" mediating an authentic, strong feelings. I tend to performance associated with life in the self-reflective approach. Based in my own performative practice in which the strength and authenticity is reflected primarily in the exchange of energy between the audience and the performer. I started to define an ideal conditions for performance to achieve this exchange, authentic experience and mutual perception of the situation within the relationship between "performer" and "spectator". In November 2015 I planned "authentic performances " series . The mutual forming of "performing" act I have anchored in the concept of identity, which is infinitely formed in relationships and the never-ending negotiation process. In the name of authenticity I am exposing myself within the anonymity of public space and waiting for the moment of identification. Waiting for concrete spectator who is already a part of me within a certain social role. Unexpected intervention into the everyday life of concrete person make the opportunity of authentic experience. Experience which is a part of his everyday life completes the action, giving the context. During the realization I got the opportunity to exhibit in the Palais de Tokyo. Aware of the significance of this opportunity for the development of my own artistic career I have decided to accepted it as a challenge. Although the terms of institution are utter contradiction of those conditions I had defined previously for authentic experience. In the name of authenticity and my principles I decided to be present in the role of spectator. To be invisible to this prestigious gallery. Be invisible to the artistic group Delta Total through which I had the opportunity to exhibit. To be inside and outside in the same time. The starting point of the last part is a necessity with the goal to get a master's degree. The necessity of outcome for jury can also betray my ideals of living authentic event. I should mediate all the experience of past events (to date 20th of April- thirty-three events) which happened in so many different places in the space of "Malá Amerika" in Brno...

Les synapses fibres moussues de l‘hippocampe entre le gyrus denté et les cellulespyramidales de CA3 sont caractérisées par leur morphologie particulière, et par leurspropriétés distinctives de transmission synaptique et de plasticité présynaptique. Cessynapses sont parfois appelées «détonatrices» pour leur rôle fonctionnel dansl‘encodage de la mémoire épisodique. Cependant, les mécanismes moléculaires à labase des propriétés spécifiques de ces synapses restent peu connus. Ce travail estcomposé de deux parties principales:1) Phénotypage des synapses fibres moussues de l'hippocampe chez les sourisVAMP7 KOVAMP7 est une protéine SNARE vésiculaire de la famille des longins, qui joue unrôle dans la croissance des neurites durant le développement. Dans le cerveauadulte, VAMP7 est enrichi dans un sous-ensemble de terminaisons nerveuses, enparticulier dans les fibres moussues de l‗hippocampe. Nous avons analysé lafonction de VAMP7 dans la libération de neurotransmetteurs par une caractérisationextensive de la transmission synaptique et des mécanismes de plasticité de cettesynapse. L'absence de VAMP7 ne cause pas de graves déficits développementauxou neuronaux (Sato et al., 2011; Danglot et al., 2012). Les mécanismesprésynaptiques de la plasticité à court terme de la fibre moussue de l‘hippocampesemblent également normaux, pour des raisons éventuelles qui seront discutées.2) Circuits du CA3 examinés par traçage viral et enregistrements de pairesNous avons développé une technique pour établir des enregistrements en pairesentre cellules en grain du gyrus denté connectées et cellules pyramidales CA3 (GCCA3),sur des cultures organotypiques de tranches d'hippocampe de souris. Pouridentifier les partenaires présynaptiques directs à une cellule pyramidale CA3 ciblée,nous avons combiné l‘électroporation cellulaire unitaire et le traçage mono-transsynaptiquebasé sur un virus de la rage recombinant et pseudotypé. Nous avonstransfecté une cellule pyramidale CA3 unique par tranche avec les plasmides codantla glycoprotéine d‘enveloppe du virus de la rage (RG), un rapporteur fluorescent, etla protéine TVA (récepteur de surface apparenté au EnvA, qui n'a pas d‘homologuechez les cellules de mammifères). Les tranches ont ensuite été infectées avec levirus de la rage recombinant et pseudotypé. Après 3-4 jours, le traçage mono-transsynaptiquerévèle les entrées présynaptiques de ce neurone unique. Ensuite, nousavons pu établir des enregistrements de paires entre les cellules en grain-CA3connectés, ainsi que de quantifier les partenaires présynaptiques de la cellulepyramidale CA3 de départ. / The hippocampal mossy fiber is characterized by its particular morphology, distinctsynaptic transmission and presynaptic plasticity. Moreover, this synapse has beencalled ―teacher‖ or ―detonator‖ for its proposed functional role in episodic memoryencoding. Nevertheless, the molecular mechanisms underlying its specific functionalproperties remain elusive. This work is composed of two main parts:1) Phenotyping Hippocampal Mossy Fiber Synapses in VAMP7 KO MiceVAMP7 is a vesicle SNARE of the longin family important in neurite growth duringdevelopment. In the adult brain, VAMP7 is enriched in a subset of nerve terminals,particularly at the hippocampal mossy fiber. We analyzed VAMP7 function inneurotransmitter release by characterizing basal and evoked transmission at thissynapse in KO mice and fully tested hypotheses relevant to short-term plasticity.Loss of VAMP7 has been previously reported not to cause major developmental orneurological deficits (Sato et al., 2011; Danglot et al., 2012). Presynapticmechanisms of short-term plasticity at the hippocampal mossy fiber also seemunaffected for potential reasons that will be discussed.2) CA3 Circuits Probed with RABV-Tracing and Paired RecordingsWe developed a technique to establish paired recordings between connected dentategyrus granule cells and CA3 pyramidal cells (GC-CA3) in mouse hippocampalorganotypic slice cultures. To identify direct presynaptic partners to a defined targetCA3 pyramidal cell, we combined single-cell electroporation (SCE) and mono-transsynaptictracing based on a pseudotyped, recombinant rabies virus (EnvApseudotyped RABV ΔG). Using SCE we transfected a single CA3 pyramidal cell perslice with the plasmids encoding: the RABV envelope glycoprotein (RG), afluorescent reporter, and TVA (the EnvA cognate surface receptor, which has nohomologue in mammalian cells). The slices were subsequently infected with EnvApseudotyped RABV ΔG. After 3-4 days, the RABV mono-trans-synaptic tracingrevealed the presynaptic inputs of that single neuron. Then, we were able toestablish paired recordings between connected GC-CA3 cells, as well as to quantifythe presynaptic partners of the starter CA3 pyramidal cell. / De mosvezel van de hippocampus kenmerkt zich door een bijzondere morfologie,uitzonderlijke synaptische transmissie en presynaptische plasticiteit. De synapswordt ook wel "leraar" of "detonator" genoemd vanwege zijn waarschijnlijke rol in decodering van het episodisch geheugen. Met behulp van SCE transfecteerde we één CA3 piramidale cel per slicemet plasmiden die coderen voor: het RABV glycoproteïne-envelop (RG), eenfluorescerende reporter, en TVA (de aan EnvA verwante oppervlakte receptor diegeen homoloog in zoogdiercellen heeft). De slices werden vervolgens geïnfecteerdmet ENVA pseudogetyped RABV ΔG. Na 3-4 dagen bracht de RABV mono-transsynaptischetracing de presynaptische ingangen van die ene neuron aan het licht.Hierna konden we gekoppelde opnames doen tussen verbonden KC-CA3 cellen.Daarnaast konden we de presynaptische partners van de starter CA3 pyramidale celkwantificeren.

Synthèse de textures dynamiques pour l'étude de la vision en psychophysique et électrophysiologie / Dynamic Textures Synthesis for Probing Vision in Psychophysics and Electrophysiology

Le but de cette thèse est de proposer une modélisation mathématique des stimulations visuelles afin d'analyser finement des données expérimentales en psychophysique et en électrophysiologie. Plus précis\'ement, afin de pouvoir exploiter des techniques d'analyse de données issues des statistiques Bayésiennes et de l'apprentissage automatique, il est nécessaire de développer un ensemble de stimulations qui doivent être dynamiques, stochastiques et d'une complexité paramétrée. Il s'agit d'un problème important afin de comprendre la capacité du système visuel à intégrer et discriminer différents stimuli. En particulier, les mesures effectuées à de multiples échelles (neurone, population de neurones, cognition) nous permette d'étudier les sensibilités particulières des neurones, leur organisation fonctionnelle et leur impact sur la prise de décision. Dans ce but, nous proposons un ensemble de contributions théoriques, numériques et expérimentales, organisées autour de trois axes principaux : (1) un modèle de synthèse de textures dynamiques Gaussiennes spécialement paramétrée pour l'étude de la vision; (2) un modèle d'observateur Bayésien rendant compte du biais positif induit par fréquence spatiale sur la perception de la vitesse; (3) l'utilisation de méthodes d'apprentissage automatique pour l'analyse de données obtenues en imagerie optique par colorant potentiométrique et au cours d'enregistrements extra-cellulaires. Ce travail, au carrefour des neurosciences, de la psychophysique et des mathématiques, est le fruit de plusieurs collaborations interdisciplinaires. / The goal of this thesis is to propose a mathematical model of visual stimulations in order to finely analyze experimental data in psychophysics and electrophysiology. More precisely, it is necessary to develop a set of dynamic, stochastic and parametric stimulations in order to exploit data analysis techniques from Bayesian statistics and machine learning. This problem is important to understand the visual system capacity to integrate and discriminate between stimuli. In particular, the measures performed at different scales (neurons, neural population, cognition) allow to study the particular sensitivities of neurons, their functional organization and their impact on decision making. To this purpose, we propose a set of theoretical, numerical and experimental contributions organized around three principal axes: (1) a Gaussian dynamic texture synthesis model specially crafted to probe vision; (2) a Bayesian observer model that accounts for the positive effect of spatial frequency over speed perception; (3) the use of machine learning techniques to analyze voltage sensitive dye optical imaging and extracellular data. This work, at the crossroads of neurosciences, psychophysics and mathematics is the fruit of several interdisciplinary collaborations.

Sonar Sea : The acoustic experience of the Baltic Sea dynamics

This thesis project aims to discuss the conditions and importance of water as a dynamic body in our environment, as water is affecting life on Earth on all levels. By focusing on the Baltic Sea, a sensitive body of water, I am exploring the acoustic characters of the sea dynamics through sound recordings at three bays in the Stockholm Archipelago. How can an acoustic exploration of the Baltic Sea dynamics mediate a sensitive relationship to our marine environments? Sound defines environments and gives indications of their current state. In this project, I intertwine an artistic approach involving our senses with scientific research of measuring to further an understanding of the relationship between humans and nature. The project explores water and sound in two parts, first as a method for listening to the sea, second as a spatial composition created for a sensory experience of the sea’s endless motion. It is my hope that my installation can stir emotions and create an understanding for the environmental challenges facing the Baltic Sea and inspire action towards prosperous natural environments where we live with and not apart from nature.
