Developing a Risk Assessment Model for non-Technical Risk in Energy Sector

Yes / Risk Management is one of the most relevant approaches and systematic applications of strategies, procedures and practices management that have been introduced in literatures for identifying and analysing risks which exist through the whole life of a product ,a process or services. Therefore, the aim of this paper is to propose a risk assessment model that will be implemented to the energy sector, particularly to power plants. This model combines the Analytic Hierarchy Process (AHP) technique with a new enhanced Balance Score Card (BSC). AHP is constructed to determine the weights and the priorities for all perspectives and risk indicators that involved in the BSC. The novelty in this paper is not only in using the BSC for risk assessment, but also, in developing a new BSC with six perspectives, which are sustainability perspective; economic; learning and growth; internal and operational business process; supply chain and customer/demand perspective. Another three contributions of this paper are firstly, including the sustainability dimension in BSC, and covering nine risk categories, which comprise 84 risk indicators that have been distributed across the six risk BSC perspectives. Secondly, assessing the non-technical risks in power plants and finally, this research will concentrate on the strategic level instead of the operational level where the majority of researches focus on latter but the former is far less researched. The created model will provide an effective measurement for the risks particularly, in the power plants sector. The results of this study demonstrate that the supply chain risks perspective is the keystone for the decision making process. Furthermore, these risk indicators with the new structure of BSC with six perspectives, help in achieving the organisation mission and vision in addition to affording a robust risk assessment model. The inputs of this model are composed from a previous stage using a modified Failure Mode and Effect Analysis (FMEA) (which has been used the Exponential Weighted Geometric Mean (EWGM)) to understand and analyse all risks, after which, the results of the developed FMEA which are the Risk Priority Numbers (RPN’s), have been used to build the AHP-BSC risk model. Dans cette thèse, nous proposons une méthodologie de conception d’architectures de communication dédiées aux réseaux de capteurs basées sur la technique de radio impulsionnelle pour les transmissions ultralarge bande (ULB). La technique impulsionnelle proposée ici repose sur la modulation de forme d’impulsion. L’approche de conception architecturale présentée dans cette thèse se focalise plus particulièrement sur la forme des impulsions et leur génération, qui revêt un intérêt majeur puisqu’elle constitue le support de l’information échangée. L’étude sur le choix de la forme d’impulsion nous a conduit à proposer deux architectures différentes. Une première architecture repose sur les polynômes orthogonaux, et plus particulièrement sur les polynômes d’Hermite, pour la génération des impulsions, et sur une architecture de corrélation pour la détection et la reconnaissance des trains d’impulsions transmis. La deuxième architecture est basée sur la transformée en paquets d’ondelettes discrète et peut être exploitée selon deux modes d’utilisation différents, mono et multiutilisateurs. L’utilisation d’une architecture de synthèse à l’émission et d’analyse à la réception ouvre une nouvelle orientation pour les communications numériques, permettant à la transformée en ondelettes d’assurer à la fois la génération des impulsions à l’émission et leur reconnaissance à la réception. Un intérêt immédiat de la technique proposée permet notamment de faciliter l’accès multiutilisateurs au canal ultralarge bande, et d’autoriser des communications simultanées (Many-to-one, des nœuds vers le puits) ou du broadcast (One-to-many, du puits vers les nœuds) sans surcharger la couche MAC. L’architecture proposée s’inscrit donc à l’interface des couches PHY et MAC et permet de relâcher les contraintes de conception spécifiques à ces couches / In this thesis, we propose a design methodology for communication architectures dedicated to wireless sensor network based on impulse radio techniques for UWB communications. The impulse technique proposed in this work relies on pulse shape modulation. The architecture design approach proposed in this thesis focuses on pulses shape and their generation, which is of major interest as it constitutes the carrier of the information exchanged. The study on the choice of pulse shape led us to propose two different architectures. The first one is based on orthogonal polynomials, more especially on the Hermite polynomials, for impulse generation, and on a correlation architecture for detection and recognition of transmitted impulses. The second architecture is based on discrete wavelet packet transform and can be used according two different modes, mono and multi-users. The use of both synthesis and analysis architectures for emitter and receiver, respectively, offers a new way for digital communications and allows the wavelet transform to ensure the impulses generation on the transmitter and their recognition on the receiver. A major interest point of the proposed technique is to facilitate the multi-users access to the ultra-wideband channel and to allow simultaneous communications (many-to-one, from the sensors to the sink) or broadcast (one-to-many, from the coordinator to the nodes) but without overloading the MAC layer. Hence, the proposed architecture is part of the interface between both PHY and MAC layers, and allows to release their specific design constraints

Essa pesquisa relata os acontecimentos em Oaxaca durante o ano de 2006, quando uma onda de protestos populares levou à tomada da capital do Estado pelos manifestantes, à dissolução do poder político e à violenta ação de desocupação imposta pelo Governo Federal do México. Segundo relatos, o papel que os professores da rede pública de Oaxaca, por meio da Seção XXII do Sindicato Nacional dos Trabalhadores da Educação do México (SNTE,) desempenharam antes e durante a revolta popular de 2006 foi fundamental para nascer a Comuna de Oaxaca. A partir de uma análise crítica dos acontecimentos e das atividades políticas conduzidas pelos professores de Oaxaca nas duas últimas décadas, período de forte efervescência política no Estado e de destacada atuação sindical docente, pretende-se localizar e descrever quais as práticas pedagógicas propostas pelos docentes da Seção XXII. Seu objetivo é compreender, a partir desse levante popular, as concepções de Educação Alternativa e Autônoma que nasceram no Estado de Oaxaca e a elaboração coletiva de uma proposta de gestão comunitária das políticas educativas, caracterizadas pelo envolvimento dos trabalhadores da Educação na construção dessas políticas. Para tanto, o estudo abordará também as concepções políticas de movimentos docentes e de movimentos indígenas no México. A análise destas práticas docentes passará pela pesquisa e descrição do contexto sociopolítico local, pelo estudo da relação que os docentes da Seção XXII estabeleceram com as comunidades indígenas, rurais e setores populares urbanos da sociedade oaxaquenha, assim como a investigação da atuação política e sindical desses professores, sempre a partir de relatos de suas práticas, experiências e propostas educativas. / This research reports the events that happened in Oaxaca during the year 2006, when a wave of popular protests led to the protesters to assume the power of the State capital, the dissolution of political power and the violent eviction action imposed by the Federal Government of Mexico. According to the records, the teachers of Oaxaca public system, through the XXII Section of the Mexican National Syndicate of Education Workers, played a fundamental role in the creation of Oaxacas Comuna before and during the 2006 popular revolution. Through a critical evaluation of the events and the political activities conducted by Oaxacas teachers in the last two decades, that represent a period of strong political unrest and memorable actuation of the teachers trade union, it is intended to describe which are the pedagogical practices proposed by the teachers of the section XXII. Its aim is to comprehend, through this popular uprising, the conceptions of the Autonomous and Alternative education that emerged in Oaxaca State, and also the collective elaboration of a community management proposal of educational policies, characterized by the involvement of the education workers in the construction of these policies. Moreover, this study will address the political conceptions of teachers and indigenous movements in Mexico. The analysis of those teaching practices will be made through the research and description of the local socio-economic context, by the study of the relation established by the professors of Section XXII with indigenous and rural communities and with Oaxacan popular urban sectors. In addiction, the teachers political and syndical actuation, always from their practical narrative, experiences and education proposals will be investigated.

Feedback Enhancement of Antibody Responses via Complement and Fc Receptors

<p>IgG, IgM and IgE in complex with antigen have the capacity to regulate specific immune responses. In this investigation, the role of Fc receptors for IgG (FcγRI, FcγRII and FcγRIII) and complement receptors 1 and 2 (CR1/2) for antibody-mediated enhancement of antibody responses are investigated.</p><p>IgM is known to efficiently activate complement and thereby enhance specific antibody responses but it is not known if this involves binding to CR1/2. Using CR1/2 deficient mice, immunized with sheep erythrocytes alone or together with specific IgM, we present evidence that IgM-mediated enhancement is completely dependent on CR1/2 expression, whereas IgG or IgE in complex with bovine serum albumin (BSA) induce strong antibody responses in CR1/2-deficient mice. Enhancement by IgE is mediated via the low affinity receptor for IgE (FcεRII, CD23). However, the receptors which are involved in IgG-mediated enhancement are not known. We find that γ-chain-deficient mice (lacking FcγRI and FcγRIII) have impaired antibody responses to IgG/BSA complexes. In contrast, FcγRIII deficient mice have normal responses, suggesting that FcγRI mediates the effect. Furthermore, IgG/BSA complexes induce up to 189-fold stronger antibody responses in FcγRIIB-deficient mice than in wild-type mice. The threshold dose of IgG/BSA required was lower, the response was sustained for longer and initiated earlier in FcγRIIB-deficient than in wild-type animals. The findings suggest that FcγRIIB acts as a "safety-valve" preventing excessive antibody production during an immune response. We show for the first time that IgG3/BSA complexes can mediate enhancement of specific antibody responses. Their effect does not involve known Fcγ receptors.</p>

On the Development of Mucin-based Biomaterial Coatings

Owing to their key role in mucosal functioning as surface barriers with biospecific interaction potentials, the mucins are interesting candidates for use as surface modifiers in biomaterials applications. In this work, “mild” fractionation procedures were used to prepare mucins of bovine (BSM), porcine (PGM), and human (MG1) origin. Biophysicochemical analysis showed the prepared mucins to differ in size, charge, conformation, and composition. In turn, these factors were shown to govern mucin adsorption on hydrophilic and hydrophobic model surfaces. To enable for detailed coating analysis, methods for the qualitative and quantitative analysis of mucin-based coatings were developed. Of particular interest, a method for the determination of the fraction of surface-exposed, presumed bioactive proteins in a complex mucin coating was described. It was shown, using microscopy and activation assays, that mucin precoating effectively suppresses the neutrophil response towards a polymeric model biomaterial. Under optimal coating conditions, all mucins performed equally well, thus indicating them to be functionally similar. Coating analysis suggested that efficient mucin surface-shielding is critical for good mucin coating performance. Following a study on the complexation of albumin with preadsorbed mucin, we investigated the effect of mucin precoating on the conformation and neutrophil-activating properties of adsorbed host proteins. We found that mucin precoating greatly reduces the strong immune-response normally caused by adsorbed proinflammatory proteins (IgG and sIgA). Detailed coating analysis revealed that the fraction of surface-exposed protein in the mucin-protein composite influences the neutrophil response. Unexpectedly low neutrophil activation for composites containing near-monolayer concentrations of exposed IgG, suggested IgG to act synergistically with mucin on the surface. Conformational analysis supported this by showing that a preadsorbed mucin layer could stabilize adsorbed IgG through complexation. Our findings link well to the complex in vivo situation and suggest that functional mucosal mimics can be created in situ for improved biomaterials performance.

Bloqueadores do sistema renina-angiotensina em ilhotas pancreáticas e fígado na obesidade induzida por dieta em camundongos / Renin-angiotensin system blockers in pancreatic islets and liver on diet-induced obesity in mice

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / As associações entre obesidade, doença hepática gordurosa não alcoólica (NAFLD) e diabetes mellitus tipo 2 (DM2) são bem estabelecidas, e o sistema renina-angiotensina (SRA) pode proporcionar uma ligação entre eles. O bloqueio do SRA em diferentes níveis pode estar relacionado a respostas na resistência à insulina, remodelagem do pâncreas e do fígado em um modelo de obesidade induzida por dieta. Camundongos C57BL/6 foram alimentados com uma dieta hiperlipídica (HF) durante oito semanas e depois tratados com alisquireno (50 mg/kg/dia), enalapril (30 mg/kg/dia) ou losartana (10 mg/kg/dia) por um período adicional de seis semanas. As drogas foram incorporadas na dieta. Avaliou-se a massa corporal (MC), pressão arterial, consumo e gasto energético (GE), metabolismo da glicose e lipídico, histopatologia pancreática e hepática, análise hormonal, imunohistoquímica, perfil gênico e/ou proteico do SRA no pâncreas, gliconeogênese hepática, sinalização da insulina, oxidação e acúmulo lipídico. Todos os inibidores do SRA reduziram significativamente o aumento da pressão arterial nos camundongos alimentados com dieta HF. O tratamento com enalapril, mas não alisquireno ou losartana, reduziu o ganho de MC e a ingestão alimentar; aumentou o GE; amenizou a intolerância à glicose e resistência à insulina; melhorou a massa de células alfa e beta; impediu a redução da adiponectina plasmática e restaurou a sensibilidade à leptina. Além disso, o tratamento com enalapril melhorou a expressão proteica nas ilhotas pancreáticas de Pdx1, GLUT2, ECA2 e do receptor Mas. O tratamento com losartana apresentou uma elevação na expressão proteica de AT2R no pâncreas. No fígado, a administração de enalapril atenuou a esteatose hepática, o acúmulo de triglicerídeos e preveniu o aumento dos níveis de PEPCK, G6Pase e do GLUT2. Do mesmo modo, o enalapril melhorou a transdução dos sinais da insulina através da via IRS-1/Akt, bem como reduziu os níveis de expressão gênica e/ou proteica de PPAR-gama, SREBP-1c e FAS. Esses resultados sugerem que a inibição da ECA com enalapril atenuou muitos efeitos deletérios provocados pelo consumo da dieta HF, incluindo: normalização da morfologia e função das ilhotas pancreáticas, proteção contra a resistência à insulina e acúmulo de lipídios no fígado. Estes efeitos protetores do enalapril podem ser atribuídos, principalmente, à redução no ganho de MC e ingestão alimentar, aumento do GE, ativação do eixo ECA2/Ang(1-7)/receptor Mas e dos níveis de adiponectina, o que promove uma melhora na ação hepática da insulina e leptina, normalização da gliconeogênese, amenizando a NAFLD. / The associations between obesity, NAFLD (non-alcoholic fatty liver disease) and diabetes are well established, and the reninangiotensin system (RAS) may provide a link among them. . The blocking of the RAS at different levels may be related to responses on insulin resistance, remodeling of the pancreas and liver in a model of diet-induced obesity. Mice (C57BL/6) were fed on a high-fat (HF) diet for 8 weeks and then treated with aliskiren (50 mg/kg/day), enalapril (30 mg/kg/day) or losartan (10 mg/kg/day) for an additional 6 weeks. The drugs were incorporated into the diet. We assessed body mass (BM), blood pressure, energy intake and expenditure (EE), glucose and lipid metabolism, pancreatic and hepatic histopathology, hormonal analysis, immunohistochemistry, the expression profile of genes and/or proteins affecting pancreas RAS, hepatic gluconeogenesis, insulin signaling and lipid oxidation and accumulation. All RAS inhibitors significantly attenuated the increased blood pressure in mice fed a HF diet. Treatment with enalapril, but not aliskiren or losartan, significantly attenuated BM gain, increased EE, enhanced the glucose intolerance and insulin resistance; improved the alpha and beta cell mass; prevented the reduction of plasma adiponectin and restored leptin sensibility. Furthermore, enalapril treatment improved the protein expression of the pancreatic islet Pdx1, GLUT2, ACE2 and Mas receptors. Losartan treatment showed the greatest AT2R expression in the pancreas. In the liver, the enalapril administration improved hepatic steatosis, triglycerides and prevented the increase hepatic protein levels of PEPCK, G6Pase and GLUT2. Additionally, enalapril improved the deleterious effects on the HF diet by upregulating the signal transduction through the IRS-1/Akt pathway, as well as downregulatin the protein levels and mRNA expression of PPAR-gamma, SREBP-1c and FAS. Our findings indicate that ACE inhibition with enalapril attenuated several of the deleterious effects of the HF diet. In summary, enalapril appears to be responsible for the normalization of islet morphology and function, a protective role against hepatic insulin resistance and lipid accumulation in the liver. These protective effects of enalapril were attributed, primarily, to the reduction in body mass gain and food intake, increasing EE, the enhancement of the ACE2/Ang (1-7)/Mas receptor axis and adiponectin levels, enhancing hepatic insulin action, leptin and gluconeogenesis, and attenuating NAFLD.

Innovative solutions leading to a more modern and faster rite are pointed. It shows the historical evolution of the Jury Court, from its earliest origins addresses up until the current development framework in various foreign jurisdictions who, in one way or another, have produced some influence on brazilian law. It concentrates on the evolution within the legal system approachin the country, taking care to highlight the headquarters of the Constitutional Court Jury in Brazil, its status as a fundamental right and constitutional principles that inform it more directly. It also highlights the procedure in stages that currently make up the rite of the Jury and presents its organization in the twenty-six state capitals of brazilian states and the Federal District, and also present the organization of the judgement court scope of the Federal Court. Innovative ideas that aim to give more speed and efficiency to the rite of the Jury in Brazil are shows, most notably the reduction of current procedural phases into one, which, as will be shown, lead to savings time and costs and become more effectiveness procedure. Besides the reduction of procedural stages and exposure of justifies reasons, several other measures are identified and claimed as necessary for procedural advancement of millennial institution that wishes to put Brazil at the top of the modernity of the Jury Court, by presenting an appropriate rite principles enshrined in its Constitution, catching up with it, a procedure with the profile of the Constitutional Democratic State.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / As associações entre obesidade, doença hepática gordurosa não alcoólica (NAFLD) e diabetes mellitus tipo 2 (DM2) são bem estabelecidas, e o sistema renina-angiotensina (SRA) pode proporcionar uma ligação entre eles. O bloqueio do SRA em diferentes níveis pode estar relacionado a respostas na resistência à insulina, remodelagem do pâncreas e do fígado em um modelo de obesidade induzida por dieta. Camundongos C57BL/6 foram alimentados com uma dieta hiperlipídica (HF) durante oito semanas e depois tratados com alisquireno (50 mg/kg/dia), enalapril (30 mg/kg/dia) ou losartana (10 mg/kg/dia) por um período adicional de seis semanas. As drogas foram incorporadas na dieta. Avaliou-se a massa corporal (MC), pressão arterial, consumo e gasto energético (GE), metabolismo da glicose e lipídico, histopatologia pancreática e hepática, análise hormonal, imunohistoquímica, perfil gênico e/ou proteico do SRA no pâncreas, gliconeogênese hepática, sinalização da insulina, oxidação e acúmulo lipídico. Todos os inibidores do SRA reduziram significativamente o aumento da pressão arterial nos camundongos alimentados com dieta HF. O tratamento com enalapril, mas não alisquireno ou losartana, reduziu o ganho de MC e a ingestão alimentar; aumentou o GE; amenizou a intolerância à glicose e resistência à insulina; melhorou a massa de células alfa e beta; impediu a redução da adiponectina plasmática e restaurou a sensibilidade à leptina. Além disso, o tratamento com enalapril melhorou a expressão proteica nas ilhotas pancreáticas de Pdx1, GLUT2, ECA2 e do receptor Mas. O tratamento com losartana apresentou uma elevação na expressão proteica de AT2R no pâncreas. No fígado, a administração de enalapril atenuou a esteatose hepática, o acúmulo de triglicerídeos e preveniu o aumento dos níveis de PEPCK, G6Pase e do GLUT2. Do mesmo modo, o enalapril melhorou a transdução dos sinais da insulina através da via IRS-1/Akt, bem como reduziu os níveis de expressão gênica e/ou proteica de PPAR-gama, SREBP-1c e FAS. Esses resultados sugerem que a inibição da ECA com enalapril atenuou muitos efeitos deletérios provocados pelo consumo da dieta HF, incluindo: normalização da morfologia e função das ilhotas pancreáticas, proteção contra a resistência à insulina e acúmulo de lipídios no fígado. Estes efeitos protetores do enalapril podem ser atribuídos, principalmente, à redução no ganho de MC e ingestão alimentar, aumento do GE, ativação do eixo ECA2/Ang(1-7)/receptor Mas e dos níveis de adiponectina, o que promove uma melhora na ação hepática da insulina e leptina, normalização da gliconeogênese, amenizando a NAFLD. / The associations between obesity, NAFLD (non-alcoholic fatty liver disease) and diabetes are well established, and the reninangiotensin system (RAS) may provide a link among them. . The blocking of the RAS at different levels may be related to responses on insulin resistance, remodeling of the pancreas and liver in a model of diet-induced obesity. Mice (C57BL/6) were fed on a high-fat (HF) diet for 8 weeks and then treated with aliskiren (50 mg/kg/day), enalapril (30 mg/kg/day) or losartan (10 mg/kg/day) for an additional 6 weeks. The drugs were incorporated into the diet. We assessed body mass (BM), blood pressure, energy intake and expenditure (EE), glucose and lipid metabolism, pancreatic and hepatic histopathology, hormonal analysis, immunohistochemistry, the expression profile of genes and/or proteins affecting pancreas RAS, hepatic gluconeogenesis, insulin signaling and lipid oxidation and accumulation. All RAS inhibitors significantly attenuated the increased blood pressure in mice fed a HF diet. Treatment with enalapril, but not aliskiren or losartan, significantly attenuated BM gain, increased EE, enhanced the glucose intolerance and insulin resistance; improved the alpha and beta cell mass; prevented the reduction of plasma adiponectin and restored leptin sensibility. Furthermore, enalapril treatment improved the protein expression of the pancreatic islet Pdx1, GLUT2, ACE2 and Mas receptors. Losartan treatment showed the greatest AT2R expression in the pancreas. In the liver, the enalapril administration improved hepatic steatosis, triglycerides and prevented the increase hepatic protein levels of PEPCK, G6Pase and GLUT2. Additionally, enalapril improved the deleterious effects on the HF diet by upregulating the signal transduction through the IRS-1/Akt pathway, as well as downregulatin the protein levels and mRNA expression of PPAR-gamma, SREBP-1c and FAS. Our findings indicate that ACE inhibition with enalapril attenuated several of the deleterious effects of the HF diet. In summary, enalapril appears to be responsible for the normalization of islet morphology and function, a protective role against hepatic insulin resistance and lipid accumulation in the liver. These protective effects of enalapril were attributed, primarily, to the reduction in body mass gain and food intake, increasing EE, the enhancement of the ACE2/Ang (1-7)/Mas receptor axis and adiponectin levels, enhancing hepatic insulin action, leptin and gluconeogenesis, and attenuating NAFLD.
