Vibrational Sum Frequency Spectroscopy Studies at the Air-Liquid Interface

<p>In this thesis the structure and hydration of small organic and amphipilic compounds adsorbed at the air-liquid interface, have been studied using the nonlinear optical technique Vibrational Sum Frequency Spectroscopy (VSFS). The second order nature of the sum frequency process makes this technique particularly surface sensitive and very suitable for interfacial studies, as molecules at the surface can be distinguished even in the presence of a vast excess of the same molecules in the bulk. Particular emphasis was given to the surface water structure and how it is affected by the presence of small model compounds such as acetic acid and formic acid, and also non-ionic surfactants with sugar based and ethylene oxide based polar headgroups. Understanding the structure of water at these interfaces is of considerable fundamental importance, and here VSFS provided unique information. Upon addition of tiny amounts of these surface active compounds, the ordered surface structure of water was found to be significantly perturbed, as revealed by the changes observed in the characteristic spectroscopic signature of the dangling OH bond of water molecules, which vibrate free in air and are present in the top monolayer. Dramatic differences between the different compounds were also observed in the bonded OH region, providing a valuable insight into the hydration of polar groups at interfaces. Additionally, by employing different polarization combinations of the laser beams involved in the sum frequency process, information about the different water species present at the surface and their average orientation were extracted. In particular an unusual state of water was found with a preferred orientation in a non-donor configuration in close proximity to the hydrophobic region formed by the hydrocarbon tails of the surfactant molecules.</p><p>The conformation and orientation of the different adsorbates were also characterized, targeting their specific vibrational frequencies. Noteworthy is the orientation of the fluorocarbon chain of ammonium perfluorononanoate (APFN), which in contrast to the hydrocarbon chains of the other surfactant molecules studied, remained constant over a wide range of surface densities. This behaviour was also observed for the anionic headgroup of sodium dodecyl sulphate (SDS). Other interesting findings were the formation of a cyclic dimer bilayer at the surface of concentrated aqueous solutions of acetic acid and the water structuring effect induced by poly(ethylene-oxide) headgroups, in spite of being themselves disordered at the air-liquid interface.</p>

Modelagem cinética da esterificação de sec-butanol com ácido acético e estudo de monitoramento em linha da reação com espectroscopia de infravermelho. / Kinetic modeling of esterification of sec-butanol with acetic acid and study of reaction on-line monitoring using near infrared spectroscopy.

A esterificação do sec-butanol com ácido acético, submetida à catálise homogênea, apresenta poucos resultados experimentais de cinética reacional e equilíbrio químico disponíveis na literatura. A presente dissertação visa determinar os parâmetros cinéticos e de equilíbrio químico desta reação de esterificação, empregando modelo cinético de 2ª ordem baseado em atividades. Os experimentos cinéticos foram realizados em reator batelada com auxílio da metodologia Karl Fischer para análise do teor de água e determinação do perfil de frações molares ao longo da reação. A principal dificuldade na modelagem desta esterificação consistiu em representar satisfatoriamente as não-idealidades da mistura em fase líquida, em espacial nas reações cuja fração molar inicial de ácido acético foi superior a 50%. Netas condições admitiu-se a ocorrência de dissociação do ácido acético levando ao surgimento de interações iônicas. O modelo empregado para obtenção das atividades, com base em frações molares, foi o NRTL. Este modelo representou bem as interações moleculares em condição de baixa acidez, mas apresentou desvios em condições de acidez e teores de água mais elevados. O estudo também visou o desenvolvimento de modelos de calibração para um método analítico on-line visando determinar o perfil de frações molares ao longo da reação. O método empregado foi a espectroscopia de infravermelho próximo (NIR), em vista à capacidade deste método analítico em determinar quantitativamente frações molares de água, além dos demais compostos envolvidos. Outro fator determinante ao emprego do NIR é a seu crescente uso no meio industrial, e o presente trabalho visou também contribuir para o uso futuro desta técnica de monitoramento no processo industrial. Modelos de calibração multivariável com conjuntos de calibração interna por validação cruzada se mostraram adequados ao sistema estudado. Calibrações externas resultaram em modelos imprecisos, não sendo possível determinar claramente os fatores responsáveis pela imprecisão. / There is a lack of literature data on the reaction kinetics and equilibrium of the homogeneously catalyzed esterification of sec-butanol with acetic acid. This work aims at obtaining the kinetic parameters and chemical equilibrium parameters of this esterification reaction by using a second order kinetic model based on activities. The kinetic experiments were performed in a batch reactor and the Karl Fischer analytical method was used to determine the molar fraction of water along the reaction progress. The main challenge of modeling this reaction was to represent the highly non-ideal mixture in liquid phase, specially for reactions runs starting with acetic acid mole fraction higher than 50%. In these conditions, it was assumed the occurrence of acetic acid dissociation that leads to ionic interactions. The thermodynamic model used to calculate the activities was the NRTL. This model predicted well the molecular interactions on low acidity conditions but presented deviations when molar fractions of acid and water were higher. The present study also aims to develop calibration models for analytical on-line method to obtain molar fraction profiles along the reaction. Near infrared spectroscopy (NIR) was used due to the possibility of quantitatively analyze water mole fraction, besides the mole fractions of the other components. Another important reason to choose NIR was its crescent use in industry, and the present work intended to contribute towards the future use of this on line monitoring technique in industrial applications. Multivariate calibration models using internal set of calibration and cross-validation seems to be suitable for this system. External set of calibration leads to less accurate models, and the causes of this lack of accuracy were not clearly identified.

Diante dos requisitos de concentração em nível de mg/kg exigidos pelos órgãos reguladores de saúde do governo, tornou-se mandatório o emprego de metodologia com limite de quantificação, precisão e exatidão adequados aos rígidos controles de processo pelos fabricantes da CAPB, bem como, dispor de técnicas convencionais com poder de resolução e proficiência pelo controle de qualidade e neste contexto inseriu-se a cromatografia a gás. Neste estudo foram estabelecidos os procedimentos analíticos que definiram as melhores condições para identificar e quantificar as impurezas MCAS e DCAS na matriz CAPB por meio da cromatografia a gás. A preparação das amostras consistiu da derivação das impurezas MCAS e DCAS a ésteres etílicos e a extração líquido-líquido em hexano para separar dos demais constituintes da matriz. Os modos de detecção acoplados à cromatografia a gás foram a ionização pela chama (GC/FID), a captura de elétrons (GC/ECD) e a espectrometria de massas (GC/MS). A validação comprovou que as metodologias são lineares entre 4 e 50 mg/kg com recuperação de 70 a 120%, apresentam limites de quantificação inferiores a 10 mg/kg e produziram médias e incertezas similares na amostra examinada, constituindo-se alternativas para a determinação de cloroacetatos em betaínas. / Dissertação (Mestrado em Tecnologia Nuclear) / IPEN/D / Instituto de Pesquisas Energeticas e Nucleares - IPEN-CNEN/SP

Características agronômicas e ecológicas de rizóbios isolados de solos ácidos e de baixa fertilidade da Amazônia.

This research work had as objectives, to accomplish morphologic characterization of rhizobia isolated from Amazonian soils; laboratory evaluations about the tolerance to acidity and toxic Al, the capacity to solubilize calcium (P-Ca) and aluminum (P-Al) phosphates, the indole-acetic acid production (AIA); the ability of the isolates to induce nodulation and to fix nitrogen under the acidity and low fertility conditions of the regional soils in greenhouse experiment; to genetically characterize the isolated ones selected using the PCR-RFLP technique. The infectivity test showed that there was significant increase in the cowpea biomass. With relationship to the phenotypic characterization, the 200 isolates showed a great diversity. Among these, 165 and 127 presented high tolerance within the media with pH 4,5 and pH 4,5 + Al, respectively. From the 200 isolates, 68 solubilized P-Ca and 47 P-Al, and 32 isolates solubilized both P-Ca and P-Al. The AIA production significantly increased (p <0,01) with tryptophan quantities added in the medium for 19 of the 92 isolates tested in the experiment. It was observed production of dry matter and nodule number and mass superior simbiotic to the control treatment and similar to the fertilized treatment for various rhizobia isolates. The 20 selected isolates evaluated presented high genotypic diversity, with the derived dendrogram PCR-RFLP profiles showing that they can be divided inside two main genders, Rhizobium and Bradyrhizobium. Associating the results of simbiotic efficiency and nodulation, it was observed that the isolates of rhizobia from Amazonian soils presented good results, and they can be recommended for tests of agronomic efficiency for cowpea bean. / Para aumentar a contribuição de rizóbia na nutrição e desenvolvimento de plantas é necessário desenvolver pesquisas de seleção de estirpes mais eficientes quanto à fixação do N2. Este trabalho teve como objetivos, realizar a caracterização morfológica de isolados de rizóbio oriundos de solos amazônicos; avaliar em laboratório, a tolerância à acidez e Al tóxico, a capacidade de solubilização de fosfato de cálcio (P-Ca) e alumínio (P-Al), a produção de acido indol-acético (AIA); a capacidade de isolados em induzirem nodulação e fixarem nitrogênio sob as condições de acidez e baixa fertilidade dos solos regionais em casa-de-vegetação; caracterizar geneticamente os isolados selecionados usando a técnica de PCR-RFLP. Os testes de infectividade mostraram que houve aumento significativo na biomassa de caupi. Quanto à caracterização fenotípica, os 200 isolados mostraram uma grande diversidade. Dentre estes, 165 e 127 isolados apresentaram elevada tolerância nos meios de cultura com pH 4,5 e pH 4,5 + Al, respectivamente. Dos 200 isolados testados, 68 solubilizaram P-Ca e 47 P-Al, sendo que 32 solubilizaram tanto o P Ca quanto o P Al. A produção de AIA aumentou significativamente com as doses de triptofano adicionadas ao meio (p<0,01) para 19 dos 92 isolados testados no experimento. Observou-se produção de matéria seca, número e massa seca dos nódulos e acúmulo de nitrogênio superiores ao tratamento testemunha e similares ao tratamento adubado para vários isolados de rizóbia. Os 20 isolados selecionados e avaliados apresentaram alta diversidade genotípica, onde o dendrograma derivado de perfis de PCR-RFLP mostrou que podem ser divididos dentro de dois gêneros principais, Rhizobium e Bradyrhizobium. Associando-se os resultados de eficiência simbiótica e nodulação, observou-se que os isolados de rizóbio de solos da Amazônia apresentaram bom desempenho e podem ser recomendados para testes de eficiência agronômica em feijão caupi.

O presente trabalho teve como objetivo avaliar, por meio da microscopia eletrônica de varredura (MEV), a capacidade de remoção da smear layer dos terços médio e apical do canal radicular utilizando soluções quelantes e desmineralizantes e, quantificar, por meio da espectrometria de absorção atômica com chama, a concentração de íons cálcio presentes nessas soluções após suas utilizações. Vinte e cinco caninos superiores foram preparados pela técnica Free Tip Preparation com 4 instrumentos acima do inicial e irrigados com hipoclorito de sódio 1% a cada troca de instrumento. Os dentes foram distribuídos aleatoriamente em 4 grupos, conforme protocolo utilizado para a irrigação final: G1 - EDTA 15%, G2 - quitosana 0,2%, G3 - ácido cítrico 10%, G4 - ácido acético 1%. O grupo controle (G5) não recebeu irrigação. Foram utilizados 5 mL de cada solução por 3 minutos. Após percorrer toda extensão do canal radicular a solução extravasada pelo forame foi coletada e encaminhada para análise espectrométrica. Os espécimes foram seccionados longitudinalmente e preparados para análise em MEV. As fotomicrografias obtidas foram avaliadas qualitativamente por três examinadores, que atribuíram escores às imagens, conforme a quantidade de smear layer. Os dados obtidos pela MEV foram analisados estatisticamente por meio do Teste de Kruskal-Wallis e Dunn. Para avaliação da espectrometria utilizou-se Tukey-Kramer (one-Way ANOVA). Os resultados mostraram que o EDTA 15%, quitosana 0,2% e ácido cítrico 10% removeram a smear layer de forma semelhante entre si e estatisticamente diferente (p<0,05) do ácido ácético 1% e controle. Não houve diferença na capacidade de limpeza das soluções quando os terços médio e apical foram comparados. A maior concentração de íons cálcio foi observada no grupo do EDTA 15% e quitosana 0,2%, sem diferença entre ambos. O grupo do ácido acético 1% apresentou as menores concentrações e o ácido cítrico 10%, concentrações intermediárias e diferentes estatísticamente dos dois grupos (p<0,01). Concluiu-se que as soluções de EDTA 15%, quitosana 0,2% e ácido cítrico 10% foram eficientes na remoção da smear layer do terço médio e apical do canal radicular. As soluções de EDTA 15% e quitosana 0,2% promoveram o maior efeito desmineralizante, seguidas pelo ácido cítrico 10% e acido acético 1%. / This study aimed to evaluate, by scanning electron microscopy (SEM), the ability to remove the smear layer from the apical and middle thirds of the root canal using chelators and demineralizing solutions, and quantify, by atomic absorption spectrophotometry flame, the concentration of calcium ions present in these solutions after their use. Twenty-five canines were prepared by Free Tip Preparation Technique with four instruments above the initial and irrigated with 1% sodium hypochlorite in each change of instrument. The teeth were randomly divided into four groups, according to the protocol used for the final irrigation: G1 - EDTA 15%, G2 - 0.2% chitosan, G3 - citric acid 10%, G4 - 1% acetic acid. The control group (G5) did not receive irrigation. We used 5 mL of each solution for 3 minutes. After irrigating the entire length of the root canal through the apical foramen, the solution was collected and sent to spectrometric analysis. The specimens were sectioned longitudinally and prepared for SEM analysis. The photomicrographs were qualitatively evaluated by three observers, who attributed scores to the images, equivalent to the amount of smear layer removal. The data obtained by SEM was statistically analyzed using the Kruskal-Wallis and Dunn tests. To evaluate the spectrometer, the Tukey-Kramer (one-way ANOVA) was used. The results showed that 15% EDTA, 0.2% chitosan and 10% citric acid had a similar smear layer removal with statistically different (p <0.05) when compared to 1% acetic acid and the control group. There was no difference between the solutions cleaning ability when the middle and apical thirds were compared. The highest concentration of calcium ions was observed in 15% EDTA group and 0.2% chitosan, with no statistical difference between them. The 1% acetic acid group had the lowest concentration and the 10% citric acid intermediate concentrations with statistical difference between each other (p <0.01). It can be concluded that the solutions of 15% EDTA, 0.2% chitosan and 10% citric acid were effective in removing the smear layer of the middle and apical thirds of the root canal. 15% EDTA solutions and 0.2% chitosan promoted the greatest effect on the demineralization, followed by 10% citric acid and 1% acetic acid.

Développement de méthodes permettant la détection et la quantification de microorganismes d'altération du vin : étude de facteurs de développement / Development of methods for the detection and quantification of spoilage microorganisms in wine : study of growing factors

Les nouvelles pratiques utilisées pour l’élaboration du vin amènent à une recrudescence des altérations microbiennes. C’est pourquoi, de nouvelles méthodes doivent être développées afin de quantifier ces microorganismes de façon précise, rapide et avec de faibles coûts. Les principales altérations du vin sont dues aux bactéries acétiques (BA) (A. aceti, A. pasteurianus, G. oxydans et Ga. liquefaciens) et à Brettanomyces bruxellensis. Par l’action d’enzymes, les 1ères transforment l’éthanol en acide acétique alors que B. bruxellensis transforme les acides hydroxycinnamiques en éthyles phénols (EP) (molécules odorantes désagréables). La cytométrie en flux couplée à la technique d’hybridation in situ en fluorescence a tout d’abord été étudiée. Aucun résultat reproductible n’a été développé pour les BA en vin rouge alors que pour B. bruxellensis, le protocole existant a été amélioré avec une quantification possible en 18 h. La PCR en temps réel a également été utilisées afin de quantifier ces microorganismes. Un protocole a été développé pour la quantification des BA en vin rouge (103 cellules/mL) avec l’utilisation d’un témoin interne microbiologique permettant de valider le rendement de l’extraction de l’ADN. Pour B. bruxellensis, trois kits commerciaux ont été analysés lors d’une étude interlaboratoires. Les quantifications se sont révélées significativement différentes des énumérations sur boite de Pétri avec une quantification des cellules mortes. De plus, il a été étudié et validé l’effet population de B. bruxellensis sur l’efficacité du SO2. Il ressort également de ces expérimentations que les cellules en état viable mais non cultivable ne produisent pas d’EP. / New practices used to elaborate wine lead to an increase of wine spoilage due to microorganisms. That is why, new technics have to be developed to quantify these microorganisms accurately, quickly and with low costs. The main wine spoilages are due to acetic acid bacteria (AAB) (A. aceti, A. pasteurianus, G. oxydans and Ga. liquefaciens) and Brettanomyces bruxellensis development. AAB transforms ethanol to acetic acid while B. bruxellensis transforms hydroxycinnamic acids to ethyl phenols (EP) (unpleasant odor molecules). In order to detect these wine spoilage microrganisms, flow cytometry coupled to fluorescent in situ hybridization has been assessed. No reproducible results have been developed for AAB in red wine while for B. bruxellensis, the existing protocol has been improved with a possible quantification after 18 h compared to 48-72 h in the previous protocol. The real-time PCR was also used to quantify these microorganisms. A protocol has been developed for the AAB quantification in red wine (103 cells/mL) with the use of a microbiological internal control to validate the DNA yield after extraction. For B. bruxellensis, three commercial kits were analyzed in an interlaboratory study. Quantifications were significantly different to the enumerations by Petri dish, with dead cell quantifications. Moreover, we demonstrated that the effectiveness of sulfite is dependent of the B. bruxellensis population. It also appears from these experiments that cells in viable but not culturable state do not produce EP.

Contribution of the adenine nucleotide carrier, porin, and sphingolipid metabolism to mitochondria membrane permeabilization in Saccharomyces cerevisiae / Contribution du transporteur adénine nucléotide, porine, et du métabolisme des sphingolipides à la perméabilization de la membrane mitochondrial de saccharomyces cerevisiae

La perméabilisation de la membrane mitochondriale externe (MOMP) est un évènement décisif lors de la balance entre la vie et la mort de la cellule. Les évènements biochimiques responsables de la MOMP ne sont pas encore complètement définis. Deux mécanismes majeurs et distincts ont été impliqués dans le contrôle de la MOMP: i) l'action des protéines de la famille de Bcl-2, qui peuvent directement s'insérer dans la membrane mitochondriale externe (OMM) et induire l'ouverture de pores; et ii) le pore de transition de perméabilité (PTP), un canal non sélectif de la membrane mitochondriale interne, qui induit un gonflement des mitochondries à la suite d'une ouverture prolongée, suivie d'une éventuelle rupture de la MOM. L'intérêt croissant pour la biologie de la mort cellulaire, entretenu par des contributions significatives d'études sur la levure dans la compréhension des mécanismes biologiques de base, ont amené l'eucaryote unicellulaire Saccharomyces cerevisiae sur le devant de la scène. La levure est dépourvue de certains régulateurs majeurs de l'apoptose, mais possède cependant des homologues des facteurs pro-apoptotiques mitochondriaux, ainsi que des orthologues des composantes moléculaires généralement attribuées au PTP, notamment le l'échangeur ADP/ATP (AAC) et la Porine (Por1). Ces caractéristiques de S. cerevisiae, ainsi que la disponibilité d'outils moléculaires et génétiques, ont fourni une excellente opportunité d'étudier les membres de la famille de Bcl-2 dans un environnement contrôlé, ainsi que la contribution du PTP et de ses composantes à la mort cellulaire. Dans ce travail, la contribution des groupes thiol de l'AAC, leur oxydation, Por1, et la possible interaction entre ces deux protéines, ont été explorées au cours de la mort cellulaire de la levure induite par l'acide acétique. Nous avons observé que l'oxydation de Aac2p, notamment la formation de ponts disulfures, ne contribuait pas au programme de mort induit par l'acide acétique. Cette conclusion est soutenue par l'absence d'un profil particulier d'oxydation d'Aac2p. Néanmoins, il avait été précédemment démontré que l'AAC était requis pour la relocalisation du cytochrome c induite par l'acide acétique, et que son absence promouvait la survie des cellules de levure. Par contre, la délétion de Por1 diminue la viabilité de levures traitées par l'acide acétique. Il a été émis l'hypothèse que les deux protéines participent à la même voie de régulation de la mort cellulaire. Pour la tester, la relocalisation du cytochrome c a été mesurée dans des mitochondries isolées de cellules Δaac1/2/3, Δpor1 et Δaac1/2/3Δpor1 suite à l'exposition à l'acide acétique. Les données obtenues suggèrent que l'absence de Por1 n'affecte pas la relocalisation du cytochrome c pendant la mort cellulaire induite par l'acide acétique, mais pourrait être importante pour sa régulation. Quand à la fois AAC et Por1 sont absentes, les mitochondries de levure peuvent toujours relarguer le cytochrome c, suggérant l'existence d'un mécanisme indépendant de l'AAC. De plus, nous avons montré que les deux protéines ont des effets distincts dans les réponses cellulaires à différents stress. En effet, l'absence des AACs contribue à l'augmentation de la résistance au stress osmotique et de l'intégrité de la paroi cellulaire. Enfin, S.cerevisiae a été utilisé comme modèle pour étudier les aspects mécanistiques relatifs à la fonction des protéines de la famille de Bcl-2. Notamment, nous avons évalué le rôle des sphingolipides sur l'action du régulateur pro-apoptotique humain Bax. Nous avons montré que l'absence de Isc1p, une inositol phospholipase C qui dégrade les sphingolipides complexes en céramides chez la levure, favorise la viabilité de cellules de levures exprimant une forme active de Bax. Nous avons ensuite révélé que cet effet n'est pas associé à des modifications de l'activité de Bax. Il semblerait plutôt que cet effet soit lié aux conséquences cellulaires de l'action de Bax. / A decisive event in the cell’s life-or-death decision is the mitochondrial outer membrane permeabilization (MOMP). The biochemical events responsible for MOMP, are not entirely defined. Two major and distinct mechanisms have been implicated in the control of MOMP: i) the action of Bcl-2 family proteins, which can directly engage the outer mitochondria membrane (OMM) and induce the opening of pores; and ii) the mitochondrial permeability transition pore (PTP), an inner membrane unselective channel that induces mitochondria swelling upon long term openings, and eventual rupture of the OMM. The growing interest in cell death biology, fostered by the relevant contributions of yeast to the understanding of basic biological processes, brought the unicellular eukaryote Saccharomyces cerevisiae into the scene. Yeast cells lack some of the major regulators of apoptosis but still possess homologues of mitochondrially enclosed pro-apoptotic factors, as well as orthologues of the molecular components generally ascribed to PTP, including the ADP/ATP carrier (AAC) and Porin (Por1). These particular features of S. cerevisiae, along with the availability of genetic and molecular tools, provided an excellent opportunity to study Bcl-2 family members in a “controlled” environment, or the contribution of the PTP and its components to cell death. In this work, the particular contribution of AAC’s thiol goups, its oxidation, Por1, and of a possible interaction between both proteins to acetic acid-induced yeast cell death, was explored. We observed that oxidative modifications of Aac2p, namely the crosslinking of thiols, do not contribute to the acetic acid-induced cell death program. Such idea is supported by the apparent absence of a particular Aac2p oxidation pattern. Nevertheless, the AAC was previously found to be required for acetic acid-induced cytochrome c release, and its absence promoted the survival of yeast cells. Deletion of Por1, on the other hand, decreased the viability of yeast cells treated with acetic acid. It was hypothesized that the two proteins could share the same pathway in the regulation of cell death. To test it, cytochrome c release was evaluated in mitochondria isolated from Δaac1/2/3, Δpor1 and Δaac1/2/3Δpor1 cells following acetic acid exposure. The data obtained suggest that absence of Por1 does not affect cytochrome c releaseduring acetic acid induced-death, but it may be important for its regulation. When both the AAC and Por1 were absent, yeast mitochondria could still release cytochrome c, raising the possibility of an AAC-independent mechanism. Furthermore, we found both proteins have distinct effects that regulate the cellular response to different stresses. Indeed, absence of the AACs somehow contributed to increased osmotic stress and cell wall resistance. Finally, S. cerevisiae was used as a model to study mechanistic aspects relative to the function of Bcl-2 family proteins. Namely, we assessed the role of sphingolipids in the action of the human pro-apoptotic regulator Bax. We found that absence of Isc1p, an inositol phospholipase C that degrades complex sphingolipids into ceramides in yeast, favored the viability of yeast cells expressing an active form of Bax. It was further revealed that this effect is not associated with changes to the action of Bax; rather, it might be related with the cellular consequences of Bax-action. A parallel with the effect of Uth1p absence in yeast cells expressing Bax suggests that the absence of Isc1p could affect the selective degradation of mitochondria by mitophagy, and thus produce a different cell death response. This work provides new insights into the physiological events underlying the contribution of mitochondrial proteins, previously associated with cell death responses, and sphingolipid metabolism to cell death induced by acetic acid and Bax, respectively. Once again, the yeast S. cerevisiae proved to be an excellent model for the research of cell life and death.

La composition phénolique du vin rouge détermine fortement sa qualité: couleur, goût, texture et la plupart des bienfaits pour la santé. Les conditions ambiantes de la vigne modulent l'équilibre hormonal endogène et l'expression de gènes qui contrôlent la voie de synthèse des composés flavonoïdes, en déterminant la composition phénolique finale du raisin. Même s'ils ont été étudiés, les effets des applications des régulateurs de croissance végétale sur l'équilibre hormonal endogène et la qualité du raisin, les effets de ces substances sur la composition et la qualité du vin sont pauvrement documentés. Le traitement des raisins destinés à la vinification avec des régulateurs de croissance végétale est un outil potentiel pour modifier la qualité des raisins et du vin rouge. Ce projet de thèse a pour objectif d’étudier l’impact d'applications de régulateur de croissance végétale sur la composition phénolique des raisins de Vitis vinifera L. cépages Cabernet Sauvignon et Carménère. L’acide abscissique, l’acide indole-3-acétique et l'acide 2-chloroethilphosphonique ont été appliqués à divers stades phénologiques du raisin, doses et conditions environnementales: Les essais ont été menés à Maipo et Cachapoal au Chili et à Bordeaux en France, dans des vignobles commerciaux et expérimentaux ainsi que sur des plantes cultivées en pots. Il a été examiné l'effet de ces traitements sur le contenu interne d'hormones, sur l'expression de gènes structuraux et régulateurs de la synthèse de composés flavonoides et sur la qualité des raisins, en particulier la composition phénolique de sa pellicule. De plus, des vinifications ont été réalisées à partir de raisins traités pour déterminer l'effet des traitements sur la composition chimique et phénolique du vin, ainsi que sur des attributs qualitatifs tels que les arômes et la texture des vins, jugés par un panel d'évaluation sensorielle. / Phenolic composition strongly determines red wine quality: color, taste, texture and most health benefits. Vineyard environmental conditions modulate endogenous hormonal balance and gene expression which control the flavonoid biosynthetic pathway leading to final grape phenolic composition. Even when the effects of plant growth regulator applications on grape endogenous hormonal balance and quality have been studied, the effect of these substances on wine composition and quality is poorly documented. The treatment of wine grapes with plant growth regulators is a potential tool in order to modify red wine phenolic composition and quality. This thesis project describes six experiments on plant growth regulator applications on developing grapes of Vitis vinifera L., cvs Cabernet Sauvignon and Carménère. Abscisic acid, Indole-3-acetic acid and 2-chloroethylphosphonic acid were applied in different phenological stages, doses and environmental conditions: Maipo and Cachapoal regions in Chile and Bordeaux region in France, commercial and experimental vineyards and plants in containers. The effect on changes in the internal hormonal content, expression of flavonoid biosynthetic and regulatory genes and grape quality, in particular grape skin phenolic composition were examined. In addition, winemaking was performed in order to assess the effect of treatments on wine chemical and phenolic composition and on wine aroma and texture attributes judged by a sensory panel.

This thesis studies the consumption of carbon dioxide in industrial processes, starting from synthesis gas, and using this building block in subsequent syntheses; ultimately integrating CO2 utilization with previously non-CO2 consuming processes.

Charakterizace vybraných bakteriálních kmenů získaných během evolučních experimentů / Characterization of bacterial strains obtained in evolutionary engineering

This diploma thesis deals with application of evolutionary engineering on PHA producing bacterial strains. Two bacterial strains, Cupriavidus necator H16 and Halomonas halophila, were chosen for the evolutionary experiments. Copper cations (Cu2+) and sodium chloride (NaCl) were chosen as the selective pressure for C. necator H16; acetic acid (AA) and levulinic acid (LA) for Halomonas halophila. The adapted strains were during long-time evolutionary experiments characterized by GC-FID and SEC-MALS. The growth of the adapted strains was studied by the mean of optical density measurement. The amount of viable cells was determined by spectral FC after their expositon to selected stress factors. Specific enzyme activities of enzymes involved in citrate and glyoxalate cycle, enzymes generating NADPH, LA metabolism enzyme and PHA biosynthesis enzymes were determined. The adapted strains were compared with the wild-type of strains. The successfull adaptation of C. necator H16 adapted to Cu2+ was detected. Biomass and PHA production of both wild and adapted H. halophila strains cultivated in lignocellulosis waste were determined. It was found out that H. halophila adapted to the LA is capable of producing more PHA than the wild strain of this bacteria.
