The Bold and the Beautiful and Generations : a comparative ethnographic audience study of Zulu-speaking students living in residences on the University of Natal's Durban campus.

This thesis is an ethnographic study of the soap opera viewing patterns and interpretations of Zulu-speaking students living in residences on the Natal University's Durban campus who watch The Bold and the Beautiful (an American soap opera) and Generations (a South African soap opera). It presents an analysis of how the viewing practices of the students compare with the findings of soap opera audience studies conducted abroad. The students' motivations and reasons for watching both soap operas are investigated. The reason for choosing black students as subjects is that I wanted to determine how a soap opera (Generations) which is comprised largely of black cast members and designed with a young black audience in mind, is interpreted and impacts on the lives of said audience, when compared with an American soap opera (The Bold and the Beautiful) which has an almost exclusively white American cast, and is popular with young black viewers in spite of the fact that it appears on the surface to be unrelated to their everyday lives. Individual one-on-one interviews were conducted with 40 students, 20 male and 20 female. The interviews were analysed to gauge how the viewing behaviour of the students differs from, or is similar to, soap opera studies conducted elsewhere in the world. It emerged that the students watch in groups and not alone, and that watching Generations and The Bold and the Beautiful is a social activity, not motivated from loneliness or isolation. The ways in which the students relate to the characters and situations of both soap operas is also examined, in an attempt to establish the role that these two shows play in the creation of the students' identities. The students displayed a tendency to be more critical of Generations than of The Bold and the Beautiful in the sense that they compared it (unfavourably) in terms of quality of production, to its American counterpart, as well as in the sense that they analysed storylines in terms of their own lived experiences and were quick to criticise Generations when they felt that it did not conform to their notions of the reality of being a black South African. They accepted situations and characters on The Bold and the Beautiful far less critically, although they did voice objections to certain characters and situations which they felt were morally questionable in terms of their understanding of right and wrong. It also became apparent that there was a greater emotional involvement with the characters on The Bold and the Beautiful than with those on Generations. The students interpretations of (and level of involvement with) situations, characters and storylines are examined, as well as the ways in which they derive pleasure from both soaps and incorporate them into their own lives. Thérapie cellulaire de l'angiogenèse tumorale : évaluation par imagerie morphologique et fonctionnelle en IRM et vidéomicroscopie de fluorescence

Introduction : L'angiogenèse tumorale conduit au développement de nouveaux vaisseaux destinés à permettre la croissance de la tumeur. Les vaisseaux tumoraux sont caractérisés notamment par des anomalies des cellules murales (cellules musculaires périvasculaires), responsables d'anomalies de la fonctionnalité et de la maturation. Dans ce travail de thèse, nous avons étudié un modèle tumoral de thérapie cellulaire par injection de cellules murales en IRM et vidéomicroscopie de fluorescence. Matériels et méthodes : Notre étude a porté sur un modèle sous cutané de carcinome épidermoïde chez la souris nude. Les animaux étaient divisés en trois groupes : contrôle (n=17), contrôle négatif (n=16) et " traité " avec injection locale de cellules murales humaines (n=17). Les animaux bénéficiaient d'une IRM et d'une exploration par vidéomicroscopie avant (J7) et après traitement (J14). Les paramètres mesurés étaient la taille tumorale (pied-à-coulisse et IRM), la densité microvasculaire (DMV par IRM, vidéomicroscopie et histologie), l'ADC, f, Dr et D* (IRM de diffusion), les variations de R2* sous air, oxygène et carbogène (IRM par effet BOLD) et " l'index leakage " (reflétant la perméabilité capillaire, en vidéomicroscopie). Résultats : Lors de la croissance tumorale, le groupe contrôle a montré une diminution des vaisseaux circulants (ou fonctionnels) qui se reflétait par une diminution du D* et du R2* sous air, une perte de la capacité à répondre au carbogène qui se reflétait par une augmentation du delta R2* sous carbogène, et une augmentation de la perméabilité capillaire qui se traduisait par un " index leakage " plus élevé. Dans le groupe traité par injection de cellules murales, nous avons observé un ralentissement de la croissance tumorale et une stabilisation de ces paramètres de microcirculation et maturation vasculaire. Conclusion : Nous avons montré un effet biologique de notre thérapie cellulaire par injection locale de cellules murales qui se traduisait par un ralentissement de la croissance tumorale, une stabilisation de l'hémodynamique microcirculatoire et de la maturation, et une perméabilité capillaire diminuée, concordants avec l'effet présumé stabilisateur et normalisateur des cellules murales sur les microvaisseaux.

EEG and BOLD-contrast fMRI in brain:cerebrovascular reactivity, suppression of neuronal activity, global and local brain injury

Abstract The purpose of the present study was to gain more insight into the blood oxygen level-dependent (BOLD)-contrast functional MRI (fMRI) in the brain and its connection to EEG, both in global and local scales of their temporal and spatial relations. BOLD signal changes were studied during hyperventilation (HV) induced EEG reactivity of intermittent rhythmic delta activity (IRDA). The BOLD signal in gray matter decreased 30% more in subjects with IRDA (N = 4) than in controls (N = 4), during the first two minutes of HV. This difference disappeared during IRDA in EEG. BOLD signal changes may provide additional information about dynamic hemodynamic changes relative to HV induced EEG reactivity. BOLD signal changes were investigated during sudden deepening of thiopental anesthesia into EEG burst-suppression level in pigs (N = 5). Positive (6–8%) or negative (-3– -8%) group average BOLD signal changes correlated to the thiopental bolus injection were seen. Positive and negative responses covered 1.6% and 2.3% of the brain voxels, respectively. BOLD signal changes in brain are associated with sudden deepening of thiopental anesthesia into EEG burst-suppression level, but they are spatially inconsistent and scarce. Somatosensory BOLD response was studied in brain before and after globally induced methotrexate (MTX) exposition in pigs (N = 4). After the MTX exposure, reduced (from 2–4% to 0–1%) or negative (-2% to -3%) BOLD responses were detected. Somatosensory BOLD-contrast response shows a slight difference in brain before and after globally induced MTX exposition. An experimental epilepsy model for development of simultaneous EEG and BOLD-contrast fMRI in the localization of epilepsy was developed and tested. Dynamic penicillin induced local epilepsy was applied in deep isoflurane anesthesia in pigs (N = 6). Relatively high (10–20%) and localized BOLD signal increase was found. The dynamic penicillin induced focal epilepsy model in deep isoflurane anesthesia with simultaneous EEG and BOLD-contrast fMRI is feasible for the development of these methods for localization of epileptic focus or foci. In conclusion, with careful experimental design and analysis, BOLD-contrast fMRI with EEG provides a potential tool for monitoring and localising functional changes in the brain.

Le pays d'Aire est un espace prospère à la frontière des comtés de Saint-Pol, de Flandre et d'Artois. Cette région rurale et céréalière est arrosée par la Lys, rivière permettant le commerce avec les grandes villes de Flandre. L'ensemble forme l'assise territoriale d'un bailliage artésien dont le siège est placé à Aire-sur-la-Lys, petite ville vivant dans l'ombre de sa puissante voisine Saint-Omer et de la cité de Thérouanne. Cet espace intermédiaire entre Ypres et Hesdin, Lille et Saint-Omer, possède toutefois d'incontestables atouts économiques et une identité originale. Aire, farouchement artésienne mais fortement influencée par la Flandre, fut toujours à la recherche d'un protecteur à la fin du Moyen Age. Ce rôle fut confié tantôt au prévôt de la collégiale tantôt au bailli. Un lignage noble originaire d'Enguinegatte entreprend de mettre la main sur la région, d'abord en menant une subtile politique matrimoniale puis en monopolisant la charge de bailli. Cet office, cumulé à la charge de chambellan local, leur permet de constituer un solide réseau local dans lequel sont impliqués l'échevinage, les chanoines mais aussi le cousinage et les bâtards du lignage. Les Viesville sont alors les véritables maîtres d'Aire et profitent de la reconversion économique de la région après les grandes pestes. La famille parvient à surmonter les coups durs reçus dans le premier quart du XVe siècle mais son influence est finalement balayée par la guerre menée en Artois par Louis XI après la mort de Charles le Téméraire. Véritable "cygne noir", cet avatar dans la lente construction de l'Etat coûte à cette famille tout ce qu'il lui avait apporté dans un premier temps. / The country of Aire is a prosperous area on the border of the counties of Saint-Pol, Flanders and Artois. This rural and cereal area is watered by the Lys river which allows trade with the big cities of Flanders. It forms the territorial basis of an Artesian bailiwick which headquarter is located in Aire-sur-la-Lys, a town living in the shade of the powerful neighbourhood of Saint-Omer and Thérouanne. This intermediate space between Ypres and Hesdin, Lille and Saint-Omer, has however indisputable economic advantages and its own identity. Aire, fiercely from Artois but strongly influenced by Flanders, was at the end of the Middle Ages always in search of a protector. This role was sometimes entrusted to the provost of the collegiate church, sometimes to the bailiff. A noble lineage native of Enguinegatte began to get hold on the region, at first by leading a subtle marital politic then by monopolizing the bailiff’s office. This status, added to the one of the chamberlain of the duke of Burgundy, allowed them to establish a robust local network in which were implied local elites, canons but also cousins and illegitimate children of the lineage. The La Viesville were then the real masters of Aire and took advantage of the economic reconversion of the region after the Great Plague. The family succeeded in overcoming the shocks received in the first quarter of the XVth century but its influence was finally annihilated by the war led in Artois by Louis XI after the death of Charles the Bold. Real " black swan ", this step in the slow construction of the State cost to this family all that it had brought before.

Signal amplification in a microfluidic immunoassay system via Binding Oligo Ladder Detection : Applying the Exazym® signal amplification to the Gyrolab® platform

Immunoassays are analytical methods that use the highly specific binding of antibodies in order to detect and quantify an analyte. The technique has become a staple in modern biopharmaceutical research and diagnostics, however the measurement of biomarkers like dysregulated cytokines require ultra-sensitive immunoassays that can detect molecules at sub pg/mL concentrations. One such method is the Exazym® signal amplification. Based on a method called Binding Oligo Ladder Detection (BOLD), it is a set of add-on reagents where a primer is conjugated to a detection antibody which is then combined with a template, polymerase and modified DNA nucleotides to generate a oligonucleotide ladder that is detected with a secondary detection antibody; this amplifies the signal by a factor of 10-100 in an existing immunoassay.  By applying this method to the Gyrolab® microfluidic immunoassay system, a sensitivity increase of 880x-1800x was achieved between a pre-synthesised BOLD product and the polymerised BOLD product. Several key factors for successful polymerisation in the microfluidic system were identified: adding the template separately before the polymerase and using a buffer with low ionic strength for the secondary detection antibody. Applying the BOLD amplification to an existing Gyrolab TNF-α assay only resulted in similar sensitivity as previous methods however. This report demonstrates that BOLD amplification can be successfully performed in a flow-through format on miniaturized affinity columns in the Gyrolab system to increase the sensitivity by orders of magnitude, where both the immunoassay and the amplification steps are automated in the system. However, further optimisation is needed for application in biomarker assays.

Resting state brain networks in young people with familial risk for psychosis

Abstract Neuropsychiatric illnesses usually become overtly manifest in adolescence and early adulthood. A critical long-term aim is to be able to prevent the development of such illnesses, which requires instruments to identify subjects at high risk of illness and to offer them effective interventions. There is an indisputable need for more sophisticated methods to enable more precise detection of adolescents and young adults who are at high risk of developing psychosis. Abnormal function in brain networks has been reported in people with schizophrenia and other psychotic disorders. Similar abnormalities have been found also in people at risk for developing psychosis, but it is not known whether this applies also to spontaneous resting state activity in young people with a familial risk for psychosis. We conducted resting-state functional MRI (R-fMRI) in 72 (29 male) young adults with a history of psychosis in one or both parents (FR) but without psychosis themselves, and 72 (29 male) similarly healthy control subjects without familial risk for psychosis. Both groups in the Oulu Brain and Mind study were drawn from the Northern Finland Birth Cohort 1986. All volunteers were 20–25 years old. Parental psychosis was established using the Care Register for Health Care. R-fMRI data was pre-processed using independent component analysis (ICA). A dual regression technique was used to detect between-group differences with p < 0.05 threshold corrected for multiple comparisons at voxel level. FR subjects demonstrated significantly decreased activity compared to control subjects in the default mode network and in the central executive network and increased activity in the cerebellum. The findings clarify previously controversial literature on the subject. The finding suggests that abnormal activity in these brain networks in rest may be associated with increased vulnerability to psychosis. The findings maybe helpful in developing more precise methods for detecting young people at highest risk for developing psychosis. / Tiivistelmä Psykoottisiin häiriöihin sairastutaan yleensä nuoruudessa tai varhaisaikuisuudessa. Psykoositutkimuksen tavoitteena on löytää uusia menetelmiä, joiden avulla kyettäisiin tunnistamaan suurimmassa psykoosiriskissä olevat nuoret, jotta heille voitaisiin tarjota sairautta ennaltaehkäiseviä hoitokeinoja. Skitsofreniaan ja muihin psykoottisiin häiriöihin sairastuneilla on havaittu aivotoiminnan poikkeavuuksia. Samankaltaisia aivotoiminnan poikkeavuuksia on havaittu myös nuorilla, jotka ovat vaarassa sairastua psykoosiin. Toistaiseksi on ollut epäselvää, onko psykoosiin sairastuneiden henkilöiden lapsilla aivohermoverkkojen toiminnan poikkeavuuksia lepotilassa. Suoritimme aivojen lepotilan MRI-tutkimuksen (R-fMRI) 72:lle (29 miestä) nuorelle aikuiselle, joiden jompikumpi vanhempi oli sairastunut psykoosin sekä 72:lle (29 miestä) nuorelle aikuiselle, joiden vanhemmat eivät olleet sairastaneet psykoosia. Molemmat tutkimusryhmät tässä Oulu Brain and Mind -tutkimuksessa olivat Pohjois-Suomen 1986 syntymäkohortin jäseniä. Tutkittavat olivat 20–25 vuoden iässä. Lepotilan toiminnallinen magneettikuvaus suoritettiin 1.5 Teslan Siemensin magneettikuvantamislaitteella. Tutkimuskohteiksi valittiin lepotilan toiminnallinen aivohermoverkko, toiminnan ohjauksesta vastaava aivohermoverkko ja pikkuaivot. Kuvantamisdataan sovellettiin itsenäisten komponenttien analyysia aivohermoverkkojen määrittämistä varten. Ryhmien välisen eron havaitsemiseen käytettiin ei-parametristä permutaatiotestiä, joka kynnystettiin tilastollisesti merkitsevään tasoon (p < 0.05). Lepotilan oletushermoverkossa ja toiminnanohjauksesta vastaavassa aivohermoverkoissa havaittiin vähäisempää aktiivisuutta ja pikkuaivoissa kohonnutta aktiivisuutta perinnöllisessä psykoosiriskissä olevilla nuorilla aikuisilla verrattuna verrokkeihin. Tutkimustulokset selkeyttivät aiempaa ristiriitaista kirjallisuutta tutkimusaiheesta. Tutkimuksessa havaittujen aivoalueiden poikkeava toiminta lepotilassa voi liittyä kohonneeseen psykoosin puhkeamisriskiin. Tutkimuslöydösten avulla voidaan todennäköisesti edesauttaa parempien kuvantamismenetelmien kehittämistä suurimmassa psykoosiriskissä olevien nuorten tunnistamiseen.

Method evaluations in spatial exploratory analyses of resting-state functional magnetic resonance imaging data

Abstract Resting-state (RS) measurements during functional magnetic resonance imaging (fMRI) have become an established approach for studying spontaneous brain activity. RS-fMRI results are often obtained using explorative approaches like spatial independent component analysis (sICA). These approaches and their software implementations are rarely evaluated extensively or specifically concerning RS-fMRI. Trust is placed in the software that they will work according to the published method descriptions. Many methods and parameters are used despite the lack of test data, and the validity of the underlying models remains an open question. A substantially greater number of evaluations would be needed to ensure the quality of exploratory RS-fMRI analyses. This thesis investigates the applicability of sICA methodology and software in the RS-fMRI context. The experiences were used to formulate general guidelines to facilitate future method evaluations. Additionally, a novel multiple comparison correction (MCC) method, Maxmad, was devised for adjusting evaluation results statistically. With regard to software considerations, the source code of FSL Melodic, popular sICA software, was analyzed against its published method descriptions. Unreported and unevaluated details were found, which implies that one should not automatically assume a correspondence between the literature and the software implementations. The method implementations should rather be subjected to independent reviews. An experimental contribution of this thesis is that the credibility of the emerging sliding window sICAs has been improved by the validation of sICA related preprocessing procedures. In addition to that, the estimation accuracy regarding the results in existing RS-fMRI sICA literature was also shown not to suffer even though repeatability tools like Icasso have not been used in their computation. Furthermore, the evidence against conventional sICA model suggests the consideration of different approaches to analysis of RS-fMRI. The guidelines developed for facilitation of evaluations include adoption of 1) open software development (improved error detection), 2) modular software designs (easier evaluations), 3) data specific evaluations (increased validity), and 4) extensive coverage of parameter space (improved credibility). The proposed Maxmad MCC addresses a statistical problem arising from broad evaluations. Large scale cooperation efforts are proposed concerning evaluations in order to improve the credibility of exploratory RS-fMRI methods. / Tiivistelmä Aivoista toiminnallisella magneettikuvantamisella (engl. functional magnetic resonance imaging, fMRI) lepotilassa tehdyt mittaukset ovat saaneet vakiintuneen aseman spontaanin aivotoiminnan tutkimuksessa. Lepotilan fMRI:n tulokset saadaan usein käyttämällä exploratiivisia menetelmiä, kuten spatiaalista itsenäisten komponenttien analyysia (engl. spatial independent component analysis, sICA). Näitä menetelmiä ja niiden ohjelmistototeutuksia evaluoidaan harvoin kattavasti tai erityisesti lepotilan fMRI:n kannalta. Ohjelmistojen luotetaan toimivan menetelmäkuvausten mukaisesti. Monia menetelmiä ja parametreja käytetään testidatan puuttumisesta huolimatta, ja myös menetelmien taustalla olevien mallien pätevyys on edelleen epäselvä asia. Eksploratiivisten lepotilan fMRI-datan analyysien laadun varmistamiseksi tarvittaisiin huomattavasti nykyistä suurempi määrä evaluaatioita. Tämä väitöskirja tutki sICA-menetelmien ja -ohjelmistojen soveltuvuutta lepotilan fMRI-tutkimuksiin. Kokemuksien perusteella luotiin yleisiä ohjenuoria helpottamaan tulevaisuuden menetelmäevaluaatioita. Lisäksi väitöskirjassa kehitettiin uusi monivertailukorjausmenetelmä, Maxmad, evaluaatiotulosten tilastolliseen korjaukseen. Tunnetun sICA-ohjelmiston, FSL Melodicin, lähdekoodi analysoitiin suhteessa julkaistuihin menetelmäkuvauksiin. Analyysissa ilmeni aiemmin raportoimattomia ja evaluoimattomia menetelmäyksityiskohtia, mikä tarkoittaa, ettei kirjallisuudessa olevien menetelmäkuvausten ja niiden ohjelmistototeutusten välille pitäisi automaattisesti olettaa vastaavuutta. Menetelmätoteutukset pitäisi katselmoida riippumattomasti. Väitöskirjan kokeellisena panoksena parannettiin liukuvassa ikkunassa suoritettavan sICA:n uskottavuutta varmistamalla sICA:n esikäsittelyjen oikeellisuus. Lisäksi väitöskirjassa näytettiin, että aiempien sICA-tulosten tarkkuus ei ole kärsinyt, vaikka niiden estimoinnissa ei ole käytetty toistettavuustyökaluja, kuten Icasso-ohjelmistoa. Väitöskirjan tulokset kyseenalaistavat myös perinteisen sICA-mallin, minkä vuoksi tulisi harkita siitä poikkeavia lähtökohtia lepotilan fMRI-datan analyysiin. Evaluaatioiden helpottamiseksi kehitetyt ohjeet sisältävät seuraavat periaatteet: 1) avoin ohjelmistokehitys (parantunut virheiden havaitseminen), 2) modulaarinen ohjelmistosuunnittelu (nykyistä helpommin toteutettavat evaluaatiot), 3) datatyyppikohtaiset evaluaatiot (parantunut validiteetti) ja 4) parametriavaruuden laaja kattavuus evaluaatioissa (parantunut uskottavuus). Ehdotettu Maxmad-monivertailukorjaus tarjoaa ratkaisuvaihtoehdon laajojen evaluaatioiden tilastollisiin haasteisiin. Jotta lepotilan fMRI:ssä käytettävien exploratiivisten menetelmien uskottavuus paranisi, väitöskirjassa ehdotetaan laaja-alaista yhteistyötä menetelmien evaluoimiseksi.

Analýza souvislostí mezi simultánně měřenými EEG a fMRI daty / Analysis of connections between simultaneous EEG and fMRI data

Electroencephalography and functional magnetic resonance are two different methods for measuring of neural activity. EEG signals have excellent time resolution, fMRI scans capture records of brain activity in excellent spatial resolution. It is assumed that the joint analysis can take advantage of both methods simultaneously. Statistical Parametric Mapping (SPM8) is freely available software which serves to automatic analysis of fMRI data estimated with general linear model. It is not possible to estimate automatic EEG–fMRI analysis with it. Therefore software EEG Regressor Builder was created during master thesis. It preprocesses EEG signals into EEG regressors which are loaded with program SPM8 where joint EEG–fMRI analysis is estimated in general linear model. EEG regressors consist of vectors of temporal changes in absolute or relative power values of EEG signal in the specified frequency bands from selected electrodes due to periods of fMRI acquisition of individual images. The software is tested on the simultaneous EEG-fMRI data of a visual oddball experiment. EEG regressors are calculated for temporal changes in absolute and relative EEG power values in three frequency bands of interest ( 8-12Hz, 12-20Hz a 20-30Hz) from the occipital electrodes (O1, O2 and Oz). Three types of test analyzes is performed. Data from three individuals is examined in the first. Accuracy of results is evaluated due to the possibilities of setting of calculation method of regressor. Group analysis of data from twenty-two healthy patients is performed in the second. Group EEG regressors analysis is realized in the third through the correlation matrix due to the specified type of power and frequency band outside of the general linear model.

The master thesis deals with the joint EEG-fMRI analysis based on a heuristic model that describes the relationship between changes in blood flow in active brain areas and in the electrical activity of neurons. This work also discusses various methods of extracting of useful information from the EEG and their influence on the final result of joined analysis. There were tested averaging methods of electrodes interest, decomposition by principal components analysis and decomposition by independent component analysis. Methods of averaging and decomposition by PCA give similar results, but information about a stimulus vector can not be extracted. Using ICA decomposition, we are able to obtain information relating to the certain stimulation, but there is the problem in the final interpretation and selection of the right components in a blind search for variability coupled with the experiment. It was found out that although components calculated from the time sequence EEG are independent for each to other, their spectrum shifts are correlated. This spectral dependence was eliminated by PCA / ICA decomposition from vectors of spectrum shifts. For this method, each component brings new information about brain activity. The results of the heuristic approach were compared with the results of the joined analysis based on the relative and absolute power approach from frequency bands of interest. And the similarity between activation maps was founded, especially for the heuristic model and the relative power from the gamma band (20-40Hz).
