Caractérisation des lymphopénies post-chirurgies cardiaques de type Fontan

La thymectomie est un geste chirurgical fréquemment nécessaire lors des interventions cardiaques en période néonatale. Il est connu que la résection du thymus entraîne une lymphopénie ne semblant pas avoir de conséquences cliniques graves. La lymphopénie constatée serait plus importante chez les patients ayant eu une chirurgie cardiaque de type Fontan. Toutefois, on ignore si la lymphopénie est uniquement secondaire à la thymectomie ou si elle est liée à ce type particulier de chirurgie cardiaque. La présente recherche porte sur 19 patients opérés selon l'approche Fontan; ils ont été comparés à 6 patients "contrôle" ayant eu une thymectomie au cours d'une chirurgie cardiaque d'un autre type. Les résultats indiquent que les patients de type Fontan accusent une diminution du nombre de cellules naïves CD4+ et CD8+ et plus particulièrement une baisse de leurs émigrants thymiques (CD45RA+CD31+/CD4+). On note en contrepartie une expansion du répertoire mémoire (CD45RO+). Ces altérations lymphocytaires sont comparables aux contrôles. Il semble donc que les anomalies lymphocytaires relevées soient reliées principalement à la thymectomie et non pas au type de chirurgie. Les infections plus importantes chez les Fontan, quant à elles, pourraient s'expliquer par une évolution post-opératoire défavorable. / The thymectomy is a surgical procedure often done during cardiac surgery. It is well known that the resection of the thymus can cause alterations of lymphocyte subpopulations without major clinical consequences. This lymphopenia could be more pronounced in patients who undergone a cardiac surgery named "Fontan". Nevertheless, it is not known if these alterations are due to the thymectomy or this specific type of cardiac surgery. We recruited 19 patients with Fontan surgery and compared them to 6 patients who had a cardiac surgery of another type. Our results show decreased CD4+ and CD8+ naive T cells populations and more specifically, low numbers of thymic emigrants (CD45RA+CD31+/CD4+). Expansion of memory repertoire (CD45RO+) is also noted. These findings are also found in the control group. Thus, perturbations of T cells populations seem to be related to the thymectomy rather than the type of cardiac surgery. Infections were more severe in the Fontan group but they can be explained by an unfavourable post-operative evolution in this group of patients.

The role of the International Organization for Migration (IOM) in the prevention of HIV-infections among mobile and vulnerable populations (MVPs) and potential emigrants in Beitbridge

This study assessed the role of the International Organization for Migration (IOM) in HIV-prevention among mobile and vulnerable populations (MVPs) and potential emigrants in Beitbridge. A mixed-method approach, which combines quantitative and qualitative approaches, was used in this action research. A total of 20 in-depth face-toface interviews were conducted with key informants and 56 self-administered questionnaires were completed by MVPs and potential emigrants in Beitbridge. The study found that a number of effective strategies are currently being implemented as a preventative measure by the IOM within MVPs and potential emigrants’ communities in Beitbridge. At the same time, however, in order to ensure sustainability of these HIVprevention initiatives, the IOM must promote long-term synergies with other strategic partners throughout the project cycle. It is recommended that, the IOM strategically position itself by moving a step further from being the sole provider of emergency humanitarian support towards devising sustainable and durable solutions among MVPs and potential emigrants. / Sociology / M.A. (Social Behaviour Studies in HIV-AIDS)

Identidad, alteridad y espacios físicos en dos novelas sobre emigrantes colombianos

Ce mémoire porte sur des aspects reliés à la migration et la représentation, notamment à la façon dont l’émigration a été racontée dans la littérature colombienne et la manière dont les émigrants ont été représentés dans les romans Lucas Guevara (1914), de Alirio Díaz Guerra, et Paraíso travel (2002), de Jorge Franco, tous deux écrivains colombiens. Nous développerons à cette fin notre analyse à partir des trois concepts: identité, altérité et espaces physiques, qui sont au cœur de ce genre de récits. Ainsi, nous abordons ces notions séparément, et en même temps, nous identifions les relations existantes entre chacune d’elles. En fait, l’un des résultats de cette étude porte sur la façon dont ces romans sur l’émigration traitent le problème de l’identité en relation avec les représentations de l’altérité et des espaces physiques. Des aspects tels que la séparation de deux traditions culturels différentes (l’hispanique versus l’anglo), la confrontation des personnages provenant d’un contexte religieux et conservateur avec l’ambiance libérale et sécularisante de New York, les conséquences du contact des émigrants avec la société étrangère et leur transformation ultérieure, la marginalité des émigrants colombiens aux États-Unis, leur manque de communication, les représentations des hommes, femmes, immigrants et natifs, ainsi que la façon dont les romans glissent quelques indices sur la condition de l’individu moderne et postmoderne, font partie des sujets analysés dans notre étude. / This study addresses issues related to migration and representation, in particular it considers how emigration has been recounted in Colombian literature and how emigrants have been represented in Alirio Díaz Guerra’s Lucas Guevara (1914) and Jorge Franco’s Paraíso travel (2002). Our analysis of the novels is based in three main concepts: identity, alterity and physical spaces, which are instrumental in the articulation in the novels plot and characters. For the purposes of this study, we treat these three concepts separately while still considering the existing connections between them. Some of the findings of this work are related to how these novels about emigrants raise the problem of identity and how identity is tightly connected to alterity, as well as to individual othering and the representation of physical spaces. Some of the topics this study considers are the following: the standoff of two different cultural traditions (Hispanic versus Anglo), the confrontation of traditionally educated Latin American individuals with a liberal and secularizing environment as that of New York City, the consequence of migration and the ulterior transformation of the characters, the marginality of Colombian emigrants in the United States of America, the problem of communication, the representation of men, women, immigrants and natives, as well as the way both novels slip some clues about the condition of the modern and postmodern individuals. / Este trabajo trata sobre aspectos relacionados con la migración y la representación, particularmente sobre la forma en que la emigración ha sido narrada en la literatura colombiana y la manera en que los emigrantes son representados en dos novelas de escritores colombianos: Lucas Guevara (1914), de Alirio Díaz Guerra, y Paraíso travel (2002), de Jorge Franco. Para ello hacemos uso de tres conceptos que son la base de nuestro estudio: identidad, alteridad y espacios físicos. La elección de estas nociones como ejes de análisis se basa en que, efectivamente, se trata de tres problemáticas que están en el centro de las narraciones. Por tal motivo, tratamos los tres conceptos por separado, sin olvidar las conexiones que existen entre ellos. Algunos de los hallazgos de nuestro análisis se relacionan con la forma en que estas novelas plantean el problema de la identidad de los emigrantes, cómo la identidad se relaciona con la alteridad y cómo ambas cosas se articulan a su vez con las descripciones y representaciones de los espacios físicos. El enfrentamiento entre tradiciones culturales diferentes (lo hispano versus lo anglo), la confrontación de dos personajes venidos de un contexto religioso y conservador con un contexto liberal y secularizante como el de Nueva York, las consecuencias de ese encuentro y la posterior transformación de los protagonistas, la marginalidad de los emigrantes colombianos en los Estados Unidos, su incomunicación, las representaciones de hombres, mujeres, inmigrantes y nativos y, en fin, la manera en que ambas novelas arrojan pistas sobre la condición de los individuos moderno y posmoderno, forman parte de los tópicos analizados en nuestro estudio.

Language Proficiency, Citizenship, and Food Insecurity among Predominantly Immigrant Caribbean Latinos in Massachusetts: A Masters Thesis

BACKGROUND: Latinos report higher food insecurity than the national average, and food insecurity has been associated with adverse health outcomes wherein Latinos experience disparities. This study quantified the independent effects of language-speaking proficiency and citizenship on increased food insecurity among a predominantly immigrant Caribbean Latino sample in Lawrence, Massachusetts. METHODS: The analytic sample comprised 574 participants aged 21-83 who visited a community health center in 2011-2013. Food insecurity was assessed via the 6-item US Household Food Security Survey. Multivariable logistic modeling (adjusted for self-reported age group, gender, education, and marital status) examined the independent associations between language proficiency and citizenship on increased food insecurity. RESULTS: One-third of participants were classified as food insecure. Most respondents were citizens (59.5%), foreign-born (92.4%; 70.3% from the Dominican Republic), and spoke monolingual Spanish (72.8%). Monolingual Spanish-speakers had marginally increased odds of food insecurity (odds ratio (OR) = 1.50, 95% confidence interval (CI): 1.00 to 2.26), compared to bilingual participants; however after adjustment this relationship was attenuated (OR = 1.25, 95% CI: 0.79 to 2.00). Non-citizenship was not associated with increased odds of food insecurity (OR=1.18, 95% CI: 0.82 to 1.68). CONCLUSION: Food insecurity in this predominantly immigrant Caribbean Latino sample was higher than the national average for Latinos. Future research on food insecurity among different Latino ethnicities is needed in order to inform targeted interventions that promote food security.

Avvikarnas oundvikliga öde : En queer läsning av Vilhelm Mobergs utvandrarserie med fokus på relationen mellan Robert och Arvid

Vilhelm Moberg (1898–1973) var en av sin tids mest inflytelserika svenska författare. De fyra romanerna i hans episka utvandrarserie hör de till de mest lästa svenska romanerna genom tiderna. Trots att forskningsfältet om Moberg är stort har hans verk hitintills inte analyserats utifrån ett queerperspektiv. Syftet med denna magisteruppsats är att göra en queer läsning av Mobergs utvandrarserie, med fokus på relationen mellan Robert och Arvid. Dessa unga drängars relation har sällan givits utrymme inom Mobergforskningen och när den omnämnts har den lästs som kamratskap. Min hypotes är att det, trots indikationerna på ett homofobt förhållningssätt hos Moberg vid tiden för utvandrarseriens tillkomst, är möjligt att läsa fram en romantisk kärlekshistoria mellan männen. I uppsatsen lyfts frågeställningen om huruvida textens djur, djuriskhet och djursymbolik öppnar upp för att läsa fram de unga männens kärlekshistoria. Tidelagstemat som i utvandrarserien är förbundet med Arvid kan förstås som en omskrivning för homosexualitet, vilket ihop med queerbetonad djursymbolik som i texten omgärdar Robert, framhäver deras icke-utskrivna begär och kärlek. Analysen synliggör den dikotomi mellan manlighet och omanlighet som i romanerna gestaltas genom Robert och hans storebror Karl Oskar. Storebrodern tillskrivs stark karaktär, individualism och arbetsstyrka, egenskaper som historiskt sett kategoriserats som manliga. Robert skrivs fram som arbetsskygg, lögnaktig och med ett infantilt drömmande om guld och rikedom, egenskaper som kategoriseras som omanliga. Analysen påvisar att i likhet med tidelagstemat kan också omanligheten läsas som en omskrivning för homosexualitet. Frågan om huruvida Mobergs föreställning om homosexualitet som ett ”olycksöde” har haft konsekvenser för gestaltningen av Robert och Arvid, genomsyrar analysen. Avsikten med detta har inte varit att smutskasta Moberg utan att lyfta det motsägelsefulla med hans homofoba hållning då det i denna uppsats fastställs att han författat en vacker kärlekshistoria om två unga män. / Vilhelm Moberg (1898–1973) was one of the most influential Swedish writers of his time. His epic The Emigrants with its four parts are among the most read Swedish novels of all time. The field of research about Moberg is large; despite this none of his works have been analyzed from a queer angle until now. The purpose of this master’s thesis (one year) is to do a queer reading of Moberg’s The Emigrants, focusing on the relationship between Robert and Arvid. The bond between these young farmhands has rarely been given attention within the research about Vilhelm Moberg; when it is mentioned in previous research, it’s perceived as camaraderie. My hypothesis is that, despite indications of a homophobic attitude in Moberg, it is possible to make visible a romantic love story between these men. In this thesis, a question regarding whether animals, animality or animal symbolism in the text makes the young men’s love story increasingly visible, is raised. The theme of bestiality in The Emigrants coincides with Arvid, and can be seen as a synonym for homosexuality. In combination with queer animal symbolism surrounding Robert, the men’s hidden desire and love is enhanced. The analysis highlights a dichotomy between masculinity and unmanliness that is portrayed through the relationship between Robert and his older brother Karl Oskar. The latter is being ascribed traditionally masculine traits, such as resilient character, individualism and physical ability for labor, whereas Robert is associated with unmanly attributes such as laziness, untrustworthiness, and an infantile longing for gold and riches. The analysis shows that Robert’s unmanliness, just as the bestiality theme, is a synonym for homosexuality. The question as to whether Moberg’s impression that homosexuality is an “unfortunate fate” has had consequences for to the portrayal of Robert and Arvid, permeates this analysis. The purpose has not been to smear Moberg but to highlight the paradox of his homophobic stance, since this thesis conclusively determines that he has written a beautiful love story involving two young men.

Étude du mécanisme par lequel la thérapie à l'IL7 induit l'expansion homéostatique des lymphocytes T CD4+

Dans les cas de lymphopénie, les lymphocytes T résiduels prolifèrent exagérément dans un phénomène appelé «expansion homéostatique périphérique» (HPE), qui est efficace pour la régénération des T CD8+, mais inefficace pour les T CD4+. L’interleukine-7 (IL7) est une cytokine homéostatique utilisée afin d’augmenter les comptes lymphocytaires T des patients lymphopéniques. Toutefois, la raison de l’expansion préférentielle des lymphocytes T CD8+ par l’IL7 demeure toujours inconnue. Nous montrons que cette expansion est due au fait que l’IL7 induit une prolifération efficace des T CD8+ périphériques (CD8+PERI) ainsi que des émigrants thymiques CD8+ (CD8+RTEs). Par contre, l’effet prolifératif de l’IL7 est restreint presqu’uniquement aux CD4+RTEs même si les CD4+PERI survivent mieux que les CD4+RTEs. De plus faibles doses d’IL7 sont nécessaires aux CD4+RTEs afin de phosphoryler STAT5 ou de proliférer comparativement aux CD4+PERI et nous démontrons que les contacts TCR/CMHII sont nécessaires à la prolifération induite par l’IL7 des CD4+RTEs en périphérie. De fait, augmenter au Flt3 ligand le nombre de cellules dendritiques périphériques d’une souris donneuse, avant de transférer ses TPERI dans des souris receveuses traitées à l’IL7 induit une prolifération significative des CD4+PERI. Nos résultats indiquent donc que l’abondance des contacts TCR/CMHII reçus dans le thymus semble contrôler la sensibilité à l’IL7 des CD4+RTEs. Finalement, l’observation que les CD8+PERI et CD8+RTEs prolifèrent pareillement pendant la thérapie à l’IL7, alors que la prolifération des T CD4+ est largement restreinte aux RTEs expliquerait pourquoi, dans les cas de lymphopénie, la régénération des T CD4+ est aussi dépendante de la thymopoïèse. / In lymphopenic settings, residual T lymphocytes typically undergo exaggerated proliferation via homeostatic peripheral expansion (HPE). While HPE efficiently regenerates CD8+ T cells, it is unable to normalize CD4+ T-cell counts. Interleukin-7 (IL7) is a homeostatic cytokine, currently used in trials in order to increase T-cell counts in lymphopenic humans. Nowadays, it is still not known why IL7 therapy is more effective toward the expansion of CD8+ T cells rather than CD4+ T cells. Here we show that CD8+ T cells preferential expansion is due to IL7-induced efficient proliferation of peripheral CD8+ T cells (CD8+PERI) and CD8+ recent thymic emigrants (CD8+RTEs). In contrast, the proliferative action of IL7 is largely restricted to CD4+RTEs although CD4+PERI survive better than CD4+RTEs. Interestingly, CD4+RTEs require lower concentrations of IL7 in order to phosphorylate STAT5 or proliferate when compared to CD4+PERI, and we demonstrate the requirement for TCR/MHCII contacts to support the IL7-induced HPE of CD4+RTEs in the periphery. Furthermore, augmenting the number of MHCII expressing cells in the periphery of donor mice by treating them with Flt3 ligand (Flt3L) prior transferring their TPERI cells in IL7 therapy-treated recipients, significantly enhances the IL7-induced proliferation of CD4+PERI. Our results indicate so far that the abundance of TCR triggering occurring inside the thymus drives IL7 responsiveness of CD4+RTEs. Moreover, the observation that CD8+PERI and CD8+RTE proliferate similarly during IL7 therapy, while proliferation of CD4+ T cells is largely restricted to RTEs, may explain why CD4+ T cells regeneration in lymphopenic settings is highly dependent on thymopoiesis.

The immune response to yellow fever vaccination in aged individuals

Mit zunehmendem Alter verringert sich die Fähigkeit des menschlichen Organismus Infektionen erfolgreich zu bekämpfen und, z.B. nach Impfung, einen protektiven Schutz aufzubauen. Es wird vermutet, dass die Alterung des Immunsystems eine Rolle dabei spielt. Wichtige Ergebnisse liefern dazu vor allem tierexperimentelle Studien, welche jedoch die Komplexität menschlicher Immunität nur bedingt abbilden. Nur ansatzweise erforscht ist der Einfluss immunologischer Alterungsprozesse auf die primäre Immunantwort im Menschen. Um ein besseres Verständnis über primäre Immunantworten im Alter zu erlangen, haben wir junge (n=11, Med=26 Jahre) und ältere (n=12, Med=60 Jahre) Erwachsene mit einem viralen Erreger experimentell infiziert und die akute Immunreaktion und Entwicklung langlebiger Protektion eingehend untersucht. Dafür verwendeten wir den attenuierten Lebendimpfstoff gegen Gelbfieber, der ein hervorragendes Modelsystem darstellt um anti-virale Primärantworten im Menschen zu erforschen. Wir konnten zeigen, dass ältere Impflinge weniger Gelbfiebervirus-(GFV)-neutralisierende Antikörper produzierten, schwächere GFV-spezifische CD8+ T-Zellantworten erzeugten und quantitativ als auch qualitativ veränderte GFV-spezifische CD4+ T-Zellantworten generierten. Zudem wiesen ältere Impflinge häufiger eine vergleichsweise späte Virämie auf. Unsere systembiologische Untersuchungen zeigten, dass die niedrige Zahl von frisch aus dem Thymus ausgewanderten naiven CD4+ T Zellen, sogenannten CD4+ Recent thymic emigrants, sowie der Mangel an dendritischen Zellen vor bzw. am Beginn der Infektion ausschlaggebend für die schlechtere Immunreaktion und niedrigere Langzeit-Immunität bei Älteren war. Daraus schließen wir, dass in älteren Menschen die Verfügbarkeit eines breiten Repertoires naiver CD4+ T-Zellen und eine effektive Induktion des angeborenen Immunsystems in der frühen Phase einer primären Infektion kritisch für die akute Abwehr viraler Erreger und die Ausbildung protektiver Immunität ist. / The immunological competence to fight infections and to generate protective immunity, for example upon vaccination, progressively declines with advancing age. Although the aged immune system has been extensively studied at steady state and in aged animal models, there is only rudimentary understanding on how aging affects the immune response to a primary infection in humans. Involving complex individual systemic immune properties, such investigations have been very challenging particularly with the given restrictions of experimental infections in humans. In our study, we explored age-related changes in human immunity during experimental, primary immunization with live-attenuated yellow fever (YF) vaccine. In 11 young (median age: 26 years) and 12 elderly (median age: 60 years) vaccinees, we assessed individual viral burden and compared humoral and cellular immunity by advanced flow cytometric analysis over the entire course of the acute infection and up to 3 years after it. We discovered that aged subjects developed fewer neutralizing antibodies, mounted diminished YF-specific CD8+ T-cell responses and showed quantitatively and qualitatively altered YF-specific CD4+ T-cell immunity. A comparatively late peak in YF viremia suggested impaired infection control and viral clearance in the elderly. Among numerous immune signatures, low in vivo numbers of naive CD4+ recent thymic emigrants (CD4+ RTE) prior immunization and peripheral dendritic cells (DCs) in the early phase of the innate response phase were indicative for reduced acute responsiveness and altered long-term persistence of human cellular immunity to YF vaccination in the elderly. Thus, we reveal by this study that essential elements of immune responses such as CD4+ RTEs and DCs affect productive immunity in the elderly, explaining conclusively diminished responsiveness to vaccination with neo-antigens and infection with de novo pathogens in aged people.

Friedrich Armand Strubberg’s semi-autobiographical novel Friedrichsburg, published in Germany in 1867, is a fountain of information about the German settlements in the Hill Country of Texas established in the years 1844-1848 by a corporation of German noblemen. The noblemen safely ensconced in their comfortable estates in Germany attempted to live up to their responsibilities and supply the settlers with basic needs, but their efforts fell woefully short. In consequence, the immigrants often were thrown upon their own devices and compelled to live from what they could learn to grow or hunt in a new land with unfamiliar climate, plants, and animals. Many hundreds perished from disease, exposure, and malnutrition. But after a painful period, the German settlements took root and began to prosper; lending a Germanic stamp to the Hill Country area of Texas that persists to the present day. In Friedrichsburg, the reader encounters many dramatic stories attendant to the foundation years of Fredericksburg, Texas, 1846/1847 when Friedrich Armand Strubberg, under the assumed name Dr. Schubbert, served as the first colonial director of the town. The situations are presented vividly and entertainingly, and although the book offers a romanticized and, in this sense, a sanitized version of the immigrants’ travails, I maintain that it contains historically accurate depictions of people and events that have been largely overlooked in other accounts of the period. The dissertation offers the first complete translation of the novel. An introduction provides an overview of German immigration in Texas, a short biography of Friedrich Armand Strubberg, and a discussion of his place in literature about Texas published in Germany in the nineteenth century. Extensive endnotes document names and episodes as they appear in the text and distinguish between what is fact and what is fiction in the novel. A bibliography of works published about Texas in the nineteenth century is supplied as an appendix. / text

Σκοπός της διατριβής ήταν να διερευνηθεί η συγκρότηση των παραδοχών, στάσεων και πρακτικών που συνδέονται με τη διαπολιτισμική ικανότητα των εκπαιδευτών ενηλίκων στο πεδίο της διδασκαλίας της ελληνικής ως δεύτερης γλώσσας σε μετανάστες εκπαιδευόμενους. Εξετάστηκε η σχέση του βαθμού διαπολιτισμικής ικανότητας των εκπαιδευτών ενηλίκων με τις παραδοχές τους σε σχέση με την επαγγελματική τους ταυτότητα, καθώς και με βασικές αρχές της εκπαίδευσης ενηλίκων. Διερευνήθηκε, επίσης, η συνάφεια της ανάπτυξης της διαπολιτισμικής ικανότητας με επιλογές επαγγελματικής ανάπτυξης που σχετίζονται με θεωρητικές αναφορές σχετιζόμενες με το πλαίσιο της μετασχηματίζουσας μάθησης. Η έρευνα βασίστηκε σε μεθοδολογική τριγωνοποίηση, αξιοποιώντας ερωτηματολόγιο που συμπληρώθηκε από 211 εκπαιδευτές που διδάσκουν την ελληνική γλώσσα ως δεύτερη ή ξένη, παρατήρηση 20 τμημάτων διδασκαλίας, συνεντεύξεις με τους εκπαιδευτές και δείγμα εκπαιδευομένων αυτών των τμημάτων (23 και 47 συνεντεύξεις αντίστοιχα) και συμμετοχική παρατήρηση των συναντήσεων και του εξ αποστάσεως διαλόγου μιας –σε εθελοντική βάση συγκροτημένης– ομάδας 18 εκπαιδευτών. Τα δεδομένα της έρευνας αποτυπώνουν την κυριαρχία, στο μεγαλύτερο μέρος των εκπαιδευτών, μονοπολιτισμικών οπτικών ως προς το στόχο διδασκαλίας της δεύτερης γλώσσας και τη διαχείριση πολιτισμικών πλαισίων αναφοράς. Οι εκπαιδευτές αυτοί υιοθετούν εκπαιδευτικές πρακτικές μετωπικής διδασκαλίας, ενώ, παράλληλα, η διάκριση ανάμεσα σε εκπαιδευόμενους «με» και «χωρίς» κουλτούρα, οδηγεί ένα μέρος τους σε έντονα αρνητικά συναισθήματα και πρακτικές υποτίμησης ορισμένων ομάδων εκπαιδευομένων. Μειοψηφική (της τάξης του 20-25%) εμφανίζεται η ομάδα των εκπαιδευτών που είναι ανοιχτοί στη διαπραγμάτευση των παραδοχών, δημιουργώντας χώρους διαλόγου, αποστασιοποίησης από τις «ασφαλείς παραδοχές» και κριτικού στοχασμού που ενδέχεται να λειτουργήσει μετασχηματιστικά. Οι εκπαιδευτές αυτοί αντιμετωπίζουν τους εκπαιδευόμενους ως ενεργούς συνδημιουργούς γνώσης, υιοθετούν σε μεγαλύτερο βαθμό συμμετοχικές εκπαιδευτικές τεχνικές, ενώ αναζητούν και αξιοποιούν την ανατροφοδότηση από εκπαιδευόμενους και κριτικούς φίλους, σε συνδυασμό με πρακτικές ενδοσκόπησης, ως στοιχεία της επαγγελματικής τους ανάπτυξης. Από τα δεδομένα της έρευνας αναδεικνύεται, τέλος, η συμβατότητα της μεθοδολογίας ανάπτυξης της διαπολιτισμικής ικανότητας με εκείνη που οδηγεί σε μετασχηματίζουσα μάθηση, ως προς χαρακτηριστικά όπως ο βιωματικός προσανατολισμός και η εστίαση στην επεξεργασία των παραδοχών αλλά και στην ανάδυση των συναισθημάτων που τις συνοδεύουν. / The purpose of this thesis was to investigate the formation of assumptions, attitudes and practices related to intercultural competence of adult educators in the field of teaching Greek as a second language to immigrant students. The thesis examined the relationship between the degree of intercultural competence of adult educators and their assumptions in relation to their professional identity and basic principles of adult learning. It also investigated the relevance between the development of intercultural competence and professional development options related to the context of transformative learning. The research was based on a methodological triangulation, utilizing questionnaire completed by 211 educators who teach Greek as a second or foreign language, observation of 20 teaching groups, interviews with trainers and trainees sample of the above mentioned groups (23 and 47 interviews respectively) and participatory observation of the meetings and the distance conversation of a voluntary structured group formed by 18 trainers . The research data illustrate that monocultural views in relation to the target of teaching a second language, as well as management of cultural frames of reference, are common for the majority of trainers. These educators adopt frontal teaching educational practices, while at the same time the distinction between learners 'with' and 'without ' culture leads a number of trainers into intense negative emotions and practices of devaluation of certain groups of learners. The group of educators that appear open to assumptions’ negotiation seems to be a minority (approximately 20-25 %). These educators keep a distance from "consolidated assumptions" and create spaces of dialogue and critical reflection, that may have a transformational effect. They treat students as active co-creators of knowledge and adopt a greater degree of participatory training techniques, while seeking and utilizing feedback from students and “critical friends”, combined with introspection practices, as elements of their professional development. From the research data emerges, finally, the compatibility of the methodology for the development of intercultural competence and the methodology that leads to transformative learning, in terms of characteristics such as experiential orientation and focus on treatment of the assumptions and the emergence of feelings that accompany assumptions.

Étude du mécanisme par lequel la thérapie à l'IL7 induit l'expansion homéostatique des lymphocytes T CD4+

Dans les cas de lymphopénie, les lymphocytes T résiduels prolifèrent exagérément dans un phénomène appelé «expansion homéostatique périphérique» (HPE), qui est efficace pour la régénération des T CD8+, mais inefficace pour les T CD4+. L’interleukine-7 (IL7) est une cytokine homéostatique utilisée afin d’augmenter les comptes lymphocytaires T des patients lymphopéniques. Toutefois, la raison de l’expansion préférentielle des lymphocytes T CD8+ par l’IL7 demeure toujours inconnue. Nous montrons que cette expansion est due au fait que l’IL7 induit une prolifération efficace des T CD8+ périphériques (CD8+PERI) ainsi que des émigrants thymiques CD8+ (CD8+RTEs). Par contre, l’effet prolifératif de l’IL7 est restreint presqu’uniquement aux CD4+RTEs même si les CD4+PERI survivent mieux que les CD4+RTEs. De plus faibles doses d’IL7 sont nécessaires aux CD4+RTEs afin de phosphoryler STAT5 ou de proliférer comparativement aux CD4+PERI et nous démontrons que les contacts TCR/CMHII sont nécessaires à la prolifération induite par l’IL7 des CD4+RTEs en périphérie. De fait, augmenter au Flt3 ligand le nombre de cellules dendritiques périphériques d’une souris donneuse, avant de transférer ses TPERI dans des souris receveuses traitées à l’IL7 induit une prolifération significative des CD4+PERI. Nos résultats indiquent donc que l’abondance des contacts TCR/CMHII reçus dans le thymus semble contrôler la sensibilité à l’IL7 des CD4+RTEs. Finalement, l’observation que les CD8+PERI et CD8+RTEs prolifèrent pareillement pendant la thérapie à l’IL7, alors que la prolifération des T CD4+ est largement restreinte aux RTEs expliquerait pourquoi, dans les cas de lymphopénie, la régénération des T CD4+ est aussi dépendante de la thymopoïèse. / In lymphopenic settings, residual T lymphocytes typically undergo exaggerated proliferation via homeostatic peripheral expansion (HPE). While HPE efficiently regenerates CD8+ T cells, it is unable to normalize CD4+ T-cell counts. Interleukin-7 (IL7) is a homeostatic cytokine, currently used in trials in order to increase T-cell counts in lymphopenic humans. Nowadays, it is still not known why IL7 therapy is more effective toward the expansion of CD8+ T cells rather than CD4+ T cells. Here we show that CD8+ T cells preferential expansion is due to IL7-induced efficient proliferation of peripheral CD8+ T cells (CD8+PERI) and CD8+ recent thymic emigrants (CD8+RTEs). In contrast, the proliferative action of IL7 is largely restricted to CD4+RTEs although CD4+PERI survive better than CD4+RTEs. Interestingly, CD4+RTEs require lower concentrations of IL7 in order to phosphorylate STAT5 or proliferate when compared to CD4+PERI, and we demonstrate the requirement for TCR/MHCII contacts to support the IL7-induced HPE of CD4+RTEs in the periphery. Furthermore, augmenting the number of MHCII expressing cells in the periphery of donor mice by treating them with Flt3 ligand (Flt3L) prior transferring their TPERI cells in IL7 therapy-treated recipients, significantly enhances the IL7-induced proliferation of CD4+PERI. Our results indicate so far that the abundance of TCR triggering occurring inside the thymus drives IL7 responsiveness of CD4+RTEs. Moreover, the observation that CD8+PERI and CD8+RTE proliferate similarly during IL7 therapy, while proliferation of CD4+ T cells is largely restricted to RTEs, may explain why CD4+ T cells regeneration in lymphopenic settings is highly dependent on thymopoiesis.
