近年社群網站Facebook成為品牌宣傳的新渠道,不同地域、年齡、社會階層或消費水準的人都可以不受實體店面的侷限,透過朋友的分享輕鬆接觸到各時尚精品品牌的資訊。於是精品品牌如何在社群人際間的傳遞過程掌握其品牌形象,還有後續構成的品牌效果是本研究欲探討的重點。 本研究以自我一致性、社會比較的角度切入,輔以平衡理論的觀點進一步檢視消費者觀看社群媒體中的朋友分享貼文是否受到品牌、朋友時尚形象與理想自我一致性高低程度不同而影響其品牌態度與購買意願。 本研究以實驗法進行,研究結果如下: 一、消費者在Facebook動態中看到時尚品牌的朋友品牌分享貼文後,品牌形象與消費者理想自我一致高會比一致低,產生更正面的品牌態度,但對購買意願無顯著影響。 二、知覺品質中介「品牌形象與理想自我一致性」對品牌態度的影響; 但對購買意願無中介效果。 三、 「朋友時尚形象與理想自我形象一致性」高低並不會影響品牌與消費者理想自我形象一致性高低對品牌態度。

The author has attempted a dubbleedged purpose, as indicated by the title. The author firstly deals with Pereboom; begining with his so-called ‘wild coincidence’-argument, by which Pereboom claims agent-causation to be unlikely. The author argues that this argument lacks both scope and strenght. The author then deals with the question of compatiblity between physics and agent-causation as related to Pereboom’s basic problematization; whether agent-causation would or would not diverge from what is expected (from any other event) given our best physical theories. This results in a strong criticism against Pereboom’s whole position, and a positive argument for agent-causation. After the first purpose is achieved, the author turns to the purpose indicated by the subtitle. The author presents a general criticism against the field of metaethics concerning the question of free will. The author also makes suggestions for a possible solution. / Författaren har, som titeln indikerar, tagit på sig ett tveeggat problem. Först hanterar författaren Pereboom; och börjar med hans så kallade ‘wild coincidence’-argument, med vilket Pereboom hävdar att agentkausalitet är osannlik. Författaren menar att detta argument saknar både omfång och styrka. Shadowboard: an agent architecture for enacting a sophisticated digital self

In recent years many people have built Personal Assistant Agents, Information Agents and the like, and have simply added them to the operating system as auxiliary applications, without regard to architecture. This thesis argues that an agent architecture, one designed as a sophisticated representation of an individual user, should be embedded deep in the device system software, with at least equal status to the GUI – the graphical user interface. A sophisticated model of the user is then built, drawing upon contemporary Analytical Psychology – the Psychology of Subselves. The Shadowboard Agent architecture is then built upon that user model, drawing both structural and computational implications from the underlying psychology. An XML DTD file named Shadowboard.dtd is declared as a practical manifestation of the semantics of Shadowboard. An implementation of the Shadowboard system is mapped out, via a planned conversion of two existing integrated systems: SlimWinX, an event-driven GUI system; and XSpaces, an object-oriented tuplespace system with Blackboard-like features. The decision making mechanism passes logic terms and contraints between the various sub-agent components (some of which take on the role of Constraint Solvers), giving this agent system some characteristics of a Generalised Constraint Solver. A Shadowboard agent (built using the system) consists of a central controlling autonomous agent named the Aware Ego Agent, and any number of sub-agents, which collectively form an integrated but singular whole agent modelled on the user called the Digital Self. One such whole-agent is defined in a file named DigitalSelf.xml – which conforms to the schema in Shadowboard.dtd - which offers a comprehensive and generic representation of a user’s stance in a 24x7 network, in particular - the Internet. Numerous types of Shadowboard sub-agents are declared.

Literacy Training in an Urban High School Professional Learning Community

The purpose of this study was to explore the essence of professional learning experiences shared by teachers who participated in a professional learning community (PLC) at a New York City high school in the South Bronx. Guided by Hord's PLC characteristics and Bruner's constructivism theories, this phenomenological study addressed the research questions of what PLC practices urban high school teachers employ to support the academic-literacy achievement of their students of low social economic status (SES); the role of administration in the PLC process; and the roles of a shared mission, values, vision, norms, and collaborative knowledge on the functioning of the PLC. Data collected from the 6 PLC teachers included semi-structured individual interviews, observations of PLC meetings over a 2-month period, participating teacher reflective journal entries, and a researcher's log. Manual data analysis consisted of reading raw data multiple times to determine patterns, themes, and relationships. Additionally, concept and descriptive coding approaches facilitated data source analysis. Gerund words and short phrases generated labels and categories that resulted symbolic representation. The results were that the urban high school teachers demonstrated Hord's PLC characteristics and Bruner's constructivism theories within their PLC's practices and principles leading to decision-making and solutions to problems such as improving teachers' literacy practices, students' literacy skills and classroom behavior, and school wide Individualized Educational Plan process. The findings of this study support the engagement of urban high school teachers in self-directed PLC activities that may promote social change by improving literacy instruction and literacy achievement among students of low SES.

This study deals with the character of Jesus in the Gospel of John and follows the social-scientific approach and aims to correct the traditional views regarding the personality of Jesus. The aim is not a personality analysis but to situate Jesus as a typical person who was a part of and functioned in the first Mediterranean world. The pivotal values of the ancient world such as honor/shame; collectivistic/dyadic personality; family and group orientation; kinship; the patronage system with the patron/broker/client relationship and the purity system are applied to reach the aim of my thesis. It is found that Jesus was a collectivistic/dyadic personality. He functioned in this family orientated as well as in a group orientated community and socially structured patronage system as a broker, totally the contrary to contemporary personality drawings of Jesus found in modern literature. / In hierdie studie, wat 'n skripsie van beperkte omvang is, word nie gepoog om 'n persoonlikheidsanalise van Jesus te doen nie. Die studie wil die probleem van die toepassings en interpretasies van Jesus in populere literatuur aanspreek. Jesus word deur moderne mense en teoloe in prentjies en idees van 'n moderne mens met moderne kategoriee, denke en perspektiewe, waardes en oordele beskryf, geteken en geinterpreteer, en dit is nie 'n werklike getroue en realistiese weergawe van Jesus as historiese mens nie. Jesus word in populere literatuur as 'n individualis geteken; sy persoon en emosies word alleenlik in moderne denke en kategoriee geinterpreteer en dit word slegs op grond van moderne individuele ervaring gedoen. Hierdie interpretasies is nie tipies van die kollektiwistiese, groep-georienteerde eerste-eeuse mediterreense persoon nie. Pratiques parentales, fréquentation d'amis déviants et consommation problématique de substances psychoactives à l'adolescence : effets modérateurs des symptômes dépressifs et du sexe

La consommation et la consommation problématique de substances psychoactives sont des phénomènes répandus à l’adolescence qui ont suscité beaucoup d’intérêt dans les pays occidentaux au cours des dernières décennies. La recherche a souligné la nécessité d’établir un modèle examinant les effets conjoints du groupe de pairs et de la famille sur ces phénomènes. Deux grandes lignées théoriques émergent de la littérature, soit les courants de la socialisation et de la sélection. De plus, de nombreuses études ont tenté d’expliquer la fréquente cooccurrence de ces phénomènes avec la symptomatologie dépressive, généralement associée à un pronostic plus lourd. Toutefois, la nature et le sens de cette association demeurent peu clairs. Une clarification des mécanismes en jeu est nécessaire afin de pouvoir mieux orienter les efforts de prévention et d’intervention. Le premier objectif de cette thèse est de contribuer à clarifier l’étiologie de la consommation problématique à l’adolescence, en examinant comment elle s’articule avec certains facteurs familiaux, comportementaux et affectifs, et avec l’association à des amis déviants et consommateurs, en testant conjointement deux modèles de médiation compétitifs. Le deuxième objectif est de clarifier l’association entre ces différentes dimensions et la symptomatologie dépressive, en testant l’effet modérateur des symptômes dépressifs dans la chaîne médiatrice proposée. Le troisième objectif est de spécifier les particularités pouvant exister entre les garçons et les filles en testant l’effet modérateur du sexe sur cette même chaîne médiatrice. Les données utilisées proviennent d’une cohorte de l’échantillon longitudinal de la Stratégie d’Intervention Agir Autrement (SIAA) comprenant plus de 3000 jeunes fréquentant des écoles de milieux majoritairement défavorisés du Québec, qui ont été suivis pendant leur secondaire (2003-2007). Lorsque testés séparément, le modèle de socialisation (Patterson) se reproduit dans notre échantillon, mais pas le modèle de sélection (Brown). Lorsque testés simultanément, les modèles structurels de régressions croisées suggèrent toutefois qu’aucun des liens postulés par les modèles ne semble se reproduire, au profit de liens médiateurs de stabilité, à l’exception d’un lien de médiation proposé par le modèle de Patterson chez les garçons. Les analyses de modération (multi-groupes) suggèrent que le sexe a bien un effet modérateur, le contrôle comportemental parental semblant particulièrement important dans l’étiologie des comportements problématiques des garçons, alors que les conflits familiaux semblent plus centraux pour les filles. Les analyses de modération suggèrent également un effet modérateur par les symptômes dépressifs, mais alors qu’il était attendu que ces symptômes exacerberaient les liens à l’étude, il apparaît qu’au contraire, aucun des liens ne se révèle significatif dans le groupe des dépressifs. Cette étude suggère donc que la symptomatologie dépressive et la consommation problématique se présentent conjointement dans un profil plus global de risque. À notre connaissance, cette thèse est la première à s’intéresser spécifiquement au rôle modérateur des symptômes dépressifs sur les liens existant entre les différentes dimensions à l’étude. Les résultats soulignent l’importance d’une intervention psychosociale précoce auprès des jeunes à risque et aux prises avec des symptômes dépressifs, en ciblant des aspects spécifiques pour les garçons et pour les filles. / Psychoactive substance use and problem substance use are common phenomena in adolescence. They have generated a sustained interest in Western countries in the past few decades. Research has highlighted the necessity of a model examining the joint effects of peer group and family on these phenomena. Two major theoretical trends have emerged from the literature, namely socialization and selection processes. Moreover, many studies have attempted to explain the observed comorbidity of these phenomena with depressive symptomatology, which is generally associated with a heavier prognostic. However, the nature and direction of this association remain unclear. Clarifying the underlying mechanisms is necessary to guiding prevention and intervention efforts. The first objective of this thesis is to contribute to clarify the nature of the etiology of problem substance use in adolescence. It examines how problem substance use revolves around familial, behavioral and affective factors and with substance using and deviant friend affiliations. To do so, it tests two competitive mediation models. The second objective of this thesis is to clarify the association between these different dimensions and depressive symptomatology by testing the moderator effect of depressive symptoms on the proposed mediation sequence. The third and last objective is to identify the particularities that may exist between boys and girls by testing the moderator effect of gender on the same mediation sequence. This work was carried out using a cohort from the New Approaches, New Solutions (NANS) longitudinal dataset, which comprises more than 3000 students attending school in mostly disadvantaged areas of Quebec who were followed throughout high school (2003-2007). The analyses carried out in this thesis provide empirical support to the socialization model but not to the selection model when both models are tested separately. However, when tested simultaneously, the structural crosslagged models provide no empirical support to the mediation associations predicted by both models, in favor of stability mediation associations, except for one association suggested by Patterson’s model for boys. Moderation analyses (multigroups) suggest indeed that gender has a moderator effect. Parental behavioral control appears particularly important in the etiology of problematic behaviors in boys, while familial conflicts seem more central for girls. Moderation analyses also suggest a moderator effect of depressive symptoms, but while it was expected that these symptoms would exacerbate the studied associations, none of the associations actually remained significant in the depressive group. This study therefore suggests that depressive symptomatology and problem substance use appear together in a more global profile of risk. To our knowledge, this thesis is the first to take a specific interest in the moderation effect of depressive symptoms on the associations between the different dimensions under study. The results underscore the importance of an early psychosocial intervention with adolescents at risk and with adolescents struggling with depressive symptoms, while targeting specific aspects for boys and girls.

The Role of APOBEC3 in Controlling Retroviral Spread and Zoonoses

APOBEC3 (A3) proteins are a family of host-encoded cytidine deaminases that protect against retroviruses and other viral intruders. Retroviruses, unlike other viruses, are able to integrate their genomic proviral DNA within hours of entering host cells. A3 proteins hinder retroviral infectivity by editing retroviral replication intermediates, as well as by inhibiting retroviral replication and integration through deamination-independent methods. These proteins thus constitute the first line of immune defense against endogenous and exogenous retroviral pathogens. The overall goal of my Master's project was to better understand the critical role A3 proteins play in restricting inter- and intra-host transmission of retroviruses. There are two specific aspects that I focused on: first, investigating the role of mouse APOBEC3 (mA3) in limiting the zoonotic transmission of murine leukemia retroviruses (MLVs) in a rural environment; second, to identify the molecular features in MLVs that confer susceptibility or resistance to deamination by mA3. For the first part of my project, we collected blood samples from dairy and production cattle from four different geographical locations across Canada. We then designed a novel PCR screening strategy targeting conserved genetic regions in MLVs and Mouse Mammary Tumor Virus (MMTV) and MMTV-like betaretroviruses. Our results indicate that 4% of animals were positive for MLV and 2% were positive for MMTV. Despite crossing the species barrier by gaining entry into bovine cells, our study also demonstrates that the bovine A3 protein is able to potently inhibit the spread of these murine retroviruses in vitro. The next question we asked was whether mA3 could also mutate and restrict murine endogenous retroviruses and thereby partake in limiting zoonotic transmission. Moloney MLV and AKV MLV are two highly homologous murine gammaretroviruses with opposite sensitivities to restriction by mA3: MoMLV is resistant to restriction and deamination while AKV is sensitive to both. Design of MoMLV/AKV hybrid viruses enabled us to map the region of mA3 resistance to the region encoding the glyco-Gag accessory protein. Site-directed mutagenesis then allowed us to correlate the number of N-linked glycosylation sites with the level of resistance to deamination by mA3. Our results suggest that Gag glycosylation is a possible viral defence mechanism that arose to counteract the evolutionary pressure imposed by mA3. Overall, my projects show the important role A3 proteins play in intrinsic immunity, whether defending the host from foreign retroviral invaders or endogenous retroviral foes.

Aberrations in Cytokine Signaling in Leukemia: Variations in Phosphorylation and O-GlcNAcylation

Tumor-induced immunosuppression can occur by multiple mechanisms, each posing a significant obstacle to immunotherapy. Evidence presented in this dissertation suggests that aberrant cytokine signaling, as a result of altered metabolism of Chronic Lymphocytic Leukemia (CLL) cells, confers a selective advantage for tumor survival and growth. Cells from CLL patients with aggressive disease (as indicated by high-risk cytogenetics) were found to exhibit prolongation in Interferon (IFN)-induced STAT3 phosphorylation, and increased levels of reactive oxygen species (ROS) in these cells reflected these signaling processes. Changes in the relative balance of phospho-STAT3 and phospho-STAT1 levels, in response to combinations of IL-2 + Toll-like receptor (TLR)-7 agonist + phorbol esters, as well as IFN, were associated with the immunosuppressive and immunogenic states of CLL cells. In addition, immunosuppressive leukemic cells were found to express high levels of proteins with O-linked N-acetylglucosamine (O-GlcNAc) modifications, due to increased metabolic activity through the Hexosamine Biosynthetic Pathway (HBP), which caused impaired intracellular signaling responses and affected disease progression. A conclusion of the studies presented here is that the intrinsic immunosuppressive properties of leukemic cells may be overcome by agents such as Resveratrol that target metabolic pathways of these cells.
