Isoleringsegenskaper hos frodvuxen gran : En studie av dess värmeledningsförmåga och funktion / Fast grown spruce insulation properties : A study of its heat conductivity and usability

Denna studie undersöker potentialen för frodvuxen gran (Picea abies) som isoleringsmaterial i historiska byggnader, med syftet att utvärdera sambandet mellan densitet och värmeledningsförmåga. Bakgrunden till studien ligger i behovet av hållbara och effektiva isoleringsmaterial som kan användas för att förbättra energiprestandan i byggnader samtidigt som de bevarar deras historiska karaktär och estetiska värde. Frodvuxen gran har växt snabbt vilket lett till en lägre densitet än lägsta värdet som finns att tillgå i den svenska standarden för olika materials värmeledningsförmågor.  Metodologin innefattade både laboratorieanalyser och fältstudier. I laboratoriedelen användes ett hot-disk instrument för att mäta värmeledningsförmåga sedan beräknades densitet och fuktkvot. Fältstudierna genomfördes för att identifiera praktiska tillämpningar och utmaningar vid användning av frodvuxen gran som konstruktionsmaterial. Dessa studier omfattade inspektioner av byggnader uppförda med frodvuxen gran och intervju med ansvarig för uppförandet av dessa byggnader för att utreda dess potentiella funktion som konstruktionsmaterial.  Huvudresultaten från laboratorieanalyserna visade ett starkt samband mellan densitet och värmeledningsförmåga, vilket indikerar att frodvuxen gran med sin lägre densitet har förbättrade isoleringsegenskaper jämfört med senvuxen gran. Fältstudierna bekräftade att frodvuxen gran kan användas som konstruktionsmaterial i byggnader, men pekade också på dess benägenhet till större rörelser vid uttorkning vilket dock går att räkna på för att undvika att problem uppstår vid torkning.  Slutsatserna visar att frodvuxen gran kan vara ett värdefullt tillskott till befintliga isoleringsmaterial tack vare dess förbättrade isoleringsförmåga. Studien rekommenderar vidare forskning för att undersöka långsiktiga nedbrytningseffekter och materialets fuktegenskaper, vilket är avgörande för dess praktiska användning i byggnader. Vidare bör forskningen även studera frodvuxen grans förmåga som putsbärare, både för ler- och kalkbruk, för att utreda virkets kompatibilitet med dessa material. / This study investigates the potential of fast-grown spruce (Picea abies) as an insulation material in historic buildings, aiming to evaluate the relationship between density and thermal conductivity. The background of the study lies in the need for sustainable and efficient insulation materials that can be used to improve the energy performance of buildings while preserving their historical character and aesthetic value. Fast-grown spruce has a lower density then the lowest available in the Swedish standard for materials heat conductivity.  The methodology included both laboratory analyses and field studies. In the laboratory part, a hot-disk instrument was used to measure thermal conductivity, followed by calculations of density and moisture content. The field studies were conducted to identify practical applications and challenges in using fast-grown spruce as a construction material. These studies involved inspections of buildings constructed with fast-grown spruce and interviews with those responsible for their construction to investigate its potential functionality as a construction material.  The main results from the laboratory analyses showed a strong correlation between density and thermal conductivity, indicating that fast-grown spruce, with its lower density, has improved insulation properties compared to slow-grown spruce. The field studies confirmed that fast-grown spruce can be used as a construction material in buildings, but also pointed out its tendency to exhibit greater movement during drying, which can be accounted for to prevent issues during the drying process.  The conclusions suggest that fast-grown spruce can be a valuable addition to existing insulation materials due to its improved insulation performance. The study recommends further research to examine long-term degradation effects and the material's moisture properties, which are critical for its practical use in buildings. Additionally, further research should investigate the ability of fast-grown spruce to serve as a substrate for plaster, both for clay and lime-based plasters, to determine the wood's compatibility with these materials.

Étude du mécanisme de protection des spermatozoïdes de mammifères par le lait

Le lait écrémé est utilisé depuis plus d’un demi-siècle comme diluant protecteur des spermatozoïdes de mammifères. Depuis quelques années, il existe une demande grandissante pour des diluants exempts de produits d’origine animale. Toutefois, le mécanisme par lequel le lait protège les spermatozoïdes n’est pas connu, ce qui rend difficile de lui trouver un substitut. Les protéines majeures du plasma séminal de taureau, les protéines « Binder of SPerm » (BSP), sont néfastes lors de la conservation de la semence. Les spermatozoïdes sont en contact avec une grande concentration de protéines BSP qui stimulent une extraction continuelle de cholestérol/phospholipides de leur membrane plasmique. Les lipoprotéines de faible densité (LDL) du jaune d’oeuf, un autre composé utilisé dans les diluants, empêcheraient les protéines BSP de se lier à la membrane des spermatozoïdes de taureaux et de stimuler un efflux des lipides membranaires, ce qui les protégerait durant la conservation. Notre hypothèse était que les protéines du lait protègent les spermatozoïdes durant la conservation en séquestrant les protéines BSP. Premièrement, nous avons démontré par filtration sur gel qu’il y a une interaction entre les protéines BSP bovines et les protéines du lait. Le lait écrémé a été fractionné en trois fractions : F1 (alpha-lactalbumine, bêta-lactoglobuline et caséine kappa), F2 (toutes les protéines du lait) et F3 (sels, sucres et petits peptides). Les protéines BSP1 et BSP5 ont une affinité plus grande pour F1 que BSP3, tandis que toutes les protéines BSP ont une affinité pour F2. Le titrage calorimétrique isotherme a permis de confirmer l’interaction entre les protéines BSP et les protéines du lait. L’association entre la protéine BSP1 bovine et les micelles de caséines est caractérisée par une constante d’affinité (Ka) de 3.5 × 10^5 M-1 et un paramètre stoichiométrique (n) de 4,5 BSP1 pour une caséine. L’association entre la protéine BSP1 bovine et l’alpha-lactalbumine (une protéine du sérum principale), est caractérisée par un Ka de 2.4 × 10^5 M-1 et une valeur “n” de 0,8. Ces résultats indiquent que le lait protège les spermatozoïdes bovins en séquestrant les protéines BSP grâce à une interaction protéine : protéine, tandis que le jaune d’oeuf les protège grâce à une interaction protéine : lipoprotéine. Deuxièmement, nous avons démontré par filtration sur gel que les protéines homologues aux BSP bovines retrouvées dans le plasma séminal de porc, d’étalon et de bélier ont une affinité avec les protéines du lait, ce qui suggère que le mécanisme de protection des spermatozoïdes par le lait pourrait être le même chez ces espèces. Troisièmement, nous avons caractérisé l’interaction entre BSP1 bovine et les LDL du jaune d’oeuf qui a un Ka de 3.4 ± 0.4 × 10^6 M-1 et une valeur de « n » de 104 BSP1 pour une particule de LDL, indiquant qu’il existe des différences entre le mécanisme de protection des spermatozoïdes par le lait et le jaune d’oeuf. Nous croyons que les résultats présentés dans cette thèse aideront à créer de nouveaux diluants ne contenant pas de produits d’origine animale afin de cryoconserver les spermatozoïdes des mammifères. / Skim milk is being used as a protective agent for mammalian semen conservation over half a century. Recently, there has been increased interest in developing extenders free of animal products. However, it is difficult to find suitable component in order to replace milk as an extender, because the mechanisms by which milk protect sperm against cooling and freezing damages during the storage is unknown. The Binder of SPerm (BSP) proteins are the major proteins of bull seminal plasma and they are harmful during sperm storage. In fact, sperm would be in contact with a large quantity of BSP proteins that induce a continuous cholesterol and phospholipids efflux from the sperm membrane during storage. When bull sperm is diluted with an extender containing egg yolk, another compound frequently used in extender, the low-density lipoproteins (LDL) present in the egg yolk prevent the binding of the BSP proteins to the sperm membrane, thus, preventing the lipid efflux from the sperm membrane induced by the BSP proteins. Our hypothesis was that milk proteins would protect sperm during storage by binding BSP proteins. First, we demonstrated by gel filtration that bovine BSP proteins could bind the milk proteins. Skim milk was fractionated into three fractions: F1 (alpha-lactalbumin and beta- lactoglobulin, the major whey proteins and kappa-casein), F2 (mainly caseins and all other milk proteins in small amounts) and F3 (salts, sugars and small peptides). Bovine BSP1 and BSP5 have more affinity for F1 as compared to BSP3 and all the BSP proteins have affinity for F2. We confirmed the interaction between bovine BSP proteins and milk proteins by isothermal titration calorimetry. The binding of BSP1 to casein micelles is characterized by an affinity constant (Ka) of 3.5 × 10^5 M-1 and of a stoichiometric parameter for the association (n) of 4.5 BSP1 per casein. The association between BSP1 and alpha-lactalbumin (one of the major whey proteins) is characterized by a Ka of 2.4 × 10^5 M-1 and a “n” value of 0.8. These results support our contention that milk can protect sperm by preventing the BSP proteins’ binding to the sperm membrane attributable to a protein : protein interaction, while egg yolk sperm protection is attributable to a protein : lipoprotein interaction. Second, our studies showed that the homologous BSP proteins found in the boar, stallion and ram seminal plasma can bind the milk proteins. These results indicate that the mechanism of sperm protection by milk in these species should be similar to the one in bovine species. Third, we characterized the interaction between bovine BSP1 protein and LDL from hen’s egg yolk. The binding was characterized by a Ka of 3.4 ± 0.4 × 10^6 M-1 and a « n » value of 104 BSP1 per LDL particle. Our results indicated that there is difference between the mechanism of sperm protection by milk and egg yolk. We believe that the results presented in this thesis may help to create new extenders free of animal product for mammal sperm preservation in liquid or frozen state.

Le récepteur CD36 : implication dans le développement de l'athérosclérose et dans le recrutement des leucocytes aux sites inflammatoires

Le CD36 est un récepteur éboueur de classe B exprimé par plusieurs types cellulaires dont les macrophages et les cellules endothéliales de la microvasculature. Le CD36 présente une haute affinité de liaison pour les ligands lipidiques tels que les lipoprotéines oxydées de basse densité (LDLox). De part sa capacité à internaliser les LDLox au niveau des macrophages et de son implication dans la formation des cellules spumeuses, le CD36 joue un rôle critique dans le développement des lésions athérosclérotiques. Nous avons testé l'hypothèse selon laquelle le EP 80317, un ligand synthétique sélectif du CD36, exerce des effets anti-athérosclérotiques chez les souris déficientes en apolipoprotéine E. Un traitement prolongé (12 semaines) avec le EP 80317 réduit fortement (de 51%) la surface des lésions athérosclérotiques par comparaison aux souris témoins. L'effet anti-athérosclérotique est associé à une diminution des taux de cholestérol plasmatique, à une réduction de l’internalisation des LDLox au niveau des macrophages et à une augmentation de l’expression des protéines impliquées dans le transport inverse du cholestérol. De plus, un traitement par le EP 80317 est également associé une diminution de l’expression aortique et plasmatique de protéines pro-inflammatoires. Nos études ont aussi montré un rôle pour le CD36 dans le recrutement des phagocytes mononucléés au niveau des lésions athérosclérotiques, tel que démontré par une réduction de l’accumulation des phagocytes mononucléés radiomarqués CD36–/– par rapport aux cellules CD36+/+. À l’échelle moléculaire, nous avons montré que les phospholipides oxydés induisent la phosphorylation de la kinase Pyk2 des podosomes des monocytes/macrophages de manière dépendante de l’expression du CD36 et de Src. Cette phosphorylation est atténuée par un traitement par le EP80317. Nos résultats appuient le rôle important du CD36 dans l’athérosclérose et suggèrent que les ligands synthétiques qui modulent la fonction du CD36 représentent potentiellement une nouvelle classe d'agents anti-athérosclérotiques. Le CD36 exprimé par les cellules endothéliales de la microvasculature est un récepteur de l’hétérodimère protéique S100A8/A9. Ces protéines s’associent à l’acide arachidonique intracellulaire (AA) des neutrophiles polymorphonucléaires (PMN) et le complexe S100A8/A9/AA peut être sécrété par les PMN activés au contact de l’endothélium. Nous avons vérifié l’hypothèse selon laquelle le CD36 exprimé par la microvasculature est impliqué dans le métabolisme transcellulaire de l’AA par la liaison du complexe S100A8/A9/AA et la réponse inflammatoire. Chez deux modèles murins d'inflammation aiguë (ischémie/reperfusion des membres inférieurs et poche d’air dorsale), nous avons observé que la réponse inflammatoire, notamment l’accumulation des PMN au niveau des sites inflammatoires, est diminuée en moyenne de 63% chez les souris CD36-/-. De même, un traitement par le EP 80317 ou par les anticorps anti-S100A8/A9 diminue chacun de 60% en moyenne l’extravasation des PMN vers les tissus inflammatoires. L’administration simultanée des deux traitements n’a aucun effet supplémentaire, et ces traitements n’exercent aucun effet chez les souris CD36-/-. Nos résultats appuient le rôle du récepteur CD36 de la microvasculature dans la régulation de la réponse inflammatoire. L’utilisation des ligands synthétiques du CD36 pourrait représenter une nouvelle avenue thérapeutique dans le traitement des réponses inflammatoires aiguës. / CD36 is a class B scavenger receptor expressed by multiple cell types such as macrophages and microvascular endothelial cells. CD36 shows a high affinity binding towards lipid-based ligands such as oxidized low-density lipoproteins (oxLDL). Macrophage CD36 has been shown to play a critical role in the development of atherosclerotic lesions by its ability to internalize oxLDL and to lead to foam cell formation. We tested the hypothesis that EP 80317, a selective CD36 ligand, exerts anti-atherosclerotic effects in apolipoprotein E-deficient (apoE–/–) mice fed on atherogenic diet. Long term treatment (12 weeks) with EP 80317 results in a striking reduction (51%) of lesion areas in EP 80317-treated apoE–/– mice. This effect was associated with a decrease in plasma cholesterol, a reduced oxLDL internalization within macrophages and an up-regulation of proteins involved in cholesterol efflux. Additionally, treatment with EP 80317 was associated with a reduced expression of vascular and plasma pro-inflammatory proteins. Our studies also showed a role of CD36 in modulating the recruitment of mononuclear phagocytes to the arterial wall, as shown by a reduced migration of radiolabeled CD36-/- macrophages into atherosclerotic lesions compared to CD36+/+ cells. At the molecular level, our studies showed that oxidized phospholipids induced the phosphorylation of the adhesion kinase Pyk2 in monocytes/macrophages, in a CD36- and Src-dependent manner. The Pyk2 phosphorylation is attenuated by treatment with EP80317. Our results strongly support the role of CD36 in atherosclerosis development and suggest that synthetic ligands featuring modulatory effect on CD36 function may represent a novel class of anti-atherosclerotic agents. CD36 expressed by microvascular endothelial cells is a receptor for the heterodimer S100A8/A9. These proteins bind intracellular arachidonic acid (AA) within polymorphonuclear neutrophils (PMN) and the complex S100A8/A9-AA may be secreted at sites of inflammation where it exerts chemotactic activities. We aimed to delineate the role of microvascular CD36, as a receptor for the S100A8/A9, in the AA transcellular metabolism and the regulation of the associated PMN trafficking to inflammatory sites. In two mouse models of acute inflammation (hind limb ischemia/reperfusion and dorsal air pouch), CD36 regulated trafficking of PMN to inflammatory sites, as shown by a mean of 63% reduction of PMN accumulation in CD36-/- mice. Treatment with EP 80317 or with S100A8/A9 antibodies reduced, each by ~ 60%, the recruitment of PMN to inflammatory sites. The combined administration of anti-S100A8/A9 and EP 80317 did not exert any additional inhibitory effect and neither treatment featured a modulatory effect in CD36-/- mice. Our results strongly support a role for microvascular CD36 in regulating PMN trafficking to inflammatory sites. Targeting CD36 might represent a novel therapeutic avenue for the treatment of acute inflammatory responses.

Generalized belief propagation based TDMR detector and decoder

Two dimensional magnetic recording (TDMR) achieves high areal densities by reducing the size of a bit comparable to the size of the magnetic grains resulting in two dimensional (2D) inter symbol interference (ISI) and very high media noise. Therefore, it is critical to handle the media noise along with the 2D ISI detection. In this paper, we tune the generalized belief propagation (GBP) algorithm to handle the media noise seen in TDMR. We also provide an intuition into the nature of hard decisions provided by the GBP algorithm. The performance of the GBP algorithm is evaluated over a Voronoi based TDMR channel model where the soft outputs from the GBP algorithm are used by a belief propagation (BP) algorithm to decode low-density parity check (LDPC) codes.

Surface-Engineered Magnetic Nanoparticles for Sample Preparation and Analysis of Proteins and Peptides

Sample preparation as an essential step in mass spectrometry-based analysis, plays a critical role in proteomics studies. Magnetic nanoparticles (MNPs) have been widely used in protein and peptide sample preparation due to their magnetic properties, biocompatibility, easy synthesis and surface functionalization. MNPs loaded with analyte or analyte modification reagent can be easily separated from the reaction medium by an externally applied magnetic field. The small size of MNPs provides high analyte loading and extraction capacity. Additionally, MNP can be decorated with different functional groups to achieve selective modification or extraction of analyte. In this study we have utilized silica coated iron oxide magnetic nanoparticles (Fe3O4@SiO2 MNPs) for protein and peptide sample preparation. Fluorescence-based methods were utilized for quantitative and qualitative characterization of N-hydrosucccinimidyl (NHS) ester groups on the surface of Fe3O4@SiO2 MNPs. Fluorophore Dansylcadaverine was conjugated to NHS ester functional groups. Fluorometric measurement of cleaved dansylcadaveine was employed to determine the number of NHS ester groups per MNPs that was found to be 2.6 × 102 and 3.4 × 103for 20 nm and 100 nm Fe3O4@SiO2 MNPrespectively. The efficiency of labeling native bovine serum albumin (BSA) by NHS ester coated Fe3O4@SiO2 MNPs was also explored in terms of maximizing the number of MNPs conjugated per BSA molecule or maximizing the number of BSA molecules conjugated per each MNP. Lysine residues of apolipoprotein B-100 (apoB-100) on the surface of intact human low density lipoprotein (LDL) were labeled by NHS ester modified Fe3O4@SiO2 MNPs in aqueous solvents at room temperature. The MNP labeledapoB-100 was treated by SDS to remove lipids and then digested using trypsin. Tryptic peptides were eluted from MNPs by cleaving disulfide linkage between labeled peptides and MNPs. LC-MS/MS analysis found 28 peptides containing labeled lysine residues. These lysine residues should be on the solvent exposed surface of LDL since the large size of MNPs prevents contact of the labeling reagent to those lysines embedded inside the structure of LDL. TCEP- immobilized Fe3O4@SiO2MNPs were fabricated and utilized for reduction of disulfide bonds in bovine pancreas insulin and two different cyclic peptides. Disulfide bonds were efficiently cleaved at room temperature in both organic and aqueous solvents confirmed by LC-MS/MS analysis of reduced/alkylated protein and peptides. Disulfide reduction and alkylation reactions was performed in one step and the reducing agent was simply separated from peptide and protein solution by magnetic separation.

Viabilidade de operação do duplo semi-reboque de cinco eixos no Brasil / Operation feasibility of the five axle twin trailer truck in Brazil

Este trabalho investiga a viabilidade de se implantar no Brasil o veículo combinado de cinco eixos constituído de um cavalo mecânico mais dois semi-reboques acoplados por um dolly intermediário e chamado aqui de Duplo Semi-Reboque (DSR). Essa investigação inclui tanto aspectos econômicos (custos operacionais) quanto operacionais (densidade das cargas, rotas, demandas, veículos utilizados), tecnológicos (veículos e implementes), legais (pesos e dimensões), de segurança (estabilidade, dirigibilidade, risco de acidentes e impacto sobre o tráfego de outros veículos) e de inter-relação com a infra-estrutura rodoviária. Como a configuração analisada é largamente utilizada nos Estados Unidos, com o nome de twin trailer truck, faz-se uma ampla revisão da literatura norte-americana. O estudo apresenta as características técnicas do twin, levanta a legislação norte-americana de carga por eixo e mostra como a busca de produtividade e o aumento da carga volumosa conduziram à necessidade de veículos mais longos. Compila artigos sobre a estabilidade, a dirigibilidade e o impacto sobre o tráfego dos veículos longos combinados. Sintetiza pesquisas comparando taxas de acidentes dos veículos combinados com os caminhões convencionais. A maioria desses estudos conduz à conclusão de que, embora os twins possam degradar ligeiramente as condições de segurança de tráfego e aumentar o desgaste elos pavimentos, essa degradação é compensada pela redução do número de viagens e do custo do transporte. Conclui-se que o veículo seria economicamente viável no Brasil para substituir caminhões trucados em rotas de demanda limitada, e que poderia também economizar custos na substituição a veículos convencionais no transporte de cargas volumosas, sem provocar maiores impactos sobre a segurança operacional. Pesquisa com frotistas mostra que quase 50% das transportadoras de carga fracionada operam cargas de densidade compatível com o DSR. A introdução do veículo, no entanto, enfrenta fortes obstáculos legais e exigiria mudança profunda na legislação dos veículos combinados longos. / This paper investigates the feasibility of introducing the American twin trailer truck in the Brazilian road transport network. The twin is a five axle configuration with one tractor and two semi-trailers, coupled by an intermediate dolly. The investigation embraces not only economic questions (operational costs), but also operational aspects (freight density and volume, vehicle choice, line extension etc), technological questions (vehicles, trailers and dollies), weights and dimensions legislation, safety impacts (handling and stability, impact of traffic operations and statistic studies of accident rates) and the twin\'s interface with the highway facilities. The American and Canadian studies about twins are reviewed. The paper presents the technical characteristics of twins, the american weights and dimensions legislation and shows how the claim for more productivity in the transport industry and the low density freight growth have induced the use of longer combinations. The study compiles the main results presented in the literature about handling, stability and the twin trailer impact on the highway traffic and analyzes statistics comparing doubles with semi-trailer accident rates. The literature review leads to the conclusion that, althrough the twins could degrade slightly the vehicle traffic safety performance and accelerate the pavement wear, this degradation is overwhelmed by the reduction of the number of vehicles on the roads and direct and indirect transportation costs. The introduction of the win trailer truck in Brazil could be advantageous in order to replace tandem trucks on low demand routes. The savings in terminal times and the higher capacitiy of this configuration would cut costs as compared to the straight truck, semi-trailer and truck-and-trailer configurations, without relevant deterioration of the operational safety. The results of a large sample with brazilian LTL companies show that almost 50% of them operate with a freight density that would be ideal for twins. The twin\'s introduction, however, faces strong legal impediments in some markets, and only would be feasible if deep changes in the Brazilian weight and dimension regulations would be undertaken.

Essa investigação inclui tanto aspectos econômicos (custos operacionais) quanto operacionais (densidade das cargas, rotas, demandas, veículos utilizados), tecnológicos (veículos e implementes), legais (pesos e dimensões), de segurança (estabilidade, dirigibilidade, risco de acidentes e impacto sobre o tráfego de outros veículos) e de inter-relação com a infra-estrutura rodoviária. Como a configuração analisada é largamente utilizada nos Estados Unidos, com o nome de twin trailer truck, faz-se uma ampla revisão da literatura norte-americana. O estudo apresenta as características técnicas do twin, levanta a legislação norte-americana de carga por eixo e mostra como a busca de produtividade e o aumento da carga volumosa conduziram à necessidade de veículos mais longos. Compila artigos sobre a estabilidade, a dirigibilidade e o impacto sobre o tráfego dos veículos longos combinados. Sintetiza pesquisas comparando taxas de acidentes dos veículos combinados com os caminhões convencionais. A maioria desses estudos conduz à conclusão de que, embora os twins possam degradar ligeiramente as condições de segurança de tráfego e aumentar o desgaste elos pavimentos, essa degradação é compensada pela redução do número de viagens e do custo do transporte. Conclui-se que o veículo seria economicamente viável no Brasil para substituir caminhões trucados em rotas de demanda limitada, e que poderia também economizar custos na substituição a veículos convencionais no transporte de cargas volumosas, sem provocar maiores impactos sobre a segurança operacional. Pesquisa com frotistas mostra que quase 50% das transportadoras de carga fracionada operam cargas de densidade compatível com o DSR. A introdução do veículo, no entanto, enfrenta fortes obstáculos legais e exigiria mudança profunda na legislação dos veículos combinados longos. / This paper investigates the feasibility of introducing the American twin trailer truck in the Brazilian road transport network. The twin is a five axle configuration with one tractor and two semi-trailers, coupled by an intermediate dolly. The investigation embraces not only economic questions (operational costs), but also operational aspects (freight density and volume, vehicle choice, line extension etc), technological questions (vehicles, trailers and dollies), weights and dimensions legislation, safety impacts (handling and stability, impact of traffic operations and statistic studies of accident rates) and the twin\'s interface with the highway facilities. The American and Canadian studies about twins are reviewed. The paper presents the technical characteristics of twins, the american weights and dimensions legislation and shows how the claim for more productivity in the transport industry and the low density freight growth have induced the use of longer combinations. The study compiles the main results presented in the literature about handling, stability and the twin trailer impact on the highway traffic and analyzes statistics comparing doubles with semi-trailer accident rates. The literature review leads to the conclusion that, althrough the twins could degrade slightly the vehicle traffic safety performance and accelerate the pavement wear, this degradation is overwhelmed by the reduction of the number of vehicles on the roads and direct and indirect transportation costs. The introduction of the win trailer truck in Brazil could be advantageous in order to replace tandem trucks on low demand routes. The savings in terminal times and the higher capacitiy of this configuration would cut costs as compared to the straight truck, semi-trailer and truck-and-trailer configurations, without relevant deterioration of the operational safety. The results of a large sample with brazilian LTL companies show that almost 50% of them operate with a freight density that would be ideal for twins. The twin\'s introduction, however, faces strong legal impediments in some markets, and only would be feasible if deep changes in the Brazilian weight and dimension regulations would be undertaken.

Experimental and numerical investigation of the mechanical behaviour of dissimilar arc and spot welds of advanced high strength steels / Etude expérimentale et numérique du comportement mécanique de soudures d'arc et de points dissemblables d'aciers avancés à très haute résistance

De nos jours, la politique écologique encourage les constructeurs automobiles à réduire le poids global du véhicule. Des tôles d'acier fines d'épaisseur différente optimisant chaque partie de l'assemblage sont utilisées et les sidérurgistes développent des aciers de plus en plus résistants à savoir l'Acier Haute Résistance Avancé (AHSS) avec un bon compromis entre résistance mécanique et ductilité (emboutissage). Lors des essais mécaniques de soudage hétérogène AHSS, des modes de fractures inhabituels sont observés, notamment le long de l'interface entre la zone affectée par la chaleur (ZAT) et la zone de fusion ou zone fondue (ZF). Ces fractures se produisent généralement avec une résistance inférieure à celle attendue pour ces soudures. Les objectifs de l'étude sont de comprendre les mécanismes de rupture au cours des essais mécaniques et de créer un modèle mécanique de FE conçu pour prédire la résistance mécanique des assemblages soudés. Tout d'abord, une étude de soudage hétérogène constituée de deux nuances d'acier bien connues d'ArcelorMittal vise à comprendre le mécanisme de défaillance et les paramètres affectant les modes de défaillance. Différentes configurations sont étudiées avec l'épaisseur. Le modèle FE est construit avec une réponse mécanique identifiée de chaque zone (matériaux de base, zones affectées par la chaleur et zone de fusion), en utilisant des modèles d'ArcelorMittal et des données expérimentales. Des critères de défaillance basés sur des dommages ductiles tenant compte de l'influence de la triaxialité sont utilisés et certains éléments cohésifs sont utilisés pour simuler une défaillance interfaciale. Deux configurations d'essais mécaniques dans le cas du soudage par résistance par points (essais de traction transversale et de traction) sont considérées. Les prédictions du modèle étaient très précises avec les modes de défaillance et les forces expérimentaux. Ensuite, cette méthode de modélisation FE a été appliquée avec succès à un boîtier de soudage par points très hétérogène comprenant un nouveau concept AHSS basse densité de troisième génération à forte teneur en aluminium et en manganèse. Les modes d'échec et les forces obtenues étaient comparables. De plus, la méthode de modélisation FE a été appliquée sur des configurations plus complexes, en particulier sur un assemblage soudé par points triple épaisseur. La robustesse du modèle pour prédire les modes de défaillance partielle et les forces d'une soudure par points triple épaisseur a été démontrée. En outre, la méthodologie de modélisation FE a été étendue à un autre type de soudage: le soudage à l'arc. Dans ce cas, deux feuilles sont soudées en configuration de chevauchement ab avec un fil d'apport. Le modèle FE permet de prédire la zone de rupture et la résistance de l'assemblage soudé. / Nowadays, ecological policy encourages carmakers to reduce the global vehicle weight. Fine steel sheets assemblies with different thickness optimizing each part of the assembly are used and steelmakers develop steels which are more and more resistant namely Advanced High Strength Steel (AHSS) with a good compromise between mechanical strength and ductility (stamping). During the mechanical tests of heterogeneous AHSS welding, unusual fracture modes are observed, in particular along the interface between the Heat Affected Zone (HAZ) and the Fusion Zone or molten zone (FZ). These fractures generally occur with lower strength than expected for these welding. The objectives of the study are to understand fracture mechanisms during mechanical testing and create a mechanical FE model is developed to be able to predict mechanical strength of the welded assemblies. Firstly, a study of heterogeneous welding constituted of two well-known steel grades of ArcelorMittal aims at understanding failure mechanism and parameters affecting the failure modes. Different configurations are studied with thickness. FE model is built with mechanical response identified of each zone (base materials, heat affected zones and fusion zone), using ArcelorMittal models and experimental data. Failure criteria based on ductile damage taking into account the influence of the triaxiality are used and some cohesive elements are used to simulate interfacial failure. Two configurations of mechanical testing in the case of Resistance Spot Welding (cross tension and tensile shear tests) are considered. Model predictions were very accurate with experimental failure modes and strengths. Then, this FE modelling method was successfully applied to a highly heterogeneous spot welding case including a new third generation low density AHSS concept with high aluminum and manganese content. Failure modes and strengths obtained were comparable. Moreover, FE modelling method was applied on more complex configurations, in particular on a triple thick spot welded assembly. The robustness of the model to predict partial failure modes and strengths of a triple thick spot weld has been demonstrated. In addition, FE modelling methodology was extended to another welding type: arc welding. In this case, two sheets are welded in ab overlap configuration with a filler wire. FE model allows predicting the failure zone and strength of welded assembly.

Molecular mechanisms involved in the protective effect of Mediterranean diet and olive oil consumption in humans

The scope of the present work was to investigate whether the protective role of the traditional Mediterranean diet (TMD), and virgin olive oil (VOO) rich in phenolic compounds (PC), towards cardiovascular disease can be mediated through gene expression changes. Two trials were performed to assess the in vivo nutrigenomic effects of TMD and VOO in healthy volunteers. The results point out: a) significant gene expression changes of those genes related with cardiovascular-risk processes after VOO ingestion; b) a down-regulation in the expression of atherosclerosis-related genes after a 3-month intervention with a TMD; and c) an olive oil PC health-protective nutrigenomic effect within the frame of the TMD. Changes in gene expression were concomitant with decreases in oxidative damage and systemic inflammation markers. Data from our studies provide further evidence to recommend both the TMD and the VOO as a useful tool for the prevention of atherosclerosis. / El objetivo de este estudio es investigar si el papel protector de la dieta Mediterránea tradicional (TMD) y del aceite de oliva virgen (VOO), rico en compuestos fenólicos (PC), puede ser mediado a través de cambios en la expresión génica. Se realizaron dos ensayos clínicos para evaluar los efectos nutrigenómicos de la TMD y del VOO, in vivo, en voluntarios sanos. Los resultados mostraron a) cambios en la expresión génica de genes relacionados con el riesgo cardiovascular tras la ingestión del aceite virgen de oliva, b) una infra-expresión en la expresión de genes relacionados con el proceso aterosclerótico tras una intervención con TMD de 3 meses y c) que los compuestos fenólicos del aceite de oliva ejercen un efecto nutrigenómico protector en el marco de la TMD. Los cambios en la expresión génica fueron coherentes.
