Efeito fotobiomodulador da terapia laser de baixa intensidade na transição inflamação-reparo em modelo de lesão pulmonar / Effect of low level laser therapy in the transition between inflammation and repair in a model of lung injury

A terapia laser de baixa intensidade (LLLT) é capaz de modular a resposta inflamatória em vários modelos experimentais, reduzindo o infiltrado de células do sistema imune e a secreção de mediadores químicos. Estudos prévios sugerem sua utilidade também no tratamento da lesão pulmonar aguda (LPA), condição caracterizada por intensa reação inflamatória em vias aéreas distais. Assim, considerando os efeitos anti-inflamatórios do laser e a fisiopatologia da LPA, formulamos a hipótese de que a LLLT 660 nm, 10 J/cm2, poderia modular o processo inflamatório nessa patologia favorecendo a transição inflamação-reparo. Foram utilizados camundongos C57BL distribuídos aleatoriamente em 4 grupos: PBS- instilado com PBS intratraqueal; PBS+LLLT- irradiado com LLLT 6 horas após instilação intratraqueal de PBS; LPS- instilado com LPS (5 mg/kg); LPS+LLLT- irradiado com LLLT 6 horas após instilação intratraqueal de LPS. Observou-se que a instilação de LPS intratraqueal levou a um aumento do número de células inflamatórias tanto na região peribronquiolar quanto no lavado broncoalveolar (LBA), detectado na histologia pulmonar e pela expressão de F4/80. Houve também aumento da transcrição e secreção de citocinas (TNF-alfa, IL-1beta, IL-6, IL-10) e quimiocinas (MCP-1) (detectadas por PCR quantitativo e ELISA). A aplicação de LLLT 6 horas após a instilação de LPS causou uma significativa diminuição tanto no influxo de células inflamatórias quanto na secreção de mediadores. Em seguida, foram caracterizadas as populações de macrófagos presentes no tecido pulmonar e observou-se nos animais do grupo LPS+LLLT, comparados ao grupo LPS, uma diminuição na expressão de mRNA de HIF-1alfa e aumento para arginase 1, sugerindo um predomínio da população de macrófagos M2. Esse efeito da LLLT foi também testado em populações de macrófagos peritoneais estimulados com LPS em cultura, porém, embora houvesse diminuição da produção de citocinas e quimiocinas, não foram observadas diferenças nos marcadores de polarização dessas células / A low-level laser therapy (LLLT) is able to modulate the inflammatory response in several experimental models, reducing the infiltration of immune system cells and the secretion of chemical mediators. There are some previous studies suggesting also useful in the treatment of acute lung injury (ALI), a condition characterized by an intense inflammatory reaction in the distal airways. Considering the anti-inflammatory effects of laser and the pathophysiology of ALI, we hypothesized that LLLT 660 nm, 10 J/cm2, could modulate the inflammatory process in this disease favoring the transition between inflammation and repair. C57BL mice were divided into four groups: PBS - intratracheal instillation of PBS; PBS+LLLT - irradiated with LLLT 6 hours after intratracheal instillation of PBS; LPS - intratracheal instillation of LPS (5 mg/kg); LPS+LLLT - irradiated with LLLT 6 hours after intratracheal instillation of LPS. Animals were sacrificed 24 hours after injury. Intratracheal LPS inoculation induced a marked increase in the number of inflammatory cells in perivascular and alveolar spaces (detected by lung histology and in bronchoalveolar lavage), and F4/80 expression (macrophage marker). There was also an increase in the expression and secretion of cytokines (TNF-alfa, IL-1beta, IL-6, IL-10) and chemokine (MCP-1) detected by ELISA and quantitative PCR. The LLLT application 6 hours after LPS induced a significant decrease in both of the inflammatory cells influx and mediators secretion. Then, we characterized the macrophages populations in lung tissue and we observed that in the LPS+LLLT group the expression of HIF-1alfa mRNA was reduced while arginase 1 was increased, which suggested that the M2 macrophages were predominant. The LLLT effect was also tested on peritoneal macrophage population in culture stimulated with LPS and there was decreased of cytokines and chemokine production, however any difference in polarization markers was found

Stably stratified atmospheric boundary layer: study trough large-eddy simulations, mesoscale modelling and observations

La capa límit atmosfèrica és l'àrea directament influenciada per la presència de la superfície de la terra i la seva alçada és d'uns centenars de metres a uns pocs quilòmetres. Durant el vespre, el refredament radiatiu estratifica establement l'aire prop del sòl i es forma el que es coneix com a Capa Límit Estable (CLE). D'avui en dia, la CLE és un règim que encara no està prou ben caracteritzat. La turbulència, que no és homogènia ni isòtropa, i la gran importància dels efectes locals com l'orografia, entre d'altres factors, dificulten l'estudi d'aquest règim. Per aquest motiu, la CLE és objecte d'especial atenció, sobretot a l'hora de millorar la seva representació en models tant de temps com de clima.Aquest treball es centra en l'estudi de la CLE mitjançant 3 eines diferents: 1) simulacions explícites de grans remolins (més conegudes com a simulacions LES), per determinar el comportament dels moviments turbulents, on les resolucions són de l'ordre de metres; 2) simulacions mesoscalars, per caracteritzar els efectes locals, on les resolucions són de l'ordre de kilòmetres; 3) anàlisi de les observacions sota aquestes condicions per tal de caracteritzar i entendre millor els fenòmens observats.En primer lloc s'estudia el rang d'estabilitats a on el model LES, que considera la teoria de Kolmogorov per la dissipació de l'energia, funciona correctament. Els resultats del model són realistes tal com mostra la seva comparació amb les mesures de dues campanyes experimentals (SABLES-98 i CASES-99). Per explorar més a fons els resultats LES i per comparar-los amb les mesures s'han utilitzat les Funcions de Distribució de Probabilitat (PDF). Aquests resultats LES són també comparables als obtinguts amb altres models LES, tal com mostra la intercomparació de models LES, més coneguda com a GABLS.Un cop desenvolupades totes les eines necessàries es fa un LES d'un cas més realista, basat en les observacions d'un màxim de vent de capes baixes (més conegut com a Low-Level Jet, LLJ). L'anàlisi combinat dels resultats LES i les mesures permet entendre millor els processos de barreja que tenen lloc a través de la inversió. Finalment, la contribució dels efectes locals s'estudia mitjançant les simulacions mesoscalars, en aquest cas centrades a l'illa de Mallorca. Durant el vespre es veu com les circulacions locals es desenvolupen a les conques (de longitud al voltant de 25km), formant-se, per exemple, vents catabàtics o LLJ com l'estudiat anteriorment. En aquest cas les simulacions es verifiquen amb imatges de satèl·lit NOAA i observacions de les estacions automàtiques de mesures, donant resultats semblants. / The atmospheric boundary layer is the area directly influenced by the presence of the Earth's surface and its height is from hundreds of meters to few kilometres. During the night, the radiative cooling stratifies the layer close to the surface and it forms the Stably-stratified Atmospheric Boundary Layer (SBL). Nowadays, the SBL is a regime not well enough characterized, yet. Turbulence, which is not homogeneous either isotropic, and the great importance of the local effects, like the orography, among other factors, make the SBL be a difficult regime to study. Even so, the SBL is an object of special attention, especially when improving its representation in numerical prediction models or climate models.This work focuses on the study of the SBL through 3 different tools: 1) Large-Eddy Simulations (LES), to determine the turbulent motions, where the resolutions are about 1m; 2) Mesoscale simulations, to characterize the local effects, where resolutions are about 1km; 3) Analysis of the observations under these conditions in order to better characterize and understand the observed phenomena.In first place, it is studied the range of stabilities where the LES model, that considers the Kolmogorov theory for the dissipation of the energy, works correctly. The results are realistic as the comparison with measures from two experimental campaigns (SABLES-98 and CASES-99) shows. To explore the results more thoroughly, and to compare the LES results to the measurements, the Probability Density Functions (PDF) have been used. The LES results are also comparable to the ones obtained with other LES models, as the intercomparison of different LES models show, better known as GABLS.Then, a more realistic case is performed using the LES model, based on observations of a Low-Level Jet (LLJ). The combined inspection of the LES results and the observations allow to better understand the mixing processes that take place through the inversion layer. Finally, the contribution of the local effects is studied through a mesoscale simulation. Here the attention is focused on the Mallorca Island. During the night, the model is able to reproduce the local circulations is a basin of a characteristic size of 25km. The main features obtained previously from the LES of the LLJ are also reproduced by the mesoscale model. These runs are verified with NOAA satellite images and observations from the automatic surface weather stations, giving that the model is able to reproduce realistic results.

Novel approaches in determining baseline information on annual disposal rates and trace element content of U.S. coal combustion residues : a response to EPA’s June 2010 proposed disposal rule

Although products of coal combustion (PCCs) such as coal ash are currently exempted from classification as a hazardous waste in the United States under the 1976 Resource Conservation and Recovery Act (RCRA), the U.S. Environmental Protection Agency (EPA) is now revising a proposed rule to modify disposal practices for these materials in order to prevent contamination of ground- and surface water sources by leached trace elements. This paper analyzes several aspects of EPA’s scientific reasoning for instating the rule, with the intent of answering the following questions: 1) Are EPA’s cited values for PCC production and disposal accurate estimates of annual totals?; 2) In what ways can EPA’s leaching risk modeling assessment be improved?; 3) What is the total quantity of trace elements contained within all PCCs disposed annually?; and 4) What would be the potential costs and feasibility of reclassifying PCCs not under RCRA, but under existing NRC regulations as low-level radioactive waste (LLRW)? Among the results of my calculations, I found that although EPA estimates for annual PCC disposal are 20% larger than industry statistics, these latter values appear to be closer to reality. Second, EPA appears to have significantly underestimated historical PCC disposal: my projections indicate that EPA’s maximum estimate for the quantity of fly ash landfilled within the past 90 years was likely met by production in the last 30 years alone, if not less. Finally, my analysis indicates that while PCCs may potentially meet the criteria for reclassification as low-level radioactive waste by NRC, the cost of such regulation would be many times that of the EPA June proposed disposal rule ($220-302 billion for PCCs disposed in 2008 alone, versus $1.47 billion per year for the Subtitle C option and $236-587 million for Subtitle D regulatory options). / text

Cohorte de patients avec le VIH/SIDA : échecs virologiques et effets de thérapies antirétrovirales sur la fonction rénale et l'hyperbilirubinémie

Le virus de l'immunodéficience humaine (VIH) est à l’origine d’une infection chronique, elle-même responsable du développement du syndrome d'immunodéficience acquise (SIDA), un état de grande vulnérabilité où le corps humain est à la merci d’infections opportunistes pouvant s’avérer fatales. Aujourd’hui, 30 ans après la découverte du virus, même si aucun vaccin n’a réussi à contrôler la pandémie, la situation s’est grandement améliorée. Conséquemment à l’arrivée de traitements antirétroviraux hautement actifs (HAART) à la fin des années 1990, la mortalité associée au VIH/SIDA a diminué et un plus grand nombre de personnes vivent maintenant avec l'infection. La présente thèse avait pour objectif d’aborder trois situations problématiques, en dépit de l’efficacité reconnue des HAART, plus particulièrement la faible charge virale persistante (LLV) et sa relation avec l’échec virologique, ainsi que les effets de certains antirétroviraux (ARV) sur les fonctions rénale et hépatique. Les objectifs précis étaient donc les suivants : 1) étudier le risque d’échec virologique à long terme chez les patients sous HAART dont la charge virale est indétectable comparativement aux patients affichant une LLV persistante; 2) évaluer sur le long terme la perte de fonction rénale associée à la prise de ténofovir (TDF) 3) étudier sur le long terme l'hyperbilirubinémie associée à la prise d’atazanavir (ATV) et ses autres déterminants possibles. Afin d’atteindre les trois objectifs susmentionnés, une cohorte de 2 416 patients atteints du VIH/SIDA, suivis depuis juillet 1977 et résidant à Montréal, a été utilisée. Pour le premier objectif, les résultats obtenus ont montré un risque accru d’échec virologique établi à >1000 copies/ml d’ARN VIH chez tous les patients qui présentaient une LLV persistante de différentes catégories durant aussi peu que 6 mois. En effet, on a observé qu’une LLV de 50-199 copies/ml persistant pendant six mois doublait le risque d’échec virologique (Hazard ratio (HR)=2,22, Intervalle de confiance (CI) 95 %:1,60–3,09). Ces résultats pourraient modifier la façon dont on aborde actuellement la gestion des patients affichant une LLV, et plus particulièrement une LLV de 50-199 copies/ml, pour laquelle aucune recommandation clinique n’a encore été formulée en raison du manque de données. Pour le deuxième objectif, on a observé une augmentation du risque de perte de fonction rénale de l’ordre de 63 % (HR=1,63; 95% CI:1,26–2,10) chez les patients sous TDF comparativement aux patients traités avec d’autres ARV. La perte de fonction rénale directement attribuable à la prise de TDF, indique que cette perte est survenue au cours des premières années de l’exposition du patient au médicament. D’une perspective à long terme, cette perte est considérée comme modérée. Enfin, pour ce qui est du troisième objectif, on a constaté que l’incidence cumulative d’hyperbilirubinémie était très élevée chez les patients sous ATV, mais que cette dernière pouvait régresser lorsque l’on mettait fin au traitement. L’hyperbilirubinémie à long terme observée avec la prise d’ATV n’a été associée à aucun effet néfaste pour la santé. Dans l’ensemble, la présente thèse a permis de mieux comprendre les trois situations problématiques susmentionnées, qui font actuellement l’objet de débats au sein de la communauté scientifique, et d’éclairer sous un jour nouveau la gestion des patients séropositifs sous traitement médicamenteux. / Human immonudeficiency virus (HIV) is a virus causing a chronic infection responsible for Acquired Immunodeficiency Syndrome (AIDS), a state of vulnerability of the body where different opportunistic infections will ultimately be fatal. About 30 years after the discovery of the virus, even if no vaccine is available to control the pandemia, situation has changed for the best. With the arrival of highly active anti-retroviral therapy (HAART) in the late 90's, a reduction in HIV/AIDS mortality rate and growing number of persons living with the infection were observed. The overall objective of this thesis was to address three problematic situations, despite recognised HAART efficacy, especially low-level viremia (LLV) and its relationship with virologic failure, and the impacts of certain antiretrovirals (ARV) on kidney and hepatic functions. The specific objectives were: 1) to study the risk of virologic failure in long-term perspective in undetectable patients under HAART in comparison to patients with persistent LLV; 2) to evaluate the long-term loss of kidney function related to tenofovir (TDF) exposure 3) to evaluate long-term hyperbilirubinemia related to atazanavir (ATV) exposure and other possible determinants. In order to address the three specific objectives, a cohort of patients 2416 living with HIV/AIDS followed in Montreal since July 1977 was used. For the first objective, analyses and results shown an increased risk of virological failure defined as >1000 copies/mL of HIV RNA, for all categories of persistent LLV as soon as 6 months of persistent duration. Persistent LLV of 50-199 copies/mL for 6 months doubled the risk of virologic failure (Hazard ratio (HR)=2,22, Confidence interval (CI) 95%: 1,60-3,09). The results shed new light for the management of patients with LLV, especially for LLV of 50-199 copies/mL, for which no clinical recommendation is currently available due to a lack of data. For the second objective, an increased risk of loss of kidney function of 63% (HR=1.63; 95% CI:1.26–2.10) associated to TDF exposure in comparison to patients taking other ARV was observed. The cumulative eGFR loss directly attribuable to TDF also shown that this loss occured during the first years of exposure. This loss was mild in a long-term perspective. For the third objective, it has been shown that the cumulative incidence of hyperbilirubinemia in ATV users was very high and that regression was possible if ATV exposure was ended. Long-term hyperbilirubinemia related to ATV use was not associated with adverse health outcome. Overall, this thesis allowed a better understanding of these three problematics currently debated in scientific literature and shed new lights on management of HIV positive patients under therapy.

Atualmente, as terapias eficazes para esta condição são escassas. Neste estudo, foram investigados os efeitos da terapia laser de baixa intensidade (LLLT) sobre a fibrose intersticial, que ocorre após obstrução ureteral unilateral (OUU) em ratos, um modelo experimental de doença renal crônica. Materiais e Métodos: Foram utilizados 32 ratos Wistar, 8 em cada grupo, machos, com 250 a 300g de peso aproximadamente e 8 semanas de idade. O rim obstruído de metade dos ratos, submetidos à OUU receberam dose única intra-operatória do LLLT (AlGaAs laser, 780 nm, 22,5 J / cm ², 30 mW, 30 segundos em cada um dos nove pontos). Após 14 dias, a fibrose renal foi avaliada pela coloração por picrosírius e medição da área transversal sob luz polarizada. Análise imunohistoquímica quantificou células do tecido renal que expressam marcadores de fibroblastos (FSP-1) e miofibroblastos (α-SMA). RT-PCR foi realizado para determinar a expressão de mRNA de genes chaves relacionados com a fibrose: TGF-β1, Smad3 e colágeno I (Col I). Resultados: No grupo OUU e tratado pelo LLLT os animais apresentaram menos fibrose renal do que os animais obstruídos (OUU). α-SMA, TGF-β1 e Smad3 foram aumentados no interstício renal de ratos OUU. LLLT reduziu a expressão de todas essas moléculas. LLLT não parece ter um efeito significativo no Col I ou FSP-1, que também foram induzidos por OUU. Conclusão: Pela primeira vez, nós mostramos que LLLT tem um efeito protetor em relação à fibrose intersticial renal. Entende-se que, atenuando a inflamação, a laserterapia pode impedir a ativação tubular e transdiferenciação, que são os dois processos principais que formam a fibrose renal no modelo OUU. / Background and Objective: Regardless of the etiology, chronic kidney disease (CKD) involves progressive widespread tissue fibrosis, tubular atrophy and loss of kidney function. At present, effective therapies to this condition are lacking. We investigated the effects of low level laser therapy (LLLT) on the interstitial fibrosis that occurs after unilateral ureteral obstruction (UUO) in rats, an experimental model of CKD. Study Design/Materials and Methods: We used 32 Wistar rats, 8 in each group, males, 250 to 300g weight and 8 weeks old. The occluded kidney of half of the Wistar rats that underwent UUO received a single intraoperative dose of LLLT (AlGaAs laser, 780 nm, 22.5 J/cm², 30 mW, 30 seconds on each of nine points). After 14 days, renal fibrosis was assessed by Sirius red staining and measurement of the cross-sectional area under polarized light. Immunohistochemical analyses quantitated the renal tissue cells that expressed fibroblast (FSP-1) and myofibroblast (α-SMA) markers. RT-PCR was performed to determine the mRNA expression of key fibrosis-related genes, namely TGF-β1, Smad3 and collagen I (Col I). Results: The UUO-LLLT animals had less severe renal fibrosis than OUU animals. α- SMA, TGF-β1 and Smad3 were increased in the renal interstitium of UUO rats. LLLT reduced the expression of all of these molecules. LLLT did not appear to have a significant effect on Col I or FSP-1, which were also induced by UUO. Conclusion: For the first time, we showed LLLT had a protective effect regarding renal interstitial fibrosis. It is conceivable that by attenuating inflammation, LLLT can prevent tubular activation and transdifferentiation, which are the two processes that mainly drive the renal fibrosis of the UUO model.

The World Health Organization (WHO) in 2009 attributed to high blood pressure (BP) was the death cause for 9.5 million people worldwide. Currently, the hypertension has become a serious public health problem. This entity is an important risk factor for congestive heart failure, cerebrovascular disease, acute myocardial infarction, nephropathy, retinopathy and peripheral vascular insufficiency. Studies have suggested that laser photobiomulation, employing a low power, acts into the inflammatory and proliferative phases of tissue repair, by modulating the inflammatory mediators synthesis as same as the Reactive Oxygen Species (ROS). According scientific publications indicate that the inflammation component is closely related to systemic arterial hypertension as well as possibly to the oxidative stress, both participates in the Hypertension genesis. The aim of this study was to verify the long-term effects of Low Level Laser Therapy (LLLT) application in Spontaneously Hypertensive Rats-SHR (Spontaneously Hypertensive Rats) through on cardiovascular autonomic modulation and oxidative stress in the blood. The experiment consisted in 3 phases: Phase I – LLLT irradiation on SHR: The experiment's phase I consisted of animal’s irradiation, when the laser group received three times LLLT applications weekly for a 7 weeks total; the sham group received three times per week of LLLT simulation for 7 weeks and a total of 21 applications. Prospective, randomized, controlled study, with 16 SHR approximately 2 months age, randomly divided into 2 groups : Sham (n = 8) and Laser (n = 8). The animals were irradiated in a prompt, onto the tail’s dorsal area, using a Diode Laser (MMOptics, São Carlos, SP, Brazil) with a wavelength (λ) of 780 ± 2 (nm), output power at 40 mW, with a 0.04 cm2 beam area, dose of 30 J/cm2 power density of 1W/cm2 and irradiation time of 90 s. In Phase II - Hemodynamic and autonomic cardiovascular evaluation: for a period of 7 weeks, consisted in the cannulation procedure, collecting and analysis. The animals were cannulated, evaluated hemodynamically and analyzed the cardiovascular autonomic modulation. Phase III - Oxidative stress analysis, were analyzed: a) protein damage; b) cell membrane damage; c) antioxidant enzyme activity; d) nitrite concentrations. Data from phase II and III were collected and statistically analyzed applying One Way ANOVA test, followed by post hoc Student - Newman Keulls and considering the significance level of p < 0.05, equivalent to an error α 0.05. The results demonstraded hemodynamic parameters of group LLLT treated showed a BP reduction, when compared with the Sham group. In laser group the diastolic arterial pressure (DAP) showed a reduction of -14 mmHg (± 143 * 4 x 157 ± 3 mmHg Sham) and mean arterial pressure (MAP) - 13mmHg (169 ± 4 * x 182 ± 4 mmHg Sham) there were statistically significant difference. Although the value of systolic arterial pressure (SAP) (196 ± 5 x 207 ± 4 mmHg) showed no differences. There was a decreased in resting HR with a statistically significant difference in the laser group compared to Sham (312 ± 14 vs. 361 ± 13 bpm sham). The spectral reviews in the field of time and frequency showed that the Laser group decreased sympathetic activity on the heart and blood vessels while compared to the Sham group. The heart rate variation was analyzed using the DP-PI ( standard deviation of the pulse interval) VAR-PI components (pulse interval variability) and it demonstrated that LLLT was effective in diminishing variation in heart rate (HR) and sympathetic activity in heart, inducing a substantial fall in blood pressure. Lasertherapy presented a rise in spectral low-frequency component in the pulse interval (LF - IP action of the sympathetic at heart), though the sham group showed up exaggeratedly decreasing (6.77 ± 4:35 and 2:31 ± 0:16 ms ² Sham) as a function of saturation variation. Thus, there was a significant reduction in sympathetic activity after LLLT using. A high-frequency band on interval pulse HF-IP (parasympathetic activity) showed no statistically significant differences between the groups and Laser Sham group. The baroreceptor sensitivity, assessed by the alpha index, signalized a significant increase in the Laser (1:07 ± 0:23 vs. 0:45 ± 0:20 ms / mmHg Sham) group, presenting an improvement in the receptors sensitivity. The baroreflex results were associated with other relevant data, the VAR - SAP (49.55 ± 15.94 * vs 70.51 ± 13:55 mmHg² Sham) and SD -SAP (6.94 ± 1.21 * vs 8.68 ± 1.11 mmHg Sham) that proved to be diminished in the laser group, indicating baroreflex improvement sensitivity concomitantly to the positive SAP variation reduction of. There were no significant differences in baseline SAP (196 ± 5 vs. 207 ± 4 mmHg Sham) between the two groups. The results in the oxidative stress and autonomic analysis demonstrated an association between increased NO production (nitrite 0:36 ± 0:03 vs 0:26 ± 0:03 nm / mg Sham) and decreased in the vascular sympathetic (LF - SAP 7.28 ± 1.63 * vs 9.86 ± 0.47 Sham), both leading to a profound vasodilatation then a significant fall in of blood pressure. Lasertherapy shown to alter the plasma parameters such as oxidative nitrite, revealing an NO increased metabolism, as described above and, moreover, accounted for a significant reduction in carbonyl plasma concentration (vs 3.93 ± 0.24, 4.75 ± 0:26 * nm / mg Sham). Our experimental study indicate that LLLT was able to reduce the oxidative stress parameters through diminishing the damage to the proteins. The enzymatic defense was analyzed by the enzyme SOD concentration in blood plasma, denoted that no significant differences (4:42 ± 0:10 4:25 ± 0:06 vs usod / mg) between groups. Thus, low level laser therapy has shown to improve cardiovascular autonomic activity as well as oxidative parameters which resulted in steadily staggeringly reduce the blood pressure of hypertensive animals. / Em razão do aumento populacional e a senescência, o número de indivíduos com Hipertensão Arterial Sistêmica (HAS) cresceu de 600 milhões em 1980 para 1,2 bilhões (OMS 2011). Lim (2012) atribuiu que a pressão arterial (PA) elevada fosse a causa mortis de 9,5 milhões de indivíduos ao redor do mundo. Atualmente, a HAS tornou-se um grave problema de saúde pública. A hipertensão é um importante fator de risco para insuficiência cardíaca congestiva, doenças cerebrovasculares, infarto agudo do miocárdio, nefropatia, insuficiência vascular periférica e retinopatia hipertensiva. Considerando publicações científicas que demonstram que o componente da inflamação e do estresse oxidativo estão intimamente relacionados à gênese da hipertensão arterial sistêmica (HAS), e que o laser com potência baixa tem efeito positivo no estresse oxidativo e apresenta ação antiinflamatória eficaz, desta forma buscamos estudar a resposta da Laserterapia na HAS. Inúmeros estudos vêm sugerindo, ao longo de décadas, que a fotobiomulação pelo laser empregado uma potência baixa, atua durante as fases inflamatórias e proliferativas da reparação tissular, modulando síntese de mediadores inflamatórios e espécies reativas de oxigênio (ROS). O objetivo deste estudo foi analisar os efeitos da aplicação do laser de baixa intensidade em ratos espontaneamente hipertensos SHR (Spontaneously Hypertensive Rats) em longo prazo na modulação autonômica cardiovascular e no estresse oxidativo sangúineo. Estudo prospectivo, randomizado e controlado com 16 ratos SHR, divididos aleatoriamente em 2 grupos: Sham (n=8) e Laser (n=8).O experimento foi dividido em três fases: Fase I – Irradiação dos animais: constituiu-se na irradiação com laser nos animais SHR, onde o grupo Laser recebeu três aplicações semanais de LBI durante sete semanas; já no grupo Sham foram realizados três simulações de aplicação semanais de Laser de Baixa Intensidade (LBI) durante 7 semanas, totalizando 21 aplicações de LBI. Os animais foram irradiados pontualmente, na região dorsal da cauda, utilizando um Laser Diodo (MMOptics, São Carlos, SP, Brasil) com comprimento de onda de λ = 780 ± 2 (nm); potência de 40 mW, área do feixe de 0,04 cm2, densidade de energia de 30 J/cm2, densidade de potência de 1W/cm2, tempo total de irradiação de 90 s de exposição. Fase II – Avaliação hemodinâmica e autonômica cardiovascular: constituiu-se nos procedimento de canulação, registro de dados e coleta de material, teve inicio após sete semanas de irradiação. Os animais canulados foram avalidados de forma hemodinâmica, bem como analisada a modulação autonômica cardiovascular. Fase III – Análises do estresse oxidativo, foram analisadas: a) danos à proteína; b) danos à membrana celular; c) atividade enzimática; d) concentração de nitrito. Os dados da fase II e III foram coletados e analisados estatisticamente através dos testes Anova One Way, seguido de Post Hoc de Student Newman-Keulls, considerando-se o nível de significância p < 0,05, equivalendo a um erro α de 0.05. Os resultados hemodinâmicos do grupo tratado com LLLT denotaram um decréscimo significativo da PA quando comparado com o grupo Sham. A pressão arterial diastólica (PAD) do grupo Laser revelou uma redução de -14 mmHg (143± 4*vs157±3 mmHg Sham) e a pressão arterial média (PAM) -13mmHg (169±4*vs182±4 mmHg Sham), a frequência cardíaca (FC) em repouso (312±14*vs361±13 bpm Sham) revelando uma diferença estatisticamente significante, porém o valor da pressão arterial sistólica(PAS) não mostrou (196±5 x 207±4 mmHg) alterações entre os grupos. As avaliações espectrais no domínio do tempo e da frequencia demostraram que o grupo Laser reduziu a atividade simpática sobre o coração e vasos sanguíneos quando comparados ao grupo Sham. A variação frequência cardíaca foi analisada através dos componentes VAR-IP (variabilidade do intervalo de pulso) e o DP-IP (desvio do intervalo de pulso) que evidenciaram que o LBI foi eficaz no decréscimo variação da FC e da atividade simpática no coração, induzindo assim a queda das pressões arteriais. A laserterapia mostrou um incremento no componente espectral baixa frequência no intervalo de pulso (BF-IP ação do simpático no coração), porém o grupo Sham apresentou-se exacerbadamente diminuído (6.77 ± 4.35 e 2.31±0.16 ms² Sham) em função da saturação da variação desse componente que foi reduzido. Desta forma, houve um importante decréscimo da atividade simpática com o uso do LBI, significando uma importante diminuição dos níveis pressóricos. A banda de alta frequência (AF-IP atividade parassimpática cardíaca) não mostrou diferenças estatísticas significantes entre os grupos Laser e grupo Sham. A sensibilidade dos barorreceptores, avaliada pelo índice alfa, demonstrou um significativo incremento da resposta no grupo Laser (1.07 ± 0.23 vs 0.45 ± 0.20 ms/mmHg Sham), revelando uma melhora na sensibilidade destes receptores. Os resultados dos barorreflexos encontravam-se associados a outro dado relevante, o componente VAR-PAS (49.55 ± 15.94* vs 70.51 ± 13.55 mmHg² Sham) e DP-PAS (6.94 ± 1.21* vs 8.68 ± 1.11 mmHg Sham) que mostrou-se diminuído no grupo Laser, indicando que a melhora da sensibilidade barorreflexa ocorreu, concomitantemente, à redução positiva da variação da PAS. Não houve diferenças estatísticas significantes na PAS basal (196±5 vs 207 ± 4 mmHg Sham) entre os dois grupos. Já os resultados encontrados na análise do estresse oxidativo e autonômica demonstraram uma associação entre o incremento da produção do óxido nitrico (NO) (nitrito 0.36 ± 0.03 vs 0.26 ± 0.03 nm/mg Sham) e redução do simpático vascular (BF-PAS 7.28 ± 1.63* vs 9.86 ± 0.47 Sham), ambos levando a uma vasodilatação com consequente queda dos níveis pressóricos arteriais. A laserterapia mostrou alterar parâmetros oxidativos como as espécies reativas de nitrogênio (RNS reactive nitrogen species), o nitrito plasmático, revelando um aumento do metabolismo do NO, como já descrito anteriormente e denotou uma diminuição significativa da concentração de carbonilas plasmáticas (3.93 ± 0.24 * vs 4.75 ± 0.26 nm/mg Sham). A defesa enzimática foi analisada através da concentração da enzima SOD no plasma sanguíneo, que não apontou diferenças significativas (4.42 ± 0.10 vs 4.25 ± 0.06 usod/mg) entre os grupos. Evidenciamos que o LBI foi capaz de reduzir este parâmetro oxidativo, reduzindo os danos às proteínas decorrente do estresse. Desta forma, concluímos que a laserterapia demonstrou resposta positiva ao melhorar a atividade autonômica cardiovascular e parâmetros oxidativos que resultaram na redução dos níveis pressóricos dos animais hipertensos.

Every reader can also read about famous aviation accidents and look at models of weather that caused those accidents. At the end of thesis there are analyses of a rawiosonde measurements related to wind shear reports. This whole thesis is focused on increasing awareness of wind shear in aviation from angle of view of both pilots and aviation meteorologist.

This thesis describes design of a cheap robot. It includes implementation of firmware of low level control unit based on microcontroller PIC. The firmware drives motors, gains sensors data and communicates with the high level control unit. Furthermore the thesis presents realisation of connection to the robotic operation system ROS and its standard structures allowing usage of existing packages for the robot teleoperation and displaying sensor data on the remote computer. The thesis finally reports experiments with the robot. The constructed prototype is the model of the robotic lawn mower, however the whole solution has universal usage.

Presents results of research in the field of low-level electromagnetic fields interacting with the higher organisms, in the frequency band from 0,01 to 30 Hz effect on the individual or social group. Provides a description of the methodology of measuring and evaluating the impact of workplace assembled measuring low-level electromagnetic fields, extremely low frequency (ELF) on a sample of respondents. It was proposed measuring device for monitoring and evaluating how respondents' reactions to changes in the geomagnetic or external - external magnetic / electromagnetic field and to work for the measurement and evaluation of changes in external magnetic fields very low levels (ELF). This work is designed to process and evaluate implemented within experimental research into the influence of low-level magnetic fields in the Earth's resonator psychophysiological parameters of human organism and its behavior and decision-making. The work confirmed the influence of low-level magnetic fields on the psychophysiological parameters of the organism to be simultaneously confirmed the hypothesis established in accordance with the objectives of work.
