Molecular characterisation of the causal agent of bacterial leaf streak of maize / Nicolaas Johannes Jacobus Niemann

All members of the genus Xanthomonas are considered to be plant pathogenic, with specific pathovars infecting several high value agricultural crops. One of these pathovars, X. campestris pv. zeae (as this is only a proposed name it will further on be referred to as Xanthomonas BLSD) the causal agent of bacterial leaf steak of maize, has established itself as a widespread significant maize pathogen within South Africa. Insufficient information about the present distribution of the pathogen is available. The main aim of the study was thus to isolate and characterise the pathogen using molecular methods. Results demonstrated that the causal agent of bacterial leaf streak disease (Xanthomonas BLSD: potentially X. campestris pv. zeae) was widely distributed within the major maize cultivation regions of South Africa. Most of the isolates collected originated from the Highveld maize production provinces (North West, Free State, Gauteng and Mpumalanga provinces) as well as from irrigated maize fields in the Northern Cape province. The XgumD gene marker was used to determine if the isolates belonged to the genus Xanthomonas. The gumD gene fragment is located within the gumB-gumM region of the operon and is conserved among Xanthomonas species. This gene fragment is partially responsible for xanthan production. This marker was amplified from all isolates and a selected number were sequenced. The marker was only able to confirm that the causal agent was a member of the genus Xanthomonas. PCR methods were used for the characterisation of the isolates. This included PCR and sequencing of ribosomal RNA- gyraseB and gumD genes. A fingerprinting method BOX-PCR was also employed. Good quality DNA of sufficient quantities was obtained from the various isolates. Amplification produced no non-specific amplification products. This resulted in good quality sequences that could be analysed using bioinformatics tools. Phylogenetic analyses of the ribosomal RNA and gyraseB genes could not detect differences amongst the 47 Xanthomonas BLSD isolates. However, these genes were able to distinguish between the type strain of these isolates and various Xanthomonas species and pathovars. From all three neighbour joining trees the Xanthomonas BLSD isolates had close association with X. axonopodis pv. vasculorum strain ATCC 35938. For the 16S rRNA gene there exists no sequence differences between Xanthomonas BLSD and X. axonopodis pv. vasculorum strain ATCC 35938. A single nucleotide difference was observed between Xanthomonas BLSD and X. axonopodis pv. vasculorum strain ATCC 35938 for the 23S rRNA gene. The gyraseB gene detected a total of six nucleotide variations between these two Xanthomonas species. For all of the phylogenetic trees there was no clustering of Xanthomonas BLSD with X. campestris pathovars. Genetic profiling (via BOX-PCR) based on present/absent analysis revealed no variations amongst the Xanthomonas BLSD isolates. All isolates shared an identical pattern produced by 12 distinct PCR products. This profiling technique did differentiate between the isolates of Xanthomonas BLSD and X. axonopodis pv. vasculorum strain ATCC 35938. Their profiles shared common bands, but differed in the number and overall pattern of the bands. These results suggest two main conclusions: (i) Xanthomonas BLSD has a clonal origin with geographical separation not impacting genetic variation. The fact that all the isolates appear to be clonal may imply that when resistant maize cultivars are developed these should be resistant to all isolates of the pathovar irrespective of their geographical origin. This is a suggestion that will have to be corroborated using more isolates and additional genetic fingerprinting techniques (ii) the Xanthomonas BLSD isolates from this study may not belong to X. campestris. L'induction du métabolisme secondaire fait partie du système de défense des plantes lors d’une attaque par un microorganisme pathogène. Les propriétés antimicrobiennes ou de signalisation des métabolites secondaires (MS) peuvent être régulées par un processus majeur et efficace, la glycosylation. Cette réaction est catalysée par les glycosyltransférases (UGTs) qui assurent le transfert d’une molécule de sucre sur un MS. Mon travail de thèse a porté sur l’étude de trois UGTs d’Arabidopsis thaliana (Arabidopsis), UGT76E12 d’une part, et UGT73B3 et UGT73B5 d’autre part, et a montré que 1) UGT76E12 est impliquée dans la mise en place de la résistance basale à la souche virulente de la bactérie Pseudomonas syringae pv. tomato, Pst DC3000, et participe au dialogue entre les voies de signalisation hormonale de l’acide jasmonique (JA) et de l’acide salicylique (SA). Des analyses par clustering hiérarchique ont révélé une co-expression forte entre le gène UGT76E12, et deux gènes codant une terpène synthase (TPS4) et un cytochrome P450 (CYP82G1), impliqués dans la biosynthèse d’un diterpène, le TMTT ((E,E)-4,8,12-triméthyletridéca-1,3,7,11-tetraène), suggérant un rôle pour UGT76E12 dans la régulation de la voie de biosynthèse des terpènes ; 2) UGT73B3 et UGT73B5 sont impliquées dans la mise en place de la résistance spécifique d’Arabidopsis à la souche avirulente Pst DC3000-AvrRpm1, notamment par la régulation de MS intervenant dans le maintien de l’état redox cellulaire au cours de la réaction d’hypersensibilité (HR). Ces deux gènes appartiennent à la classe des « SA-early induced-genes » et présentent un profil co-expression avec le gène TOLB-related et deux gènes codant des glutathion S-transférases (GSTU7 et GSTU24) impliqués dans la réponse au stress oxydant et la détoxication des MS. Nos résultats suggèrent un rôle pour UGT73B3 et UGT73B5 dans la glycosylation de métabolites oxydants associés à la mise en place de la HR. / Plant secondary metabolism induction is part of an integrated defense system after pathogen infection. Signalling and antimicrobial properties of secondary metabolites (SM) can be regulated by an efficient process named, glycosylation. This reaction is ensured by glycosyltransferases (UGTs) which catalyze the transfer of a sugar moiety on a SM. My PhD work was divided in two parts, the first on UGT76E12 and the second part on UGT73B3 and UGT73B5, three UGTs of Arabidopsis thaliana (Arabidopsis) and it demonstrated that 1) UGT76E12 is involved in basal resistance establishment during plant challenge with the virulent strain of Pseudomonas syringae pv. tomato (Pst DC3000), and participates to the hormonal crosstalk between jasmonic acid (JA) and salicylic acid (SA). Clustering analyses revealed a strong co-expression between UGT76E12 and two genes encoding a terpene synthase (TPS4) and a cytochrome P450 (CYP82G1), both involved in the biosynthesis of a diterpene named TMTT ((E,E)-4,8,12-trimethyltrideca-1,3,7,11-tetraene), which suggests a role for UGT76E12 in terpene biosynthesis pathway regulation. 2) UGT73B5 and UGT73B3 are involved in the establishment of Arabidopsis specific resistance against the avirulent strain of Pst DC3000-AvrRpm1, especially in the regulation of MS playing a role in redox homeostasis status during the hypersensitive response (HR). These two genes are part of SA-early induced-genes and showed co-expression patterns with the TOLB-related gene and two genes encoding glutathione S-transferases (GSTU7 and GSTU24) involved in oxidative stress responses and in MS detoxification. Our results suggest a role for UGT73B3 and UGT73B5 in the glycosylation of oxidant MS associated with HR establishment.

Le manioc (Manihot esculenta L. Crantz) est cultivé dans toute la zone intertropicale. Compte tenu de ses utilisations potentielles, le développement de la culture du manioc a considérablement augmenté au niveau mondial, mais il est encore considéré comme une culture de subsistance en particulier dans les régions pauvres où il est cultivé par les petits producteurs. La production du manioc est fortement contrainte par des facteurs biotiques et abiotiques, dont la bactériose vasculaire du manioc (CBB) et la nécrose bactérienne du manioc (CBN) qui sont deux maladies causées par les bactéries Xanthomonas axonopodis pv. les manihotis (Xam) et X. cassavae (Xc), respectivement. CBB est considérée comme la principale maladie bactérienne, notamment au Venezuela où la CBB a été signalée pour la première fois dans les années 70. Dans les années 90, des études ont été menées pour élucider la variabilité génétique de Xam dans différentes régions du pays, au moyen d’outils moléculaires, mettant en évidence un degré élevé de polymorphisme parmi les souches analysés.Le sujet de cette thèse porte sur la situation de CBB au Venezuela 20 ans après. Quelle est la diversité génétique actuelle de Xam au Venezuela? Quelle est la structure génétique des populations de Xam dans ce pays, et comment diffèrent-elles des souches de Xam recueillies dans les années 90? En outre, étant donné que Xam et Xc provoquent des symptômes semblables sur feuilles de manioc et présentent des caractéristiques physiologiques et morphologiques similaires, nous avons également visé à développer un nouvel outil de diagnostic moléculaire permettant une détection rapide et fiable de Xam et de Xc tout étant capable de les discriminer l’une vis à vis de l’autre.Pour atteindre nos objectifs, nous avons d'abord établi une PCR-duplex comme outil de détection moléculaire des xanthomonades infectant le manioc. Sur la base de l'analyse in silico des séquences du génome de 66 souches de Xam et une de Xc, nous avons pu sélectionner 6 paires d'amorces candidates spécifiques de Xam et six autres pour Xc. Nous avons pu développer un test de PCR-duplex qui a été validé en testant 53 souches de Xam et 25 souches de Xc issues de différents pays, 18 souches non cibles contrôles et cinq souches épiphytes associées au manioc. Cette technique représente un outil utile pour détecter et différencier Xam et Xc provenant de cultures in vitro mais aussi fonctionnelle à partir de tissues de plantes infectés.Deuxièmement, nous avons donc évalué la diversité des populations de Xam à l’aide de l’étude de microsatellites (ou MLVA pour « Multiple loci VNTR analysis »). Une enquête de terrain menée dans six États du Venezuela a permis d'évaluer la présence de la maladie, son statut et nous a permis d'établir une collection de souches de Xam dont l’analyse détaillée de la diversité a pu être réalisée. Au total nous avons isolé 202 souches de Xam issues de six localités situées dans quatre états. À l'aide d'un schéma 14-MVLA, nous avons analysé 12 populations et mis en évidence un indice élevé de la diversité génétique au sein de ces populations et entre elles, et principalement dans l'est du pays.Le développement de ce type de recherche est essentiel pour une gestion raisonnée des cultures dans le monde. Conjugué aux politiques agricoles existantes, il nous permettra de mieux comprendre les agents pathogènes d'importance agricole et les mécanismes impliqués dans leur établissement dans le temps et à travers l’espace. L'objectif à long terme est d'appliquer des mesures de contrôle efficaces et durables, ce qui conduira à des mesures de quarantaine plus efficaces pour prévenir la propagation de la maladie. / Cassava (Manihot esculenta L. Crantz) belongs to the group of roots and tubers and is cultivated in the tropics worldwide. This species is a nutritional alternative in many populations where no optimal crop production, general conditions nor technological support exist. Considering its potential uses, the global development of cassava crop has increased significantly but it is still considered a subsistence crop in poor regions lead by smallholder producers. Cassava is affected by biotic and abiotic factors during its life cycle, which heavily limiting its optimal performance. A variety of pests and diseases are known to affect cassava production. Among them are those caused by Xanthomonas axonopodis pv. manihotis (Xam) and Xanthomonas cassavae (Xc), causal agents of Cassava Bacterial Blight (CBB) and Cassava Bacterial Necrosis (CBN) diseases, respectively. CBB is considered the major bacterial disease that affects cassava crop worldwide which is also the case in Venezuela where it was first reported in the 70s. Since the 90s, studies were conducted to elucidate Xam genetic variability in different regions in the country, by means of different molecular tools available at that time. A high degree of polymorphism among the isolates was reported, whether collected from the same or different fields. The Xam population was distributed into eight clusters and no correlation was observed between genetic diversity and geographic origin.Our questions deal with the situation of CBB in Venezuela 20 years later : what is the current genetic diversity of Xam populations in Venezuela? what is the genetic structure of Xam populations and how do they differ with respect to Xam strains collected in 90s. Moreover, because Xam and Xc cause similar symptoms on cassava leaves and display similar physiological and morphological characteristics, we also aimed at developing a new molecular diagnostic tool allowing for fast and reliable detection of Xam and able to discriminate with Xc.To achieve our goals, we first established a duplex-PCR as a molecular detection tool of cassava-infecting xanthomonads. Based on in silico analysis of the genome sequences of 66 Xam and 1 Xc strains, we were able to select 6 Xam and 6 Xc primers pairs candidates, of which one set of primers for each was selected for further studies. We were able to develop a duplex-PCR assay that was validated upon testing 53 Xam strains and 25 Xc strains from different countries, 18 non-target strains, and 5 epiphytic strains associated to cassava, proving this technique a useful tool to detect and differentiate Xam and Xc from in vitro cultures and in planta.Secondly we assessed the diversity of Xam populations through a variable number of tandem repeat analysis (MLVA). A field survey conducted in six states in Venezuela enabled to evaluate the occurence of the disease, its status and allowed us to establish a strain collection for detailed diversity analysis. We isolated 202 Xam strains from six localities, localized in four states. Using a MVLA14-scheme, we analyzed 12 populations highlighting a high index of genetic diversity among and within populations, mainly in the east of the country.The development of this type of research is essential in the management of crops in the world and coupled with the existing agricultural policies, it will allow us to have a deeper understanding of pathogens of agricultural importance and the mechanisms involved in their establishment over time and across regions. The long-term objective of this is to apply control measures that are effective in time, thus establishing more stringent quarantine measures to prevent the spread of the disease.

Study of defects in PV modules : UV fluorescence and Thermographic photography for Photovoltaics (PV) Field Application

For a PV plant it is of fundamental importance that the operation of the PV modules is free from faults or at least that the faults can be detected early, to ensure efficient electricity production. Some defects such as cracks can be seen in visible light while microcracks and damage to the silicon material can only be seen through special lighting. This study focuses on the most common defects in photovoltaic (PV) systems. Compare the infrared (IR) technology with the new ultraviolet (UV) fluorescence image technique for PV characterization, based on their accuracy and uncertainty factors under an experimental field investigation. In this study, first a literature study was conducted to the most common defects in PV system and their impact on electricity generation. Then a simulation model of a PV system was created in PVsyst and exported to Microsoft Excel which was used to evaluate how different defects at different stages of the PV cell's life cycle impact electricity generation, performance parameters and economic exchange. Furthermore, experiments with UV and IR was implemented at a PV system located in Dalarna and some PV modules at MDH. It was conducted that occurrence of snail tracks, delamination and hot spots in combination with bypass failures and non-functioning cell will affect the economic profitability in the long run and the payback time will increase since their impacts on electricity generation and performance parameters are huge. The worst case is when PV modules are affected by the fault in bypass diode and non-functioning cell which result to a payback time longer than the module's lifetime and huge amount electricity losses in different bypass diodes configurations. Since UV and IR are two different methods that are performed in two different ways, different errors occurred during the measurements. The biggest external factor was the weather that determined if the experiment could be implemented. The IR method gave decent results and was quicker to use, but the UV method highlighted some defect which could not be seen with the IR technology.

A captação de maltose em bactérias é feita por um sistema transportador do tipo ABC composto por uma proteína ligadora de substrato (MalE), duas proteínas transmembrana e uma ATPase. No presente trabalho descrevemos a clonagem, expressão e análise bioquímica e estrutural da proteína MalE da bactéria fitopatogênica Xanthomonas axonopodis pv citri (Xac) O gene malE de Xac foi clonado em vetor de expressão pET28a, a proteína recombinante foi expressa em Escherichia coli e purificada por cromatografia de afinidade ao níquel. Amostras da proteína solúvel foram analisadas quanto à estrutura secundária, interação com possíveis ligantes, estabilidade frente a diferentes condições físico-químicas. Ensaios de cristalização possibilitaram a obtenção de cristais em diferentes condições, um deles apresentou grupo espacial P6122, mas não foi possível resolver a estrutura. Com base nas estruturas conhecidas de ortólogos de MalE, geramos um modelo estrutural para a proteína de Xac e foram feitas análises quanto à interação com trealose e maltose. Modelos estruturais dos componentes transmembrana (LacF e LacG) e ATPase (UgpC) do sistema transportador de maltose de Xac também foram gerados. Os resultados representam uma contribuição importante para o conhecimento sobre a fisiologia e sistemas de transporte de Xac. / Maltose uptake in bacteria is mediated by an ABC transporter comprising a substrate binding protein (MalE), two transmembrane proteins, and one ATPase. In the present study, we describe the cloning, expression and biochemical as well as structural analyses of the MalE protein of the phytopagen Xanthomonas axonopodis pv citri (Xac). The malE gene of Xac was cloned in the pET28a expression vector, the recombinant protein was expressed in Escherichia coli and, subsequently, purified by nickel affinity chromatography. Samples of soluble protein were analyzed regarding secondary structure, interaction with putative ligants and stability under different physico-chemical conditions. Crystallization trials were carried out under different conditions, one particular condition yielded crystal with a P6122space group, but the structure was not solved. Based on known ortholog structures, a structural model for Xac MalE was obtained allowing interaction with modeled threhalose and maltose. Structural models the transmembrane (LacF and LacG) and ATPase (UgpC) components were also obtained. The present results represent an important contribution to the knowledge of the physiology and transporter systems found in Xac.

Contribution to renewable systems grid connected : control, stability analysis and reliability / Contribution aux systèmes renouvelables connectés en réseau : contrôle, analyse de la stabilité et fiabilité

Cette thèse a pour but l'analyse d'un système photovoltaïque connecté au réseau électrique en prenant en compte le contrôle, l'étude de la stabilité et la fiabilité. Un onduleur de type 2-Niveau a été comparé avec un onduleur multi-niveaux appelé Neutral Point Clamped. Les avantages et désavantage de chaque topologie ont été analysé en considérant l'efficacité énergétique, l'optimisation de l'injection de l'énergie sur le réseau électrique et la fiabilité du système. Pour le contrôle du courant de sortie de l'onduleur, ont été proposées deux solutions : un contrôle basé sur la théorie de la platitude et un contrôle par passivité. Ces deux différents contrôles sont comparés par rapport à la robustesse, la complexité et le nombre de capteurs utilisés. Il a été montré que les deux contrôles sont capables de gérer la problématique de la résonance du filtre LCL. Pour augmenter l'efficacité de l'algorithme MPPT dans une configuration Distributed-MPPT avec la connexion en série des deux sorties des convertisseurs boost, une nouvelle technique a été proposé pour l'équilibrage des tension d'entrée d'un onduleur NPC. En utilisant un outil appelé TPtool, un étude de la stabilité large signal par "Higher-Order-Singular-Value-Decomposition" a été présenté et comparé avec une méthode basée sur les modèles Takagi-Sugeno pour des systèmes non-linéaires. Finalement, l'onduleur 2-Niveaux est comparé avec deux multi-niveaux différents (NPP et NPC) en termes de disponibilité, en prenant en compte les niveaux de redondance des convertisseurs. Pour analyser la disponibilité, a été considérée la théorie des chaines de Markov et pour l'implémentation, le logiciel GRIF a été utilisé / The aim of this PhD thesis is to analyze a PV-grid connected system in terms of control, stability and reliability. A comparison between a classical 2-Level inverter and a multilevel NPC is presented. The advantages and weakness of both the converters are analyzed with respect to the power efficiency, optimization of the energy injection to the grid and reliability of the system. In order to control the inverter output current, two different solutions are proposed: flatness-based control and passivity-based control. These controls are compared in terms of robustness, complexity and number of sensors used. It is shown that both the controls may manage the resonance problems due to a LCL filter. For increasing the efficiency of the MPPT in a configuration Distributed-MPPT with connections in series of the boost converters outputs, a novel technique for the dc voltages balancing of a NPC inverter is proposed. A large stability analysis using "Higher-Order-Singular-Value-Decomposition" is presented and compared with Takagi-Sugeno approach for nonlinear systems. Finally, 2-Level inverter is compared with two multilevel inverters (NPC and NPP) in terms of availability, considering the redundancy levels of the converters. To analyze the systems availability, the Markov chains theory is considered and it is implemented on GRIF

Evaluation of a Flat-Plate Photovoltaic Thermal (PVT) Collector prototype

This Master thesis, in collaboration with Morgonsol Väst AB, was completed as a part of the Solar Energy engineering program at Dalarna University. It analyses the electrical and thermal performance of a prototype PVT collector developed by Morgonsol Väst AB. By following the standards EN 12975 and EN ISO 9806 as guides, the thermal tests of the collector were completed at the facility in Borlänge. The electrical performance of the PVT collector was evaluated by comparing it to a reference PV panel fitted next to it. The result from the tests shows an improved electrical performance of the PVT collector caused by the cooling and a thermal performance described by the linear efficiency curve ηth=0.53-21.6(Tm-Ta/G). The experimental work in this thesis is an initial study of the prototype PVT collector that will supply Morgonsol Väst with important data for future development and research of the product.

Bifacial photovoltaic systems established in a Nordic climate : A study investigating a frameless bifacial panel compared to a monofacial panel

The aim with this project was to study the power output from a frameless bifacial photovoltaic (PV) system relative to a traditional monofacial PV system with a frame. A general overview of how the geographical conditions affects the energy utilization of different PV systems is investigated throughout the project. Also, the study examined if further comparisons and evaluations, between PV systems, can be better established. The two examined solar parks, installed under different conditions, are located in Uppsala and Enköping, Sweden. In order to fulfill the aim and compare the different PV systems, three cases were analyzed. To increase the credibility of a comparison between the two cities, a sensitivity analysis considering the weather condition was executed. In case one, the result indicates that a bifacial panel is 5.2% and 3.6% more advantageous than a traditional monofacial panel during summer and winter, respectively. In case two, the frameless, more tilted and elevated bifacial panel is 58% and 680% more advantageous than a traditional monofacial panel during summer and winter, respectively. Also, in case three, the frameless, more tilted and elevated bifacial panel is 19% and 76% more advantageous than a bifacial panel with frame during summer and winter, respectively. When installing a new solar park, it is important to consider the location’s specific features since these affects the energy yield of the PV system. Future installations, which are installed with the intention to evaluate certain properties, is suggested to be installed with more initially comparable conditions in mind.

Investigating the autonomy of sensor nodes, and the dimensioning of a battery and a photovoltaic panel. For the validation of the solution, is used the sensor node developed in this work to collect data in external areas for periods of time. Applying strategies for dimensioning an energy system to aiming the needs of a sensor node in an external environment. An energy-saving strategy has been developed with sleep mode that allows higher work cycles to the NSS. The results are the finding that the NSS worked in the period, through the voltage level of the battery, which is related to the energy of the same. It can be affirmed that the NSS functioned and the objective of monitoring quantities in outdoor environments. / A disserta??o descreve o desenvolvimento de um elemento de rede sem fio aut?nomo, que possibilita a pesquisadores a coleta de informa??es em ambientes externos. Investigando a autonomia de n?s sensores, e o dimensionamento de uma bateria e de um painel fotovoltaico. Para a valida??o da solu??o, utiliza-se o n? sensor desenvolvido para a realiza??o de coletas de grandezas em ?reas externas por per?odos de tempo. Aplicando estrat?gias de dimensionamento de um sistema de energia visando as necessidades de um n? sensor em ambiente externo. Apresenta-se os resultados de estudo de coorte em um assentamento rural no Acre, Brasil, onde 509 indivíduos contribuíram com 489,7 pessoas-ano de seguimento. A incidência de malária por Plasmodium vivax e Plasmodium falciparum foi de 30,0/100 e 16,3/100 pessoas-ano, respectivamente. A morbidade por malária se associou fortemente ao desmatamento e agropecuária e diminuiu após 5 anos de residência no local; além disso ocorreram conglomerados espaciais significantes de casos de malária vivax e falciparum em áreas de ocupação recente. Não houve associação entre alótipos do receptor FcgRIIa, promotor Duffy ou anticorpos contra a PvMSP-1, e malária no seguimento. As respostas humorais e celulares foram mais freqüentes contra as porções C- e N-terminal da PvMSP-1 respectivamente, porém sem relação com as variantes de PvMSP-1 infectantes. O espectro clínico dos episódios de malária foi variado; enquanto cefaléia, febre e mialgia foram sintomas freqüentes, 29,4% dos episódios eram assintomáticos. Os resultados mostram que a aquisição de imunidade clínica é adquirida em áreas de baixa transmissão e que mudanças ambientais causadas nos assentamentos rurais perpetuam a transmissão de malária. Estes achados são de extrema importância para o controle da malária na Amazônia. / Here a cohort study in a frontier settlement in Acre, Brazil, where 509 subjects contributed 489.7 person-years of follow-up, is described. Incidence rates for Plasmodium vivax and Plasmodium falciparum malaria were 30.0/100 and 16.3/100 person-years at risk, respectively. Malaria morbidity was strongly associated with land clearing and farming, and decreased after 5 years of local residence. Besides, there was significant spatial clustering of vivax and falciparum malaria in areas of recent occupation. No significant association was found between FcgRIIa allotype, Duffy promoter type or presence of antibodies against PvMSP-1 and malaria in the follow-up. Humoral and cellular responses were more common against the C- and N-terminal portions of PvMSP-1 respectively, but did not match the PvMSP-1 variants found in infecting parasites. The clinical spectrum of malaria episodes varied widely; while headache, fever and myalgia were the most frequent symptoms, 29.4% of the episodes were asymptomatic. The results show that clinical immunity is acquired under low malaria transmission and environmental changes occurring in the settlements perpetuate malaria transmission. These findings are of utmost importance for malaria control in the Amazon.
