The role of chemistry and strut porosity and the influence of serum proteins in modulating cellular response to bone graft substitutes

The objective of this thesis was to investigate the role of hydroxyapatite and silicate-substituted hydroxyapatite synthetic bone graft substitute (SBG) material properties in modulating the processes of protein adsorption and desorption, and their combined role in the subsequent regulation of cell attachment, proliferation and differentiation on the surfaces of these materials in vitro. As a result of their purported role in promoting osteogenic behaviour in vivo the materials parameters selected for investigation were chemistry (stoichiometric hydroxyapatite (HA) versus 0.8wt% silicate-substituted hydroxyapatite (SA)) and strut porosity (20% versus 30% strut porosity). Cell attachment and response to different SBG was assessed to samples in the ‘as received’ condition as well as after a series of sequentially varied pre-treatments with solutions of phosphate buffered saline or cell culture media either unsupplemented or in combination with mixed serum proteins and/or Fibronectin (Fn). This enabled investigation of the effect of sample chemistry and strut porosity on mixed serum protein interactions and Fn adsorption under both competitive and non-competitive conditions, and the study of subsequent regulation of cell attachment and response as a consequence of pre-treatment. Results showed that serum protein interactions were key to modulation of cell response to chemistry, and there was evidence that for Fn this may be related to conformational changes in the adsorbed protein rather than its level of enrichment in the protein interlayer. In terms of the materials properties investigated strut porosity was found to be the most dominant factor in the regulation of cell response, where SBG with 30% strut porosity promoted human mesenchymal stem cell (hMSC) osteoblastic differentiation. Moreover hMSC response to SBG with 30% strut porosity seemed to be less sensitive to pre-treatment. In conclusion, the results of these experiments indicate that strut porosity more directly influences the cellular response to HA and SA BGS than chemistry in vitro. Moreover, the role that Fn and other serum proteins have in regulating this response is dependent on the physiological environment and BGS chemistry.

Cette thèse concerne l’auto-assemblage d'alcanes semi-fluorés (FnHm) et de fluorocarbures (FCs) aux interfaces et leurs interactions avec des phospholipides (PLs) en 2D et 3D. Nous avons étudié les sphérulites formées dans des films de diblocs FnHm. La morphologie de ces sphérulites, concentrique ou radiale, est contrôlée par la longueur des blocs Fn et Hm et la vitesse de refroidissement. Les nanodomaines de diblocs FnHm dans les monocouches de Langmuir sont incompressibles et forment des gels physiques 2D, même à pression nulle. Les blocs Hm sont cristallisés et inclinés d’ ~30°C par rapport à la normale à la surface. La réflectivité de neutrons a montré que l’albumine adsorbée sur des monocouches de PL est désorbée par un FC gazeux. Ce résultat pourrait permettre de lutter contre l’inactivation du surfactant pulmonaire par les protéines sériques. L’ajout de diblocs à des films de PLs accroit l'élasticité des monocouches. Nous avons préparé des microbulles stables à parois de PLs/FnHm, les diblocs agissant en co-surfactants. / This thesis focuses on the self-assembly of semi-fluorinated alkanes (FnHm) and fluorocarbons (FCs) at interfaces and their interactions with phospholipids (PLs) in 2D and 3D. 2D Spherulites were identified in FnHm films for the first time. Their morphology, ring-banded or radial, was controlled by varying block lengths and cooling rate. Nanodomains of FnHm in monolayers formed incompressible 2D physical gels, even at zero surface pressure. The Hm segments are crystalline and titled by 30°C to the normal to the surface. Neutron reflectivity showed that albumin adsorbed on PLs monolayers is desorbed by exposure to FC gas, which opens the potential use of FCs to treat the inactivation of the lung surfactant by serum proteins. Incorporating FnHm into PL monolayers increases their elasticity. Small, stable microbubbles of PLs/FnHm were obtained. FnHm diblocks function as co-surfactants for stabilizing microbubbles.

Etude des petits ARNs extracellulaires pour le diagnostic de cancer du rein à cellules claires / Study of circulating small RNAs for diagnostic clear cell renal cell carcinoma

Le cancer du rein est un problème majeur de santé publique. Un diagnostic précoce améliore les chances de survie. Le diagnostic repose essentiellement sur les examens d’imagerie comme l’échographie, la tomodensitométrie et l’IRM. Ces examens sont parfois associés à la biopsie et sont couteux et parfois invasifs. De plus, l’imagerie n’est pas capable de faire la distinction entre les tumeurs bénignes et les tumeurs malignes et entre les sous-types histologiques de carcinome à cellules claires qui est le plus fréquent. Il n’existe pas dans le cancer rénal de marqueur comme la PSA dans le cancer de la prostate ou la Foetoprotéine et l’HCG dans le cancer du testicule. Le but de cette étude est concentré sur la recherche des marqueurs mARNs ou miARNs dans les liquides biologiques (sérum, plasma, urine) pour le cancer du rein à cellules claires. Nous avons montré que les petits ARNs dans le sérum et les urines et l’intégrité des ARNs dans les urines étaient des outils diagnostiques dans le cancer du rein à cellules claires. Si ces petits ARNs circulants sont validés, on peut éventuellement imaginer l’intérêt pratique en clinique comme la détection des petits ARNs circulants dans l’urine pour prédire le cancer, la classification de la tumeur pour aider le clinicien, la prédiction d’une récidive ou d’une progression soit après néphrectomie soit au cours d’un traitement médical / Kidney cancer is a major public health problem. Early diagnosis improves the chances of survival. The diagnosis is mainly based on the imaging tests such as ultrasound, CT and MRI. These tests are sometimes associated with biopsy and are expensive and sometimes invasive. In addition, the imaging test is not able to distinguish between benign and malignant tumors and between histological subtypes of renal cell carcinoma. There don’t exist tumor marker in renal cancer such as PSA in prostate cancer or Fetoprotein and HCG in testicular cancer. For this purpose, the study has focused on finding markers mRNAs and miRNAs in biological fluids (serum, plasma, urine) for clear cell renal cell carcinoma. We have shown that the small cell-free RNAs in serum and urine and the integrity of cell-free RNA in urine were diagnostic tools for clear cell renal cell carcinoma. If these circulating small RNAs are validated, we can possibly imagine the interest in clinical practice as the detection of circulating small RNAs in urine to predict cancer, the classification of the tumor to help the clinician, the prediction of recurrence and progression after nephrectomy or during medical treatment

A Fotoquimioterapia (FQT) vem sendo usada no tratamento de várias doenças, inclusive câncer, com a vantagem de ser menos agressiva, possuindo efeitos colaterais negativos desprezíveis. Esta técnica envolve a combinação de uma fonte de luz visível e de um fotossensibilizador (FS), que é reativo apenas sob a irradiação com luz adequada, e que se acumula preferencialmente no tecido doente. Uma ideia para deixar essa técnica mais eficiente é potencializar esta última característica do FS com uso de um carreador que tenha afinidade com tecidos doentes. Pensamos na hipótese de usar a albumina do soro como carreador uma vez que ela é uma proteína intrínseca do organismo, tem alta afinidade com os tecidos tumorais e possui sítios para ligar vários compostos extrínsecos. Como modelo usamos a albumina de soro bovino (ASB). Como FS utilizamos acridina laranja, nitrofurantoína, corantes ciânicos com dois cromóforos e protoporfirina IX (PpIX) sintética e extraída de glândulas Harderianas de ratos da linhagem Wistar (PpIX endógena). Foram analisadas as características de ligação entre os FS e a ASB e seus efeitos nas características fotofísicas dos FS em função da sua estrutura, utilizando espectroscopias de absorção ótica, fluorescência estática e resolvida no tempo e a técnica de microscopia de imagens por tempo de vida da fluorescência (FLIM). Foi mostrado que a ligação com ASB induz ao deslocamento batocrômico dos espectros de absorção e da fluorescência, aumentando a intensidade e o tempo de vida de fluorescência dos FS. Características como carga positiva, estrutura linear e tamanho pequeno da molécula do FS favorecem sua ligação com a ASB. Uma comparação entre PpIX sintética e endógena mostrou que a formação de um complexo da PpIX com estruturas anfifílicas também aumenta a probabilidade da sua ligação com a ASB. Os resultados obtidos mostraram que a ASB se liga efetivamente aos FS de diferentes estruturas e pode ser usada como carreador dos mesmos para tecidos tumorais. / Photochemotherapy (PCT) is used for the treatment of various diseases, including cancer, with the advantage of being less aggressive with reduced adverse effects. This technique includes the use of a visible light source and a photosensitizer (PS), which is reactive only under light exposure and localizes preferably in abnormal tissues. To make this technique more efficient it is necessary to increase the PS content in these tissues using a carrier which possesses affinity with these tissues and is able to bind PS. A good opportunity is to use serum albumin as a carrier since it is an intrinsic protein, has high affinity to tumor tissues and possesses sites for binding of various extrinsic compounds. In this work the bovine serum albumin (BSA) was used as a carrier model. Acridine orange, nitrofurantoin, cyanine dyes with two chromophores and protoporphyrin IX synthetic and extracted from Harderian glands of Wistar line rats (endogenous) were used as PS. The characteristics of the PS binding with BSA and the effect of this interaction on PS photophysical properties were studied using optical absorption, steady-state and time resolved fluorescence spectroscopies, and fluorescence lifetime image microscopy (FLIM). It was demonstrated that PS binding with BSA produces a bathochromic shift in the PS absorption and fluorescence spectra and increases its fluorescence intensity and lifetime. The positive charge, linear structure and small size of the PS molecule favors its binding with BSA. A comparison between synthetic and endogenous PpIX demonstrates that complexation of PpIX molecule with an amphiphilic structure increases the probability of its binding with BSA. The obtained results demonstrate that BSA binds effectively PS with different molecular structures and it is promising for their transport to tumor tissue.

Ewes subjected to the same semi-intensive nursing system were followed up. The animals were evaluated according to the following stages: 10 days before parturition (dbp) and 15 days postpartum (dpp), 30 (dpp), 60 (dpp), and 90 (dpp). Blood metabolites were evaluated starting from the stage previous to parturition and whey metabolites were evaluated in the subsequent stages. A screening of the ewes followed up in this study (12 healthy and 18 with subclinical mastitis) was performed after a clinical and bacteriological examination. During lactation, maintaining the same screening criteria, 11 healthy and 20 infected mammary glands were selected; the milk for whey extraction was collected from these glands. Energy profile metabolites (non-esterified fatty acids [NEFAs], ß-hydroxybutyrate [BHB], fructosamine, cholesterol and triglycerides), protein profile (total protein, albumin, urea and creatinine) and mineral profile (iron, copper, zinc, magnesium, total calcium, ionized calcium, sodium, and potassium) were measured in the blood serum. Calcium, sodium and potassium ions, as well as NEFAs and BHB were measured in the whey. Blood biochemistry revealed an influence (P<0,05) of the peripartum and lactation periods on the blood concentrations of NEFAs, BHB, cholesterol, albumin, urea, ionized calcium. An analysis of the whey also revealed an influence on the potassium ion. Ewes with subclinical mastitis showed higher (P<0,05) blood levels of cholesterol, albumin and copper; higher sodium ion concentrations and NEFAs, and lower potassium ion in whey. Good physical score of ewes observed during this study, combined with the biochemical findings, allowed us to conclude that there was a larger energy requirement in the first month of lactation; however, this requirement was not enough to trigger any metabolic disorder or the emergence of ketonemia, and these discrete changes were more apparent in ewes with subclinical mastitis. / Este estudo teve por objetivo avaliar o metabolismo energético, proteico e mineral de ovelhas Santa Inês hígidas e com mastite subclínica acompanhadas durante o final da gestação e na lactação. Foram acompanhadas ovelhas submetidas ao mesmo sistema de criação semi-intensivo. Os animais foram avaliados conforme os momentos a seguir: 10 dias que precedeu o parto (dap) e 15 dias após o parto (dpp), 30 (dpp), 60 (dpp) e 90 (dpp). Os metabólitos sanguíneos foram avaliados a partir do momento que antecedeu ao parto e os metabólitos no soro lácteo nos momentos subsequentes. Após exame clínico e bacteriológico foi realizada a triagem das ovelhas acompanhadas neste estudo, sendo 12 hígidas e 18 com mastite subclínica. Durante a lactação, mantendo os mesmos critérios de triagem, foram selecionadas 11 glândulas mamárias sadias e 20 infectadas, das quais foi colhido o leite para obtenção do soro lácteo. Foram mensurados no soro sanguíneo os metabólitos do perfil energético (ácidos graxos não esterificados (AGNEs), ß-hidroxibutirato (BHB), frutosamina, colesterol e triglicérides), ao perfil proteico (proteína total, albumina, uréia e creatinina) e ao perfil mineral (ferro, cobre, zinco, magnésio, cálcio total, cálcio ionizado, sódio e potássio). No soro lácteo foram mensurados os íons cálcio, sódio e potássio, bem como dos AGNEs e BHB. A bioquímica sanguínea revelou haver influência (P<0,05) do período do periparto e da lactação sobre as concentrações sanguíneas dos AGNEs, BHB, colesterol, albumina, uréia, cálcio ionizado e no soro lácteo sobre o íon potássio. As ovelhas portadoras de mastite subclínica apresentaram valores sanguíneos superiores (P<0,05) de colesterol, albumina e cobre e no soro lácteo teores superiores do íon sódio e dos AGNEs e inferiores do íon potássio. O bom escore corporal das ovelhas observado durante o estudo aliado aos achados bioquímicos permitiu concluir ter ocorrido maior requerimento energético no primeiro mês da lactação, porém não o suficiente para desencadear qualquer transtorno metabólico e o aparecimento de um quadro de cetonemia, sendo estas discretas alterações mais expressivas nas ovelhas com mastite subclínica.

Análise das dosagens do fator de crescimento endotelial vascular no plasma e fluidos peritoneais de pacientes com câncer epitelial de ovário / Analysis of measurements of vascular endothelial growth factor in plasma and peritoneal fluids of patients with epithelial ovarian cancer

A carcinogênese epitelial ovariana tem sido foco de estudos científicos em todo o mundo desenvolvido. A angiogênese tumoral ovariana é um processo multifatorial que resulta em vários produtos pró-angiogênicos. Entre eles, o fator de crescimento vascular endotelial (VEGF) é predominante. Os objetivos deste estudo foram relacionar as dosagens do VEGF dos fluidos peritoneais, do plasma periférico e do infundíbulo pélvico aos níveis de citorredução em pacientes operadas de adenocarcinoma epitelial de ovário (CEO); formular um modelo probabilístico de citorredução e utilizar estas dosagens para estimar a probabilidade do desfecho de citorredução. Além disto, foi criada uma nova variável chamada carga de VEGF. Pelo procedimento step-wise a citorredução foi melhor descrita pela carga de VEGF, mas faltou Normalidade aos resíduos, não sendo possível a adequação de um modelo matemático. Porém, a curva ROC, forneceu uma área sob a curva de 0,84, com sensibilidade de 71,4 % e especificidade variando de 69,5 a 73,9%. O ponto de corte ótimo foi 15,52 log de picograma de carga de VEGF. A odds-ratio (OR) calculada para citorredução ótima descrita pela carga de VEGF foi de 11 (IC= 2,59 ; 46,78). No grupo com estágio avançado (III e IV), a OR foi de 6 (IC= 1,15; 31,22). Apesar do pequeno número de casos, esta nova variável pode vir a ser um auxiliar na determinação de situações onde cirurgia citorredutora deixa de ser a pedra fundamental do tratamento primário do CEO e a indução quimioterápica passe a ter o principal papel na citorredução química antes da cirugia nestes casos. / Epithelial ovarian carcinogenesis has been the focus of scientific studies in developed world. The ovarian tumor angiogenesis is a multifactorial process that results in pro-angiogenic products. Among them, vascular endothelial growth factor (VEGF) is predominant. This study aimed to relate VEGF levels in peritoneal fluids, peripheric plasma and pelvic infundibular plasma to debulking levels in patients operated on for epithelial ovarian carcinoma (EOC), formulate a probabilistic model for debulking and use these measurements to estimate the probability of the outcome of debulking. Moreover, it was created a new variable called burden of VEGF. For step-wise procedure, cytoreduction was better described by of burden VEGF, but missed Normality of residuals, so the adequacy of a mathematical model was not possible. Nevertheless, the ROC curve provided an area under the curve of 0.84, with sensitivity of 71.4 % and especificity varying from 69.5 to 73,9%. The optimal cutoff point was 15.52 log of picograms of VEGF burden. An odds-ratio for optimal cytoreduction described by the VEGF burden was 11 (CI= 2.59; 46.78). In the group with advanced stages (III & IV), the OR was 6 (CI= 1.15; 31.22). Apart from the small number of cases, this new variable might help to determine situations where cytoreductive surgery leaves behind the cornerstone of primary treatment of CEO and the chemotherapic induction comes to have main role in chemical cytoreduction prior to surgery in these cases.

Tratamento do linfoma canino com poliquimioterapia seguida ou não de transplante autólogo de medula óssea /

Resumo: Dezoito cães com diagnóstico citopatológico e/ou histopatológico de linfoma foram avaliados quanto às alterações clínicas e clinico-patológicas. Os animais foram submetidos ao protocolo quimioterápico de Madison-Wisconsin e avaliados clinicamente a cada sessão de quimioterapia quanto à sua resposta ao tratamento. Cinqüenta por cento dos cães apresentaram a forma multicêntrica da doença e 33% a forma cutânea. A manifestação clínica mais comum foi a linfadenomegalia superficial, acompanhada dos sinais sistêmicos de hiporexia, apatia e perda de peso. As principais alterações hematólogicas foram anemia normocítica normocrômica, trombocitopenia e leucocitose, associadas às síndromes paraneoplásicas. Elevações nas atividades séricas de lactato desidrogenase foram observadas em 58% dos cães e a hiperproteinemia ocorreu em 38% dos animais, sendo atribuída principalmente ao aumento de globulinas séricas. Vinte e sete por cento dos animais atingiram remissão completa da doença e 33% atingiram sobrevida de seis meses. Houve correlação positiva entre o valor do hematócrito e o tempo de sobrevida para os cães que morreram, entretanto, o mesmo não foi observado para o valor de lactato desidrogenase. Sugere-se a ocorrência da síndrome da lise tumoral aguda para sete animais dado o intervalo de tempo observado entre a quimioterapia e o óbito e o avançado estágio da doença. A ausência de resposta às tentativas de reindução da remissão, observada em quatro cães, pode estar relacionada com o fenômeno de resistência tumoral aos agentes quimioterápicos. / Abstract: Clinical and laboratorial evaluation were performed in eighteen dogs with cytopathological and/or histopathological lymphoma diagnosis. Animals underwent the Madison-Wisconsin chemotherapy protocol and were evaluated clinically at each session performed weekly. The multicentric and cutaneous forms of the disease were observed in 50% and 33% of the dogs, respectively. The most common clinical sign was superficial lymphadenomegaly, combined to systemic signs of hyporexia, apathy and weight loss. The main hematological changes were anemia, thrombocytopenia and leukocytosis, associated with paraneoplastic syndromes. Elevated serum activity of lactate dehydrogenase was observed in 58% of the dogs and hyperproteinemia in 38%, attributed to serum globulin increase. Complete remission was achieved in 27% of the animals and the six-month survival rate was 33%. There was a positive correlation between the hematocrit and survival time in dogs that died, however, there was no correlation between lactate dehydrogenase and survival time. The time interval between chemotherapy and death and the advanced stage of the disease in seven animals suggest the occurrence of acute tumor lysis syndrome. Funcionalização da superfície de nanopartículas superparamagnéticas encapsuladas por quitosana para a imobilização de proteínas / Surface functionalization of superparamagnetic nanoparticles encapsulated by chitosan for protein immobilization

A nanociência e a nanotecnologia vêm abrindo inúmeros desenvolvimentos de dispositivos e sistemas em escala nanométrica, com novas organizações moleculares, propriedades e funções distintas. Nesse contexto, as nanopartículas magnéticas poliméricas são compósitos formados por materiais magnéticos com tamanhos de partículas entre 1 e 100 nm combinados com polímeros funcionais. São materiais bem conhecidos e têm sido amplamente estudados devido às suas aplicações em diversas áreas tecnológicas. Nas áreas biológica e médica, as aplicações incluem separação e imobilização de enzimas e proteínas, melhoria nas técnicas de imagem de ressonância magnética para diagnóstico e sistemas de liberação controlada de fármacos. Neste trabalho, proteínas foram imobilizadas na superfície de um biopolímero combinado com partículas superparamagnéticas de magnetita para formar o compósito magnético. Utilizou-se o biopolímero quitosana, reticulada e funcionalizada com glutaraldeído, aplicável em ensaios biológicos. Obtiveram-se 3 tipos de compósitos magnéticos, os quais foram nomeados QM1Glu, QM2NaGlu e QM3Glu. Foram caracterizados por difratometria de raios X, microscopia eletrônica de varredura, magnetometria de amostra vibrante, calorimetria exploratória diferencial, termogravimetria e espectroscopia por infravermelho. Foram avaliados quanto à imobilização das proteínas albumina de soro bovino (SAB), colágeno e tripsina. A imobilização das proteínas no biopolímero ocorreu em 30 min de incubação. O compósito magnético de quitosana não funcionalizada (QM3) também foi avaliado. Para a tripsina verificou-se que QM3 apresentou maior potencial de imobilização do que QM3Glu. Após 30 dias, QM3-Trip e QM3Glu-Trip ainda apresentavam a tripsina ativada. Foram demonstradas a atividade e a cinética enzimática da QM3Glu-trip com o substrato BApNA. / Nanoscience and nanotechnology have opened up numerous developments of devices and systems on the nanometer scale, with new molecular organization, properties and functions. In this context, the polymeric magnetic nanoparticles are composites formed by magnetic materials with a particle size between 1 and 100 nm combined with functional polymers. They are well-known and have been widely studied because of its applications in various technology areas. Applications on the biological and medical areas include separation and immobilization of enzymes and proteins, improved techniques of magnetic resonance imaging and diagnostic systems for controlled drug delivery. In this work, proteins were immobilized on the surface of a biopolymer combined with superparamagnetic particles of magnetite. The biopolymer chitosan was used, cross-linked and functionalized with glutaraldehyde, applicable to the biological assays. Three types of magnetic composites were obtained, which were called QM1Glu, QM2NaGlu and QM3Glu. They were characterized by X-ray diffraction, scanning electron microscopy, vibrating sample magnetometry, differential scanning calorimetry, thermogravimetry and infrared spectroscopy. They were evaluated concerning the immobilization of the proteins bovine serum albumin (BSA), collagen and trypsin. The study showed that the immobilization of proteins on the biopolymer occurred in 30 min of incubation. The magnetic composite of nonfunctionalized chitosan (QM3) was also evaluated. For trypsin, it was found that the immobilization potential of QM3 was higher than that observed for QM3Glu. After 30 days, the trypsin of the QM3-Trip and QM3Glu-Trip was still with activity. The activity and the enzyme kinetics of the QM3Glu-Trip with the substrate BApNA were demonstrated.

Aspectos hematológicos, bioquímicos, morfológicos e citoquímicos de células sangüíneas em Viperídeos neotropicais dos gêneros Bothrops e Crotalus mantidos em cativeiro / Hematological, biochemical, morphological and cytochemical aspects of blood cells in neotropical Viperidae from Bothrops and Crotalus genus mantained in captivity

Este estudo apresenta um painel de dados hematológicos, bioquímicos, morfológicos e citoquímicos para um grupo de serpentes dos gêneros Bothrops e Crotalus mantidas em cativeiro no Instituto Butantan. Para tanto, amostras de sangue foram colhidas da veia coccígea ventral e processadas de acordo com métodos padronizados. Os resultados alcançados foram analisados para determinar diferenças interespecíficas e sexuais. A avaliação citoquímica incluiu Sudan Black B (SBB), Ácido Periódico de Schiff (PAS) e Benzidina Peroxidase (BP). Os resultados hematológicos e bioquímicos mais relevantes indicaram um número significativamente mais alto de basófilos em B. jararaca e níveis mais altos de cálcio nas fêmeas da maioria das espécies. Os azurófilos marcaram positivamente para todas as colorações. A diferenciação entre trombócitos e linfócitos foi facilmente obtida com PAS. Os basófilos marcaram positivamente apenas com PAS em Bothrops alternatus. A marcação em heterófilos variou consideravelmente entre as espécies. Ultra-estruturalmente, os leucócitos foram semelhantes ao já descrito em literatura com algumas variações no tipo dos grânulos dos basófilos e heterófilos. Os grânulos dos basófilos apresentaram-se redondos em sua maioria, com tamanho e densidade variadas, enquanto que os nos heterófilos foram heterogêneos em forma, tamanho e densidade. / This study reports a panel for hematological, biochemical, morphological and cytochemical data for a group of captive snakes belonging to the genus Bothrops and Crotalus mantained at Instituto Butantan, São Paulo, Brazil. Blood samples were collected from ventral coccigeal vein and were processed according to standard protocols. Cytochemical staining including Sudan Black B (SBB), Periodic Acid-Schiff (PAS) and Benzidine peroxidase (BP) were conducted. Blood values were evaluated to determine interspecific and sex differences. We found a significant increase of basophils in Bothrops jararaca and higher levels of calcium in females. Azurophils stained positively for all stains and a differentiation between lymphocytes and thrombocytes was easily obtained with PAS stain. Basophils stained positively only with PAS in Bothrops alternatus. Heterophils staining varied between species. The ultrastructure of leucocytes were similar within described in literature, with some variations on granules of basophils and heterophils. Basophils granules were round, with heterogeneous size and density; whereas, heterphils granules were heterogeneous in shape, size and density.

Efeito da liofilização sobre a estrutura e mudanças de fase da albumina bovina modificada por reação com metoxi-polietilenoglicol / Effect of lyophilization on the structure and phase changes of PEGylated-bovine serum albumin.

A conjugação por polietilenoglicol (PEG) mascara a superfície das proteínas e aumenta o tamanho molecular do polipeptídio, reduzindo assim sua ultrafiltragem renal, impedindo a aproximação de células processadoras de antígenos ou anticorpos e reduzindo a degradação por enzimas proteolíticas. O PEG transfere para as moléculas suas propriedades físico-químicas e, conseqüentemente, modifica também a biodistribuição e a solubilidade de drogas peptídicas e não peptídicas. As soluções de proteínas são facilmente desnaturadas (muitas vezes irreversivelmente) pelo aparecimento de numerosos eventos que podem afetar a estabilidade das soluções, tais como: aquecimento, agitação, congelamento, mudanças no pH e exposição a interfaces ou agentes desnaturantes, resultando geralmente na perda da eficácia clínica e aumento do risco de efeitos colaterais adversos. A solução prática para o dilema da estabilidade da proteína é a remoção da água. A liofilização é o método mais comumente utilizado para a preparação de proteínas desidratadas, as quais, teoricamente, devem apresentar uma estabilidade adequada por um longo período de armazenagem em temperaturas ambientes. A proteína utilizada neste estudo foi a albumina sérica bovina (BSA), amplamente estudada no campo da bioquímica. Através da espectroscopia Raman associada com análise térmica por DSC, análise colorimétrica, e a determinação do teor de umidade, verificou-se que o congelamento rápido (30 &#176;C/min.) favoreceu a manutenção da estrutura conformacional da proteína após a liofilização, porém aumentou o tempo de secagem primária em sete horas em relação ao congelamento lento (2,5 &#176;C/min.). Após a modificação da albumina bovina por reação com o metoxi-PEG verificou-se que a BSA-PEG (1:0,25) apresentou um menor grau de alteração estrutural e conseqüentemente uma menor variação das características físico-químicas, além de otimizar as condições de liofilização e armazenamento da proteína quando comparada com a BSA-PEG (1:0,5) . / PEG conjugation masks the proteins surface and increases the molecular size of the polypeptide, thus reducing its renal ultrafiltration, preventing the approach of antibodies or antigen processing cells and reducing the degradation by proteolytic enzymes. The PEG conveys to molecules its physico-chemical properties and therefore modifies also biodistribution and solubility of peptide and non-peptide drugs. This property opens new techniques in biocatalysis and in pharmaceutical technology where many insoluble drugs are solubilized by PEG conjugation and thus more easily administered. Aqueous protein solutions are readily denatured (often irreversibly) by numerous stresses arising in solution, e.g., heating, agitation, freezing, pH changes, and exposure to interfaces or denaturants, usually resulting in lost of clinical efficacy and increase the risk of adverse side effects. Even if its physical stability is maintained, a protein can be degraded by chemical reactions (e.g., hydrolysis and deamidation), many of which are mediated by water. The practical solution to the protein stability dilemma is to remove the water. Lyophilization is most commonly used to prepare dehydrated proteins, which, theorecally, should have the desired long-term stability at ambient temperatures. The protein used in this study was the bovine serum albumin (BSA), largely studied in the biochemical field. Through Raman spectroscopy associated with thermal analysis using DSC, Colorimetric analysis and the determination of water content It was observed that the fast freezing (30 &#176;C/min.) favored the maintenance of the conformational structure in the protein after lyophilization, however increased the primary drying in seven hours with regard to the slow freezing (2,5 &#176;C/min.). After the modification of bovine serum albumin with methoxy-PEG it was observed that the BSA-PEG (1:0,25) showed a lower degree of structural alterations and consequently a lower variation on the physical-chemical characteristics, moreover optimized the conditions during the lyophilization process and storage of the protein when it was compared to BSA-PEG (1:0,5).
