The miltefosine (MT) is an alkylphospholipid that has been approved for the treatment of breast cancer metastasis and visceral leishmaniasis, although the mechanism of action at the molecular level is poorly understood. Electron paramagnetic resonance (EPR) spectroscopy of the lipid spin lebel analog of stearic acid (5-DSA) and the maleimide derivative spin label (6-MSL) covalently bound to membrane proteins showed that the MT causes a large increase in the molecular dynamics of erythrocyte membranes (ghosts) and detergent resistant membranes (DRMs) prepared from erythrocyte membranes. In the vesicles of lipid raft constituents, it was shown that 20 mol% sphingomyelin could be replaced by 20 mol% MT with no change in the molecular dynamics. Furthermore, the effect of MT on DRMs was more pronounced than in erythrocyte ghosts, supporting the hypothesis that MT is a lipid raft modulator. At the reported MT-plasma concentrations found during the treatment of leishmaniasis (31-52μg/mL), our measurements in blood plasma indicated a hemolytic level of 2-5% and also showed that the MT concentration that changes the erythrocyte membrane fluidity to an extent that is detectable by EPR spectroscopy causes about 46% hemolysis. Subsequently, EPR studies performed with the same spin labels in the membrane of Leishmania (L.) amazonensis (promastigote) showed changes similar to those found in erythrocyte membranes. Cytotoxic effects on the parasites were also evaluated to investigate the relationships between the cytotoxic potential of MT and its ability to alter membrane fluidity. The EPR data showed that the minimum concentration of MT required to cause a change in the parasite membrane occurred near the values of MT concentration which inhibits 50 % of cell growth (IC50); thus, there is a correlation between the cytotoxicity and changes in the membrane. Although these III membrane alterations can be detected using a spin-labeled lipid, our experimental results indicated that MT interacts predominantly with the protein component of the membrane. Cell lysis was also detected by analyzing the supernatants of centrifuged samples for the presence of spin-labeled membrane fragments and cytoplasmic proteins. Using a method for the rapid incorporation of MT into the membrane, these effects were measured immediately after treatment under the same range of MT concentrations that cause cell growth inhibition. Cytotoxicity, estimated via microscopic counting of living and dead cells, indicated ∼ 70% cell death at the concentration of MT at which EPR spectroscopy detected a significant change in membrane dynamics. After this initial impact on the number of viable parasites, the processes of cell death and growth continued during the first 4 h of incubation. The EPR spectra of spin-labeled membrane-bound proteins were consistent with more expanded and solvent-exposed protein conformations, suggesting a detergent-like action. Thus, MT may form micelle-like structures around polypeptide chains, and proteins with a higher hydrophobicity may induce the penetration of hydrophilic groups of MT into the membrane, causing its rupture. / A leishmaniose cut ˆanea ´e uma doenc¸a tropical negligenciada que afetamilh˜oes de pessoas em todo mundo, representando um grave problema de sa´ude p´ublica. Os dados de RPE demonstraram que a concentrac¸ ˜ao m´ınima necess´aria de MT para causar alterac¸ ˜ao na membrana do parasito ocorreu pr ´oximo dos valores de concentrac¸ ˜ao que inibe 50% do crescimento celular (IC50), existindo, portanto, uma correlac¸ ˜ao entre as alterac¸ ˜oes na membrana e a citotoxicidade. Embora o marcador de spin lip´ıdico tamb´em tenha detectado alterac¸ ˜oes na fluidez da membrana, os experimentos de RPE indicaram que a MT atua predominantemente no componente prot ´eico da membrana. A lise celular tamb´em foi detectada atrav´es da an´alise dos sobrenadantes das amostras centrifugadas onde foram encontrados fragmentos de membranas marcadas e tamb´em prote´ınas citoplasm´aticas. Usando um m´etodo para a r ´apida incorporac¸ ˜ao da MT na membrana, os efeitos de citotoxidade foram medidos imediatamente ap´os o tratamento e na mesma faixa de concentrac¸ ˜oes de MT que causam a inibic¸ ˜ao do crescimento celular. A citotoxicidade, estimada atrav´es da contagem microsc´opica das c´elulas vivas e mortas, indicou ∼ 70% de morte celular na concentrac¸ ˜ao de MT em que a espectroscopia de RPE detectou uma alterac¸ ˜ao significativa na dinˆamica da membrana. Ap´os o primeiro impacto no n´umero de parasitas vi ´aveis, o processo de morte e crescimento celular continuou durante as primeiras quatro horas de incubac¸ ˜ao. Os espectros de RPE de prote´ınas de membrana marcadas com o 6-MSL foram consistentes com conformac¸ ˜oes de prote´ına mais expandidas e expostas ao solvente, sugerindo uma ac¸ ˜ao t´ıpica dos detergentes. Nossa interpretac¸ ˜ao ´e que a MT pode formar estruturas semelhantes das micelas ao redor das cadeias polipept´ıdicas, expandindo as prote´ınas e que as prote´ınas com maior hidrofobicidade poderiam induzir a penetrac¸ ˜ao de grupos hidrof´ılicos da MT no interior da membrana, causando sua ruptura e consequentemente a morte da c´elula.

Were analyzed the effect of these two methods as well as the effect of additional thermal treatment of the samples prepared by the combustion method, on the physical properties of YCrO3 by several spectroscopic and structural techniques: X-ray diffraction (XRD), scanning electron microscopy and transmission (SEM and TEM), Fourier transform spectroscopy and infrared, diffuse reflectance, susceptibility and electron paramagnetic resonance (EPR). Images (SEM) show that the powders YCrO3 synthesized by the two methods of synthesis have similar morphologies, with average grain size of approximately 160 nm. Through the images was found that applying heat treatment to the sample obtained by combustion synthesis at the same temperature as the precursor obtained by the polymeric precursor method, the structural characteristics of the material are similar. The spectra diffuse reflectance of the different samples exhibit two intense bands, which correspond to electronic transitions of ion Cr3+ from the ground state 4A2g to the excited states, 4A2g → 4T2g and 4A2g → 4T1g two minor bands corresponding to the transitions 4A2g → 2Eg and 4A2g → 2T1g were observed in the spectra. Were calculated from the spectra the crystal field parameters Dq and Racah, B and C for all samples, with average values of 3.03 for Dq, 542 cm-1 and 3,144 cm-1 for B and C respectively. Even with these spectra and the theory of Kubelka-Munk was calculate the optical gap Eg, with a mean value of 3.7 eV, coinciding with those reported in the literature for YCrO3 prepared by other methods. The dependence of the spectrum parameters (EPR), factor g, intensity, and line width as a function of temperature was also studied from 100 K to 300 K. The results show that, near 140 K, the Néel temperature for the material, a magnetic phase transition occurs from the antiferromagnetic phase to the paramagnetic phase, in full agreement with the results obtained from measurements of susceptibility. Our results are discussed and compared with other reported in the literature. / Neste trabalho descreve-se a síntese da ortocromita de ítrio YCrO3 policristalina por dois diferentes métodos: precursores polímericos modificado (MPP) e combustão em solução (CS) com e sem tratamento térmico adicional à 1300 °C. O efeito desses dois métodos, bem como o efeito do tratamento térmico adicional às amostras preparadas pelo método de combustão, foram analizados as propriedades físicas do YCrO3 por diversas técnicas espectroscópicas e estruturais: difracção de raios-X (DRX), microscopia eletronica de varredura e transmissão (MEV e MET), espectroscopias de transformada de Fourier no infravermelho e de reflectância difusa, susceptibilidade e ressonância paramagnética eletrônica (RPE). As imagens (MEV) mostram que os pós de YCrO3 sintetizados através dos dois métodos de síntese possuem morfologias semelhantes, com tamanho médio de grãos de aproximadamente 160 nm. Através das imagens foi constatado que a aplicação de um tratamento térmico para a amostra obtida por combustão, na mesma temperatura síntese que a obtida pelo método de precursores de polímericos, não alteran significativamente as propriedades estructurais do sistema. Os espectros de reflectância difusa das diferentes amostras apresentam duas bandas intensas, as quais correspondem às transições eletrônicas do ion Cr3+ desde o estado fundamental 4A2g para os estados excitados 4T2g e 4T1g. Duas bandas menores correspondentes às transições 4A2g → 2Eg e 4A2g → 2T1g também puderam ser observadas nos espectros. A partir dos espectros foram calculados os parâmetros de campo cristalino Dq e de Racah, B e C, para todas as amostras, com valores médios de 3.03 para Dq, 542 cm-1 e 3.144 cm-1 para B e C respectivamente. Ainda com estes espectros e a teoria de Kubelka-Munk calculamos o gap óptico Eg, com valor médio de 3,7 eV, coincidindo com os reportados na literatura para o YCrO3 preparado por outros métodos. Também foi estudada a dependência dos parâmetros do espectro de RPE, fator g, intensidade e largura de linha em função da temperatura, desde 100 K até 300 K. Os resultados mostram que, próximo de 140 K, temperatura de Néel para este material, ocorre uma transição de fase magnética, desde a fase antiferromagnética para a fase paramagnética, em pleno acordo com os resultados obtidos através das medidas de susceptibilidade. Os nossos resultados são discutidos e comparados com outros reportados na literatura.

A hidroxiapatita e o esmalte dental são, na atualidade, alguns dos materiais mais pesquisados pela técnica de espectroscopia por Ressonância Paramagnética Eletrônica ? RPE. O esmalte dental, que possui cerca de 97% de sua massa constituída por cristais de hidroxiapatita, se apresenta como o tecido humano preferencial para dosimetria de radiação com o uso desta técnica, principalmente no caso de exposições acidentais da população em geral. A análise de hidroxiapatitas sintéticas tem por finalidade auxiliar o entendimento do espectro do esmalte dental bem como desenvolver um dosímetro de radiação com boa eficiência e eficácia. O espectrômetro em banda-K utiliza uma freqüência intermediária aos espectrômetros em banda-X e em banda-Q, possuindo uma melhor resolução que banda-X sem as dificuldades, devido à alta sensibilidade, do banda-Q. O presente trabalho tem como objetivo analisar espectros em banda-K de um bloco de esmalte irradiado e de hidroxiapatitas sintéticas, comparando com espectros de banda-Q e banda-X presentes na literatura, e ainda verificar a possibilidade de se fazer dosimetria de radiação com o uso deste aparelho. / Hydroxyapatite and tooth enamel are, nowadays, some of materials mostly investigated by the spectroscopic technique of Electron Paramagnetic Resonance ? EPR. Tooth enamel, that have about of 97% of its mass constituted by hydroxyapatite crystals, appears itself as the preferential human tissue for radiation dosimetry using EPR, mainly in case of accidental expositions by ordinary personnel. The analyses of synthetic hydroxyapatites have as the main goal to help the understanding of tooth enamel?s spectrum seeking the developing of a radiation dosimeter with good efficiency and accuracy. The K-band EPR spectrometer uses an intermediate frequency between the spectrometers in X-band and Q-band, possessing a better resolution than the X-band without presenting the Q-band?s difficulties. In this preliminary work the EPR spectra of a small block tooth enamel and synthetic hydroxyapatites irradiated were analyzed in K-band, comparing the results with X- and Q-band with published results, the possibility of performing radiation dosimetry at K-band was verified.

Os radicais nitróxidos tem sido muito utilizados em estudos de sistemas biológicos através do método de marcagem de spin. Apesar do grande número de publicações sobre aplicações, o número de estudos das propriedades intrínsecas dos nitróxidos ê bem reduzido. A importância de tais estudos é grande uma vez que o estudo de um sistema simples permite avaliar e interpretar os resultados dos sistemas biológicos, geralmente mais complicados Assim no presente trabalho estudou-se o espectro do RPE do 4-hidroxi-2,2,6,6-tetrametil piperidina-1 oxil (I) introduzido como impureza na sua matriz diamagnética 4-hidroxi-2,2,6,6-tetrametilpiperidina (II). Este processo de dopagem muito utilizado em física do estado sólido permitiu a determinação dos tensores g e A. Os valores obtidos gxx = 2.0098, gyy = 2.0061, gzz = 2.0024, e A xx = 5.3 G, Ayy = 7.0 G, Azz = 35.0 G, encontram-se dentro do esperado sendo que o valor de go = 2.0061 e Ao = 15.8 G evidencia a interação do fragmento N-O com ponte de hidrogênio (de hidroxila na posição 4 de uma molécula vizinha). A existência de dois centros paramagnéticos inequivalentes é interpretada corno devido a presença de duas possíveis conformações do fragmento N-O. As larguras das linhas de RPE observadas são grandes e em certas orientações do campo magnético ocorre o desdobramento da transição eletrônica num dubleto devido a interação com o hidrogênio da hidroxiIa que faz ponte com o N-O. A temperatura de cerca de -140&#176C e observada uma mudança sensível nos espectros de RPE evidenciando a desaparição da estrutura hiperfina na região de Axx e Ayy. Esta mudança está relacionada com o congelamento da rotação dos grupos metila afetando assim a interação dos prótons destes grupos com o elétron do fragmento N-O. O sistema estudado parece ser extremamente interessante do ponto de vista do estudo de influência de prótons na relaxação eletrônica do radical nitroxido. É evidenciada a forte dependência das propriedades paramagnéticas do nitróxido em relação à estrutura da vizinhança do fragmento N-O. / Nitroxide radicals are being used in many studies of biological systems through the spin label method. Despite the great number of publications involving the application of nitroxides only a reduced number of work s about nitroxides themselves is known. These studies are important because the knowledge of the properties of simple systems could be used in the interpretation of results in biological systems, normally mare complicated. In the present work the EPR spectra of 4-hydraxy-2,2,6,6-tetramethyl piperidine-l oxyl (I) introduced as impurity in the diamagnetic matrix of 4-hidraxy-2,2,6,6-tetramethyl piperidine (II) were studied. This process commonly used in solid state physics made it possible the determination of the g and A tensors. The values obtained : gxx = 2.0098, gyy = 2.0061, gzz = 2.0024, and A xx = 5.3 G, Ayy = 7.0 G, Azz = 35.0 G are typical for nitroxides. The mean values go = 2.0061 and Ao 15.8G are consistent with the presence af hydrogen bond to the N-O fragment (this bond is made with the proton of a hydroxyl in position 4 of a neighbour molecule). The existence of two magnetically inequivalent centers is interpreted so that two different conformations for the N-O fragment are possible. The line widths of the EPR lines are quite great and for some orientations of the magnetic field a splitting of the electronic transition in to a dublet occur. This splitting is due to the interaction of the electron in the N-Q fragment with the hydroxyl hydrogen which is bonded to the nitroxide group. At a temperature around -140&#176C a change in the EPR spectra is observed, when the hyperfine nitrogen structure in the region of A and A disappear. This change x y is related to the freezing of the rotation of the methyl groups affecting the interaction of the protons of these groups with the electron in the N-O. The system studied in the present work seems to be very interesting from the point of view of the influence of the protons in the electronic relaxation in the nitroxide radical. ln this context the strong dependency of the properties of the nitroxide relative to the structure of the neighbouring of the N-O fragment become evident.

Engineering the phase behaviour of high performance inkjet colorants

Dyes for inkjet printing are typically of the chromonic type. Chromonic mesophases have gained considerable attention as a well-defined group of lyotropic mesogens with different properties from conventional amphiphiles. While extensive research has been dedicated to the field of surfactant liquid crystals, structural and aggregation studies of chromonics have only emerged as a topic of interest within the last few years. The liquid crystalline structures in aqueous solutions of commercial Cu - phthalocyanine and black dyes have been examined using a combination of optical microscopy, UV-vis spectroscopy, nuclear magnetic resonance, wide- and small-angle X-ray diffraction and electronic paramagnetic resonance with a view to examining the phase behaviour of the chromonic mesophases formed over a broad range of concentrations and temperatures. These studies were performed in order to resolve outstanding problems concerning structural properties of these systems. Optical microscopy allowed us to identify the liquid crystalline phases and to construct the phase diagram. The observations show that both of these dyes form nematic mesophase above 15% wt / wt% dye. The small-angle diffraction data confirmed that the nematic phase for the black dye is maintained throughout the 16-25% composition range. A further increase in concentration leads to the formation of the hexagonal phase. The Cu – phthalocyanine dye also formed a nematic phase at low concentrations, with the aggregates undergoing a phase transition to an orientationally ordered chromonic liquid crystal phase at high dye concentration. These studies showed that this ordered phase possessed hexagonal symmetry. The wide-angle X-ray results demonstrated that aggregation involved π-π stacking of the molecules into columns. An additional reflection at ca. 6.8Å was observed for the black dye, which is believed to arise from “head – to – tail” packing of the molecules within the aggregates (a similar phenomenon observed in other azo dyes, e.g. Edicol Sunset Yellow).The densities of both dyes were measured over the studied range of concentrations. This enabled us to calculate the parameters of the aggregates within the hexagonal mesophase. A comparison between the area of the molecule and the cross-section of the aggregates showed that the aggregates of both dyes were the unimolecular stacks.

Cette thèse porte sur le développement de nouvelles approches par marquage de spin suivi par spectroscopie RPE. Cette technique est bien adaptée pour suivre la dynamique structurale des protéines. Son principe repose sur l'insertion d'un radical nitroxyde, en un (ou plusieurs) site(s) choisi(s) d'une protéine et permet de sonder localement la structure de la protéine étudiée grâce aux différentes techniques de RPE (en onde continue et impulsionnelle).Dans une première partie, cette technique a été appliquée à la caractérisation de la dynamique structurale de l'IF1 de levure, un peptide inhibiteur de l'ATP-synthase. L'utilisation des spectroscopies de RPE et de dichroïsme circulaire a permis de montrer qu'IF1 de levure dimérise par sa partie médiane et que la partie C-terminale est désordonnée.La seconde partie est plus méthodologique et a pour but d'étudier et de caractériser un marqueur nouvellement synthétisé afin d'élargir les potentialités du marquage de spin. En effet, cette technique est notamment limitée par la faible diversité spectrale offerte par les sondes disponibles (trois raies). Le nouveau marqueur donne un spectre RPE à six raies grâce à la présence d'un noyau magnétique dans l'environnement du radical. Greffé sur une protéine modèle, nous avons montré que ce nouveau marqueur est tout autant capable de rendre compte de variations structurales qu'un marqueur classique. La superposition des signatures spectrales (trois raies + six raies) montre qu'il est possible de différencier les deux signatures spectrales et de sonder simultanément deux sites d'une protéine et de son partenaire. / This thesis focuses on the development of new approaches for site-directed spin labeling followed by EPR spectroscopy. This technique is well suited to monitor the structural dynamics of proteins. The insertion of a nitroxide radical, in one (or several) selected site(s) of a protein, allows probing the structure of the protein using different EPR spectroscopy approaches (continuous wave and pulsed).In a first part, this technique has been applied to characterize the structural dynamics of the yeast IF1, an inhibitory peptide of the ATP-synthase. Using EPR and circular dichroïsm spectroscopies we showed that yeast IF1 dimerizes by its central part and that the C-terminal part remains disordered.The second part is more methodological and the aim is to study and characterize a newly synthesized spin label in order to expand the potential of site-directed spin labeling. In particular, the technique is limited by the poor spectral diversity offered by the available labels (three lines). The new label gives a six lines EPR spectrum thanks to the presence of a magnetic nucleus in the environment of the radical. Grafted on a model protein, we demonstrated that this new label is as able as classical ones to report on structural variations. The superposition of the spectral signatures (three lines + six lines) showed that it is possible to differentiate the two spectral signatures and to probe two sites of a protein and its partner simultaneously.

Etude pluridisciplinaire d'une hydrogénase : mécanisme et optimisation des propriétés catalytiques / multidisciplinary study of a hydrogenase : mechanism and optimization of catalytic properties

Les hydrogénases sont des métalloenzymes qui catalysent la conversion réversible du dihydrogène en protons et en électrons. Durant ma thèse, je me suis focalisé sur certains aspects du fonctionnement de l’hydrogénase à [NiFe] hétérodimérique de Desulfovibrio fructosovorans. Nous avons montré que contrairement au mécanisme communément admis d’inactivation aérobie de l’enzyme, l’O2 n’est pas incorporé en tant que ligand au niveau du site actif mais agit plutôt comme un simple oxydant. Ce résultat remet en question le mécanisme proposé pour expliquer la tolérance naturelle à l’O2 de certaines hydrogénases. L’analyse à l’aide d’un modèle cinétique des voltamogrammes cycliques complexes obtenus avec 16 variants a montré que les vitesses d’(in)activation en conditions anaérobies peuvent être accélérées de plusieurs ordres de grandeurs. Nous avons aussi montré et expliqué la corrélation entre ces vitesses et la tolérance à l’O2. Nous avons étudié une série de mutants qui produisent H2 beaucoup plus lentement que l’enzyme sauvage. Nous avons montré que la vitesse de cette réaction est déterminée par celle de l’étape de diffusion du H2, qui est lente dans les mutants. Finalement, nous nous sommes intéressés à une thréonine appartenant à la voie putative de transfert des protons. Nous avons démontré que cet acide aminé est effectivement impliqué dans le transport des protons. Il joue aussi un rôle crucial dans la stabilisation des intermédiaires formés au cours du cycle catalytique et probablement dans la détermination des vitesses de transfert électronique et de diffusion à travers le canal. / Hydrogenases are metalloenzymes which catalyse the reversible conversion of dihydrogen into protons and electrons. In my work, I focused on some aspects of the catalytic mechanism of the heterodimeric NiFe hydrogenase from Desulfovibrio fructosovorans. We demonstrated that, contrary to the commonly accepted mechanism of aerobic inactivation, the attacking O2 is not incorporated as an active site ligand but rather acts as an electron acceptor. This finding calls for a re-examination of the mechanism for O2 tolerance of the natural O2 tolerant NiFe hydrogenases. We also described a simple analytical model that we used to analyse the complex voltammetric signals of 16 mutants obtained by substituting an amino acid near the active site. We demonstrated that this substitution can accelerate anaerobic inactivation and reactivation by up to three orders of magnitude. We also demonstrated and explained the correlation between these rates and O2-tolerance. We studied mutants whose H2-production activity is impaired. We found that the rate limiting step of this reaction is the diffusion of hydrogen out of the enzyme, through the hydrophobic channel. Finally, we focused on a threonine belonging to the putative proton transfer pathway. We demonstrated that this amino acid is indeed implicated in proton transport. It may also play a crucial role in the stabilization of intermediates formed during the catalytic cycle, and probably also in determining the rate of electron transfer and diffusion along the gas channel.

Os hormônios peptídicos Angiotensina II (Ang II) e bradicinina (BK) ativam transdução de sinal através da ligação a Receptores Acoplados a Proteínas G (GPCR). Este trabalho propõe o estudo de propriedades conformacionais, através de espectroscopia de fluorescência da Ang II e BK e de seus análogos contendo o marcador de spin ácido 2,2,6,6-tetrametilpiperidina-1-oxil-4-amino-4-carboxílico, TOAC (TOAC1-Ang II, TOAC3-Ang II, TOAC0-BK, TOAC3-BK). Os peptídeos foram estudados em solução (efeito do pH) e também na presença de membranas modelo, micelas e bicamadas, formadas por anfifílicos zwitteriônicos ou aniônicos. Foi monitorada a fluorescência intrínseca dos resíduos aromáticos (Tyr4 na Ang II e Phe5 e Phe8 na BK). O efeito de supressão da fluorescência pelo TOAC foi utilizado para obter informação sobre a proximidade desse resíduo aos grupos fluoróforos. Foi observada dependência da fluorescência com o pH e regiões de pKs dos grupamentos ionizáveis. Os espectros evidenciaram também a interação peptídeo-membrana modelo. Interações mais fortes ocorreram entre os peptídeos e membranas com carga superficial negativa, evidenciando a importância de interações eletrostáticas para a ligação. Porém, interações hidrofóbicas também estão envolvidas, como verificado pela ligação dos peptídeos a membranas zwitteriônicas. Estudos com variação de pH também mostraram o papel dessa variável na interação peptídeo-membrana e a alteração de pKs de resíduos ionizáveis decorrentes da interação. A titulação com concentrações crescentes de membranas permitiu o cálculo das constantes de associação. A ligação a membranas é função da conformação dos peptídeos. Em particular, a presença de TOAC na posição 3 parece diminuir a afinidade desses análogos por membranas. Supressão de fluorescência foi efetuada empregando três diferentes abordagens: 1) supressão pela molécula aquossolúvel acrilamida, 2) supressão da fluorescência de fosfolipídeos contendo o fluoróforo NBD em diferentes posições da molécula pelos análogos marcados com TOAC, 3) supressão da fluorescência dos peptídeos por ésteres metílicos do ácido esteárico contendo o grupamento nitróxido em diferentes posições da cadeia. Esses estudos permitiram determinar a localização dos peptídeos na interface água-membrana. Medidas de anisotropia de fluorescência também evidenciaram a ligação dos peptídeos a membranas, revelando maior imobilidade dos mesmos nessas condições. Foi ainda estudado um peptídeo que contém os resíduos 92-100 (fEL1) do receptor AT1 de Ang II humano. Predições baseadas na estrutura cristalina da rodopsina estimam que essa seqüência localiza-se na primeira alça extra-celular do receptor. A seqüência contém a Tyr92, considerada um resíduo importante para a ligação hormônio-receptor. Resultados preliminares sugeriram que fEL1 interage com Ang II e TOAC1-Ang II, mas não com TOAC3-Ang II. Este último resultado provavelmente deve-se à dobra causada por TOAC que restringe a liberdade de movimento do esqueleto peptídico. Essa característica provavelmente determina a falta de atividade biológica de TOAC3-Ang II e TOAC3-BK, enquanto os análogos marcados no N-terminal retém atividade parcial (Nakaie et al., 2002). Tem sido proposto que peptídeos ligantes de GPCR se ligariam à bicamada lipídica e atingiriam seu receptor através da difusão pela bicamada. Em solução aquosa essas moléculas são flexíveis, existindo um equilíbrio dinâmico entre várias conformações. A ligação à bicamada lipídica estabilizaria uma ou algumas conformações, entre elas aquela que o ligante adota ao ligar-se ao receptor. O presente estudo contribui para a compreensão, a nível molecular, do processo de interação entre os hormônios peptídicos e membranas lipídicas. / The peptide hormones Angiotensin II (Ang II) and bradykinin (BK) trigger signal transduction by binding to G Protein Coupled Receptors (GPCR). This work proposes the study of conformational properties of Ang II and BK, as well as their analogues containing the spin label 2,2,6,6-tetramethylpiperidine-1-oxyl-4-amino-4-carboxylic acid, TOAC (TOAC1-Ang II, TOAC3-Ang II, TOAC0-BK, TOAC3-BK) making use of fluorescence spectroscopy. Studies were performed in solution (effect of pH) and also of the interaction between the peptides and model membranes - micelles and bilayers - formed by amphiphiles, either zwitterionic or negatively charged. The intrinsic fluorescence of aromatic residues (Tyr4 in Ang II and Phe5 and Phe8 in BK) was monitored. Fluorescence quenching by the TOAC-carrying analogues provided information about the proximity between TOAC and the fluorophores. The fluorescence was pH-dependent and evinced regions corresponding to pKs of ionizable groups. Peptide-model membrane interactions were also examined. Stronger interactions were detected between the peptides and membranes formed by negatively charged amphiphiles, pointing to the importance of electrostatic interactions for binding. However, hydrophobic interactions were also involved, as suggested by the fact that the peptides also bound to zwitterionic membranes. Variable pH studies showed the effect of this parameter on peptide-membrane interaction. The peptide-membrane interactions promoted changes in the pKs of ionizable residues. Titrations with increasing membrane concentrations allowed calculation of binding constants. Binding to membranes is a function of peptide conformation. In particular, TOAC at position 3 seems to decrease the affinity of both Ang II and BK for membranes. Fluorescence quenching studies made use of three different approaches: 1) quenching by water soluble acrylamide, 2) quenching of the fluorescence of phospholipids carrying the fluorescent group NBD in different positions by the spin-labeled TOAC-bearing analogues, 3) quenching of the peptides fluorescence by methyl esters of stearic acid containing the nitroxide moiety at different positions in the acyl chain. These studies indicated that the peptides are located at the water-membrane interface. Measurements of fluorescence anisotropy also evinced binding of the peptides to the membranes and showed that the peptides undergo more restricted motion under these conditions. A peptide containing residues 92-100 (fEL1) of the Ang II AT1 human receptor was also studied. Predictions based on rhodopsin crystalline structure estimate that this sequence is located in the receptor´s first extra-cellular loop. Preliminary results suggest that fEL1 interacts with Ang II and TOAC1-Ang II, but not TOAC3-Ang II. This latter result is probably due to the TOAC-induced bend that restricts the freedom of motion of the peptide backbone. This feature is probably the cause of lack of biological activity of TOAC3-Ang II and TOAC3-BK, while the N-terminally labeled analogues retain partial activity (Nakaie et al., 2002). GPCR-binding peptides have been proposed to bind to the lipid bilayer and reach their receptors by diffusion in the bilayer. In aqueous solution these molecules exist as a dynamic equilibrium between various flexible conformations, among them, the receptor-bound conformation. The present study provides contributions for the understanding, at the molecular level, of the interaction between the peptide hormones and lipid membranes.

Preparação e caracterização de catalisadores de V2O5 suportados em TiO2. / Preparation and characterization of V2O5 catalyst supported TiO2.

Uma série de amostras do sistema catalítico V2O5/TiO2 foi preparada pelo método sol-gel com diferentes teores da fase ativa (de 0 a 9% em massa de V2O5) e calcinada em diferentes temperaturas (de 250 a 6000C). Sistema similar foi obtido pelo método de impregnação para a comparação. Os catalisadores foram caracterizados quanto suas propriedades texturais e estruturais pelas técnicas de Adsorção de N2 a 77K, Difratometria de Raios-X (DRX), Espectroscopia Raman, Ressonância Paramagnética Eletrônica (EPR), Espectroscopia de Fotoelétrons Excitados por Raios-X (XPS) e Espectroscopia de Absorção de Raios-X (XAS). Testes catalíticos para a verificação da atividade e seletividade foram realizados frente à reação de conversão do metanol. O sistema obtido pelo método sol-gel apresentou elevada área superficial e porosidade, as quais foram ampliadas com o aumento do teor de vanádio e reduzidos com o aumento da temperatura de calcinação. O sistema preparado por impregnação apresentou as mesmas propriedades texturais do suporte comercial. A análise de DRX identificou a existência de TiO2 nas formas cristalográficas anatásio e rutilo, não foi identificada a presença de V2O5 na forma cristalina. No sistema preparado por impregnação a estrutura cristalográfica do suporte se mantém, independente do teor de vanádio, porém a partir de 6% de V2O5 foi identificado à presença de V2O5 cristalino. Os espectros de Raman mostraram a presença de dois grupos superficiais de vanádio, os vanadilos monoméricos e os vanadatos poliméricos mesmo em elevados teores de vanádio, nos sistemas preparados pelo processo sol-gel. Nos catalisadores obtidos por impregnação foram identificados modos vibracionais referentes ao V2O5 cristalino.Os resultados obtidos por XAS confirmaram os obtidos por DRX e indicaram a presença de vanádio V4+ e V5+, porém com simetria diferente da forma cristalina V2O5. A determinação da razão entre os átomos V/Ti superficiais feita por XPS mostrou ser crescente com o aumento do teor de vanádio. Os espectros de EPR indicaram a presença de pelo menos três famílias de íons V4+ em diferentes simetrias, duas delas inseridas na matriz da titânia na fase rutilo e a outra dispersa na superfície. O sistema obtido por impregnação apresentou poucos íons V4+. Os testes catalíticos revelaram que a atividade e seletividade catalítica foram praticamente as mesmas para os sistemas preparados pelos dois métodos. Entretanto, a estabilidade catalítica do sistema obtido via sol-gel foi superior ao sistema preparado por impregnação. / A series of samples of the catalytic system V2O5/TiO2 was prepared by the sol-gel method with different proportions of the active phase (from 0 to 9% in mass of V2O5) and calcined in different temperatures (from 250 to 6000C). A similar system was obtained by the impregnation method for comparison. The catalyst were characterized by their textural and structural properties by Adsorption of N2 at 77K, X-Ray Diffraction (XRD), Raman Spectroscopy, Electronic Paramagnetic Resonance (EPR), X-Ray Photoelectron Spectroscopy (XPS) and X-Ray Absorption Spectroscopy (XAS). Catalytic tests for the verification of the activity and selectivity were performed by the conversion reaction of the methanol. The system obtained by the sol-gel method presented a high superficial area and porosity, which were enlarged by the increase of vanadium proportion and reduced with increase of the calcinations temperature. The system prepared by impregnation presented the same textural proprieties of the commercial support. XRD analysis identified the existence of TiO2 in two crystallographic forms, anatase and rutile. It was not identified the presence of V2O5 in the crystalline form. In the system prepared by impregnation the crystallographic structure of the support keeps, independent of the vanadium content, however starting from 6% of the vanadia was identified crystalline V2O5. Raman spectra showed the presence of two superficial groups of vanadium, the monomeric vanadyls and the polymeric vanadates even for elevated contents of vanadium, in the system prepared by the impregnation method vibrational modes were identified referring to V2O5 crystalline. The results obtained by XAS confirmed the obtained by XRD and indicated the presence of V4+ and V5+, however with different symmetry from the V2O5 in a crystalline form. The determination of the proportion between the surface V/Ti atoms determined by XPS showed to be increased with the increase of the vanadium content. EPR spectra indicated the presence of at least three V4+ion families in different symmetries; two of them inserted in the matrix of titania in the rutile phase and to other dispersed on the surface. The system obtained by impregnation presented few V4+ ions. The catalytic tests reveled that the catalytic activity and selectivity were practically the same for the systems prepared by both methods. However, the catalytic stability of the system obtained via sol-gel was superior to the system prepared by impregnation.

APLICACIÓN DE TÉCNICAS ESPECTROSCÓPICAS IN SITU AL ESTUDIO DE REACCIONES DE INTERÉS MEDIOAMBIENTAL: ELIMINACIÓN DE ÓXIDOS DE NITRÓGENO

[EN] The Selective Catalytic Reduction of nitrogen oxides (SCR-NOx) is nowadays a very relevant process for reducing NOx emissions in diesel vehicles, which must comply with increasingly restrictive European regulations. In this thesis the reaction mechanism and active centers in Cu-zeolite catalysts with different structures has been investigated. For his purpose two in situ spectroscopic techniques were mainly used, being Nuclear Magnetic Resonance NMR and Electron Paramagnetic Resonance EPR, which allowed the detection of reaction intermediates and identification of Cu active species. In particular we have studied the SCR-NOx reaction using NH3 as the reducing agent and Cu-zeolites catalysts with the chabazite structure. The preliminary study of the interaction of the catalysts with NH3 has shown the formation of several Cu-NH3 complexes with different stability. At SCR typical reaction temperatures (250 ° C), ammonia forms NH4+ ions in Brønsted acids centers of the zeolite and stable Cu+-(NH3) complexes, while Cu2+ remains isolated at the 6R plane, which is precisely the active site. However, when studying species formed on Cu-zeolites in the presence of the reaction mixture NH3/NO/O2, it appears that NH4+ ions are consumed during reaction and the formation of various intermediates including nitrite/nitrate species that decompose at T > 250 ° C to the reaction products (N2 and H2O). We also investigated the mechanism of the SCR-NOx reaction using C3H8 as the reducing agent and Cu-zeolites of different topology with medium and large pore systems. The results show the formation of a hydrocarbon activated species in the Cu2+ which is related to the catalytic activity. Furthermore, isolated Cu2+ is an active site that is reduced to Cu+ during reaction, and then re-oxidized to Cu2+ in the presence of O2. Finally, the hydrothermal stability of Cu-zeolites has been studied, since it is a prerequisite for its application as SCR catalysts, comparing the very hydrothermally stable Cu-SSZ-13 zeolite and the Cu-ZSM-5 which is completely disabled after hydrothermal treatment. The findings suggest the origin of the deactivation to be a change in the coordination of isolated Cu2+ in the Cu-ZSM-5 type to form CuAlOx species which are inactive in the SCR-NOx reaction. / [ES] La Reducción Catalítica Selectiva de óxidos de nitrógeno, SCR-NOx (acrónimo del inglés Selective Catalytic Reduction of NOx) es un proceso muy importante actualmente para la reducción de las emisiones de NOx en vehículos diésel, que deben ajustarse a nuevas normativas europeas más restrictivas. En la presente tesis doctoral se ha investigado el mecanismo de reacción y los centros activos en catalizadores de Cu en zeolitas (Cu-zeolitas) con distintas estructuras. Para ello se han utilizado fundamentalmente dos técnicas espectroscópicas in situ, la resonancia magnética nuclear RMN y la resonancia paramagnética electrónica EPR, que han permitido la detección de intermedios de reacción y la identificación de especies de Cu activas. En concreto se ha estudiado la reacción SCR-NOx utilizando NH3 como reductor y catalizadores Cu-zeolitas con estructura chabazita. Los resultados obtenidos en el estudio preliminar de la interacción del catalizador con el NH3 muestran la formación de distintos complejos de Cu-NH3 con diferente estabilidad. A la temperatura de reacción (250 °C), el amoniaco forma iones NH4+ en centros ácidos Brønsted de la zeolita y complejos Cu+(NH3) estables, y el Cu2+ permanece aislado en el plano de los anillos 6R, que es precisamente el centro activo. Sin embargo, cuando se estudian las especies formadas con la mezcla de reacción NH3/NO/O2 en las Cu-zeolitas, se observa que los iones NH4+ se consumen en el transcurso de la reacción y la formación de varios intermedios incluyendo nitritos/nitratos que descomponen a T > 250 °C a los productos de reacción (N2 y H2O). También se ha investigado el mecanismo de la reacción SCR-NOx utilizando C3H8 como reductor y Cu-zeolitas de distinta topología con tamaños de poros medio y grande. Los resultados obtenidos evidencian la formación de una especie activada del hidrocarburo en el Cu2+ que está relacionada con la actividad catalítica. Además el Cu2+ aislado es un centro activo que se reduce a Cu+ en el transcurso de la reacción, y se re-oxida posteriormente a Cu2+ en presencia de O2. Finalmente, se ha estudiado la estabilidad hidrotermal de las zeolitas con cobre, puesto que es una condición indispensable para su aplicación como catalizadores SCR, comparando la zeolita Cu-SSZ-13 muy estable hidrotermalmente, y la Cu-ZSM-5 que se desactiva por completo tras el tratamiento hidrotermal. Los resultados obtenidos apuntan que el origen de la desactivación es el cambio en la coordinación del Cu2+ en la Cu-ZSM-5 para formar especies tipo CuAlOx inactivas en la reacción. / [CAT] La reducció catalítica selectiva d'òxids de nitrogen, SCR-NOx (acrònim del anglès Selective Catalytic Reduction of NOx) és un procés molt important actualment per a la disminució de les emissions de NOx en vehicles dièsel, que deuen ajustar-se a les normatives europees més restrictives. En la present tesi doctoral s'ha investigat el mecanisme de reacció i els centres actius en catalitzadors de Cu en zeolites (Cu-zeolites) amb diferents estructures. Per a dur a terme aquesta tasca s'han utilitzat fonamentalment dos tècniques espectroscòpiques in situ, la ressonància magnètica nuclear RMN i la ressonància paramagnètica electrònica EPR, les quals han permès la detecció d'intermedis de reacció i la identificació d'espècies de Cu actives. Concretament s'ha estudiat la reacció SCR-NOx emprant NH3 com a reductor i catalitzadors Cu-zeolita amb estructura chabacita. Els resultats obtinguts en l'estudi preliminar de la interacció del catalitzador amb NH3 mostren la formació de diversos complexes de Cu-NH3 amb diferent estabilitat. A la temperatura de reacció (250 °C), l'amoníac forma ions NH4+ en centres àcids Brønsted de la zeolita i complexes Cu+(NH3) estables, a més, el Cu2+ roman aïllat en el plànol dels anells 6R, que és precisament el centre actiu. No obstant, quan s'estudien les espècies formades amb la mescla de reacció, NH3/NO/O2 en les Cu-zeolites, s'observa que els ions NH4+ es consumeixen durant la reacció i la formació de diversos intermedis incloent nitrits/nitrats que descomponen, a T > 250 °C, als productes de la reacció (N2 y H2O). També s'ha estudiat el mecanisme de la reacció SCR-NOx utilitzant C3H8 com a reductor i Cu-zeolites amb diferent topologia amb mides de porus mitges i grans. Els resultats obtinguts evidencien la formació d'una espècie en forma activa del hidrocarbur al Cu2+ que està relacionada amb l'activitat catalítica. A més a més, el Cu2+ aïllat és un centre actiu que és redueix amb el transcurs de la reacció, i es re-oxida posteriorment a Cu2+ en presència de O2. Finalment, s'estudia l'estabilitat hidrotermal de les zeolites amb coure, puix que és una condició indispensable per a la seua aplicació com a catalitzadors SCR, comparant la zeolita Cu-SSZ-13 que és molt estable hidrotermalment, i Cu-ZSM-5 que es desactiva completament després del tractament hidrotermal.
