Elucidating and Mapping Heat Tolerance in Wild Tetraploid Wheat (Triticum turgidum L.)Ali, Mohamed Badry Mohamed 2010 December 1900 (has links)
Identifying reliable screening tools and characterizing tolerant germplasm sources is essential for developing wheat (Triticum aestivum L.) varieties suited for the hot areas of the world. Our objective was to evaluate heat tolerance of promising wild tetraploid wheat (Triticum turgidum L.) accessions that could be used as sources of heat tolerance in common- and durum-wheat (Triticum durum) breeding programs.
We screened 109 wild tetraploid wheat accessions collected by the International Center for Agriculture Research in the Dry Areas (ICARDA) from the hottest wheat growing areas in Africa and Asia, as well as, two common wheat checks for their response to heat stress by measuring damage to the thylakoid membranes, flag leaf temperature depression (FLTD), and spike temperature depression (STD) during exposure to heat stress for 16 beginning at anthesis. Measurements were taken on the day of anthesis then 4, 8, 12, and 16 days post anthesis (DPA) under controlled optimum and heat-stress conditions. Individual kernel weight (IKW) and heat susceptibility index (HSI) measurements were also obtained. Prolonged exposure to heat stress was associated with increased damage to thylakoid membranes, as indicated by the high ratio of constant fluorescence (O) to peak variable fluorescence (P).
A positive and significant correlation was found between O/P ratio and both FLTD and STD under heat-stress conditions. A negative and significant correlation was found between FLTD and HSI and between STD and HSI based on the second and third measurements (4 and 8 DPA). Correlations obtained after the third measurement were not significant because heat-stress accelerated maturity and senescence.
For a pedigree-based mapping strategy a family approach was then developed by crossing and back-crossing heat-tolerant and heat-susceptible germplasm. A set of 800 lines resulting from the pedigree-based family approach was phenotyped using FLTD, chlorophyll content and yield and its components under heat stress. Genotyping of these lines was accomplished using simple sequence repeat (SSRs) markers. Some QTLs associated with heat stress tolerance were identified. This study identified potential heat-tolerant wild tetraploid wheat germplasm and QTL conditioning heat tolerance that can be incorporated into wheat breeding programs to improve cultivated common and durum wheat.

Politics in extraordinary times : a study of the reaction of political parties and elites to terrorismChowanietz, Christophe 06 1900 (has links)
Cette thèse porte sur le comportement des élites politiques durant les périodes de crise
nationale et plus particulièrement sur leurs réactions aux attentats terroristes. Elle
démontre que les crises terroristes sont tout comme les conflits militaires ou
diplomatiques propices aux unions nationales et notamment aux ralliements des partis
d’opposition auprès du gouvernement.
L’analyse statistique d’actes terroristes s’étant produits dans cinq états
démocratiques (Allemagne, Espagne, États-Unis d’Amérique, France et Royaume-Uni)
entre 1990 et 2006 révèle que l’ampleur d’un attentat en termes de pertes humaines ainsi
que la répétition de ces attentats influencent dans une large mesure la réaction des élites
politiques. Ainsi plus l’ampleur d’un attentat est élevée, plus la probabilité d’un
ralliement est grande. En revanche, la multiplication des attentats augmente la possibilité
de dissension entre l’opposition et le gouvernement. Par ailleurs, l’opposition est plus
susceptible de se rallier au gouvernement lorsque l’attentat est perpétré par des terroristes
provenant de l’étranger. L’analyse quantitative indique également que l’existence d’un
accord formel de coopération dans la lutte antiterroriste entre le gouvernement et
l’opposition favorise l’union des élites. Enfin, les données analysées suggèrent que la
proportion des ralliements dans les cinq pays est plus importante depuis les attentats du
11 septembre 2001.
Une analyse qualitative portant exclusivement sur la France et couvrant la période
1980-2006 confirme la validité des variables identifiées dans la partie quantitative, mais
suggère que les élites réagissent au nombre total de victimes (morts mais aussi blessés) et
que la répétition des actes terroristes a moins d’impact lors des vagues d’attentats. Par ailleurs, les analyses de cas confirment que les élites politiques françaises sont plus
susceptibles de se rallier quand un attentat vise un haut-fonctionnaire de l’État. Il apparaît
également que les rivalités et rancœurs politiques propre à la France (notamment suite à
l’arrivée de la gauche au pouvoir en 1981) ont parfois empêché le ralliement des élites.
Enfin, cette analyse qualitative révèle que si l’extrême gauche française soutient
généralement le gouvernement, qu’il soit de gauche ou de droite, en période de crise
terroriste, l’extrême droite en revanche saisit quasi systématiquement l’opportunité
offerte par l’acte terroriste pour critiquer le gouvernement ainsi que les partis de
l’establishment.
La thèse est divisée en sept chapitres. Le premier chapitre suggère que le
comportement des élites politiques en période de crises internationales graves (guerres et
conflits diplomatiques) est souvent influencé par la raison d’État et non par l’intérêt
électoral qui prédomine lors des périodes plus paisibles. Le second chapitre discute du
phénomène terroriste et de la littérature afférente. Le troisième chapitre analyse les
causes du phénomène d’union nationale, soumet un cadre pour l’analyse de la réaction
des élites aux actes terroristes, et présente une série d’hypothèses. Le quatrième chapitre
détaille la méthodologie utilisée au cours de cette recherche. Les chapitres cinq et six
présentent respectivement les résultats des analyses quantitatives et qualitatives. Enfin, le
chapitre sept conclut cette thèse en résumant la contribution de l’auteur et en suggérant
des pistes de recherche. / This thesis focuses on the behaviour of political elites during periods of national crisis
and particularly on their reactions to acts of terrorism. It demonstrates that terrorist crises,
much like military and diplomatic conflicts, represent a fertile ground for rallies around
the flag.
The statistical analysis of terrorist events that occurred in five democracies
(France, Germany, Spain, the United Kingdom and the United States of America) from
1990 to 2006 indicates that the magnitude of the attack in terms of fatalities and the
repetition of attacks influence in large measures the reaction of the political elite. The
higher the magnitude is, the more likely the rally. However, the repetition of attacks
increases the likelihood of dissention between opposition and government. Moreover, the
opposition is more likely to support the government when the attack is perpetrated by
terrorists originating from abroad. The quantitative analysis also indicates that the
existence of a formal antiterrorist pact between government and opposition increases the
likelihood of a rally. Finally, the data suggest that elites are more likely to rally around
the flag since the events of 9/11.
A qualitative analysis focusing solely on France and examining cases from the
period 1980-2006 confirms the validity of the variables identified in the quantitative part
but suggests that elites react to the total number of victims (including wounded) and that
the repetition has less impact during waves of attacks. Furthermore, the case studies
confirm that French political elites are particularly prone to rally when high-ranking
representatives of the state fall victim to the terrorists. In addition, it appears that political
rivalries and resentment (in particular following the Left’s return to power in 1981) have on occasion thwarted the rallying of elites. Finally, this analysis indicates that whereas
the far-left generally supports the government (left-wing or right-wing), the far-right uses
almost every opportunity to criticize the government and the parties of the establishment.
The thesis is divided into seven chapters. Chapter one suggests that during intense
foreign policy crises (military and diplomatic) the behaviour of the political elite is often
influenced by the national interest, rather than the electoral interest that predominates in
quieter periods. Chapter two discusses the terrorist phenomenon and the corresponding
literature. Chapter three analyses the rally-around-the-flag phenomenon, lays out a
framework for analysing the reaction of elites, and presents a series of hypotheses.
Chapter four details the methodology used. Chapters five and six present respectively the
results of the quantitative and qualitative analyses. Finally, chapter seven concludes with
a summary of the author’s contribution and suggests avenues of research.

Enligt styrdokumenten är hållbar utveckling en nödvändig komponent i utbildning. Materialet erhölls genom kvalitativa djupintervjuer med 6 lärare, inkluderar förskollärare, grundskollärare, gymnasielärare, representanter från Håll Sverige Rent, Naturskolan som har av erfarenhet i praktiskt arbete med hållbar utveckling inom framgångsrikt Grön Flaggs certifieringsprogram. Det analyserades och sammanställdes sedan i löpande text. Genom SWOT- analys identifieras och kategoriseras starka sidor, svaga sidor, möjligheter och hot bland faktorer som främjar utbildning för hållbar utveckling. Starka sidor och möjligheter betraktas som framgångsfaktorer. Resultaten visar vilka faktorer som är viktiga för att främja och förbättra utbildning för hållbar utveckling utifrån lärarens perspektiv. Den studien belyser framgångsfaktorer för utbildning för hållbar utveckling i praktiskt arbete. Min förhoppning är att min undersökning ska öka förutsättningarna för att utveckla en internationell dialog och samarbete inom kompetensutveckling inom utbildning för hållbar utveckling.

Aproximadamente 500.000 novos casos ocorrem todo ano, freqüentemente em países
pobres e em desenvolvimento. No Brasil, a leishmaniose visceral é causada pela
Leishmania chagasi, que é transmitida principalmente pelo inseto vetor Lutzomyia
longipalpis. Após 72 h de alimentação sanguínea num hospedeiro infectado, parasitas
procíclicos são vistos aderidos ao intestino do inseto via flagelo. Esta adesão é vital para a
evolução do ciclo de transmissão do parasita. Em vetores espécie–específicos foi proposto
um papel para um lipofosfogligano (LPG) de superfície de Leishmania na adesão, porém,
em vetores permissíveis, tem-se sugerido que esta adesão seja independente de LPG.
Com o interesse de identificar e caracterizar moléculas que possam ter uma
participação na interação parasito-vetor, nós iniciamos o estudo de duas moléculas de L.
chagasi denominadas Flag e Sup, pertencentes à família de proteínas pequenas miristiladas
(SMP’s), que anteriormente haviam sido implicadas em adesão ao tubo digestivo de
flebotomíneos.
Amplificamos e seqüenciamos o gene Flag de L. chagasi e observamos por
alinhamento múltiplo o alto grau de conservação deste gene entre as espécies de
Leishmania. Proteínas Flag e Sup recombinantes foram produzidas e utilizadas para
produção de anticorpos policlonais em coelhos. Vimos por Western Blot de extrato protéico
de várias espécies de Leishmania, que os anticorpos monoclonal e policlonal anti-Flag/MBP
e o anticorpo policlonal anti-Sup, reconhecem as proteínas em todas as espécies,
evidenciando a conservação destas.
A
imunolocalização
destas
proteínas
mostrou
que
a
proteína
Flag
está
preferencialmente localizada na forma promastigota na bolsa flagelar e no flagelo, e, em
amastigota, na bolsa flagelar e no flagelo interno curto, enquanto que a proteína Sup
mostrou localização em toda a superfície do corpo em ambas as formas.
Experimentos de RT-PCR mostraram que há transcrição diferencial de Flag e Sup
quando comparamos entre as formas evolutivas amastigotas e promastigotas. Flag tem uma
expressão parecida nas duas formas evolutivas enquanto Sup é mais expresso em
amastigotas.
Ensaios de inibição ex-vivo incubando previamente L. chagasi com anticorpo
monoclonal anti-Flag ou policlonal anti-Sup, mostrou redução significativa de parasitas
aderidos ao intestino de L. longipalpis
Na tentativa de identificar um potencial receptor para o parasita no intestino de L.
longipalpis, observamos a marcação de uma proteína de 27 kDa no extrato protéico do vetor
utilizando parasita biotinilado em experimentos de “ligand blot”. / Visceral leishmaniasis is an illness of high mortality if not treated. Approximately
500,000 new cases occur per year, frequently in poor countries. In Brazil, visceral
leishmaniasis is caused by Leishmania chagasi, transmitted by the insect vector Lutzomyia
longipalpis. After 72h of blood feeding in an infected host, procyclic parasites are seen
adhered to insect gut by the flagellum. This adhesion is vital for the completion of the
parasite cycle of transmission. In species-specific vectors a role for a surface
lipophosphoglycan (LPG) of Leishmania in the adhesion has been considered, while in the
permissible vectors, this adhesion may be independent of LPG.
Aiming to identify and characterize molecules that may be involved in the parasite-
vector interaction, we initiated a study of two molecules of L. chagasi, Flag and Sup, which
belong to the small myristylated protein family (SMP's), that have previously been implicated
in adhesion to the sandfly midgut.
We amplifyed and sequenced the Flag gene of L. chagasi and, using multiple
alignments, observed a high degree of conservation of this gene among Leishmania species.
Recombinant protein Flag and Sup were produced and used for polyclonal antibodies
production in rabbits. By Western Blot of total protein extract of some species of Leishmania,
we observed that the anti-Flag monoclonal and polyclonal antibodies and the anti-Sup
polyclonal antibody, recognized proteins in all the analyzed species, evidencing their
conservation.
Immunolocalization of these proteins showed that Flag is preferentially located in the
flagellar pocket and flagellum in the promastigote form and in the flagellar pocket and
internalized short flagellum in the amastigote, while Sup showed localization on the cell
surface in both forms.
Experiments of RT-PCR showed that there is a differential transcription of Flag and
Sup between amastigotes and promastigotes. Flag has similar levels of expression in both
evolutive forms while Sup is more expressed in amastigotes.
Assays of ex-vivo inhibition with L. chagasi previously incubated with monoclonal
antibodies anti-Flag or anti-Sup showed a significant reduction of the number of adhered
parasites to the L. longipalpis midgut.
In an attempt to identify a potential receptor for the parasite in the intestine of L.
longipalpis, we observed the labelling of a protein of 27 kDa in the protein extract of the
vector using biotinylated parasites in experiments of "ligand blot".

The album served as a reference to arise a integrated arts movement based on the authenticity of the maranhense way of life , condensing the aesthetic , artistic, political and cultural effort, from regional musicality, but inserted in the proposal to be a poetical expression Brazilian music. Based on theoretical like Pierre Bourdieu, Renato Ortiz, Raymond Williams, this research intend to understand how Bandeira de Aço (Flag Steel) served as a symbol of regional cultural identity integrated to the idea of nation, following the flow and movement around the structure of Brazilian popular music (MPB), between the national- popular and industrialization of the disk market. The first chapter discusses the transition of representations of cultural identity in Maranhão, before linked to tradition and literary canons, until achieve to the appropriations of popular culture. The second chapter discusses how popular music was used in the social construction of the idea of the Brazilian nation, through the market of symbolic goods and cultural products with the advent of mass media to the consolidation of the phonographic industry market in the 70's. The third chapter analyzes the album Bandeira de Aço (Flag Steel) hard, trying to understand the relationship of appropriation of artistic material (songs) into a commercial product (the disk) and how it served to structure the symbolic field called "maranhense popular music." / No Maranhão, desde os anos 1980, criou-se pelos meios de comunicação de massa o denominativo música popular maranhense para definir a produção musical dos compositores e intérpretes maranhenses após o lançamento do LP Bandeira de Aço, gravado por Papete, em 1978, e lançado pelo selo Discos Marcus Pereira. O disco serviu de referência para que surgisse um movimento de artes integradas à base da autenticidade do modo de vida maranhense, condensando-se no esforço estético, artístico, político e cultural, de musicalidade regional, mas inserida na proposta de ser uma expressão poético-musical brasileira. Fundamentada em teóricos como Pierre Bourdieu, Renato Ortiz, Raymond Williams, esta pesquisa quer compreender de que maneira Bandeira de Aço serviu como símbolo de identidade cultural regional integrada à ideia de nação, acompanhando o fluxo e os movimentos em torno da estruturação da música popular brasileira (MPB), no trânsito entre o nacional-popular e a industrialização do mercado de discos. The generalization of Lyapunov Exponents for flows in flag bundles is used to obtain the differenciability of certain linear combinations of the Lyapunov spectra. This specific combinations that are differentiable are determined by the caracterization of the finest Morse decomposition of the flows on flag bundles. This diploma thesis named The Personality of Jan Rys as a journalist and his work in the Flag magazine describes the personality of Jan Rys in all aspects of his life and work. In the introductory part it is set into the historical context where the state of protectorate journalism is described focusing on illegal and collaborant and activistic journalists. Part of the chapter concentrates on subsequent retribution courts. Another chapter focuses on the historical context where the origin and formation of fascism in our country is described and concentrates also on antisemitism in a Protectorate. The main part is focused on the character of Jan Rys, while his work in Vlajka movement is described whose leader he was until 1942, when he was sent at first to Terezin and than to Dachau, where he stayed till the end of war as well as his literature and his journalistic work. In the journalistic work it deals mainly with journal the Vlajka in the period when Jan Rys was an editor in chief, attention is paid to Rys' articles and their focus. A separate chapter analyzes a court trial with Jan Rys and other representatives of Vlajka, the seventh chapter concludes with the judgement of the National Court, which sentenced him to death by hanging.

Assessment of Retroviruses as Potential Vectors for the Cell Delivery of PrionsRahimi Khameneh, Shabnam January 2012 (has links)
Transmissible spongiform encephalopathies (TSEs) or prion diseases are a class of fatal brain disorders better known as Creutzfeldt-Jacob Disease (CJD) in humans, bovine spongiform encephalopathy (BSE) in cattle, scrapie in sheep, and chronic wasting disease (CWD) in deer and elk. The infectious agent responsible for these diseases is a misfolded prion protein capable of catalyzing a conformational change in normal cellular prion proteins (PrPC) into aberrant disease-causing structural isoforms (PrPSc). Although the etiological agent for TSEs has clearly been defined as PrPSc, there are important gaps in our understanding of how these proteins target and invade brain tissue. It remains to be established how ingested PrPSc ultimately reach the brain and also to understand why these tissues are particularly targeted, notwithstanding that several other tissues highly express prion proteins. Certain viruses, retroviruses in particular, efficiently hijack host proteins and can carry these proteins with them when they are released from a cell. Several lines of evidence have shown that prions and retroviruses can interact and associate at various stages of the retroviral replication cycle. Of special interest is that most retroviruses can cross the blood-brain barrier and could therefore deliver host-derived proteins to neuronal cells. In view of these observations, this thesis investigates whether retroviruses can act as vectors to capture prions from an infected cell and deliver them to a susceptible target cell.
In this work, I have cloned human and mouse prion cDNAs from PBMCs and the murine cell line NIH 3T3. Either a FLAG epitope tag or the eGFP reporter protein cDNA was inserted into a region of the prion cDNA that is predicted to be amenable to such genetic insertions without affecting protein folding or expression. I then confirmed using both fluorescent and confocal microscopy and that the recombinant proteins had a similar cell distribution to the endogenous prion protein. Using Western blot analysis, I then showed that endogenous and overexpressed prion proteins can be detected in co-transfected cells producing HIV and murine leukemia virus (MLV) retroviral particles. Finally, I went on to show that prions are also present at high levels in HIV and MLV retroviral particles released from these cells.
This work constitutes the first step in determining whether retroviruses can act as vectors for prion dissemination. Establishing a strong and clear association between retroviruses, pathogenic prions and prion disease would provide the rationale for preventive measures to be taken directly against retroviruses in order to protect humans and animals that have been newly exposed to PrPSc-infected products or those who are genetically predisposed to develop prion diseases. Anti-retroviral drugs could also be potentially used to delay disease progression and reduce prion transmission in human and animal tissues. The availability of such a treatment would constitute a significant advancement because there is currently no cure or treatment for prion diseases.

Previous issue date: 2015 / Resumo: A primeira parte do trabalho é dedicada ao estudo da formalidade geométrica em variedades flag. Uma Estrutura Riemanniana (M,g) é geometricamente formal se g possui a propriedade que todos os produtos wedge de formas harmônicas são harmônicos. Tal métrica g é chamada formal. Vamos analisar esse fato quando M é uma variedade flag usando métodos topológicos. Na verdade, mostraremos que muitas variedades flag não admitem nenhuma métrica formal g. Na segunda parte do trabalho, calcularemos os números de Chern de várias variedades flag e vamos usá-los para classificar algumas estruturas quase complexas invariantes. Além disso, mostraremos, com o auxílio do Teorema de Kodaira, que os números de Chern satisfazem algumas relações impostas pelo Teorema de Hirzebruch-Riemann-Roch / Abstract: The first part of work is dedicated to the study of geometric formality on flag manifolds. A Riemannian Structure (M,g) is geometrically formal if g has the property that all wedge products of harmonic forms are harmonic. Such metric g is called formal. We are going to analyse this fact when M is a flag manifold using topological methods. Indeed, we will show that many flag manifolds do not admit a formal metric g. In the second part of work, we will calculate Chern numbers of many flag manifolds and we are going to use them to classify some invariant almost complex structures. Furthermore, we will show with help of the Kodaira Theorem that the Chern numbers satisfy some relations imposed by the Hirzebruch-Riemann-Roch Theorem / Doutorado / Matematica / Doutor em Matemática

Soit G un schéma en groupes réductif, connexe et déployé sur l’anneau d’entiers d’une extension finie du corps de nombres p-adiques. Un théorème important dans la théorie des groupes c’est le théorème de localisation, ce qui a été démontré par A. Beilinson et J. Bernstein, et par J.L. Brylinsky et M. Kashiwara. Il s’agit d’un résultat de D-affinité pour la variété de drapeaux de la fibre générique de G. En caractéristique mixte un progrès important se trouve dans les travaux de C. Huyghe et T. Schmidt. Ils donnent une réponse partielle en considérant des caractères algébriques. Les premières quatre chapitres de cette thèse sont consacrés à étendre cette correspondance (le théorème de localisation arithmétique) pour des caractères arbitraires. Dans les chapitres cinq et six, nous traiterons l’objectif principal de cette thèse qui concerne les représentations localement analytiques. Nous montrerons que pour un caractère algébrique, qui est de plus dominant et régulier, la catégorie des représentations admissibles localement analytiques, à caractère central, c’est équivalente à une catégorie de modules arithmétiques coadmissibles et équivariants sur la famille des modèles formels de la variété de drapeaux rigide. / Let G be a split connected, reductive group scheme over the ring of integers of a finite extension of the field of p-adic numbers.. An important theorem in group theory is the localization theorem, demonstrated by A. Beilinson and J. Bernstein, and by J.L. Brylinsky and M. Kashiwara. This is a result about the D-affinity of the flag variety of the generic fiber of G). In mixed characteristic an important progress is found in the work of C. Huyghe and T. Schmidt. They give a partial answer by considering algebraic characters. The first four chapters of this thesis are dedicated to extending this correspondence (the arithmetic localization theorem) for arbitrary characters. In chapters five and six, we will treat the principal objective of this thesis, which concerns admissible locally analytic representations. We will show that for an algebraic character, which is dominant and regular, the category of admissible locally analytic representations, with central character, it is equivalent to the category of coadmissible equivariant arithmetic modules over the family of formal models of the rigid flag variety.
