Nesta dissertação, tratamos do estudo das bifurcações de um modelo bi-dimensional de presa-predador, que estende e aperfeiçoa o sistema de Lotka-Volterra. Tal modelo apresenta cinco parâmetros e uma função resposta não monotônica do tipo Holling IV: $$ \\left\\{\\begin \\dot=x(1-\\lambda x-\\frac{\\alpha x^2+\\beta x +1})\\\\ \\dot=y(-\\delta-\\mu y+\\frac{\\alpha x^2+\\beta x +1}) \\end ight. $$ Estudamos as bifurcações do tipo sela-nó, Hopf, transcrítica, Bogdanov-Takens e Bogdanov-Takens degenerada. O método dos centros organizadores é usado para estudar o comportamento qualitativo do diagrama de bifurcação. / In this work are studied the bifurcations of a bi-dimensional predator-prey model, which extends and improves the Volterra-Lotka system. This model has five parameters and a non-monotonic response function of Holling IV type: $$ \\left\\{\\begin \\dot=x(1-\\lambda x-\\frac{\\alpha x^2+\\beta x +1})\\\\ \\dot=y(-\\delta-\\mu y+\\frac{\\alpha x^2+\\beta x +1}) \\end ight. $$ They studied the sadle-node, Hopf, transcritic, Bogdanov-Takens and degenerate Bogdanov-Takens bifurcations. The method of organising centers is used to study the qualitative behavior of the bifurcation diagram.

Eficácia em longo prazo das gliflozinas versus gliptinas no tratamento do diabetes mellitus tipo 2 após falência da metformina como monoterapia: revisão sistemática e metanálise em rede / Long-term efficacy of gliflozins versus gliptins in the treatment of type 2 diabetes mellitus after metformin failure as monotherapy: systematic review and network meta-analysis

A metformina é a droga de escolha no tratamento inicial do diabetes mellitus tipo 2 (DM2). Não existe consenso na literatura sobre qual seria a segunda melhor opção terapêutica após a falência desta em longo prazo. Objetivo: avaliar a eficácia em longo prazo de gliflozinas e gliptinas após a falência do tratamento primário com metformina no DM2. Material e métodos: foi realizada uma revisão sistemática para o maior tempo de tratamento nas bases de dados bases Embase, Pubmed (via Medline), Lilacs e Cochrane Library e metanálise em rede com as sulfoniluréias (glimepirida e glipizida) como meta comparador. Desfechos: eficácia da medicação (valor final da HbA1c e porcentagem de pacientes com HbA1c < 7%), variação de peso e frequência de pacientes com hipoglicemia. Resultados: O maior tempo de segmento foi de quatro anos. Foram selecionados um artigo com empagliflozina, um artigo com dapagliflozina e um artigo com saxagliptina com dados faltantes. Após um ano de tratamento, mais de 50% dos pacientes estavam com HbA1c > 7%. O perfil de eficácia em quatro anos da empagliflozina (23%) foi melhor que da dapagliflozina (5%) e saxagliptina (7%), porém com valores de HbA1c não estatisticamente significantes (7,4 e 7,3% entre as gliflozinas), sem dados para a saxagliptina. Entretanto, a empagliflozina foi superior à glimepirida no período de quatro anos (diferença média padronizada/DMP: 0,40, intervalo de confiança/IC95%: 0,23- 0,56). A variação de peso permaneceu estável após um ano de tratamento, com vantagem em quatro anos para a empa (DMP: 1,56, IC95%: 1,23- 1,88). A frequência de pacientes com hipoglicemia não diferiu entre empagliflozina e dapagliflozina (razão de chances: 1,53, IC95%: 0,80- 2,91) e foi significativamente menor do que em relação às sulfoniluréias. Conclusões: a falência da segunda terapia com gliflozinas ocorre em menos de um ano de tratamento ( > 50% dos pacientes com HbA1c > 7%). A empagliflozina obteve um controle glicêmico melhor em relação às sulfoniluréias, porém semelhante à dapagliflozina. A perda de peso foi mantida por quatro anos, com superioridade para empagliflozina. Houve uma baixa frequência de hipoglicemia nas gliflozinas em comparação com as sulfoniluréias. Mais estudos são necessários para avaliar a eficácia de gliptinas e gliflozinas em longo prazo, após a falência terapêutica com metformina / Metformin is the first-choice treatment in people with type 2 diabetes (TD2). There is no consensus in the medical literature about which drug would be a second-best option of treatment in the case of metformin failure in long-term. Objective: to assess the long-term efficacy of gliflozins and gliptins once metformin has failed as the primary treatment for TD2. Materials and methods: a systematic review was performed considering the longest period found in Embase, Pubmed (via Medline), Lilacs and Cochrane Library databases and also network meta-analyses using sulfonylureas (glimepiride and glipizide) as a meta comparator. Clinical outcomes where efficacy of medical treatment (final value of HbA1c and percentage of patients with HbA1c < 7%), weight variation and frequency of patients with hypoglycemia. Results: the longest period of the segment was 4 years. It was selected 1 article on empagliflozin, 1 article on dapagliflozin, and 1 article on saxagliptin with missing data. After one year of treatment, over 50% of the patients presented HbA1c > 7%. Efficacy rate in 4 years of empagliflozin (23%) was better than dapagliflozin (5%) and saxagliptin (7%), however presenting statistically non-significant values for HbA1c (7.4 and 7.3% between gliflozins), and missing data for the saxaglifozin. Nonetheless, empagliflozin performed better than glimepiride in the 4-year period (standardized mean difference SMD 0.4, confidence interval CI 95% 0.23 to 0.56). Weight variation remained stable after one year of treatment, presenting empagliflozin better results in the 4-year period (SMD 1.56, CI 95% 1.23 to 1.88). The frequency of patients with hyperglycemia did not vary for empagliflozin and dapagliflozin (odds ratio 1.53, CI 95% 0.8 to 2.91), and it was significantly lower when compared to the sulfonylureas. Conclusions: the failure of the secondary treatment using gliflozins occurs in less than one year of treatment (less than 50% of the patients presenting HbA1c > 7 %). Empagliflozin offered a better glycemic control compared to the sulfonylureas, but similar to dapagliflozin. The weight loss was maintained for 4 years, being empagliflozin the one with better results. There was a low frequency of hypoglycemia for the gliflozins when compared to the sulfonylureas. Further studies are required to evaluate the efficacy of gliptins and gliflozins in the long-term after metformin failure

Padrões histopatológicos e deposição de colágenos durante a progressão da fibrose hepática como fatores prognósticos da atresia de vias biliares / Histopathological and collagens deposition patterns during hepatic fibrosis progression as prognostic factors in biliary atresia

Atresia de vias biliares (AVB) é uma hepatopatia colestática específica da criança, de etiologia desconhecida, com evolução para fibrose hepática precoce. AVB é a principal causa de cirrose na infância e principal indicação de transplante hepático pediátrico (Tx). Compreender os fatores envolvidos na progressão da fibrose é fundamental para estabelecer tratamentos efetivos nas hepatopatias crônicas. Identificar padrões histopatológicos associados ao prognóstico da AVB permitiria melhor planejamento dos centros de transplante e adequado aconselhamento familiar. OBJETIVO: Estabelecer padrões de marcadores histopatológicos e de imunofluorescência para colágenos em biópsias hepáticas iniciais e finais de pacientes com AVB submetidos a tratamento cirúrgico. Correlacionar esses marcadores com o prognóstico da doença, definido com base no tempo de evolução até realização do Tx. MÉTODO: Avaliação histológica de alterações biliares e fibrose hepática e histomorfometria da fibrose marcada por picrossírius e da deposição dos colágenos tipos I, III, IV e V marcados por imunofluorescência indireta (IF), em biópsias hepáticas iniciais e finais de 36 pacientes com AVB submetidos à hepatoportoenterostomia de Kasai (KPE) e ao Tx nos últimos 20 anos em nossa instituição. RESULTADOS: A mediana das idades de realização da KPE foi de 12,5 semanas (6-20) e do Tx foi de 27 meses (6-120). Reação ductular e malformação de placa ductal foram mais intensas nas biópsias iniciais (p < 0,05), enquanto fibrose hepática e ductopenia apresentaram padrão progressivo (p < 0,001), sem correlações com a idade de realização da KPE nem com o tempo de evolução até Tx. A morfometria da fibrose hepática marcada pelo picrossírius nas biópsias iniciais apresentou correlação positiva com a idade da KPE (p = 0,01), mas não com a idade do Tx (p = 0,24). A deposição perissinusoidal dos colágenos dos tipos III e V foi mais intensa nas biópsias iniciais (p < 0,01), enquanto os colágenos dos tipos I e IV apresentaram padrão de deposição progressiva (p < 0,01). Pacientes com maior deposição perissinusoidal de colágeno tipo I nas biópsias iniciais apresentaram curva de tempo de evolução até Tx sugerindo pior prognóstico (p = 0,04). CONCLUSÃO: Marcadores histopatológicos de alterações biliares, fibrose hepática e deposição de colágenos apresentaram características evolutivas distintas nas fases inicial e final da AVB, sem correlação com o tempo de evolução até Tx. A morfometria da deposição perissinusoidal de colágeno tipo I em biopsias iniciais marcadas por IF pode ser correlacionada ao tempo de evolução até Tx em pacientes com AVB operada / Biliary atresia (BA) is a specific cholestatic liver disease of unknown etiology that affects children and progresses to early hepatic fibrosis. BA is the main indication of pediatric liver transplantation (LTx). Understanding the factors involved in the progress of fibrosis is essential to establish effective treatment to chronic liver disease. Histopathological markers in liver biopsies could be useful to predict progression to end stage disease and to make it possible to improve planning in transplantation centers and parental orientation. OBJECTIVE: To establish histopathological or immunohistochemical markers in initial or final liver biopsies of BA patients and correlate those markers with prognosis, as defined by progression time lapse until LTx. METHOD: Histological analysis of multiple parameters of biliary alterations and morphometrical assessment of liver fibrosis were performed, besides indirect immunofluorescence assays (IF) for type I, III, IV and V collagens in initial and final liver biopsies of 36 patients with BA submitted to Kasai hepatoportoenterostomy (KPE) and LTx in the last 20 years in a single center. RESULTS: The median of the ages at KPE was 12.5 weeks (6-20) and at LTx was 27 months (6-120). Ductular reaction and ductal plate malformation were more severe in the initial biopsies (p < 0.05), while ductopenia and liver fibrosis were more severe in final biopsies (p < 0.001), though without correlation with age at KPE nor with progression time lapse until LTx. Morphometrical assessment of liver fibrosis marked by picrosirius red in initial biopsies demonstrated positive correlation with age at KPE (p = 0.01) but not with age at LTx (p = 0.24). The perisinusoidal deposition of type III and V collagens was more extended in the initial biopsies (p < 0.01), while type I and IV collagens deposition indicated progression (p < 0.01). Patients with large amounts of perisinusoidal type I collagen in the initial biopsies had worse progression time curve until LTx (p = 0.04). CONCLUSION: Biliary alterations, liver fibrosis and collagens deposition demonstrated distinctive progression findings in the initial or final phases of the BA, without prognostic correlation. Morphometrical assessment of perisinusoidal deposition of type I collagen by IF in the initial biopsies can be correlated with progression time until LTx in patients with post-surgical BA

Mécanismes moléculaires impliqués dans la formation de biofilm à l’interface eau-composés organiques hydrophobes / Molecular mecanisms involved in the bacterial biofilm formation at the water-hydrophobic organic compound interface

Les composés organiques hydrophobes (HOC), une grande famille de molécules naturelles ou d’origine anthropique incluant les lipides et les hydrocarbures, constituent une part significative de la matière organique dans les écosystèmes marins. Du fait de leur faible solubilité dans l’eau, les bactéries qui les dégradent requièrent la mise en place de fonctions cellulaires spécifiques permettant d’augmenter la fraction assimilable de ces HOC. La formation de biofilms à l’interface eau-HOC est une de ces stratégies adaptatives. C’est le cas pour Marinobacter hydrocarbonoclasticus SP17, modèle d’étude utilisé au laboratoire, qui est capable de former des biofilms sur un large spectre de HOC métabolisables tels que les alcanes, les triglycérides et les alcools gras. Le but de mes recherches consistait à améliorer la compréhension du processus d’adhésion et de développement des biofilms sur les HOC, à travers la caractérisation fonctionnelle de 10 gènes candidats mis en évidence lors d’analyses d’expression en protéomique et en transcriptomique. Pour mener à bien ce projet, des outils génétiques et une caractérisation fonctionnelle propre à chaque gène ont dû être développés. L’étude fonctionnelle du gène MARHY2686 a relevé son implication dans la formation de biofilm sur les alcanes. La co-expression de MARHY2686 et des gènes adjacents MARHY2687 et MARHY2685 en transcriptomique, leur distribution phylogénétique et leur conservation de la synthénie suggèreraient que ces trois gènes soient impliqués dans le même processus biologique. D’après l’identité forte de 36 % qui existe entre la protéine MARHY2686 et une protéine périplasmique AdeT d’un système de pompe d’efflux tripartite d’Acinetobacter baumanii, cette protéine, en association avec MARHY2687 et MARHY2685, pourrait faire partie d’un système de ce type. Par ailleurs, des observations ont permis d’envisager une implication potentielle de ce gène dans l’assimilation des HOC ou dans l’accumulation des réserves lipidiques intracellulaires. M. hydrocarbonoclasticus SP17 utilise les pili de type IV lors de la formation de biofilm sur les HOC. Ces appendices interviennent lors de l’adhésion de cette souche à des HOC ainsi que dans un processus de détachement d’un support hydrophobe. Les pili pourraient soit intervenir directement pour permettre à la bactérie de se détacher de la surface à laquelle elle s’est adhérée, soit indirectement par l’action de bactériophages. La présence d’une mobilité de type twitching sur les HOC a pu être également envisagée. Enfin, le rôle du système de sécrétion de type VI (T6SS), connu pour permettre à la bactérie d’interagir avec une cellule hôte, lors de la formation de biofilm mono-spécifique sur HOC, où aucun autre microorganisme que M. hydrocarbonoclasticus SP17 n’est présent, a été étudié. / Hydrophobic organic compounds (HOC), a large family of naturally-produced or anthropogenic molecules including lipids and hydrocarbons, represent a significant part of organic matter in marine ecosystems. Because of their low solubility in water, bacteria that degrade those compounds require the establishment of specific cell functions to increase their biodisponibility. Biofilm formation in water-HOC interface is one of these adaptations. The model of bacteria used in our laboratory, Marinobacter hydrocarbonoclasticus SP17, is able to form a biofilm on a wide range of HOC, such as alkanes, fatty alcohols and triglycerides, in order to use them as a carbon and energy source. The main purpose of my work was to broaden the knowledge of how bacteria adhere to and from biofilms on HOC, through the functional characterization of 10 candidate genes highlighted during proteomic and transcriptomic studies. Genetic tools and a gene-specific functional characterization have been developed in order to carry out this project. Functional study conducted on MARHY2686 revealed its involvement in the formation of biofilm on alkanes. Co-expression of MARHY2686 and the adjacent genes MARHY2687 and MARHY2685 durnig transcriptomic analysis together with their phylogenetic distribution and synteny conservation suggest that these three genes are involved in the same biological process. According to the high peptide sequence identity between MARHY2686 and AdeT, a periplasmic protein of a tripartite efflux pump system of Acinetobacter baumanii, MARHY2686 in combination with MARHY2687 and MARHY2685 could be the components of such a system. Other phenotypic observations would consider the involvement of MARHY2686 either in the assimilation of HOC or in the accumulation of intracellular lipid reserves. M. hydrocarbonoclasticus SP17 uses type IV pili during biofilm formation on HOC. These appendages are involved in the adhesion of this strain to and in a detachment process from HOC. Type IV pili could either act directly to allow bacteria to detach from the surface to which it is adhered, or indirectly through the action of bacteriophages. The presence of twitching motility on HOC has also been suggested. Finally, the role of the type VI secretion system (T6SS), a well-known protein system which allows interactions between bacteria and host cells, during the formation of a mono-species biofilm on HOC where no other microorganism than M. hydrocarbonoclasticus SP17 is present, has been studied.

Résolution de l’équation de transport de Boltzmann pour les phonons et applications / Solving Boltzmann transport equation for phonons and applications

Cette thèse est consacrée à l’étude de la dynamique et du transport des phonons via la résolution de l’équation de transport de Boltzmann (ETB) pour les Phonons. Un ‘solveur’ Monte Carlo dédié à la résolution de l’ETB des phonons dans l’espace réciproque, prenant en compte tous les processus d’interactions Normaux et Umklapp à trois-phonons, est proposé. Une prise en compte rigoureuse des lois de conservation de l’énergie et de la quantité de mouvement est entreprise. Des relations de dispersion réalistes, intégrant tous les modes de polarisations, sont considérées. Le calcul des taux d’interactions à trois-phonons de tous les processus Normaux et Umklapp est effectué en utilisant l’approche théorique due à Ridley qui ne nécessite qu’un unique paramètre semi-ajustable pour chaque mode de polarisation, nommément : le coefficient de couplage anharmonique représenté par les constantes de Grüneisen. Les taux d’interactions ainsi calculés ne servent pas uniquement à la résolution de l’ETB des phonons, mais ont permis aussi une analyse complète des canaux de relaxation des phonons longitudinaux optiques de centre de zone. Cette analyse a montré que le canal de Vallée-Bogani est négligeable dans le GaAs, et que vraisemblablement les temps de vie des phonons LO de centre de zone dans l’InAs et le GaSb rapportés dans la littérature sont fortement sous-estimés. Pour la première fois à notre connaissance, un couplage de deux solveurs Monte Carlo indépendants l’un dédié aux porteurs de charges (Thèse E. Tea) et l’autre dédié aux phonons, est effectué. Cela permet d’étudier l’effet des phonons chauds sur le transport des porteurs de charges. Cette étude a montré que l’approximation de temps de relaxation surestime souvent l’effet bottleneck des phonons. Le ‘solveur’ Monte Carlo est étendu pour résoudre l’ETB des phonons dans l’espace réel (en plus de l’espace réciproque), cela a permet d’étudier le transport des phonons et ainsi de la chaleur. La théorie généralisée de Ridley est toujours utilisée avec des particules de simulations qui interagissent les unes avec les autres directement. Les règles de conservation de l’énergie et de la quantité de mouvement sont rigoureusement respectées. L’effet des processus Umklapp sur la quantité de mouvement totale des phonons est fidèlement traduit; tout comme l’effet des interactions sur les directions des phonons, grâce à une procédure prenant en compte les directions vectorielles respectives lors d’une interaction, au lieu, de la distribution aléatoire usuellement utilisée. Les résultats préliminaires montrent la limite de l’équation analytique de conduction de la chaleur. / This work is dedicated to the study of phonon transport and dynamics via the solution of Boltzmann Transport Equation (BTE) for phonons. The Monte Carlo stochastic method is used to solve the phonon BTE. A solution scheme taking into account all the different individual types of Normal and Umklapp processes which respect energy and momentum conservation rules is presented. The use of the common relaxation time approximation is thus avoided. A generalized Ridley theoretical scheme is used instead to calculate three-phonon scattering rates, with the Grüneisen constant as the only adjustable parameter. A method for deriving adequate adjustable anharmonic coupling coefficients is presented. Polarization branches with real nonlinear dispersion relations for transverse or longitudinal optical and acoustic phonons are considered. Zone-center longitudinal optical (LO) phonon lifetimes are extracted from the MC simulations for GaAs, InP, InAs, and GaSb. Decay channels contributions to zone-center LO phonon lifetimes are investigated using the calculated scattering rates. Vallée-Bogani’s channel is found to have a negligible contribution in all studied materials, notably GaAs. A comparison of phonons behavior between the different materials indicates that the previously reported LO phonon lifetimes in InAs and GaSb were quite underestimated in the literature. For the first time, to our knowledge, a coupling of two independent Monte Carlo solvers, one for charge carriers [PhD manuscript, E. TEA], and one for phonons, is undertaken. Hot phonon effect on charge carrier dynamics is studied. It is shown that the relaxation time approximation overestimates the phonon bottleneck effect. The phonon MC solver is extended to solve the phonon’s BTE in real space simultaneously with the reciprocal space, to study phonon and heat transport. Ridley’s generalized theoretical scheme is utilized again with simulation particles interacting directly together. Energy and momentum conservation laws are rigorously implemented. Umklapp processes effect on the total phonon momentum is thoroughly reproduced, as for the anharmonic interactions effect on resulting phonon directions. This is thanks to a procedure taking in consideration the respective vector directions during an interaction, instead of the randomization procedure usually used in literature. Our preliminary results show the limit of the analytic macroscopic heat conduction equation.

Objective: to assess the long-term efficacy of gliflozins and gliptins once metformin has failed as the primary treatment for TD2. Materials and methods: a systematic review was performed considering the longest period found in Embase, Pubmed (via Medline), Lilacs and Cochrane Library databases and also network meta-analyses using sulfonylureas (glimepiride and glipizide) as a meta comparator. Clinical outcomes where efficacy of medical treatment (final value of HbA1c and percentage of patients with HbA1c < 7%), weight variation and frequency of patients with hypoglycemia. Results: the longest period of the segment was 4 years. It was selected 1 article on empagliflozin, 1 article on dapagliflozin, and 1 article on saxagliptin with missing data. After one year of treatment, over 50% of the patients presented HbA1c > 7%. Efficacy rate in 4 years of empagliflozin (23%) was better than dapagliflozin (5%) and saxagliptin (7%), however presenting statistically non-significant values for HbA1c (7.4 and 7.3% between gliflozins), and missing data for the saxaglifozin. Nonetheless, empagliflozin performed better than glimepiride in the 4-year period (standardized mean difference SMD 0.4, confidence interval CI 95% 0.23 to 0.56). Weight variation remained stable after one year of treatment, presenting empagliflozin better results in the 4-year period (SMD 1.56, CI 95% 1.23 to 1.88). The frequency of patients with hyperglycemia did not vary for empagliflozin and dapagliflozin (odds ratio 1.53, CI 95% 0.8 to 2.91), and it was significantly lower when compared to the sulfonylureas. Conclusions: the failure of the secondary treatment using gliflozins occurs in less than one year of treatment (less than 50% of the patients presenting HbA1c > 7 %). Empagliflozin offered a better glycemic control compared to the sulfonylureas, but similar to dapagliflozin. The weight loss was maintained for 4 years, being empagliflozin the one with better results. There was a low frequency of hypoglycemia for the gliflozins when compared to the sulfonylureas. Further studies are required to evaluate the efficacy of gliptins and gliflozins in the long-term after metformin failure

Décrivant l'ascension sociale d'un groupe de chevaliers valant par leur seule compétence, le "Poème du Cid" proclame le modèle d'une seigneurie personnelle destinée à corriger les effets néfastes d'un régime fondé essentiellement sur la 'nature'. Vouée à servir, quant à elle, les aspirations centralisatrices de la couronne, l'historiographie alphonsine tente d'imposer le modèle d'un ordre royal. Deux discours, donc, différents tant dans leur forme que dans les valeurs qu'ils véhiculent. Et pourtant, le "Poème du Cid" constitue l'essentiel du matériau narratif de l'"Histoire d'Espagne" pour l'histoire de Rodrigue Diaz et d'Alphonse VI. Quels furent, dans ces conditions, les procédés de transfert du discours épique au discours historiographique? Une étude minutieuse des similitudes que présentent les différentes versions de l'"Histoire d'Espagne" ("Chronique de vingt rois", "Version sancienne", "Chronique de Castille") permet de dégager les deux critères de sélection de l'information narrative: sa vraisemblance et sa fonctionnalité. Quatre opérations traduisent les mécanismes d'intégration de la geste à l'histoire: la segmentation, l'uniformisation, l'abréviation et surtout l'amplification. Sur ces remaniements repose l'essentiel du propos monarchique: réaliser la centralisation juridique et valoriser l'acte exemplaire de dépendance du sujet face au seigneur naturel. L'analyse des écarts entre les textes nous informe, à un autre niveau, de l'évolution du discours historiographique lui-même. Si le propos de l'historiographie reste inchangé, en revanche, la voix des destinateurs du récit s'exprime de façon plus personnelle. Ainsi, le texte accorde une large place à la noblesse chevaleresque, ces hommes désireux de gravir la hiérarchie des états en entrant au service du roi et qui, dans le dernier tiers du XIVe siècle, formeront les grandes Maisons Trastamare.

Les sites apuriniques/apyrimidinique (AP) représentent une forme de dommage à l’ADN hautement mutagène et ce type de dommage peut survenir spontanément ou être induit par une variété d’agents. Afin de préserver la stabilité génomique, deux familles d’endonucléases de type AP, endo-IV et exo-III, sont nécessaires pour contrecarrer les effets mutagènes des sites AP. Malgré l’identification de membres des deux familles dans plusieurs organismes unicellulaire tels que E.coli et S. cerevisiae, aucun membre de la famille endo-IV n’a été identifié chez les organismes multicellulaires à l’exception de C. elegans et de C. briggsae. Nous avons donc décidé d’investiguer l’importance biologique de APN-1 chez C. elegans par l’utilisation d’une approche de knockdown du gène. Dans notre étude, nous avons montré que le knockdown du gène apn-1 chez C. elegans, en utilisant des ARN d’interférence (ARNi), cause une accumulation de mutations spontanées et induites par des drogues résultant en un délai de l’éclosion des œufs ainsi que par une diminution de la survie et de la longévité des vers adultes. De plus, nous avons montré que cette accumulation de mutations mène à un délai dans la progression du cycle cellulaire durant l’embryogénèse, représentant possiblement une explication du délai dans l’éclosion des œufs. Nous avons montré qu’il y avait une augmentation du niveau de mutations dans la gorge des vers, sans toutefois pouvoir confirmer la distribution de APN-1 qui possède une étiquette GFP. Les animaux transgéniques APN-1-GFP n’exprimaient pas suffisamment de la protéine de fusion pour permettre une visualisation à l’aide d’un microscope à fluorescence, mais la protéine a été détectée par immunobuvardage de type western. Les animaux transgéniques APN-1-GFP étaient instables et avaient des phénotypes concordants avec les défauts génétiques. En conclusion, il semble que C. elegans aie évolué afin de retenir un niveau de base de APN-1 jouant ainsi un rôle versatile afin de maintenir l’intégrité génétique d’autant plus que cet organisme semble manquer plusieurs enzymes de la voie de réparation par excision de base. / Apurinic/apyrimidinic (AP) sites are a form of highly mutagenic DNA damage that arise either spontaneously or by a variety of DNA damaging agents. To preserve genomic stability two AP endonuclease families, endo-IV and exo-III, evolved to counteract the mutagenic effect of AP sites. While members of both families were identified in multiple unicellular organisms, notably E. coli and S. cerevisiae, no members of the endo-IV family were identified in multicellular ones, with the exception of C. elegans and its close relatives, particularly C. briggsae. We set out to investigate the biological importance of APN-1 in C. elegans using gene knockdown approach. In our study, we showed that the knockdown of C. elegans apn-1 gene, using RNAi causes the accumulation of spontaneous and drug induced mutations, resulting in a delay in egg hatching, decreased survival and longevity. Furthermore, we have showed that the accumulated mutations lead to delays in cell cycle progression during early embryogenesis, thus providing a possible explanation for the observed delay in hatching. Although we showed increased mutations in the gut of the worm, we were unable to confirm APN-1 distribution tagged with GFP. The transgenic APN-1-GFP animal did not express enough of this fusion protein to be visualized by fluorescent microscopy, although it was detected by Western blot analysis. The transgenic animals over-expressing APN-1-GFP were unstable and showed phenotypes consistent with genetic defects. In conclusion, it would seem that C. elegans has evolved to retain a balanced level of APN-1, which plays a versatile role in maintaining genetic integrity, since this organism lacks a full complement of the enzymes in the base-excision repair pathway.
