Removal of hexavalent chromium in wastewater using granular ferric hydroxides

This study took place in Malawi, south eastern Africa. Metal contamination of water and soil is a threat to the environment and human health and is a serious concern in many countries including Malawi. Blantyre is the city of commerce where most of the industry is located along the banks of the main rivers. Some of the industries produce wastewater, which due to poor access to wastewater treatment plants, is discharged without treatment into the environment. A match factory in Blantyre uses chromium as a colouring agent for match heads and very high concentrations of chromium(VI) have been measured downstream the factory with concentrations up to 56 mg/l, which is much higher than the WHO guidelines of 0.05 mg/l.  Chromium(VI) mainly occurs as chromate CrO42- and dichromate Cr2O72- ions. They are both toxic and carcinogenic and can cause mutations and chromosomal aberrations.   The aim of the study was to evaluate the efficiency of Granular Ferric Hydroxide (GFH) in adsorbing and removing chromium from an aqueous phase. Wastewater was collected from the match factory and by optimizing relevant parameters the removal efficiency was maximized. The parameters optimized were pH, dosage and contact time and the effect of initial concentration. After respective experiment, all samples were analysed for chromium using microwave plasma atomic emission spectroscopy (MP-AES).   The optimum pH was chosen to 8.0, the dosage to 17.4 g GFH/l and the contact time to 2 hours for a 95% removal of total chromium in undiluted wastewater. The removal efficiency of the GFH was 2.84 mg Cr/g GFH. To implement wastewater purification with GFH a number of practical issues have to be taken into consideration. Above all, a sufficient stirring method has to be devised as the removal efficiency is much affected by improper stirring. / Denna studie genomfördes i Malawi, sydöstra Afrika. Metallföroreningar i vatten och mark är ett stort problem i många länder, inklusive Malawi. Landets centrum för industri och handel ligger i Blantyre där de flesta av industrierna ligger längs med floderna. En del industrier genererar utsläppsvatten och på grund av dålig tillgång till vattenreningsteknik och vattenreningsanläggningar släpps mycket av det förorenade vattnet obehandlat ut i naturen. En tändsticksfabrik i Blantyre använder kromsalter för att färga tändstickshuvudena röda och höga koncentrationer av sexvärt krom har uppmätts nedströms fabriken. De uppmätta koncentrationerna var upp till 56 mg/l vilket är betydligt högre än WHO:s riktvärde på 0,05 mg/l. Sexvärt krom förekommer som kromat CrO42- och dikromat Cr2O72- joner vilka båda är mycket toxiska och cancerframkallande och därmed ett hot mot miljö och människor. Syftet med studien var att undersöka om granulära järnoxider (GFH) kunde användas som en adsorbent för att rena utsläppsvatten från kromater. Utsläppsvatten hämtades från tändsticksfabriken och genom att optimera relevanta parametrar kunde GFHns adsorbtionseffektivitet maximeras. Parametrarna som optimerades var pH, dos, kontakttid och initialkoncentration. Efter respektive experiment mättes kromkoncentrationen med mikrovågsplasma atomemissionsspektroskopi (MP-AES) Det optimala pH-värdet valdes till 8.0, dosen till 17.4 g GFH/l med en kontakttid på 2 timmar för en 95.3% reningsgrad på outspätt utsläppsvatten. Adsorptionseffektiviteten på GFHn var 2.84 mg Cr/g GFH. För att implementera vattenrening med GFH behöver flera praktiska aspekter tas med i beaktning. Framförallt är det viktigt med en bra omrörning för att GFHn ska kunna adsorbera effektivt.

Šiame eksperimente buvo nagrinėjama pH įtaka Cr (VI) ir Cu (II) jonų pašalinimui iš vandeninių tirpalų, naudojant metalinę geležį Fe (0), kai tirpalų pH vertės buvo nuo 2 iki 6, purtymo procese, uždaruose ir atviruose induose. Kadangi Cr (VI) ir Cu (II) jonų koncentracijų sumažėjimas tirpale priklauso nuo geležies paviršiaus, tai labai svarbu ją tinkamai paruošti: išvalyti ir aktyvuoti. Maksimalus Cr (VI) jonų pašalinimo efektyvumas buvo esant pH vertei 2, o Cu (II) jonų atveju, kai pH vertė 4 uždaruose induose ir atviruose induose – chromo atveju pH 6, vario atveju, kai pH 4. Pašalinimo įvertinimui naudojamos kalibraciniės kreivės pagal kurias apskaičiuojamos likusiųjų jonų koncentracijos, matuojant tirpalų optinius tankius. Metodas yra pigus ir patikimas, nes pašalinima didelė dalis sunkiųjų metalų jonų. / The effects of pH on Cr (VI) and Cu (II) ions removal from solutions by Fe (0) was investigated by batch shaking process. Since the presence of ferrous iron in the solution is mainly responsible for Cr (VI) and Cu (II) reduction the surface of iron must be purified and activated. The maximum Cr (VI) removal efficiency was found at pH 2 and in Cu (II) case it was pH 4 without oxygen and in case with open jar it was for Cr (vi) removal pH 6 and for Cu (II) removal pH 4. Remained concentrations of Cu (II) and Cr (VI) ions were investigated using calibrated graphs and analysis of optical density. This study confirmed that this method is cheap because we can use wasted iron and effective of removing most of heavy metal ions from solutions.

As mucopolissacaridoses apresentam uma história natural progressiva, causada por defeitos no metabolismo dos glicosaminoglicanos. Frequentemente graves, as mucopolissacaridoses encurtam de forma considerável a expectativa de vida do paciente. Apesar de que em muitos casos a função intelectual é normal, morbidade neurológica considerável pode ser causada por compressão medular secundária ao acúmulo de glicosaminoglicanos nas meninges. O tratamento deste problema pode requerer a descompressão medular através de laminectomia cervical. A terapia de reposição enzimática endovenosa, para o tratamento de mucopolissacaridose, reduz o acúmulo lisossômico e alivia muitos dos sintomas da doença, porém não oferece benefício direto para o sistema nervoso central uma vez que não atravessa a barreira hemato-encefálica. Esta limitação da reposição enzimática endovenosa levou alguns pesquisadores a trabalhar com uma nova opção de via de liberação medicamentosa de alcance direto no sistema nervoso central, aproveitando o extenso contato que existe entre o líquido cefaloraquidiano e as meninges e com as granulações aracnoideas, para o tratamento de algumas doenças de depósito lisossomal. Estudos em modelos animais têm sido conduzidos e com resultados promissores. Este trabalho se propõe a estudar uma nova via de administração da enzima recombinante, diretamente no espaço liquórico que foi utilizada em dois pacientes com MPS I e em um paciente com MPS VI, com acesso a esta terapêutica através de uso compassivo individual aprovado pelo Comitê de Ética, no Hospital de Clínicas de Porto Alegre. Até então, apenas estudos animais haviam sido realizados abordando esta via de acesso em doenças de depósito lisossomal e estes pacientes foram os primeiros indivíduos com MPS no mundo a receber terapia de reposição intratecal para o tratamento de compressão medular sintomática por depósito de glicosaminoglicanos. Em 2005 um paciente adulto com MPS I apresentando compressão medular foi incluído em um protocolo de terapia de reposição enzimática intratecal por uso compassivo no Hospital de Clínicas de Porto Alegre. Em 2006 uma menina com MPS I apresentando compressão cervical medular sintomática também recebeu terapia de reposição enzimática intratecal por uso compassivo no Hospital de Clínicas de Porto Alegre. Em 2007 um menino com MPS VI e com compressão medular cervical também foi tratado através de reposição enzimática intratecal por uso compassivo neste mesmo hospital. / The mucopolysaccharidoses present a progressive natural course caused by defects on glycosaminoglycan degradation pathways. Usually severe, the mucopolysaccharidoses considerably shorten patient lifespan. Although in many cases the cognitive function is preserved, considerable neurological morbidity can be present due to spinal cord compression which is secondary to glycosaminoglycan storage in the meninges. Treatment for this complication usually requires surgical intervention with cervical laminectomy for thickened meninges removal. Enzyme replacement therapy used for the treatment of mucopolysaccharidoses reduces lysosomal storage and ameliorates many somatic symptoms but does not provide any direct benefit to central nervous system as the enzyme does not cross the blood-brain-barrier. Due to this limitation of intravenous enzyme replacement therapy some researchers have been working with an alternative option of enzyme delivery with direct action on central nervous system through the extensive close contact provided by cefalo-spinal fluid and meniniges and arachnoid villosities, to the treatment of some lysosomal disorders. Animal model studies have been conducted and some promising results have been achieved. This study intends to present an alternative route for the administration of a recombinant enzyme, directly in the cefalo-spinal fluid, which was used in two patients with mucopolysaccharidosis I and one patient with mucopolysaccharidois VI. These patients gained access to this therapy by individual compassionate use enrollment approved by local Ethics Board at Hospital de Clínicas de Porto Alegre. So far, only animal model trials had been conducted with the use of this administration route in lysosomal storage diseases, and these were the first three patients with mucopolysaccharidoses and cord compression to receive intrathecal enzyme replacement therapy in the world. In 2005, an adult mucopolysaccharidosis I patient presenting cervical cord compression was enrolled in a compassionate use trial of intrathecal enzyme replacement therapy, at the Hospital de Clínicas de Porto Alegre. In 2006, a girl with mucopolysaccharidosis I presenting spinal cord compression was also enrolled in a compassionate use trial of intrathecal enzyme replacement therapy, at the Hospital de Clínicas de Porto Alegre. Teve por objetivo pesquisar a representação social do professor de Monte Santo-BA acerca do aluno com uma doença genética rara: a mucopolissacaridose tipo VI. Para desenvolvê-lo, foram observados os princípios da pesquisa qualitativa e do estudo de caso coletivo, tomando como orientação teórico-metodológica a teoria das representações sociais. Participaram do estudo 7 professores, de ambos os sexos, que trabalham em 3 escolas municipais, localizadas em 3 diferentes povoados da cidade de Monte Santo-BA, e que lecionam ou que já lecionaram para tais alunos. Os procedimentos de coleta de dados foram a observação e as entrevistas semiestruturadas. Após a coleta, submeteu-se todo material à análise de conteúdo, o que possibilitou identificar a representação social, segundo os professores, sobre alunos com uma doença incomum. Os resultados alcançados mostram que, segundo os referidos docentes, esses alunos não têm uma doença, mas apenas uma deficiência física, já que ela é visível; eles são bons alunos, pois, apresentam facilidade para aprender; são sociáveis, têm ajuda da família, são curiosos e participativos. São praticamente normais, embora apresentem um atributo depreciativo visível que leva a uma baixa atratividade física, em geral, e facial, fato que, do ponto de vista dos professores, constitui-se numa aparência anormal. São considerados alunos com necessidades educacionais especiais, simplesmente por apresentarem condição de deficiência física; esta condição, para os docentes, requer um professor especialista, haja vista que, segundo seu ponto de vista, a educação especial ainda é uma modalidade da educação paralela ao ensino convencional, oferecida unicamente em salas de recursos multifuncionais. A educação especial também é entendida como sinônimo de educação inclusiva e significa somente a inserção de alunos, em condição de deficiência, na sala de aula comum. Esses achados demonstram que os professores compreendem a mucopolissacaridose tipo VI apenas como uma deficiência física e que eles foram e ainda estão sendo formados com ideias superficiais acerca da educação especial, na perspectiva da educação inclusiva; isso leva à formação de profissionais sem capacitação para promover a inclusão de fato, na medida em que os próprios formadores desses novos docentes estão distantes desse processo. Se a educação almeja professores que lecionem com qualidade para todos os alunos, eles também precisam ser formados com qualidade, fato que requer o compromisso de todos: escola, família, serviços da saúde, transporte, trabalho; ademais, esse compromisso implica investimentos do governo mediante políticas públicas, tendo em vista que se a finalidade é a educação para todos, então, a responsabilidade deve ser de todos / ABSTRACT This coursework is the result of reflections regarding my professional practice as a teacher in a hospital of the University Hospital Professor Edgard Santos, located in Salvador, Bahia, It was aimed to research about the social representation of teachers from Monte SantoBA town, in which there is a student with a genetic rare illness: mucopolysaccharidosis type VI. To develop th is search, the principles of qualitative research and the collective case study were observed and used, also having the theoretical and methodological orientation to the theory of social representations. The study participants included 07 teachers, men and women, working in 03 public schools located in 03 different villages in the town of Monte SantoBA that had already taught or still teach these students. As data collection, it was used the observation and semistructured interviews. After collecting the info, it was possible to indentify the social representation when it came to the teachers’ part, regarding the students who sufferent from that unusual disease. Among the results achieved, it was found that for teachers these students do not have a disease, but a deficiency, related to physical par t, as it is visible. They are good students, because they do have ability to learn, they are sociable, have family help, are curious and participative. They are practically normal, although they have this visible deformation that is not atractive when it comes to facial and physical formation, in which, in the teachers' point of view, it is considered to be an abnormal appearance. They are considered students with special educational needs, just by being a person on disability status, and for teachers, these students need the help of an expert teacher. Given that, in their view, special education is still education that must be sidelined to formal education, and is mostly just offered in schools that count with multifunctional facilities. Also, special education is understood as a synonym for inclusive education, and only means placing students in deficiency condition in a standard classroom. These findings show that teachers have little knowledge of MPS VI, and they have been and are still being formed wrongly and with vague ideas about the special education from the perspective of formation of professionals who are not able to include in fact, as the trainers themselves of these new teachers are far from this process. If education aims teachers who can teach all students with quality, they also need to be formed with quality; which requires the commitment of all: school, family, health services, education, transportation, labor, government investment and public policy, because if the purpose is education for all, then, the responsibility must be everyone’s.

Structures de semiconducteurs II-VI à alignements de bande de type II pour le photovoltaïque / II-VI semiconductor heterostructures with type-II band alignments for photovoltaics

Ce travail porte sur l'étude d'hétérostructures de semiconducteurs II-VI à alignements de bande de type II, en particulier sous forme de superréseaux. Il s'agit d'un système qui peut être prometteur pour une application photovoltaïque, et c'est dans cette optique qu'est orienté ce travail. Une première partie traite ainsi d'une réflexion conceptuelle sur l'apport des interfaces de type II au photovoltaïque.Nous présentons ensuite une étude sur la croissance de CdSe et ZnTe par épitaxie par jets moléculaires, sur différents substrats. Ces matériaux sont particulièrement intéressants et adaptés pour cette application car ils ont un gap direct, quasiment le même paramètre de maille, un alignement de bandes de type II, et le CdSe une bande interdite compatible avec le spectre solaire. Mais en contrepartie il s'agit de semiconducteurs binaires qui n'ont aucun atome en commun, de sorte que la croissance d'échantillons avec des épaisseurs précises à la monocouche près constitue un vrai défi. Pour cette raison nous avons procédé à une étude fine des interfaces grâce à des analyses de diffraction de rayons X et de microscopie en transmission, qui nous permet de conclure sur la nature chimique des atomes à proximité des interfaces.Vient ensuite une étude poussée de spectroscopie sur les effets des interfaces de type II sur les porteurs de charges, à travers leur énergie et cinétique de recombinaison. Nous avons développé un modèle analytique qui permet d'ajuster précisément toutes les caractéristiques observées en relation avec ces interfaces, et qui témoigne d'un mécanisme de séparation des charges très efficace. Nous montrons par la suite que ces effets observés sont des caractéristiques intrinsèques de toutes les interfaces de type II, indépendamment des matériaux et des structures, et que ceux-ci nous permettent d'extraire avec précision les valeurs des décalages de bandes entre différents matériaux à alignement de type II. / This work focuses on the study of II-VI semiconductor heterostructures with type II band alignments, especially in the form of superlattices. This is a system that can be promising for photovoltaic applications, and my work is presented in this perspective. Thus the first part deals with a conceptual reflection on the contribution of type II interfaces for photovoltaics.In a second step I present a study on the growth of CdSe and ZnTe by molecular beam epitaxy on various substrates. These materials are particularly interesting and suitable for this application because they have a direct bandgap, are almost lattice-matched, present a type II band alignment, and CdSe shows a bandgap compatible with the solar spectrum. But in return these are binary semiconductors which have no atoms in common, so that the growth of samples with specific thicknesses close to the monolayer is challenging. For this reason we conducted a detailed study at the interfaces through analysis of X-ray diffraction and transmission electron microscopy, which allows us to conclude on the chemical nature of the atoms near the interfaces.This is followed by a detailed spectroscopy study on the effects of type II interfaces on the charge carriers through their energy and kinetics of recombination. We have developed an analytical model that allows to precisely adjust all the features observed in relation to these interfaces, and shows a very efficient charge separation mechanism. We show later that these effects are inherent characteristics of all interfaces of type II, regardless of materials and structures, and that they allow us to accurately extract the values of band offsets between different materials with type II band alignments.

Fundação de Amparo a Pesquisa do Estado de Minas Gerais / This work was to study the influence of the magnetic field on the reduction of hexavalent chromium, removal of COD and total chromium, addition of cell growth in batch (batch reactor), using a mixed culture from located in a tanning city of Franca-SP (AMCOA). The influence of static magnetic field generated by a device that produces an electric current through a conductor, producing a magnetic field around the conductor, like a magnet. This equipment produces a magnetic field pulsed square wave of constant intensity and frequency. In the first step of adjusting the mixed cultures of chromium VI obtained a promising response. Thereafter, a preliminary test was performed to analyze the behavior of the influence of the magnetic field in the reduction of chromium VI and diameter of the reactor, obtaining 71% reduction of chromium VI with a larger diameter, without the presence of magnetic field has seen a reduction of 53%, with an initial concentration of 100 mg/l of chromium and a time of 4 hours of process. In other stages of the project, performing the tests with different frequencies of the magnetic field (1,2,5 and 10 Hz) and doing an analysis related to cell death of micro-organisms, showed that for frequencies of 10 Hz, there was cell death presents some cultures, which was confirmed in a lower frequency. The results were satisfactory when used in a magnetic field frequency of 5 Hz, maximal reduction of chromium VI reducing the time about 4 hours of processing, better removal of COD and total chromium using a volume of 500 ml and concentration of 100 mg/l initial chromium. Checking the influence of different frequency magnetic field, the initial effluent volumes (300 and 1000 ml) and initial concentration of chromium VI (50, 150 and 200 mg/l). The higher the initial concentration the lower the chromium reduction of chromium VI, larger volumes and a reduction of chromium VI and removal of COD and total chromium is not as significant as the use of smaller volumes. / Neste trabalho foi realizado o estudo da influência do campo magnético sobre a redução de cromo hexavalente, remoção de cromo total e DQO, além do crescimento celular em modo descontínuo (reator batelada), utilizando-se de uma cultura mista proveniente do curtume AMCOA localizado na cidade de Franca-SP. Foi avaliada a influência de campo eletromagnético estático gerado por um equipamento que produz uma corrente elétrica, passando por um condutor, produzindo um campo magnético ao redor do condutor, como se fosse um ímã. Este equipamento produz um campo magnético pulsante com onda quadrática de intensidade e frequência constantes. Na primeira etapa de adaptação das culturas mistas ao cromo VI obteve-se uma resposta promissora. Posteriormente, foi realizado um teste preliminar para analisar o comportamento da influência do campo magnético na redução de cromo VI e diâmetro de reator, obtendo 71% de redução de cromo VI com um maior diâmetro, sem a presença de campo magnético obteve uma redução de 53%, com uma concentração inicial de 100 mg/L de cromo e com um tempo de 4 horas de processo. Nas outras etapas do projeto, realizando os testes com diferentes frequências de campo magnético (1, 2, 5 e 10 Hz) e fazendo uma análise relacionada à morte celular dos micro-organismos, mostrou-se que para frequências de 10 Hz, havia morte celular de algumas culturas presentes, o que não foi verificado em uma frequência menor. Os resultados foram satisfatórios quando se utiliza uma frequência de campo magnético de 5 Hz, com máxima redução de cromo VI reduzindo o tempo em torno de 4 horas de processo, com melhor remoção de cromo total e DQO, utilizando um volume de 500 mL e concentração de 100 mg/L de cromo inicial. Verificando a influência de diferentes frequências de campo magnético, volumes iniciais de efluente (300 e 1000 mL) e concentrações iniciais de cromo VI (50, 150 e 200 mg/L). Quanto maior a concentração inicial de cromo menor foi a redução de cromo VI, e em volumes de efluentes maiores a redução de cromo VI e remoção de cromo total e DQO não foram tão significativas quanto em se aplicar volumes menores. / Mestre em Engenharia Química

Distribuição e conservação dos genes que codificam as proteínas VgrG e Hcp em espécies de Aeromonas / Distribution and conservation of genes that encode protins HCP and verg in Aeromonas species

Aeromonas spp. são bastonetes Gram negativos amplamente distribuídos nos ambientes aquáticos, com relatos de isolamento em água de abastecimento público e alimentos. Este micro-organismo possui potencial de causar doenças intestinais e extraintestinais cuja patogenicidade está associada a sua virulência multifatorial. Diversos determinantes de virulência de Aeromonas já foram identificados, incluindo sistemas de secreção de proteínas. O sistema de secreção tipo VI (SST6) é o mais recente sistema de secreção de proteínas identificado em bactérias cuja presença em estirpes no gênero Aeromonas pode implicar atividades de citotoxicidade para o hospedeiro, pois esse sistema é capaz de injetar moléculas efetoras dentro da célula, interferindo diretamente nos processos celulares. A fim de determinar a presença e analisar a distribuição dos genes hcp e vgrG codificadores das proteínas efetoras do SST6 em Aeromonas spp. o presente estudo examinou 119 cepas isoladas de diversas origens pela técnica da PCR após o desenho de oligonucleotídeos iniciadores específicos. Objetivamos ainda analisar a variabilidade genética interespecífica dos genes hcp e vgrG a partir de dados de sequenciamento. Os resultados obtidos indicaram a distribuição dos genes vgrG e hcp em 46% das cepas de Aeromonas hydrophila e Aeromonas caviae de diferentes origens. Entre as cepas de A. hydrophila a maior frequência foi observada nas cepas isoladas de humanos, onde todas foram positivas para os iniciadores que amplificaram um produto de 541 pb do gene vgrG e 418 pb do gene hcp. Entre as cepas de A. caviae, a incidência de genes vgrG e hcp foi mais elevada nas cepas isoladas de alface (60%) e peixes (50%). As cepas analisadas de origem ambiental apresentaram índice total de 36% de positividade, apresentando frequência de 60% e 22% em A. hydrophila e A. caviae, respectivamente. Os dados obtidos da análise de cepas de origem alimentar mostraram a presença dos genes vgrG e hcp em 67% (A. hydrophila) e 60% (A. caviae) das cepas isoladas de folhas de alface. Nas cepas isoladas de queijo os genes foram encontrados em 67% e 12,5% das cepas de A. hydrophila e de A. caviae, respectivamente. O alinhamento múltiplo entre as sequências dos segmentos dos genes hcp e vgrG obtidas no sequenciamento indicou grau de identidade nucleotídica de 75 a 100% entre as sequências de hcp e 80 a 100% entre as sequências de vgrG. Em conclusão, nossos resultados indicaram que os iniciadores desenhados foram capazes de detectar suas sequências alvo em cepas de A. caviae e outras espécies de Aeromonas, sugerindo a existência de homologia entre os genes nas diferentes espécies, confirmada após sequenciamento de DNA. Os dados indicaram que esses genes estão distribuídos em várias espécies de Aeromonas e em cepas isoladas de diversas fontes. Ressaltamos a prevalência de cepas de A. hydrophila PCR-positivas em isolados clínicos, sugerindo a participação do SST6 no complexo universo da virulência multifatorial que permeia esse micro-organismo / Aeromonas species are Gram negative bacilli distributed widely in aquatic environments, with reports of isolation of this microorganism in water for public supply and food. Aeromonas have the potential to cause intestinal and extra intestinal infections whose pathogenicity is associated with multifactorial virulence. A number of virulence determinants have already been identified in Aeromonas, including protein secretion systems. The type VI secretion system (T6SS) is the most recent pathway to secrete proteins identified in bacteria. The presence of T6SS in Aeromonas strains may involve activities of cytotoxicity to the host, since this system is capable of injecting effectors molecules into the cell, interfering directly with a variety of cellular processes. The present study examined 119 strains of different origins by PCR, after the design of specific primers, to determine the distribution of vgrG and hcp genes encoding the effector proteins of T6SS in Aeromonas spp. We aimed to further analyze the interspecific sequence variation of hcp and vgrG genes based on sequencing data. The results show the presence of hcp and vgrG genes in 46% of A. hydrophila and A. caviae strains from different sources. All A. hydrophila strains isolated from humans were positive for the primers used to amplify a product of 541 bp and 418 bp of vgrG and hcp genes, respectively. Among A. caviae strains, the incidence of hcp and vgrG genes was high in the strains isolated from lettuce (60%) and fish (50%). The overall PCR-positive rate of strains from environmental source was 36%, with a frequency of 60% and 22% in A. hydrophila and A. caviae, respectively. The data obtained from analysis of food-borne strains showed the presence of hcp and vgrG genes in 67% (A. hydrophila) and 60% (A. caviae) of strains isolated from lettuce, while in the strains isolated from cheese the frequency was 67% (A. hydrophila) and 12.5% (A. caviae). The multiple alignment of hcp and vgrG sequences obtained revealed nucleotide identity rate between 75-100% among the hcp sequences and 80-100% in vgrG sequences. In conclusion, our results indicate that the primers designed were able to detect their target sequences in strains of A. caviae and other Aeromonas species, suggesting the existence of homology between genes in different species, as confirmed after DNA sequencing. The data indicate that these genes are distributed in various Aeromonas species from different sources. We emphasize the prevalence of PCR-positive A. hydrophila strains in clinical samples suggesting the involvement of T6SS in the complex universe of multifactorial virulence, which permeates this microorganism

The MA thesis briefly describes the history of the Vatican Council II and analyses the reaction of Czechoslovakian press to the Vatican Council II. The theoretical part aims at outlining the history, the course, the results and the importance of the Vatican Council II for the Catholic Church and also for the whole world. The theoretical opening is then reflected in the practical part of the MA thesis. It helps the reader to understand the issue. The theoretical opening pays attention to some important life moments of Pope John XXIII., ``the Council Maker{\crqq}. It focuses on the council idea, the definition of the council goals and setup time work before the council. It follows clarification of some organization issues during the council. It mentions its rule of procedure description. Another part of the theoretical part contains the development of all the council meetings and their results. There is also a list of council documents and evaluation of the Vatican Council II´s importance at the end of the theoretical part. The practical part of the MA thesis contains analysis of newspaper articles, namely from the Rudé právo and the Lidová demokracie dailies, covering the Vatican Council II. The monitored issues were: the space dedicated by the above dailies to the Council, the richness, content variety and balance of the coverage. The aim of the practical part and the MA thesis as a whole was the content analysis of the two 1960s daily newspapers and their reaction to the development and the result of the Council. It also contains content analysis comparison of these two newspapers and also identification of the extent of difference in objectivity and the ideological pressure on the news.
