Resumo:A leishmaniose visceral (LV) é uma doença crônica que pode ser fatal para os humanos. A doença é causada por um parasita intracelular, Leishmania infantum e é transmitida pela picada do mosquito flebótomo. Em cães com LV observa-se uma intensa reação inflamatória crônica no fígado, baço, pele, medula óssea e gânglios linfáticos. A atividade arginase é importante na LV, pois o aumento dessa enzima pode contribuir para a multiplicação do parasita e para a redução da síntese de óxido nítrico (NO), predispondo o macrófago à infecção. A prostaglandina E2 (PGE2) pode regular a produção do fator de necrose tumoral alfa (TNF-α) e da interleucina-10 (IL-10), porém nenhum estudo ainda foi realizado em cães com LV. O presente estudo teve como objetivo avaliar a atividade arginase em macrófagos aderentes de cultura de linfonodos de 23 cães naturalmente infectados e 18 saudáveis, e os níveis de NO e PGE2 no sobrenadante dessas culturas. O efeito regulatório de PGE2 na produção de TNF-α e IL-10 também foi avaliado em cultura de leucócitos totais de linfonodo. Esse estudo ajuda a esclarecer o mecanismo da resposta imunológica na LVC. / Abstract:Visceral leishmaniasis (VL) is a chronic disease that can be fatal to humans and dogs. The disease is caused by the intracellular parasite Leishmania infantum and is transmitted by the bite of the sandfly (phlebotomines). In dogs, VL is observed as an intense chronic inflammatory reaction in the liver, spleen, skin, bone marrow and lymph nodes. Arginase activity is important in VL because an increase of this enzyme may contribute to the multiplication of the parasite and a reduction of nitric oxide (NO) synthesis, predisposing a macrophage to infection. Prostaglandin E2 (PGE2) can play a regulatory role in the production of tumor necrosis factor (TNF-α) and interleukin-10 (IL-10), however, there have been no studies in dogs with LV. This study aimed to evaluate the arginase activity in adherent macrophages cultivated from the lymph nodes of 18 healthy and 23 naturally infected dogs and to examine levels of NO and PGE2 in the supernatant of these cultures. The regulatory effect of PGE2 on the production of TNF-α and IL-10 was also evaluated in supernatants of total lymph node leukocytes cultures. Previous issue date: 2014-10-10 / A leishmaniose visceral (LV) ? end?mica no Brasil, sendo a regi?o nordeste a que apresenta maior incid?ncia, apesar de nos ?ltimos 30 anos ter aumentado o n?mero de casos em outras regi?es do pa?s. A LV na Am?rica Latina ? resultante da infec??o por Leishmania infantum. No entanto, nem todas as pessoas infectadas desenvolvem doen?a; na verdade, a maioria apresenta resolu??o espont?nea da infec??o sem sintomas caracter?sticos de LV. Tradicionalmente a avalia??o do perfil imunol?gico tem sido realizada utilizando a reestimula??o de c?lulas mononucleares de sangue perif?rico em cultura. Esses estudos revelaram que pacientes com LV apresentavam inibi??o tanto da prolifera??o linfocit?ria quanto da resposta pr?-inflamat?ria anti-Leishmania. O presente trabalho teve por objetivo avaliar a resposta imune na LV sintom?tica, na cura p?s-tratamento e assintom?tica. Para isso, analisamos caracter?sticas imunofenot?picas relacionadas ? ativa??o, Treg e mem?ria de linf?citos, por citometria de fluxo, assim como a produ??o de citocinas ex vivo ou em cultura de sangue total. Para os volunt?rios com LV, foi realizado um estudo longitudinal com reavalia??o imunol?gicas aos 4 e 14 meses ap?s a cura cl?nica. O grupo controle incluiu pessoas provenientes de regi?o end?mica, sendo dividido em 2 grupos: Controle Positivo, formado por pessoas que apresentaram sorologia e?ou PCR anti-Leishmania positivos e Controle Negativo formado por pessoas com sorologia e PCR anti-Leishmania negativos. Durante a LV os linf?citos CD4 apresentam um maior perfil de ativa??o e mem?ria, al?m de serem maiores produtores de citocinas em cultura, quando comparado aos linf?citos CD8, contudo essa ativa??o n?o ? Leishmania espec?fica, visto que ocorreu tanto em aus?ncia (CD4+CD25+:10,60%, CD8+CD25+:5,36%; p<0,0001) quanto em presen?a de ant?genos sol?veis de Leishmania (SLA), (CD4+CD25+: 10,20%, CD8+CD25+:3,28%; p=0,0003). H? ativa??o de linf?citos durante a LV (CD4+CD69+: 4,9%), quando comparado aos grupos Controle Positivo (CD4+CD69+: 1,96% p=0,0045) e Negativo (CD4+CD69+: 1,35% p=0,0062), mas esta ativa??o tamb?m n?o ? Leishmania espec?fica. O perfil de ativa??o linfocit?ria permanece elevado mesmo 14 meses ap?s o fim do tratamento, por?m ap?s a cura a ativa??o ? Leishmania espec?fica (CD4+CD25+ em aus?ncia de SLA: 8,44%, presen?a de SLA: 10,70% p=0,0279). Linf?citos CD8+CD25+ foram capazes de produzir IFN-? em presen?a de ant?geno tanto em Controles Positivos (aus?ncia de SLA: 5,17%, presen?a de SLA: 9,52% p=0,0391) como em LV curados (Curado 4 meses: aus?ncia de SLA: 3,90%, presen?a SLA: 10,70% p=0,0098). C?lulas presentes no sangue total de pessoas com LV ativa s?o capazes de produzir IFN-? em resposta ao SLA (IFN-? em aus?ncia de SLA: 3,61 pg?mL e em presen?a de SLA: 44,26 pg?mL; p=0,0020), assim como LV recuperado (IFN-? em aus?ncia de SLA: 2,29 pg?mL e presen?a de SLA: 139,80 pg?mL; p=0,0005). Contudo o elevado n?vel de IL-10 parece estar inibindo a atividade pro-inflamat?ria de IFN-? e TNF-? em pacientes na fase sintom?tica. Contrariamente as demais citocinas pro-inflamat?rias, a cultura de sangue total do grupo LV ativa n?o apresentou produ??o de IL-2 Leishmania espec?fica (em aus?ncia de SLA: 2,42 pg?mL e em presen?a de SLA: 2,56 pg?mL). parasita. / Visceral leishmaniasis (VL) is endemic in many countries, including Brazil. The
protozoan Leishmania infantum, is the etiological agent of VL, and is transmitted
by the bite of female sandflies during the blood meal. The majority of subjects
when exposed to the parasite do not develop the disease, because of
development of Th1 cellular responses. Those who have develop signs of VL
such as fever, weight loss, hepatosplenomegaly, have impairment of the cellular
immune response, specific to the Leishmania antigens. We evaluated whether
the specififc anergy during symptomatic VL, may be associated with changes in T
cells costimulatory molecules or their ligands in CD14+ monocytes. There is an
increase in CTLA-4 porcentage on CD4+ T lymphocytes (p=0.001) and ICOS on
CD4+ and CD8+ T cells (p=0.002 to CD4+ and p=0.003 to CD8+), after
stimulation by soluble Leishmania antigen (SLA) during active visceral
leishmaniasis, and that there is a higher percentage of these molecules ex vivo,
when comparing symptomatic to recovered individuals (p=0.04 to CTLA-4 in
CD4+, and p=0.001 to ICOS in CD4+ and p=0.026 to CD8+). Moreover, we
found a high gene expression of CTLA-4, OX-40 and ICOS during active VL.
CD40, CD80, CD86, HLA-DR and ICOSL molecules do not suffer changes during
disease. There is IFN-? production by the peripheral blood cells, after SLA
stimulation, by peripheral blood cells in symptomatic subjects; however, there is a
decrease of the ratio IFN-?/IL-10, which is reversed after clinical recovery. The
impairment of some costimulatory molecules pathways during symptomatic VL
could inhibit the ability of phagocytes to kill Leishmania and could facilitate their
survival and the proliferation inside macrophages.

Fatores que interferem na sensibilidade do teste parasitológico no diagnóstico de leishmaniose visceral canina / Influence factories the sensitivity parasitological diagnosis of canine leishmaniasis visceralSousa, Marcus Vinícius Caetano de 18 December 2012 (has links)
This study aimed to investigate the sensitivity of parasitological testing when using aspirates or imprints of spleen, lymph nodes and bone marrow in function of time and correlate the parasite load of dogs with clinical symptoms. We used 31 seropositive dogs that were euthanized from the CCZ of Uberlândia. In the UFU s Laboratory of Pathology, the blades were fabricated and evaluated under a light microscope, 100X objective. The slides were evaluated for at most one hour and, if it was positive, the time spent for diagnosis in one, five, 20, 30 and 60 minutes were recorded. In the determination of the parasite load, the amastigotes were displayed and grouped in one, 10, 50, 100, and 500, 1000 and even 1500 views in 100 fields. Among the 31 animals analyzed, 11 (35.48%) were symptomatic, 17 (54.83%) oligosymptomatic and three (9.67%) asymptomatic. In the analysis by PAAF, difference was read of 20 minutes with one minute (p = 0.001), and the test sensitivity with time was greater at 60 minutes than at 20 minutes (p = 0.04). In the analysis of imprint, the test sensitivity was higher with time at 20 minutes than at five (p = 0.03). There was no statistical difference between the two techniques used in collecting samples for each tissue (bone marrow p = 1.0, p = 0.76 lymph nodes, spleen p = 0.57). There was no difference between tissues in the sensitivity of diagnoses made by PAAF, as well as imprint. The worm burden of both techniques demonstrated that asymptomatic animals have lower parasite load than in relation to symptomatic. / O presente estudo objetivou verificar a sensibilidade do teste parasitológico quando se utilizou aspirados ou imprints de baço, linfonodos e medula óssea, em função do tempo, além de correlacionar o resultado, a carga parasitária dos cães com a sintomatologia clínica. Foram utilizados 31 cães sororreagentes e eutanasiados provenientes do CCZ de Uberlândia. No Laboratório de Patologia da UFU, foram confeccionadas as lâminas e avaliadas em microscópio óptico, objetiva 100X. As lâminas eram avaliadas por no máximo uma hora e quando positiva registrou-se o tempo dispensado para o diagnóstico em um, cinco, 20, 30 e 60 minutos. Na determinação da carga parasitária as formas amastigotas eram visualizadas e agrupadas em uma, 10, 50, 100, 500,1000 e até 1500 visualizações em 100 campos. Dos 31 animais analisados 11 (35,48%) eram sintomáticos, 17 (54,83%) oligossintomáticos e três (9,67%) assintomáticos. Na análise por PAAF houve diferença de leitura de 20 minutos com um minuto (p= 0,001), assim como a sensibilidade do teste foi maior com o tempo de 60 minutos do que com 20 minutos (p=0,04). Na análise de imprint a sensibilidade do teste foi maior com o tempo de 20 minutos do que com cinco (p=0,03). Não houve diferença estatística entre as duas técnicas aplicadas na coleta de amostras para cada tecido (medula óssea p= 1,0; linfonodo p=0,76; baço p= 0,57). Não houve diferença na sensibilidade dos diagnósticos realizados por PAAF entre os tecidos, assim como por imprint. A carga parasitária de ambas as técnicas demonstraram que os animais assintomáticos possuem menor carga parasitária do que em relação aos sintomáticos. / Mestre em Ciências Veterinárias

Associações entre qualidade muscular, adiposidade, saúde vascular e funcionalidade de indivíduos obesosRech, Anderson January 2014 (has links)
Os níveis de obesidade vêm aumentando, no Brasil e no mundo, de maneira exponencial. A condição de excesso de tecido adiposo pode gerar um prejuízo funcional, bem como alterações em uma série de parâmetros de saúde cardiovascular. A quantidade e qualidade muscular apresentada por um indivíduo obeso podem ser determinantes na maneira como o organismo lida com esses desafios. Dessa forma, o objetivo do presente estudo foi verificar a existência de associação entre variáveis de força, qualidade e quantidade muscular, funcionalidade, saúde vascular e adiposidade em indivíduos obesos. Para isso, trinta e quatro indivíduos obesos, com idade entre 20-60 anos, sedentários, não fumantes e não diabéticos, se voluntariaram a participar do estudo. Após prévia familiarização, foram realizados testes de força nos extensores de joelho (contração isométrica voluntária máxima - CIVM ; 1 repetição máxima - 1RM; potência média) e funcionalidade (teste de caminhada de 6 minutos - TC6M; sentar e levantar em 30 segundos - SL30; get up and go). A massa muscular (EMQUA) e echo intensity do quadríceps femoral (EIQUA) foram medidas por ultrassonografia, bem como diferentes parâmetros de adiposidade e função endotelial (avaliada por dilatação mediada por fluxo - DMF). Foram verificados os coeficientes de correlação de pearson entre as variáveis acima citadas e adotou-se um p<0.05 para associação significativa. Foi encontrada uma associação negativa significativa entre a EIQUA e todos os testes de força analisados, bem como com os testes funcionais. A EMQUA também apresentou correlação significativa, porém positiva, com os testes de força e funcionalidade. A EIQUA apresentou uma associação significativa com a gordura visceral, bem como com a gordura subcutânea da coxa. Além disso, a gordura subcutânea abdominal apresentou associação significativa com a DMF. Os dados do presente estudo mostraram haver uma associação significativa entre a quantidade e qualidade muscular e o desempenho em testes de força e funcionais em indivíduos obesos. Ainda, importantes parâmetros de adiposidade se correlacionaram com a EIQUA e com a DMF, um importante marcador de saúde endotelial. Dessa forma, estartégias de intervenção (por exemplo, treinamento de força), que sejam capazes de modificar os parâmetros acima descritos, devem ser priorizadas quando o objetivo for uma melhora na qualidade de vida de indivíduos obesos. / Obesity levels are increasing in Brazil and all over the world in an exponential way. The adipose tissue excess may affect functionality, as well as another health parameters. The skeletal muscle quantity and quality in obese individual may be determinant in how that the organism deals with theses problems. Thus, the objective of the present study was to verify the association between strength, muscle quantity and quality, functionality, vascular health and adiposity in obese subjects. Thirty four obese subjects, aged 20-60 years, sedentary, non-smokers and non-diabetic, volunteered to participate in the study. After a familiartization session, the knee extensors strength (isometric maximal volunatry contraction - IMVC; on repetition maximum - 1RM ; average power) and functional (6 minutes walk test - TC6M; sit to stand up test - 30SL; get up and go) tests were performed. The quadriceps femoris muscle thickness (EMQUA) and echo intensity (EIQUA) were evaluated by ultrasonography, as well as different adiposty parameters and vascular function. The pearson correlation coefficent were verified between all variables cited above and a P<0.05 were adopted. There was found a negative significant association between the EIQUA and all the strength tests performed, as well as all functional tests. The EMQUA presented significant positive association with all strength and functional tests. Additionally, the EIQUA presented a significant negative association with the visceral adipose tissue, as well as the thigh subcutaneous adipose tissue. Besides that, the abdominal subcutaneous adipose tissue was associated positively with the vascular function. The data presented here have shown a significant association between the muscle quantity and quality and the performance in functional and strength tests in obese subjects. In addition, important adiposity parameters correlated with the EIQUA and vascular function. Thus, intervention strategies (such as strength training), that are capable of modifying these parameters, should be prioritized when the objective is to enhance the quality of life in an obese person.

Efeito do tratamento da apneia do sono com CPAP sobre a circunferência da cinturaSimões, Débora Bohrer January 2013 (has links)
Objetivo: Verificar o efeito em longo prazo do tratamento da apneia do sono com CPAP na circunferência da cintura em pacientes com apneia do sono. Métodos: Foram incluídos indivíduos com apneia do sono de ambos os sexos, idade superior a 18 anos, com indicação de uso de CPAP que aderiram ou não ao tratamento. Todos foram submetidos à polissonografia de noite inteira pelo método convencional em laboratório do sono afiliado a universidade entre 01/01/2007 e 01/06/2012. O grupo com CPAP realizou pelo menos três polissonografias. O grupo controle foi composto por pacientes que repetiram a polissonografia após pelo menos seis meses, que relataram não receber qualquer tipo de tratamento e que mantinham o IAH na segunda polissonografia. Resultados: No grupo CPAP foram incluídos 77 pacientes e no grupo controle, 43 pacientes. O tempo de acompanhamento foi respectivamente de 2,5 e 2,3 anos. Foi observada uma redução significativa na circunferência abdominal no grupo CPAP, comparando com controles através do teste t de Student (P= 0,024). O modelo logístico binário para prever redução da circunferência abdominal após o período de acompanhamento foi significativo (P <0,001), utilizando-se como regressores: uso do CPAP, sexo, idade> 45 anos, IMC≥ 30 kg/m2 e IAH≥ 30. O R2 de Nagelkerke foi de 0,36, indicando que essas variáveis explicam 36% da variação da redução da circunferência abdominal. Conclusão: O uso de CPAP se associa à redução da circunferência abdominal, independentemente de sexo, idade e índice de apneia-hipopneia basais, bem como da diferença de índice de massa corporal. Este achado justifica pesquisas futura sobre o papel da apneia do sono no volume de gordura visceral. / Objective: Verify the long term effect of sleep apnea treatment with CPAP on waist circumference in patients with sleep apnea. Methods: Individuals of both genders, older than 18 years that adhered to CPAP use or not were included. All underwent a full-night polysomnography by the conventional method in a university-affiliated sleep laboratory between 01/01/2007 to 01/06/2012. The group using CPAP underwent at least three polysomnographies. The control group consisted of patients with moderate to severe OSA who repeated a polysomnography after at least six months. Only individuals who reported not receiving any treatment and in which the AHI continued in the same OSA categories at the second polysomnography were included. Results: The CPAP group included 77 and the control group, 43 patients. Follow up time was, respectively, 2.5 and 2.3 years. A significant reduction in waist circumference was observed in the CPAP group, comparing with controls by Student’s t test (P= 0.024). The binary logistic model to predict decreased waist circumference after follow up period was significant (P<0.001), using as regressors: CPAP use, gender, age>45 years, BMI≥30 Kg/m2, and AHI≥30. The Nagelkerke R Square was 0.36, indicating that these variable explain 36% of the variance of waist circumference reduction. Conclusion: Long-term CPAP therapy is associated with a reduction in waist circumference, regardless of sex, age and basal apnea-hypopnea as well as the difference in body mass index.

Progressivamente, tem sido observada a urbanização da doença, associada ao processo migratório para as cidades de médio e grande porte, sendo evidente a concentração dos casos nesses centros urbanos. A letalidade pela LV nos últimos anos vem aumentando gradativamente, principalmente em pessoas vivendo com HIV/AIDS. . Nesses pacientes, a LV apresenta pior prognóstico caracterizado por recidivas frequentes e maior letalidade. Ao mesmo tempo, o número de relatos de reativação de LV em pessoas submetidas à imunossupressão para fins terapêuticos tem aumentado. O presente estudo descritivo transversal foi desenvolvido em amostra de conveniência com o objetivo principal de estimar a prevalência da infecção assintomática por Leishmania spp. e pacientes imunodeficientes do Hospital Universitário de Brasília. Foram avaliados 146 pacientes divididos em três grupos: pessoas vivendo com HIV/AIDS (48 participantes); imunossuprimidospelo tratamento de doenças reumáticas (47 participantes) e; pacientes submetidos a transplante renal (51 participantes). Amostras de sangue obtidas dos participantes foram submetidas à detecção de DNA do parasito por meio da PCR- ITS1 e detecção de anticorpos específicos anti- Leishmania por meio dos testes de aglutinação direta DAT, imunofluorescência indireta IFAT e o teste imunocromatográfico rápido baseado no antígeno recombinante rK39. Foi encontrada uma positividade empelo menos um dos testes em 23 dos 146 pacientes, sendo estimada uma prevalência de 17,2% (IC95% 11,6 a 24,4). Vinte e dois participantes apresentaram positividade no teste ELISA-rK39 (15,0%); cinco (3,4%) participantes apresentaram positividade no DAT; um participante apresentou positividade no teste rápido ICT-rK39 (0,6%) e um participante (0,6%) teve resultado positivo no teste de imunofluorescência indireta. Nenhuma amostra teve resultado positivo na PCRITS1 e em todas as amostras foi possível amplificar o gene de actina humana. A concordância bruta entre os testes foi baixa e os resultados sugerem a necessidade de uso combinado de múltiplos testes para melhorar a capacidade de detecção da infecção assintomática por Leishmania spp. Na população com déficit de imunidade. / The propagation of visceral leishmaniasis (VL) has been described in many regions of Brazil. Gradually, the urbanization of the disease has been observed, associated to the migration process of people to cities of medium and large size, the concentration of many cases being evident in those urban centers. The lethality of VL in recent years has increased gradually, mostly in people with HIV/AIDS. In these patients, VL has a worse prognosis which is characterized by frequent relapses and increased mortality. At the same time, the number of reports of reactivation of VL in people submitted to immunosuppression for therapeutic purposes has increased. This cross-sectional study was conducted in a sample of convenience with the main objective of estimating the prevalence of asymptomatic infection with Leishmania spp. and immunodeficient patients treated at the University Hospital of Brasília. This study evaluated 146 patients divided into three groups: people living with HIV/AIDS (48 participants), those immunosuppressed by treatment of rheumatic diseases (47 participants), and patients undergoing kidney transplant (51 participants). Blood samples taken from the participants were subjected to the detection of parasite DNA by PCRITS1 and detection of specific anti-Leishmania antibodies through direct agglutination test (DAT), indirect immunofluorescence (IFAT), the rapid test based on rK39 antigen (ICT-rK39 test) and the immunoassay based on recombinant antigen rK39 (ELISA-rK39). Positivity was found in at least one of those tests in 23 of 146 patients, an estimated prevalence of 17.2% (IC 95% 11.6 a 24.4). Twenty-two participants tested positive on ELISA-rK39 (15,0%); five (3.4%) participants tested positive on DAT; one participant was positive on ICT-rK39 (0.6%) and one participant (0.6%) was positive on IFAT. No sample tested positive on PCR-ITS1 and in all samples it was possible to amplify the gene of human actin. The crude agreement between tests was low and the results suggest the need for combined use of multiple tests to improve the capacity to detect asymptomatic infection by Leishmania spp. in the immunodeficientpopulation.

Vitamin C is Not Related to Resting Fat Oxidation in Healthy, Non-Obese AdultsJanuary 2014 (has links)
abstract: ABSTRACT Vitamin C plays an important role in fatty acid metabolism because it is required for carnitine synthesis. Vitamin C has been shown to have an inverse relationship with weight and body fat percent in a number of studies. However, there has been limited research exploring the relationship between vitamin C status and fat oxidation. This cross-sectional study investigates the relationship between plasma vitamin C and fat oxidation in 69 participants and between plasma vitamin C and body fatness in 82 participants. Participants were measured for substrate utilization via indirect calorimetry while at rest and measured for body fatness via DEXA scan. Participants provided a single fasting blood draw for analysis of plasma vitamin C. Results did not show a significant association between vitamin C and fat oxidation while at rest, therefore the data do not support the hypothesis that vitamin C status affects fat oxidation in a resting state. Associações entre qualidade muscular, adiposidade, saúde vascular e funcionalidade de indivíduos obesosRech, Anderson January 2014 (has links)
Os níveis de obesidade vêm aumentando, no Brasil e no mundo, de maneira exponencial. A condição de excesso de tecido adiposo pode gerar um prejuízo funcional, bem como alterações em uma série de parâmetros de saúde cardiovascular. A quantidade e qualidade muscular apresentada por um indivíduo obeso podem ser determinantes na maneira como o organismo lida com esses desafios. Dessa forma, o objetivo do presente estudo foi verificar a existência de associação entre variáveis de força, qualidade e quantidade muscular, funcionalidade, saúde vascular e adiposidade em indivíduos obesos. Para isso, trinta e quatro indivíduos obesos, com idade entre 20-60 anos, sedentários, não fumantes e não diabéticos, se voluntariaram a participar do estudo. Após prévia familiarização, foram realizados testes de força nos extensores de joelho (contração isométrica voluntária máxima - CIVM ; 1 repetição máxima - 1RM; potência média) e funcionalidade (teste de caminhada de 6 minutos - TC6M; sentar e levantar em 30 segundos - SL30; get up and go). A massa muscular (EMQUA) e echo intensity do quadríceps femoral (EIQUA) foram medidas por ultrassonografia, bem como diferentes parâmetros de adiposidade e função endotelial (avaliada por dilatação mediada por fluxo - DMF). Foram verificados os coeficientes de correlação de pearson entre as variáveis acima citadas e adotou-se um p<0.05 para associação significativa. Foi encontrada uma associação negativa significativa entre a EIQUA e todos os testes de força analisados, bem como com os testes funcionais. A EMQUA também apresentou correlação significativa, porém positiva, com os testes de força e funcionalidade. A EIQUA apresentou uma associação significativa com a gordura visceral, bem como com a gordura subcutânea da coxa. Além disso, a gordura subcutânea abdominal apresentou associação significativa com a DMF. Os dados do presente estudo mostraram haver uma associação significativa entre a quantidade e qualidade muscular e o desempenho em testes de força e funcionais em indivíduos obesos. Ainda, importantes parâmetros de adiposidade se correlacionaram com a EIQUA e com a DMF, um importante marcador de saúde endotelial. Dessa forma, estartégias de intervenção (por exemplo, treinamento de força), que sejam capazes de modificar os parâmetros acima descritos, devem ser priorizadas quando o objetivo for uma melhora na qualidade de vida de indivíduos obesos. / Obesity levels are increasing in Brazil and all over the world in an exponential way. The adipose tissue excess may affect functionality, as well as another health parameters. The skeletal muscle quantity and quality in obese individual may be determinant in how that the organism deals with theses problems. Thus, the objective of the present study was to verify the association between strength, muscle quantity and quality, functionality, vascular health and adiposity in obese subjects. Thirty four obese subjects, aged 20-60 years, sedentary, non-smokers and non-diabetic, volunteered to participate in the study. After a familiartization session, the knee extensors strength (isometric maximal volunatry contraction - IMVC; on repetition maximum - 1RM ; average power) and functional (6 minutes walk test - TC6M; sit to stand up test - 30SL; get up and go) tests were performed. The quadriceps femoris muscle thickness (EMQUA) and echo intensity (EIQUA) were evaluated by ultrasonography, as well as different adiposty parameters and vascular function. The pearson correlation coefficent were verified between all variables cited above and a P<0.05 were adopted. There was found a negative significant association between the EIQUA and all the strength tests performed, as well as all functional tests. The EMQUA presented significant positive association with all strength and functional tests. Additionally, the EIQUA presented a significant negative association with the visceral adipose tissue, as well as the thigh subcutaneous adipose tissue. Besides that, the abdominal subcutaneous adipose tissue was associated positively with the vascular function. The data presented here have shown a significant association between the muscle quantity and quality and the performance in functional and strength tests in obese subjects. In addition, important adiposity parameters correlated with the EIQUA and vascular function. Thus, intervention strategies (such as strength training), that are capable of modifying these parameters, should be prioritized when the objective is to enhance the quality of life in an obese person.

Objetivo: Verificar o efeito em longo prazo do tratamento da apneia do sono com CPAP na circunferência da cintura em pacientes com apneia do sono. Métodos: Foram incluídos indivíduos com apneia do sono de ambos os sexos, idade superior a 18 anos, com indicação de uso de CPAP que aderiram ou não ao tratamento. Todos foram submetidos à polissonografia de noite inteira pelo método convencional em laboratório do sono afiliado a universidade entre 01/01/2007 e 01/06/2012. O grupo com CPAP realizou pelo menos três polissonografias. O grupo controle foi composto por pacientes que repetiram a polissonografia após pelo menos seis meses, que relataram não receber qualquer tipo de tratamento e que mantinham o IAH na segunda polissonografia.
