Cette thèse, une édition traduite et commentée de deux livres de l'Histoire romaine (livres 57 et 58), s'inscrit dans un programme d'édition critique complète et moderne de cette œuvre. Dans ce diptyque faisant suite au cycle augustéen des livres 51 à 56, Dion Cassius retrace, avec le recul d'un sénateur romain de l'époque sévérienne, le principat de Tibère et ses soubresauts, des mutineries des légions en Pannonie et en Germanie à la disparition subreptice de l'empereur, en passant par les morts tragiques des héritiers présomptifs Germanicus et Drusus et la trahison de Séjan. Particulièrement attentif aux questions institutionnelles, l'historien grec s'attache à montrer comment le successeur d'Auguste poursuit l'œuvre de fondation du Principat entreprise par ce dernier en même temps qu'il la pervertit et s'éloigne progressivement de l'idéal politique défini par Mécène au livre 52, en particulier dans les relations qu'il instaure avec les sénateurs. Combinant structure biographique et trame annalistique, approche chronologique et distorsions temporelles, le récit de Dion vient tantôt corroborer tantôt nuancer les témoignages antérieurs des écrivains latins Suétone et Tacite sur la personnalité de Tibère et les événements marquants de son principat. Bien que longtemps considérés par les historiens modernes comme une source d'appoint pour la connaissance de cette période, les livres 57 et 58 de l'Histoire romaine, malgré leur état partiellement lacunaire, apportent de surcroît un éclairage essentiel sur les circonstances entourant la chute de Séjan, pour lesquelles nous ne disposons pas du témoignage des Annales de Tacite. À côté de notre travail d'établissement du texte et de traduction, nous proposons donc un commentaire qui s'efforce de dégager, en trois temps consacrés successivement à la structure narrative des deux livres, à la construction d'une figure d'empereur à la fois singulière et archétypale, et, enfin, à la tonalité distanciée du récit, l'originalité stylistique et l'intérêt historique du point de vue développé par Dion Cassius sur la vie politique et les institutions romaines du Haut-Empire. / This thesis is a translated edition of Books 57 & 58 of Cassius Dio's Roman History, with a philological and historical commentary. The latter edition of Cassius Dio's work is now outdated, and so are the French translations based upon it. Our work constitutes a part of a larger programme that aims to provide an up-to-date edition with a translation of the complete works of Cassius Dio. In books 57 & 58, the Greek historian, living under the Severans, follows thoroughly the evolution of the Tiberian Principate, with a particular focus on the political crisis from military rebellions in Pannonia and Germania to Sejanus' conspiracy. His analyses are founded both on his literature searches and on his own political experience as a Roman senator, and reveal an accurate knowledge of the institutional realities of the Early Principate. The main goal of the two books is to show how Tiberius, as the successor of Augustus, completes the founding work of his great predecessor while debasing the political ideals defined by Maecenas in book 52. In this train of thought, Dio pays special attention to the relationships between the Emperor and the Senators and how they evolve. Combining biographical patterns with an annalistic framework, the narration provides an original point of view on the figure of Tiberius, beside the earlier testimonies of Suetonius and Tacitus which remain incomplete with regard to the fall of Sejanus. Accordingly, the present work focusses on three main areas, including first the narrative structure, then the profiling of Tiberius as a political leader in relation to other rulers, and finally the distanced and ironic view on the political and human comedy.

Rezonanční přenos energie v prostředí hydrogelové matrice / Resonance energy transfer in the environment of the hydrogel matrix

The diploma thesis deals with resonance energy transfer in hydrogel matrix enviroment. In the theoretical part, all the important features of these systems and their components are summarized and all the principles for explaining the method used are described. The work also includes a short literary background research. In the experimental part, the resonance energy transfer was measured in micellar solutions and hydrogels in 0.15 M NaCl medium. The hydrogels were formed on the basis of interaction of the surfactant with the oppositely charged polyelectrolyte. As a positively charged surfactant, carbethopendecinium bromide (Septonex) was chosen and hyaluronan as the negatively charged polyelectrolyte. Perylene with fluorescein and perylene were selected as fluorescent pairs for RET. It has been found that increasing concentration of the acceptor molecule increases the intensity of RET, in other word the rate of acceptor binding to the donor. In the study of fluorescent pair of perylene with DiO was observed, that it was not appropriate to use fluorescence probe DiO at higher concentrations. Experiments were carried out to determine whether RET changes in the time from the point of mixing the donor and acceptor fluorescence probes together. This phenomenon has not been confirmed. In hydrogels, the effect of increasing surfactant concentration and molecular weight of hyaluronan was observed. The resulting analysis showed that the molecular weight of hyaluronan has a significant effect on RET efficiency in hydrogels.

Identification of genetic loci associated with different responses to high-fat diet-induced obesity in C57BL/6N and C57BL/6J substrains

We have recently demonstrated that C57BL/6NTac and C57BL/6JRj substrains are significantly different in their response to high-fat diet-induced obesity (DIO). The C57BL/6JRj substrain seems to be protected from DIO and genetic differences between C57BL/6J and C57BL/6N substrains at 11 single nucleotide polymorphism (SNP) loci have been identified. To define genetic variants as well as differences in parameters of glucose homeostasis and insulin sensitivity between C57BL/6NTac and C57BL/6JRj substrains that may explain the different response to DIO, we analyzed 208 first backcross (BC1) hybrids of C57BL/6NTac and C57BL/6JRj [(C57BL/6NTac × C57BL/6JRj)F1 × C57BL/6NTac] mice. Body weight, epigonadal and subcutaneous fat mass, circulating leptin, as well as parameters of glucose metabolism were measured after 10 wk of high-fat diet (HFD). Genetic profiling of BC1 hybrids were performed using TaqMan SNP genotyping assays. Furthermore, to assess whether SNP polymorphisms could affect mRNA level, we carried out gene expression analysis in murine liver samples. Human subcutaneous adipose tissue was used to verify murine data of SNAP29. We identified four sex-specific variants that are associated with the extent of HFD-induced weight gain and fat depot mass. BC1 hybrids carrying the combination of risk or beneficial alleles exhibit the phenotypical extremes of the parental strains. Murine and human SC expression analysis revealed Snap29 as strongest candidate. Our data indicate an important role of these loci in responsiveness to HFD-induced obesity and suggest genes of the synaptic vesicle release system such as Snap29 being involved in the regulation of high-fat DIO.

Cassius Dio, human nature and the late Roman Republic

This thesis builds on recent scholarship on Dio’s φύσις model to argue that Dio’s view of the fall of the Republic can be explained in terms of his interest in the relationship between human nature and political constitution. Chapter One examines Dio’s thinking on Classical debates surrounding the issue of φύσις and is dedicated to a detailed discussion of the terms that are important to Dio’s understanding of Republican political life. The second chapter examines the relationship between φύσις and Roman theories of moral decline in the late Republic. Chapter Three examines the influence of Thucydides on Dio. Chapter Four examines Dio’s reliance on Classical theories of democracy and monarchy. These four chapters, grouped into two sections, show how he explains the downfall of the Republic in the face of human ambition. Section Three will be the first of two case studies, exploring the life of Cicero, one of the main protagonists in Dio’s history of the late Republic. In Chapter Five, I examine Dio’s account of Cicero’s career up to the civil war between Pompey and Caesar. Chapter Six explores Cicero’s role in politics in the immediate aftermath of Caesar’s death, first examining the amnesty speech and then the debate between Cicero and Calenus. Chapter Seven examines the dialogue between Cicero and Philiscus, found in Book 38. In Section Four is my other case study, Caesar. Chapter Eight discusses Caesar as a Republican politician. In Chapter Nine, I examine Dio’s version of the mutiny at Vesontio and Caesar’s speech. Chapter Ten examines Dio’s portrayal of Caesar after he becomes dictator and the speech he delivers to the senate. The Epilogue ties together the main conclusions of the thesis and examines how the ideas explored by Dio in his explanation of the fall of the Republic are resolved in his portrait of the reign of Augustus.

This thesis is a historical and historiographical commentary on Book 57 (Chs. 1-17.8) of Cassius Dio's Roman History. It comprises two sections, an Introduction followed by the Commentary itself. The introduction is sub-divided into three chapters. The first of these introductory chapters (The Roman Historian at Work) presents a discussion of the historical material available for Dio's Tiberian narrative, and a discussion of the factors which were instrumental in Dio's writing and shaping his narrative of the reign of Tiberius. The second chapter (Dio on Tiberius) is an analysis of Dio's portrayal of Tiberius and of the historian’s understanding of Tiberius in the historical context of the early Principate. These chapters are followed by some brief Notes on the Text of Book 57, which considers the manuscript tradition of Book 57, and comments on portrayal of the reign of Tiberius in the Dionian tradition, and in particular the Excerpta Constantiniana, Xiphilinus, and Zonaras. The second part of the thesis, the commentary, presents an analysis of Dio's narrative from both historical and historiographical perspectives.

Cette thèse, une édition traduite et commentée de deux livres de l’Histoire romaine (livres 52 et 53), s’insère dans un programme d’édition critique complète et moderne de cette œuvre. Dans ces livres, Cassius Dion, historien grec et sénateur romain sous les Sévères, s’appuie sur sa connaissance de la politique romaine pour décrire en termes institutionnels la transition entre République et Empire. Les questions institutionnelles l’intéressaient tout particulièrement et il avait accès à une documentation que négligent ou ignorent les sources parallèles : il nous livre souvent un point de vue inédit sur l’histoire de Rome. C’est particulièrement vrai dans les livres 52 et 53 : ces livres sont centrés sur le passage de la République au Principat et ils consacrent une large place à l’analyse de ce changement de régime et des mutations institutionnelles qu’il entraîne. À travers une série de discours probablement fictifs, placés dans la bouche des protagonistes de cette « révolution » (par exemple Auguste, Agrippa, Mécène), Dion met en place un vaste débat sur « le meilleur régime », qui puise à de multiples traditions : certaines sont bien connues et ont été étudiées, comme la tradition historiographique, qui commence avec Hérodote et que l’on retrouve ensuite dans les tragédies comme dans la philosophie. Mais la perspective historique et institutionnelle se double d’une perspective proprement littéraire : le travail de Dion se nourrit en effet aussi de deux traditions, celle du débat philosophique sur le meilleur régime politique et celle de la déclamation rhétorique. The last French edition of Cassius Dio’s Roman History is now obsolete. Moreover there is no modern translation of this text. For a few years an up-to-date edition, with a translation, has been in progress, which includes our work. In Books 52 and 53, the author, a Greek historian and a Severian senator, describes the evolution of the Roman institutions between Republic and Principate. His analyses are based on his own political knowledge, his official duties and on his study of his Greek predecessors (such as Polybius or Dionysius of Halicarnassus). His point of view is most of the time original. In books 52 and 53, he focuses on the monarchic transition and the changes that affected the constitution and the institutions. The most important actors of the “revolution” (Augustus, Agrippa and Maecenas) are delivering probably fictive speeches that illustrate this change. There is also in these books an interesting literary work. Dio is to be considered as an heir of two traditions: the debate over the best constitution (as old as Herodotus at least) and the rhetoric declamation. Both of them are at work in the speeches of books 52 and 53. The well-known debate between Maecenas and Agrippa (Book 52) has thoroughly been studied but rarely in connection with the other speeches of the Augustan books. Besides, the literary part of these texts has been, most of the time, used to criticize them. This work lays stress on two (in our opinion) linked questions: how valuable is the historical testimony of Cassius Dio on this key period? And what do the speeches bring (besides the rhetorical display) to Dio’s historiographical project?

A coleta de dados ocorreu atrav?s de um question?rio inicial de caracteriza??o do grupo, de dois testes (pr? e p?s-testes) em rela??o ao conte?do matem?tico estudado na Unidade de Aprendizagem, de registros das comunica??es on-line entre professor e discentes e das entrevistas com seis alunos que interagiram mais intensamente com o professor. A an?lise dos dados quantitativos e qualitativos permite concluir que houve um crescimento do desempenho dos alunos envolvidos. Quanto mais intensas s?o as intera??es professor-aluno, melhores s?o as condi??es para que as aprendizagens aconte?am. A pesquisa tamb?m mostra a import?ncia do uso de recursos de comunica??o da rede Internet pelo professor para apresenta??o de propostas de atividades, orienta??es e acompanhamento ao trabalho dos alunos no sentido de auxili?-los na reconstru??o de seus conhecimentos em atividade n?o-presencial.

Reconhecendo que tal vis?o fragmentada do conhecimento cient?fico ? nociva para a forma??o do esp?rito cient?fico do aluno, te?ricos t?m proposto a interdisciplinaridade como forma de atenuar essa fragmenta??o escolar, restabelecendo v?nculos entre as disciplinas. Como sujeitos ativos na educa??o, os professores s?o fundamentais para o sucesso desse processo, pois somente a partir da atua??o conjunta do grupo docente ser? poss?vel a implanta??o de uma interdisciplinaridade escolar capaz de superar as dificuldades da fragmenta??o. Para compreender a rela??o entre a atua??o docente e a fragmenta??o dos saberes, esta pesquisa realizou entrevistas com professores das disciplinas cient?ficas do Ensino M?dio, visando reconhecer neles a percep??o que t?m sobre a realidade da educa??o cient?fica escolar, buscando descobrir as causas, conseq??ncias e manifesta??es da fragmenta??o dos saberes neste n?vel de ensino. As entrevistas foram realizadas com seis professores de uma escola da rede particular de ensino da regi?o metropolitana de Porto Alegre. A partir de uma abordagem qualitativa, analisamos o material proveniente das entrevistas utilizando a An?lise Textual Discursiva. A pesquisa possibilitou a constata??o de que as concep??es que os professores t?m da sua disciplina e das ci?ncias em geral est?o intimamente ligadas ao modo como atuam em sala de aula. Pudemos perceber que os docentes reconhecem a necessidade de um trabalho interdisciplinar e apontam o di?logo entre si como fator determinante para que ocorra a interdisciplinaridade na escola. Entretanto, apesar de admitirem que ? preciso mudar o modo de agir, os entrevistados indicam a falta de tempo, a falta de incentivo da escola e a grande quantidade de conte?dos exigida pelo curr?culo como elementos que impedem a efetiva??o da educa??o interdisciplinar.
