Efeito do extrato de Azadirachta indica (nim) sobre resposta de hipersensibilidade mediada por ácido salicílico em células de Rubus fruticosus / Effect of Azadirachta indica extract (neem) on hypersensitivity response mediated by salicylic acid in cells of Rubus fruticosus.

Veronica Paviani 01 June 2010 (has links)
Plants, like other organisms, have the capacity to defend themselves against attack by pathogens. One of the responses triggered by pathogen recognition by plant cells is the hypersensitive response (HR), which involves the immediate death of cells in the primary site of infection, providing resistance to the pathogen growth. In this regard, it has been well established that mitochondria are involved in cell death. The neem tree (Azadirachta indica) is known due to its medicinal and insecticidal properties; studies on the insecticidal action of this plant had been restricted to the analysis of their action mechanisms on insects and their effects on rural workers who use neem-based products. However, its impact on plant systems has not been addressed. Considering previous results from our laboratory and literature data we assessed the effects of neem as elicitor, particularly the mechanisms leading to the phenomenon of plant resistance. The neem extract (EB) was prepared from the seeds, characterized biochemically by quantification of phenolic compounds, sugars and proteins. The extract showed strong dose-dependent antioxidant activity (IC50 of 14.85 mg/mL). EB concentrations of 0.1-5 mg/mL, alone or in association with 1 mol/L or 1 mmol/L SA (salicylic acid), were used for the biological assays. For cell death assays, EB was employed in concentrations of 0.1 and 5.0 mg/mL, alone or in association with 1 mol/L SA, during 0-8 hours. EB (0.1 mg/mL), alone or in association with 1 mol/L SA, induced Rubus fruticosus cell death more efficiently than EB alone or in association with 5 mg/mL SA. After 8 hours, a 64% of death of cells elicited with 0.1 mg/mL EB and 71% of death of cells elicited with 0.1 mg/mL EB in association with SA, was observed. Decrease in ROS generation and production of endogenous SA, as well as increased production of phenolic compounds, was observed in intact cells elicited with EB alone. However, when cells were elicited with EB in association with SA, a decreased production of phenolic compounds and an increased production of endogenous SA, was observed. In isolated mitochondria, it was measured oxygen consumption, membrane potential and ROS production for EB alone or in association with 1 mmol/L SA. In either conditions, EB decreased oxygen consumption by the respiratory chain, an effect more pronounced in association with SA: ~79 % inhibition for state 3 and ~ 62 % for state 4 respiration. Also, either neem alone or in association with SA decreased mitochondrial membrane potential, as well as ROS generation to an extent of 55-20% in the presence of antimycin A and 39-10% in the presence of rotenone; in association with SA, EB decreased ROS at 5, 1 and 0.5 mg/mL. Together with our previous study, these results suggest that neem seeds extract has a protective effect on Rubus fruticosus cells by scavenging, via phenolic compounds, reactive oxygen species generated by SA, thereby decreasing its action as cell death inducer.

Estudo do efeito de respostas de hipersensibilidade sobre a parede celular em cultura de células de amora-preta (Rubus fruticosus) / study of the effects of hypersensitive response on cell wall in blackberry-black cell culture (Rubus fruticosus)

Fernando Aparecido Mariano de Souza 23 February 2007 (has links)
Like the other organisms, plants have the ability to self-defend through recognition of pathogens (hypersensitive response - HR), causing immediate cell death at the primary infection site, thus offering resistance to their grown. The HR is characterized by necrosis of tissues in this site via many signals still not completely elucidated, like formation of free radicals including H2O2 and reinforcement of cell wall. The aim of this study was to establish the relationship between these signals in blackberry-black cell culture (Rubus fruticosus). The experimental conditions for analysis of cell wall, reactive oxygen species (ROS) and H2O2, were established. Acid polysaccharide (rhamnoglucuronogalactan, F-I), salicylic acid (SA), and methyl jasmonate (MeJA), well established effectors of the defense response, were used as elicitors. ROS and H2O2 production was activated by F-I and SA, followed by release of fragments like disaccharides from the cell wall, apparently due to its degradation. By contrast, a small production of ROS and H2O2 was observed in presence of MeJA, as well as an increase of high molecular weight fragments, that may function as signals for reinforcement of cell wall, enzyme induction and production of others defense molecules. Together, the two elicitors SA and MeJA inhibited the ROS production, caused by MeJA, while sustaining release of the extra cellular compounds of high molecular weight.

However, due to the need for mimicking the in vivo process, that consists in several interdependent steps there are difficulties in setting the optimal conditions for each situation performed. The improvement of in vitro maturation (IVM) protocols through supplementation with different molecules can increase the efficiency of the culture medium and improve the competence of oocytes for fertilization and embryogenesis. A promising approach is the realization of mediated delivery of nanocarriers molecules. Tretinoin (TTN, all- trans retinoic acid - ATRA ) is a natural retinoid and active metabolite of vitamin A with important action in cell proliferation and differentiation and embryonic development under both in vivo and in vitro conditions. TTN acts on the cytoplasmic maturation process, in the initial embryonic development and oocyte competence, improving the quality of embryos generated. The combinations of tretinoin with polymeric nanoparticles are alternatives to enhance the solubility and chemical stability of this molecule, allowing controlled release and decreased degradation. The objective of this study was to evaluate the effects of IVM medium supplementation with tretinoin-loaded lipid-core nanocapsules (TTN-LNC) at concentrations of 0.25, 0.5 and 1 μM, by analysis of embryonic development until the blastocyst stage, production of reactive oxygen species (ROS), and expression of genes related to apoptosis and pluripotency. As main results, TTN-LNC 0.25 μM increased the rate of blastocyst production and reduced ROS production. In addition, TTN and TTN-LNC induced a lower gene expression of Bax and SHC1, suggesting beneficial effects on the development of embryos. The results indicate that nanoencapsulation allows the use of a lower dose of TTN-LNC with consequent obtention of higher percentages of blastocyst production and decreased production of ROS, making nanoembriology a potential tool for improving bovine embryos IVP.

Edge effects: poetry, place, and spiritual practices

Bubel, Katharine 01 May 2018 (has links)
"Edge Effects: Poetry, Place, and Spiritual Practices” focusses on the intersection of the environmental and religious imaginations in the work of five West Coast poets: Robinson Jeffers, Theodore Roethke, Robert Hass, Denise Levertov, and Jan Zwicky. My research examines the selected poems for their reimagination of the sacred perceived through attachments to particular places. For these writers, poetry is a constitutive practice, part of a way of life that includes desire for wise participation in the more-than-human community. Taking into account the poets’ critical reflections and historical-cultural contexts, along with a range of critical and philosophical sources, the poetry is examined as a discursive spiritual exercise. It is seen as conjoined with other focal practices of place, notably meditative walking and attentive looking and listening under the influence of ecospiritual eros. My analysis attends to aesthetics of relinquishment, formal strategies employed to recognize and accept finitude and the non-anthropocentric nature of reality, along with the complementary aesthetics of affirmation, configuration of the goodness of the whole. I identify an orienting feature of West Coast place, particular to each poet, that recurs as a leitmotif for engagement of such aesthetics and related practices. In chapter one, I consider a group of Jeffers’s final poems as part of a project he designated “our De Natura,” attending especially to his affinity for stones and stars. In chapter two, I investigate both Roethke’s and Hass’s configurations of ecospiritual eros in accord with their fascination for flora, while in chapter three, I employ the concepts of “aura” and “resonance” to explicate Levertov’s meditations on the “coming and going” Mount Rainier-Tacoma and Zwicky’s reflective iterations of the sea. / Graduate / 2019-04-04

Santamaría Blasco, María Lourdes 27 October 2008 (has links)
(2008). Figuras del exceso y políticas del cuerpo. Ironic Multiplicity: Fernando's "Pessoas" Suspended in Kierkegaardian Irony

Hale, Michelle Pulsipher 19 March 2004 (has links) (PDF)
This thesis applies Søren Kierkegaard's understanding of irony as outlined in his master's thesis, The Concept of Irony, to the literary works of Fernando Pessoa. Recently Kierkegaardian scholarship has opened possibilities for non-traditional interpretation of Kierkegaard's dissertation and pseudonymous "aesthetic" texts by reading them in the ironic tone in which they were written. This paper offers a similar re-reading of the poetic and prose works Pessoa attributes to his heteronyms. Kierkegaard's presentation of Socrates as irony serves as a model for how Pessoa sustains the heteronymic project by balancing the use of rhetorical irony within the works of the heteronyms with simultaneous use of "Socratic" irony relating to both the heteronyms and their literary contributions. Pessoa "controls" irony by bringing his heteronyms into his historical reality whereby he posits subjectivities for them. The necessary element of eros as it is identified with Socrates and thereby with irony is defined negatively as the desire for that which one is lacking and is sustained by the distance inherent in desire. Irony-eros as desire is present in the works of each of Pessoa's poetic heteronyms, gains for them corporeality, and characterizes the relationship the reader has with those works. Pessoa, like Socrates, is unable to extend controlled irony to his personal life and remains in the negativity of desire. Bernardo Soares and O Livro do Desassossego challenge traditional notions of reality since Soares feels with equal intensity the reality of his actuality and that of his imagination. Kierkegaard holds that the imagination provides the thinker with various possibilities or ideals. The thinker must then actualize the ideal. Kierkegaard's pseudonyms offer possible life-views as do Pessoa's heteronyms. The distance of irony is essential, for in reflecting on the life-views, the reader must not be able to see the author in that reflection. Unlike Kierkegaard, Pessoa successfully distanced himself from his heteronyms by multiplying and deferring his identity. Adept in Socratic midwifery Pessoa establishes the subjectivity of other "Pessoas" through whom he offers his readers possibilities. Pessoa's ironic existence proves the self is indefinable and unassimilable to any System.

Teknikens dialektik : Ambivalenser och brott i Herbert Marcues teknikfilosofi / The Dialectics of Technology : Ambivalences and Ruptures in Herbert Marcuses' Philosophy of Technology

Hall, Mikael January 2022 (has links)
In this thesis ”The Dialectics of Technology: Ambivalences and Ruptures in Herbert Marcuses’ Philosophy of Technology” I present a thorough analysis of Herbert Marcuses’philosophy of technology as it developed from the late 1940s until his death in 1979. Whereas, previous studies have tried to reduce his philosophy of technology to one coherent project, I argue that his philosophy of technology is characterised by ambivalences and ruptures and therefore cannot be reduced to such a coherent totality. Marcuses’ philosophy of technology vacillates between what I, with Andrew Feenbergs’ concepts, callan instrumentalist and substantialist notion of technology. An instrumentalist notion is one where technology is understood merely as a tool to be wielded by external actors, in Marcuses’ case that of the capitalist class and the state. In contrast to this, Marcuse also precents a substantialist philosophy of technology, where it is imbued with its own agency and direction. The previous scholarship has largely reduced Marcuses’ philosophy to one of these positions, rather than emphasising how both of them are present through out the text. Furthermore, I argue that his substantialist understanding of technology itself is characterised by ruptures and ambivalences, where technology is viewed both as inherently emancipatory and as inherently subjugating. It is in regard to this duality I argue that his philosophy of technology should be understood as dialectical, in the sense that technology at the same time can be a central part of human subjection and one of the most important vehicles for emancipation. In relation to this, I agree with Samir Gandesha that one of the most fruitful directions Marcuses’ philosophy opens is an understanding of technology as bearer of a historical substance, that is to say that technology is imbued with certain goals, desires and world-views beyond those of the wielder, while at the same time understanding that this substance can be changed through historical processes. Beyond the more exegetic presentation of Marcuses’ philosophy and the critique against the previous scholarship, I argue for critical theory, and especially Marcuses’, relevance for current debates around technology and automation. In the current debates between left wing accelerationist cheerleaders of technology and those radicals more sceptical of technologies emancipatory potential, an investigation into the nature of technology itself is severely lacking. While, the former uncritically embraces the technology arisen in capitalist society and views it as a direct path towards a utopian post-scarcity communism, the latter solely focuses on technology as a tool of capitalist class power. In relation to this, I argue that the historical and dialectical understanding of technology that can be salvaged from Marcuses oeuvre would be a welcome and useful addition to the debate.

The nature of love a phenomenological approach

Schroeder, Samantha 01 May 2012 (has links)
As I hope to show, a philosophical study of love is highly relevant today, since the sciences have not adequately answered the perennial question: What is love?; Since the time of Socrates, the idea of love and the conception of the human heart have been devalued by thinkers who, by definition, are known as "lovers of wisdom." Considered pejoratively as "the passions," the subject of emotion was deemed inferior to thought centered upon the human faculty of reason. Many studies in the sciences, from biology to psychology, claim to have pointed us to the source of the human experience of love--but do they help us to understand love properly? In order to provide a full consideration of love in my philosophical research, I will focus my analysis on love under the philosophical lens of phenomenology. Known as the study of firsthand human experience, phenomenology became the influential school of thought for many German philosophers in the early twentieth century. My research will closely examine the writings of Max Scheler, Dietrich von Hildebrand, and Jean-Luc Marion within the context of this tradition. Moving from a justification of love in philosophy to the topic of self-love, I hope to define effectively what it means to love another. I shall also attempt to disambiguate the common assumptions regarding the nature of love. Is there a fundamental difference between the phenomenon of "falling in love" and of love itself? I question whether love, in its essence, is defined by the element of choice--of a willful emotional giving of oneself to another--and whether it can be distinguished from a passive feeling and an active loving will. I aim to bring the human affective sphere into the full light of philosophical inquiry, considering whether love is a moral act of the will that involves a total participation of the self--in mind, body, and spirit. Love is arguably the most powerful of the human emotions, one that elevates the human sphere of emotions and the ethical existence beyond simple desire.

Me by Your Name / Echoes of the Past : A Study of the Role and Thematization of Ancient Greek Eros in the Novel and Film Call Me by Your Name

Hall, Nina January 2019 (has links)

Pagán Cánovas, Cristóbal 25 June 2009 (has links)
This dissertation discusses Aristotle's approach to metaphor, Conceptual Metaphor Theory and Blending Theory, and proposes that the latter constitutes a change of paradigm. I use Blending Theory to model a conceptual pattern generalizing over several case studies of verbal figuration, in different periods of Greek love poetry: ancient Greek lyric, the emergence of the arrows of love in the archaic and classical periods, medieval folksongs, and two 20th century poets, Ritsos and Elytis. All these poetic images share a conceptual pattern that can be modelled with a generic integration network of EROTIC EMISSION. Variation in the realization of the pattern crucially relies on assigning the role of the emitter to the loved person or to an external agent, and on the instantiation of the EMISSION image schema, like light, wind, or an object. Culture and context impose further constraints that can be studied systematically.
