Effets des fluorures sur l'activité antimicrobienne des antibiotiques contre Staphylococcus aureus et Pseudomonas aeruginosa

L’émergence des souches bactériennes résistantes aux antibiotiques est un phénomène inquiétant, qui se répand à travers le monde. Staphylococcus aureus et Pseudomonas aeruginosa sont des bactéries pathogènes opportunistes multi résistantes qui peuvent causer plusieurs maladies. Cependant, ces bactéries deviennent difficiles à traiter avec des antibiotiques sans occasionner de toxicité. Alors pour trouver des solutions, c’est nécessaire de développer de nouvelles molécules afin de combattre les agents pathogènes résistants. Grâce à leur action pharmacologique, les fluorures exercent un certain effet antibactérien au niveau de l'émail des dents; donc, leur association aux antibiotiques pourrait bien a méliorer l’activité antimicrobienne. De ce fait, nous nous sommes proposés d’étudier les activités in vitro de la vancomycine (VAN), l’oxacilline (OXA), la ceftazidime (CFT) et la méropenème (MER) libre ou associée au fluorure de sodium (NaF) et fluorure de lithium (LiF) qui ont été évaluées sur des souches S.aureus et P.aeruginosa sensibles et résistantes, par la méthode de la microdilution en bouillon, déterminant leur concentration minimale inhibitrice (CMI), leur concentration minimale bactéricide (CMB), leur courbe cinétique (Time-Kill). Leur cytotoxicité sur les globules rouges humains, et leur stabilité à la température de 4°C et 22°C ont été étudiées. Les associations des antimicrobiens aux dérivés des fluorures ont montré une amélioration de l’effet des antibiotiques par la réduction des leurs concentrations et toxicité pour traiter correctement ces pathogènes résistants. Par conséquent, des antibiotiques associés aux dérivés de fluorure pourraient devenir une option de traitement contre des souches résistantes afin de diminuer la toxicité causée par de fortes doses des antibiotiques conventionnels. / The emergence of bacterial strains resistant to antibiotics is a worrying phenomenon, which has spread worldwide. Staphylococcus aureus and Pseudomonas aeruginosa are multi-resistant opportunistic pathogenic bacteria that can cause many diseases. However, they have become difficult to treat with antibiotics without causing toxicity to humans. It is therefore necessary to develop new molecules against resistant pathogens to combat this problem. Due to their pharmacological action, fluorides have an antibacterial effect on tooth enamel which suggests that their combination with antibiotics could improve antimicrobial activity. Thus, we proposed to study in vitro activities of vancomycin (VAN), oxacillin (OXA), ceftazidime (CFT) and meropenem (MER) alone or combined with sodium fluoride (NaF) and lithium fluoride (LiF ) that were evaluated against resistant and susceptible strains of S. aureus and P. aeruginosa by antimicrobial susceptibility testing , in determining their minimum inhibitory concentration (MIC), minimum bactericidal concentration (MBC), and kinetics (Time-Kill). The evaluation of cytotoxicity on human red blood cells, and determination of stability at two different temperatures (4°C, 22°C) were also performed. Associations of antibiotics with fluoride derivatives showed an improvement by increasing the effect of antibiotics with minimal levels necessary to achieve the same result by reducing their toxicity and to properly kill these resistant pathogens. Therefore, antibiotics associated with fluoride derivatives could become a treatment option against resistant strains and at the same time reduce the toxicity caused by high doses of conventional antibiotics.

Reaktionerna blev starka, vilket kan bero på att <i>Kill Bill</i> är en actionfilm där kvinnorna har en annan roll än den lättklädda medhjälparen. Då kvinnorna står i fokus var filmen som gjord för en analysav hur just kvinnan gestaltas i filmens värld. Syftet med uppsatsen är att undersöka om de patriarkala strukturerna förändras när typiskt manliga roller intas av kvinnor. Analysen inriktar sig främst på filmen <i>Kill Bill</i>, men diskuterar också det sammanhang i vilket filmen skapats och lanserats det vill säga filmindustrin i Hollywood som knappast gjort sig känd för några jämställdhetssträvanden. The analysis is mainly concentrated on the film <i>Kill Bill</i>, but will also discuss the context in which it was made - the Film Industry in Hollywood which has hardly been known for any ambition to achieve gender equality.</p>

Denna studie syftar till att synliggöra olika bedrägliga beteenden som riktar sig mot innovatörer. I studien diskuteras möjligheterna till att etablera en funktion som skyddar innovatörer mot en typ av övergrepp som blir allt vanligare. Hur skall subtil manschettbrottslighet begränsas när den är både riskfri och lönsam? En vägledande fråga i diskussionen har varit: Skall innovatörer ha samma skydd för sin upphovsrätt som andra fria yrkesutövare, eller ska de fortsätta få vara villebråd utan skydd av rättsväsendet? Resultatet bygger på dokumentanalyser och en kvalitativ studie om upphovsmän till patent, samt intervjuer med personer inom branschen innovationer och entreprenörskap. Resultat baseras bl a på den kvalitativa studien och beskrivs genom fakta om innovatörerna Dr. Håkan Lans och M.D. Johan Ullman, vilka båda fått sina fall uppmärksammade i massmedia. Resultatet av analysen visar att* Uppfinnare är en minoritet som saknar rättsskydd i Sverige* Likhet inför lagen gäller inte för innovatörer och upphovsmän* Det är nästintill undantagslöst den ekonomiskt starkaste som vinner patentstrider Ett par av slutsatserna är att risken för upptäckt styr omfattningen av subtil ekobrottslighet. När det är riskfritt att bedriva sådan verksamhet ökar antalet brott. Det finns metoder att begränsa organiserad ekonomisk brottslighet, bedrägerier och kränkningar. Slutdiskussionen ger förslag till hur man kan skydda den svagare parten och skapa förutsättningar för uppfinnare att verka, för en större industriell tillväxt och därmed fler nya arbetstillfällen i Europa.

Applications of geographic information science in the archaeological research of the Fincastle Kill Site (D1Ox 5) Alberta, Canada, and Tel Beth-Shemesh, Israel

Many scientists have used the expediency of geographic information science (GIS) for archaeological analyses, such as predictive site location modeling and producing topographical site surveys. However, the use of GIS to explore the spatial relationships among the architecture, geography and site artifacts has rarely been done. This research focuses on visualizing and analyzing these relationships using GIS. The sites of Tel Beth Shemesh, Israel and the Fincastle Kill Site (DIOx 5), north-east of Taber, Alberta, were used as case studies, as they were very differnt types of sites. Based on field measurments and by using specific GIS applications and software, components of these sites were reconstructed in virtual space as GIS models. Other recorded field data were used as input parameters into the models in order to attain the most accurate representations and analyses of the sites. The analysis at Fincastle Kill Site used two types of GIS models: 1) a viewshed model to assess possible bison hunting techniques and 2) surface interpolation models that delineated correlations between high density and low density areas of archaeological remains. The investigation at Tel Beth-Shemesh used a GIS model to store, visualize, interpret and assess the quality and accuracy of the field data recorded during 2001 - 2004 excavations. Predominately, the work in this thesis did not aim at answering any profound questions about the archaeology of either site, although in some cases it did, but rather focused on developing useful GIS tools for the archaeologist. Ransomware-attacker : En kvalitativ studie kring informationssäkerhetsarbetet inom mindre svenska kommuner

Ransomware-attacker har blivit ett allt större hot i och med samhällets ständigt pågående digitalisering. Denna studie undersöker vilka faktorer som är viktiga för att förhindra ransomware-attacker mot mindre svenska kommuner. För att åstadkomma detta genomfördes semistrukturerade intervjuer med sex olika respondenter. De som intervjuades arbetade alla i mindre svenska kommuner och hade god insyn och kunskap kring kommunens IT- och informationssäkerhetsarbete. Materialet analyserades sedan utifrån Protection Motivation Theory (PMT). Studien diskuterar både kommunernas attityd till informationssäkerhet samt konstaterar vilka säkerhetsåtgärder som utmärker sig som viktigast. Dessa var skyddade säkerhetskopior, utbildning samt kontinuitetsplaner kopplade till just IT-attacker. / Ransomware attacks have become an increasing threat with the ongoing digitalization of society. This study investigates what factors are important to prevent ransomware attacks against smaller Swedish municipalities. To accomplish this, semi-structured interviews were conducted with six different respondents. The interviewees all worked in smaller Swedish municipalities and had good insight and knowledge of the municipality's IT and information security work. The material was then analyzed using Protection Motivation Theory (PMT). The study discusses both the municipalities' attitude to information security and notes which security measures stand out as most important. These were protected backups, education and continuity plans linked to IT attacks. The paper then concludes with suggestions for further research.

Bestimmung der in vitro Aktivität von Clindamycin, Imipenem, Metronidazol und Piperacillin/Tazobactam gegenüber sensiblen und resistenten Bacteroides fragilis Stämmen mittels Absterbekinetik

Obligat anaerob wachsende Bakterien sind an einer Vielzahl von Infektionen beteiligt. Dabei ist Bacteroides fragilis einer der wichtigsten opportunistischen Erreger unter den Anaerobiern. Bei Verdacht auf eine Infektion durch obligate Anaerobier muss nach Materialentnahme für die mikrobiologische Diagnostik unverzüglich eine kalkulierte Therapie eingeleitet werden. Oft ist eine chirurgische Therapie notwendig, die ebenso wie eine adäquate Antibiotikatherapie entscheidend für den Verlauf der Erkrankung ist. Wichtige Substanzen für eine Therapie bei Infektionen mit Beteiligung von B. fragilis sind Clindamycin, Imipenem, Metronidazol und Piperacillin/Tazobactam. Um Aussagen zur in vitro Wirksamkeit dieser Antibiotika gegenüber obligaten Anaerobiern treffen zu können, wurden in der vorliegenden Arbeit die Aktivitäten von verschiedenen Konzentrationen des jeweiligen Antibiotikums auf das Wachstum von sensiblen und resistenten B. fragilis Stämmen mittels Absterbekinetik untersucht. In Abhängigkeit von der zuvor ermittelten minimalen Hemmkonzentration des jeweiligen Antibiotikums wurden die Stämme in 2 Gruppen eingeteilt. Die erste Gruppe umfasst alle Stämme mit einer MHK ≤ 8 µg/ml. In der zweiten Gruppe sind die Stämme mit einer MHK > 8 µg/ml zusammengefasst. Die einzelnen Stämme wurden mit einem Vielfachen der minimalen Hemmkonzentration (MHK) beziehungsweise einem Vielfachen der im menschlichen Blutplasma maximal erreichbaren Konzentration (Cmax) des jeweiligen Antibiotikums inkubiert und die Bakterienkonzentration zu definierten Zeitpunkten ermittelt. Dadurch können sowohl die Wirksamkeit unterschiedlicher Antibiotikakonzentrationen als auch verschiedene Antibiotikaklassen miteinander verglichen und Aussagen zu Empfehlungen für kalkulierte Therapien getroffen werden.

This paper takes a cultural materialist approach in analyzing the hegemonic purpose of using Harper Lee’s To Kill a Mockingbird in American education. Ideas from critical race theory and Lee’s second novel, Go Set a Watchman, are used to reveal obfuscated aspects of Mockingbird’s narrative. These aspects have been repurposed to fit a Eurocentric palate, and have let the book achieve success under the guise of being a progressive and multiculturalist work. Mockingbird’s narration, marked by childlike innocence, has been used to obfuscate Eurocentric ignorance of racial and economic inequality. The text has also been used to divert blame from those in power onto those oppressed by a hegemonic system. Racism is in Mockingbird inaccurately described as an individual moral issue, rather than a system of discrimination which is deeply ingrained in every aspect of U.S. society. The liberal moderate ideology which informs Atticus character has historically been ignored due to his unquestionable, near-mythical position as a moral role model. The paper finds that Mockingbird has been used as part of a greater Eurocentric narrative which positions the Civil Rights Movement as a white movement of moral improvement.

Study Design and Dose Regimen Evaluation of Antibiotics based on Pharmacokinetic and Pharmacodynamic Modelling

Current excessive use and abuse of antibiotics has resulted in increasing bacterial resistance to common treatment options which is threatening to deprive us of a pillar of modern medicine. In this work methods to optimize the use of existing antibiotics and to help development of new antibiotics were developed and applied. Semi-mechanistic pharmacokinetic-pharmacodynamic (PKPD) models were developed to describe the time course of the dynamic effect and interaction of combinations of antibiotics. The models were applied to illustrate that colistin combined with a high dose of meropenem may overcome meropenem-resistant P. aeruginosa infections. The results from an in vivo dose finding study of meropenem was successfully predicted by the meropenem PKPD model in combination with a murine PK model, which supports model based dosage selection. However, the traditional PK/PD index based dose selection was predicted to have poor extrapolation properties from pre-clinical to clinical settings, and across patient populations. The precision of the model parameters, and hence the model predictions, is dependent on the experimental design. A limited study design is dictated by cost and, for in vivo studies, ethical reasons. In this work optimal design (OD) was demonstrated to be able to reduce the experimental effort in time-kill curve experiments and was utilized to suggest the experimental design for identification and estimation of an interaction between antibiotics. OD methods to handle inter occasion variability (IOV) in optimization of individual PK parameter estimates were proposed. The strategy was applied in the design of a sparse sampling schedule that aim to estimate individual exposures of colistin in a multi-centre clinical study. Plasma concentration samples from the first 100 patients have been analysed and indicate that the performance of the design is close to the predicted. The methods described in this thesis holds promise to facilitate the development of new antibiotics and to improve the use of existing antibiotics.
