Biological control of the two-spotted spider mite, Tetranychus urticae Koch (Acari : tetranychidae).

The two-spotted spider mite (TSM), Tetranychus urticae Koch, is an important pest of many greenhouse and field crops worldwide. The development of resistance in TSM populations to chemical acaricides, allied with public health concerns about pesticide residues, has motivated the search for alternative control measures to suppress the pest. Hyphomycetous fungi are promising agents for mite control and the fungus Beauveria bassiana (Bb) (Balsamo) Vuillemin was investigated in this study as a biocontrol agent. The principal objectives of this study comprised: a) screening Bb strains for their pathogenicity against T. urticae; b) testing the effect of adjuvants on the efficacy of Bb; c) studying the effect of plant type on persistence of Bb and the efficacy of control of Bb against T. urticae; d) evaluating the field efficacy of Bb applications against T. urticae; e) testing the compatibility of Bb with selected fungicides; and f) assessing the synergy between Bb and soluble silicon for T. urticae control. Screening bioassays of sixty-two strains of Bb identified the two most effective strains, PPRI 7315 (R289) and PPRI 7861 (R444), that caused mortality levels of more than 80% of adult mites at 9 d post-inoculation with 2 × 108 conidia ml-1. These strains performed significantly better than the Bb commercial strain PPRI 5339, in laboratory bioassays. The two strains also attacked mite eggs, causing 53.4% and 55.5% reduction in egg hatchability at 2 × 108 conidia ml-1 respectively. However, PPRI 7861 showed relatively higher production of conidia in culture and was, therefore, selected for further trials under greenhouse and field conditions. Greenhouse evaluations of the effects of two adjuvants (Break-thru® and a paraffin oil-based emulsion) on efficacy of Bb demonstrated a higher efficacy of the biocontrol agent (BCA) when it was applied with Break-thru® or the oil solution than with water alone. Moreover, Bb conidia applied in Break-thru® solution resulted in greater control of TSM than conidia applied in the mineral oil. There was also a dose-response effect and the control of TSM by Bb increased when the concentration of conidia was increased. The control of TSM by Bb in beans (Phaseolus vulgaris L), cucumber (Cucumis sativus L.), eggplant (Solanum melongena L.), maize (Zea mays L.) and tomato (Solanum lycopersicum L.) was tested in greenhouse trials. On these crops, the persistence of conidia declined over time. The rate of decline was significantly higher on maize. However, TSM mortality was positively correlated with the amount of conidia deposited on leaves immediately after spraying, rather than their persistence over time. Higher levels of mortality of TSM due to Bb application were observed on beans, cucumber and eggplants, suggesting that the type of crop must be taken into consideration when Bb is applied as a BCA. Field efficacy of Bb against mites was evaluated in two trials on eggplants. Based on assessment of population densities of mites and leaf damage assessments; both trials showed that the strain PPRI 7861 controlled TSM in the field. Two commonly used fungicides, azoxystrobin and flutriafol, were investigated in vitro tests on culture medium and laboratory bioassays on detached bean leaves (Phaseolus vulgaris L.) for their effects on Bb. Azoxystrobin (a strobilurin) was less harmful to Bb while flutriafol was found to be inhibitory. Another important finding of this study was the substantial enhancement of Bb efficacy by soluble silicon. When Bb was combined with soluble Si, the control of TSM was better than when either of the two products was applied alone. Moreover, application of soluble Si as a plant fertilizer in hydroponic water nutrient increased accumulation of peroxidase, polyphenoloxidase and phenylalanine ammonia-lyase enzymes in leaves of plants infested with TSM. Increased activity of these defense enzymes in leaves deters feeding behaviour of mites. Avaliação dos fatores de risco no processamento de café verde para o aparecimento de ocratoxina A (OTA) / Evaluation of risk factors in green coffee processing for ochratoxin A (OTA)

Dentre os perigos relacionados com a produção de café verde, a ocratoxina A (OTA) é considerada significativa nesse produto. Com o objetivo de estudar a origem da OTA no processamento de café verde e avaliar os possíveis fatores de risco (região, etapas do processamento, fungos, insetos e ácaros) que poderiam contribuir na presença de OTA, foram amostradas três fazendas nos seguintes municípios: Bom Jardim (RJ), Espírito Santo do Pinhal (SP) e Cornélio Procópio (PR), onde havia a produção de café verde pela via seca e úmida. Foram coletadas 109 amostras de grãos de café, em diversas etapas do processamento, no período de maio a agosto de 2002. As amostras foram colhidas de acordo com fluxograma estabelecido e as etapas estudadas foram: cereja (campo), via seca, via úmida, armazém e café de varrição. Dessas amostras, 10 apresentaram OTA, que variou na sua quantificação de 3 a 101 ng.g-1. A maior prevalência de OTA foi a do café de varrição 57%, a via úmida apresentou uma prevalência de 10,3% e a via seca 7%. Na etapa cereja o fungo potencialmente toxigênico mais encontrado foi Penicillium spp, e quanto aos insetos, as larvas de Diptera e os Homoptera; na via úmida foram encontradas linhagens do Aspergillus seção Nigri e Circundatti e Penicillium spp, a infestação predominante foi a broca do café e os ácaros da família Acaridae; na etapa via seca registrou-se o isolamento de fungos do gênero Penicillium spp e Aspergillus seção Nigri , a broca do café foi a infestação predominante junto com os ácaros da família Acaridae, na etapa de armazém foram isolados Penicillium spp e Aspergillus seção Nigri, o inseto predominante foi a broca do café e os ácaros da família Acaridae; no café de varrição foram isolados Penicillium spp e Aspergillus seção Nigri e a broca do café estava presente junto com insetos da Ordem Diptera. Os resultados de cada etapa foram comparados por Análise de Variância em um nível de significância de 95%. Na análise de fatores de risco o café de varrição foi o mais significativo (p< 0,001). / The ochratoxin A (OTA) is considered to be among the most significant coffee contaminants with related hazards. With the purpose to study the origin of the OTA in the processing of green coffee and to evaluate the possible risk factors (region, step of processing, fungi, insects and mites) that could contribute in the OTA presence, had been selected three regions (states) of Brazil: Bom Jardim/RJ, Espírito Santo do Pinhal/SP and Cornélio Procópio/PR. A total of 109 samples of coffee beans were collected in conformity with flow chart established at different stages of ripening and processing: cherry (field), dry method, wet method, and warehouse and varrição coffee, from May to August of 2002. Of these samples 10 had presented OTA, which varied in its quantification from 3 to 101 ng.g-1. The biggest prevalence of OTA was the varrição coffee 57%, the wet method presented a prevalence of 10,3% and dry way 7%. In the stage cherry fungi potentially toxigenic was more found Penicillium spp, the insects had been the larvae of Diptera and the Homoptera; in the wet way it was found Aspergillus section Nigri and Circundatti and Penicillium spp, the predominant infestation was the coffee berry borer and the mite of the Acaridae family; in the stage wet methods spp was registered the isolation the Penicillium spp and Aspergillus section Nigri, the predominant infestation was the coffee berry borer together with the mites of the Acaridae family; in the stage of warehouse had been isolated Penicillium spp and Aspergillus section Nigri, and the predominant insect was the coffee berry borer and mites of the family Acaridae; in the varrição coffee had been isolated Penicillium spp and Aspergillus section Nigri the coffee berry borer was the most present insects together of Diptera. The results of each step had been with compared by Analysis of Variance. In the analysis of risk factors the varrição coffee\" was the factor most strongly related to the risk of the presence of OTA (p<0,001 ).

Les performances du végétal et des arthropodes dont il constitue le microhabitat dépendent des températures de surface foliaire. Celles-ci peuvent dévier fortement de la température de l’air et présenter des niveaux d’hétérogénéité différents selon l'échelle spatiale considérée. La feuille atténue les températures extrêmes en rapprochant son amplitude de variation journalière de celle de la température de l’air. Cependant, cette réponse diminue l’hétérogénéité des températures de surface foliaire et les capacités de thermorégulation comportementale des arthropodes à l'échelle de la feuille. Les températures moyennes de surface foliaire atténuent peu le réchauffement, et déterminent localement la performance photosynthétique du végétal. De l’échelle de la feuille à celle de la canopée, les plantes montrent des réponses différentes au réchauffement. Ce type de changement d'échelle est primordial pour améliorer notre compréhension de l'impact des changements climatiques. / Plant performance and leaf-dwelling arthropods are impacted by leaf surface temperatures. Leaf surface temperatures can show important deviation from air temperature and present different levels of heterogeneity depending on the spatial scale. The leaf buffers temperature extremes by getting closer in amplitude to air temperature. However, this physiological response decreases the heterogeneity of temperatures at the leaf surface and the opportunities for arthropods to behavioraly thermoregulate in this microclimate. Mean temperatures at the leaf surface show low buffering abilities in response to warming and locally determine photosynthetic performance. From the leaf to the canopy scale, plants show different responses to warming and scaling is crucial to increase our understanding of the impact of global warming.

A visual semiotic analysis of the hidden meanings, myths and ideologies in Old Mutual South Africa's CSR 2.0 corporate advertising

Abstracts in English and Afrikaans / The aim of this exploratory study was to investigate the underlying meanings of Old Mutual South Africa’s (OMSA’s) CSR 2.0 corporate advertisements. It specifically examined the underlying myths and ideologies conveyed through the texts. This qualitative study was conducted by means of a visual semiotic analysis (Machin 2012) on eight CSR 2.0 corporate advertisements, which were purposively selected. A theoretical framework to measure visual CSR 2.0 corporate advertisements was developed, which aided in the sampling, data analysis and interpretation processes. The findings of the study indicated that by positioning its CSR 2.0 positively in terms of change and transformation, progress and equal opportunities for all citizens, OMSA is fostering stakeholder-organisation relationships. In addition, it was also found that OMSA adopts established representational tropes of CSR 2.0 and use them repetitively that firmly entrenches the intended/encoded message to the viewer. / Die doel van hierdie verkennende studie was om die onderliggende betekenis van CSR 2.0 korporatiewe advertensies in Ou Mutual Suid-Afrika te ondersoek. Dit is spesifiek gemoeid met die onderliggende mites en ideologieë wat deur middel van die tekste oorgedra word. Die kwalitatiewe studie is uitgevoer deur middel van 'n visuele semiotiese ontleding (Machin 2012) op agt CSR 2.0 korporatiewe advertensies, wat doelbewus gekies is. 'n Teoretiese raamwerk, om visuele CSR 2.0 korporatiewe advertensies te meet, is ontwikkel, dit het bygedra tot die steekproefneming, data-analise en interpretasie prosesse. Die bevindinge van die studie dui daarop dat deur CSR 2.0 positief in terme van verandering en transformasie, vooruitgang, en gelyke geleenthede vir alle burgers te posisioneer, OMSA verhoudings met belanghebbendes kan bevorder.
