Étude des facteurs influençant la susceptibilité à l'infection au VIH chez des femmes africaines

Chez la femme, la majorité des cas d’infection au VIH sont acquis lors de relations hétérosexuelles. Cependant, très peu d’informations sont disponibles concernant l’immunité locale naturelle du tractus génital féminin, les facteurs influençant la susceptibilité à l’infection au VIH dans ce compartiment, ainsi que la réponse immunitaire de la muqueuse enclenchée après l’infection. Le but de notre projet est donc d’étudier certains facteurs pouvant être impliqués dans la susceptibilité à l’infection au VIH, afin de mieux comprendre l’immunité du tractus génital féminin. Nous avons, dans un premier temps, analysé le rôle du polymorphisme des gènes HLA-G et HLA-E sur la susceptibilité au VIH dans une population de femmes zimbabwéennes. La présence de l’allèle HLA-G*0105N, en combinaison avec le génotype HLA-EG/HLA-EG, était associée avec une diminution du risque d’infection. Puis, dans une étude cas-contrôle de travailleuses du sexe (TS) du Bénin, nous avons mesuré l’expression de HLA-G soluble au niveau du plasma. Nous avons observé une différence significative dans l’expression de HLA-G soluble, celle-ci étant plus faible dans le groupe des TS VIH positives comparé aux groupes de TS VIH négatives et de femmes VIH négatives de la population générale. Nous avons aussi analysé l’expression de cytokines et chimiokines dans le sérum et le tractus génital des participantes de l’étude du Bénin. Nous avons constaté que chez les TS VIH positives il y avait une expression plus élevée des chimiokines MPC-3, IP-10 et MIG dans le tractus génital et le sérum comparativement aux deux autres groupes. Les patrons d’expression des cytokines variaient selon les compartiments : le niveau de TNF-α et IFN-γ était plus élevé dans le tractus génital des TS VIH positives, alors que le niveau d’IL-2, d’IL-10 et de TNF-α était plus faible dans le sang des TS VIH positives, comparativement aux deux autres groupes. Ainsi, au niveau du tractus génital des femmes VIH positives, il semble y avoir une activation chronique du système immunitaire dans le but de favoriser la dissémination/perpétuation du virus. Les patrons d’expression différents entre le milieu systémique et génital nous montrent que l’immunité présente dans un compartiment n’est pas nécessairement le reflet de l’autre. Nous avons aussi observé une augmentation significative des niveaux d’IL-4, de MIP-1α, de MIP-1β et de MCP-1 dans le sérum des TS VIH négatives. Ces personnes, hautement exposées mais non infectées, semblent démontrer une plus grande capacité à enclencher une réponse immunitaire précoce pour empêcher la dissémination du virus. Notre étude a donc permis d’acquérir de nouvelles connaissances sur l’immunité du tractus génital féminin en relation avec l’infection au VIH. / Initial exposure to HIV during heterosexual transmission occurs in the female genital tract. However, little is known about the local immunity, the factors influencing the susceptibility to HIV infection and the immune response in the female genital tract against HIV infection. The aim of this study is to analyse some factors that could be implicated in the susceptibility to HIV infection and to analyse, in part, the immunity present in the female genital tract. We investigated the role of HLA-G and HLA-E in the susceptibility to HIV infection in a cohort of Zimbabwean women. We found that the presence of HLA-G*0105N allele in combination with the genotype HLA-EG/HLA-EG was associated with a decrease in the risk of HIV infection. We also measured the expression of soluble HLA-G in a study of commercial sex workers (CSW) in Benin. Levels of soluble HLA-G were lower in the HIV-1-infected CSWs compared to those observed in both the HIV-1-uninfected CSWs and the HIV-1-uninfected women from the general population at low risk of infection. We also analysed the chemokine and cytokine expression patterns in the serum and female genital tract of the three groups of women. HIV-1-infected CSWs had significantly higher blood and genital levels of the chemokines IP-10, MCP-3 and MIG compared with those in both the HIV-1-uninfected CSW and non-CSW groups. HIV-1-infected CSWs had significantly higher genital mucosal levels of the cytokines TNF-α and IFN-γ compared with those in both the HIV-uninfected CSW and non-CSW groups. In contrast, the serum levels of the cytokines IL-2, IL-10 and TNF-α were lower in HIV-1-infected CSWs compared with those in the other groups. This suggests the presence of a constant immune cells recruitment and immune activation in the female genital tract in order to favour perpetuation and dissemination of the virus. Our results also demonstrate the important difference between the systemic and the mucosal immunity. We also observed a significant increase in the levels of IL-4, MIP-1α, MIP-1β and MCP-1 in the serum of the HIV-1-uninfected CSWs. It seems that these highly-exposed and yet uninfected women can have a better capacity to mount an early immune response against HIV. This study gives us new insights of the mucosal immunology of HIV infection.

Effects of physicochemical properties of fibrous feed on feeding behaviour and gut health of growing and finishing pigs.

The broad objective of the study was to determine feeding behaviour and gut health of pigs fed different fibres at varying inclusion levels. Maize cob, maize stover, sunflower hulls, veld grass, sawdust, lucerne and dried citrus pulp were used in growing and finishing pig diets to provide a wide range of physicochemical properties. Time spent eating, drinking, lying down, sitting/standing and other activities was observed using video cameras. Blood samples were collected at the end of the trial for both growing and finishing pigs for analysis of glycated haemoglobin, albumin, globulin, total protein, creatine kinase, urea and uric acid. Intestinal segments were collected at the end of trial for growing pigs to determine mucosal architecture of the intestines. Digestible energy (DE), bulk density (BD), acid detergent fibre (ADF) and water holding capacity (WHC) were the most important variables predicting time spent on different behavioural activities in growing pigs (P < 0.001). Water holding capacity, neutral detergent fibre (NDF) and ADF were the most important variables involved in predicting time spent on different behavioural activities in finishing pigs (P < 0.001). Physicochemical properties influenced time spent on different behavioural activities and they provided relationships with time spent on different behavioural activities in both growing and finishing pigs. Glycated haemoglobin, albumin, globulin, total protein and uric acid were factors that influenced time spent eating in growing pigs (P < 0.05). In finishing pigs, only serum total protein was selected as the best predictor variable influencing time spent eating (P < 0.05). The blood metabolites were correlated with time spent eating and drinking. They provided threshold values with time spent eating and drinking by pig. Hence, they can also be used as potential biomarkers that modulate neuronal pathways which reduce time spent eating and drinking. In this study, bulk density (BD) and NDF were the best predictor variables influencing villi height (VH) and apparent villi surface area (AVSA) in pigs (P < 0.05). Grouped pigs fed fibrous diet spent more time eating, lying down, standing, walking and fighting (P < 0.05). Skin lesions appeared the most on neck and shoulder region followed by chest, stomach and hind leg region, and finally head region (P < 0.05). Fibrous diet did not reduce aggressive behaviours, rather aggressive behaviours emanated out of frustration when queuing at the feeder. It was concluded that physicochemical properties of fibrous diets and nutritionally-related blood metabolites influence feeding behaviour. Efectes de la suplementació dietètica amb proteïnes plasmàtiques sobre la resposta immunitària en un model d’inflamació pulmonar aguda en ratolí

El concentrat de plasma assecat per polvorització (SDP) i el d’immunoglobulines (IC) són suplements dietètics que han estat àmpliament utilitzats en l’alimentació dels animals de granja i que han estat proposats com alternativa a l’ús dels antibiòtics com a promotors del creixement, especialment durant el període posterior al deslletament. Aquests suplements incrementen la taxa de creixement degut, en part, a que atenuen la resposta del sistema immunitari. Per exemple, s’ha pogut observar que la suplementació dietètica amb proteïnes plasmàtiques redueix la sobreestimulació immunitària en un model de malaltia inflamatòria intestinal. El sistema immunitari mucosal comú connecta els llocs inductors (en aquest cas la mucosa intestinal) amb els llocs efectors (com les mucoses naso- i broncoalveolar o la genito-urinària), que facilita el moviment de cèl•lules immunitàries entre teixits allunyats anatòmicament i permet que la dieta moduli la resposta associada a infeccions extraintestinals. Així doncs, ens hem plantejat si la suplementació dietètica amb proteïnes plasmàtiques podia modular la resposta immunitària en una inflamació pulmonar aguda. Primerament es va establir una pauta adequada per obtenir un model d’inflamació pulmonar robust i reproduïble. Seguidament s’han estudiat els efectes dels suplements SDP i IC sobre la resposta immunitària en aquest model d’inflamació, en ratolins acabats de deslletar. S’han analitzat cèl•lules i mediadors inflamatoris involucrats tant en la resposta innata, com en la resposta adaptativa. Finalment, també s’han estudiat els efectes dels pinsos experimentals sobre diferents elements que intervenen en la resolució de la resposta inflamatòria, com les cèl•lules reguladores i les citocines antiinflamatòries. L’administració de LPS produeix una migració de leucòcits i un alliberament massiu de citocines i quimiocines proinflamatòries, que recluten i activen monòcits i neutròfils a l’espai alveolar i al teixit pulmonar. A més estimula la immunitat adaptativa, en la que hi ha un increment considerable de limfòcits Th activats, així com una gran producció de citocines. Ambdues dietes disminueixen el reclutament de leucòcits, sobretot els neutròfils i els monòcits. Tot i que la resposta innata és més extensa, les dietes són més efectives sobre la resposta adaptativa, on redueixen les poblacions limfocitàries estimulades per l’LPS tant a pulmó com a sang. Els suplements també atenuen de forma notable l’efecte de l’LPS sobre l’expressió de citocines i quimiocines proinflamatòries a l’espai alveolar. Els efectes de l’SDP i l’IC estan mediats per canvis en l’expressió de citocines antiinflamatòries, ja que augmenten la concentració de IL-10 a pulmó i a jejú, i la del TGF-β a pulmó. Ambdues dietes redueixen el balanç entre els limfòcits Th activats i els limfòcits T reguladors, d’aquesta manera atenuen la magnitud de la resposta inflamatòria i augmenten la concentració de mediadors essencials per la reparació del teixit. Els resultats d’aquest estudi indiquen que els suplements amb proteïnes plasmàtiques poden limitar la resposta immunitària pulmonar i podrien ser útils en la prevenció i atenuació de les malalties inflamatòries extraintestinals. / The concentrated spray-dried plasma (SDP) and immunoglobulin (IC) are dietary supplements that have been widely used in the diet of farm animals and proposed as an alternative to the use of antibiotics as growth promoters, especially in the subsequent weaning period. These supplements increase the growth rate, in part because they attenuate the response of the immune system. The common mucosal immune system connects the inductive sites (in this case the intestinal mucosa) with the effector sites (as nasopharyngeal and bronchoalveolar mucosa or genito-urinary), which facilitates the movement of immune cells between anatomically distant tissues and allows the diet to modulate the associated response to intestinal infections. Consequently, the aim of the present thesis was to evaluate whether the dietary supplementation with plasma proteins could modulate the immune response in an acute lung inflammation. First, a suitable pattern was established in order to obtain a robust and reproducible inflammatory lung model. Next, the effects of SDP and IC supplementation on the immune response in this inflammation model were studied in newly weaned mice. Cells and inflammatory mediators, involved either in innate response as in adaptive response, have been analyzed. Finally, the effects of experimental feed on different elements involved in the resolution of the inflammatory response, such as regulatory cells and inflammatory cytokines, have also been assessed. Both diets decreased the recruitment of leukocytes. Although the innate response is more extensive, the diets are more effective on the adaptive response. The supplements also significantly attenuated the effect of LPS on the expression of pro-inflammatory cytokines and chemokines in the alveolar space. The effects of SDP and IC are mediated by changes in the expression of anti-inflammatory cytokines. Both diets reduced the balance between activated Th cells and regulatory T cells, in such a way that, the magnitude of the inflammatory response is attenuated and the concentration of essential mediators for tissue reparation is increased. In conclusion, the results of this study indicate that plasma protein supplementation is able to limit the pulmonary immune response and could be useful in the prevention and attenuation of the extraintestinal inflammatory diseases.

Isso posto, foi formulado o presente estudo com o fito principal de fazer a avaliação das respostas imunes humorais e celulares em diferentes intervalos pós-desafio com o VBI de aves previamente vacinadas ou não, realizando-se a mensuração de anticorpos no soro e na lágrima, e a quantificação da expressão de genes relacionados às respostas imunes na superfície traqueal, a fim de correlacionar tais parâmetros com o estado de proteção ao desafio. Os resultados demonstraram que os aumentos significativos nos níveis de anticorpos lacrimais dos isótipos IgG e IgA nas aves previamente vacinadas e também na expressão dos genes relacionados às respostas imunes, sobretudo o CD8, a Granzima A e o IFNg foram correlacionados negativamente com um ou mais parâmetros de alterações patológicas traqueais. Constatou-se também, que a memória das respostas imunes humorais e cito-mediadas conferida por uma única vacinação contra a BI no primeiro dia de idade é dependente da dose vacinal administrada / Abstract: Avian infectious bronchitis virus (IB) is a worldwide infectious disease which causes significant economic losses in poultry industry. The innate and acquired immune responses, including whether there mediated by both cellular and humoral factors that are induced after infection or vaccination with IB virus (IBV) are characterized by their higher complexity and for the relevant aspects that are still poorly known, with respect to the elements able to exercise one or more actions against the pathogen and that culminate in the restriction of its propagation and also on your clearance of the host organism. So, this project was formulated with the main done to make the evaluation of cellular and humoral immune responses at different intervals post-immunization or postchallenge with IBV poultry previously vaccinated or not, performing the measurement of antibodies in serum or tears and quantitation of the expression of genes related to immune responses in tracheal surface, correlating these parameters with the protection against IBV. The results showed that both significative increase of IgG and IgA isotypes in tears of previously vaccinated chickens and the expression levels of genes related to immune responses, especially CD8, Granzyme A and IFN g were negatively correlated with one or more parameters of pathological lesions at trachea. Activation and Role of Memory CD8 T Cells in Heterologous Antiviral Immunity and Immunopathology in the Lung: A Dissertation

Each individual experiences many sequential infections throughout the lifetime. An increasing body of work indicates that prior exposure to unrelated pathogens can greatly alter the disease course during a later infection. This can be a consequence of a phenomenon known as heterologous immunity. Most viruses invade the host through the mucosa of a variety of organs and tissues. Using the intranasal mucosal route of infection, the thesis focused on studying modulation of lymphocytic choriomeningitis virus (LCMV)-specific memory CD8 T cells upon respiratory vaccinia virus (VV) infection and the role of these memory CD8 T cells in heterologous immunity against VV and altered immunopathology in the lung. The VV infection had a profound impact on memory T cells specific for LCMV. The impact included the up-regulation of CD69 expression on LCMV-specific CD8 memory T cells and the activation of their in vivoIFN-γ production and cytotoxic function. Some of these antigen-specific memory T cells selectively expanded in number, resulting in modulation of the original LCMV-specific T cell repertoire. In addition, there was a selective organ-dependent redistribution of these LCMV-specific memory T cell populations in secondary lymphoid tissue (the mediastinal lymph node and spleen) and the non-lymphoid peripheral (the lung) organs. The presence of these LCMV-specific memory T cells correlated with IFN-γ-dependent enhanced VV clearance, decreased mortality and marked changes in lung immunopathology. Thus, the participation of pre-existing memory T cells specific for unrelated agents can alter the dynamics of mucosal immunity. This is associated with an altered disease course in response to a pathogen. The roles for T cell cross-reactivity and cytokines in the modulation of memory CD8 T cells during heterologous memory CD8 T cell-mediated immunity and immunopathology were investigated. Upon VV challenge, there were preferential expansions of several LCMV-specific memory CD8 T cell populations. This selectivity suggested that cross-reactive responses played a role in this expansion. Moreover, a VV peptide, partially homologous to LCMV NP 205, stimulated LCMV-NP205 specific CD8 T cells, suggesting that NP205 may be a cross-reactive epitope. Poly I:C treatment of LCMV-immune mice resulted in a transient increase but no repertoire alteration of LCMV-epitope-specific CD8 T cells. These T cells did not produce IFN-γ in vivo. These results imply that poly I:C, presumably through its induced cytokines, was assisting in initial recruitment of LCMV-specific memory CD8 T cells in a nonspecific manner. VV challenge of LCMV-immune IL-12KO mice resulted in activation and slightly decreased accumulation of LCMV-specific CD8 T cells. Moreover, there was a dramatic reduction of in vivoIFN-γ production by LCMV-specific IL-12KO CD8 T cells in the lung. I interpreted this to mean that IL-12 was important to augment IFN-γ production by memory CD8 T cells upon TCR engagement by antigens and to induce further accumulation of activated memory CD8 T cells during the heterologous viral infection. This thesis also systematically examined what effect the sequence of two heterologous virus challenges had on viral clearance, early cytokine profiles and immunopathology in the lung after infecting mice immune to one virus with another unrelated viruses. Four unrelated viruses, [LCMV, VV, influenza A virus or murine cytomegalovirus (MCMV)], were used. There were many common changes observed in the acute response to VV as a consequence of prior immunity to any of three viruses, LCMV, MCMV or influenza A virus. These included the enhanced clearance of VV in the lung, associated with enhanced TH1 type responses with increased IFN-γ and suppressed pro-inflammatory responses. However, immunity to the three different viruses resulted in unique pathologies in the VV-infected lungs, but with one common feature, the substitution of lymphocytic and chronic mononuclear infiltrates for the usual acute polymorphonuclear response seen in non-immune mice. Immunity to influenza A virus appeared to influence the outcome of subsequent acute infections with any of the three viruses, VV, LCMV and MCMV. Most notably, influenza A virus-immunity protected against VV but it actually enhanced LCMV and MCMV titers. This enhanced MCMV replication was associated with enhanced TH1 type response and pro-inflammatory cytokine responses. Immunity to influenza A virus appeared to dramatically enhance the mild lymphocytic and chronic mononuclear response usually observed during acute infection with either LCMV or MCMV in non-immune mice, but LCMV infection and MCM infection of influenza A virus-immune mice each had its own unique features. Thus, the specific sequence of virus infections controls the outcome of disease.

PSBE. A população elegível correspondeu a todos os trabalhadores da empresa. Esses participaram do exame odontológico periódico entre os anos estudados, podendo ter realizado um ou mais exames nesse período. Teve como objetivo avaliar sexo; idade; realização do primeiro exame; cargo ocupado, sendo os trabalhadores agrupadas em administrativo ou produção; e condição de urgência; além das condições de saúde bucal, analisando prevalência e severidade da doença periodontal, através do Índice Comunitário de Necessidade de Tratamento Periodontal em 1999 e do Índice Periodontal Comunitário e Perda de Inserção Periodontal nos outros anos; prevalência e localização de lesão de mucosa; uso e necessidade de prótese. Com base nos resultados, pôde-se notar que houve uma predominância de trabalhadores do sexo masculino e poucos apresentavam condição de urgência. A média de idade nos anos de 1999, 2002 e 2003 foi de 39 anos (Desvio Padrão ± 9). Ao se avaliar as condições de saúde bucal, em relação à doença periodontal, o sextante superior anterior foi o que apresentou maior percentual de saúde enquanto que o sextante inferior anterior foi o mais acometido por cálculo dental, variando de 63,5 a 74,7%. O percentual de trabalhadores sem alteração de mucosa variou de 70,6% a 90,7%, com o passar dos anos, sendo o palato o local mais acometido, e o tipo mais freqüente foi a lesão vermelha, que variou de 5,8% a 17,5%. Na arcada superior o percentual de trabalhadores que não faziam uso de prótese foi de 37,7% a 42,7%, já na arcada inferior este percentual variou de 45,4% a 53%, sendo a prótese unitária a mais usada em ambas arcadas, com exceção de 2000 que teve a parcial removível como a mais utilizada na arcada inferior. A prótese que os trabalhadores mais tinham necessidade era a fixa, em ambas as arcadas, variando de 16,6% a 28,4%. A partir da análise dos resultados pôde-se concluir que o percentual de trabalhadores que apresentaram condição de urgência foi muito baixo, com pequena variação ao longo dos anos; a doença periodontal permaneceu estável, estando controlada nos anos estudados; houve uma redução no percentual de indivíduos com alterações de mucosa; e uma grande quantidade de trabalhadores fazia uso e não tinha necessidade de prótese, provavelmente devido ao percentual elevado de cobertura assistencial. / Salvador

Isso posto, foi formulado o presente estudo com o fito principal de fazer a avaliação das respostas imunes humorais e celulares em diferentes intervalos pós-desafio com o VBI de aves previamente vacinadas ou não, realizando-se a mensuração de anticorpos no soro e na lágrima, e a quantificação da expressão de genes relacionados às respostas imunes na superfície traqueal, a fim de correlacionar tais parâmetros com o estado de proteção ao desafio. Os resultados demonstraram que os aumentos significativos nos níveis de anticorpos lacrimais dos isótipos IgG e IgA nas aves previamente vacinadas e também na expressão dos genes relacionados às respostas imunes, sobretudo o CD8, a Granzima A e o IFNg foram correlacionados negativamente com um ou mais parâmetros de alterações patológicas traqueais. Constatou-se também, que a memória das respostas imunes humorais e cito-mediadas conferida por uma única vacinação contra a BI no primeiro dia de idade é dependente da dose vacinal administrada / Avian infectious bronchitis virus (IB) is a worldwide infectious disease which causes significant economic losses in poultry industry. The innate and acquired immune responses, including whether there mediated by both cellular and humoral factors that are induced after infection or vaccination with IB virus (IBV) are characterized by their higher complexity and for the relevant aspects that are still poorly known, with respect to the elements able to exercise one or more actions against the pathogen and that culminate in the restriction of its propagation and also on your clearance of the host organism. So, this project was formulated with the main done to make the evaluation of cellular and humoral immune responses at different intervals post-immunization or postchallenge with IBV poultry previously vaccinated or not, performing the measurement of antibodies in serum or tears and quantitation of the expression of genes related to immune responses in tracheal surface, correlating these parameters with the protection against IBV. The results showed that both significative increase of IgG and IgA isotypes in tears of previously vaccinated chickens and the expression levels of genes related to immune responses, especially CD8, Granzyme A and IFN g were negatively correlated with one or more parameters of pathological lesions at trachea. Flagelina, a subunidade estrutural dos flagelos bacterianos, pode ser empregada em estratégias vacinais como carregadora de epitopos antigênicos fusionados ou como adjuvante. Neste trabalho avaliamos a imunogenicidade e o potencial vacinal das flagelinas Salmonella enterica sorovar Typhimurium em camundongos BALB/c após administração pelas vias oral ou intraperitoneal. Os resultados indicam que animais imunizados pela via oral com linhagens flageladas de S. Typhimurium não desenvolvem respostas de anticorpos, sistêmicos ou secretados, anti-flagelina, por outro lado, ativam respostas celulares específicas para as flagelinas em função da via de administração utilizada. Observamos ainda que as diferentes flagelinas testadas apresentam efeito adjuvante quando co-administradas pela via nasal com a fímbria CFA/I de ETEC. Em suma, os resultados apresentados contribuem para um melhor conhecimento sobre as propriedades imunológicas e adjuvantes das flagelinas de S. Typhimurium e agregam informações para o uso racional dessas proteínas em formulações vacinais. / Flagellin, the structural subunit of bacterial flagella, can be used in vaccine development as a fused antigenic epitope carrier or as an adjuvant. In this work we evaluated the immunogenicity and vaccine approach of S. Typhimurium flagellin in BALB/c mice after administration by oral or intraperitoneal route. Our results indicate that mice immunized by oral route with flagellated S. Typhimurium strains do not develop any anti-flagellin antibody response, be it serum or secreted. On the other hand, specific anti-flagellin cellular response was observed depending on the chosen route of immunization with S. Typhimurium attenuated strain. Furthermore, all flagellins tested proved to be efficient adjuvants when co-administrated with CFA/I fimbriae by nasal route. Together, these results contribute to a better understanding of the immunological and adjuvant properties of S. Typhimurium flagellins and also provide information on the rational use of these proteins in vaccine development.
