Diferentes condições podem alterar as características fisiológicas, metabólicas, nutricionais e morfológicas. Assim, o objetivo desse estudo foi analisar o efeito do treinamento físico, destreinamento e especialmente destreinamento combinado com a suplementação de leucina sobre o peso corporal, composição corporal, homeostase glicêmica e expressão e fosforilação da via de sinalização da proteína alvo da rapamicina em mamíferos (mTOR). Quarenta e seis ratos Wistar adultos machos foram divididos em 4 grupos: 31 ratos foram treinados em esteira por 8 semanas e 15 foram usados como controle (ratos sedentários). Destes, 15 ratos foram eutanasiados imediatamene após o treinamento (grupo treinado = 8 e grupo controle =7), 8 continuaram treinando, 16 foram mantidos sem exercícios (grupo destreinado = 8 e grupo destreinado suplementado com leucina = 7), e os 7 ratos restantes foram usados como controle (ratos sedentários) por mais 6 semanas antes de serem eutanasiados. Os animais do grupo destreinado foram alimentados com uma dieta contendo ~18% de proteínas, suplementados ou não com 5% de leucina. A composição corporal, o consumo alimentar, o volume de adipócitos, a glicose de jejum, o teste oral de tolerância à glicose (TOTG), o teste oral de tolerância à insulina (TTI), a atividade da citrato sintase, as concentrações séricas de leptina, adiponectina, aminoácidos e ácidos graxos não esterificados (AGNE) e a expressão e fosforilação da proteína mTOR foram avaliados. Os resultados de peso corporal, gordura corporal, coxins adiposos epididimal e retroperitoneal, volume de adipócitos, consumo alimentar, concentração sérica de leptina e TOTG foram significantemente menores nos animais treinados do que nos sedentários e destreinados (P < 0,05). Nenhuma diferença significante foi observada nos valores de glicemia de jejum, TTI e AGNE entre os três grupos (P > 0,05). Por outro lado, o grupo treinado apresentou valores significantemente mais altos para a atividade da citrato sintase do que os grupos sedentário e destreinado (P < 0,05). Nenhuma diferença significante (P > 0,05) foi identificada na comparação entre os grupos destreinados (com ou sem suplementação de leucina), incluindo a expressão e fosforilação da proteína mTOR. O exercício moderado exerce um efeito protetor sobre a composição corporal e a homeostase glicêmica, enquanto que a interrupção do treinamento físico teve um efeito negativo sobre essas variáveis. Os resultados sugerem que a suplementação crônica de leucina parece não reduzir os efeitos deletérios do destreinamento. / Different conditions may alter physiological, metabolic, nutritional, and morphological characteristics. Thus, the aim of this study was to analyze the effects of training, detraining, and especially detraining combined with leucine supplementation on body weight, body composition, glicemic homeostasis and expression and phosphorylation of mammalian target of rapamycin (mTOR) protein-signaling pathway in epididymal adipose tissue in rats. Fourty-seven male adult Wistar rats were divided into 4 groups: 32 rats were trained on a treadmill for 8 weeks and 15 were used as controls (untrained rats). Of these, 15 rats were euthanized immediately after training (trained group = 8 and control group = 7), 8 continued in training, 16 were caged without exercise (detrained group = 8 and detrained group plus leucine supplementation = 7), and the remaining 7 rats were used as controls (untrained rats) for more 6 weeks before being euthanized. Animals of the detrained groups were fed with a ~18% protein diet supplemented or not with 5% leucine. Body composition, food intake, adipocytes size, fasting glucose, oral glucose tolerance test (OGTT), insulin tolerance test (ITT), citrate synthase activity (CS), serum leptin, adiponectin, amino acids and non-esterified fatty acids (NEFA) concentrations and expression and phosphorylation of mammalian target of rapamycin (mTOR) protein were measured. Body weight, body fat, epididymal and retroperitoneal fats depots, adipocytes size, food intake, serum leptin concentration and OGTT were significantly lower in trained animals than in untrained and detrained (P < 0.05). No significant differences in the fasting glucose, ITT and NEFA among the three groups were observed (P > 0.05). On the other hand, the trained group showed a significantly higher score in the citrate synthase activity than both untrained and detrained groups (P < 0.05). No significant differences (P > 0.05) were identified in the comparisons between detrained groups (with and without leucine supplementation), including of the mTOR protein expression and phosphorylation. Moderate exercise exerted a protective effect on the body composition and glucose homeostasis while sudden discontinuation of physical training had a negative effect on such variables. Our results suggest that chronic leucine supplementation not seems to decrease the deleterious effects caused by detraining.

A hipertensão arterial cursa com hiperativação de neurônios glutamatérgicos (excitatórios) e depressão de neurônios gabaérgicos (inibitórios) em áreas centrais autonômicas. Avaliamos em ratos SHR e WKY os efeitos temporais do treinamento aeróbio (T) sobre a expressão/funcionalidade das vias gabaérgicas e glutamatérgicas no núcleo paraventricular do hipotálamo (PVN), núcleo do trato solitário (NTS) e bulbo ventrolateral rostral (RVLM), correlacionando-as a dados hemodinâmicos. SHRs apresentavam elevada PAM e FC e níveis elevados de RNAm de GAD67 no NTS. T promoveu bradicardia de repouso (T2 SHR e T8 WKY) e redução da PAM nos SHR (T8). O aumento de GAD em SHR e WKY induzido pelo T correlacionava-se com a redução da FC basal. T determinou aumento da razão inibição/excitação nos WKY e não a alterou nos SHR. Alterações nas expressões gênicas foram confirmadas por alterações similares na expressão protéica. Assim, o aumento da inibição gabaérgica essencialmente no PVN de WKY e SHR treinados é um fator determinante para a instalação da bradicardia de repouso. / Hypertension is accompanied by hyperactivity of glutamatergic (excitatory) and depression of gabaergic (inhibitory) neurons in autonomic areas driving cardiovascular control. Evaluated in SHR and WKY rats the time effects of T on cardiovascular parameters and on the expression/activity of gabaergic and glutamatergic pathways in the hypothalamic paraventricular nucleus (PVN), nucleus of solitary tract (NTS) and rostroventrolateral medulla (RVLM), correlating the hemodynamic data. SHRs exhibited elevated BP and HR and high GAD67 mRNA levels within the NTS. T caused resting bradycardia (T2 SHR and T8 WKY) and BP basal in SHR (T8). In both groups, T-induced elevated GAD expression was correlated with baseline HR reduction. T caused in the WKY augmentation of inhibitory/excitatory ratio, and did not change it in the SHR. Gene expression changes were confirmed by similar changes in protein expression. So, the increased gabaergic inhibition within the PVN of trained WKY and SHR is a main factor determining the appearance of resting bradycardia.

Influência do gene da enzima conversora de angiotensina sobre as respostas metabólicas induzida pelo treinamento físico aeróbio em camundongos diabéticos / Influence of angiotensin converting enzime gene on metabolic responses induced by aerobic physical training in diabetic mice

O objetivo do presente estudo foi avaliar os efeitos do treinamento físico (TF) sobre as alterações do metabolismo energético induzidas pelo diabetes em camundongos com 1 e 3 cópias do gene da enzima conversora de angiotensina (ECA). Para isso, camundongos machos C57BL/6 geneticamente modificados com 1 e 3 cópias do gene da ECA foram induzidos ao diabetes por estreptozotocina (STZ), e separados em 4 grupos de acordo com a realização ou não de TF com natação: 1 cópia sedentário (1S, n=14), 1 cópia treinado (1T, n=15), 3 cópias sedentário (3S, n=10), e 3 cópias treinado (3T, n=11). Os principais resultados foram: não houve diferença no ganho de peso corporal, porém o peso do depósito de gordura retroperitoneal e diâmetro de adipócito foram menores no grupo 3T comparado ao 1S, 1T e 3S; menor consumo de ração diária do 1S comparado ao 1T, 3S e 3T; maior consumo de oxigênio de repouso do 3S comparado ao 1T e 3T; melhor capacidade de realizar exercício físico e menor glicemia de jejum do 1T comparado ao 1S e 3S (1T=305±34 vs. 1S=479±47 e 3S=496±49 mg/dl); maior concentração de leptina no 1T comparado ao 1S (1T=5,65±0,33 vs 1S=4,07±0,43 ng/ml) xi e maior atividade lipolítica no 1T comparada aos grupos 1S, 3S e 3T (1T=288±50 nmol/106cels/h vs. 1S=79±37, 3S=123±37 e 3T=50±20 nmol/106cels/h); Não houve diferença estatística entre os grupos na tolerância à glicose, na atividade da enzima lipogênica ácido graxo sintase, na atividade da enzima citrato sintase, na tipagem de fibras e na razão capilar/fibra muscular do músculo sóleo. Esses resultados demonstraram que as adaptações metabólicas promovidas pelo TF no diabetes são parcialmente associadas ao genótipo da ECA / The main purpose of this study was to evaluate the effects of physical training (PT) on changes in energy metabolism induced by diabetes in mice harboring 1 and 3 copies of angiotensin converting enzyme (ACE) gene. For this, transgenic adult male mice (C57BL/6) with 1 or 3 copies of ACE gene were induced to diabetes by Streptozotocin (STZ) and separated into groups sedentary or physical trained with swimming: 1 copy sedentary (1S, n = 14), 1 copy trained (1T, n = 15), 3 copies sedentary (3S, n = 10) and 3 copies trained (3T, n = 11). The main results were: body weight gain was not different among groups, but 3T group reduced retroperitoneal fat pad and adipocyte diameter compared to 1S, 1T and 3S groups; daily food intake was lower in 1S compared to 1T, 3S and 3T groups; resting oxygen uptake was higher in 3S compared to 1T and 3T; physical exercise ability increased and fasting glucose decreased only in 1T group compared to 1S and 3S (1T=305±34 vs. 1S=479±47 e 3S=496±49 mg/dl); serum leptin increased in 1T compared to 1S (1T=5,65±0,33 vs 1S=4,07±0,43 ng/ml), and lipolytic activity was higher in 1T compared to 1S, 3S and 3T groups (1T=288±50 nmol/106cels/h vs. 1S=79±37, 3S=123±37 e 3T=50±20 nmol/106cels/h). There were no statistical differences among groups in glucose tolerance, lipogenic activity of fat acid synthase, activity of citrate synthase enzyme, fibre type composition and capillary-to-fibre ratio of soleous muscle. These results demonstrated that metabolic adaptations promoted by PT in diabetes are partially associated with ACE genotype

Notre société occidentale fait face actuellement à une hausse de l’espérance de vie, ce qui amène à un vieillissement de la population. Malgré les avancées médicales, il s’opère une dégradation des fonctions cognitives et des capacités physiques, responsable d’une baisse de la qualité de vie et de la perte d’autonomie. De plus, le nombre de pathologies dégénératives, dont le premier facteur de risque est l’âge, ne cesse d’augmenter. A l’heure actuelle, il n’existe encore aucun moyen pharmacologique permettant de contrer efficacement le déclin cognitif et physique lié à l’âge. Ainsi, l’objectif de ce travail de thèse était de tester l’impact de méthodes non-pharmacologiques sur la cognition, particulièrement au niveau de la mémoire de travail et des fonctions exécutives, la qualité de vie, et l’autonomie. Nous nous sommes concentrés sur les entraînements cognitifs et physiques, en émettant l’hypothèse principale qu’une combinaison de ces deux sphères au sein d’un même entraînement serait plus efficace qu’un entraînement seul (i.e. cognitif ou physique) dans la mesure où ils engendreraient des bénéfices différents mais complémentaires. Ce travail de thèse s’organise donc autour de trois études : l’Etude 1, qui a pour but d’investiguer l’impact d’un entraînement cognitif simple comparé à un entraînement combiné cognitif-et-physique sur la cognition ; l’Etude 2, qui compare l’impact de ces deux mêmes entraînements mais cette fois à un niveau fonctionnel cérébral ; l’Etude 3 qui investigue l’impact d’une prise en charge multidimensionnelle dans la maladie d’Alzheimer. Les résultats de ce travail de thèse nous montrent, en ce qui concerne le vieillissement normal, des bénéfices des deux types d’entraînement au niveau comportemental, en termes de transfert et de maintien à long-terme. En revanche, si l’entraînement combiné a engendré un transfert plus important, l’entraînement cognitif seul produit un meilleur maintien des bénéfices dans le temps. Par rapport au niveau fonctionnel cérébral, des bénéfices ont été montrés sur la composante P100, qui reflète le recrutement attentionnel uniquement pour le groupe d’entraînement cognitif seul. Ensuite, nous avons observé des bénéfices dans le vieillissement pathologique au niveau de la cognition. Pour conclure, ce travail de thèse nous a permis de montrer des effets bénéfiques des entraînements cognitifs et/ou physiques dans le vieillissement normal et pathologique, même si à l’heure actuelle nous ne pouvons pas conclure par une supériorité de l’entraînement combiné comparé à un entraînement cognitif. / Our western society is currently facing an increase in life expectancy, which is leading to a growing aging population. Despite medical advances, there is a deterioration of cognitive functions and physical abilities, which is responsible for a decline in life quality and a loss of autonomy. In addition, the number of neurodegenerative diseases, whose first risk factor is age, is increasing. Today, there is no pharmacological means to effectively counter agerelated cognitive and physical decline. Thus, the objective of this thesis was to test the impact of non-pharmacological methods on cognition, particularly on working memory and executive functions, quality of life, and autonomy. We focused on cognitive and physical training, with the main hypothesis that a combination of these two aspects within the same training would be more effective than a single workout (ie cognitive or physical) insofar as they would generate different but complementary benefits. This thesis work is organized around three studies : Study 1, which aims to investigate the impact of a simple cognitive training compared to a combined cognitive-and-physical training on cognition ; Study 2, which compares the impact of these two same trainings but this time at a cerebral functional level ; Study 3 which investigates the impact of multidimensional intervention in Alzheimer's disease. The results of this thesis work showed, with regard to normal aging, the benefits of both types of training at the behavioral level, in terms of transfer and long-term maintenance. On the other hand, if combined training has resulted in greater transfer, cognitive training alone produces better benefits maintenance over time. Relative to brain function level, benefits were shown on the P100 component, which reflects attentional recruitment solely for the cognitive training group alone. Then, we observed benefits in pathological aging at the level of cognition. To conclude, this thesis work allowed us to show the beneficial effects of cognitive and / or physical training in normal and pathological aging, even if at present we cannot conclude for the superiority of combined training compared to cognitive training.

Ett sätt att få barn, både överviktiga och icke överviktiga, att bli mer aktiva är att engagera dem på lektionerna i idrott och hälsa i skolan. Syftet med denna studie var att ta reda på om det sker något förebyggande arbete för de eleverna med övervikt, på idrottslektionerna. Data samlades in genom att skicka ut enkäter till rektorer, på grundskolor i Skåne, med uppmaning att de skulle vidarebefordra enkäten till idrottslärarna i år 3 och år 5. Sammanlagt deltog 57 idrottslärare i studien. Resultatet visade att 43 respondenter anpassar sina lektioner i ämnet idrott och hälsa så att elever med övervikt kan vara med i samma utsträckning som övriga elever. 28 svarar att de försöker motverka övervikt bland sina elever. Utifrån vad resultatet visade engagerar sig de flesta lärarna i elevernas övervikt, vilket är bra. Men det kan bli bättre!</p> / <p>The reports about overweight among children are increasing by day and it has developed into a large health problem in Sweden as well as in the rest of the western world. One way to inspire children to be more physically active is to stimulate them at the physical education classes in school. The aim of this study was to find out if the physical education teachers are working actively to prevent weight gain in overweight children. Data was collected through a questionnaire sent to headmasters at primary schools in Skåne, Sweden. The respondents were physical education teachers working with children in grade three and five. A total of 57 teachers participated in the study. The result was that 43 respondents adapt their lessons to make overweight children as involved as not overweight children. 28 teachers reported that they actively try to prevent overweight. The result shows that most of these teachers are committed to preventing overweight among children.</p>

Fysisk aktivitet och hälsa : Leder fysisk aktivitet i form av träning till en bättre självupplevd hälsa i både arbetslivet och på fritiden?

Bakgrund: Sambandet mellan fysisk aktivitet och psykisk hälsa har under senare åren studerats allt oftare.  Fysisk aktivitet kan förebygga olika sjukdomstillstånd och många rapporter stöder att motion är bra för människan. För att kroppen ska fungera optimalt behövs fysisk aktivitet. Stillasittande yrken och dålig motion är vardag för många. Folk verkar bli ohälsosammare tidigare och fysisk aktivitet skulle kunna vara en del av en lösning i en hälsosammare livsstil. Det finns ett samband med träning och bättre hälsa och förmåga . Enligt min frågeställning tror jag att en regelbunden träning ger en friskare individ i sin vardagliga sysselsättning (arbete/studier). Fler studier på området behövs för att upplysa människor om de förbättringar man själv kan göra för sin hälsa och sitt inre välmående. Syfte: Studiens syfte var att undersöka om den självupplevda effekten av fysisk aktivitet är positiv i form av konkreta hälsoförbättringar.  Metod: Jag har använt en kvalitativ metod med semistrutkurerade intervjuer. Innehålls analys har använts för att analysera det manifesta innehållet i texten. Resultat: Samtliga av testpersonerna beskriver konkreta hälsoförbättringar i både den fysiska vardagen och den psykiska vardagen. Stressnivåerna hade minskat, ryggont hade minimerats och välmåendet hade förbättrats hos samtliga. Glädjen att arbeta och umgås hade höjts och flera av testpersonerna nämnde blodsockernivåerna som en viktig faktor som hade förbättrats. Diskussion: För att behålla en bra hälsa i arbetslivet och på fritiden behövs regelbunden fysisk aktivitet. Konklusion: Fysisk aktivitet borde lyftas fram ännu mer och användas mer i preventionssyfte än vad man gör idag. En bättre fysisk förmåga ger helt enkelt en friskare vardag. / Background: The relationship between physical activity and mental health has in recent years been studied more frequently. Physical activity can help prevent various diseases and many reports supports that exercise is good. For the body to function optimally physical activity is needed. Sedentary occupations and poor exercise is for many a lifestyle. People seem to be more unhealthy . Physical activity could be part of the solution to a healthier lifestyle. There is an connection between exercise and better health and ability. In my question, I think that a regular exercise provides a healthier individual in their everyday jobs (work / studies). More studies in this area is needed to enlighten people about the improvements you can do yourself to their health and inner wellbeing. Objective: Purpose of the study was to investigate whether the self-perceived effect of physical activity is positive in terms of concrete improvements in health. Method: The content analysis was used to analyze the manifest content of the text. Results: All of the participants describe specific health improvements in both the physical and mental everyday everyday life. Stress levels had decreased pain, back pain was minimized and well-being had improved. The joy of working and socializing had been increased and several of the participants mentioned their blood sugar levels was key factor that had improved. Discussion: In order to maintain a good health at work and during leisure regular physical activity is needed. Conclusions: Physical activity should be promoted more and used more in prevention purposes than they do today. A better physical ability simply provides a healthier living. / <p>2013-06-04</p>

Magistriniame darbe nagrinėjama Visagino savivaldybės kūno kultūros ir sporto sistema, kaip Lietuvos sistemos grandis, analizuojamos Visagino kūno kultūros ir sporto sistemos raidos kryptys, problemos ir modernizavimo galimybės. Literatūros analizė parodė, kad kol kas mažai teorinio įdirbio kūno kultūros ir sporto sistemos funkcionavimo srityje. Daugelis sistemos dalių yra iškelta nacionaliniu lygiu ir daug mažiau dėmesio kreipiama į savivaldos lygį. Todėl nėra susiformavęs inovatyvus požiūris į kūno kultūros ir sporto sistemos funkcionavimą valstybinėse institucijose ir ypatingai savivaldybėse. Tai aktuali problema, nes valstybė neturi tiesioginio sąlyčio su visuomene ir negali tiesiogiai vykdyti savo kūno kultūros ir sporto politikos. [toliau žr. visą tekstą] / This Master thesis is devoted to the analysis of Visaginas City Municipality physical education and sports system as a segment of Lithuania’s sports system, the trends of Visaginas City physical education and sports development, problems and modernization possibilities. The analysis of literature sources revealed that there is little theoretical knowledge applied in the functioning of physical education and sports system. Most segments of the system are relevant on the national level, thus, less attention is paid to self-government level. Therefore, there is no innovative attitude developed towards the functioning of physical education and sports system in state institutions, especially in municipalities. This is an acute problem because the state does not have a direct contact with society and cannot directly implement physical education and sports policy. Physical education and sports system is the unity of specific segments connected to an individual’s physical nature, i.e. physical education, preparation for physical load and work activities, active physical activities and free time organization, development and possibilities of functional reserves of human organism as well as a more specific direction – the best possible result in sports and/or attempt for physical perfection. Motion interactive games for children with motor disorders : motivation, physical activity, and motor control / Rörelsestyrda spel i träning av barn med motoriska nedsättningar : motivation, fysisk aktivitet och motorisk kontroll

As motion interactive games have become more widespread the interest in using these games in rehabilitation of children with motor disorders has increased among both clinical professionals and the families of these children. The general aim of this thesis was to evaluate the feasibility of using interactive games in rehabilitation of children to promote motivation for practice, physical activity, and motor control. A systematic review of published intervention studies was conducted to obtain an overview of existing research and the current levels of evidence for using interactive games in motor rehabilitation of children. Sixteen studies met the inclusion criteria, out of these three were randomized controlled trials while half were case series or case reports. Thirteen studies presented positive findings, which indicated a promising potential. However, more convincing research is needed. Commercially available motion interactive games have only been used in a few studies on motor control, and in none of these home based practice was provided. Moreover, no earlier studies have evaluated if these games may increase motivation for training and daily physical activity among children with disabilities. To address these issues a feasibility intervention including 15 children in the ages 6-16 years and with mild to moderate cerebral palsy was conducted. Each child was provided with a Sony PlayStation2â and the EyeToyâ games in Play3, and was recommended to practice with the provided games for at least 20 minutes/day during four weeks. The intervention was evaluated with gaming diaries, physical activity monitors (SenseWear Armband), interviews with the parents, and the clinical motor tests Movement Assessment Battery for Children-2 (mABC-2), Bruininks-Oseretsky Test of Motor Proficiency subtest 5:6, and the 1 Minute Walk Test. In addition, 3D motion analysis was used to evaluate effects on quality of goal-directed arm movements towards virtual and real objects, respectively. Motivation for practice and compliance of training were high, although declining somewhat during the course of the four weeks. The children’s physical activity increased significantly during the intervention. However, four children were excluded from this analysis due to lack of complete data from the physical activity monitors. According to mABC-2 the children’s motor performance improved, but there were both floor and ceiling effects, indicating a low sensibility of this test. The two additional motor tests showed only non-significant progress. Results from the 3D motion analysis suggest that the children improved movement precision when playing the games, movement smoothness when reaching for real objects, and used a more economic reaching strategy with less trunk involvement. In the interviews the parents expressed the view that motion interactive games promote positive experiences of physical training and add elements of social interaction to the training. They also experienced less urge to take on a coaching role. The training provided by the games was considered unspecific and there was a desire for individualized games to better address the unique rehabilitative need of each child. In conclusion, it is feasible to use motion interactive games in home rehabilitation for children with cerebral palsy to promote short term motivation for practice and general physical training. Specific effects on motor control need to be further explored and there is also a need for reliable tests that are adequate and sensitive enough to capture changes in movement control. In future development of interactive games for rehabilitation purposes, it is a challenge to preserve the motivational and social features of games while at the same time optimizing an individualized physical training.
