Doing food-knowing food : an exploration of allotment practices and the production of knowledge through visceral engagement

The original contribution of this thesis is through its conceptualisations of human more-than- human encounters on the allotment that break down the boundaries of subjectivities. This work extends knowledge of cultural food geography by investigating how people engage with the matter of the plot and learn to grow food. The conceptual tool by which this occurs is set out as processes of visceral learning within a framework of mattering. Therefore this work follows the material transformations of matter across production consumption cycles of allotment produce. This is examined through processes of bodily adaptions to the matter of the plot. The processes of growing your own food affords an opportunity to focus on the processes of doing and becoming, allowing the how of food growing to take centre stage (Crouch 2003, Ingold 2010, Grosz 1999). Procuring and producing food for consumption is enacted through the human more-than-human interface of bodily engagement that disrupts dualisms and revealing their complex inter-relationships, as well as the potential of visceral research (Roe 2006, Whatmore 2006, Hayes-Conroy 2008). Therefore, this is an immersive account of the procurement of food and the development of food knowledge through material, sensory and visceral becomings, which occur within a contextual frame of everyday food experiences. This study is contextualised in the complexities of contemporary food issues where matters of access, foodism and sustainability shape the enquiry. However the research is carried out at a micro-geographies lens of bodily engagements with food matter through grow your own practices on allotments. Growing food on new allotments is the locus of procurement reflecting a resurgence in such activities following from the recent rise in interest in local food, alternative food networks (AFNs) and food as a conduit for celebrity in the media (Dupuis & Goodman 2005, Lockie & Kitto 2000, Winter 2003). Moreover, the current spread of the allotment is examined as transgressing urban/rural divides and disrupting traditional perceptions of plot users. This allows investigations into spaces where community processes can unfold, providing a richly observed insight into the broadened demographics of recent allotment life.

Using Your Imagination to Pursue Goals: Diminishing the Effects of Visceral Temptations

Consumers consistently set goals for themselves. Despite good intentions, consumers often deviate from their goals. If consumers understand the benefits that arise from goal success, then why do most consumers fail to accomplish goals? Often, temptations are more appealing than achievement of goals; temptations are tangible while the benefits of a goal are difficult to grasp. An individual who uses his/her imagination to visualize goal success makes the goal more present-minded and attainable (Oettingen 2000). Thus, imagination facilitates self-efficacy, the belief in one’s ability to reach a goal. Higher self-efficacy, then, provides an individual with the willpower to achieve a goal (Taylor, Pham, Rivkin, and Armor 1998). Whereas previous work has examined temptations’ relationship with goals (e.g. Fedorikhin and Patrick 2010; Wilcox, Vallen, Block, and Fitzsimons 2009; Zhang, Huang, and Broniarczyk 2010; etc.), the scope of this dissertation study differs. Rather, the research aim is to identify how consumers can overcome visceral temptations. Thus, the main objectives include: contributing new perspectives on goal research by merging the literatures on imagination and visceral cues, outlining how imagination regulates the impact of visceral temptations, and identifying the underlying mechanism that explains how imagination regulates the relationship between visceral cues and ad-evoked thoughts, through self-efficacy.

Para isso, discutimos a LV no Brasil enquanto um problema de saúde pública a partir da distribuição de casos no Planalto Ocidental Paulista. O objetivo desta tese é definir e analisar os padrões espaciais e epidemiológicos da LV na escala regional e local, tendo como recorte empírico os municípios de Araçatuba, Presidente Prudente e Votuporanga, no estado de São Paulo, Brasil. Partimos da leitura da paisagem epidemiológica da LV e da descrição da narrativa geográfica dos serviços de saúde para a elaboração de um raciocínio geográfico de maior complexidade, com base em diferentes conceitos da Geografia, tais como: paisagem, espaço, sítio-situação e escala. Utilizamos a análise espacial, com ferramentas dos Sistemas de Informação Geográfica (SIG) e modelagem espacial, os quais permitiram delinear dois padrões distintos da LV no estado de São Paulo: a região do Planalto Ocidental Paulista e a região Inerte, onde a primeira possui maior intensidade de casos da doença e é definida, inicialmente, por variáveis ambientais, embora ações humanas influenciem em sua dispersão. We discussed VL as a public health problem in Brazil, analyzing the distribution of cases in the Paulista Western Plateau region. This thesis aims to define and analyze the spatial and epidemiological patterns of VL at the regional and local scale, focusing on the municipalities of Araçatuba, Presidente Prudente, and Votuporanga, all of which are located in the Brazilian state of São Paulo, Brazil. We present the epidemiological landscape of VL and the description of the geographical narrative of public health services aiming to achieve a more complex geographic reasoning based on different concepts of Geography, such as landscape, site-situation, and scale. We employed spatial analysis, for instance Geographic Information System (GIS) and spatial modeling tools, which allowed us to identify two VL patterns in the state of São Paulo: the region of the Paulista Western Plateau and the Inert region, where the first has the highest intensity of cases, and it is, initially, defined by environmental factors, although human actions influence the disease cases spreading. Para a averiguação da possibilidade de confirmação da inconstitucionalidade, na prática da eutanásia de animais, (principalmente quando se tem no Brasil um país endêmico na doença), foi necessário confrontar essa inconstitucionalidade. O confronto se deu com os direitos fundamentais contidos na Constituição Federal de 1988, assim como na Declaração Universal dos Direitos dos Animais e Lei n. 9.605/1998 (Lei de Crimes Ambientais), principalmente porque o ato de eutanásia vem respaldado pelo Decreto n. 51.838, de 14 de março de 1963, que deste originou a Portaria Interministerial n. 1.426, de 11 de julho de 2008, do Ministério da Agricultura, Pecuária e Abastecimento e Ministério da Saúde, onde esta determina a proibição do tratamento de animais acometidos pela leishmaniose visceral e aquela a eutanásia dos mesmos. Não bastando, tais normas também possuem conteúdo que cerceia a autonomia da prática profissional do médico veterinário. No aspecto processual, a inconstitucionalidade da eutanásia de animais vêm sendo reconhecida, na menção disposta em algumas decisões judiciais, que quando não expressam claramente a fala do magistrando demonstrando a inconstitucionalidade, a tem pelo como prática ilegal. Para os aspectos processuais foram apresentadas algumas decisões do Tribunais brasileiros, teorias inconstitucionais utilizadas nas teses de contestação, dos entes públicos, na tentativa de justificar a prática da eutanásia de animais tratáveis, como algo legal, também as manifestações processuais do MPE e MPF sobre o tema. Assim, foi possível visualizar e constatar a inconstitucionalidade da eutanásia face os prejuízos causados aos animais e população (como detentora do direito a um meio ambiente saudável e ecológicamente equilibrado, livre de maus tratos e crueldade a sua fauna).

Fatores de risco e análise espacial para a leishmaniose visceral no município de Juazeiro - Bahia - Brasil /

Orientador: Mary Marcondes / Banca: Rosemeri de Oliveira Vasconcelos / Banca: Raimundo Souza Lopes / Banca: Estevam Guilherme Lux Hoppe / Banca: José Eduardo Tolezano / Resumo: O objetivo do presente estudo foi determinar a presença de anticorpos anti-Leishmania sp. no soro e de DNA de Leishmania infantum chagasi no sangue periférico das diversas espécies de animais domésticos, residentes em um raio de 250 metros de distância de seres humanos portadores de leishmaniose visceral (LV), bem como avaliar as características ambientais, em uma região da cidade de Juazeiro (BA), na tentativa de determinar os possíveis fatores de risco para a ocorrência da doença. Para tanto, após a seleção de 21 casos humanos de LV distribuídos em seis bairros do município, realizou-se a sorologia e a reação em cadeia de polimerase em tempo real (qPCR) de soro e sangue total, respectivamente, de 498 animais, sendo 383 (76,91%) da espécie canina, 84 (16,87%) felina, 15 (3,00%) caprina, 12 (2,42%) equídea e 4 (0,80%) ovina. A prevalência de LV, considerando-se os resultados sorológicos e da qPCR, foi de 6,43% (32/498), sendo 5,75% (22/383) na população de cães e 11,90% (10/84) na de gatos. Vinte cães (5,22%) e 10 (11,90%) gatos foram sororreagentes e as outras espécies não apresentaram anticorpos anti-Leishmania sp. Apenas dois (0,40%) dos 498 animais apresentaram DNA do parasita no sangue, ambos da espécie canina e sem a presença de anticorpos séricos anti-Leishmania sp. Os gatos apresentaram duas vezes mais chances de se tornarem portadores da LV que os cães (OR=2,21; IC=1,1-4,87; p=0,043), e os felinos domiciliados apresentaram um risco 9,57 vezes maior de se infectarem que os gatos semidomiciliados (OR=9,57; IC=2,21-41,37; p=0,0006). O acesso a rua não foi associado com a doença em cães, e o sexo e a faixa etária não constituíram um risco para os animais contraírem esta zoonose. A presença de mais de um animal por domicílio aumentou em 3,46 vezes as chances de infecção na população... (Resumo completo, clicar acesso eletrônico abaixo) / Abstract: The aim of the present study was to determine the presence of antibodies anti-Leishmania sp. in serum and DNA of Leishmania infantum chagasi in peripheral blood of domestic animals living within a 250 m perimeter from human cases of visceral leishmaniasis (VL), as well as to evaluate the environmental characteristics, in the region of the city of Juazeiro, Bahia, in an attempt to determine possible risk factors for the occurrence of the disease. For this, after the selection of 21 human cases of VL distributed in six neighborhoods of Juazeiro, serology and real time polymerase chain reaction (qPCR) in whole blood were performed in 498 animals, being 383 (76.91%) dogs, 84 (16.87%) cats, 15 (3%) goats, 12 (2.42%) equids, and 4 (0.8%) sheep. The prevalence of VL, considering serological and qPCR results, was 6.43% (32/498), being 5.75% (22/383) dogs and 11.9% (10/84) cats. Twenty (5.22%) dogs and 10 (11.9%) cats were seroreactive and the others species did not present antibodies anti-Leishmania sp. Only two (0.40%) out of 498 animals presented the parasite DNA in blood, both dogs without the presence of anti- Leishmania antibodies. Cats were two times more likely to have VL than dogs (OR = 2.21; 95% CI, 1.1 - 4.87; p = 0.043), and the domiciled cats were more likely to be positive for VL than semi-domiciled (OR=9.57; 95% CI, 2.21 - 41.37; p=0.0006). Access to the street was not associated to the disease in dogs, and sex and gender did not constitute a risk factor for the development of VL. The presence of more than one animal per household increased in 3.46 times the odds of infection in the population (OR=3,46; IC=1,49-8,06; p=0,0025). Um total de 41 cães portadores de leishmaniose visceral, foram divididos em dois grupos. O primeiro composto por cães sem sintomas neurológicos (n=31) e o segundo grupo composto por cães com sintomas neurológicos (n=10). Amostras de encéfalo foram coletadas e armazenadas em formalina tamponada, para realização de imunoistoquímica para a pesquisa de formas amastigotas da Leishmania sp., linfócitos T CD3+, CD4+ e CD8+ e macrófagos. Não foram observadas formas amastigotas de Leishmania sp. Linfócitos T CD3+ estavam presentes em 24/31 (77,4%) dos cães sem sintomas neurológicos e em todos os cães do segundo grupo (p=0,0011). Linfócitos T CD4+ e CD8+ raramente foram observados, apresentando imunomarcação para CD4+ em 10/41 (24,3%) dos cães e em metade dos animais do grupo neurológico (p=0,0090). A presença de CD8+ foi detectada em 4/10 (40%) dos cães com doenças neurológicas (p=0,0021). Macrófagos foram observados em 38/41 (92,6%) cães, sem diferença estatística significante entre os dois grupos (p= 0,7664) / Abstract: Visceral leishmaniasis in dogs is described as a chronic disease whose main symptoms are progressive weigth loss, dermatologic lesions and, recently, the disease has been associated to neurologic disorders in dogs. A total of 41 Leishmania chagasi infected dogs were divided into two groups. The first composed of dogs without neurological signs (n=31) and the second by dogs with neurological disorders (n=10). Brain samples were collected, stored in 10% buffered formalin and subjected to immunohistochemical examination for amastigotes forms of Leishmania sp, CD3+, CD4+ and CD8+ T lymphocytes and macrophages. Imunnohistochemistry not revealed amastigote any forms of Leishmania sp. CD3+ T lymphocytes were present in 24/31 (77,4%) dogs without neurological signs and in all dogs from the second group (p=0.0011). CD4+ and CD8+ were rarely observed, with CD4+ immunostaining in 10/41 (24,3%) dogs, from which half of them had neurological disease (p=0.0090). The presence of CD8+ was detected only in 4/10 (40%) dogs from neurological group (p=0.0021). Orientador: José Luiz Laus / Banca: Márcia Rita Fernandes Machado / Banca: Leucio Camara Alves / Resumo: Avaliaram-se as alterações oftálmicas, protéicas do humor aquoso, sorológicas e histopatológicas em cães infectados experimentalmente com Leishmania (Leishmania) chagasi por via conjuntival. Selecionaram-se dez cães saudáveis após exame clínico, prova sorológica de ELISA para Leishmania sp e microscopia de luz de esfregaços de medula óssea. Dois grupos de cães compuseram o estudo. O Grupo Inoculado (GI) foi constituído por sete animais, que receberam 150æL de uma solução de cultura de Leishmania (Leishmania) chagasi por via conjuntival, e o Grupo Controle (GC) por três animais, inoculados com solução salina fisiológica, pela mesma via. Os cães foram monitorados quanto às alterações oculares às 2, 4, 6, 12 e 24h após a instilação. Em seguida, diariamente por uma semana, e a partir daí, uma vez por semana, até que se completassem os 60 dias pós-infecção. Amostras de sangue e de humor aquoso de ambos os olhos e esfregaços de medula óssea foram coletados aos 60 dias, para sorologia, análise protéica e parasitologia, respectivamente. Todos os animais do GI mostraram sinais oculares unilaterais ou bilaterais, particularmente opacidades corneais. Não somente os resultados dos anticorpos anti-Leishmania sp no soro e no humor aquoso, como também a parasitologia, resultaram negativos. A histopatologia dos olhos de cães do grupo infectado por Leishmania revelou resposta inflamatória caracterizada por infiltração de macrófagos, linfócitos e células plasmáticas, notadamente na terceira pálpebra, na conjuntiva e na glândula lacrimal. Edema, congestão e perivasculite difusa envolvendo o trato uveal, igualmente foram observados. / Abstract: The aim of this study was to evaluate the ocular signs protein of aqueous humour, serological and structural changes in the dogs' eyes infected experimentally by the conjunctival route of exposure with Leishmania (Leishmania) chagasi. A total of 10 healthy dogs were submitted to clinical examination, assayed by the ELISA test for anti- Leishmania sp antibodies and also microscopic examination of bone marrow smears. Two groups of dogs were studied. The first, Inoculated Group (IG) consisted of seven animal which one received a 150æL of Leishmania (Leishmania) chagasi suspension by the conjunctival route and the Control Group (CG) consisted of three animals inoculated by the saline physiological solution at the same route. The dogs were ophthalmology monitored at 2, 4, 6, 12 e 24 hours pos instilation. After this period, they were monitored once a day for one week and then, once a week until 60 days were complete. Blood samples, aqueous humor of both eyes and bone marrow smears were collected at 60 days for serology , protein of aqueous humour analyses and parasitological examination. All animals from IG showed unilateral or bilateral ocular signs, particularly corneal opacification. Not only the results of anti- Leishmania sp antibodies in sera and aqueous humor but also the parasitological examinations were negative. The histopathologic evaluation of eyes in the Leishmaniainfected dogs revealed inflammatory response characterized by macrophages infiltration, lymphocytes and plasma cells particularly on the third eyelid, the conjunctive tissue and lacrimal gland. Differentiation of extraembryonic endoderm stem cell lines and parietal endoderm into visceral endoderm : the art of XEN cells

The extraembryonic endoderm of mammals is essential for nutritive support of the foetus and patterning of the early embryo. Visceral and parietal endoderm are major subtypes of this lineage with the former exhibiting most, if not all, of the embryonic patterning properties. Extraembryonic endoderm (XEN) cell lines derived from the primitive endoderm of mouse blastocysts represent a cell culture model of this lineage, but are biased towards parietal endoderm in culture and in chimaeras. Here, I further characterise XEN cells and show that these cell lines exhibit high levels of heterogeneity. In an effort for XEN cells to adopt visceral endoderm character different aspects of the in vivo environment were mimicked. I found that BMP4 and laminin promote a mesenchymal-to-epithelial transition of XEN cells with upregulation of epithelial markers and downregulation of mesenchymal markers. Gene expression analysis showed the differentiated XEN cells most resembled extraembryonic visceral endoderm. Correspondingly, inhibition of Erk and BMP signalling drives XEN cells toward parietal endoderm fate. Finally, I show that BMP4 treatment of freshly isolated parietal endoderm from Reichert’s membrane promotes its visceral endoderm differentiation. This suggests that parietal endoderm is still developmentally plastic and can be transdifferentiated to a visceral endoderm in response to BMP. Generation of visceral endoderm from XEN cells uncovers the true potential of these blastocyst-derived cells and is a significant step towards modelling early developmental events ex vivo.
