Teresinha Lermen Donatti 04 August 2009 (has links)
The aim of the present study was to evaluate the final height, bone mineral density (BMD) and bone mineralization markers of adults with steroid responsive Idiopathic Nephrotic Syndrome (NS) in childhood and adolescence and to examine the influence of the disease, its co-morbidities and the patients\' target height in the final height and mineralization results. Patients and Methods: We have analyzed initial and final anthropometric data of 60 patients (41 male and 19 females) and / or their records, with a minimum age of nineteen years or fully developed pubertal status (P4G4 in males and menarche in females). BMD (g/cm2) was evaluated in 26 patients and in 35 controls, with a concomitant analysis, of serum levels of 25-OH Vitamin D (25(OH)D), Parathyroid Hormone (PTH); C-terminal telopeptide of type I collagen (CTx) and aminoterminal propeptide of type 1 procollagen (P1NP) and Osteocalcin (OC) Results: Mean age at first consultation was 5.3 years (SD: 2.4 yrs) and at last consultation was 20.4 yrs (SD: 3.0 yrs). The mean cumulative dose of prednisone was 1254 mg/kg (SD: 831.39 mg/kg). The mean initial height SDS was -0.60; (SD: 1.0) the final height SDS was -0.64; (SD: 0.92), (t-test: p=0.72). The final height SDS showed correlated significantly only with the initial height SDS and the target height SDS. Six patients achieved a final height SDS <-2 and this finding showed a strong correlation to the initial height SDS and to the target height SDS in the male patients. The patients\' and control subjects L1L4 head of the femur and the total femur BMD and BMD SDS did not differ significantly. 6 patients and 2 control subjects showed a BMD SDS <-2 (low bone mass) while 2 patients and 1 control subjects showed a BMD SDS <-2.5 (osteoporosis). Patients with BMD SDS <-2 received 2189 mg / kg of prednisone over 13 years while those with a BMD SDS <-2.5 received 2510 mg / kg prednisone for 14 years (p = 0.01 vs BMD SDS -2 ). No other studied variable correlated significantly with BMD. The studied bone biomarkers showed similar results in patients and control subjects without a significant correlation with disease activity, co-morbidities, and BMD or height parameters. Conclusion: 1. the initial and final height SDS were strongly correlated to the height target. 2. INS and its co-morbidities did not prevent the patients to reach their target height 3. The patients\' BMD and bone mineralization markers did not differ when compared to controls. 4. The 6 patients with low bone mass (2 with osteoporosis) used a total dose of prednisone for a longer period of time in relation to those with an adequate BMD 5. There was no correlation between bone mineralization markers, disease activity and its co-morbidities, final height and BMD of adult patients with INS in childhood and adolescence

Chan, Thomas 07 1900 (has links)
Osteoarthritis (OA) is a disease affecting joints and it has a very strong incidence in the population. It is characterized by articular cartilage degradation, an abnormal bone remodelling cycle and subchondral bone sclerosis. The aetiology of this disease is still unknown although OA is now considered as a joint disease involving all the tissues of the joint. The importance of bone in OA pathogenesis is now considered as fundamental and thus bone represent an interesting target for treatments. Tomodensitometric studies have shown an abnormal structure and organisation of the bone tissue of OA patients. In parallel, primary human OA osteoblast (Ob) cultures grown from the tibiofemoral subchondral bone show an abnormal phenotype and a reduced mineralization in vitro. The Wnt signalling pathway, known primarily for its important role in embryogenesis, is of utmost importance in osteogenesis and it has been shown to regulate terminal Ob differentiation. Transforming growth factor-β1 (TGF-β1), known to act on proliferation and on early phases of the differentiation of Ob, is overexpressed by OA Ob and could also modulate the Wnt signalling activity. Furthermore, two populations of OA patients can be discriminated in vitro by the production of prostaglandins E2 (PGE2) of their Ob. In a study on colorectal cancer, PGE2 was shown to modulate the canonical Wnt signalling pathway. Our hypothesis is that canonical Wnt signalling is diminished in OA Ob. This reduction is responsible for the poor mineralization and the abnormal phenotype of human OA Ob. Furthermore, DKK2, overexpressed in OA Ob and whose expression is controlled by TGF-β1, is responsible for the diminution of Wnt signalling activity, and PGE2 can partly correct this situation. The general goal of this study is twofold: 1) to demonstrate the role of TGF-β1, DKK2 and of PGE2 in the Wnt signalling activity; 2) to demonstrate the link between TGF-β1 and DKK2 and their effect on the abnormal OA Ob phenotype. We have shown in this study that the canonical Wnt signalling pathway is altered in OA Ob and that this was responsible for the altered phenotype observed in OA Ob. Also, we have shown that among the mediators of the Wnt signalling pathway, the expression of antagonist Dickkopf-1 (DKK1) was similar between OA and normal Ob. In contrast, the antagonist DKK2 was overexpressed and the expression of the agonist Wnt7B was low in OA Ob. Moreover, PGE2 increased Wnt signalling activity in OA Ob. The inhibition of DKK2 expression also increased Wnt signalling activity and corrected the abnormal phenotype along with increasing the mineralization of OA Ob. The inhibition of TGF-β1 expression, also overexpressed in OA Ob, also resulted in the correction of the phenotype and increased the mineralization of OA Ob. Inhibiting TGF-β1 expression also led to DKK2 inhibition. As the contrary was not observed, this demonstrated that TGF-β1 could regulate DKK2 expression. To conclude, Wnt signalling is reduced in OA Ob and this is due to elevated DKK2 levels in these cells. High levels of TGF-β1 in OA Ob increased DKK2 expression which could be responsible, at least partially, for their altered phenotype. PGE2 was shown to also increase Wnt signalling activity in OA Ob. Taken together these data suggest that such alterations in OA Ob could be responsible for the abnormal bone structure observed in OA patients.

Etude structurale et gîtologique du gisement de sulfures massifs à métaux de base de Hajjar : contribution à l'histoire tectono-métamorphique des Guemassa orientales, Maroc / Structural and gitological study of the Hajjar massive base metal sulfide deposit : contribution to the tectono-metamorphic history of Eastern Guemassa, Morocco

Admou, Safouane 08 December 2018 (has links)
The massive sulphide deposits hosted in the Moroccan Hercynian massifs of Jebilets and Guemassa are Volcanogenic Massive Sulfide (SMV), deformed and metamorphosed. However, an assessment of the contribution of this deformation to their current structure and the eventual impact on the economy and in terms of exploration is missing. Therefore, this contribution aims mainly to define the structural context of these mineralizations, in order to deduce a structural model relating to Hajjar's VMS.The VMS of Hajjar, belonging to the domain of Guemassa (Western Meseta). It is hosted in volcanosedimentary terrain composed of pelites, sandstones and lentils and limestone bars intruded by felic and basic sills and dykes. The main volcanic bodies correspond to the rhyolitic plugs. The Hajjar deposit is composed of 3 mineralized bodies: main body (CP); north-eastern body (CNE); western body (CWD) and extreme western body(CEWD.). The Hajjar deposit is affected by a single XY plane of subvertical flattening of directions N0 to N45. The host rock and sulphide bodies have the same silicate metamorphic assemblages, which exhibit either a "static" granoblastic texture when the strain is low or pre to syn-tectonic features when the foliation is strongly expressed. The texture shapes, and aggregates of the biotite and andalousite (± suspected cordierite) are typical of HT/LP “contact” metamorphism in the hornfels facies. The syn-tectonic assemblage is composed of quartz + chlorite + white micas (± calcite) and partially replaced the former biotite and andalousite blasts. The data from the N’Fis block show the occurrence of a single sub-vertical XY plane oriented N130. The biotites of contact metamorphism are elongated parallel to the foliation. These data imply that the Hajjar MSD and the N’Fis block are affected by a single foliation which encompasses a HT/LP contact metamorphism. The significant inflexion of foliation from Imarine outcrops to Hajjar VMS is due to major right-lateral ENE-trending transpressive shear zone at Hajjar. Near the Hajjar mine, such a regional shear zone is recognized for the first time, and is a structural key feature of the Hajjar district. The mineralization of Hajjar is strongly deformed and metamorphosed. It is affected by folding, foliation and mylonitic bands within a regional scale shear zone. Structures such as pyrrhotite-rich ribbons clearly pre-date the deformation and the HT/LP contact metamorphism. The early stage is characterized by the precipitation of pyrrhotite, pyrite, arsenopyrite, sphalerite, and galena, whereas the synkinematic stage constitutes major concentrator event characterized by the dissolution and recrystallization of early paragenesis with enrichment in Zn, Cu. The late post-kinematic stage is responsible for the neoformation of sulphides, namely chalcopyrite, pyrite and some traces of sphalerite. We have demonstrated a tectonic thickening within the fold hinge zone. pre-syntectonic metamorphism favors ductile behavior and sulfide recrystallization (Po, Cpy, and Sph). The polymetallic veins argue for hydrothermal fluid-assisted remobilization during deformation. All of our studies lead to the proposal of a 3D tectono-metamorphic model for the VMS of Guemassa and proposals to search extensions of the mineralization.

Le gisement d’or du Tocantinzinho (province aurifère du Tapajós) relations entre déformation, hydrothermalisme et minéralisation / Tocantinzinho gold deposit (Tapajós Gold Province) relationship between deformation, hydrothermal alteration and mineralisation

Borgo, Ariadne 23 February 2017 (has links)
The Tocantinzinho deposit is located on the Tapajós Gold Province and is the largest gold deposit within Province, with 53,9 tons of gold. Its formation begins with a granodioritic magmatism around 2005Ma, followed by a granitic magmatism 10 Ma latter. The Tocantinzinho granite is composed by two main facies, syenogranite (1996±2Ma) and monzogranite (1989±1Ma), and by aplite and pegmatite bodies, suggesting a fluid-rich magmatism at shallow depth. Andesite dikes (1998±8Ma) are intrusive in both rocks. Sharp fragments of those rocks along contacts and minor mingling with granitic magma suggest a multiphase magmatism at distinct timing. The first dikes have intruded within granite when it was crystalizing, thus a minimum age of 1975Ma was estimated. Cooling rates of plutonic rocks vary from 3.6 to 14.7°C/Ma, with an average of 7.5°C/Ma, suggesting vertical exhumation processes were minor. The elongated geometry of granite along with sin-magmatic strike-slip tectonics of andesite corroborate the predominance of horizontal movements. Geochemical analysis show high-K calk-alkaline affinity and niobium anomaly indicator of two possible geotectonic settings for these rocks: Andean-type continental arc or post-collisional one. Considering the genetic relationship between magmatism, strike-slip faults, and low cooling rates, a post-collisional setting is more likely. The geochemical signature, ages and style of tectonism allow us to compare those rocks with the ones from Creporizão Intrusive Suite (1997-1957Ma). Dacite dikes (1992±2Ma) cut across all other rocks, but the temporal relationship among them remains misunderstood, due to the geochemical signature similar to the anorogenic rocks, suggesting it belongs to a distinct and latter magmatic series. Indeed, the dated zircons were probably inherited from host rocks. The mineralized area is restricted to a domain constrained by two major sinistral strike-slip N100°-130E°E faults that comprises the Tocantinzinho granite and sub-volcanic rocks, which were hydrothermally altered, brittle deformed and mineralized during two phases. The first one is characterized by breccias and microfractures infilled with muscovite (1864±5Ma) and pyrite, which contains low gold grades and are restricted to the Tocantinzinho granite. The second phase was controlled by strike-slip and normal tectonics generating tension gashes veins and pull apart breccias infilled with quartz, chlorite, calcite, albite, rutile, pyrite, galena, sphalerite, chalcopyrite, and gold. The gold grade can reach up to 70 ppm in some sulfide-rich veins. These structures are parallel and mainly trends N30-60°E, showing textures and orientated minerals typical of syn-tectonic infilling. Based on petrographical features and argon ages two hypothesis were proposed for the ore genesis: the first one consider a genetic relationship between magmatism and ore fluids for first mineralization stage and the second hypothesis consider a reactivation of pre-existing faults by an extensional tectonism related to the Maloquinha Intrusive Suite magmatism (ca.1880Ma) for this phase. The second mineralization phase is considered as formed as consequence of tectonic reactivation at ca. 1880Ma, in both hypothesis. Both phases in both hypothesis were classified as magmatic-hydrothermal ore mineralization and might be classified as intrusion-related gold systems. However, new field works are important in order to identify and characterize the nature and source of hydrothermal fluids, as well as ore dating and new geochemical and geochronological data of sub-volcanic rocks are imperative to better understand the genesis and evolution of the Tocantinzinho gold deposit. Such results, strongly linked to the fact that the tectonic control seem significant, may help for future exploration and exploitation programs.

Adubação nitrogenada em pastagem tropical por meio de compostos orgânicos / Nitrogen supply of tropical pasture with organic compounds

Wendel, Carolina Façanha 02 October 2012 (has links)
The present work was based in the hypothesis that it is possible to obtain similar agricultural productivity levels applying organic compounds on soil in comparison with mineral nitrogen application. The experiment was conducted in pots with Brachiaria brizantha cv. Marandu in two soil types, with clay and sandy clay loam texture. The objective was to evaluate if estimated doses of nitrogen (N) from waste compost, sewage sludge compost and and pig manure compost, when applied on the soil, was equivalent to 150 mg dm-3 of mineral Nitrogen fertilizer. The doses of organic composts applied were based in estimations of N mineralized of 30, 15 and 7%, correspondent, respectively, to doses of 500, 1000 and 2143 mg dm-3 of total N. The grass was harvested three times, on each time the chlorophyll level, the number of leaves, number of branches, leaf area, biomass and total N were measured in the aerial part of the plants. The results were submitted to variance analysis, and regression analysis. It was possible, trough the N fertilization by organic composts, to obtain similar and even superior results, in comparison to the mineral N, depending on type of compost, the applied dose and the time frame. The treatment with sewage sludge compost in the dose of 1000 mg dm-3 of N was superior to the mineral N in the first harvest, similar in the second and superior in the third harvest of the grass. The waste and pig manure composts reached similarity with the mineral fertilization in the second harvest, in the dose of 2143 mg dm-3, and in the third harvest, in the dose of 1000 mg dm-3 of N. The mineralization fractions of N found for the composts that were the most similar the mineral fertilization were, in average, of 15% for the sewage sludge compost and 7% for the waste and pig manure compost

Molecular aspects of the effect of transforming growth factor-beta 1 (TGF-β1) in the signaling pathways in vitro biomineralization.

Donato, Tatiani Ayako Goto 11 March 2014 (has links)
This in vitro study aimed to evaluate the molecular effects of TGF-β1, with different supplementation time periods on the establishment of MC3T3-E1 cells, comparing with cells treated with AA+β-GP supplemented with Dex and/or TGF-β1, without or with neutralization of TGF-β1 receptors. The gene expression of the TGF-β1 and Smad3 were analyzed, as well as the osteoblast differentiation and biomineralization. The cells treated with TGF-β1 without neutralization of receptors have had inhibitory effect on some important stages of osteoblast differentiation and biomineralization in vitro, but expressed some important mineralization markers. Mineral nodules were observed in all treatments of cells with their TGF-β1 receptors neutralized, but there was a decrease in the expression of some important genes. The results confirm the complexity of the pathway signaling of TGF-β1, showing that there are gaps for understand the mechanisms of this molecule in the biology of osteoblasts.

Transformações do N derivado do fertilizante no solo e a eficiência de utilização pela cultura da cana-de-açúcar cultivada em solo coberto por palha / Transformations of N derived from fertilizer in soil and use efficiency by sugarcane grown in straw-covered soil

Megda, Michele Xavier Vieira 26 June 2013 (has links)
The addition of plant residue with high C/N ratio to the soil in a \"green cane\" system affects the balance of input/output processes of N in a soil-plant-atmosphere system. Thus, the presence of straw modifies the agroecosystem requiring adjustments in management of nitrogen fertilization. Accordingly, four studies were developed to: (i) evaluate the nutritional and productive characteristics of sugarcane crops to identify the source and N rates of greater agronomic efficiency; (ii) quantify the sugarcane N recovery from fertilizers; (iii) assess the potential inhibition of chloride ion in reaction to soil nitrification, (iv) evaluate mineralization and immobilization rates of fertilizer-N and their interaction with the native soil N. We conducted a field experiment on a Typic Hapludox with sugarcane, in the 2009/2010 harvest. The experimental design was a randomized block with four replications and nine treatments with nitrogen sources: ammonium sulfate, YaraBela NitromagTM, ammonium nitrate and urea at 100 kg N ha-1, ammonium chloride at N doses: 50, 100, 150 and 200 kg ha-1 and control. Microplots were installed with application of nitrogen sources labeled 15N at 100 kg N ha-1. Nitrogen fertilizers were applied manually over straw. Subsequently, previously collected soil samples were incubated aerobically for 21 days. The experimental design was completely randomized with four replications and the treatments consisted of ammonium sulfate or ammonium chloride at 100 mg N kg-1 with potassium chloride at a chlorine dose 100 or 200 mg kg-1. We also evaluated doses of ammonium chloride (50, 100, and 200 mg N kg-1). Forms of mineral-N were determined by Flow Injection Analysis (FIA) system. Soil samples were incubated aerobically for 20 weeks. The experimental design was completely randomized with three replications and six treatments, comprising a 3x2 factorial (amidic-N, ammonium- N or without N versus with or without cane residue). Nitrogen sources (enriched with 2 atom% 15N) were applied to the soil surface at a N dose of 100 mg kg-1 and residue sugarcane incorporated at 5.2 g kg-1. The N supply in ammonium chloride form resulted in decreased yield of sugar and stalks. Nitric-ammonium sources promoted higher activity of nitrate reductase in sugarcane leave; however, they did not affect total N accumulation. The sugarcane recovery of fertilizer-N was approximately 60% in early stages, dropping to about 20% near the maturity stage. The chloride ion reduced nitrate concentration in soil due to the anion action in the nitrification reaction. The sugarcane residue incorporation to the soil showed higher N immobilization rates and the use of ammonium sulfate resulted in higher N mineralization rates of native soil N compared to urea.

Influence de l'herbier de Zostera noltei sur la méiofaune benthique et la géochimie de sédiments intertidaux du Bassin d'Arcachon / Influence of Zostera noltei meadows on benthic meiofauna and geochemistry of the intertidal sediment of Arcachon Bay

Cesbron, Florian 13 March 2015 (has links)
This thesis aimed to study the geochemical microenvironments created by Zostera noltei meadows and its root system in Arcachon Basin and to evaluate the effect of these microenvironments 1) on living benthic foraminifera ecology, a group of organisms present in large densities in sediment which impact on marine biogeochemical cycles remain poorly known and 2) on the chemical species and associated benthic fluxes. To fulfill these objectives, a multidisciplinary strategy was developed. The study of benthic foraminiferal ecology at centimeter scale, conducted in sediments with or without vegetation in February and July 2011, has shown the presence of three major species with specific metabolisms. Zostera noltei meadows seem to influence the densities and the living depth of heterotrophic (Ammonia tepida) and mixotrophic (Haynesina germanica) calcareous species, present in the sediment surface. The third species (Eggerella Scabra), which anaerobic metabolism is still to discover, is found throughout the sedimentary column and seems to prefer the organic matter coming from the degradation of the seagrass. In surface sediment, the contribution of these three species to aerobic remineralization was estimated at 7%, i.e. 5 times more than the maximum rates previously recorded in marine environments. The presence of the seagrass also influences geochemistry where enriched iron and depleted sulfide structures are observable in 2D thanks to DET-DGT gels developed here. Dissolved phosphorus depended on seagrass uptake and was highly concentrated only as scattered spots. A slice of sediment, taken face to face with the DET-DGT gel, was also conducted during this study aiming to map the solid phase and foraminiferal density. Despite this work is still in progress, methods such as X-ray microfluorescence and microtomography showed promising perspectives. New research pathways have been opened through technological developments and innovative approaches combining physiology, ecology, and geochemistry.

Estudos petrográficos e geocronológicos do prospecto Mankombiti, Cinturão de Fíngoè, Província de Tete, noroeste de Moçambique / not available

Mahanjane, Laura Nilza Mendes 27 November 2013 (has links)
The Fíngoè belt is a tectonic unit of West Gondwana Supercontinent, comprised of supracrustal rocks formed at approximately 1.33 Ga. These rocks are carriers of Au, Cu, Zn, Pb and Fe mineralization. With about 150 km long and oriented WSW- ENE, this belt extends from west of the Atchiza Mount to about 30 km east of the Fíngoè village in the north Mozambique, and consists of an extensive variety of metasedimentary and metavolcanic rocks. The Fíngoè metallogenic belt provides specialization for Au and Cu, which have been exploited in the past in various locations. Many of these records are associated with magnetite and malachite. Currently, the prospecting and mineral exploration have been conducted by the African Queen Mines Ltd mining company, through a project called the King Solomons, located in the central part of it. The geology of the studied area shows a predominance of carbonate rocks, granites and gabbros. The carbonate rocks are considered important in the genesis of the skarn deposits type and deposits of replacement for base metals and Au due to its highly reactive nature. The geochronological data performed in this work indicate a precise age of the 1079.1 ± 8.2 Ma for the intrusive granitic rock associated with mineralization, which could be interpreted as the age of the skarn and consequently of the mineralization. However the age obtained for the hydrothermal alteration that affected the gabbro at about 657 ± 36 Ma and the Pb-Pb model age suggested by the model of Stacey and Kramers (1975) for primary chalcopyrite mineralization at 725 Ma, suggesting that the mineralization of the Mankombiti prospectus was developed during the Neoproterozoic time. For understanding the processes that were involved in the skarn-type mineralization processes, two hypotheses are here considered: (i) could be admitted the presence of i ntrusive granitic rocks at 700 Ma, situated in depth, which have not yet been characterized, as responsible for the heat source required, (ii) may be admitted occurrence of an extensional event at 700 Ma to produce a thinning of the lithosphere and the uplift of the asthenosphere, producing high thermal flows generating the mineralization fluids.

Isso evidencia a necessidade de mais estudos acerca da dinâmica do N nesses ambientes, principalmente em florestas em restauração / Nitrogen (N) is an essential element for all organisms; however, the most abundant nitrogen compound, dinitrogen (N2), is assimilable only by a few microorganisms. This makes N limiting, which reflects in its availability via organic matter (OM) mineralization and nitrification, which produce ammonium (NH4+) and nitrate (NO3-), respectively. In soils, these processes are regulated by factors such as OM and soil water content, pH and temperature. In forest ecosystems, tropical and boreal, these processes are very relevant and still poorly understood, especially in secondary forest previously deforested. The land use change causes alterations in cycling and availability of N, regulating factors, and emission of nitrous oxide (N2O), a greenhouse gas. Forest recovery patterns are quite distinct among different rainforests, as seen for Atlantic and Amazon rainforest, with very high levels of mineralization in young forests (10 years) in Amazonia (20.9 μg N2O-N g-1 SWD h-1) and low in the Atlantic Forest (3.2 μg N2O-N g-1 SWD h-1). Nitrification had a similar pattern between them, with low rates in both young forests (0.6 and 6.8 μg N2O-N g-1 SWD h-1 in the Atlantic and Amazon forest, respectively), indicating a conservative N cycling, avoiding losses through gas emission and NO3- leaching. However, the N2O emission in restoration area in the Atlantic Forest was higher than in pristine (22 and 2.5 μg N2O-N g-1 SWD h-1, respectively) due to the higher soil temperature, which stimulates microbial production of N2O. In terms of pristine forest, the nitrification rate was very different between the Atlantic forest and Amazonian forest (0.08 and 15.9 μg N2O-N g-1 SWD h-1, respectively), showing the different regulatory factors of each tropical region, such as rainfall regime, vegetal composition, soil types, etc. In boreal forests, OM, soil water content and soil pH were the main regulators, limiting the availability of N in conifer-dominated well-drained forests compared to peat-dominated poorly drained forests. In this way, we verified that mineralization and, mainly, nitrification are very relevant processes in the control of N, and even in extremely different ecosystems, the regulating factors are often the same. This evidences the need for more studies about N dynamics in these environments, especially in restoration forests
