Ludic Landscapes: Liberating Landscape Architecture through the Brilliance of Carnival

In the age of an overworked society, struggling to emotionally connect, play is essential. Ludic Landscapes, explores the reenchantment of undervalued landscapes, in particular the industrial site of the Canadian Pacific Railway yards in Winnipeg, Manitoba, Canada. If carnivals are landscapes devoted to play, as rail yards are landscapes devoted to work and industry, what happens when play is brought into a landscape of work? This practicum examines how ludic activities can be integrated into a landscape previously devoted to work - resulting in a reenchantment of an industrial landscape. The term reenchantment implies a positive change in perspective, an emergence of magic, toward something formerly disregarded. The explorations of undervalued and overlooked landscapes within the urban environment begin to unearth a curious history. Many carnival rides of the past germinate from technology developed for work, industry and progress. When the carnival is liberated of its artifice, its heart is shown to be machine technology utilized for work. It is possible to put these amusement rides through one more evolution, to make them not only fun, but also functional. The marriage of work and play results in remediating and reenchanting the landscape through ludic activity, spectacle and wonder. This is a poetic act.

A aplicação tópica de corticosteróides intranasal é amplamente reconhecida como primeira linha de tratamento antiinflamatório. Espera-se que a maioria dos sprays nasais prescritos como drogas de ação local, ainda não possuam patente permitindo o aumento de cópias genéricas desses medicamentos, levando a concorrência e redução de preço. Estudos de bioequivalência para sprays nasais estão ainda em discussão. Geralmente, os estudos para essa finalidade usa modelos de intervenção terapêutica a longo prazo, com altos custos para o paciente e longo tempo de duração. O objetivo deste trabalho foi mostrar a aplicabilidade da provocação nasal com histamina e rinomanometria em estudos de bioequivalência para sprays nasais. Trata-se de um estudo aberto, cruzado aleatorizado, utilizando dois períodos e duas seqüências para avaliar a equivalência farmacodinâmica entre duas formulações de sprays de dipropionato de beclometasona de manufaturamento distinto. Após estímulo nasal com histamina (0,5 mg/ml em ambas narinas), 25 voluntários saudáveis foram submetidos a rinomanometria anterior nos tempos 0; 15; 30 e 60 minutos para estabelecimento do fluxo, pressão e resistência basal de cada câmara nasal. Os voluntários foram submetidos à utilização do spray nasal com a droga teste (T) ou referência (R) de acordo com o esquema de randomização, e a área sobre a curva foi analisada. De acordo com os parâmetros estudados os resultados obtidos mostraram diferença significativa entre as duas formulações (p= 0,31) indicando a equivalência terapêutica entre as drogas T e R. Assim, estes resultados sugerem que provocação nasal com histamina em indivíduos saudáveis e o emprego da rinomanometria como método quantitativo das alterações provocadas nas câmaras nasais possam ser aplicáveis em estudos farmacodinâmicos de bioequivalência para sprays nasais / Abstract: Allergic rhinitis is a common condition with widespread morbidity, increased medical treatment costs, reduced work productivity and lost school days. Topically delivered intranasal corticosteroids are widely recognized to be the first-line anti-inflammatory treatment. It is expected that many of the most-prescribed nasal sprays for local action drugs will go off patent, allowing the increase of the generic copies of these medications, with more products competition and price reduction. The bioequivalence studies for nasal sprays are still in discussion. Usually the studies designs for this purpose use a long-term therapeutic intervention models using patients with high costs and time duration. This study was designed to demonstrate the feasibility of rhinomanometry in bioequivalence studies for nasal sprays. Study design, an open, randomized, crossover study, using two periods and two sequences to evaluate pharmacodynamic equivalence between two formulations of beclometasone dipropionate spray. After nasal challenge with histamine (0.5mg/ml, in both nostrils), 25 healthy volunteers were submited to an anterior rhinomanometry at the time 0; 15; 30 and 60 minutes building a baseline of flow, pression and resistance of nasal chamber. Then, the vontuteers were submited to nasal drug spray (Test (T) or Reference(R)), according to randomized schedule and the Are Under Curve (AUC0-t) analyzed. ABSTRAKT Detta kandidatarbete består av en enda frågeställning som undersöks med stöd av relevant forskning och författarens egen prövande gestaltning. De begrepp som fokuseras på i detta arbete är situerad kunskap och intra-aktion, men även ett antal andra relevanta medietekniska begrepp tas upp. Arbetet ämnar undersöka provokationer när det gäller ljud och bild som står i kontrast till varandra samt hur begreppen intra-aktion och situerad kunskap kan hjälpa medietekniker att medvetet påverka människors upplevelse av konstinstallationer. I detta kandidatarbete så har fyra installationer designats och producerats samt att experiment utförts på ett antal testpersoner. Installationerna i detta kandidatarbete är skapade med Logic Pro X (Apple Inc 2017) och visades för testpersonerna med hjälp av en skärm och tillhörande ljud genom ett par hörlurar. The concepts that are focused on in this bachelor thesis are situated knowledge and intra-action, but also a number of other relevant media technical concepts are addressed. The goal is to investigate provocations in terms of sound and image that stand in contrast to each other and how the concepts of intra-action and situated knowledge can help media technicians to consciously influence people's experience of art installations. In this bachelor thesis, four installations have been designed and produced, and experiments have been performed on a number of test subjects. La confirmation d’une hypersensibilité médicamenteuse est importante car la plupart des cas allégués ne sont pas confirmés. Le diagnostic repose sur l’interrogatoire et le bilan allergologique, ce dernier comprenant surtout des tests in vivo. Ces tests ne sont pas dénués de risque. Ce travail se propose de répondre avec une méthodologie originale (pour le domaine de l’hypersensibilité médicamenteuse), à des questions en suspens visant la nécessité, les modalités et l’utilité du bilan allergologique, en prenant principalement mais pas exclusivement le modèle des hypersensibilités aux ß-lactamines. J’ai utilisé plusieurs approches méthodologiques, appliquées à une large base de données d’hypersensibilités médicamenteuses. Dans un premier temps, j’ai exploité les données cliniques rétrospectives afin de construire 2 modèles de diagnostic d’hypersensibilité aux ß-lactamines et j’ai ensuite testé leurs performances diagnostiques sur un échantillon prospectif de patients. Les modèles atteignent globalement une sensibilité de 50%, ce qui est difficilement acceptable, dans un contexte iatrogénique. Secondairement, j’ai réalisé le passage des protocoles de tests de provocation aux ß-lactamines, d’une étape avec paliers purement empiriques, à un protocole basé sur des données issues d’une analyse de survie. Autres 3 articles ont suivi une méthodologie similaire : les patients ayant eu un bilan allergologique négatif pour un médicament donné ont été re-contactés et interrogés au moyen d’un questionnaire. Le service rendu au patient a été calculé par le taux de patients ayant repris (sans réaction) le médicament autorisé (plus de 90% pour 3 classes médicamenteuses analysées). / Most alleged cases of hypersensitivity reactions following drug administration are actually ruled out by drug allergy work up. The diagnosis is based on clinical history and allergy tests, mainly in vivo tests. These tests carry a considerable risk of iatrogeny. The purpose of this thesis was to address some unmet needs regarding the need, technical aspects and utility of the drug allergy work up, using an original methodology applied to the drug hypersensitivity field. It focuses mainly (but not only) on drug hypersensitivity reactions to ß-lactams. I used different statistical methods, applied to a large database, the Drug Allergy and Hypersensitivity Database. First (Article 1), I used retrospective clinical data to build 2 models for ß-lactam hypersensitivity diagnosis. I then tested these models on a prospective sample, in order to analyze their diagnostic performances. The overall sensitivity of the 2 models is around 50%, which is unacceptable in an iatrogenic context. Second (Article 2), I worked on empirical protocols of drug provocation tests and I identified steps for data-driven, evidence-based protocols by means of survival analysis. The Articles 3, 4 and 5 were conceived following a similar methodology: patients with a negative drug allergy work-up for a certain drug were called and questioned on whether they had been exposed to this same drug, following allergy tests. High negative predictive values of these tests, with more than 90% of patients tolerating subsequent administration without any hypersensitivity reaction, were obtained for 3 different drug classes.
