L’émergence des déterminants de résistances plasmidiques aux quinolones (PMQR), auxquels appartient le gène qnrD, participent de manière significative à la sélection des résistances de haut-niveau aux permet les réparations de l’ADN lors des stress soumis aux bactéries, et d’autre part, que les aminosides, une autre classe d’antibiotiques que les fluoroquinolones, induisaient la réponse SOS chez Escherichia coli. En effet, nous avons montré que les petits plasmides-qnrD chez E. coli, induisent la formation de monoxyde de nitrogène et l’inhibition de la voie de détoxification Hmp-dépendante. Ces processus génèrent des lésions à l’ADN qui s’ajoutent à celles occasionnées par les aminosides concourant à activer la réponse SOS chez E. coli. L’ensemble de nos résultats montrent que l’émergence de la résistance aux fluoroquinolones peut être occasionnée par l’exposition d’E. coli à une autre classe d’antibiotiques, ici les aminosides. / The emerging plasmid-mediated quinolones resistance (PMQR) determinants significantly participate in the selection of high-level of resistance to the major antibiotics fluoroquinolones, leading to numerous clinical failures. In this study, we reported for the first time that PMQR expression could be triggered by the fluoroquinolones but also by another major class of antibiotics, the aminoglycosides. We were able to show that this unique cross selection of antibiotic resistance was the consequence of the PMQR determinant qnrD being SOS-regulated in a RecA-LexA dependent manner. We demonstrated that sub inhibitory concentration of aminoglycoside induced nitric oxide formation associated with the repression of the Hmp-mediated detoxification pathway, resulting in the induction of the SOS response and thus up-regulation of the PMQR. Overall, our findings revealed an unexpected antibiotic resistance cross-selection with low aminoglycosides concentrations promoting emergence of fluoroquinolones resistance.

The NGOs are also frequent users of advertisement as a method of publicizing their actions. This research aims to investigate the discourse of NGOs on mass media, focusing advertisement, and we consider such advertisement as products consumed in a symbolic way. Hence, the goal of this research is to understand the interactions between such advertisements and the culture of consumption. In this research, we assume that consumption, with its broad manifestations and social representations, has great influence over the production of NGOs public narratives under the form of advertisements. For the purpose of the research, we address the following aspects of the problem: the context of civil society and citizenship; the field of publicity; the overlapping of the Third Sector with the marketing, propaganda, and adv ertisement sector; and the concepts and paradigms involving the Environmental Movement, because we have chosen, as a corpus for the discourse analysis of publicity, ads from SOS Mata Atlântica Foundation, an environmental NGO active in Brazil since 1987. The chosen methodology is the French tradition of Discourse Analysis (DA), applied both on the publicity narratives and on the Mission of the Foundation. We carried out a comparative study between the discourses produced during the establishment of the Foundation and the contemporary discourses. The analysis demonstrates the existence of a tension zone between the publicity discourse and the Mission of the NGO. As a result, we explain the move of the Mission towards the publicity narrative. This move has brought the Foundation discourse closer to the rhetoric of the market, showing an intention to produce a symbolic image of the Foundation in the same way the market rhetoric produces the brand of goods: focusing on publicity communication to build its "brand" from a discourse already consumed by society instead of publicizing their cause. / As Organizações Não Governamentais (ONGs), desde a década de 1980, vêm ganhando notoriedade no Brasil, decorrente de suas atuações no âmbito social e socioambiental, em resposta a demandas da sociedade brasileira, sendo também anunciantes assíduas no cenário publicitário. A pesquisa se propôs a investigar os discursos publicitários de ONGs, produtos consumidos de forma 0simbólica, na busca das prováveis mutações e interações desses com a cultura de consumo. Entendemos que o consumo, com suas abrangentes manifestações e representações sociais, exerce grande influência na produção das narrativas publicitárias de ONGs. Para o desenvolvimento da pesquisa, abordamos o contexto da sociedade civil e da cidadania; o campo da publicidade; os imbricamentos do Terceiro Setor com a publicidade do mercado; e os conceitos e paradigmas que envolvem o Movimento Ambiental, pois escolhemos como corpus para análise dos discursos publicitários, os anúncios da Fundação SOS Mata Atlântica, ONG ambientalista atuante no Brasil desde 1987. A metodologia utilizada foi a Análise do Discurso de linha francesa - AD - aplicada nas narrativas publicitárias e na Missão da Fundação, no sentido da realização de um estudo comparativo entre os discursos produzidos no período da constituição da Fundação e os discursos contemporâneos. A análise demonstrou a existência de uma zona de tensão entre o discurso publicitário e a Missão da ONG. Entendemos o deslocamento da Missão da narrativa publicitária, que aproximou o discurso da Fundação da retórica utilizada pelo mercado, como resultado de uma intenção de produção simbólica da imagem da ONG, na mesma forma das marcas de mercadorias, privilegiando na comunicação publicitária a construção de sua "marca", a partir de um discurso já consumido pela sociedade, em detrimento da divulgação de sua causa.

A leptospirose é uma zoonose de importância mundial, e é causada por bactérias patogênicas do gênero Leptospira, pertencente à ordem Spirochaetales. Os seres humanos são hospedeiros acidentais e os surtos de leptospirose ocorrem em grandes centros urbanos após enchentes contaminadas por urina de ratos. Existem poucas informações a respeito de como Leptospira spp lida com situações de estresse induzidas pelo hospedeiro e pelo ambiente. O ferro é um íon essencial para a maioria dos seres vivos. A regulação de genes envolvidos por seu aporte e estoque na célula bacteriana é mediada por proteínas da família de reguladores transcricionais, Fur (ferric uptake regulator). L. interrogans sorovar Copenhageni possui quatro ortólogos para Fur, que foram alvo de estudo deste trabalho. A caracterização destes genes foi realizada através de estudos evolutivos, determinação do seu padrão de expressão em modelo animal e análise de modelagem estrutural. Durante o andamento do mestrado, ensaios paralelos revelaram resultados promissores na análise de expressão de genes relacionados ao sistema SOS, um mecanismo de resposta bacteriano a danos no material genético. Assim sendo, o estudo de caracterização de expressão em modelo animal suscetível e resistente à doença foi ampliado. Ensaios de qRT-PCR de cDNAs provenientes de pulmão, rim e fígado permitiram a identificação de dois genes que foram expressos quase que constitutivamente ao longo de toda a infecção em todos os órgãos e organismos estudados: fur979 e recA. Os demais foram requeridos em dias específicos da infecção. Quanto aos componentes do sistema SOS, observamos padrão de expressão específico para o rim, no quinto dia após a infecção. Para os estudos evolutivos de Fur foi gerada uma árvore filogenética que revelou o agrupamento de duas sequências da família Fur de Leptospira interrogans sorovar Copenhageni em ramos fechados com sequências muito similares a proteína Fur e Zur de Escherichia coli. Os outros dois ortólogos agruparam com as proteínas correspondentes nas demais espécies de Leptospira. Uma destas sequências apresentou padrão evolutivo específico dentre as espécies patogênicas. A modelagem da estrutura terciária, confirmou o padrão evolutivo obtido em nossa inferência filogenética. / Leptospirosis is a worldwide zoonosis caused by pathogenic bacteria from the genus Leptospira, order Spirochetales. Human beings are accidental hosts, and leptospirosis outbreaks occur in large urban centers after contact with contaminated waterby rodent urine. There are few informations concerning the mechanisms employed by Leptospira sppto deal with the stress induced by the host and the environment Iron is an essential ion to most of living beings. The regulation of genes involved in its uptake and maintenance in the bacterial cell is mediated by the transcriptional regulator family proteins, Fur (ferric uptake regulators). L. interrogans serovar Copenhageni possesses four orthologues for Fur, which were the focus of this work. The characterization of Leptospira Fur genes was done through evolutive studies, determination of their expression pattern on animal model and structural modeling analysis. In parallel, some experiments presented promising results for the expression analysis of genes related to the SOS system, a bacterial response mechanism to DNA damage. Therefore, the gene expression characterization on susceptible and resistant animal model was amplified. qRT-PCR experiments of cDNA from lung, kidney and liver allowed the identification of two genes expressed almost constitutively during the infection in all organs and organisms : fur979 and recA. The others were required in specific days of the infection. Curiously, the SOS system components showed specific expression pattern in the fifth day after inoculation, in kidney. For the Fur evolutive studies, a phylogenetic tree was inferred, revealing the clustering of two Fur family sequences from Leptospira interrogans serovar Copenhageni in closed branches with very similar sequences to Fur and Zur proteins from Escherichia coli. The other two orthologues clustered with corresponding proteins in the other Leptospira species. One of these sequences presented a specific evolutive pattern among pathogenic species. The tertiary structure modeling confirmed the evolutive pattern obtained in our phylogenetic inference.

<p>The purpose of this project is to study the women involved in the SOS Children’s Villages; the influence of the association on their views on life, as seen from a human-rights and a religious perspective. Questions such as why they chose to work with the association and what it gives them to do so are treated in this study.</p><p>My goal has been to study and present different aspects of something so important, but yet so unfamiliar. The method used in this project is minor field studies, which means visiting the villages, living with the families and observing their daily lives. The means of acquiring the information necessary for this study is by qualitative interviews with the mothers.</p><p>A theoretical framework has been used as a complement to the study, and it is also used to bring greater understanding to the SOS mothers and how they have shaped their lives.</p><p>My ambition with this essay is to bring out their individual experiences on how they view their own lives at present and what meaning life has given them. I have used seven of the 14 interviews conducted.</p><p>The outcome shows that several factors play important roles as to why the mothers decided to work with the association. Such factors were socioeconomic and sociopsyhologic factors. Other conclusions that were drawn after this field trip were that the women are very vulnerable to the social situation in Lebanon. They need someone to support them since the men are the primary providers. If the women do not find someone to marry they need to find another source for provision and the SOS children’s Association is one way to go. The sense of Coherence that these women had was indeed strong, they felt meaningfulness in what they did, they had comprehended the situations at hand and could manage the situations as predictable, and they have the confidence to know that everything will work out in the best way possible.</p> / Uppsatsen har givits ut som bok 2009 med titeln: "Lebanese SOS Children's Villages: Stories of the village mothers" av VDM Verlag, Saarbrücken.

Syftet med denna studie är att studera mediernas rapportering kring ett antal specifika händelser kopplade till SOS Alarm. Med vår undersökning avser vi att bidra till kunskapen om hur tidningarna Aftonbladet och Dagens Nyheter framställer SOS Alarm i sina nyhetspubliceringar. Mediernas rapportering är viktig att studera då mediebilden av SOS Alarm kan påverka allmänhetens förtroende för denna viktiga samhällsaktör. Detta är viktigt på grund av att den bild och på det vis som medierna framställer olika händelser påverkar människor, vår sociala och kulturella kontext, att se och uppfatta verkligheten vi lever i. Medierna utgör ett kraftfullt verktyg för människors åsikter, värderingar och påverkar människors beteende. Studien har använt sig av kvalitativ textanalys och inriktat sig på 43 tidningsartiklar från Aftonbladet och Dagens Nyheter. Med grunden i valda teorier diskuteras de resultat som undersökningen visar, med fokus på dramatisering. Vår slutsats är att båda tidningarna, men främst Aftonbladet har skapat en vinklad bild av olika händelser, genom sin frekventa användning av dramatisering i texterna. Vinklingen visade sig vara negativ, vilket beror på valet av ord och sammanhang som tidningarna gjort. / This study aims to study the media’s reporting on a number of specific events linked to the SOS Alarm. In our study, we wish to gain an insight into how the newspaper Aftonbladet and Dagens Nyheter presents SOS Alarm in their news releases. Media reporting is important to study because the media image of SOS Alarm can affect public confidence in this actor of society. This is important because the image and the way media produce different events affect people, our social and cultural context, to see and perceive the reality we live in. The media is a powerful tool for people’s options, values and affect people’s behavior. The study has used the method of quality content analysis and focused on 43 newspaper articles from Aftonbladet and Dagens Nyheter. With the foundation of the selected theories discussed the results that the investigation concludes with a focus on the dramatization. Our conclusion in the analysis of reporting on SOS Alarm is that both papers, but mainly Aftonbladet has created a biased and partially inaccurate picture of different events by the frequent use of dramatizations in the texts. The bias proved out to be negative, which depends on the choice of words and context in which the newspapers have done.

Orientador: Marcelo Carvalho Minhoto Teixeira / Resumo: Neste trabalho são propostos novos métodos de controle chaveado para uma classe de sistemas não lineares incertos utilizando a decomposição em soma de quadrados. Inicialmente é apresentada uma revisão dos conceitos e projetos de controladores baseados em desigualdades matriciais lineares (do inglês Linear Matrix Inequalities - LMIs) e a decomposição em soma de quadrados (do inglês Sum of Squares - SOS), buscando evidenciar as diferenças e vantagens das metodologias para a área de controle. Comumente são utilizados modelos fuzzy para realizar a análise da estabilidade e projeto de controladores para sistemas não lineares, e estes modelos podem ser classificados de acordo com a parte consequente linear ou polinomial. Busca-se neste trabalho evidenciar as diferenças entre os dois modelos fuzzy e a metodologia para projeto de controladores. Para o caso de sistemas cujas dinâmicas podem ser descritas apenas por funções polinomiais, serão consideradas incertezas politópicas. Então, visando flexibilizar o projeto utilizando um controlador composto por um único ganho polinomial e aumentar a região de factibilidade, são propostos controladores com ganhos polinomiais chaveados. O objetivo desta lei de chaveamento é minimizar a derivada da função de Lyapunov empregada no projeto. Considerando uma classe de sistemas não lineares mais geral, são propostos controladores com ganhos chaveados para modelos fuzzy polinomiais. (Resumo completo, clicar acesso eletrônico abaixo) / Abstract: In this manuscript new control methods are proposed for a class of uncertain nonlinear systems using a sum of squares decomposition. Initially is presented a revision of concepts and control design procedures based on Linear Matrix Inequalities (LMIs) and on sum of squares (SOS) evidencing the differences and advantages of these methodologies in the control system design. Fuzzy models are commonly used to perform stability analysis and controller design for nonlinear systems, and can be classified by a linear or polynomial consequent model. A goal of this dissertation is to compare these two methodologies in the control system design of a class of uncertain nonlinear systems. For the case of systems whose dynamics can be described only by polynomial functions will be also considered polytopic uncertainty. Therefore, in order to make the design more flexible than that obtained with only one controller with polynomial gain and increase the feasibility region, a new procedure for designing controllers with switched polynomial gains is proposed. The purpose of this switching law is to minimize the time derivative of the Lyapunov function employed in the design. For a more general class of nonlinear systems, controllers with switched gains for polynomial fuzzy models are proposed. The proposed methodology does not require the knowledge of the membership functions for an implementation of the control law. Caracterization of the SOS response in Leptospira interrogans sorovar Copenhageni / Caracterização da resposta SOS em Leptospira interrogans sorovar Copenhageni

Leptospira is a basal genus in an ancient group of bacteria, the spirochetes. The pathogenic species are responsible for leptospirosis, a disease with worldwide distribution and of public health importance in developed tropical countries. L. interrogans serovar Copenhageni is the agent for the majority of human leptospirosis in Brazil. In this work, we used a great variety of experimental approaches to characterize the SOS system in this serovar, to identify its impact in general DNA damage response, as well as to assess the DNA repair toolbox owned by pathogenic and saprophytic leptospires. We identified an additional repressor LexA, acquired by lateral gene transfer, exclusively in serovar Copenhageni. We also observed that UV-C irradiation led to massive death of cells and blockage of cell division in the survivors. Both repressors were active and we identified the sequences responsible for binding to promoters. However, the LexA1 SOS box was redefined after a de novo motif search on LexA1 ChIP-seq enriched sequences. This regulator was able to bind to at least 25 loci in the genome. DNA damage also caused a massive rearrangement of metabolism: increase in expression was observed in transposon and prophage genes, in addition to DNA repair pathways and mutagenesis inducers; on the other hand, motility, general metabolism and almost all virulence genes were repressed. Two induced prophages provided several proteins with useful functions. We also assessed the DNA repair-related genes presented by the three species of Leptospira: the saprophytic L. biflexa, the facultative pathogen L. interrogans and the obligatory pathogen L. borgpetersenii. There are more diversity and redundancy of repair genes in L. interrogans in comparison with the other species. Lateral gene transfer seems to be an important supplier of DNA repair functions. In addition, leptospires share characteristics of both Gram-positives and Gram-negatives bacteria. Representative genes from several different pathways were induced during infection of susceptible mice kidneys, suggesting DNA repair genes are active while causing disease. All these data suggest mobile genetic elements are the major forces in leptospiral evolution. Moreover, during DNA damage response, several SOS-dependent and independent mechanisms are employed to decrease cell growth and virulence in favor of controlled induction of mechanisms involved in genetic variability. / Leptospira é um gênero basal em um grupo já considerado um dos mais ancestrais, as espiroquetas. As espécies patogênicas são responsáveis pela leptospirose, uma doença presente em todo o mundo e de principal importância em países tropicais em desenvolvimento. L. interrogans sorovar Copenhageni é o agente da maior parte dos casos no Brasil. Nesse trabalho, utilizamos diversas abordagens experimentais para caracterizar o sistema SOS nesse sorovar, identificar seu impacto na resposta geral a danos no DNA, assim como avaliar as funções de reparo de DNA disponíveis em leptospiras patogênicas e saprofíticas. Identificamos um repressor LexA adicional, adquirido por transferência horizontal e exclusivo do sorovar Copenhageni. Observamos também que irradiação por UV-C causou significativa morte celular e bloqueio da divisão celular dos sobreviventes. Ambos os repressores são ativos e identificamos as sequências que utilizam para se ligar aos promotores dos genes regulados. Entretanto, o SOS box de LexA1 foi redefinido após uma busca de novo por motivos enriquecidos nas sequências recuperadas por ChIP-seq. Esse regulador ligou-se ao menos a 25 locais do genoma. A maioria desses alvos teve aumento de expressão após UV-C. Danos no DNA também causaram um importante rearranjo metabólico: houve aumento de expressão em transposons e profagos, além de indutores de mutagênese e vias de reparo; por outro lado, mobilidade, crescimento celular e quase todos os fatores de virulência foram reprimidos. Dois profagos induzidos durante essa resposta, possivelmente proporcionam algumas proteínas de funções importantes. Nós também avaliamos a presença de genes envolvidos no reparo de DNA em três espécies de leptospira: L. biflexa, L. interrogans e L. borgpetersenii. L. interrogans é a espécie com maior diversidade e redundância de genes de reparo. Além disso, transferência horizontal parece ser um importante fornecedor de funções de reparo nesse gênero. Leptospiras também apresentam genes característicos tanto de bactérias Gram-positivas quanto Gram-negativas. Genes representando diferentes vias de reparo foram induzidos durante infecção em modelo animal, sugerindo que essas vias estão ativas no curso da doença. Todos esses dados, em conjunto, sugerem que elementos genéticos móveis são de extrema importância na evolução do gênero e das vias de reparo. Assim, durante a resposta a danos no DNA, diversos mecanismos dependentes e independentes de SOS são empregados para frear o crescimento celular e virulência em favor da indução controlada de mecanismos para aumentar variabilidade genética.

It is characterized by small fruit trees that have great therapeutic potential. The leaves of E. uniflora are rich in hydrolyzable tannins, and the ellagitannin oenothein B has been studied for its remarkable antitumor activity, representing a possibility for the treatment of some cancers. However, there is a need for evaluation of possible mutagenic, genotoxic and toxic effects, which may result in damage to the human body. Three doses of oenoteina B (25, 50 and 75 mg/kg) were used in the micronucleus test, and the same doses along with mitomycin C. Five doses of oenoteina B were used in SOS inductest (10, 20, 50, 100 and 200 μg/plate) to evaluate the possible genotoxic or antigenotoxic action and examining the cytotoxic or anticytotoxic activity of this tannin. Our data showed that oenoteina B did not exhibit genotoxic effects, and furthermore, this ellagitannin demonstrated protective action against micronucleus formation induced by mitomycin C in mice. The possible use of oenoteina B as a chemopreventive and/or therapeutic agent still needs further studies using different tests for genotoxicity, applied to longterm, pre- and post-treatments, to investigate the mechanism of action of this compound. / A espécie Eugenia uniflora L. pertencente à família Myrtaceae, conhecida por pitangueira, é caracterizada por árvores frutíferas de pequeno porte que possuem grande potencial terapêutico. As folhas de E. uniflora são ricas em taninos hidrolisáveis, sendo que o elagitanino oenoteina B tem sido estudado por apresentar notável atividade antitumoral, representando uma possibilidade para o tratamento de alguns tipos de câncer. Contudo, há a necessidade de uma avaliação dos possíveis efeitos mutagênicos, genotóxicos e tóxicos, que podem provocar no organismo humano. Foi realizado o teste do micronúcleo, no qual foram utilizadas três doses do tanino (25, 50 e 75 mg/Kg) e as mesmas doses juntamente com mitomicina C. Também foram utilizados diferentes doses de oenoteina B no induteste SOS (10, 20, 50, 100 e 200 μg/placa) com objetivo de avaliar a possível ação genotóxica ou antigenotóxica e ainda analisar a atividade citotóxica ou anticitotóxica deste tanino. Nossos dados mostraram que oenoteina B não exibe efeitos genotóxicos e, além disso, este elagitanino demonstra ação protetora contra a formação de micronúcleos, induzidos por mitomicina C em camundongos. O possível uso de oenoteina B como um agente quimiopreventivo e/ou terapêutico ainda necessita de mais estudos usando diferentes testes de genotoxicidade, aplicados em longo prazo, além de ensaios com pré e póstratamentos, para a investigação do mecanismo de ação deste composto.
